It is well understood that insufficient oxygen within plant root zones can greatly diminish plant productivity. However, little is known about the effect of elevated root zone oxygen concentrations. Tomato (Lycopersicon lycopersicum Mill., cv. Trust) seedlings were grown in nutrient solutions containing dissolved oxygen (DO) concentration ranging from 5.3 to 40 mg L−1 for 4 weeks. There were no visible symptoms observed on the leaves or stems in any of the treatments. Leaf chlorophyll content was higher in the 40 mg L−1 treatment than with 20 and 30 mg L−1 DO treatments. Two weeks from the start of the experiment, roots in the 40 mg L−1 treatment exhibited stunted growth, became thicker, and had fewer side and fine roots compared to roots in the lower levels of DO treatment. Almost all the measured growth parameters (fresh and dry weights of root, stem, and leaf, leaf area, stem diameter) were significantly reduced in plants grown in the 40 mg L−1 treatment compared to plants in the lower level of DO treatments, except that the plant height increased with the increasing DO concentration. Root respiration increased linearly with increasing DO concentration; however, there was no effect on leaf net CO2 exchange rate. It is suggested that it was safe to enrich root zone DO to as high as 30 mg L−1, although the growth benefit was minor by increasing DO from ambient air saturated level (∼8.5 mg L−1) to 30 mg L−1. Higher than 30 mg L−1 could cause reduction in tomato plant growth.  相似文献

This research included two independent yet build up objectives: (i) to examine the effects of Mn concentration in the irrigation water on the yield and fruit quality of greenhouse-grown bell pepper exposed to heat stress; and (ii) to investigate in a laboratory experiment the role of Mn addition on oxidative and anti-oxidative components of fruit apoplast. The field experiment included four concentrations of Mn in the irrigation water (0, 0.2, 0.6 and 1.0 mg L−1). The effect of Mn application rate on vegetative growth and total fruit yield was insignificant and despite low Mn concentrations in zero-Mn-fed plants, no visible foliar Mn symptoms were observed. A quadratic regression was obtained between high-quality fruit yield and Mn concentration in fruit. Based on the quadratic equation, maximum high-quality yield was obtained when fruit-Mn concentration approached 44 mg kg−1 DW. The correlation also indicates that variations in high-quality yield could be explained by differences in fruit-Mn concentration. The combination of low-Mn nutrition and high air temperature which prevailed during the experiment (summer season) induced severe pale spots and blemished fruits; approximately 50% of fruits were infected in zero-Mn-fed plants. In a laboratory experiment with detached-fruit under high temperature the H2O2 concentration increased whereas ascorbic acid concentration decreased in the apoplast. In Mn-fed fruit the H2O2 concentration was attenuated and the activity of ascorbate oxidase was suppressed, with the result that apoplastic ascorbic acid concentration increased. In light of the known crucial role of Mn in enzyme activities and in detoxification of oxygen free-radicals, the pale spots might be related to Mn effect on oxidative stress-related apoplastic activities. The relationships between incidence of pale spots and plant-Mn concentration may support this hypothesis, but further research is needed to validate it.  相似文献

There has been an increased demand for landscaping plants in Lebanon as a result of numerous reconstruction projects. Sustainable landscape regulations have created a need for regionally adapted taxa, especially those with low water requirements. Therefore, water use of container-grown plants and the impact of fertilization on water use were studied in the following native species: Cercis siliquastrum L. (six mother trees), Malus trilobata Schneid (two mother trees) and Acer syriacum Boin and Gaill (one tree). Two-year-old containerized seedlings were grown at The Ohio State University (Columbus, USA) under two fertilizer rates: 25 or 100 mg N L−1 of 21 N–3.1 P–5.9 K water soluble fertilizer. Water use estimates were made by saturating the containers early in the morning, allowing them to drain for 1 h, weighing them and re-weighing approximately 5 h later. Although there were differences in seedling heights, those grown at 25 mg N L−1 were taller than those at 100, there were few differences in water use per seedling. In August, Cercis seedlings grown under 100 mg N L−1 had higher height adjusted water use (g water cm−1 height h−1, a method for standardizing water use among different sized plants) than those grown under 25 mg L−1. However, there were no differences in height adjusted water use in September attributed to fertilizer rates. In September, Acer seedlings had higher water use cm−2 leaf surface area under 25 than 100 mg N L−1. There were no differences in water use among the progeny from the six Cercis mother trees. However, the seedlings from one Malus tree had higher water use cm−2 leaf surface area than those from the other tree, even though the extant trees were separated by less than 20 m.  相似文献

The effects of application method and concentration of gibberellic acid (GA3), paclobutrazol and chlormequat on black iris performance were assessed. Plants (10 cm high, 4 ± 1 leaves) were sprayed with 125, 250, 375 or 500 mg L−1 or drenched with 0.25, 0.5, 1 or 2 mg L−1 GA3. In a second experiment, the plants were sprayed with 100, 250, 500 or 1000 mg L−1 or drenched with 0.25, 0.5, 1 or 2 mg L−1 paclobutrazol. Other plants were sprayed with 250, 500, 1000 or 1500 mg L−1 or drenched with 100, 250, 375 or 550 mg L−1 chlormequat. In each experiment, the control treatment consisted of untreated plants. Results indicated that the tallest plants (37.3 cm) in the GA3 experiment were those sprayed with 250 mg L−1. The most rapid flowering (160 days after planting) occurred when a 375 mg L−1 GA3 spray was used, whereas flowering was delayed to 200 days using 1 mg L−1 GA3 drench. Drenching with 1 mg L−1 GA3 increased height of the flower stalk by 7 cm compared to the control. Though relatively slow to flower, plants drenched with 1 mg L−1 GA3 had long and rigid stalks, which were suitable as cut flowers. Number and characteristics of the sprouts were not affected by GA3. All paclobutrazol sprays resulted in leaf falcation. A 500 or 1000 mg L−1 paclobutrazol spray resulted in severe and undesirable control of plant height, drastic reduction in stalk height and weight, and delayed flowering. Plants drenched with 0.25 or 1 mg L−1 paclobutrazol were suitable as pot plants. Chlormequat reduced plant height only at the highest drench concentration, which also reduced flowering to 70%. No leaf falcation was observed with GA3 or chlormequat. Fusarium mairei, a paclitaxel-producing fungal endophyte, its effects on taxoid synthesis in Taxus cells were investigated via adding the fungal endophyte culture supernatants (FECS) to the suspension cultures of Taxus cuspidata. The concentration of FECS was determined on its total carbohydrate. When 100 mL of Taxus cell suspension cultures were treated with several dosages of FECS (4, 6 and 8 g) at day10, the cultures treated with 6 g FECS produced the highest yield of paclitaxel (5.84 mg L−1), which was 1.8-fold the yield from controls (3.14 mg L−1). The major elicitor element in FECS was an oligosaccharide of 2 kDa. In addition, the cultures treated with 6 g FECS resulted in 25.86 mg L−1 of the precursor 10-deacetylbaccatin III (10-DAB) accumulation, which was 11 times that of control cultures (2.32 mg L−1), and 4.7 times higher than that of cultures treated with 200 μM methyl jasmonate (MJ) (5.43 mg L−1). These results indicate that FECS favors to stimulate 10-DAB accumulation more effectively than paclitaxel accumulation.  相似文献

Two field experiments were carried out in Egypt during two successive seasons (2007/2008 and 2008/2009). This study aimed to investigate the response of growth, yield quality and some metabolic constituents of onion (Allium cepa L. cv. ‘Giza 20’) to foliar application of putrescine (Put; 25, 50 and 100 mg L−1) and glutamine (Glut; 50, 100 and 200 mg L−1), the former a diamine and the latter an amino acid, either alone, or in combination. Foliar application of Put and Glut, either alone or in combination, significantly increased plant height, number of leaves, fresh weight of leaves/plant, fresh and dry weight/plant, leaf area, leaf area/plant, bulb length, bulb diameter and weight, as well as yield of onion and quality of bulbs. Total soluble sugars, sulfur compounds, total soluble phenols, total free amino acids and total photosynthetic pigment content in leaves were increased by increasing Put and/or Glut concentrations up to 100 and 200 mg L−1, respectively. Generally, foliar application of Put at 100 mg L−1 and Glut at 200 mg L−1 singly, or combined, effectively increased bulb yield and quality. In conclusion, the yield-contributing characters and quality of onion could be improved by application of Put and/or Glut.  相似文献

Lychnis (Caryophyllaceae) consists of about 30 species distributed throughout the temperate regions of the Northern Hemisphere, from East Asia to Europe. Many Lychnis spp. have high ornamental value and cultivated as pot or garden plants. In the present study, in vitro chromosome doubling of several Lychnis spp. was examined in order to widen their variability in horticultural traits. Initially effect of various spindle toxin treatments [100, 500 or 1000 mg l−1 colchicine (COL), 10, 20 or 50 mg l−1 oryzalin (ORY), or 1, 5, 10 mg l−1 amiprophos-methyl (APM)] of nodal segments of a triploid genotype of L. senno (3x) was investigated on survival of nodal segments and chromosome doubling in nodal segment-derived plantlets. Significantly higher percentage (75.0%) of surviving segments after spindle toxin treatment was obtained in 10 mg l−1 ORY treatment. Flow cytometry (FCM) analysis of leaf tissues showed that 9.4–13.8% of plantlets, which were derived from 10 to 20 mg l−1 ORY, or 5 mg l−1 APM treatments, were hexaploid (6x) or ploidy chimera (3x + 6x, 4x + 6x, 5x + 6x, 3x + 4x + 6x). The results obtained by FCM analysis were confirmed by chromosome observation in root tip cells. Thus 10 mg l−1 ORY treatment of nodal segments is suitable for in vitro chromosome doubling of triploid L. senno. Efficient chromosome doubling was also achieved in diploid L. fulgens (2x) and L. sieboldii (2x) by treating nodal segments with 10 mg l−1 ORY: 68.9–88.7% of nodal segments survived after ORY treatment, and 24.7–26.5% of plantlets derived from ORY-treated nodal segments were tetraploid (4x) or ploidy chimera (2x + 4x) in both species. These results indicate that the in vitro chromosome doubling method established for triploid L. senno may be applicable to a wide range of Lychnis spp. Tetraploid L. fulgens and L. sieboldii showed a compact plant form, and had thick stems and deep green leaves compared with the diploid mother plants. Elevated levels of nitrate-nitrogen (NO3-N) in the surficial aquifer above the drinking water quality standard, i.e. maximum contaminant limit (MCL; 10 mg L−1), have been reported in some part of central Florida citrus production regions. Soils in this region are very sandy (sand content >95%), hence are vulnerable to leaching of soluble nutrients and chemicals below the rooting depth of the trees. The objective of this research was to develop N and irrigation best management practices for citrus in sandy soils to maintain optimal crop yield and quality, and to minimize potential leaching of nitrate below the root zone. Six years of field experiment was conducted in a high productive (mean fruit yield > 80 Mg ha−1yr−1) >20-year-old ‘Hamlin’ orange trees [Citrus sinensis (L.) Osbeck] on ‘Cleopatra mandarin’ (Citrus reticulata Blanco) rootstock grown on a well drained Tavares fine sand (hyperthermic, uncoated, Typic Quartzipsamments) in Highland county, FL. Nitrogen rates ranged from 112 to 280 kg ha−1 yr−1 applied as fertigation (FRT), water soluble granular (WSG), 50:50 mix of FRT and WSG, and controlled-release fertilizer (CRF). Tensiometers were used to monitor the soil water content as a basis to schedule optimal irrigation. Fruit yield response over the entire range of N rates was greater for the FRT and WSG sources as compared to that for the WSG + FRT or CRF sources. Using the regression analysis of the fruit yield in relation to N rate, the optimum N rate appeared to be at 260 kg ha−1 yr−1. Based on fruit production response in this study, the N requirement for production of 1 Mg of fruit varied from 2.2 to 2.6 kg across four N sources. This study demonstrated an increased N uptake efficiency, as a result of best management of N and irrigation applications. The optimal N and K concentration in the 4–6-month-old spring flush leaves were 26–30, and 15–18 g kg−1, respectively. However, fruit yield response showed no significant relationship with concentrations of P in the 4–6-month-old spring flush leaves over a range of 0.8–2.4 g kg−1. Ornithogalum ulophyllum Hand.-Mazz. with beautiful white flowers is an important medicinal and ornamental plant of the Middle Eastern countries and need exploitation for commercial propagation. The study reports in vitro mass proliferation of bulblets achieved from twin scales and “in vitro regenerated bulblet” explants on MS medium supplemented with various concentrations of BAP–NAA. The best regeneration on twin scales and “in vitro regenerated bulblets” was obtained on MS medium containing 2 mg l−1 BAP–0.5 mg l−1 NAA and 2 mg l−1BAP–1 mg l−1 NAA, respectively. However, bulb scales seemed to be more potent for bulblet regeneration. A large number of the developing bulblets rooted on the regeneration medium. Remaining non-rooting bulblets were rooted on MS medium containing 1 mg l−1 NAA. All plants were acclimatized in the environmental chamber for 4 weeks and were transferred to the greenhouse for flowering. Regenerated bulblets developed into morphologically normal plants.  相似文献

The in vitro formation of newly formed adventitious buds and shoots from internodal branch segments was studied on 12-month-old plants of Citrus aurantium L. cv. Brazilian. The effects of 6-Benzyladenine (BA) and α-Naphthalene acetic acid (NAA) treatments were evaluated on adventitious bud and shoot regeneration. High rates of bud initiation and shoot development were obtained both with BA supplemented medium, in the range from 1 mg L−1 to 3 mg L−1, and with 0.1 mg L−1 NAA supplemented medium. NAA concentrations above 1 mg L−1 significantly reduced bud initiation and shoot elongation. The results obtained using different in vitro culture vessels such as Petri dishes, tubes and glass culture jars were compared. The highest adventitious bud induction was observed in Petri dishes for internodes cultured in 2 mg L−1 BA supplemented medium, with 95% responsive explants forming 9.0 ± 2.4 adventitious buds. The adventitious buds observed in Petri dishes reached a maximum height of 1 mm, with no further development, while some of the adventitious shoots cultured in tubes and glass culture jars grew over 1 cm in height. A shoot regeneration gradient of the internodes collected along the branch axis was noticed, with basal ones exhibiting higher regeneration frequency.  相似文献

Difficulties to develop an easy and reproducible protocol to get healthy and well formed plants from somatic embryos of papaya (Carica papaya L.) had included low germination, callus production at the base of the embryo radicle and the occurrence of hyperhydric plantlets among others, and by consequence unsuccessful transfer to the field. With the aim of improving a propagation method, the effects of light quality, gelling agent and phloridzin concentration on the germination of somatic embryos of hermaphrodite C. papaya L. var. Maradol were studied. Somatic embryos were grown on half strength MS medium, with the addition of Chen vitamins [Chen, M.H., Wang, P.J., Maeda, E., 1987. Somatic embryogenesis and plant regeneration in Carica papaya L. tissue culture derived from root explants. Plant Cell Rep. 6, 348–351], solidified with three distinct gelling agents: Sigma® Agar–Agar, Difco® Bacto agar and Phytagel®; supplemented with phloridzin and exposed to different light qualities: blue (54 μmol m−2 s−1), red (65 μmol m−2 s−1), gro-lux (68 μmol m−2 s−1), red + blue, white (32 μmol m−2 s−1) and wide spectrum (49 μmol m−2 s−1) during a period of 4 weeks. Results show that light quality and gelling agent had important effects on germination and plant growth, while 3.0 mg L−1 phloridzin had an important role on germination as well as in root development. Somatic embryos exposed to white light, culture medium solidified with 3.0 mg L−1 phytagel and 3.0 mg L−1 phloridzin showed longer roots. Meanwhile, germination and plant length were promoted on an improved culture medium solidified with 7.5 g L−1 Difco® Bacto agar, 3.0 mg L−1 phloridzin and exposed to gro-lux lamps. Under these conditions, 70% of somatic embryos germinated and developed normal roots without hyperhydricity. The regenerated plantlets with well developed roots and shoots were successfully transferred to a greenhouse with a survival rate of 95%.  相似文献

As a precondition for lilac mass propagation, the optimal shoot-multiplication medium for Syringa × hyacinthiflora ‘Luo Lan Zi’ was ascertained mainly based on clustered microshoot inducement and large leaf area establishment in 6-benzyladenine (BAP) (1.00 mg L−1) and zeatin (Z) (0.10 mg L−1) combination. Medium supplied with lower level of BAP (0.50 mg L−1) and auxin (IAA) (0.25 mg L−1) was not suitable for lilac shoot proliferation, but it could be competent for long-term preservation of the un-rooted shoots so that subsequent proliferation culture could be carried out at anytime. In addition, excess height growth which resulted in low transplanting survival rate was effectively controlled by decrease in node number when paclobutrazol (PBZ) was applied in rooting medium at a concentration of 1.00 mg L−1 after taking into account the effects on shoot height, rooting, persistent leaf area and PBZ carry-over. An important overwintering treatment was to use a plastic chamber covering for plants in the greenhouse prior to field planting to ensure adequate biomass of stem and underground parts not only in the current growing season but also in the subsequent years.  相似文献

Dendrobium nobile Lindl. is a popular temperate Chinese orchid commonly marketed as a traditional medicinal plant. Seedlings of Dendrobium nobile Lindl. produced floral buds (33.3–34.8%) precociously on a defined basal medium (1/2 MS) containing paclobutrazol (PP333) at 0.5 mg L−1 or thidiazuron (TDZ) at 0.1 mg L−1 within 4 months of culturing. The frequency of floral buds formation can be further increased to 95.6% by growing seedlings in a PN (PP333 0.3 mg L−1 + NAA 0.5 mg L−1)-containing medium followed by transfer onto 1/2 MS medium with PP333 and TDZ (PP333 + TDZ). However, flower developed was deformed under 25 °C but it developed fully when grown in a lower temperature regime (23 °C/18 °C, light/dark) for 45 days. Under optimal condition, in vitro flowering was observed about 6 months after seed sowing.  相似文献

Inadequate watering of potted ornamental plants during retail can cause a decline in quality as well as plant death. Abscisic acid (ABA) is the main phytohormone controlling plant responses to drought stress, including regulation of stomatal opening and closure. ABA may become available for the greenhouse industry in 2010. The objective of this study was to determine whether exogenous ABA applications can be used to induce stomatal closure and reduce water loss from the substrate. We drenched salvia (Salvia splendens F. Sellow. ex Roem & Shult.) ‘Bonfire Red’ with 50 mL of ABA solutions at concentrations of 0, 250, 500, 1000, and 2000 mg L−1, after which plants were no longer irrigated. ABA drenches slowed down substrate water loss by reducing transpiration in a dose-dependent manner. Stomata closed within 3 h after ABA treatment, decreasing transpirational water loss. However, ABA drenches also caused abscission of lower leaves, resulting in approximately 60% leaf loss with 2000 mg L−1 ABA. ABA drenches with 250 mg L−1 and 500 mg L−1 increased the shelf life of salvia ‘Bonfire Red’ by 3 days, without excessive leaf abscission. ABA can be used to increase the shelf life of salvia, but the lowest effective concentration should be used to minimize leaf abscission.  相似文献

Most of the Japanese plum (Prunus salicina) cultivars grown in Israel produce relatively small fruit. Application of 2 l solution tree−1 of 25 mg l−1 2,4-dichlorophenoxypropionic acid (2,4-DP) as butoxyethyl ester (Power™), 15 mg l−1 3,5,6-trichloro-2-pyridyloxyacetic acid (3,5,6-TPA) as free acid (Maxim®), or 25 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) + 30 mg l−1 naphthaleneacetic acid (NAA) (0.3% Amigo™) at the beginning of pit-hardening, when fruitlet diameter was ca. 22 mm, caused an appreciable and significant increase in fruit size. The yield of large fruit per cv.: ‘Kesselmen’ (100% increase), ‘Songold’ (100%), ‘Black Diamond’ (800%), ‘Royal Diamond’ (160%) and ‘Royal Zee’ (100%). As a result, the total yield of all five cultivars was also increased dramatically. Anatomical studies with ‘Songold’ revealed that the main effect of these synthetic auxins was via direct stimulation of fruit cell enlargement. The above auxins had no negative effect either on fruit quality at harvest (and after 1 week in shelf-life), or on return yield in the following year.  相似文献

Kosteletzkya virginica, a perennial dicot halophytic species of the Malvaceae, is native to American salt marsh. It was introduced into China as a potential species to improve coastal wetlands and to develop ecologically sound saline agriculture. K. virginica adapts excellently to the tidal-flat habitats in China's east coast, with multiple eco-benefits; in particular, its seed oil could be used to produce biodiesel. The purpose of this study was thus to develop a standardized protocol to induce a high frequency of callus and subsequent plantlet regeneration system for a K. virginica breeding program with the final objective of applying transgenic techniques to improve seed oil yield. The embryonic axes of K. virginica were used as explants for callus induction, shoot induction from the callus and then adventitious root induction from the shoots on nine culture media with different hormone combinations. The best results were achieved on the following media: (1) 93.94% callus induction on MS medium supplemented with 1.0 mg L−1 indole-3-acetic acid (IAA), 0.3 mg L−1 kinetin, 30 g L−1 sucrose and 8 g L−1 agar; (2) 65.83% shoot induction on 1/2MS medium supplemented with 0.1 mg L−1 IAA, 0.5 mg L−1 zeatin, 30 g L−1 sucrose and 8 g L−1 agar; (3) 96.67% rooting on MS medium containing 30 g L−1 sucrose and 8 g L−1 agar. The survival rate of plantlets by organogenic regeneration was 85% after being transplanted into potting soil in flowerpots and placed in the greenhouse. This experiment indicates that we established successful callus induction and plant regeneration protocols for K. virginica.  相似文献

Lebanon's native flora is threatened by loss of natural habitat to rural and urban development and the increased demand of plant materials for landscaping. Despite Lebanon's floristic richness, most taxa used for landscaping are non-native. This study was done to determine if Cercis siliquastrum (L.) is amenable to container production. Therefore, six open pollinated seeds sources native to Lebanon were grown under two fertilizer rates to study growth, N, P, K uptake efficiency, and partitioning. Two-year-old seedlings were planted in 11 L containers in a 3:1 pine bark:compost substrate. Seedlings within each seed source or mother tree were grown at either 25 or 100 mg N L−1 from 21N–3.1P–5.9K water-soluble fertilizer. Seedlings of all sources grown under 25 mg N L−1 had greater dry weight than those grown at 100 mg. Nutrient loading occurred in plants under the high fertilizer rate, although total plant N, P, and K content were not affected by fertilizer rate. There were significant differences in mineral nutrient uptake and nutrient use efficiencies among the seed sources. The results show that C. siliquastrum is amenable to container production. The great variation in growth rate and nutrient use efficiency among the limited number of seed sources studied suggest that significant improvement can be made through mother tree selection and/or clonal propagation of superior individual plants within a source.  相似文献

A romaine-type lettuce (Lactuca sativa L.) cv. Corsica was cultivated during three successive crop seasons (late-spring, late-autumn and late-winter) in the same soil of an experimental greenhouse in S.W. Peloponnese, Greece. Seven long-term fertilization treatments were tested for their effect on plant growth and nitrate concentration in the external lettuce leaves. Treatments included: three different doses of organic fertilization (composted sheep manure) applied at the start of each crop season, three different doses of inorganic N fertilization applied via fertigation during each crop season, and a control treatment in which no fertilizer was applied. A drip irrigation system was used to water all plants. The highest nitrate levels were observed in the medium and maximum inorganic fertilization treatments (572–664 mg kg−1) in all crop seasons. They were significantly higher compared to the respective organic fertilization treatments (253–435 mg kg−1) and all other fertilization treatments (148–435 mg kg−1). Crop season affected lettuce growth more than nitrate accumulation in the lettuce leaves: lettuce biomass production was the smallest and most uniform in the late-autumn season and did not respond to the fertilization treatments tested (ranging from 409 to 439 g plant−1), while in the late-spring season biomass production was the highest and most variable (561–841 g plant−1), it correlated with nitrate concentration in the leaves and in the medium and maximum inorganic fertilizer doses it significantly exceeded production from all other fertilization treatments (827–841 g plant−1). Following the three crop seasons the residual availability of N, P and K was clearly enhanced in the soil receiving the organic compared to the inorganic fertilization. Nitrate concentration in lettuce leaves was far below the upper limits set by the European Commission in all fertilization treatments throughout the three crop seasons, a result attributed mainly to the sufficient level of light intensity and duration throughout the year in Southern Greece.  相似文献

The main cherry cultivar grown in the warm climate of Israel, ‘Bing’, produces relatively small fruit. Over three consecutive years (2003–2005), application of 50 mg l−1 2,4-dichlorophenoxypropionic acid [2,4-DP; as its butoxyethyl ester (Power™)], 10 mg l−1 3,5,6-trichloro-2-pyridyloxyacetic acid [3,5,6-TPA; as the free acid (Maxim®)], or 25 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) plus 30 mg l−1 naphthaleneacetic acid (NAA; 0.3% Amigo™), at the beginning of pit-hardening when fruitlet diameter was ca. 13 mm caused appreciable and significant increases in fruit size and total yield, except when the crop load was heavy. Anatomical studies revealed that the main effect of these synthetic auxins was via direct stimulation of fruit cell enlargement. The above auxins had no negative effect on fruit quality, either at harvest or after 1 month of storage at 0 °C, or on return yield in the following year.  相似文献

Protocols for in vitro regeneration and production of in vitro-propagated plants and a transformation system were developed for Mirabilis jalapa (Nyctaginaceae). Among the types of explants and the different media tested, consistent shoot regeneration was obtained only from nodal segments grown in a regeneration medium consisting of Murshashige and Skoog medium supplemented with 2 mg l−1 6-benzyladenine, 2 mg l−1 zeatin and 1 mg l−1 indole acetic acid. Regeneration efficiency was dependent on the type of plant – white or pink flowers – used as the source of explants. Stable transformation was obtained following inoculation of nodal segments with Agrobacterium tumefasciens strain EHA105, which harbours the binary plasmid pAD1339 containing both nptII and gus genes under the control of the 35S promoter. Transformation was confirmed by PCR and Southern blot analysis of genomic DNA from mature regenerated plants. β-Glucuronidase (GUS) activity was observed only in tissues regenerated from in vitro-grown plants and not in tissues originating from greenhouse-grown plants. GUS expression was not uniform in regenerated leaves and showed a chimera pattern.  相似文献
