The swimming crab (Portunus trituberculatus) is an economically important species in Asian aquaculture. Implementing growth‐related traits of P. trituberculatus into genetic breeding programmes is an ongoing effort. We used a previously published genetic linkage map of P. trituberculatus, containing 55 simple sequence repeat (SSR) markers and 172 amplified fragment length polymorphism (AFLP) techniques to map quantitative trait loci (QTL) for growth‐related traits in a single full sibling F2 family. Ten growth‐related traits were measured for QTL mapping. Composite interval mapping identified 9 QTL on the female map and 16 on the male map. Individual QTL with additive effects explained 11–38% of the phenotypic variance for various traits using the female parent's map, and from 1% to 21% using the male parent's map. Two QTL explaining a large percentage of variation in body weight were detected on chromosome 17 on the female map, and on chromosome 16 on the male map, and contributed 38% and 18% of the phenotypic variance respectively. This is the first study to report the detection and positioning of major QTL affecting growth in a true crab species (Brachyura). The mapping of growth‐related QTL in this study raises the possibility of improving the growth of P. trituberculatus through marker‐assisted selection.  相似文献

The γ‐aminobutyrate type A receptor‐associated protein (GABARAP) is a ubiquitin‐like modifier implicated in membrane trafficking and fusion events involving the γ‐aminobutyrate type A receptor, autophagy and apoptosis. In this study, the gene encoding GABARAP was cloned from swimming crab Portunus trituberculatus (PtGABARAP) based on the expression sequence tag (EST). The full‐length cDNA of 664 bp includes a 5′ untranslated region (UTR) of 87 bp, a 3′ UTR of 223 bp with a poly(A) tail, and an open reading frame (ORF) of 354 bp encoding a polypeptide of 117 amino acids with a predicted molecular weight of 13.96 kDa. The deduced amino acid sequence shares high similarity (93%–100%) with GABARAPs from other species and includes a conserved Atg8 domain. In a phylogenetic analysis PtGABARAP clustered with GABARAPs from other species, and more widely with other GABARAP family proteins. The impact of elevated ocean acidification (OA) on P. trituberculatus behaviours was investigated, and real‐time RT‐PCR revealed that PtGABARAP expression was up‐regulated after OA exposure. Ocean acidification also caused crabs anxiety‐like behaviours, like the shoal average speed increase, preference for dark environment (scototaxis) and fast exploration. The results indicated that GABARAP might be involved in the interactions of GABAA receptors and elevated‐CO2 seawater.  相似文献

The purpose of this study was to investigate the effects of inbreeding on the immune responses and antioxidant status of Portunus trituberculatus juveniles. Results showed that inbreeding affected the total haemocyte count, and phagocytic, pro‐phenoloxidase (propo), phenoloxidase and antibacterial activities decreased after the seventh generation. Antioxidant status showed a similar pattern: total antioxidant capacity, superoxide dismutase (SOD) activity and GSH/GSSG in the cell‐free haemolymph and hepatopancreas decreased, while catalase activity in the cell‐free haemolymph increased (P < 0.05). There were no significant differences in α2‐macroglobulin and bacteriolytic activities in the cell‐free haemolymph and glutathione peroxidase activity in the cell‐free haemolymph and hepatopancreas among nine inbreeding generations. Gene expression levels of proPO and crustin in haemocytes and SOD in haemocytes and hepatopancreas also decreased significantly as the inbreeding generations increased. The results suggest that a high level of inbreeding could severely reduce the physiological health of P. trituberculatus. Our obtained data would be particularly useful for P. trituberculatus breeding programmes.  相似文献

A 30‐day experiment was conducted to investigate the effects of long‐term low‐salinity stress on the growth performance and osmotic related chlorine ion channel ligand regulation: gamma‐aminobutyric acid (GABA) content and GABAA receptor‐associated protein (GABARAP) expression in Portunus trituberculatus. The salinity levels of both the control group and the experimental group were 30 and 12 psu respectively. After rearing for 30 days, the specific growth rate and survival rate were compared between the two groups, and salinity 6 psu was used to test the salinity tolerance. The results were as follows: (a) Both the specific growth rate and survival rate were significant lower in the experimental group (p < 0.05) after 30 days; (b) After challenge with salinity 6 psu for 72 hr, the crabs of experimental group had a 100% survival rate, whereas the crabs of the control group were all dead within 48 hr; (c) The content of GABA and the gene expression level of GABARAP in experimental group were significant different from control group (p < 0.05) after challenge via salinity 6 psu. In the control group, the GABA content increased rapidly from 9.96 ± 2.09 to 42.00 ± 5.94 µg/g; however, in the experimental group, it only increased to 27.82 ± 2.55 µg/g; the gene expression of GABARAP in the experimental group increased to the maximum at 24 hr, then decreased and stabilized at 48 hr, suggesting that GABA and GABARAP were trigged during the early stage of low‐salinity stress resistance.  相似文献

The purpose of the present study was to investigate the effects of salinity stress on immune responses and evaluating indicators in swimming crab Portunus trituberculatus. The crabs (150 ± 8.5 g in body weight) were exposed to different salinities as 21, 26 and 31‰ (control) for 6 days. The results showed the total haemocyte counts (THC) and prophenoloxidase (proPO) activity in the haemocytes decreased significantly in the treatment groups after 6 hr, and reached the lowest levels at 12 hr. The phenoloxidase (PO) activity in the plasma increased significantly and peaked at 12 hr, then recovered to control level after 24 hr. The phagocytic per cent of haemocyte, antibacterial and bacteriolytic activities in the plasma decreased significantly in the treatment groups, and reached the lowest level at 12 hr, then recovered to control level after 72 hr. The dopamine (DA) and 5‐hydroxytryptamine (5‐HT) contents in the plasma increased significantly and peaked at 12 hr, then recovered to control level, while the noradrenaline (NE) content in the plasma had no significant change throughout the duration of the experiment. The DA and 5‐HT receptors were significantly up‐regulated in the treatment groups. The highest value of mRNA expression of DA and 5‐HT receptors occurred at 12 hr and recovered to control level after 24 hr. In addition, the cAMP and protein kinase A (PKA) contents in the haemocytes increased significantly and peaked at 12 hr, then recovered to control level after 72 hr. The phospholipase C (PLC) and protein kinase C (PKC) contents in the haemocytes increased significantly and peaked at 12 hr, then resumed to control level after 24 hr. These results speculated that biogenic amine (DA and 5‐HT) is likely to play an important role in immune modulation via cAMP/PKA signalling pathway or PLC/PKC signalling pathway when P. trituberculatus is exposed to low salinity and these results will provide scientific data for immune evaluation.  相似文献

Despite recent advances in culture technology for East Asian common octopus Octopus sinensis paralarvae using upwelling systems, securing suitable feed for the paralarvae is an unresolved issue. The zoea of the swimming crab Portunus trituberculatus is a good candidate for paralarval feed because of the high fecundity of the adult females. To investigate the effects of supplying P. trituberculatus zoeae and their feeding method on paralarvae, we cultured paralarvae with supplying different combination ratios of zoeae and Artemia (10:0, 7:3, 5:5, 3:7 and 0:10), and with or without supplementing rotifers using small‐scale (3‐L) upwelling systems. Paralarval survival rate and growth were improved when zoeae were supplied as the main feed, but reduced when the proportion of Artemia exceeded half the whole preys. Supplementing rotifers did not affect the paralarval survival and growth. Subsequently, paralarvae were cultured by supplying zoeae (partially augmented by Artemia) using three large (1‐kl) upwelling systems to assess their feeding effectiveness in juvenile octopus production. Paralarvae could be cultured at high survival rates of 77.1 ± 5.1% to reach benthic juveniles at 23 days after hatching. In conclusion, supplying P. trituberculatus zoeae augmented with Artemia under an upwelling culture system has great potential for juvenile octopus production.  相似文献

Insulin‐like growth factor (IGF) plays a key role in the complex system that regulates bony fish growth, differentiation, and reproduction. In the current study, recombinant tongue sole IGF‐1 and IGF‐2 were obtained using the Pichia pastoris expression system and their comparative bioactivities were investigated. Tricine–SDS–PAGE and western blot analysis showed that the recombinant tongue sole IGFs were secreted into the culture medium and had a molecular weight of 8.7 kDa. The optimal incubation time and pH for recombinant expression of IGFs were 36 hr and 5.0 respectively. Functional analysis demonstrated that both recombinant tongue sole IGF‐1 and IGF‐2 significantly promoted cell proliferation of MFC‐7 in vitro. In addition, the recombinant tongue sole IGF‐1 and IGF‐2 proteins could suppress hepatic mRNA levels of igf‐1 and igf‐2 in vitro, which showed that they have similar physiological functions. Taken together, the biologically active recombinant tongue sole IGF‐I and IGF‐II proteins will allow us to further investigate their physiological roles in growth regulation of this species. Furthermore, the present results also hinted at the potential application of these two recombinant IGF‐I and IGF‐II proteins into the tongue sole farming industry.  相似文献

A 60‐day feeding trial was conducted to determine the growth performance and expression of insulin‐like growth factor 1 gene (IGF‐I gene) in Labeo rohita fingerlings fed with either raw, soaked or fermented sweet potato leaf meal (SPLM) by completely replacing de‐oiled rice bran (DORB), following a completely randomized design. Seven isonitrogenous (30%) and isocaloric (1.8 MJ/100 g) diets were prepared by replacing DORB with 50% and 100% raw, soaked and fermented sweet potato leaf meal, maintaining DORB‐containing diets as a control. Weight gain %, SGR (specific growth rate) and PER (protein efficiency ratio) were significantly (p < 0.05) higher when 100% DORB was replaced by fermented SPLM in comparison to other treatment groups. The fermented and soaked SPLM‐fed groups had registered with lower FCR value. The expression of growth regulating gene IGF‐I mRNA and RNA/DNA ratio was found to be significantly higher (p < 0.05) in soaked and fermented SPLM‐fed groups. In this study, the body protein and lipid composition did not vary significantly (p > 0.05). Hence, the study concludes that the fermented sweet potato leaf meal using Chaetomium globosum can replace 100% DORB in the diet of Labeo rohita without any detrimental effect on growth performance.  相似文献

The histiophagous scuticociliate Philasterides dicentrarchi is the aetiological agent of scuticociliatosis, a parasitic disease of farmed turbot. Curcumin, a polyphenol from Curcuma longa (turmeric), is known to have antioxidant and anti‐inflammatory properties. We investigated the in vitro effects of curcumin on the growth of P. dicentrarchi and on the production of pro‐inflammatory cytokines in turbot leucocytes activated by parasite cysteine proteases. At 100 μm , curcumin had a cytotoxic effect and completely inhibited the growth of the parasite. At 50 μm , curcumin inhibited the protease activity of the parasite and expression of genes encoding two virulence‐associated proteases: leishmanolysin‐like peptidase and cathepsin L‐like. At concentrations between 25 and 50 μm , curcumin inhibited the expression of S‐adenosyl‐L‐homocysteine hydrolase, an enzyme involved in the biosynthesis of the amino acids methionine and cysteine. At 100 μm , curcumin inhibited the expression of the cytokines tumour necrosis factor‐alpha (TNF‐α) and interleukin‐1 beta (IL‐1β) produced in turbot leucocytes activated by parasite proteases. Results show that curcumin has a dual effect on scuticociliatosis: an antiparasitic effect on the catabolism and anabolism of ciliate proteins, and an anti‐inflammatory effect that inhibits the production of proinflammatory cytokines in the host. The present findings suggest the potential usefulness of this polyphenol in treating scuticociliatosis.  相似文献

A sixty‐day feeding trial was conducted to determine the ascorbic acid (AA) requirement for growth of striped catfish, Pangasianodon hypophthalmus juveniles. Seven iso‐nitrogenous and iso‐energetic (370 g protein per kg and 19.6 MJ/kg) purified diets were prepared with different levels of ascorbic acid such as control (0), T1 (17.5), T2 (35), T3 (70), T4 (175), T5 (350) and T6 (700) mg ascorbic acid (L‐ascorbyl‐2‐polyphosphate) equivalent per kg diet. Fish with a mean body weight of 3.2–3.4 g were stocked (fifteen fish per tank) in triplicates following a completely randomized design. Each group was fed to satiation twice a day for 60 days. Significant differences were observed in growth, survival, body composition and metabolic enzymes activities with different dietary ascorbic acid levels. Maximum weight gain, specific growth rate (SGR) and protein efficiency ratio (PER) were found in fishes fed with 35 mg AA per kg diet, supported by best feed conversion. Fish fed a diet containing vitamin C had the highest activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) compared to those fed with vitamin C‐depleted diets. In this study, based on using broken‐line regression analysis, the dietary vitamin C requirement for growth of P. hypophthalmus juveniles was estimated to be in the range of 46–76 mg AA per kg, depending on the criterion used, growth and liver storage. Our results will be helpful for the formulation of cost‐effective ascorbic acid incorporated diets for striped catfish, P. hypophthalmus.  相似文献

An 8‐week study was conducted to evaluate the effects of different feeding patterns with dietary Clostridium butyricum supplementation on growth performance, antioxidant and non‐specific immune responses in freshwater prawn, Macrobrachium rosenbergii (0.39 ± 0.001 g). There were four feeding methodologies: feeding basal diet continuously (P1); feeding diet with 500 mg/kg C. butyricum continuously (P2); feeding diet with 500 mg/kg C. butyricum 5 days after 2 days of basal diet (P3) and feeding diet with 500 mg/kg C. butyricum 2 days after 5 days of basal diet (P4). The results revealed that prawns in P3 had the highest weight gain rate (WGR) and lowest feed conversion ratio (FCR) than the other groups. Haemolymph total protein levels and superoxide dismutase activity increased significantly in P2, P3 and P4 groups, while malondialdehyde content and anti‐superoxide anion levels decreased significantly compared to control. The mRNA expression of intestinal dorsal and Toll in P2 and P3 groups decreased significantly compared to control. Prawns in P3 exhibited improved growth performance, increased antioxidant capacity and enhanced immune function. We concluded that feeding diet with 500 mg/kg C. butyricum for 5 days after 2 days of basal diet was recommended for M. rosenbergii.  相似文献

A female swimming crab (Portunus trituberculatus) with fully developed ovaries is considered one of the most nutritious types of seafood. The fishing yield of P. trituberculatus in China has always been high; however, its economic value has been low and unreliable due to poor ovarian development and low nutritional value. Artificial fattening is an effective way to improve the value of wild‐caught crabs, and this study focused on analysing the ovarian development and nutritional quality of the female P. trituberculatus during an artificial fattening period. Therefore, this study provides scientific evidence on the effects of artificial fattening on female P. trituberculatus and can guide the development of nutritional quality regulations for wild‐caught female crabs during the fattening process. The results showed that the ovaries of the wild female P. trituberculatus developed gradually. The ovarian morphology became bulkier, while the gonadosomatic index (GSI) and total edible yield (TEY) were significantly improved after artificial fattening. Significant changes in the nutritional components of the edible tissues were observed, and the moisture content in the ovaries and hepatopancreas decreased continually, while both the protein and fat content in the ovaries significantly increased. Moreover, the fat content in the hepatopancreas significantly increased. The saturated fatty acids(ΣSFA) in the ovaries and hepatopancreas had an increasing trend after fattening, while ΣPUFA and Σn‐3PUFA first decreased and then increased. In contrast, the ratio of Σn‐3PUFA to Σn‐6PUFA decreased steadily. However, the fatty acid composition of the muscles did not change significantly. The levels of all the amino acids increased continuously during the fattening process, and EAA increased significantly in the early stages of the fattening period. Consequently, all amino acids became nonlimiting amino acids after fattening. Furthermore, the ratio of EAA/NEAA in the ovaries increased continually until the end of the fattening period. On the other hand, the fatness and TEY increased significantly after artificial fattening. In addition, the nutritional value significantly improved with the exception of the high‐quality fatty acids in the ovaries and hepatopancreas, which were slightly lower in both.  相似文献

The aim of this study was to evaluate the effects of dietary lipids on protein‐sparing and lipoprotein lipase (LPL) mRNA expression in culture using 360 juvenile soft‐shelled turtles (Pelodiscussinensis) (initial weight 4.26 ± 0.14 g). The turtles were allotted to six diets with three duplicates for 60 days. A control diet with 46% protein and 55% fishmeal (CD) and five isonitrogenous diets with 41.3% protein and 45% fishmeal (F, S, L1, L2 and L3) were used, containing the following three lipid types: fish oil, soybean oil and mixed oils (soybean oil: fish oil = 1:1). The results showed that the survival rate was not affected by dietary lipids (P > 0.05). The highest weight gain and lowest feed coefficient ratio were seen in the L3 diets (P < 0.05). Turtles fed with L2 and L3 diets had lower superoxide dismutase activities, higher alanine aminotransferase activities and higher cholesterol concentrations than those exposed to other diets (P < 0.05). Hepatic LPL activity and LPL mRNA expression were higher in the L3 diets than in the other diets (P < 0.05). Overall, there were obvious protein‐sparing effects of dietary lipids and LPL mRNA expression was stimulated by high dietary lipids in soft‐shelled turtles in this study.  相似文献

An 8‐week feeding trial was conducted with extruded practical diets containing 320 g/kg of crude protein from plant‐based sources to determine the effects of L‐lysine on growth performances, muscle‐growth‐related gene (myoD, myogenine and myostatin) expression and haemato‐biochemical responses in juvenile genetically improved farmed tilapia (GIFT). Five isonitrogenous and isoenergetic diets were formulated to contain graded levels of lysine (14.3, 16, 17.5, 19 and 20.5 g/kg of diet). Each diet was randomly assigned to triplicate groups of 30 juvenile fish (5.2 ± 0.1 g), which were fed thrice a day (9:00, 13:00 and 17:00 hr). Maximum growth performances were observed in fish fed with lysine at 19 g/kg of diet. There was no significant (p > .05) effect on whole‐body composition and amino acids profile by dietary lysine supplementation. Significant (p < .05) changes were observed in relative expression of muscle‐growth‐related genes namely myoD and myogenine and in plasma metabolites by dietary lysine supplementation. In contrast, the relative expression of myostatin was not affected by dietary lysine supplementation. Broken‐line regression analysis and second‐order polynomial regression analysis of weight gain and N gain against dietary lysine levels showed that the dietary lysine requirement for juvenile GIFT tilapia was 18–18.3 g/kg of diet and 19.3–19.5 g/kg of diet, respectively.  相似文献

A 60‐day feeding trial was conducted to optimize the inclusion level of de‐oiled rice bran (DORB) in the diet of Labeo rohita fingerlings. Six iso‐nitrogenous (260g/kg), iso‐lipidic (70g/kg) and iso‐energetic (15 MJ/Kg) diets were prepared with different inclusion level of DORB viz., T33, T38, T43, T48, T53 and T58. One hundred and eighty fingerlings (9.8 ± 0.5 g) were randomly distributed in seven experimental groups in triplicates following a completely randomized design. The weight gain %, specific growth rate, protein efficiency ratio and feed efficiency ratio were found similar in 33%, 38% and 43% DORB fed groups. The expression of both insulin‐like growth factors I and II (IGF‐I and II) was significantly (p < 0.05) higher in T33, T38 and T43 groups. Based on second order polynomial regression analysis (y = ?0.0059x2 + 0.4573x ? 4.8404, R2 = 0.95), the expression of IGF‐I was found to be maximum at 39.5% inclusion level of DORB. Significantly (p < 0.05) lower muscle RNA content, RNA‐DNA ratio and higher feed conversion ratio were found in T58 group. Hence, from these results, it can be concluded that the optimum inclusion of DORB in the diet (260g/kg CP and 70g/kg lipid) of L. rohita should be 39.5% to support maximum nutrient utilization and growth performance.  相似文献

A previous proteomic study examining the plasma acute‐phase response of rainbow trout to sterile inflammation highlighted an unidentified 9.5‐kDa spot using 2D‐PAGE, which was dramatically increased. The 15 amino acid sequence obtained from this protein spot allowed rapid amplification of cDNA ends PCR to generate a 443‐bp nucleotide sequence that was 98.6% similar to type‐4 ice‐structuring protein LS‐12 from Atlantic salmon Salmo salar Linnaeus. Quantitative reverse translation PCR and an ELISA were used to measure gene expression and plasma concentrations of LS‐12 following experimental intraperitoneal injection of rainbow trout with either 106 or 108 colony‐forming units (CFU) of Flavobacterium psychrophilum. There was no significant change in the plasma concentration of LS‐12 up to 15 days post‐infection in any group. Hepatic LS‐12 gene expression was significantly reduced at 3 and 6 days (p < 0.001) post‐infection in fish injected with 108 CFU of F. psychrophilum relative to control fish, while branchial or head kidney expression was unchanged. Infected fish had significantly increased hepatic gene expression of serum amyloid A, confirming an acute‐phase response. Under the conditions used, LS‐12 is not a positive acute‐phase protein in rainbow trout.  相似文献

In recent years, more and more attentions have been paid to the development and application of probiotics in aquaculture, and viable probiotics have been extensively studied, while rare information was available about inactivated probiotics in aquaculture. Therefore, in this study, a feeding trial was designed to investigate the effect of dietary supplementation of heat‐inactivated probiotic Bacillus clausii DE5 on growth performance, immune response and key immune genes expression in head kidney and intestine in grouper Epinephelus coioides. Fish were fed for 60 days with control diet (C) and two experimental diets containing 1.0 × 108 CFU/g live (T1) and heat‐inactivated (T2) B. clausii DE5, respectively. The probiotic treatments did not affect the final weight, weight gain (WG) and specific growth rate (SGR) of E. coioides at days 30 and 60 (> .05), while both heat‐inactivated and live B. clausii DE5 significantly decreased the feed intake and feed conversion ratio (FCR) at day 60 (< .05). Serum lysozyme activity and complement C3 level in the two probiotic treatments were significantly higher than those in the control (< .05). The lysozyme activity and complement C3 level at day 60 were significantly higher than those at day 30 (< .05), while no significant interaction effect between diet and administration date was observed. Moreover, the heat‐inactivated B. clausii DE5 significantly improved the expression of TLR5, pro‐inflammatory cytokines (IL‐8 and IL‐1β) and TGF‐β1 in head kidney and intestine (< .05), while the live probiotic did not show any significant effect on the expression of these key immune‐related genes in head kidney and intestine. These results indicate that dietary supplementation of heat‐inactivated B. clausii DE5 effectively improved feed utilization and both the local and systemic immune responses of E. coioides.  相似文献

An experiment was designed to evaluate the requirement of dietary protein for Reba carp, Cirrhinus reba fingerlings using semi‐purified diet. Five isocaloric diets were prepared containing 20%, 25%, 30%, 35% and 40% level of protein using casein, gelatin as a major protein source. The experiment was conducted for 105 days in triplicates to evaluate the growth, survival, feed utilization, carcass composition of C. reba. Mean weight gain was highest (p < .05) in T3. Specific growth rate was highest in T3 and lowest in T1. Similarly, significantly (p < .05) highest net fish yield were recorded in T3. No significant difference was recorded in survivability. Feed conversion ratio, protein efficiency ratio, feed conversion efficiency and protein conversion efficiency were significantly (p < .05) higher for T3 compared with other treatments. Highest carcass protein and lipid values were recorded in T3. So, conclusion can be drawn that 30% crude protein is optimum for overall performance. Furthermore, broken‐line analysis for specific growth rate, final biomass and net fish yield indicated that dietary protein level of 28.148 to 28.253% is optimum for the best growth performance of fingerling C. reba.  相似文献

A 9‐week study was conducted to evaluate the potential of processed animal protein (PAP) in comparison with soybean meal (SM) to replace fish meal (FM) in practical diets for European catfish, Silurus glanis, on growth performance, liver transaminase activities and expression of growth‐related genes. Seven isonitrogenous (440 g kg‐1 crude protein) and isoenergetic (16.70 MJ/kg) practical diets were formulated by replacing 0 (control), 30%, 60% and 100% FM with either SM or PAP. Each diet was randomly assigned to triplicate groups of 25 fish per cages fixed in the pond. Fish fed the diet substituted 100% FM from SM or PAP had lower (p < .05) growth performance, feed efficiency and protein retention compared to control and other groups. The decreased growth performance occurred concomitant with a decline in the expression level of liver GHR/IGF‐I axis genes. The gene expression and activity of liver protein metabolism enzymes also positively correlated with growth performance. Findings of this study indicated that both SM and PAP were equally effective in replacing FM up to 60% in the practical diet of Silurus glanis. Total replacement of FM either with SM or PAP induced negative influences on growth and feed utilization.  相似文献
