CASE HISTORY: A skeletal disease characterised by lameness, limb deformities and reduced growth rate occurred over two successive years in lambs born on a commercial sheep farm in Marlborough. A genetic aetiology was considered likely following exclusion of other known causes of rickets and because of the progressive nature of the disease, even after affected animals were transferred to another property. CLINICAL FINDINGS: Affected lambs appeared normal at birth but developed clinical signs during the first 2 months of life. The most severely affected animals either died or were euthanised within the first year of life, but some survived to breeding age. Serum biochemistry revealed hypocalcaemia, hypophosphataemia and increased concentrations of 1,25 dihydroxyvitamin D. The mean serum 25 hydroxyvitamin D concentration was similar to that of control lambs. PATHOLOGICAL FINDINGS: Gross lesions included enlarged costochondral junctions, bilateral irregularity of articular surfaces on humeral heads due to collapse of subchondral bone, thickened cortices in long bones and irregular thickening of physeal cartilages. Microscopically, tongues of hypertrophic chondrocytes extended from physes into metaphyseal regions; metaphyseal trabeculae were thick, disorganised and often lined by wide osteoid seams. Osteoclastic activity was excessive both in cortical and trabecular bone. DIAGNOSIS: Inherited rickets in Corriedale sheep. CLINICAL RELEVANCE AND CONCLUSIONS: This disease is likely to be present in several Corriedale sheep flocks in New Zealand and may have been misdiagnosed as arthritis or other diseases causing lameness and/or poor growth. A defect in end-organ responsiveness to 1,25 dihydroxyvitamin D is the likely mechanism. This disease of sheep may be a useful model for studying vitamin D metabolism and the treatment of inherited forms of rickets in human beings.  相似文献

OBJECTIVE: To investigate possible genetic influences on susceptibility or resistance of sheep to Johne's disease. DESIGN: A field and laboratory study of two fine-wool Merino flocks with a high prevalence of disease due to Mycobacterium avium subsp paratuberculosis infection. PROCEDURE: Adult sheep were phenotypically classified as having severe, mild or no disease on the basis of clinical, pathological and cultural tests for paratuberculosis, and as positive or negative in tests for humoral immunity (agar gel immunodiffusion test) or cell mediated immunity (skin test for delayed type hypersensitivity). Correlations with phenotype were sought for polymorphisms at loci within selected immune function genes (NRAMP, MHC complex, IFN-gamma, lysozyme, leukaemia inhibiting factor). RESULTS: Possible associations of particular NRAMP and MHC alleles with susceptibility or resistance to Johne's disease were detected. CONCLUSION: If the results of this preliminary study are confirmed in further work, then the use of rams with "resistant" genotypes may assist in the control of Johne's disease in infected flocks.  相似文献

The canine gastric mucosa is known to be a habitat for various Helicobacter species. So far, five Helicobacter species have been described from the canine gastric mucosa, but histological studies have demonstrated a greater variety. In order to gain more information on diversity of canine gastric mucosa colonising helicobacters, biopsy samples of four pet dogs were examined by DNA-based techniques. PCR with a primer pair binding specifically to the 16S rDNA of the species of the genus Helicobacter and generating a fragment of approximately 400 bp indicated the presence of Helicobacter strains in the stomachs of the four dogs. PCR products were cloned into Escherichia coli DH10B and PCR-re-amplified 16S rDNA fragments were subjected to amplified ribosomal DNA restriction analysis (ARDRA) employing restriction enzyme HhaI. Restriction profiles indicated the presence of at least two different Helicobacter species in two dogs. Partial sequences of 16S rDNA of six clones were compared with sequences available in the EMBL data bank. Two sequences obtained from different dogs were identical with the corresponding sequences of Helicobacter pylori strains. Three sequences showed highest but moderate similarity values to H. pylori (96.6-98.0%) and one sequence to Helicobacter salomonis (97.3%). In contrast to previous reports our data implicate that the gastric mucosa of dogs may be colonised by strains of H. pylori or a very closely related species but they also confirm indications for the presence of so far uncultivated species of Helicobacter.  相似文献

Sulfur influences on rumen microorganisms in vitro and in sheep and calves   总被引:2,自引:0,他引:2  
When continuously cultured ruminal microbes were given orchardgrass hay and sufficient sulfuric acid or hydrochloric acid to maintain a pH of 5.5, fermentation and numbers of protozoa were reduced compared with cultures whose pH was controlled with phosphoric acid. Likewise, when sulfur-deficient, purified diets were supplied to cultures, less methane (mmol X liter-1 X d-1), 3.2 vs 32.6, was produced and fewer cellulolytic bacteria (log10/ml), 5.8 vs 7.2 were present than when cultures were given the same diet supplemented with .3% elemental sulfur. The rumen of sheep fed the .04% sulfur diet had reduced digesta weights (1.69 vs 3.2 kg) compared with sheep fed the diet with .34% sulfur at the same intake. There also was reduced methanogenesis 12.3 vs 25.8 mmol X liter-1 X d-1) and reduced numbers of cellulolytic bacteria (7.4 vs 8.4 log10/ml) in sulfur-deficient sheep in comparison to sulfur-supplemented sheep. In growing calves, the same types of bacteria predominated in the rumen, but more facultative anaerobic bacteria were isolated from calves fed .04% sulfur than from calves fed diets with .34 to 1.72% sulfur. None of the dietary levels of sulfur appeared toxic. Regardless of treatment, volatile fatty acids were more predominant than lactic acid as end-products of fermentation of ruminal microbes in fermenters, sheep and calves. The greater methanogenesis and the greater cellulolytic bacterial numbers of sulfur-supplemented sheep compared with sulfur-deficient in vitro cultures, is interpreted to be the result of recycling of sulfur to the rumen in sheep where it is efficiently scavengered by ruminal bacteria.  相似文献

Literature relating to the effects of the tapeworm Moniezia expansa on sheep is reviewed. While there are many claims of adverse effects by this organism, none of these withstand critical scrutiny. The balance of evidence indicates that M. expansa infections are generally harmless, even when the tapeworms are present in large numbers in young lambs, and that there is no justification for treating sheep for M. expansa on the basis of any likely benefit to the health or production of the animals. No published experimental results can be found to support claims that heavy M. expansa infections sometimes exacerbate losses from other causes. Anthelmintics for investigative use, and some aspects of the classification, life history and biology of M. expansa and of its intermediate host are also discussed.  相似文献

Estimates of genetic parameters for growth traits in Kermani sheep   总被引:3,自引:0,他引:3  
Birth weight (BW), weaning weight (WW), 6-month weight (W6), 9-month weight (W9) and yearling weight (YW) of Kermani lambs were used to estimate genetic parameters. The data were collected from Shahrbabak Sheep Breeding Research Station in Iran during the period of 1993-1998. The fixed effects in the model were lambing year, sex, type of birth and age of dam. Number of days between birth date and the date of obtaining measurement of each record was used as a covariate. Estimates of (co)variance components and genetic parameters were obtained by restricted maximum likelihood, using single and two-trait animal models. Based on the most appropriate fitted model, direct and maternal heritabilities of BW, WW, W6, W9 and YW were estimated to be 0.10 +/- 0.06 and 0.27 +/- 0.04, 0.22 +/- 0.09 and 0.19 +/- 0.05, 0.09 +/- 0.06 and 0.25 +/- 0.04, 0.13 +/- 0.08 and 0.18 +/- 0.05, and 0.14 +/- 0.08 and 0.14 +/- 0.06 respectively. Direct and maternal genetic correlations between the lamb weights varied between 0.66 and 0.99, and 0.11 and 0.99. The results showed that the maternal influence on lamb weights decreased with age at measurement. Ignoring maternal effects in the model caused overestimation of direct heritability. Maternal effects are significant sources of variation for growth traits and ignoring maternal effects in the model would cause inaccurate genetic evaluation of lambs.  相似文献

Ocular infections by chlamydiae are associated with ocular disease manifestations such as conjunctivitis and keratitis in humans and animals. Limited evidence exists that members of the order Chlamydiales can also cause ocular disease in sheep. In the current study, the prevalence of chlamydiae in the eyes of sheep was investigated by using PCR methods. Data obtained in sheep by broad-range 16S rRNA order Chlamydiales-specific PCR were compared to the prevalence of antibodies against chlamydiae detected by a competitive enzyme-linked immunosorbent assay (cELISA). Flocks tested included a clinically healthy flock and two flocks suffering from ocular disease and with histories of Ovine Enzootic Abortion (OEA). PCR detected DNA of Chlamydophila (Cp.) abortus and Cp. pecorum in the eyes of both healthy and sick animals but also identified Chlamydia (C.) suis and a variety of uncultured chlamydia-like organisms. Good correlation was found between the presence of Cp. abortus DNA in sheep conjunctival samples and seropositivity detected by cELISA. Despite these findings, no association was found between the presence of chlamydial DNA in the sheep conjunctival samples and the onset of clinical disease. These results suggest that the biodiversity of chlamydiae in the eyes of sheep is greater than that previously thought. Further investigations are needed to determine whether a causal relationship between infection by chlamydiae and ocular disease exists in these animals.  相似文献

The objective of this study was to estimate genetic parameters for reproductive traits in Shall sheep. Data included 1,316 records on reproductive performances of 395 Shall ewes from 41 sires and 136 dams which were collected from 2001 to 2007 in Shall breeding station in Qazvin province at the Northwest of Iran. Studied traits were litter size at birth (LSB), litter size at weaning (LSW), litter mean weight per lamb born (LMWLB), litter mean weight per lamb weaned (LMWLW), total litter weight at birth (TLWB), and total litter weight at weaning (TLWW). Test of significance to include fixed effects in the statistical model was performed using the general linear model procedure of SAS. The effects of lambing year and ewe age at lambing were significant (P?<?0.05). Genetic parameters were estimated using restricted maximum likelihood procedure, under repeatability animal models. Direct heritability estimates were 0.02, 0.01, 0.47, 0.40, 0.15, and 0.03 for LSB, LSW, LMWLB, LMWLW, TLWB, and TLWW, respectively, and corresponding repeatabilities were 0.02, 0.01, 0.73, 0.41, 0.27, and 0.03. Genetic correlation estimates between traits ranged from ?0.99 for LSW–LMWLW to 0.99 for LSB–TLWB, LSW–TLWB, and LSW–TLWW. Phenotypic correlations ranged from ?0.71 for LSB–LMWLW to 0.98 for LSB–TLWW and environmental correlations ranged from ?0.89 for LSB–LMWLW to 0.99 for LSB–TLWW. Results showed that the highest heritability estimates were for LMWLB and LMWLW suggesting that direct selection based on these traits could be effective. Also, strong positive genetic correlations of LMWLB and LMWLW with other traits may improve meat production efficiency in Shall sheep.  相似文献

This study reports on the phenotypic and genetic (co)variance components for reproductive traits in Zandi sheep, using between 1,859 and 2,588 records obtained from 577 ewes. The data were collected from the Khojir Breeding Station of Zandi sheep in Tehran, Iran from 1994 to 2008. The basic traits were litter size at birth (LSB), litter size at weaning (LSW), litter mean weight per lamb born (LMWLB), and litter mean weight per lamb weaned (LMWLW), and the composite traits were total litter weight at birth (TLWB) and total litter weight at weaning (TLWW). Genetic analyses were carried out using the restricted maximum likelihood method that was explored by fitting the additive direct genetic effects and permanent environmental effects of the ewes as random effects and the ewe age at lambing and lambing year as fixed effects for all of the investigated traits. Akaike’s information criterion was used to choose the most appropriate model. LSB, LSW, LMWLB, LMWLW, TLWB, and TLWW direct heritability estimates were 0.07, 0.05, 0.12, 0.10, 0.08, and 0.14, respectively. The estimated fractions of variance due to the permanent environmental effects of the ewe ranged from 0.03 for LMWLB to 0.08 for LMWLW and TLWW. Corresponding repeatability estimates ranged from 0.10 for LSW to 0.22 for TLWW. Direct genetic correlations varied from ?0.61 for LSB–LMWLB to 0.88 for LSB–LSW and LSB–TLWB. Results indicate that genetic change depends not only on the heritability of traits, but also on the observed phenotypic variation; therefore, improvement of non-genetic factors should be included in the breeding programs.  相似文献

Genetic parameters for carcass traits in lambs at weaning (average age of 128 days) measured by ultrasound (n = 1821) and computer tomography (CT) (n = 234), and response to selection for ultrasound eye muscle depth (UMD) and carcass LEAN weight, were estimated. The research flock comprised a meat line (ML) and a control line (CL) of Norwegian White Sheep. The ML was crossed with Texel from 1998, and selected for UMD from 1993 to 2001, and for LEAN weight from 2001 to 2004. For CT scanning, a mean of 23 images was taken per animal. Genetic parameters were estimated with univariate and bivariate mixed-animal models using AIREML, including all animals with records and their relatives. The statistical models included fixed effects, live weight or age of lamb at weaning (covariate), and a random genetic effect.

Heritability estimates for weight of LEAN, FAT and BONE were 0.57, 0.29 and 0.51 using model corrected for live weight. The heritability estimates were lower when these traits were adjusted for age. High genetic correlation was found between LEAN and UMD (0.70), and between carcass FAT and ultrasound fat depth (UFD) (0.82). The genetic trend for UMD regressed on year of birth was significantly greater for ML than CL in 2004.  相似文献
