Genetic diversity of 54 accessions of Aegilops tauschii from five countries was assessed using sequence-tagged microsatellites (or simple sequence repeats, SSRs) and amplified fragment
length polymorphisms (AFLPs). In the case of AFLP analysis, a total of 256 amplification products obtained, 234 of them were
polymorphic across all the 54 accessions. A total of 224 fragments were obtained from the 24 SSR primers and 219 of fragments
were polymorphic across all the genotypes screened. Based on both AFLP and SSR markers, the highest percentage of polymorphisms
were obtained in Iranian and accessions of unknown origin. The highest polymorphic information content (PIC) value was observed
for SSRs (0.82) while the highest marker index (MI) value was for AFLPs (8.5) reflecting the hyper-variability of the first
and the distinctive nature of the second system. Principal co-ordinate analysis (PCO) revealed congruent patterns of genetic
relationships for both data sets, but did not group accessions strictly according to their geographical origins. Poor correlation
was found between AFLP and SSR marker loci. This low association may be due to low number of AFLP and SSR markers. These results
show that molecular markers can help to organize the genetic variability and expose useful diversity for breeding purposes. 相似文献

Genetic diversity among 40 accessions (Coffea canephora) of robusta coffee genepool available in India was determined in comparison with 14 representative samples from a C. canephora core collection and three accessions of C. congensis, using amplified fragment length polymorphism (AFLP) and simple sequence repeats (SSR) markers. Both these molecular approaches were able to generate unique fingerprints for each of the accessions analysed. All the 12 SSR primers used in the present study were found polymorphic, with an average of six alleles per primer pair. Comparative analysis revealed the higher amount of diversity in representatives from a core collection than in the Indian genepool. Moreover, a total of 205 polymorphic AFLP bands were scored in all the 57 accessions analysed. The genetic relationship among 57 accessions was compared on the basis of SSR and AFLP polymorphisms. Genetic similarity dendrograms showed high correlation between the two marker systems. This study clearly established the high amount of diversity present in core samples, which is not represented in Indian genepool. Furthermore, the three accessions of C. congensis did not exhibit any significant diversity from other robusta accessions supporting the school of thought that C. congensis forms a biotype of C. canephora. The potential use of SSRs and AFLP markers in genetic diversity analysis for better ex situ management and also for exploitation of diversity in breeding programmes is discussed. 相似文献

Opaque endosperm is the main phenotypic indicator for waxy rice, but other phenotypic and genotypic variation among waxy rice
accessions has largely been ignored. Previous studies showed that wide diversity in starch physiochemical properties exists
in both indica and japonica waxy rices, especially for starch gelatinization temperature (GT) which could be divided into a high- and a low-GT group.
In the present study, amplified fragment length polymorphism (AFLP) and inter-simple sequence repeat (ISSR) molecular markers
were employed to examine genetic diversity and relationships of 56 waxy rice accessions. A total of 358 AFLP fragments were
amplified with five primer combinations, showing a high level of polymorphism (78.3%). A total of 190 ISSR bands were generated
with a single primer and a primer pair, showing a very high level of polymorphism (92.2%). The genetic distance matrices obtained
from the two sets of markers were significantly correlated (r = 0.731, P = 0.004). The dendrogram generated with combined AFLP and ISSR markers could clearly differentiate the indica and japonica groups. Newly released varieties and breeding lines within each subspecies tended to be clustered together, whereas landraces
were more distantly placed in the dendrogram. Only one AFLP band was found specific to the indica type, while no specific bands were found for starch GT. The implications for the conservation and breeding of waxy rice are

Sweet potato (Ipomoea batatas L.) is the fifth most important crop in the developing countries after rice, wheat, maize and cassava. The amplified fragment
length polymorphism (AFLP) method was used to study the genetic diversity and relationships of sweet potato accessions in
the germplasm collection of Sokoine University of Agriculture, Morogoro and Sugarcane Research Institute, Kibaha, Tanzania.
AFLP analysis of 97 sweet potato accessions using ten primer combinations gave a total of 202 clear polymorphic bands. Each
one of the 97 sweet potato accessions could be distinguished based on these primer combinations. Estimates of genetic similarities
were obtained by the Dice coefficient, and a final dendrogram was constructed with the un-weight pair-group method using arithmetic
average. AFLP-based genetic similarity varied from 0.388 to 0.941, with a mean of 0.709. Cluster analysis using genetic similarity
divided the accessions into two main groups suggesting that there are genetic relationships among the accessions. Principal
Coordinate analysis confirmed the pattern of the cluster analysis. Analysis of molecular variance revealed greater variation
within regions (96.19%) than among regions (3.81%). The results from the AFLP analysis revealed a relatively low genetic diversity
among the germplasm accessions and the genetic distances between regions were low. A maximally diverse subset of 13 accessions
capturing 97% of the molecular markers diversity was identified. We were able to detect duplicates accessions in the germplasm
collection using the highly polymorphic markers obtained by AFLP, which were found to be an efficient tool to characterize
the genetic diversity and relationships of sweet potato accessions in the germplasm collection in Tanzania. 相似文献

Kenaf (Hibiscus cannabinus L.) is one of the world's most economically important fiber crops. In order to identify different varieties, and investigate its diversity and genetic relationships, twenty-three kenaf accessions and two accessions of its relative, roselle (H. sabdariffa var. altissima), were analyzed by morphological characterization and AFLP fingerprinting. It is very difficult to identify kenaf accessions based merely on morphological characters, due to their limited variation. For the AFLP study, a total of 505 polymorphic markers (out of 560) were produced by six selected AFLP primer combinations. The AFLP fingerprinting was effective in identifying all kenaf accessions included in the study. Kenaf and roselle are independent species with close relationships, and great genetic diversity was also detected among the kenaf accessions with different origins, based on the analysis of the AFLP markers. The AFLP analysis strongly supports the opinion that kenaf originated in Africa. It also demonstrated that the dissemination of kenaf was from Africa through Asia to Central and North America. 相似文献

Couepia is a genus distributed in tropical regions of America. The nut of some Couepia species is used as fresh fruit and oil source for local communities. Despite its critical situation of conservation and its
economic potential, there is a lack of information on the genetic variability of Couepia species. This study examines AFLP variation among 40 accessions of Couepia collected in the Colombian Amazonian region, representing two species: Couepia
dolichopoda and Couepia
subcordata. The individuals were examined for 96 markers generated from four EcoRI/MseI primer pairs, with 80% polymorphism across all accessions studied. According to cluster analysis, 40 accessions were grouped
into two major clusters, corresponding to the two species analyzed, except one case whose situation is discussed. In C. dolichopoda accessions, significant correlation between the clustering pattern and the geographical origin was detected; the extent of
variation within and among its collect sites was examined by AMOVA. The knowledge about the genetic variability of the accessions

Forty-four accessions of cultivated peanut (Arachis hypogaea L.) representing sixbotanical varieties of two subspecies along with three accessions ofthe wild relative A. monticola Krapov et Rigoni were evaluated for their genetic relationships using theAFLP marker technology. Fifteen AFLP primer pairs (EcoRI/MseI) generated 28distinct polymorphic markers that were employed to develop uniqueprofiles of all accessions and to construct a phenogram. The resultsshowed that the botanical varieties aequatoriana and peruviana werecloser to subspecies hypogaea than subspeciesfastigiata Waldr. to which they belong, and the wildA. monticola was notdistinct from the cultivated A.hypogaea. A total of 8117 suitable SSR-contaning ESTs were acquired by screening from a Malus EST database, among which dinudeotide SSRs were the most abundant repeat motif, within which, CT/TC followed by AG/GA were
predominant. Based on the suitable sequences, we developed 147 SSR primer pairs, of which 94 pairs gave amplifications within
the expected size range while 65 pairs were found to be polymorphic after a preliminary test. Eighteen primer pairs selected
randomly were further used to assess genetic relationship among 20 Malus species or cultivars. As a result, these primers displayed high level of polymorphism with a mean of 6.94 alleles per locus
and UPGMA cluster analysis grouped twenty Malus accessions into five groups at the similarity level of 0.6800 that were largely congruent to the traditional taxonomy. Subsequently,
all of the 94 primer pairs were tested on four accessions of Pyrus to evaluate the transferability of the markers, and 40 of 72 functional SSRs produced polymorphic amplicons from which 8
SSR loci selected randomly were employed to analyze genetic diversity and relationship among a collection of Pyrus. The 8 primer pairs produced expected bands with the similar size in apples with an average of 7.375 alleles per locus. The
observed heterozygosity of different loci ranged from 0.29 (MES96) to 0.83 (MES138), with a mean of 0.55 which is lower than 0.63 reported in genome-derived SSR marker analysis in Pyrus. The UPGMA dendrogram was similar to the previous results obtained by using RAPD and AFLP markers. Our results showed that
these EST-SSR markers displayed reliable amplification and considerable polymorphism in both Malus and Pyrus, and will contribute to the knowledge of genetic study of Malus and genetically closed genera. 相似文献

Iwateyamanashi (Pyrus ussuriensis var. aromatica) is one of the Pyrus species which grows wild in Japan. The number of Iwateyamanashi trees has been decreasing, so conservation and evaluation
is urgently needed. Over 500 accessions of Pyrus species collected from Iwate in northern Tohoku region are maintained at Kobe University as an Iwateyamanashi germplasm collection.
In order to investigate the genetic diversity, five SSR (simple sequence repeat) markers, developed from Japanese and European
pear were examined for 86 Pyrus individuals including 58 accessions from Iwate. These SSR loci could discriminate between all the Iwate accessions except
for 10 that bear seedless fruit, as well as determine the genetic diversity in Iwateyamanashi germplasms. High levels of variation
were detected in 41 alleles and the mean observed heterozygosity across 5 loci was 0.50 for the Iwate accessions. Seedless
accessions sharing identical SSR genotype with the local pear variety “Iwatetanenashi” were supposed to have been propagated
vegetatively via grafting. In an UPGMA phenogram, Japanese pear varieties (P. pyrifolia) were clustered into two groups with some Iwate accessions including seedless ones. Another 38 Iwate accessions were not
clustered clearly, and there was no clear relationship between these accessions and geographical distribution or morphological
characters. Allele frequency revealed that the Iwate accessions were genetically more divergent than the Japanese pear varieties.
Most Japanese pears possessed a 219 bp deletion at a spacer region between the accD and psaI genes in the chloroplast DNA (cpDNA), but other Pyrus species and two Iwateyamanashi trees did not. In the Iwate accessions, 79.3% had a deletion type cpDNA and others had a standard
type cpDNA without deletion. These results are indicative of the wide range of genetic diversity in the Iwate accessions which
include Japanese pear varieties. A combination of SSR and cpDNA analyses revealed high heterogeneity in Iwateyamanashi and
coexistence of Iwateyamanashi and hybrid progeny with P. pyrifolia. These could be reasons for the wide range of continuous morphological variation described previously. 相似文献

Asian cotton (Gossypium arboreum L.) was once widely cultivated in China. It has also been a valuable source of genetic variation in modern cotton improvement. In this study, the genetic diversity of selected G. arboreum accessions collected from different regions of China was evaluated by microsatellite (simple sequence repeats, SSRs) analysis. Of the 358 microsatellite markers analyzed, 74 primer pairs detected 165 polymorphic DNA fragments among 39 G. arboreum accessions examined. Twelve accessions could be fingerprinted with one or more SSR markers. With the exception of two accessions, DaZiJie and DaZiMian, genetic similarity coefficients among all accessions ranged from 0.58 to 0.87 suggesting high level of genetic variation in the G. arboreum collections. The UPGMA dendrogram constructed from genetic similarity coefficients revealed positive correlation between cluster groupings and geographic distances. In addition, comparison of the microsatellite amplification profiles of the diploid G. arboreum and tetraploid Gossypium hirsutum L. found that size distribution of amplified products in G. arboreum was dispersive and that of G. hirsutum was relatively concentrated. The genetic variation and relationships among 31 accessions of Phaseolus vulgaris L., and two representatives of Vigna unguiculata L., were evaluated by AFLP analysis. A total of 263 DNA fragments across all materials were scored using nine primer combinations, averaging 32 per primer. More than 95% of the amplification products showed polymorphism, indicating high variation at the DNA level among these accessions. Pair-wise genetic similarity (Jaccard's coefficient) ranged from 0.553 to 0.840, with a mean of 0.765. Twenty-three accessions (70%) clustered into three groups. A majority of the commercial cultivars (91%) clustered within a single group, whereas the landraces were distributed along all the variation. An apparent correlation with phaseolin types was detected. Results of this study suggest that Brazilian landraces truly represent the overall genetic variability of Phaseolus vulgaris, confirming the multiple origins of these materials, and their potential as a source of variation for breeding programs. 相似文献

The cultivated types of Brassica rapa L. em. Metzg. consist of morphologically distinct subspecies such as turnip, turnip rape, Chinese cabbage, pak choi and pot
herb mustard which are classified as ssp. rapa, ssp. oleifera, ssp. pekinensis, ssp. chinensis and ssp. nipposinica (syn. ssp. japonica), respectively. We attempted to elucidate the phylogenetic relationships among the cultivated types of B. rapa. Thirty-two accessions from the Eurasian Continent were analyzed using AFLP markers with a cultivar of B. oleracea as an outgroup. In total, 455 bands were detected in the ingroup and 392 (86.6%) were polymorphic. The Neighbor-Joining tree
based on the AFLP markers indicated that the accessions of B. rapa were congregated into two groups according to geographic origin. One group consisted of ssp. rapa and ssp. oleifera of Europe and Central Asia and the other included all the subspecies of East Asia. Our results suggest that cultivars from
East Asia were probably derived from a primitive cultivated type, which originated in Europe or in Central Asia and migrated
to East Asia. This primitive cultivated type was probably a common ancestor of ssp. rapa and ssp. oleifera. The Neighbor-Joining tree also shows that leafy vegetables in East Asia such as ssp. pekinensis, ssp. chinensis and ssp. nipposinica were differentiated several times from the distinct cultivars of ssp. oleifera in East Asia. 相似文献

Amplified fragment length polymorphism (AFLP) markers were used to characterize the genetic diversity within and among natural populations and cultivars of Hedysarum coronarium. Twelve populations within Tunisia were evaluated with three AFLP primer combinations. A total of 207 reproducible bands was detected of which 178 (86%) were polymorphic. The great discriminative power of AFLP markers and their ability to represent genetic relationships among Hedysarum plants was demonstrated. Genetic diversity within and among populations was assessed through Principal Component Analysis (PCA) and cluster analysis by using the Neighbor-joining clustering algorithm. AFLP technology has provided evidence of a high degree of intra- and inter-population genetic diversity in H. coronarium. AFLP banding patterns provided molecular markers correlated with the plants’ geotropism. In addition, AFLP markers can differentiate wild accessions from cultivars. Wild annual Cicer gene pools contain valuable germplasm for chickpea improvement programs. Previous research showed that duplication might
exist in accessions collected from these gene pools, which would hinder chickpea breeding and related research. AFLP (amplified
fragment length polymorphism) markers were used to fingerprint the world collections of the primary and secondary gene pools
including C. reticulatum Lad., C. bijugum K.H. Rech., C. judaicum Boiss. and C. pinnatifidum Jaub. et Sp. Duplicates were detected in a total of 24 accessions in both the gene pools, highlighting the necessity to fingerprint
the germplasm. Genotypic difference was detected as gene pool specific, species specific and accession specific AFLP markers.
These were developed into fingerprinting keys for accession identification between and within species and gene pools. Use
of AFLP markers to detect duplicates and to identify accessions is a reliable method which will assist in the characterisation
and use of wild annual Cicer germplasm in chickpea improvement programs. We recommend the procedure presented in this paper as a standard approach for
the precise genetic identification and characterisation of future world collections of wild Cicer, to keep germplasm integrity and to benefit chickpea breeding and related research programs. 相似文献

The present study, using RAPD analysis, was undertaken to characterize genetic variation in domesticated cowpea and its wild progenitor, as well as their relationships. The materials used consisted of 26 domesticated accessions, including accessions from each of the five cultivar-group, and 30 wild/weedy accessions, including accessions from West, East and southern Africa. A total of 28 primers generated 202 RAPD bands. One hundred and eight bands were polymorphic among the domesticated compared to 181 among wild/weedy cowpea accessions. Wild accessions were more diverse in East Africa, which is the likely area of origin of V. unguiculata var. spontanea. Var. spontanea is supposed to have spread westward and southward, with a loss of variability, loss counterbalanceed in southern Africa by introgressions with local perennial subspecies. Although the variabilty of domesticated cowpea was the highest ever recorded, cultivar-groups were poorly resolved, and several results obtained with isozyme data were not confirmed here. However primitive cultivars were more diverse than evolved cultivars, which still suggests two consecutive bottlenecks within domesticated cowpea evolution. As isozymes and AFLP markers, although with a larger number of markers, RAPD data confirmed the single domestication hypothesis, the gap between wild and domesticated cowpea, and the widespread introgression phenomena between wild and domesticated cowpea. 相似文献

Dead seeds of a fodder beet cultivar ‘Elvetham’ stored under ambient conditions since 1880 were compared to a homonymous sample
preserved in an on-farm situation in Denmark. DNA was isolated from single seeds and successfully applied to Amplified Fragment
Length Polymorphism (AFLP) analysis of the accessions. Six primer pairs were used to determine the similarity between the
two accessions based on 112 polymorphic bands. Furthermore, similarity among seven cultivars of fodder beets representing
the main types used in Scandinavia at the end of the 19th century was determined. This analysis was based on 152 polymorphic
bands. Differentiation among the seven cultivars was determined to a mean G
ST value of 0.438, while G
ST between the two ‘Elvetham’ accessions was 0.266. A principal coordinate analysis based on jaccards similarity index illustrates
that the two ‘Elvetham’ accessions are different from each other. The differentiation is higher than the value found between
two separate ‘Eckerndorfer’ accessions. The results indicate that the cultivated accession has changed. Additionally, the
value of applying old dead seed material for documentation in gene banks is demonstrated.
During the analysis it was found that DNA isolated from seeds and leaves behaved differently in the AFLP process, however,
the two fractions assigned to their common accession. 相似文献

Portuguese wheat landraces, ‘Arrancada’ were collected from the Aveiro region, Portugal before the 1950s. We found in eight
accessions of `Arrancada' hexaploid wheat with the long glume phenotype. We assessed the comparative genetic diversity among
Portuguese `Arrancada' wheat and Triticum petropavlovskyi Udacz. et Migusch. using AFLP assays and discuss the origin of long glumed `Arrancada' wheat. With the four primer pairs
a total of 4885 visible bands were scored corresponding to 99 AFLP markers as putative loci, of which 55 markers (54%) were
polymorphic. UPGMA clustering and PCO grouping showed that long glumed ‘Arrancada’ wheat and T. petropavlovskyi were genetically diverse. Long glumed ‘Arrancada’ hexaploid wheat separated into two clusters (groups) in both the UPGMA
dendrogram and in PCO analysis. Four long glumed accessions fell in the cluster of tetraploid wheat. A similar argument could
be made for another four accessions which belong to the cluster of hexaploid wheat. The substantial level of genetic variation
indicated that long glumed ‘Arrancada’ wheat and T. petropavlovskyi originated independently. It is most likely that the P-gene of long glumed ‘Arrancada’ hexaploid wheat was introduced from T. turgidum ssp. polonicum (L.) Thell. to T. aestivum via natural introgression or breeding. We suggest that the long glumed ‘Arrancada’ hexaploid wheat did not originate from

Genetic diversity throughout the rapeseed (Brassica napus ssp. napus) primary gene pool was examined by obtaining detailed molecular genetic information at simple sequence repeat (SSR) loci
for a broad range of winter and spring oilseed, fodder and leaf rape gene bank accessions. The plant material investigated
was selected from a preliminary B. napus core collection developed from European gene bank material, and was intended to cover as broadly as possible the diversity
present in the species, excluding swedes (B. napus ssp. napobrassica (L.) Hanelt). A set of 96 genotypes was characterised using publicly available mapped SSR markers spread over the B. napus genome. Allelic information from 30 SSR primer combinations amplifying 220 alleles at 51 polymorphic loci provided unique
genetic fingerprints for all genotypes. UPGMA clustering enabled identification of four general groups with increasing genetic
diversity as follows (1) spring oilseed and fodder; (2) winter oilseed; (3) winter fodder; (4) vegetable genotypes. The most
extreme allelic variation was observed in a spring kale from the United Kingdom and a Japanese spring vegetable genotype,
and two winter rape accessions from Korea and Japan, respectively. Unexpectedly the next most distinct genotypes were two
old winter oilseed varieties from Germany and Ukraine, respectively. A number of other accessions were also found to be genetically
distinct from the other material of the same type. The molecular genetic information gained enables the identification of
untapped genetic variability for rapeseed breeding and is potentially interesting with respect to increasing heterosis in

We used amplified-fragment-length polymorphism (AFLP) markers to evaluate genetic variation in a set of bread wheat (Triticum aestivum L.) landraces and improved materials. Landraces collected from different geographic and agro-ecological zones in Pakistan
in 1987, 1989 and 1991 were separated into two groups based on their geographic origins: northern (Himalaya) and south-western
(Balochistan) Pakistan. Six AFLP primer combinations detected 453 AFLP markers in the 43 landrace accessions and four high-yield
varieties (HYVs). Of these, 225 (49.67%) were rare (shared with < 5% of all accessions). Among these rare alleles, 23 (10.22%)
were common in the Himalaya (shared with > 10% of accessions collected there) but were not found in Balochistan. We conclude
that there is a higher probability of collecting rare alleles at overall, but which are in contrast locally common ones in
the Himalayan region. Gene diversity was 0.17 in the Himalayan group and 0.15 in the Balochistan group. Considerable genetic
variability was found in both groups. Accessions from different agro-ecological zones were indistinguishable by cluster analysis,
indicating intensive seed trading within the country. Cluster analysis indicated that the landraces and the HYVs are genetically
distinct; suggesting that genetic erosion of wheat landraces has been unlikely taken in place. This study provides an example
of how analysis of existing materials and data, can serve as a basis for future collection planning and conservation policies. 相似文献
