In order to investigate the functions of the gene PsG6PDH and the mechanisms underlying freezing tolerance of Populus suaveolens, the recombinant expression vector pET-G (pET30a-G6PDH), which contained full encoding region of PsG6PDH gene, was established. The recombinant was identified by lawn-PCR and double enzyme digestion and then transformed into expression host XA90 and induced by isopropyl-â-D-thiogalactoside (IPTG) to express 100 kD polypeptide of G6PDH fusion protein. The results showed that the expressed amount of the fusion protein culminated after 1 mmol•L–1 IPTG treatment for 4 h and that pET-G product was predominately soluble and not extra-cellular secreting.  相似文献

Optimizing SSR-PCR system of Panax ginseng by orthogonal design   总被引:1,自引:0,他引:1  
An orthogonal design was used to optimize SSR-PCR amplification system using Panax ginseng genomic DNA as template. Four levels of five factors (DNA template, Taq DNA polymerase, Mg^2+, primer, and dNTP) and annealing temperature have been tested separately in this system. The results demonstrated the reaction efficiency was affected by these factors. Based on the results, a stable, productive and reproducible PCR system and cycling program for amplifying a ginseng SSR locus were obtained: 20 μL system containing 1.0 U Taq DNA polymerase, 2.0 mmol·L^-1 Mg^2+, 0.2 mmol·L^-1 dNTPs, 0.3 μmol·L^-1 SSR primer, 60 ng· μla^-1 DNA template, performed with a program of 94℃ for 5 min, 94℃ for 30 s, annealing at 56.3℃ for 30 s, 72℃ for 1 min, 37 cycles, finishing at 72℃ for 7 min, and storing at 4℃.  相似文献

The effects of Timentin and cefotaxime (Cef) on shoot regeneration of the London plane tree (Platanus acerifolia Willd.) and their use for the suppression of Agrobacterium tumefaciens in Agrobacterium-mediated genetic transformation were compared. Shoot regeneration was significantly reduced on the media with Cef at concentrations from 100 to 500 mg·L–1.  Spatial distribution of sporocarps of Suillus pictus A.H. Smith and Thiers was studied in a plot of 6 × 12 m in size established in a stand of Pinus koraiensis Sieb. et Zucc. in Kyoto, Japan for 4 years, and the biomass of mycorrhizas was examined in the last year. S. pictus was dominant in both sporocarp and ectomycorrhizal community in the study plot. The number of S. pictus sporocarps ranged from 0.94 to 1.26 m−2 (surface area) in the study plot and did not vary very much during the study period. Sporocarps of S. pictus occurred in clumps and the distributions of clumps were generally random. As the spots of sporocarp occurrence changed gradually from year to year, the distributions of sporocarps that occurred successively in 2-year periods overlapped, especially when analyzed in 9-m2 unit size using the m * –m method. Mycorrhizas of S. pictus were distributed in more subplots than its sporocarps. Distribution of mycorrhizas and sporocarps of S. pictus generally overlapped well. The biomass of mycorrhizas and mycelia in the mycorrhizas of S. pictus was estimated at 15.5 g DW m−2 and 6.2 g DW m−2 (surface area) in this plot, respectively. The biomass of mycorrhizas and mycelia in the mycorrhizas supporting the production of one sporocarp (average dry weight was 0.86 g) of S. pictus was evaluated as about 16.4 and 7.3 g DW, respectively, in this plot. The cutting seedlings ofLiriodendron chinense xtulipifera were treated with the different concentrations of auxin (treatment1: IBA of 50 g·kg−1+NAA of 300 g·kg−1; treatment2; IBA of 100 g·kg−1+NAA of 300 g·kg−1). The biomass nutrient element contents for different organs (root, stem, leaf) of cutting seedling ofLiriodendron chinense xtulipifera were measured by the dry method, Kjeldahl method and Atomic Absorption Spectroscopy method. The result showed that the biomass of root, stem, and leaf of the cutting seedling treated with auxin was all remarkably increased. The contents of element C in root, stem and leaf had no significant difference between the control and auxin treatments, while the contents of N, P, K and Ca in stem were much lower than that in leaf and root. Variance analysis showed that for the same organ with different concentration treatment of auxin, the four nutrient elements (N, P, K, and Ca) had no significant difference in contents, while there existed significant or very significant difference in contents of the four nutrient elements in different organs with the same concentration auxin treatment. The N, P, K and Ca contents were very low in cutting seedlings; as a result, additional fertilizer should be applied to the seedlings when they were planted in the field. Lignins are cell wall phenolic heteropolymers that result from the oxidative coupling of three monolignols bearing p-coumaryl (H), coniferyl (G), and sinapyl (S) units, in a reaction mediated by peroxidases. Here, we report the existence of a relationship between the Σβ-O-4/ΣO-4 end monomer ratio of the linear lignin fraction, released through the specific cleavage of the alkyl ether linkages by thioacidolysis, and the G/S ratio of lignins, when this was estimated in differentially evolved vascular land plants. Most importantly, in the case of angiosperms, Gnetales, and lycopods, the Σβ-O-4/ΣO-4 end monomer ratio was apparently predictable from the proportions at which the G and S units were mixed. In the case of G lignins (present in basal gymnosperms and ferns), the Σβ-O-4/ΣO-4 end monomer ratio decayed exponentially to increase the O-4-linked dihydroconiferyl alcohol (DHCA) content. The results obtained suggest that the Σβ-O-4/ΣO-4 end monomer ratio of the linear lignin fraction depends intimately on the lignin monomer composition, and, therefore, on the chemical nature of the radicals derived from three monolignols (coniferyl, dihydroconiferyl, and sinapyl alcohols), whose gain have been finely tuned during land plant evolution.  相似文献

UGPase gene related with wood cellulose synthesis was transferred into C. acuminata using the method of Agrobacte- rium-mediated genetic transformation, and an efficient transformation system was developed for C. acuminata on the basis of evaluations of several factors affecting Agrobacterium-mediated DNA transfer rate. The highest transformation rate was achieved when pre-cultttred leaf explants were infected with an Agrobacterium culture corresponding to OD600 (0.5) for 10 min, and cultured on explant regeneration medium for three days. The results of Southern hybridization showed that genomic DNA of the kanamycin-resistant shoots to an UGPase gene probe substantiated the integration of the transgene. Transformation efficiency (6%) was achieved under the optimized transformation procedure, This system should facilitate the introduction of important useful genes into C, acuminata.  相似文献

Embryo of lilacs (Syringa L) culture in vitro and the rapid propagation were studied. The orthogonal experiments, including the selection of basal medium, embryo age and other factors such as sugar, benzyladenine (BA), naphthalene acetic acid (NAA) and glutamine (Gln), were carried out. The results indicated that the optimal medium for embryo culture was Monnier medium supplemented with NAA (0.001 mg.L^-1), BA (0.1 mg.L^-1), sugar (50 g.L^-1), and Gin (400 mg.L^-1), with a germination rate of 91.7% at least; the optimal embryo age was 50 d; and Gln had significant effects on the germination rate of embryo.Moreover, the optimal medium for subculture was MS BA (2 mg.L^-1) NAA (0.001 mg.L^-1) Gln (0.5 mg.L^-1), with the propagation coefficient of 3.6 at least.  相似文献

Twigs of 2–3-year-old Platanus occidentalis L. were used as experimental material to find the causes for the contamination and browning in the initial stages of tissue cultures. To compare the degree of browning of explants picked off from different growing seasons, the experimental material was excised from trees on each of the first ten days in January, March, May and July, 2006. The results indicated that the contamination and browning rates of the material cut off in January (14.2% and 30.6%, respectively) and March were somewhat lower than those in July. The pretreatment of soaking the explants in different anti-oxidants and absorbents at the same time could diminish some side effects. The pretreatment of using 10 g·L–1 vitamin C reduced the contamination and brown-ing rate effectively. An orthogonal experiment showed that the optimal factor and level arrangement is 0.5 mg·L–1 BA, 2.0 g·L–1 ac-tive carbon and 1.5 g·L–1 PVP which resulted in a browning rate of only 16.5%. In general, sampling period, physical properties and pretreatment of explants are the main factors responsible for the contamination and browning of material in the initial stages of P. occidentalis tissue cultures.  相似文献

Cotton bollworm, Helicoverpa armigera Hubner selected for five generations with Bacillus thuringiensis insecticidal crystal protein Cry1Ac in the laboratory developed 12.98-fold resistance. Resistance and susceptible populations were mass crossed to study the dominance of resistance. The Cry1Ac—selected (BCR) population showed 5.8-fold resistance to Cry1Aa and 5.04-fold resistance to Cry1Ab. The degree of dominance (D) was 0.34 and 0.40 for the R × S and S × R hybrids, respectively, which indicates incomplete recessive character of Cry1Ac resistance in the population. The estimated realized heritability (h 2) and response quotient (Q) of resistance for Cry1Ac were 0.52 and 0.15, respectively. This indicated the lower phenotypic variation in the selected population. The resistance risk assessment based on h 2 indicated that the resistance would increased tenfold after <9 generations for Cry1Ac in the resistant population. The results show the ability of H. armigera to develop resistance against Cry1Ac and cross-resistance to Cry1Aa and Cry1Ab.  相似文献

Plant growth, morphology and nutritive value under shade can differ between temperate grasses. Therefore, the aim of this study was to quantify the dry matter (DM) production, sward morphology, crude protein (CP%), organic matter digestibility (OMD) and macro-nutrient concentrations (P, K, Mg, Ca and S) in a grazed cocksfoot (Dactylis glomerata L.) pasture under 10-year-old Pinus radiata D. Don forest. Four levels of light intensity were compared: full sunlight (100% photosynthetic photon flux density-PPFD), open + wooden slats (∼43% PPFD), trees (∼58% PPFD) and tree + slats (∼24% PPFD). The mean total DM production was 8.2 t DM ha−1 yr−1 in the open and 3.8 t DM ha−1 yr−1 in the trees + slats treatment. The changes in cocksfoot leaf area index (LAI) were related to variations in morphological aspects of the sward such as canopy height and tiller population. CP% increased as PPFD declined with mean values of 18.6% in open and 22.5% in the trees + slats treatment. In contrast, the intensity of fluctuating shade had little effect on OMD with a mean value of 79 ± 3.2%. The mean annual macro-nutrient concentrations in leaves increased as the PPFD level declined mainly between the open and the trees + slats treatments. It therefore appears that heavily shaded dominant temperate pastures in silvopastoral systems limit animal production per hectare through lower DM production rates and per animal through reduced pre-grazing pasture mass of lower bulk density from the etiolated pasture.  相似文献

To compare the responses to repeated flooding and drought of Salix gracilistyla, which grows on coarse gravel substrates, and Salix subfragilis, which grows on fine silt or clay substrates, we measured pre-dawn leaf water potential (Ψw pd), osmotic adjustment (Ψw tlp, Ψo sat), and biomass production of cuttings under greenhouse conditions. The experimental design involved a control and four treatments that crossed 1 or 3-week flooding (F) with 1 or 2-week droughts (D). Ψw pd was reduced after 2 weeks of drought when preceded by 1 week of flooding. Neither species increased osmotic adjustment in response to increased duration of drought between repeated 3-week flooding. Moreover, a decrease in the ratio of leaf biomass to total biomass or an increase in the ratio of root biomass to total biomass with longer drought repetitions was not observed for either species. The root ratio of S. gracilistyla was more strongly inhibited by flooding than that of S. subfragilis. The shoot-to-root ratio of S. subfragilis was higher than that of S. gracilistyla in all F combinations. The hypertrophied lenticel ratio of S. gracilistyla after 1 week of flooding was nearly the same as that after 3 weeks of flooding, whereas values for S. subfragilis after 1 week of flooding were lower than those after 3 weeks of flooding. The low allocation to roots and the generation of hypertrophied lenticels by S. gracilistyla in response to flooding, as compared with S. subfragilis, seem to be related to the different habitat substrate conditions of the two species.  相似文献

In alley cropping systems, fast growing leguminous trees are pruned to reduce competition with crops for light and to provide organic inputs for crop nutrition. Tree regrowth depends on non-structural carbohydrate reserves in the remaining tree parts. In this study, the dynamics of starch and soluble carbohydrates in roots and stems of completely pruned (all shoots removed), partially pruned (one branch retained on the pruned stump) and unpruned Erythrina poeppigiana (Walp.) O.F. Cook and Gliricidia sepium (Jacq.) Kunth ex Walp. trees were studied under humid tropical conditions in Turrialba, Costa Rica. Measurements on starch and soluble carbohydrates in roots and stems were made at 0, 2, 6 and 12 weeks after pruning during both a “rainy” and a “dry” season. In general, the dynamics of non-structural carbohydrates in roots and stems of pruned E. poeppigiana and G. sepium trees were similar. Starch concentration was highest in unpruned trees and higher in roots than in stems of pruned trees. The effect of pruning intensity was first observed in stems, and starch reserves were more depleted in stems than in roots, an effect more evident during the “dry” season. The critical tree regrowth stage for starch mobilisation was that of vigorous sprout development at six or four weeks after pruning particularly in completely pruned trees. At this time, fine root biomass and length and nodule biomass in pruned trees decreased. Survival of fine roots and nodules was greater in partially pruned than in completely pruned trees. Starch accumulation in roots recommenced at 12 weeks after pruning in G. sepium, and later than 12 weeks after pruning in E. poeppigiana roots. This study showed that E. poeppigiana responded better to pruning regimes than G. sepium. Recovery of trees after pruning is better when trees are partially pruned than when completely pruned.  相似文献

The bark ofPteroceltis tatarinowii is a raw material for manufacturing Xuan Paper. The effects of Ca2+ concentrations on the accumulation of mineral elements in the bark, leaf and root ofPteroceltis tatarinowii were studied under controlled conditions. The types of Hoagland nutrient solution with three Ca2+ concentrations levels (200, 400 and 600 μg·g−1) and a control (without Ca2+ were designed to culturePteroceltis tatarinowii. After 6 months, contents of seven mineral elements including Ca, K, Mg, Mn, Zn, Cu and Na in the root, leaf and bark were analyzed. The results indicated that Ca accumulations content in the root, leaf and bark had positively relation with Ca2+ concentrations (200, 400, 600 μg·g−1), and the order of the Ca content in the three components was root>leaf>bark. Ca content in the root treated with 600 μg·g−1 Ca2+ concentrations was 5.5 times as high as that of the control, and about 1.4 times as high as that of the root treated in 200 and 400 μg/g Ca2+ concentrations respectively. On the contrary, K and Mg contents in the root, leaf and bark were negatively related to Ca2+ concentrations, especially in the bark, and their accumulation trend followed the order of leaf>root>bark. K content in the bark treated with 600 μg·g−1 Ca2+ concentrations was 39.3% of that of the control, and was 79.0% and 91.8% of that of the bark treated with 200μg·g−1 and 400μg·g−1 Ca2+ concentrations respectively; Mg content in the bark treated with 600μg·g−1 Ca2+ concentrations was 23.4% of that of the control, and was 27.1% and 35.4% of that of the bark treated with 200 and 400 μg·g−1 Ca2+ concentrations respectively. Compared with the control, the general tendency of Mn, Zn and Cu content decreased with increasing of Ca2+ concentrations and their contents were in the order: root>leaf>bark. To clarify the biosynthetic pathway for syringyl lignans, especially syringyl tetrahydronaphthalene lignans and formation of the C2–C7′ linkage, production of (+)-lyoniresinol (LYR) and its predicted intermediates [syringaresinol (SYR), 5,5′-dimethoxylariciresinol (DMLR), and 5,5′-dimethoxysecoisolariciresinol (DMSLR)] in Lyonia ovalifolia var. elliptica was investigated by means of feeding experiments with radiolabeled precursors. Following individual administration of l-[U-14C]phenylalanine (Phe), [8-14C]sinapyl alcohol (SA), and [8,8′-14C]SYR to excised young shoots of L. ovalifolia and their subsequent metabolism, free [14C]lignans and [14C]lignan glycosides were extracted with methanol from stems and leaves and were divided into ethyl acetate-soluble fractions (lignans) and aqueous fractions (lignan glycosides), respectively. Using a combination of xylanase, cellulase, and β-glucosidase, the glycosides were hydrolyzed to liberate [14C]lignans as aglycones. l-[U-14C]Phe was incorporated into (+)-[14C]SYR [stem 0.38%, 8% enantiomeric excess (e.e.)], (−)-[14C]SYR (leaves 2.75%, 72% e.e.), (+)-[14C]DMLR (stem 0.07%, 18% e.e. and leaves 0.009%, 58% e.e.), (−)-[14C]DMSLR (stem 0.03%, 46% e.e. and leaves 0.05%, 20% e.e.), (+)-[14C]LYR (leaves 0.013%, 22% e.e.) and glycosides of (+)-[14C]LYR (stem 0.036%, 50% e.e.) in 24h. Based on the percent incorporation and enantiomeric composition of the lignans, the biosynthetic pathway of (8R,8′R)-(+)-LYR was proposed as follows: a nonselective dehydrogenative dimerization of sinapyl alcohol yields (±)-SYR, which is reduced with low specificity to give (8R,8′R)-(+)-DMLR. This is cyclized to directly give (+)-LYR as well as reduced again to (8R,8′R)-(−)-DMSLR. Although further transformation of (−)-DMSLR also leads to the formation of (+)-LYR, cyclization could be a main pathway for (+)-LYR biosynthesis. A study was conducted to determine the fitting soil moisture for the normal growth of two-year-old W. sinensis (Sims) Sweets by using gas exchange technique. Remarkable threshold values of net photosynthetic rate (Pn), transpiration rate (Tr) and water use efficiency (WUE) were observed in the W. sinensis leaves treated by various soil moisture and photosynthetic available radiation (PAR). The fitting soil moisture for maintaining a high level of Pn and WUE was in range of 15.3%-26.5% of volumetric water content (VWC), of which the optimal VWC was 23.3%. Under the condition of fitting soil moisture, the light saturation point of leaves occurred at above 800μmol.m^2.s^-1, whereas under the condition of water deficiency (VWC, 11.9% and 8.2%) or oversaturation (VWC, 26.5%), the light saturation point was below 400μmol.m^-1.s^-1. Moreover, the light response curves suggested that a special point of PAR occurred with the increase in PAR. This special point was considered as the turning point that indicated the functional transition from stomatal limitation to non-stomatal limitation. The turning point was about 600, 1000, 1000 and 400 μmol.m^-2.s^-1, respectively, at VWC of 28.4%, 15.3%, 11.9% and 8.2%. In conclusion, W. sinensis had higher adaptive ability to water stress by regulating itself physiological function.  相似文献

Rhyzopertha dominica (Fabricius) has developed extensive pesticide resistance in the last several decades. We have developed a supercritical fluid extraction method for Trigonella foenum-graecum L. (TFG) and studied the contact toxicities of the extracts to R. dominica. The extraction method was designed with orthogonal experiments to preserve and collect all the possible active components. Contact toxicity and efficiency of extraction were used as standard values to optimize extraction conditions, which were achieved at 55°C under 25 Mpa of pressure. The extraction efficiency for 200 g of dry sample reached 6.21% with 30 ml of 95% alcohol. Extracts loaded on filter paper showed dose and time dependent toxicities to adult R. dominica with a LC50 value of 65.02 μg/cm2 after 3 days post treatment. Our extensive in vivo studies indicated the extracts from Trigonella foenum-graecum seeds have high efficacy against pesticide resistant R. dominica. The active ingredient(s) from the extract shows promise as a novel pesticide candidate.  相似文献

Experiments were conducted to study plant regeneration through direct somatic embryogenesis using mature zygotic embryo and cotyledonary explants from seeds of Melia volkensii stored for <3 and >12 months. Explants were cultured on Murashige and Skoog (MS) medium supplemented with BAP, NAA and 2,4-D (0.5, 1.0 and 2.0 mg l−1) alone, and BAP (0.5, 1.0, 2.0 and 4.0 mg l−1) in combination with 2,4-D or NAA (0.2 and 0.5 mg l−1). After 4 weeks in culture, up to 60% of cotyledonary explants from the seeds stored for <3 months produced direct somatic embryos on BAP (0.5–4.0 mg l−1) in combination with 2,4-D (0.2 mg l−1). The number of somatic embryos ranged from 5 to 14 per explant in BAP (0.5 mg l−1) and 2,4-D (0.2 mg l−1) combination. Only 20% of cotyledonary explants from seeds stored for >12 months produced somatic embryos. Mature zygotic embryos failed to produce any somatic embryos. Subcultures of somatic embryos from cotyledonary explants of seeds stored for <3 months formed clusters of shootlets on semi solid MS and 1/2 MS media. After 6 weeks of subculture on multiplication MS media augmented with BAP (0.5 mg l−1) and IAA (0.2 mg l−1), 70% of the shoot tips formed 4–7 shoots per explant. Up to 33% of the multiplied shoots were rooted in MS medium supplemented with 2.0 mg l−1 IBA. Plantlets developed normally into seedlings in the greenhouse.  相似文献

 Two novel stilbenoids, laevifonol (an ε-viniferin-ascorbic acid hybrid compound) and laevifoside (an O-glucoside of ampelopsin A) were isolated from the heartwood of Shorea laeviforia. Their structures were elucidated by nuclear magnetic resonance spectroscopic evidence including HMQC, HMBC, and NOESY. The novel compounds and other known stilbenoids from S. laeviforia were evaluated for inhibitory activity against rat liver 5α-reductase. Positive inhibitory activities were observed in resveratrol dimers and tetramers. No inhibitory activity was detected in laevifonol and ampelopsin A glucosides, laevifoside, or hemsleyanoside, whereas inhibitory activity was seen in their aglycon. These results suggest that the hydrophilic moiety in these compounds may inhibit action with the hydrophobic active site of the enzyme. Effects of side chain hydroxyl groups on pyrolytic β-ether cleavage of phenolic model dimers were studied with various deoxygenated dimers under pyrolysis conditions of N2/400°C/1 min. Although phenolic dimer with hydroxyl groups at the C α and C γ positions was much more reactive than the corresponding nonphenolic type, deoxygenation at the C γ -position substantially reduced the reactivity up to the level of the nonphenolic type. These results are discussed with the cleavage mechanism via quinone methide intermediate formation, which is activated through intramolecular hydrogen bonds between C α and C γ hydroxyl groups.  相似文献
