Immunologic factors related to survival and performance in neonatal swine   总被引:1,自引:0,他引:1  
Logistic regression was used to develop models predicting preweaning survival in 334 neonatal swine. Measured risk factors included birth weight, litter size (live born), dam parity, serum IgG concentration, serum ELISA titers recognizing common gram-negative core antigens, and serum concentrations of the third component of complement. Larger birth weights were associated with increased probability of preweaning survival. The highest mortality was observed in litters with more than 12 pigs. Pigs with serum concentration of the third component of complement (C3) in the lowest stratum, less than 20% adult pooled C3 standard (APC3), had reduced mortality, compared with high (greater than 38% APC3) and middle (20 to 38% APC3) groups. Associations between all other variables, including total serum IgG concentration and preweaning survival were not significant. Few pigs had hypogammaglobulinemia, less than 3% of the study population had serum IgG concentrations less than 1 g/dl. Of all measured variables, only birth weight and dam parity were significant predictors of preweaning gain. Larger pigs and pigs born to third or greater parity dams had more preweaning gain than other pigs.  相似文献

Serologic recognition of common lipopolysaccharide core antigens has been related to enhanced resistance to gram-negative bacterial disease in several species. Class-specific titers (IgG, IgM) were determined by direct ELISA, using intact Escherichia coli (J5) as a plate antigen. Serum samples were obtained from 224 neonatal swine between the ages of 36 and 60 hours. The mean (+/- SEM) log10 IgG titer against gram-negative core antigens was 1:1,713 +/- 0.4718 and the mean log10 IgM titer was 1:202 +/- 0.5644. The IgG titer was directly related with litter size, birth weight, and serum total IgG concentration; IgM titer was directly related with dam parity and serum total IgG concentration.  相似文献

Background

Intake of colostrum after birth is essential to stimulate intestinal growth and function, and to provide systemic immunological protection via absorption of Immunoglobulin G (IgG). The birth order and weight of 745 piglets (from 75 litters) were recorded during a one-week period of farrowing. Only pigs weighing greater than 0.68 kg birth weight were chosen for the trial. Sow colostrum was collected during parturition, and piglets were bled between 48 and 72 hours post-birth. Piglet serum IgG and colostral IgG concentrations were determined by radial immunodiffusion.

Results

Sow parity had a significant (P < 0.001) effect on sow colostral IgG concentration, being 5% higher in multiparous females. Sow colostral IgG concentration explained 6% and piglet birth order accounted for another 4% of the variation observed in piglet serum IgG concentration (P < 0.05); however, birth weight had no detectable effect. Piglet serum IgG concentration had both a linear (P < 0.05) and quadratic effect (P < 0.05) on % survival. Piglets with 1,000 mg/dl serum IgG or less (n=24) had a 67% survival; whereas, piglets with IgG concentrations between 2250 to 2500 mg/dl (n=247) had a 91% survival. Birth order had no detectable effect on survival, but birth weight had a positive linear effect (P < 0.05). Piglets weighing 0.9 kg (n = 107) at birth had a 68% survival rate, and those weighing 1.6 kg (n = 158) had an 89% survival.

Conclusion

We found that the combination of sow colostrum IgG concentration and birth order can account for 10% of the variation of piglet serum IgG concentration and that piglets with less than 1,000 mg/dl IgG serum concentration and weight of 0.9 kg at birth had low survival rate when compared to their larger siblings. The effective management of colostrum uptake in neonatal piglets in the first 24 hrs post-birth may potentially improve survival from birth to weaning.  相似文献

Ontogeny of the third component of complement (C3) was monitored in 10 neonatal swine, using a radial immunodiffusion technique. Significant differences in mean serum C3 concentrations, expressed as percentage of C3 concentration in a pooled standard drawn from 15 adult swine, were not observed between serum samples collected before pigs suckled and at 2 days of age (P = 0.2583). Serum C3 concentrations increased significantly between 2 and 7 days of age (P less than 0.0001) and 7 and 14 days of age (P less than 0.0001). Concentrations comparable with those in adults were observed at 14 days of age and significant changes were not observed thereafter. Acquisition of adult concentrations of C3 appeared to be a function of endogenous production by the neonate, rather than by passive colostral transfer.  相似文献

New-born calves, artificially fed colostrum or native colostral whey, either dried or preserved by another method, had good health and good weight gains (between 0.05 and 0.60 kg). No greater differences were observed between the groups of calves given three times the colostrum of their mothers, calves given mixed colostrum, and calves fed colostral whey powder. In all groups only individual differences in IgG content in the blood serum were observed after 48 hours from birth. Hypogammaglobulinaemia occurred in individual cases both in calves given small amounts of colostrum or colostral whey and in calves given sufficient quantities. The time that had elapsed from birth to the first drinking did not exert any greater influence upon the IgG level in the blood; the decisive factor was the amount of colostrum taken in by the calf in the first dose. The rate of the absorption of IgG1, IgG2, IgA, and IgM from colostrum and the concentration of the immunoglobulins in the serum depended on the quantity of colostrum in the first dose and were not influenced to any greater degree by the amount of colostrum given to the calves in further doses. The amount of IgG in the blood serum of calves corresponded approximately to the level of colostral antibodies to the virus PI-3. The antibodies to the virus PI-3 and small quantities of IgG were observed also in the serum to new-born calves before drinking colostrum.  相似文献

The localization of immunoglobulins IgG, IgM and IgA in tissue sections prepared from the ileum of neonatal and adult swine were compared. Eighty percent of the immunoglobulin-containing lymphoid cells in the lamina propria of conventional adult German Landrasse swine were IgA-positive with lower numbers of IgM cells and occasionally an IgG cell. Anti-μ and α-chain reagents also stained the cytoplasm of the crypt epithelial cells. By comparison to these adult control tissues, the ileum of unsuckled neonates contained no immunoglobulins although after the ingestion of colostrum, the entire cytoplasm of the villus epithelial cells stained intensely when tested for IgG with only faint staining for IgM and IgA. On the other hand, IgA and IgM were readily localized on what appears to be only the apical border of the crypt epithelial cells but in contrast to the adult, the cytoplasm of these cells was unlabelled. IgG was absent from the crypt region. We interprete these findings to indicate an important, selective role for the villus epithelium in the absorption into the neonatal circulation of colostral IgG and probably IgA and IgM, and a specialized role for the crypt epithelium in adsorbing colostral IgA and IgM; possibly by complexing with mucin-bound secretory component.  相似文献

Background: Plasma transfusions have been used clinically in the management of neonates with failure of passive transfer. No studies have evaluated the effect of IV serum transfusions on serum IgG concentrations in dairy calves with inadequate transfer of passive immunity.
Hypothesis: A commercially available serum product will increase serum immunoglobulin concentration in calves with inadequate transfer of colostral immunoglobulins.
Animals: Thirty-two Jersey and Jersey-Holstein cross calves with inadequate colostral transfer of immunoglobulins (serum total protein <5.0 g/L).
Methods: Thirty-two calves were randomly assigned to either control (n = 15) or treated (n = 17) groups. Treated calves received 0.5 L of a pooled serum product IV. Serum IgG concentrations before and after serum transfusion were determined by radial immunodiffusion.
Results: Serum protein concentrations increased from time 0 to 72 hours in both control and transfused calves and the difference was significant between the control and treatment groups ( P < .001). Mean pre- and posttreatment serum IgG concentrations in control and transfused calves did not differ significantly. Median serum IgG concentrations decreased from 0 to 72 hours by 70 mg/dL in control calves and increased over the same time interval in transfused calves by 210 mg/dL. The difference was significant between groups ( P < .001). The percentage of calves that had failure of immunoglobulin transfer 72 hours after serum transfusion was 82.4%.
Conclusions and Clinical Importance: Serum administration at the dosage reported did not provide adequate serum IgG concentrations in neonatal calves with inadequate transfer of colostral immunoglobulins.  相似文献

OBJECTIVE: To quantify absorption of colostral IgG by healthy neonatal foals and to test the hypothesis that delayed ingestion of macromolecules prolongs the duration of intestinal permeability to immunoglobulins (Ig) in newborn foals. ANIMALS: Thirteen mixed breed foals. PROCEDURE: Foals were randomly assigned to two treatment groups, which were fed either a glucose-electrolyte solution or a commercial milk replacer for 12 h after birth, before being fed a known amount of colostral IgG. A control group was fed a known amount of colostral IgG from birth. The efficiency of IgG absorption was calculated following determination of plasma IgG concentration for each foal. RESULTS: Foals given colostrum immediately after birth transferred approximately 51% of ingested IgG into their vascular space. Delayed colostral ingestion significantly reduced the amount of IgG absorbed by foals. Withholding macromolecules for 12 h had no effect on the subsequent efficiency of IgG absorption. CONCLUSIONS: Colostrum should be supplied to foals within 12 h of birth for best uptake of Ig. The type of fluid administered to foals before the ingestion of colostrum does not influence subsequent absorption of Ig, suggesting that the process of gut closure in foals is not mediated by a finite capacity for macromolecular uptake.  相似文献

In a newly established closed specific pathogen-free (SPF) swine herd, gilt/sow suckling and weaned pig rotavirus specific antibody titers were followed for three lactations by enzyme-linked immunosorbent assay (ELISA) to gain insight into the dynamics of herd antibody titers to group A rotavirus. Among gilts/sows, serum antirotavirus IgG titers increased during each lactation with a subsequent drop in titer between farrowings. Serum antirotavirus IgM titers declined during each lactation and with subsequent parity. Serum antirotavirus IgA titers remained constant during lactations and among parities. In colostrum and milk, antirotavirus IgA antibody was abundant. Differences in titer were not noticed between gilts and second litter sows but third litter sows had significantly higher titers than the first two groups. Antirotavirus IgG was high in colostrum but nearly nonexistent in milk. This titer did not vary significantly within or among parities. There was a linear regression in the titers of baby pig serum antirotavirus IgG from the post colostral sample through to seven weeks old, after which titer began to increase. No difference in baby pig serum antirotavirus IgG was noted among the three litters. Serum antirotavirus IgA and IgM were undetectable in baby pig sera after 2-3 weeks of age. Coproantibody to rotavirus was sporadically present in pig feces for 2-3 weeks after birth with highest titers in the IgA fraction. We conclude that although it is probable that age resistance of pigs to rotavirus diarrhea occurs, humoral immunity as measured by ELISA rotavirus antibody titers may not be intimately involved in virus clearance since in our studies baby pigs passively received large amounts of antibody but still excreted pathogenic virus. The finding of increasing levels of serum antirotavirus IgG in gilt/sow serum suggest that exposure to antigen of dams occur without significant increases in antirotavirus IgG titers in either colostrum, milk, or baby pig serum.  相似文献

A total of 36 gilts were used to assess the effects of Cr tripicolinate supplementation on immune response in sows and their offspring during the periparturient and neonatal period. Gilts were raised from weaning to reproductive age on diets with either 0 (-Cr) or 200 (+Cr) ppb supplemental Cr from CrPic. Subsequently, 22 gilts (9 -Cr and 13 +Cr) in parity 1 and 16 sows in parity 2 (7 -Cr and 9 +Cr) underwent immune status testing. Only sows that completed all procedures in parity 1 were included in parity 2. Sows were immunized with ovalbumin about 3 wk (d 0), and again 14 d later for gilts, prior to anticipated farrowing, and serum was collected on d 0 and at 14-d intervals for a total of four samples. Serum was collected from five to six pigs/litter at 24 h after birth, three or six pigs/litter the day after weaning (25 d of age) in parity 1, and three pigs/litter the day of weaning (20 d of age) in parity 2. Milk was collected at 1 h (colostrum), 6.5 d (early), and 19 d (late) after farrowing. The only effect of Cr on total immunoglobulin (Ig) concentration was on sow serum IgG (21.7 and 24.1 mg/mL for -Cr and +Cr, respectively; P = 0.08) and IgM (11.0 and 12.5 mg/mL; P = 0.06) on d 0. No effect (P > 0.15) of Cr was observed on the IgG antibody response to ovalbumin, but Cr was associated (P < 0.10) with a decreased IgM antibody response to ovalbumin beginning on d 14. In parity 2, colostral total IgG increased (80.6 and 92.4 mg/mL for parity 1 and 2, respectively; P = 0.06), which was reflected in the neonates at 24 h after birth (33.6 and 39.7 mg/mL; P = 0.01) and at weaning (7.3 and 13.3 mg/mL; P < 0.001). Supplementation of Cr tripicolinate had minimal effects on humoral antibody response of the dam or its transfer to the neonate; however, parity greatly influenced the concentrations of immunoglobulins in the milk and their transfer to the neonate.  相似文献

603 neonatal piglets (German Landrace) were investigated for passive immunization affected by birth order, birth weight, litter size and parity. Concentrations of immunoglobulins in the blood serum of piglets on the first day of life served as measure for passive immunization. Blood samples were drawn at 12, 18 and 24 hours post partum. Quantitative measurements of immunoglobulin classes were carried out by radial immunodiffusion. There was a significant effect of birth order, because of the rapid changes in colostrum composition between onset of birth and the last piglet of each litter. The effect of birth weight was only partly significant and different for the immunoglobulin classes. There was no significant effect of litter size on passive immunization. Parity had a significant positive effect on IgG- and IgA-concentrations in sow's colostrum with litter two to six, parity 1 and 2 lead to highest IgM concentrations in the blood serum of piglets.  相似文献

Variation in serum IgG and IgM concentration was determined in three homozygous SLA-defined strains of miniature swine (SLAa, SLAc and SLAd) and one recombinant strain SLAg (ABcDd) as part of a study of SLA and other genetic effects on immune response. Data were obtained from 119 8-week-old piglets from 29 litters by 12 sires and analyzed using a SAS linear model for the effects of SLA haplotype, sire, dam, litter, sex, season of birth and sow parity. SLA haplotype (P less than 0.10) and other genetic effects due to sire (P less than or equal to 0.001) and dam (P less than or equal to 0.002) contributed to the variation in serum IgG concentrations. Season of birth and sow parity also affected IgG concentration as did litter effects. Least squares mean comparisons indicated that pigs of the dd, dg and gg haplotypes had significantly higher serum IgG than did pigs of the other haplotypes. Heritability estimates for IgG, calculated by paternal half-sib correlation, ranged from 0.31 to 0.27, indicating that selection for increased serum IgG concentrations would be possible. For serum IgM concentrations, only the effect of litter was significant at P less than or equal to 0.001 and P less than or equal to 0.009 by the radial immunodiffusion test read at 24 or 48 h. Since sire variance components estimates were negative, heritabilities were not calculated for IgM and are assumed to be zero.  相似文献

Presuckle colostral samples and lamb serum samples taken 36 h postpartum were assayed for immunoglobulin G1 (IgG1) concentration (mg/ml) using single radial immunodiffusion. Breeds sampled included Polypay (P), Rambouillet (R), Targhee (T), Columbia (C), Finnish Landrace (F) and Finn crosses (Fx). Sources of variation examined in IgG1 concentration in colostrum (dam trait) included dam's sire breed, dam's sire, age of ewe and number of lambs born. All sources of variation were statistically significant. Least-squares means of IgG1 levels for sire breed were 80, 64, 67, 64, 72 and 69 mg/ml for P, R, T, C, F and Fx breed groups, respectively. A fetal stimulus may exist to increase the mass of IgG1 in colostrum available for multiple births (61, 69 and 77 mg/ml for single, twin and triplet, respectively). Ewe age was a significant source of variation because of a high mean concentration of IgG1 in the yearling's colostrum (100 mg/ml), whereas only slight differences occurred between the other age groups (65 to 67 mg/ml), except for the 7-yr older group (53 mg/ml). Sources of variation examined in IgG1 concentration of lamb serum at 36 h postpartum (lamb trait) included lamb's sire breed, lamb's sire, age of dam, birth type and sex, with dam's colostral IgG1 concentration and day born as covariates. Sire within breed, birth type and the two covariates were significant. Least squares means for sire breed were 36, 32, 33, 32, 31 and 32 mg/ml of serum for P, R, T, C, F and Fx groups, respectively. Lamb serum IgG1 decreased as birth type increased. The heritability of IgG1, estimated by paternal half-sib analyses, was .19 +/- .12 for colostrum and .18 +/- .06 for lamb serum.  相似文献

The effect of postnatal acid-base status on the absorption of colostral immunoglobulins by calves was examined in 2 field studies. In study 1, blood pH at 2 and 4 hours after birth was related to serum IgG1 concentration 12 hours after colostrum feeding (P less than 0.05). Decreased IgG1 absorption from colostrum was associated with respiratory, rather than metabolic, acidosis, because blood PCO2 at 2 and 4 hours after birth was negatively related to IgG1 absorption (P less than 0.05), whereas serum bicarbonate concentration was not significantly related to IgG1 absorption. Acidosis was frequently observed in the 30 calves of study 1. At birth, all calves had venous PCO2 value greater than or equal to 60 mm of Hg, 20 of the calves had blood pH less than 7.20, and 8 of the calves had blood bicarbonate concentration less than 24 mEq/L. Blood pH values were considerably improved by 4 hours after birth; only 7 calves had blood pH values less than 7.20. Calves lacking risk factors for acidosis were examined in study 2, and blood pH values at 4 hours after birth ranged from 7.25 to 7.39. Blood pH was unrelated to IgG1 absorption in the calves of study 2. However, blood PCO2 was again found to be negatively related to colostral IgG1 absorption (P less than 0.005). Results indicate that postnatal respiratory acidosis in calves can adversely affect colostral immunoglobulin absorption, despite adequate colostrum intake early in the absorptive period.  相似文献

Effects of farm management, breed, mare age, gestation duration, and climatologic factors on colostral specific gravity, colostral IgG concentration, and foal serum IgG concentration were evaluated. Climatologic variables measured were daily maximal, minimal, and mean air temperature, precipitation, average relative humidity, and total solar radiation. Presuckle, postpartum colostrum samples were collected from 140 Standardbred, 94 Thoroughbred, and 59 Arabian mares from January through June during 1985 and 1986. Thoroughbred (farm A, n = 61; farm B, n = 33) and Arabian (farm C, n = 45; farm D, n = 14) mares were located in Ocala, Fla; Standardbred mares (farm E) were in Montgomery, NY. Mares from farms A, B, D, and E foaled in box stalls, and mares from farm C foaled in sand paddocks. Mares with premature lactation greater than 12 hours were not included in the study. Foals were clinically normal at birth and suckled colostrum without assistance within 2 hours of parturition. Specific gravity of presuckle colostrum samples was measured by use of an equine colostrometer. Blood samples were collected 18 hours after parturition from 253 of the 293 foals (n = 45, 25, 32, 13, 138 on farms A through E, respectively) to determine serum concentration of IgG. The IgG concentrations in colostrum and serum were measured by single radial immunodiffusion. Data were analyzed by multiple regression or chi 2 analysis. BACKGROUND: Lactoferrin is a colostral glycoprotein with antimicrobial properties. HYPOTHESES: (1) Serum lactoferrin and immunoglobulin G (IgG) concentrations are correlated and increase in healthy foals after ingestion of colostrum; (2) compared to healthy foals, ill foals will have lower lactoferrin concentrations that correlate with their IgG concentration, neutrophil count, the diagnosis of sepsis, and survival; and (3) plasma concentrations of lactoferrin will be less than serum concentrations. ANIMALS: Healthy foals (n = 16), mature horses (n = 10), and ill foals 1-4 days old (n = 111) that were examined for suspected sepsis were used for blood collection. Colostrum was obtained from 10 healthy mares unrelated to the foals. METHODS: Blood was obtained from the healthy foals at birth and 1-3 days of age and from the ill foals at admission. Serum IgG was quantified by single radial immunodiffusion (SRID). Lactoferrin concentrations in colostrum and blood were determined by an enzyme-linked immunosorbant assay. The sepsis score, blood culture results, neutrophil counts, and survival were obtained on ill foals. RESULTS: The mean colostral lactoferrin concentration was 21.7 microg/mL. Compared to values at birth, serum IgG (18+/-2 versus 2,921+/-245 mg/dL, SEM) and lactoferrin (249+/-39 versus 445+/-63 ng/mL, SEM) concentrations were significantly greater in healthy foals 1-3 days old. Serum lactoferrin concentration in 1-3-day-old healthy foals was not different from mature horses or ill foals. IgG and lactoferrin concentrations were significantly correlated only in healthy foals. Serum lactoferrin concentrations were significantly lower in ill neutropenic foals. The serum IgG concentration was significantly lower in ill foals as compared to healthy foals. Only serum IgG was significantly less in ill foals with a positive sepsis score and in nonsurvivors, Plasma lactoferrin concentrations were lower than serum concentrations, although values were significantly correlated. CLINICAL IMPORTANCE: Although both serum IgG and lactoferrin concentrations increase in healthy foals after ingestion of colostrum, only serum IgG is significantly correlated with the sepsis score and outcome.  相似文献

OBJECTIVE: To determine sensitivity and specificity of 4 methods to assess colostral IgG concentration in dairy cows and determine the optimal cutpoint for each method. DESIGN: Cross-sectional study. ANIMALS: 160 Holstein dairy cows. PROCEDURES: 171 composite colostrum samples collected within 2 hours after parturition were used in the study. Test methods used to estimate colostral IgG concentration consisted of weight of the first milking, 2 hydrometers, and an electronic refractometer. Results of the test methods were compared with colostral IgG concentration determined by means of radial immunodiffusion. For each method, sensitivity and specificity for detecting colostral IgG concentration < 50 g/L were calculated across a range of potential cutpoints, and the optimal cutpoint for each test was selected to maximize sensitivity and specificity. RESULTS: At the optimal cutpoint for each method, sensitivity for weight of the first milking (0.42) was significantly lower than sensitivity for each of the other 3 methods (hydrometer 1, 0.75; hydrometer 2, 0.76; refractometer, 0.75), but no significant differences were identified among the other 3 methods with regard to sensitivity. Specificities at the optimal cutpoint were similar for all 4 methods. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that use of either hydrometer or the electronic refractometer was an acceptable method of screening colostrum for low IgG concentration; however, the manufacturer-defined scale for both hydrometers overestimated colostral IgG concentration. Use of weight of the first milking as a screening test to identify bovine colostrum with inadequate IgG concentration could not be justified because of the low sensitivity.  相似文献

Seven Friesian calves were fed colostrum for four days beginning within 24 hours of birth, and milk thereafter. The concentration of alpha-lactalbumin in serum was measured by specific radioimmunoassay and compared to IgG assayed by electroimmunodiffusion. Serum concentrations of alpha-lactalbumin peaked at 387 +/- 85 ng ml-1 within eight hours of initial intake of colostrum, declining to 12 +/- 3 ng ml-1 by day 6. IgG rose steadily to 17 mg ml-1 by 48 hours of birth and remained relatively constant thereafter. The temporal pattern of alpha-lactalbumin in serum following colostrum intake confirms previous studies suggesting reduced absorption of colostral proteins between 24 and 36 hours. The presence of variable amounts of alpha-lactalbumin in serum even after 17 days, however, indicates limited transfer of milk-derived proteins across the gut at this time. The data further show that cessation of maximal gut transfer does not relate to molecular weight of transferred protein.  相似文献

OBJECTIVE: To determine the amount of colostral IgG required for adequate passive transfer in calves administered colostrum by use of oroesophageal intubation and evaluate the impact of other factors on passive transfer of colostral immunoglobulins in calves. ANIMALS: 120 Holstein bull calves. PROCEDURES: Calves were randomly assigned to specific treatment groups on the basis of volume of colostrum administered and age of calf at administration of colostrum. Colostrum was administered once by oroesophageal intubation. Equal numbers of calves received 1, 2, 3, or 4 L of colostrum, and equal numbers of calves received colostrum at 2, 6, 10, 14, 18, or 22 hours after birth. Serum samples were obtained from calves 48 hours after birth for IgG determination by radial immunodiffusion assay. Effects of factors affecting transfer of colostral immunoglobulins were determined by use of a stepwise multiple regression model and logistic regression models. RESULTS: A minimum of 153 g of colostral IgG was required for optimum colostral transfer of immunoglobulins when calves were fed 3L of colostrum at 2 hours after birth. Substantially larger IgG intakes were required by calves fed colostrum > 2 hours after birth. CONCLUSIONS AND CLINICAL RELEVANCE: Feeding 100 g of colostral IgG by oroesophageal intubation was insufficient for adequate passive transfer of colostral immunoglobulins. At least 150 to 200 g of colostral IgG was required for adequate passive transfer of colostral immunoglobulins. Use of an oroesophageal tube for administration of 3 L of colostrum to calves within 2 hours after birth is recommended.  相似文献
