In situ denitrification (DNT) and denitrification enzyme activity (DEA) were measured in a Mediterranean riparian forest soil during two periods under contrasting soil moisture conditions in order to investigate the factors that affect denitrification through the year. Results showed that in summer, soil moisture limited denitrification throughout the entire soil profile, whereas in winter, anaerobic conditions in the soil were more favourable for denitrifiers. The potential for denitrification was larger at shallow depths (<30 cm), and neither nitrate nor organic carbon limited denitrification significantly. Some denitrification was measured during winter at depths below 30 cm, suggesting that a reduction of groundwater nitrate could occur in some areas of this riparian forest during the wet period. In summer, low denitrification, together with high mineralization rates, brought about an increase of soil N, which could be leached to the stream channel during rainfall events. This study suggests that Mediterranean riparian soils act as sources or sinks of dissolved nitrogen depending on the period of the year.

Riparian buffers, located in the transition zone between terrestrial and aquatic ecosystems, are a hotspot for nitrogen (N) removal through denitrification. Earthworms are abundant in riparian buffers and may enhance denitrification. This study investigated earthworm demographics of three earthworm functional groups (anecic, epigeic, and endogeic) and denitrifier activity in temporarily flooded and non-flooded riparian soils from April to October 2012 in southern Quebec, Canada. Nine earthworm species, mostly endogeic, were found in the temporarily flooded soil, while only six earthworm species were found in the non-flooded soil. On average, there were 11.7 times more earthworms with 12.4 times greater biomass (P<0.05) found in the temporarily flooded soil than in the non-flooded soil. The denitrification enzyme activity (DEA) was of similar magnitude in temporarily flooded and non-flooded soils, with temporal variation associated with rainfall patterns. Endogeic earthworm biomass was positively correlated (P<0.05) with DEA, while epigeic earthworm biomass was positively correlated (P<0.05) with 16S rRNA gene copies and nosZ gene copies from bacteria, indicating an association between earthworm functional groups and denitrifier activity in riparian soils. Stepwise multiple regressions showed that DEA in riparian soils could be predicted using soil moisture, inorganic N concentration, and earthworm functional groups, suggesting that endogeic and epigeic earthworms contributed to denitrifier activity in riparian soils.

In the recent decades, most rivers and lakes in the Taihu Basin have experienced degradation from an excess of nutrients. The presence of the nitrogen in water contributes to the increase of eutrophication. The riparian zones are associated with these watercourses and can effectively reduce any excess nitrogen. Soil denitrification is the most significant process in the transfer of nitrogen, which migrates from the terrestrial to the aquatic ecosystem. The relationship between soil denitrification and soil characteristics is well documented. However, the degree of soil denitrification and the main impact factors during different processes within the riparian zones due to gradual changes in the surroundings are not well understood. The present study selected four types of riparian soils that are contained in three different watersheds. The soil denitrification potential was determined within these soils using the acetylene block technique. The results indicate that, among the local factors studied, the soil denitrification potential increased with the intensity of anthropogenic activities, which varied significantly within the basin. This variation indicated a trend in the soil denitrification potential: cropland?>?woodland?>?grassland?>?bareland. Results suggest that soil moisture, nitrate-nitrogen concentration, and microbial biomass carbon concentration are the dominant factors that influence the riparian soil denitrification potential in the Tiaoxi watershed, while soil organic matter is the major factor for soil denitrification potential in the Hexi watershed and nitrate-nitrogen concentration is the dominant factor in the Tianmuhu watershed.

The dynamics of biological denitrification in riparian soil is still poorly understood. We studied the spring‐time pattern of denitrifying enzyme activity (DEA) and the rate of denitrification (DNT) in two hydromorphic riparian soils, one a mollic Gleysol and the other a terric Histosol. The average DEA ranged from 73 to 1232 ng N g?1 hour?1, and DNT ranged from 4 to 36 ng N g?1 hour?1. Both DEA and DNT diminished with increasing depth in both soil types. This decrease corresponded to a decrease in total and K2SO4‐extractable organic carbon and K2SO4‐extractable mineral nitrogen. The DEA and DNT differed in their dynamics. The former had no evident pattern in subsurface horizons but increased with temperature at the end of spring in surface and structural horizons. The DNT diminished as the soil dried in the mollic Gleysol when the water table fell. In the terric Histosol, the water table was still too high at the end of spring to affect the DNT. The results suggest that the vertical pattern of denitrification is related to that of organic carbon content. This organic carbon content determines biological activity and the supply of carbon and nitrous oxides. In biologically active horizons temperature drives the dynamics of DEA, whereas soil moisture drives the dynamics of DNT. Our results show the importance of the dynamic soil–water relationship in controlling denitrification within the riparian zone.

Excess nitrate (NO3-) in lakes and streams has deleterious effects for environmental and human health. Nitrate concentrations have become problematic in agricultural watersheds due to increased use of fertilizers and improper management of livestock wastes. Research has indicated that the planting and/or preservation of riparian buffer zones can be an effective means of reducing pollution from agricultural fields (Osborne and Kovacic, 1993; Jordan et al., 1992; Simmons et al., 1992). Biological denitrification is the most desirable means of nitrate attenuation as the microbial conversion of NO3- removes nitrate from the watershed in the form of N gases. Despite the inherent value of biological denitrification, a comprehensive review discussing the role of this process in removing nitrate from riparian zones is lacking. In this paper we examine the results and conclusions of past research on the topic of denitrification in riparian zones and make recommendations for future research in this area. The need for subsurface denitrification assays in riparian zones is emphasized.

Plant effects on ecosystem processes are mediated through plant-microbial interactions belowground and soil enzyme assays are commonly used to directly relate microbial activity to ecosystem processes. Live plants influence microbial biomass and activity via differences in rhizosphere processes and detrital inputs. I utilized six grass species of varying litter chemistry in a factorial greenhouse experiment to evaluate the relative effect of live plants and detrital inputs on substrate-induced respiration (SIR, a measure of active microbial biomass), basal respiration, dissolved organic carbon (DOC), and the activities of β-glucosidase, β-glucosaminidase, and acid phosphatase. To minimize confounding variables, I used organic-free potting media, held soil moisture constant, and fertilized weekly. SIR and enzyme activities were 2-15 times greater in litter-addition than plant-addition treatments. Combining live plants with litter did not stimulate microbial biomass or activity above that in litter-only treatments, and β-glucosidase activity was significantly lower. Species-specific differences in litter N (%) and plant biomass were related to differences in β-glucosaminidase and acid phosphatase activity, respectively, but had no apparent effect on β-glucosidase, SIR, or basal respiration. DOC was negatively related to litter C:N, and positively related to plant biomass. Species identity and living plants were not as important as litter additions in stimulating microbial activity, suggesting that plant effects on soil enzymatic activity were driven primarily by detrital inputs, although the strength of litter effects may be moderated by the effect of growing plants.

From the global change perspective, increase of atmospheric CO2 and land cover transformation are among the major impacts caused by human activities. In this study, we are addressing the combined issues of the effect of CO2 concentration increase and plant type on soil microbial activities by asking how annual and perennial plant groups affect soil microbial processes under elevated CO2. The experimental design used a mix of species of different growth forms for both annuals and perennials. Our objective was: (1) to determine how two years of annual or perennial plant cover and CO2 enrichment could affect Mediterranean soil microbial processes; (2) to test the resistance and the resilience of these soil functional processes after a natural perturbation. We determined the effects of 2 years atmospheric CO2 enrichment on soil potential respiration (SIR), denitrification (DEA) and nitrification (NEA) activities. We could not find any significant effect of CO2 increase on SIR, DEA and NEA. However, we found a strong effect of the plant cover type, i.e. annuals versus perennials, on the potential microbial activity related to N cycling. DEA and NEA were significantly higher in soil under annual plants while SIR was not significantly different. To determine whether these changes would survive a natural perturbation, we carried out a rain event experiment once the experimental treatments (i.e. different plant cover and atmospheric CO2 concentration) were stopped. The soil potential respiration, as expressed by the SIR, was not affected and remained stable. DEA rates converged rapidly under annuals and perennials after the rain event. Under both annuals and perennials NEA increased significantly after the rain event but remained significantly higher in the soil with annual plants. The relative change of the soil microbial processes induced by annual and perennial plants was inversely related to the density and the diversity of the corresponding microbial functional groups.

Ecosystem and biogeochemical responses to anthropogenic stressors are the result of complex interactions between plants and microbes. A mechanistic understanding of how plant traits influence microbial processes is needed in order to predict the ecosystem-level effects of natural or anthropogenic change. This is particularly true in wetland ecosystems, where plants alter the availability of both electron donors (e.g., organic carbon) and electron acceptors (e.g., oxygen and ferric iron), thereby regulating the total amount of anaerobic respiration and the production of methane, a highly potent greenhouse gas. In this study, we examined how plant traits associated with plant inputs of carbon (photosynthesis and biomass) and oxygen (root porosity and ferric iron on roots) to mineral soils relate to microbial competition for organic carbon and, ultimately, methane production. Plant productivity was positively correlated with microbial respiration and negatively correlated to methane production. Root porosity was relatively constant across plant species, but belowground biomass, total biomass, and the concentration of oxidized (ferric) iron on roots varied significantly between species. As a result the size of the total root oxidized iron pool varied considerably across plant species, scaling with plant productivity. Large pools of oxidized iron were related to high CO2:CH4 ratios during microbial respiration, indicating that as plant productivity and biomass increased, microbes used non-methanogenic respiration pathways, most likely including the reduction of iron oxides. Taken together these results suggest that increased oxygen input from plants with greater biomass can offset any potential stimulation of methanogenic microbes from additional carbon inputs. Because the species composition of plant communities influences both electron donor and acceptor availability in wetland soils, changes in plant species as a consequence of anthropogenic disturbance have the potential to trigger profound effects on microbial processes, including changes in anaerobic decomposition rates and the proportion of mineralized carbon emitted as the greenhouse gas methane.

Global nitrogen cycling is being altered by anthropogenic disturbances including invasion by non-native species. European and Asian earthworms have invaded northern temperate forests in North America with dramatic consequences for litter thickness, forest floor plant diversity, and soil nitrogen cycling. Invasive earthworms present at the boundary of terrestrial and aquatic ecosystems (i.e., riparian zones) may alter the flux of nitrogen into adjacent aquatic ecosystems. We examined how nitrogen cycling in riparian soil responds to amendments of invasive earthworms or artificial earthworm burrows. In earthworm-free riparian plots (0.25 m2), we established treatments of invasive earthworms (60 g fresh mass·m−2), artificial burrows (120 m−2), or control plots and sampled the plots after 30 days. Before and after treatment application we measured major soil characteristics (water-filled pore space, organic matter, and pH), nitrogen pools (exchangeable NH4+ and NO3), and nitrogen transformation rates (net N-mineralization, net nitrification, and denitrification). Exchangeable NH4+ and NO3 changed through time but did not differ among treatments. Net N-mineralization and net nitrification rates did not change through time and were similar across all treatments. However, denitrification rates in plots with added earthworms were 4 times greater than rates in control and burrow-only plots, which represents a large rapid increase in gaseous nitrogen flux out of these riparian soils. For all response variables, artificial burrows responded similarly to control plots, suggesting that earthworm biological activity (i.e., feeding, excretion, and mucus production) rather than physical effects (i.e., burrowing and soil aeration) drove the changes in nitrogen cycling. Examination of soil nitrogen pool and flux measurements suggest that this increase in denitrification was coupled with NH4+ consumption by nitrifying bacteria, but future studies are needed to confirm this hypothesis. We conclude that the activity of invasive earthworms in riparian zones can increase the flux of N out of riparian zones, but the hydrologic context of the riparian zone (e.g., pore-water residence time) ultimately controls whether denitrification or nitrate leaching is the dominant flux of N.

Effects of vegetation and nutrient availability on potentail denitrification rates were studied in two volcanic, alluvial-terrace soils in lowland Costa Rica that differ greatly in weathering stage and thus in availability of P and base cations. Potential denitrification rates were significantly higher in plots where vegetation had been left undisturbed than in plots where all vegetation had been removed continuously, and were higher on the less fertile of the two soils. The potential denitrification rates were correlated strongly with respiration rates, levels of mineralizable N, microbial biomass, and moisture content, and moderately well with concentrations of extractable NH inf4 sup+ , Kjeldahl N, and total C. In all plots, denitrification rates were stimulated by the removal of O2 and by the addition of glucose but not by the addition of water or NO inf3 sup- .This is Paper 2772 of the Forest Research Laboratory, Oregon State University

Riparian zones are important features of the landscape that can buffer waterways from non-point sources of nitrogen pollution. Studies of perennial streams have identified denitrification as one of the dominant mechanisms by which this can occur. This study aimed to assess nitrate removal within the riparian zone of an ephemeral stream and characterise the processes responsible, particularly denitrification, using both in-situ and laboratory techniques. To quantify rates of groundwater nitrate removal and denitrification in-situ, nitrate was added to two separate injection-capture well networks in a perched riparian aquifer of a low order ephemeral stream in South East Queensland, Australia. Both networks also received bromide as a conservative tracer and one received acetylene to inhibit the last step of denitrification. An average of 77 ± 2% and 98 ± 1% of the added nitrate was removed within a distance of 40 cm from the injection wells (networks with acetylene and without, respectively). Based on rates of N2O production in the network with added acetylene, denitrification was not a major mechanism of nitrate loss, accounting for only 3% of removal. Reduction of nitrate to ammonium was also not a major pathway in either network, contributing <4%. Relatively high concentrations of oxygen in the aquifer following recent filling by stream water may have reduced the importance of these two anaerobic pathways. Alternatively, denitrification may have been underestimated using the in-situ acetylene block technique. In the laboratory, soils taken from two depths at each well network were incubated with four nitrate-N treatments (ranging from ambient concentration to an addition of 15 mg N l−1), with and without added acetylene. Potential rates of denitrification, N2O production and N2O:N2 ratios increased with nitrate additions, particularly in shallow soils. Potential rates of denitrification observed in the laboratory were equivalent in magnitude to nitrate removal measured in the field (mean 0.26 ± 0.12 mg N kg of dry soil−1 d−1), but were two orders of magnitude greater than denitrification measured in the field with added acetylene. The relative importance of assimilatory vs. dissimilatory processes of nitrate removal depends on environmental conditions in the aquifer, particularly hydrology and its effects on dissolved oxygen concentrations. Depending on seasonal conditions, aquifers of ephemeral streams like the study site are likely to fluctuate between oxic and anoxic conditions; nevertheless they may still function as effective buffers. While denitrification to N2 is a desirable outcome from a management perspective, assimilation into biomass can provide a rapid sink for nitrate, thus helping to reduce short-term delivery of nitrate downstream. Longer-term studies are needed to determine the overall effectiveness of riparian buffers associated with ephemeral streams in mitigating nitrate loads reaching downstream ecosystems.

Limitations to the respiratory activity of heterotrophic soil microorganisms exert important controls of CO2 efflux from soils. In the northeastern US, ecosystem nutrient status varies across the landscape and changes with forest succession following disturbance, likely impacting soil microbial processes regulating the transformation and emission of carbon (C). We tested whether nitrogen (N) or phosphorus (P) limit the mineralization of soil organic C (SOC) or that of added C sources in the Oe horizon of successional and mature northern hardwood forests in three locations in central New Hampshire, USA. Added N reduced mineralization of C from SOC and from added leaf litter and cellulose. Added P did not affect mineralization from SOC; however, it did enhance mineralization of litter- and cellulose- C in organic horizons from all forest locations. Added N increased microbial biomass N and K2SO4-extractable DON pools, but added P had no effect. Microbial biomass C increased with litter addition but did not respond to either nutrient. The direction of responses to added nutrients was consistent among sites and between forest ages. We conclude that in these organic horizons limitation by N promotes mineralization of C from SOC, whereas limitation by P constrains mineralization of C from new organic inputs. We also suggest that N suppresses respiration in these organic horizons either by relieving the N limitation of microbial biomass synthesis, or by slowing turnover of C through the microbial pool; concurrent measures of microbial growth and turnover are needed to resolve this question.

Purpose

Denitrification has been extensively studied in soils from temperate zones in industrialized countries. However, few studies quantifying denitrification rates in soils from tropical and subtropical zones have been reported. Denitrification mechanisms in tropical/subtropical soils may be different from other soils due to their unique soil characteristics. The identification of denitrification in the area is crucial to understand the role of denitrification in the global nitrogen (N) cycle in terrestrial ecosystems and in the interaction between global environmental changes and ecosystem responses.

Materials and methods

We review the existing literature on microbially mediated denitrification in tropical/subtropical soils, attempting to provide a better understanding about and new research directions for denitrification in these regions.

Results and discussion

Tropical and subtropical soils might be characterized by generally lower denitrification capacity than temperate soils, with greater variability due to land use and management practices varying temporally and spatially. Factors that influence soil water content and the nature and rate of carbon (C) and N turnover are the landscape-scale and field-scale controls of denitrification. High redox potential in the field, which is mainly attributed to soil oxide enrichment, may be at least one critical edaphic variable responsible for slow denitrification rates in the humid tropical and subtropical soils. However, soil pH is not responsible for these slow denitrification rates. Organic C mineralization is more important than total N content and C/N in determining denitrification capacity in humid subtropical soils. There is increasing evidence that the ecological consequence of denitrification in tropical and subtropical soils may be different from that of temperate zones. Contribution of denitrification in tropical and subtropical regions to the global climate warming should be considered comprehensively since it could affect other greenhouse gases, such as methane (CH4) and carbon dioxide (CO2), and N deposition.

Conclusions

Tropical/subtropical soils have developed several N conservation strategies to prevent N losses via denitrification from the ecosystems. However, the mechanisms involved in the biogeochemical regulation of tropical and subtropical ecosystem responses to environmental changes are largely unknown. These works are important for accurately modeling denitrification and all other simultaneously operating N transformations.

The denitrifying enzyme activity (DEA), denitrification potential (DP) and anaerobic respiration (RESP) together with chemical characteristics were measured in three contrasting soils collected from experimental arable plots that had been subjected to long-term (21–23 years) fertilizer treatments. The plots sampled were either unfertilized or had received either annual inorganic NPK, manure and lime, or inorganic NPK and manure treatments. Addition of inorganic NPK, manure and lime led to large increases in the DEA for two of the three soils, but in the absence of lime, inorganic NPK and manure caused only small increases in DEA compared to unfertilized soils. Both DP and RESP were increased by the addition of inorganic NPK, manure and lime, but were substantially decreased by fertilizer treatments without lime. In most cases there was a simple relationship between soil pH and either DEA and DP, with those treatments that reduced soil pH also leading to reduced denitrification and vice versa. The effects of artificially increasing the pH to a value close to the pH in unfertilized soils (6.3) by addition of NaOH to the soils that had received inorganic NPK, and which had the lowest soil pH values, were to increase substantially DEA, DP and RESP. In soil from one of the sites that had been stored for 5 weeks, the DP values responded differently between the fertilizer treatments. The DP value was lowest in the soil that had inorganic NPK and manure, higher in the soil that received inorganic NPK, manure and lime and it was the highest in unfertilized (control) soil. The soil pH values for these treatments were 4.47, 5.79 and 6.58, respectively. However, when the soil pHs were adjusted by addition of either H2SO4 or NaOH to give a range between pH 2 and 12, the DP values from all three fertilizer treatments showed almost identical responses. The optimum pH value for DP was between 7 and 8 for all three fertilizer treatments. Substrate-induced respiration values from all fertilizer treatments showed a similar trend to DP when the soil pHs were modified. The results show that soil pH was an important factor which in the studied soils controls the microbial community in general and the community of denitrifiers in particular. However, denitrifiers showed a high pH resilience leading to no marked change of the pH optimum for potential denitrification.

In land-based wastewater treatment systems (LTS), denitrification is an important nitrogen removal process. We investigated the factors limiting the denitrifying population in a forested LTS, by studying the individual and combined effects of soil aeration, water content, nitrate and carbon on denitrification enzyme activity (DEA). The size of the soil denitrifying population in the LTS appeared to be limited by soil aeration, and limiting oxygen availability increased the denitrifying population above that observed in the field. Furthermore, we found that wastewater irrigation altered the short-term response of denitrifiers to anaerobic soil conditions. Under low oxygen conditions, denitrifiers in the wastewater-irrigated soils produced enzymes sooner and at a greater rate than soils without a history of wastewater irrigation. We propose that the size of the denitrifying population cannot be expected to be large in free-draining, coarsely textured soils even when provided with additional nitrogen and water inputs. Nitrogen (N) and carbon (C) mineralisation are triggered by pulses of water availability in arid and semi-arid systems. Intermittent streams and their associated riparian communities are obvious ‘hot spots’ for biogeochemical processes in arid landscapes where water and often C are limiting. Stream landscapes are characterized by highly heterogeneous soils that may respond variably to rewetting. We used a laboratory incubation to quantify how N and C mineralisation in rewetted soils and sediments from an intermittent stream in the semi-arid Pilbara region of north-west Australia varied with saturation level and substrate addition (as ground Eucalyptus litter). Full (100%) saturation was defined as the maximum gravimetric moisture content (%) achieved in free-draining soils and sediments after rewetting, with 50% saturation defined as half this value. We estimated rates and amounts of N mineralised from changes in inorganic N and microbial respiration as CO2 efflux throughout the incubation. In soils and sediments subject to 50% saturation, >90% of N mineralised accumulated within the first 7 d of incubation, compared to only 48% when soils were fully saturated (100% saturation). Mineralisation rates and microbial respiration were similar in riparian and floodplain soils, and channel sediments. N mineralisation rates in litter-amended soils and sediments (0.73 mg N kg−1 d−1) were only one-third that of unamended samples (3.04 mg N kg−1 d−1), while cumulative microbial respiration was doubled in litter-amended soils, suggesting N was more rapidly immobilized. Landscape position was less important in controlling microbial activity than soil saturation when water-filled pore space (% WFPS) was greater than 40%. Our results suggest that large pulses of water availability resulting in full soil saturation cause a slower release of mineralisation products, compared to small pulse events that stimulate a rapid cycle of C and N mineralisation-immobilization.

The aim of this study was to investigate temporal and spatial patterns of denitrification enzyme activity (DEA) and nitrous oxide (N2O) fluxes in three adjacent riparian sites (mixed vegetation, forest and grass). The highest DEA was found in the surface (0–30 cm) soil and varied between 0.7±0.1 mg N kg–1 day–1 at 5°C and 5.9±0.4 mg N kg–1 day–1 at 15°C. There was no significant difference (P >0.05) between the DEA in the uppermost (0–30 cm and 60–90 cm) soil depths under different vegetation covers. In the two deepest (120–150 cm and 180–210 cm) soil depths the DEA varied between 0.0±0.0 mg N kg–1 day–1 at 5°C and 4.4±0.9 mg N kg–1 day–1 at 15°C and was clearly associated with the accumulation of buried organic carbon (OC). Two threshold values of OC were observed before DEA started to increase significantly, namely 5 and 25 g OC kg–1 soil at 10–15°C and 5°C, respectively. In the three riparian sites N2O fluxes varied between a net N2O uptake of –0.6±0.4 mg N2O-N m–2 day–1 and a net N2O emission of 2.5±0.3 mg N2O-N m–2 day–1. The observed N2O emission did not lead to an important pollution swapping (from water pollution to greenhouse gas emission). Especially in the mixed vegetation and forest riparian site highest N2O fluxes were observed upslope of the riparian site. The N2O fluxes showed no clear temporal trend.
