The European pear (Pyrus communis) carries the S-RNase-mediated gametophytic self-incompatibility (GSI) system. The S-haplotype is conferred by an S-locus, which contains the style-specific expressed S-RNase, and the pollen-specific expressed F-box genes (SFB). Both the S-RNase and the SFB genes are multi-allelic and each is characteristic of one of the S-haplotypes. Therefore, they are ideal markers for molecular S-genotyping. In this work, for the first time, seven haplotype-specific SFBs were isolated from European pears. Particular primers for each of these SFBs were generated, thus providing an additional tool for S-genotyping of European pear cultivars.  相似文献

In this work we report the cloning and identification of S-RNase alleles responsible for gametophytic self-incompatibility (GSI) of ‘Rocha’ pear and of 13 other European pear cultivars that might be used as its pollinators. Partial sequences of S-RNase alleles were amplified by PCR with specific primers hybridising in conserved regions of previously identified S-RNase alleles of Pyrus communis, cloned and sequenced and the S-genotype of eight pear cultivars was fully determined. Three cultivars (‘General Léclerc’ (SqSl), ‘Tosca’ (SbSl) and ‘Alexandrine Douillard’ (SbSk)) shared no S-alleles with ‘Rocha’ (SaSe) and shall be totally compatible with this cultivar. None of the cultivars analysed showed an identical amplification pattern to the one observed in ‘Rocha’, so the other cultivars shall be at least semi-compatible. One new allele was identified in P. communis cv. ‘Beurré d’Avril’ (designated as St). The determination of both S-RNase alleles of cvs ‘Rocha’, ‘Beurré Precoce Morettini’ (SeSk) and ‘Tosca’ and the identification of one S-RNase allele in cvs ‘Carapinheira’ (Sb), ‘Amêndoa’ (Se), ‘Pérola’ (Sk) and ‘Beurré d’Avril’ (St) are important contributions for the effort recently developed worldwide to establish groups of sexual compatibility among European pears.  相似文献

The vegetative and reproductive performances of ‘Coscia’ pear (Pyrus communis L.) grown on nine rootstocks [OHF 69, OHF 97, OHF 217, OHF 333, OHF 513 and BP 1 (P. communis), clonal seedling (Davis AxB) of Pyrus betulifolia and quince BA 29 and EMA (Cydonia oblonga)] were compared during a 9-year period. The trial was conducted at the Experimental Station ‘Avnei Eitan’ in northern Israel (elevation 400 m above sea level, a.s.l.), on a well-drained soil with pH 7.1. Trees were planted in December 1998, spaced at 4.0 m × 2.0 m and trained with a central axis. The most vigorous trees were on P. betulifolia seedlings, followed by the four OHF rootstock (69, 97, 333, 513) and BP 1 (with no significant difference between them). All the above rootstocks demonstrated greater vigor than OHF 217 and quince BA 29 or EMA. The highest cumulative yields per tree were harvested from trees on the four OHF rootstocks (69, 97, 333, 513) and P. betulifolia followed by the BP 1. The two quince rootstocks, as well as OHF 217, had the lowest cumulative yield and the lowest yield of large fruit. A positive correlation was found between the vigor of the tree, as affected by the rootstock, and both the total yield and the fruit size. We conclude that in a warm climate, yield efficiency is not the only parameter that should be taken into account, and building a strong tree for a weak cultivar is the first requirement for establishing an orchard.  相似文献

Plant height, a vigor trait, in 1-year-old seedlings made from Japanese pear (Pyrus pyrifolia) cultivars/selections was measured using 994 individuals from 29 families. The family mean of plant height was negatively correlated (r = −0.72**) to the inbreeding coefficients (F). The regression of the family mean (Fm) on the F value (Fm = 130 − 104F) showed that inbreeding depressions were 8%, 20%, and 40% for F = 0.1, 0.25, and 0.5, respectively. According to the regression, the family mean at F = 0 was estimated at 130 cm. These results showed that the vigor was greatly influenced by inbreeding in Japanese pear. Within-family variances, the genetic segregation of offspring in a family, differed according to family. The proportions of offspring with plant height above 130 cm (estimated Fm for F = 0) were extremely low, i.e., 0–17% for 0.5 ≤ F < 0.60 and 0–8% for F = 0.75.  相似文献

Few records are available about local Tunisian pear cultivars characterized by low chilling requirements and adaptation to dry conditions. In this work, seven SSRs derived from apple were successfully transferred to 25 local Tunisian pear genotypes and 6 common varieties of Pyrus communis cultivated in Europe. The 7 SSRs used amplified a total of 36 fragments. All the microsatellites except one seem to amplify more than one locus in some of the genotypes studied. Only 12 different fingerprinting patterns could be distinguished among the 25 Tunisian cultivars studied indicating a high number of synonymies. The mean expected and observed heterozygosities in the 25 Tunisian cultivars analyzed averaged 0.71 indicating a high level of genetic diversity among the local Tunisian pear germplasm. These markers will be useful to optimize the conservation of this highly threatened germplasm.  相似文献

Seven microsatellite loci (SSR) developed in apple were used for the identification of 63 European pear cultivars. A total of 46 fragments were amplified, with an average of 6.6 alleles per SSR. Only one microsatellite amplified more than one locus. The mean expected and observed heterozygosities over the six single-locus SSRs averaged 0.68 and 0.44, respectively, and the number of effective alleles per loci was 3.43. The amplified fragments produced 61 different fingerprinting patterns that allowed to unequivocally distinguish all the varieties analyzed. Only two varieties, which derive from mutations, could not be distinguished from the original variety. Cluster analysis of the estimated genetic similarity, grouped the varieties according to their pedigree and their geographic origin. The variability detected with the SSRs in European pear varieties was low when compared with the variability detected in other fruit crops in the Rosaceae.  相似文献

Fruit thinning in pear is feasible for mitigation of water stress effects. However, it is not well known how fruit quality at harvest and after cold storage is affected by pre-harvest water stress. Even less is known about the effects of fruit thinning on quality under these circumstances. To elucidate these, we applied deficit irrigation (DI) and fruit thinning treatments to ‘Conference’ pear over the growing seasons of 2008 and 2009. At the onset of Stage II (80 and 67 days before harvest in 2008 and 2009, respectively), two irrigation treatments were applied: full irrigation (FI) and DI. FI trees received 100% of crop evapotranspiration (ETc). DI trees received no irrigation during the first three weeks of Stage II to induce water stress, but then received 20% of ETc to ensure tree survival. From bud-break until the onset of Stage II and during post-harvest, FI and DI trees received 100% of ETc. Each irrigation treatment received two thinning levels: no thinning leaving commercial crop load (∼180 fruits tree−1), and hand-thinning at the onset of Stage II leaving a light crop load (∼85 fruits tree−1). Under commercial crop loads, DI trees were moderately water-stressed and this had some positive effects on fruit quality. DI increased fruit firmness (FF), soluble solids concentrations (SSC) and acidity at harvest while no changes were observed in fruit maturity (based on ethylene production). Differences in FF and acidity at harvest between FI and DI fruit were maintained during cold storage. DI also reduced fruit weight loss during storage. But fruit size was reduced under DI. Fruit thinning under DI resulted in better fruit composition with no detrimental effect on fresh-market yield compared to un-thinned fruit. Fruit size at harvest and SSC values after five months of cold storage were higher in fruit from thinned trees than fruit from un-thinned trees. Fruit thinning increased fruit ethylene production, indicating advanced maturity. This may lead to earlier harvest which is desirable in years with impending drought. Fruit thinning is therefore a useful technique to enhance pear marketability under water shortage.  相似文献

Argyranthemum frutescens is a commercially important ornamental species with extensive breeding programmes in several countries. Since it is vegetatively propagated there is a great need for tools for identification of cultivars. Vegetatively propagated species require clean-up from diseases, often performed through meristem-tip cultures. Forty-three cultivars of A. frutescens propagated by meristem-tip culture and traditional cuttings were analyzed for genetic relatedness and possible somaclonal variation using AFLP. Five primer combinations resulted in a relatively high degree of polymorphism. Ten molecular markers generated by one primer combination could distinguish between all 43 cultivars. Differences in fingerprints between meristem-tip culture and cuttings from the same cultivars were found, but the proportion of fragments being specific for either tissue culture or cuttings was relatively low. Some cultivars that did not display somaclonal variation as judged by the AFLP-fingerprints may still be genetically unstable since some morphological variation was observed in the true to type test.  相似文献

Taxonomic revision of Dendrobium moniliforme complex is presented. D. moniliforme complex is characterized by the even slim stems, bracts with brownish zone, semi-spherical anther cap and the hairy disc of lip. Dendrobium tosaense, Dendrobium officinale and Dendrobium guangxiense were excluded by having membranous bracts lacking brownish zone, anther cap conical and bifid. Two species are recognized in this complex, i.e., D. moniliforme and Dendrobium wilsonii. D. wilsonii differs from D. moniliforme by having elliptic leaves about 1.3–2 cm wide, dorsal sepal 3.0–4.0 cm long, 0.6–0.9 cm wide, petals elliptic to oblong, 3.0–4.0 cm long, 1.0–1.5 cm wide, lip elliptic to ovate–lanceolate, 2.6–3 cm long, 1.2–1.5 cm wide.  相似文献

The essential oils of aerial parts of Achillea gypsicola Hub.-Mor., Hypericum scabrum L., Satureja hortensis L., and Origanum acutidens (Hand.-Mazz.) Letswaart were analyzed in this study by GC and GC–MS and their oils were tested for toxicity against broadbean weevil (Bruchus dentipes). A. gypsicola oil contained camphor (40.17%), 1,8-cineole (22.01%), piperitone (11.29%), borneol (9.50%) and α-terpineol (1.56%) as major components. A total of 74 components were identified by GC–MS in H. scabrum oil, including α-pinene (9.26%), terpinen-4-ol (5.12%), camphor (5.94%), δ-cadinene (4.52%), pulegone (4.45%), γ-muurolene (4.12%), pinocarvone (3.97%) and β-caryophyllene (3.42%) as predominant components. The essential oils of O. acutidens and S. hortensis were characterized by high contents of carvacrol (86.99% and 55.74%), γ-terpinene (0.71% and 20.94%), p-cymene (1.95% and 12.30%), α-terpinene (0.13% and 2.04%) and β-caryophyllene (1.30% and 1.08%). All of the essential oils were toxic to adults of B. dentipes and insect mortality increased with increasing concentration of each oil. The oils (20 μl dose) brought about 100% mortality in 36 h. Although desirable insecticidal activities against the pest were achieved with the oils from all four plant species, S. hortensis and O. acutidens oils were more effective, particularly after 6 h of treatment. The current results concluded that the essential oils, in particular O. acutidens and S. hortensis oils, may be used as potential botanical insecticides against B. dentipes.  相似文献

The response of ‘Conference’ pear subjected to branch bending in the content levels of various metabolites in its fruit was investigated. The fruits in commercial maturity were sampled in 2004 and 2005 from branches bent in the late summer of 2003 (the summer treatment), from branches bent in the late spring of 2004 (the spring treatment) and from control branches. The content levels of carbohydrates, organic acids and phenolic compounds were compared among treatments in two successive years. The fruit revealed various responses in content levels of metabolites. In the first year after bending, no significant differences were found in the content of each carbohydrate, but in 2005 by far the highest content level of glucose and fructose and the lowest content level of sucrose were found in fruits from the control. The control fruit showed significantly higher content levels of malic acid and lower content levels of some phenolics (chlorogenic and vanillic acid) in the first year after bending, but in the next year the opposite reaction occurred—the control fruit had the lowest content level of malic acid and the highest content level of epicatechin, quercetin-3-d-galactoside and quercetin-3-β-d-glucoside. The comparison of the two bending treatments alone in 2004 showed that the summer treatment often produced a slightly higher value of each phenolic in comparison to the spring treatment. However, in 2005 the significantly highest content of chlorogenic acid was in fruit from the spring treatment. Sorbitol, as well as citric acid, catechin and sinapic acid showed no clear tendency among treatments, neither in 2004 nor in 2005. It is suggested that these variations of ‘Conference’ fruit subjected to different bending treatments could not be the result of bending alone, but that they could be indirectly affected by other physiological responses of the fruit tree. However, it seemed that variations are affected by the time of bending and by the year-to-year, and such responses can be attributed to the ‘Conference’ genotype only.  相似文献

The S-allele characterisation of ‘Alzina’ and ‘Garondès’, two local almond cultivars from the island of Majorca, by a multidimensional approach has allowed the confirmation of the presence of the Sf-allele and the identification of a new allele not previously described in almond, S36. When these cultivars were phenotypically evaluated, both showed a self-incompatible phenotype and were cross-incompatible, as assessed by artificial self- and cross-pollinations and fruit sets after field pollinations, confirming that their Sf-allele is in its active form, Sfa. Thus a new CGI group in almond is proposed and named XXVIII. These results confirm the wide diversity of S-alleles in almond both at genotypic and phenotypic levels, as well as their similarity with the S-alleles from other close Prunus species. This similarity suggests the possibility of allele introgression between species or allele identity by descent from a common ancestor.  相似文献

Agrobacterium tumefaciens-mediated transformation has been one of the methods used to generate transgenic plants in bell pepper. An alternate transformation method that avoids/minimizes tissue culture would be beneficial for the improvement of bell pepper due to its recalcitrant nature. In this report, transgenic bell pepper plants have been developed by a tissue-culture-independent A. tumefaciens-mediated in planta transformation procedure. In the present study, two open pollinated varieties viz., Arka Gaurav and Arka Mohini were used for transformation. Agrobacterium strain EHA105 harboring the binary vector pCAMBIA1301 that carries the genes for β-glucuronidase (uid A) and hygromycin phosphotransferase II (hpt II) was used for transformation. GUS histochemical analysis of T0 and T1 plants at various stages of growth followed by molecular analysis using PCR, Southern analysis and RT-PCR allowed selection of transgenics. The method resulted in 17.8% and 11.4% of the T0 plants in Arka Gaurav and Arka Mohini being selected as chimeric and 35.0% and 29.7%, respectively, were identified as stable transformants in the T1 generation based on PCR analysis.  相似文献

The study and development of transformation technology with new selection schemes is important for various fundamental studies and for crop trait improvement via genetic engineering. Here we have shown that hygromycin resistance is an effective system for plum genetic transformation. Embryonic axes of mature seeds were co-cultivated with Agrobacterium strain LBA4404 containing the pC1381 plasmid carrying the hygromycin phosphotranferase gene (hpt) and β-glucuronidase (GUS) gene or with strain EHA105 containing the plasmid pC1301 carrying the same marker and reporter genes. The latter strain containing a pC2301 plasmid carrying the neomycin phosphotransferase gene (nptII) gene was used as a control. Infected explants were placed on shoot induction medium containing either 5 mg L−1 hygromycin or 75 mg L−1 kanamycin for selection. Green shoots developed from the explants under hygromycin pressure. These shoots showed continued and vigorous growth and development upon transfer onto fresh hygromycin medium. PCR using hpt sequence primers, and Southern blot analysis using a probe from the hpt gene, confirmed the presence of the transgenes and their stable integration in regenerated plants. Full transgenic plants were obtained in a greenhouse. Hygromycin selection was very effective and no escapes were observed. The study demonstrated that hygromycin resistance can be used as an effective selectable marker for plum transformation. The new system developed here is important and useful for multiple gene transformation in plum.  相似文献

There has been limited research on measuring potential differences in leaf gas exchange of Arracacha (Peruvian parsnip, Arracacia xanthorriza Bancroft) cultivars, as affected by different environments, as well as its relation to storage root-yield. The present paper reports field measurements of leaf CO2 assimilation rates (A) for five contrasting cultivars grown at two different high-altitude locations. Using a design of plots chosen at random with three repetitions, commercial root production was determined in the two locations at different altitude (1580 and 1930 m). Daily leaf gas exchange was repeatedly monitored with a portable open-mode infrared gas analyzer at different times in both locations during the growth cycle. Root-yield, leaf area and dry weight were measured. Significant differences in leaf photosynthetic rate and in specific leaf area (SLA) were observed among cultivars. Cultivars with high SLA, had high CO2 assimilation. Mean (An) and total (Atot) of CO2 assimilation and SLA were significantly correlated with storage root-yield across cultivars and locations. The three cultivars with the greatest commercial root production also had the highest maximum values for A and the highest specific leaf area, indicating that these two parameters can be used to select for highly productive cultivars of A. xanthorriza.  相似文献

In the current work attempts were made to investigate culture of leaf explants derived from in vitro seedlings of two sweet orange (Citrus sinensis (L.) Osbeck) cultivars, Bingtangcheng and Valencia. Effects of several factors, including culture medium, lighting condition, explant age and genotype on regeneration response were examined based on three parameters, percentage of explants producing shoots, mean number of shoots per explant and shoot forming capacity. Culture of the explants on shoot-inducing media (SIM) composed of MT salts supplemented with different growth regulators gave rise to disparate shoot regeneration, in which SIM1 (MT + 0.5 mg L−1 BA + 0.5 mg L−1 Kinetin + 0.1 mg L−1 NAA + 3% sucrose + 0.8% agar, pH 5.8) was shown to be the most effective medium for direct induction of shoots from leaf explants. Highly significant difference in the response of shoot bud regeneration was noted between the two cultivars, with Bingtangcheng being more responsive than Valencia. Culture of explants from fully developed leaves led to better shoot regeneration capacity in comparison to undeveloped ones. However, the two lighting conditions used herein did not cause significant difference in shoot regeneration. Phenotypic observation and randomly amplified polymorphic DNA (RAPD) analysis confirmed that all the regenerated plants from both genotypes were genetically identical to their donor plants, suggesting absence of detectable genetic variation in the regenerated plants. The data presented here demonstrated that direct initiation of plants from leaf explants has been successfully accomplished. To our knowledge, this is the first report on direct regeneration of shoots from leaf explants in Citrus, which will provide an alternative source for citrus genetic manipulation in the future.  相似文献

The effects of different types of chitosan on seed germination and protocorm development were determined for two orchid species, Dendrobium bigibbum var. compactum and Dendrobium formosum. Six chitosan types derived from polymer or oligomer chitosan each with 70, 80 or 90% levels of deacetylation (P70, P80, P90, O70, O80 and O90, respectively), were evaluated as direct medium supplements at 0, 10, 20, 40 or 80 mg/L in modified VW medium by following seed germination and protocorm growth for 12 weeks. Chitosan of all six tested types and four concentrations were found to significantly enhance the proportion of D. formosum seeds that germinated, when compared to these germinated without chitosan. In contrast, chitosan caused no enhanced germination rate was noted for D. bigibbum var. compactum with all tested chitosans and doses tested. However, almost all types of chitosan at 10 mg/L, except O90, were able to significantly improve the growth of D. bigibbum var. compactum protocorms, whilst 10 or 20 mg/L of P70 chitosan was the best formula to enhance the growth of D. formosum protocorms. It is concluded that chitosan responses in seed germination and protocorm development were somewhat species and developmental stage dependent. Therefore, the appropriate chitosan application for each plant species should be evaluated first before use.  相似文献

A new virus species designated as Grapevine leafroll associated virus-Pr (GLRaV-Pr), which is classified in a distinct phylogenetic group of the genus Ampelovirus (Closteroviridae), was recently characterized from Greek grapevine cultivars. Elimination studies of GLRaV-Pr were carried out in two grapevine cultivars, ‘Mantilaria’ and ‘Prevezaniko’, co-infected with Grapevine rupestris stem pitting associated virus (GRSPaV, Flexiviridae). Both viruses were detected by nested RT-PCR assays. Virus elimination was achieved by combining in vitro thermotherapy with meristem (≤0.2 mm) or shoot tip culture (≤0.5 cm). The survival and regeneration rate of meristems was very low. On the other hand, high survival rates were observed in the cultured shoot tips accompanied with high elimination rates for both viruses. Data obtained in this study indicate that virus elimination depends on the genotype of grapevine. The results confirmed that sanitation is easier for species of the Closteroviridae family than for GRSPaV, whereas it seems that eradication of GLRaV-Pr and GRSPaV is feasible even with larger plant tissue parts if combined with an appropriate thermotherapy profile in vitro.  相似文献
