As a precondition for lilac mass propagation, the optimal shoot-multiplication medium for Syringa × hyacinthiflora ‘Luo Lan Zi’ was ascertained mainly based on clustered microshoot inducement and large leaf area establishment in 6-benzyladenine (BAP) (1.00 mg L−1) and zeatin (Z) (0.10 mg L−1) combination. Medium supplied with lower level of BAP (0.50 mg L−1) and auxin (IAA) (0.25 mg L−1) was not suitable for lilac shoot proliferation, but it could be competent for long-term preservation of the un-rooted shoots so that subsequent proliferation culture could be carried out at anytime. In addition, excess height growth which resulted in low transplanting survival rate was effectively controlled by decrease in node number when paclobutrazol (PBZ) was applied in rooting medium at a concentration of 1.00 mg L−1 after taking into account the effects on shoot height, rooting, persistent leaf area and PBZ carry-over. An important overwintering treatment was to use a plastic chamber covering for plants in the greenhouse prior to field planting to ensure adequate biomass of stem and underground parts not only in the current growing season but also in the subsequent years.  相似文献

‘Picual’ olive cuttings were grown in a greenhouse under saline conditions in 2 L plastic pots containing perlite. Plants were irrigated with a nutrient solution plus 75 mM NaCl and 0, 2.5, 10 or 40 mM CaCl2. Vegetative growth, leaf and root Na+ and Ca2+ concentrations were measured. Na+ toxicity symptoms were observed in plants non-treated with Ca2+. Shoot length was higher in Ca2+ treated plants, although shoot growth was reduced at 40 mM CaCl2, probably due to the high total ion concentration reached in the external solution. Ca2+ supply linearly increased leaf and root Ca2+ concentration and decreased leaf Na+ concentration. However, there were no differences in root Na+ concentration. Results indicate Ca2+ may take part in the Na+ exclusion mechanism, mainly preventing Na+ transport to the shoot, that may be an important ability for survival under saline conditions.  相似文献

Changes caused by NaCl-induced salinity on several growth parameters and ions accumulation have been measured in five olive (Olea europaea L.) cultivars (‘Chemlali’, ‘Chetoui’, ‘Koroneiki’, ‘Arbequina I18’, and ‘Arbosana I43’) growing in a greenhouse in nutrient solution pot experiment. One-year-old plants were transplanted to sand–perlite (1:1) culture, and were irrigated with half-strength Hoagland nutrient solution containing NaCl at various levels (0.5, 50, 100 and 200 mM). Salinity induced significant decrease in growth parameters, but to a different extent in each cultivar. Leaf growth and total leaf area per plant were significantly affected by all salinity treatments in all studied cultivars, being ‘Arbequina I18’ the most sensitive cultivar. Leaf drop phenomenon was observed from 60 days after salt application at high salinity treatments, mainly in Arbequina I18. Contrary to leaf area, leaf thickness increased progressively during the experiment. ‘Chemlali’ developed thicker leaves at the two highest salinity treatments when compared to the other cultivars. Na+ and Cl concentrations were higher in roots than in shoots and leaves in most of the cultivars investigated. The effectiveness of Na+ exclusion mechanism in the roots differed significantly among studied cultivars, working effectively in ‘Chemlali’ (by inhibiting translocation of Na+ to the aerial part) and being much less efficient in ‘Arbequina I18’. Furthermore, leaf abscission can be considered as an additional tolerance mechanism of olive cultivars allowing the elimination of leaves that had accumulated Na+ and Cl ions. Tolerance to salinity stress was as follows: ‘Chemlali’ > ‘Chetoui’ > ‘Arbosana I43’ > ‘Koroneiki’ > ‘Arbequina I18’. This order of salt tolerance was indicated by lower reduction in plant growth parameters (shoot elongation, trunk diameter, total plant dry weight, internodes length, and total leaf area), the increase of leaf thickness, and by the effectiveness of the exclusion mechanism of Na+ and Cl in the root system.  相似文献

An in vitro plant regeneration protocol for pansy (Viola wittrockiana) cultivar ‘Caidie’ from petioles was established as following: callus induction on a half-strength MS medium supplemented with 0.45 μmol l−1 2,4-d plus 8.9 μmol l−1 BA, callus subculture on medium F (1/2MS with 4.5 μmol l−1 2,4-d, 2.7 μmol l−1 NAA and 0.44 μmol l−1 BA) and then on medium T (1/2MS with 4.5 μmol l−1 2,4-d, 2.7 μmol l−1 NAA and 2.2 μmol l−1 BA), shoot regeneration on medium D3 (MS media supplemented with 2.9 μmol l−1GA3, 23.6 μmol l−1 AgNO3, 0.02% active charcoal and 4.5 μmol l−1 TDZ), shoot multiplication on medium M (half-strength MS medium containing NAA 1.1 μmol l−1, TDZ 9.1 μmol l−1 and GA3 8.7 μmol l−1), and then shoot elongation and rooting on medium R (MS medium supplemented with 1.1 μmol l−1 NAA and 1.1 μmol l−1 BA). Salt tolerance of five cultivars of Capsicum annuum L. Early Jalapeno, Golden Treasure, NuMex Sweet, NuMex Joe E. Parker, and Santa Fe Grande, two cultivars of C. chinense Jacq. Habanero and Pimienta De Chiera, and one accession of C. annuum, NMCA 10652, were evaluated in a field study. Seedlings were transplanted in late May to field raised beds containing loamy sand soils in a semi-arid environment. Plants were well irrigated throughout the experiment. Three saline solution treatments, prepared by adding NaCl, MgSO4, and CaCl2 to tap water at different amounts to create three salinity levels of 0.82 dS m−1 (control, tap water), 2.5 dS m−1, and 4.1 dS m−1 electrical conductivity (EC), were initiated on 15th June and ended in late August. Among the eight varieties, NMCA 10652 had the highest survival percentage at 100% in the 4.1 dS m−1 treatment, followed by ‘Early Jalapeno’, ‘NuMex Sweet’, ‘Pimienta De Chiera’, ‘Santa Fe Grande’, ‘Golden Treasure’, and ‘NuMex Joe E. Parker’. ‘Habanero’ had the lowest survival at 28%. Compared to control, final shoot dry weight of the plants irrigated with saline solution at 4.1 dS m−1 was reduced by 92% in ‘Habanero’, followed by ‘Golden Treasure’ at 80%. For fruit fresh weight in 4.1 dS m−1 vs. control, ‘Habanero’ had the highest reduction at 86%, followed by ‘Golden Treasure’ at 74%, while NMCA 10652 and ‘Santa Fe Grande’ had the least at 26% and 19%, respectively. NMCA 10652, the most tolerant to salinity, had the lowest leaf Na+ accumulation, while ‘Habanero’, the most sensitive to salinity, had the highest Na+ in the leaves. For leaf Cl, ‘Early Jalapeno’ had the highest, while ‘Habanero’ had the lowest Cl accumulation in the leaves. Generally, sensitive varieties accumulated more Na+ and/or Cl in leaves, except for ‘Early Jalapeno’, which was relatively tolerant to salinity but had high Na+ and Cl accumulation in leaves.  相似文献

Plant production under salinity requires increased capacity for K+ homeostasis. For this purpose, supplementary K2SO4 in the nutrient solution and grafting on a tolerant rootstock were employed in two experiments to test whether grafting, potassium and their interactions can alleviate salinity stress in tomato (Solanum lycopersicum L.). In Exp-ion, plants were cultivated for 122 days to compare different ionic compositions: EC 9 dS m−1 in ECall (by macro-nutrients) and in ECNaCl (by 64.2 mM NaCl), EC 12 dS m−1 in ECK (ECNaCl + 25.8 mM K+). Exp-K+ was established to compare K+ concentrations of 6, 16 and 36 mM at 150 mM NaCl. In both Experiments, ‘ZS-5’, selected as a salt sensitive cultivar, was either self-grafted or grafted onto the cultivar ‘Edkawi’, reported as salt tolerant. Yield and growth, minerals, gas exchange, soluble sugars, and proline were analyzed. Different ionic treatments affected almost all characteristics considered while differences between rootstocks were rarely observed. No pronounced differences were found in shoot growth, yield and gas exchange between ECall and ECNaCl. ECK did not show any salinity alleviative effects but inhibited even growth compared with the other treatments. In Exp-K+, 16 mM K+ increased plant growth, leaf soluble sugars and proline concentrations. 36 mM K+ did not further reduce upper leaf Na+ although leaf K+ concentration increased significantly. The results indicated that the response of tomato plant to NaCl stress was principally attributed to the osmotic component in Exp-ion, excessive K+ showed no mitigating effect on fruit yield and shoot growth. However, 16 mM K+ in the root environment enhanced the salt adaptive capacity of plants stressed at 150 mM NaCl. The use of the tolerant rootstock resulted in no ameliorative effects, owing to its susceptibility to blossom-end rot, failure in enhancing photosynthesis, and ineffectiveness of restraining the long-distance transport of Na+.  相似文献

There is little available information on the effects of temperature and CO2 enrichment on stomata anatomical characteristics of plants. Effect of these two microclimates was studied on five rose (Rosa spp.) cultivars, viz. ‘First Red’ (used as check), ‘Arjun’, ‘Raktima’, ‘Raktagandha’ and ‘Pusa Pitamber’. Budded, single-stemmed rose cultivars having five lateral buds were grown in controlled environment growth cabinets under enriched CO2 (1000 μmol mol−1) and optimum (28/18 °C, T0) or high (35/25 °C, T1) temperature for 50 days. All observations were made on the abaxial leaf surface. Significant increases in stomatal density (68.7%), index (29.6%) and epidermal cell density (37.3%) were recorded in plants grown at high temperature over control with CO2 enrichment. The cultivars responded differently in terms of length and width of guard cell and stoma (pore) under high temperature, however, the values averaged over treatments showed a significant reduction in these parameters. Further, number of stomata per leaf was higher (28.3%) in plants grown at high temperature, except First Red. A reduction in mean leaf area (26.7%) and dry mass (32.0%) was recorded at high rather than optimum temperature. The specific leaf area was maximum in Arjun (87%) while in First Red, a 14% reduction was noted at high temperature.  相似文献

In order to study the effect of brassinosteroids on the amelioration of the inhibitory effect of salinity on strawberry plants, a short-term experiment was conducted in greenhouse to test different concentrations of 24-epibrassinolide (24-EBL) (0.0, 0.5, and 1 μM) by foliar application on some agro-physiological properties, such as shoot dry weight, and root dry weight, stomatal conductance (SC), leaf relative water content (LRWC), leaf chlorophyll reading values (LCRV) and membrane permeability (MP) of strawberry ‘Fern’ and ‘A6’ cultivars irrigated with salt water (35 mM NaCl). 24-EBL solutions were applied twice during late afternoon hours with 7 d intervals using a hand-held sprayer. Plant shoot dry weight, root dry weight, SC, LRWC and LCRV were reduced by 29–33%, 45–15%, 71–55%, 11–13%, and 12–13% for ‘A6’ and ‘Fern’ cultivars at 35 mM (without 24-EBL applied), respectively, as compared to the nonsaline treatment, but MP increased 40% and 12%. An exogenous supply of 24-EBL was found to be successful in alleviating of the inhibitory effects of salt stress on plant growth parameters and nutrient contents. 24-EBL (1 μM) application under saline condition significantly increased shoot and root dry matter, SC, LRWC and LCRV of plants, and alleviation effects of 1 μM 24-EBL application was 20%, 15%, 122%, 5.8%, and 10.9% for ‘A6’ and 47%, 8.0%, 83%, 33.3% and 6.0% for ‘Fern’ cultivars, respectively. Macro-micro element content of plant leaf and root increased with increase 24-EBL except for Na under salinity stress. These results support the view that supplementary 24-EBL application can overcome the effects of salinity stress on plant growth and growth parameter under saline conditions.  相似文献

Malformation of emerging leaves with distortion of leaf tips, a condition known as tipburn, is frequently observed in strawberry. Calcium (Ca) deficiency has been considered the main cause of tipburn. The aim of the present study was to analyse the relationship between leaf mineral composition and the incidence of tipburn in three short-day strawberry (Fragaria x ananassa Duch.) cultivars (‘Ventana’, ‘Camarosa’ and ‘Candonga’) submitted to different concentrations of Ca. The studies were conducted in a hydroponic system in a greenhouse. Calcium was added as Ca(NO3)2 at 2 mM, 3 mM, 4 mM and 5 mM. A completely randomized block design (4 Ca concentrations × 3 cultivars) with three replications was used. Each replicate consisted of 12 plants grown in a polyethylene bag (100 cm × 18 cm × 3 cm) filled with coconut peat. Crown diameter and tipburn incidence were evaluated throughout the experimental period, and at the end of the experiment leaf mineral composition was assessed. In general, plants with larger crown diameters had a greater incidence of tipburn. The ‘Candonga’ cultivar had the smallest incidence of tipburn, while the ‘Camarosa’ and ‘Ventana’ cultivars were more susceptible. There was no correlation between level of Ca applied and incidence of tipburn. The incidence of tipburn was associated with foliar K:Ca and K:Mg ratios. Ratios above 3.40 for K:Mg and 1.77 for K:Ca represented a risk of more than 50% of tipburn incidence, when overall means for all cultivars and levels of Ca were used.  相似文献

A protocol for clonal propagation of Rosa clinophylla, a rare and endangered species but very important for breeding purposes had been standardized through in vitro axillary bud culture. Although cytokinins alone were able to induce shoot buds, but their proper growth and number could be increased only when they were used in combination with GA3. However, there was shoot tip necrosis and leaf fall in the proliferated shoots. AgNO3 at 58.85 μM proved effective to avoid shoot necrosis and yellowing of leaves. Activated charcoal (AC) at 250 mg l−1 was found necessary at all the stages of shoot multiplication as well as rooting. Ninety percent rooting could be achieved in 1/2 MS medium supplemented with 4.92 μM IBA and 250 mg l−1 AC. Rooted plantlets were hardened and transferred to the field successfully with 80% survival rate.  相似文献

‘Swingle’ citrumelo [Citrus paradisi MacFaden × Poncirus trifoliata (L.) Raf.] has been extensively used as a rootstock in several citrus growing regions of the World, including Southern Brazil where ‘Rangpur’ lime (Citrus limonia Osbeck) is still the predominant variety despite being affected by several important pathogens. In this case, ‘Swingle’ citrumelo is used to produce nursery trees to establish new orchards or to be inarched in adult and healthy groves in order to change the rootstock. We report herein a system to produce trees on ‘Swingle’ citrumelo more rapidly by budding onto non-rooted cuttings, as well as assessing potential to rapidly multiply ‘Swingle’ through rooting of non-budded cuttings. Therefore, two potential products are described: budded trees and rooted rootstock cuttings. ‘Valencia’ sweet orange [Citrus sinensis (L.) Osbeck] was budded at different heights on cuttings derived from eight-month old rootstocks. Grafted and additional non-budded cuttings were then treated with indole-3-butyric acid (500 mg L−1) or left untreated before rooting. Three types of cuttings were evaluated: softwood, semi-hardwood and hardwood. The use of nursery trees derived from pre-budded hardwood cuttings of ‘Swingle’ citrumelo is an alternative grafting method on this cultivar. Softwood cuttings with one leaf pair were considered the most adequate material for rapid multiplication of ‘Swingle’ citrumelo by cutting. This could be particularly useful for inarching production or conventional budding after transplant of cutting-derived rootstocks.  相似文献

Olive (Olea europaea L.) is the major fruit tree in the Mediterranean region, often grown in locations where plants are exposed to increased salinity. To determine the effect of NaCl on shoot and root growth, dry matter allocation, leaf Na+ and K+ concentration, electrolyte (EL) and K+ leakage (KL), seven olive cultivars of different origins were grown in nutrient solution containing 0, 33, 66, 100 or 166 mM NaCl for three months. The general effect of salinity was linear and quadratic decrease of observed plant growth parameters. Different responses of tested cultivars to applied levels of salinity were found for stem dry weight, shoot length and number of leaves. As salinity increased, growth of ‘Manzanillo’ declined sharply, whereas ‘Frantoio’ was the most tolerant to growth reduction in most of the observed growth parameters. Allometric analysis showed that biomass allocation under salinity stress was similar in all cultivars, but the slope between shoot weight and total plant weight decreased as salinity increased. Since the higher allocation in roots was not found, it seems that salinity only slowed the above ground plant canopy growth. Sodium concentration in leaves of all cultivars increased as salinity increased with the highest increment reached when the salinity of nutrient solution was raised from 100 to 166 mM NaCl. Significant differences among genotypes were found in leaf Na+ and K+ concentration and K+:Na+ ratio, but they were not related to the growth rate. Generally, ‘Frantoio’ and ‘Oblica’ accumulated less Na+ and were able to maintain higher K+:Na+ ratios as compared to other genotypes. Electrolyte leakage and KL linearly increased with increasing salinity and the magnitude of the response depended upon the olive cultivar.  相似文献

This work examined the effect of nitrogen source and medium buffering on the micropropagation of Eucalyptus marginata Donn ex Sm. The number of shoots was increased when media contained 2-(N-morpholino) ethanesulfonic acid (MES) but this increase was minor and only applied to one of the two clones tested. Highest root production was obtained when the medium contained 7.5 mM nitrogen in a ratio of 2NO3:1NH4+ and was buffered with 10 mM MES. In the rooting medium the pH was influenced most significantly by the nitrogen source, and then whether the medium was buffered. The media pH remained relatively constant when nitrate was the sole nitrogen source and this was assisted by the addition of 10 mM MES. Lower concentrations (<10 mM) of MES were less effective in buffering media over a four-week culture period in both shoot multiplication and rooting medium.  相似文献

The regenerability of three ornamental species—Lysimachia christinae, Lysimachia rubinervis and Lysimachia nummularia ‘Aurea’, were investigated using in vitro leaves and shoot tips. 6-Benzylaminopurine (BAP) and α-naphthalene acetic acid (NAA) added to Murashige and Skoog (MS) medium were tested for their effect on organogenesis. On the medium, shoot regeneration occurred directly without callus formation. In these species, L. christinae developed the highest regeneration rate and numbers of shoots/explant from shoot tips (100%, 12.25) and leaf bases (100%, 13.01) on the MS medium containing 3.0 mg l−1 BAP and 0.1 mg l−1 NAA. For L. rubinervis, the highest shoot induction rate and number of shoots/explant were obtained from shoot tip (100%, 16.87–17.20) on the MS medium with 0.1 mg l−1 NAA and 3.0–5.0 mg l−1 BAP. L. nummularia ‘Aurea’, however, showed the highest regeneration rate and number of shoots/explant (100%, 12.73) from leaf bases on MS medium supplemented with 1.0 mg l−1 BAP and 0.1 mg l−1 NAA. All in vitro shoots rooted well on half macronutrient MS medium containing 0.1 mg l−1 NAA. After acclimatization, transplanted plantlets grew normally and flowered in the field.  相似文献

In vitro bud and shoot organogenesis was investigated for the ornamental plants Eucalyptus erythronema var. erythronema, E. stricklandii and their interspecific hybrids cv. ‘Urrbrae Gem’ and ‘Hybrid 2.5’ by using 0.0, 0.1, 0.25, 0.5 or 1.0 μM BAP on apex and leaf explants. Callus developed on all explants and increased with all concentrations of BAP without significant differences between BAP concentrations. Buds formed on apex and leaf explants of E. erythronema and E. cv. ‘Urrbrae Gem’ especially with 1.0 μM BAP, but these buds rarely developed into shoots. Bud clusters formed on E. erythronema and E. cv. ‘Urrbrae Gem’ apex and leaf explants whereas E. stricklandii and ‘Hybrid 2.5’ produced fewer, individual buds on the explant. Shoots regenerated from apex explants of all genotypes with all levels of BAP, whereas few shoots of any genotype regenerated from leaf explants regardless of the number of buds formed. Shoots from apex explants could be multiplied successfully. Light microscopy showed meristems developed within the callus, and at the callus and bud surfaces. However, few shoots developed considering the level of bud and meristem formation. This report is the first for successful shoot organogenesis and multiplication in an ornamental eucalypt.  相似文献

Twenty-five genotypes of cowpea (Vigna unguiculata L. Walp.) were tested for salt-tolerance at the vegetative growth stage in pot in the greenhouse experiments at salinity levels of 0, 85, and 170 mM NaCl. Plant survival was the main criterion for classifying genotypes. Other criteria included the ion concentration (Na+ and Cl) in root and shoot and biomass accumulation. Four local accessions (‘Paceño’, ‘Tardón’, ‘Sonorense’, and ‘Cuarenteño’), three accessions from California (‘CB46’, ‘CB27’, and ‘CB3’), and one accession from the International Institute of Tropical Agriculture (IITA) (‘IT82D-889’) survived at concentrations of both 85 and 170 mM NaCl and were classified as salt-tolerant, while ‘IT96D-666’, ‘IT89KD-288’, and ‘IT93K-734’ from IITA were classified as salt-sensitive. One local accession (‘Sesenteño’), three accessions from IITA (‘PEPH-V Wes-85’, ‘IT86D-719’, and ‘IT95K-1090-12’), and one accession from California (‘CB5’) were classified as moderately salt-tolerant. Eight accessions from IITA (‘IT96D-733’, ‘IT90K-277-2’, ‘IT91K-93-10’, ‘IT91K-118-20’, ‘IT90K-284-2’, ‘IT95K-1088-4’, ‘IT89KD-391’, and ‘IT94K-437-1’) and one from California (‘CB88’) were classified as moderately salt-sensitive. Biomass was affected by both 85 and 170 mM NaCl in all groups of genotypes, however, salt-tolerant and moderately salt-tolerant genotypes showed higher biomass than genotypes classified as moderately salt-sensitive and salt-sensitive. In all genotypes Cl concentration was higher in shoots than roots and increased as salinity increased. Similarly Na+ concentration increased with increasing salinity. However, in salt-tolerant and moderately salt-tolerant genotypes, Na+ concentration was more in roots than shoots, while in moderately salt-sensitive and salt-sensitive genotypes, Na+ was higher in shoots than roots.  相似文献

Many cool season garden crops, including Viola × wittrockiana Gams. (pansy), exhibit reduced flowering outdoors during the warm summer months. Twelve pansy cultivars varying in summer garden performance were grown under either 20 ± 1.5 or 30 ± 1 °C (air temperature) to determine growth and flowering responses to prolonged high-temperature exposure and to identify selection criteria to screen pansies for flowering heat tolerance. Increasing temperature from 20 to 30 °C increased leaf number below the first flower on ‘Crystal Bowl Primrose’ and ‘Skyline White’ only. Flower bud number reduction at 30 °C versus 20 °C varied from 20% for ‘Crystal Bowl Purple’ to 77% for ‘Majestic Giants Red and Yellow’. Flower diameter reduction at 30 °C versus 20 °C ranged from 14% for ‘Skyline Beaconsfield’ to 44% for ‘Super Majestic Giants Ocean’. The percentage reduction in total color (flower number × estimated flower area) ranged from 60% for ‘Crystal Bowl Primrose’ to 88% for ‘Majestic Giants Rose Shades’. Based on a weighted base selection index, ‘Super Majestic Giants Canary’ and ‘Delta Yellow’ were identified as the most heat-tolerant cultivars, while ‘Super Majestic Giants Ocean’ and ‘Majestic Giants Rose Shades’ were identified as the most heat-sensitive. In a second experiment, root and shoot dry mass were determined after 10, 20, or 30 d when grown at 20 or 30 °C. Relative growth rate and root:shoot ratio were also calculated. After 30 d, ‘Crystal Bowl Primrose’, ‘Crystal Bowl Sky Blue’ and ‘Skyline White’ relative growth rates were lower at 30 °C versus 20 °C. Root:shoot ratio on day 30 was lower at 30 °C compared to 20 °C for six cultivars, but similar across temperature for five cultivars and higher for ‘Crystal Bowl Primrose’. Flower bud number at first flower was positively correlated with branch number, shoot dry mass at flowering, but not correlated with root dry mass at flowering, and negatively correlated with flower diameter and root:shoot ratio (either at flowering, or after 10, 20 or 30 d at 30 °C), suggesting that these traits may be useful when screening pansies for flowering heat tolerance.  相似文献

Continuous and rapidly proliferating axillary shoots were raised from axillary buds in secondary branches of adult field culms and nursery grown 1-year-old tissue culture-raised plants of Bambusa vulgaris ‘Striata’. Shoots continuously proliferated in a MS medium containing 4 mg L−1 6-benzyladenine (BA). The effects of indole butyric acid (IBA) levels, a pretreatment with thidiazuron (TDZ) (1-phenyl-1-([1,2,3-thidiazol-5-yl])urea) and illumination on rooting, were investigated after 6 months of shoot proliferation. A rooting medium with IBA at 3 mg L−1 was optimum for root induction. Shoots of adult field culms that were proliferated in the presence of BA when induced to root in this medium resulted in 40% rooting in 27 days. In vitro shoots raised from 1-year-old tissue cultured plants showed 92% rooting under the same conditions. Rooting was enhanced when the relatively difficult-to-root in vitro shoots from adult field culms were pretreated with 0.5 mg L−1 TDZ for two to three subcultures before placing in the root induction medium. Continuously illuminated shoots pretreated with TDZ for three subcultures showed 100% rooting compared to 83% rooting of shoots that were exposed to a 12 h photoperiod. These findings have been applied in the large-scale propagation of this species.  相似文献

Molecular characterization using RAPD analysis was carried out in eight cut flowers and two pot plant cultivars of chrysanthemum. Three of them (‘Refocus’, ‘Red Reagan’, and ‘Sheena Select’) were established in vitro and the occurrence of somaclonal variation was studied using the same molecular technique. Two induction media (MS + 0.1 mg l−1 NAA + 0.1 mg l−1 BA, and MS + 2.0 mg l−1 IAA + 0.5 mg l−1 Kinetin), and two proliferation media (MS + 0.1 mg l−1 NAA + 0.2 mg l−1 BA, and MS + 4.0 mg l−1 IAA + 2.0 mg l−1 Kinetin) were employed in order to evaluate the effect of the medium composition in the shoots’ stability. Likewise, the effect of the culture age was considered in assessing genetic stability. Monthly subcultures were carried out, identifying the origin and history of the shoots, throughout a nine-month proliferation period followed by acclimatization. Molecular markers were obtained in every subculture cycle and from the acclimatized plants. Only one shoot from the 7th subculture of the cultivar ‘Refocus’ showed a different band pattern. The use of RAPD for chrysanthemum cultivar characterization and somaclonal variation detection is discussed.  相似文献
