Summary In artificial inoculations it was found thatFusarium oxysporum f.sp.tuberosi is able to penetrate through potato leaves. The fungus was isolated from the stem of infected plants from all inoculated cultivars. Tubers were completely destroyed if they were inoculated before sprouting. Not any destruction was observed if inoculation was made before young sprouts are longer than 1–3 cm; plants from these sprouts were infected. Lesioned tubers were more sensitive than unlesioned ones, and tuber rot and sprout damage were increased significantly.  相似文献

Summary Offspring were produced from a cross between two long-day-adaptedSolanum phureja clones which carried resistance to tuber soft rot (Erwinia carotovora subsp.atroseptica). In tests carried out on the produce of field-grown plants raised from tubers, over fifty per cent of the 173 offspring were found to be highly resistant. Assessments were also carried out of tuber yield, mean tuber weight, tuber number, shape, regularity, flesh colour, texture of the steamed flesh, fry colour, after-cooking blackening, sprout length after storage and overall dormancy. There were statistically significant differences between clones for all characters (P<0.001). Twelve of the clones were selected on the basis of high resistance, yield, tuber weight, regularity of shape and absence of after-cooking blackening. The value of resistant long-day-adapted diploid material for commercial breeding is discussed.  相似文献

Summary Erwinia carotovora in potato stocks can be detected more efficiently by the tuber incubation method in which the test tubers are first induced to rot under anaerobic conditions (followed by detection isolation of the bacteria on a selective growth medium or by serological techniques) than by direct lenticel sampling. However, the efficiency of detection is affected by both the extent of decay at sampling time and the incubation temperature. Detection ofE. carotovora is poor from extensive lesions developed at temperature of 25 C or above. Furthermore, detection ofE. carotovora var.atroseptica is more frequent than that ofE. carotovora var.carotovora at temperatures below 22 C in tubers contaminated by both organisms, whereas at higher temperatures the reverse is true. Only within a narrow temperature range, close to 22 C is there an equal chance of detecting both varieties.  相似文献

Summary Various methods of screening for resistance to root-knot nematodes were compared and evaluated. Seedling populations ofSolanum spp., grown in clay pots and plastic tubes with silver sand and inoculated with juveniles ofMeloidogyne chitwoodi andM. hapla, showed large differences in the number of egg masses on roots 7 weeks after inoculation. The differences were reproducible when re-testing was done with cuttings and plants from tubers. No resistance toMeloidogyne spp. was observed with ten potato cultivars when grown in clay pots, plastic tobes or closed containers. Plants from tubers in growth pouches developed a large two-dimensional root system, and after inoculation with juveniles the infection process could be observed over 8 weeks. A method of infecting potato tuber tissue withMeloidogyne is described, using tuber slices in Petri dishes as a potential screening test for tuber resistance.  相似文献

Summary Seed tubers, cv. Désirée, derived from stocks treated annually with thiabendazole were infected with thiabendazole-resistant strains ofHelminthosporium solani andPolyscytalum pustulans. Samples of seed tubers were either untreated or immersed for 5 min in fungicide suspensions of thiabendazole, imazalil or thiabendazole plus imazalil and planted on four farms in 1988 and 1989. After harvest, tuber samples from each treatment were treated with thiabendazole and stored for 6 months. Applying imazalil or thiabendazole plus imazalil to seed tubers decreased the severity of silver scurf and skin spot on stored progeny tubers. Thiabendazole applied to seed tubers or to progeny tubers after harvest did not affect the severity of either disease, but post-harvest treatment decreased the incidence of black scurf after storage.  相似文献

Summary In 1983 and 1984 there were reports in Norway of more severe late blight infection than usual on tubers of cv. Pimpernel, a cultivar without R-genes known to be somewhat resistant to both haulm and tuber infection. Results from routine tests confirmed earlier experience that cv. Pimpernel shows stable resistance to tuber late blight, although the ranking of cultivars may vary between years. In experiments during three harvest seasons tubers of different cultivars were inoculated with isolates ofP. infestans obtained from stocks of cv. Pimpernel with severe tuber late blight and with other isolates. In these experiments the isolates from cv. Pimpernel caused more rot on tubers of Pimpernel than did other isolates.  相似文献

Summary After three weeks curing at 10°C, potato tubers cv. Record were stored at 4°C under different controlled atmospheres (CA) for six months to study the effect on crisp fry colour, sprout growth and rotting. Combinations of low levels of CO2 (0.7–1.8%) and low levels of O2 (2.1–3.9%) gave a significantly lighter crisp colour, low sprout growth and fewer rotted tubers compared with 0.9% CO2 and 21.0% O2. Tubers stored in these conditions. showed a significantly higher weight loss and shrinkage after reconditioning. High CO2: low O2 combinations during storage completely inhibited sprout growth and caused the darkest crisp colour, but after reconditioning tubers gave the same level of sprouting and crisps as light as the other CA combinations. Furthermore these combinations, especially CO2 at 10 or 15%, increased the onset of rotting. Also our results showed that at low concentrations of CO2 (0.7–1.6%), and low O2 (2–2.4%) there was an increase in tuber rotting.  相似文献

The goal of these studies was to evaluate the influence of tuber age on the expression of resistance to Phytophthora infestans in five selected potato genotypes (three cultivars and two clones) varying in tuber resistance and maturity type, and also to select the best time for testing potato tubers for resistance to blight. Resistance was evaluated by inoculating whole tubers at 13 different times as they progressively aged during growth and storage. Tuber age was expressed as weeks after planting. Tubers were rated from week 13 (late July) until week 43 (late February) for tubers planted on 27–28 April, over three consecutive seasons (2001–2003). Analysis of variance performed on tuber resistance data showed significant effects of genotype, year and tuber age. Significant contributions of genotype × year, genotype × tuber age, year × tuber age and year × genotype × tuber age interactions were also detected. A slight increase in tuber resistance with tuber age was observed for cvs Bzura, Sokół and Irga, while the opposite trend was observed for the clones 97-A-63 and DG 92-227. Changes in tuber resistance with age of tested genotypes were not related to their maturity type. The period of most stable expression of tuber resistance was observed when tubers were tested between the 16th and 28th week after planting.  相似文献

Summary Four potato lines of cv. Désirée that express the pectate lyase (PL) isoenzyme 3 ofErwinia carotovora subsp.atroseptica were examined in a 4-year field experiment with respect to plant development, tuber yield and resistance of tuber tissue toErwinia soft rot. The PL3 degrades plant cell wall pectin into unsaturated oligogalacturonates eliciting plant defence responses. In one line, enzyme expression was controlled by the CaMV 35S promoter (C) and in three lines it was driven by the potato patatin B33 promoter (D). Plant development of the D-lines in field plots was not distinguishable from that of the non-transgenic counterpart. Also tuber yield was not too different. By contrast, plants of the C-line were smaller than those of the nontransformed counterpart and also showed reduced tuber yield. There were no significant differences in dry mass, starch and protein content of tuber tissue between PL transgenic and non-transgenic potatoes. But compared with the latter, field-grown tubers expressing the PL displayed an enhanced resistance toErwinia soft rot. Thus, average rotting caused byEc-bacteria was diminished in tubers of PL-transgenic lines by 34.1%. The resistance of tubers toEc soft rot was significantly correlated with the PPO activity in tuber tissue.  相似文献

Summary The intercellular volume within tuber tissue increases with tuber maturation. At the same time, both tuber injury after mechanical load and lentical perforation after water uptake by tubers in wet peat decreases. The degree of tuber injury is correlated with intercellular space (r=−0.667,n=16) and with water saturation of tissue (r=0.697,n=16). The tubers lose water during storage. In wet soil wilted tubers take up significantly higher amounts of water than turgid tubers, and lenticel perforation is much more frequent. Tuber maturation in autumn and low water loss during storage enhance the cushioning effect of the intercellular gas volume of tuber tissue against outside mechanical load and internal tension after water uptake.  相似文献

Summary The resistance to gangrene (caused byPhoma exigua var.foveata) of progenies from two hybridisation schedules involving cultivars resistant, susceptible, or of intermediate reaction to gangrene was assessed. Two methods of sampling tubers from field-grown single-plant plots of 18 clones representing each progeny were used: one tuber was taken from each plant, and a bulk sample of 18 tubers was selected at random from the whole progeny. Gangrene resistance was assessed after incubating tubers which had been rolled in cornmeal-sand inoculum ofP. exigua var.foveata, and was expressed as a percentage of tubers infected. Results from the two methods of sampling correlated closely, and agreed well with tests in a subsequent year on 10-tuber samples of each individual clone. It is concluded that an 18-clone sample adequately represented each progeny, and that the mid-parent gangrene score satisfactorily predicted the resistance of the resulting progeny. There are indications that susceptibility tended to be dominant. The implication of these findings to breeding for resistance to gangrene is discussed.  相似文献

Summary Various haulm treatments and harvest times differentially influenced the incidence of storage rots in potato tubers (Solanum tuberosum L.) in field experiments in Sweden performed over 4 years. The incidence of gangrene caused byPhoma foveata Foister (Phoma exigua var.foveata (Foister) Boerema) increased in cv. Bellona after spraying haulms with diquat as did the amount of infection caused byFusarium avenaceum (Corda ex. Fr.) Sacc. in all three cultivars (Bintje, Bellona, and Magnum Bonum). The lowestP. foveata infection levels occurred after pulling haulms. Late harvesting increased the frequency ofP. foveata in cvs Bintje and Bellona both at 2 and 4 wks interval between haulm treatment and harvest. Tuber and visible stem infection caused byP. foveata were correlated for cvs Bintje and Bellona, but not for cv. Magnum Bonum. The possible relation between tuber and stem infection is discussed.  相似文献

Summary Variation in aggressiveness to tubers among isolates ofPhytophthora infestans sampled from three potato growing regions in the Netherlands was compared. Variation in the ability to infect tubers of cv. Bintje was found between isolates of each of the three regional populations. The most aggressive isolate of the old population matched the average level of the new population in its ability to infect tubers. As a consequence, the commonly used reference isolate VK 6C can no longer be considered to be representative of the present population ofP. infestans. Therefore it is recommended that testing tuber resistance for the official list of potato cultivars with this isolate should be discontinued. Tuber infection and subsequent spread of the fungus in the tuber tissues were not found to be correlated. The components of tuber pathogenicity studied were not correlated to pathogenicity factors in the foliage, as measured under growth chamber conditions.  相似文献

Summary Potato tubers were inoculated with two biotypes ofPhytophthora infestans then stored at 3,7, 10 and 15°C. Image analysis quantified average reflective intensity (ARI) of diseased tissue from cut surfaces of sample tubers. Tuber tissue infection and infection rate were measured by calculating Mean ARI of samples. Average tuber tissue infection and infection rate was minimal at 3°C (P.i.-US8 orP.i.-US1). Tuber tissue infection increased at temperatures >3°C, from 220 Mean ARI seven days after inoculation (dai) to 190–150 Mean ARI 50 dai (depending on cultivar and biotype ofP. infestans). Rate of tuber tissue infection caused byP.i.-US1 at 7°C was about zero in cv. Snowden but greater than −0.2 ARI day−1 (cvs Russet Burbank and Superior). Rate of late blight infection in tuber tissue generally increased with temperature from −0.2 ARI day−1 (at 7°C) in all cultivars to a maximum of −0.8 ARI day−1 (10°C).  相似文献

Summary Potato tuber dormancy is usually defined as lasting from tuber initiation until a sprout of 2 mm long has been formed under storage conditions optimal for sprouting. We tried to find out whether there is a period during which buds of seed tubers do not grow and whether different batches of seed take the same time to grow sprouts 2 mm long. We measured changes in number of leaf primordia and length of tuber buds of cvs Diamant and Désirée over two years. After early haulm pulling, buds did not grow for at least 60 days (‘Diamant’) or 95 days (‘Désirée’). Buds in both cultivars and two tuber weights of ‘Diamant’ took about 20 days from the estimated onset of sprouting to grow 2 mm long. We question whether this period is always similar and thus whether the moment sprouts 2 mm long have formed is a good criterion for the end of dormancy.  相似文献

The nutrient and antinutrient components of tubers from seven cultivars ofDioscorea alata were determined. The average crude protein content ofD. alata tubers was 7.4%. Starch (75.6–84.3%) was the predominant fraction of the tuber dry matter. Significant differences in crude protein and starch contents were observed among cultivars. Vitamin C content of the yam tubers ranged from 13.0 to 24.7 mg/100 g fresh weight. The results showed yams to be reasonably good sources of minerals. Phytic acid contents of the yams were low, with values ranging from 58.6 to 198.0 mg/100 g dry matter. Total oxalate levels in yam tubers were found to be in the range of 486–781 mg/100 g dry matter, but may not constitute a nutritional concern since 50–75% of the oxalates were in the water-soluble form. The overall results are suggestive of the nutritional superiority of yams compared to other tropical root crops.  相似文献

Summary Formulae which determined tuber surface area from tuber dimensions and weight were tested by using samples from three potato cultivars. Surviving populations of a labelled isolate ofErwinia carotovora ssp.atroseptica following planting, and populations of a fluorescent pseudomonad following dusting with a bacterial powder formulation were used to demonstrate the application of these formulae to the calculation of bacterial density on the tuber surface. Results obtained by washing whole tubers using the formulae to calculate the surface area were shown to be comparable in sensitivity with those obtained by dilution-plating of periderm discs of 1cm2.  相似文献

Summary The severity of late blight tuber rot in five potato cultivars (Green Mountain, Sebago, Dorita, AC Brador and Island Sunshine) resulting from inoculation with US-1/US-8 combinations of Canadian isolates ofPhytophthora infestans was evaluated. In addition, the potential for mixed (A1∶A2) inoculations to result in oospore formation was examined. In studies conducted in 1996 and 1997, cv. Dorita (and Island Sunshine in 1997) developed the least amount of surface or internal necrosis relative to the other cultivars following co-inoculation with US-1 and US-8 isolates ofP. infestans. Differences also were revealed among isolate combinations used for inoculation. The combination of a US-1 isolate from Prince Edward Island with a US-8 isolate from New Brunswick consistently produced the least amount of necrosis in tubers of the various cultivars. Summary Seed quality in a crop harvested in 1984 was compared in tubers which either, (a) were unsprouted prior to harvest (normal), (b) had formed secondary chain tubers prior to harvest (primary), (c) secondary chain tubers. Normal and primary tubers formed substantial sprout growth during storage, while there was little sprout growth from secondary tubers. Plants derived from these seed tuber types differed, in terms of emergence, number of emerged stems, and tuber number. It is concluded that the tuber types can be considered to vary in terms of physiological age; normal and primary tubers appear to be physiologically older than the secondary tubers. This variability in seed condition can lead to variability in the performance of the subsequent crop.  相似文献

The use of industrial-type linear accelerators may be an alternative treatment for controlling sprouts in potatoes during long-term storage. The technical aspect of treatment has been described and may include largescale treatment of bulk potatoes as they are moving along a system of conveyors either to or from storage. The objective of the study was to evaluate the use of a linear accelerator for long-term sprout control under commercial storage conditions and the effects on tuber quality. An 18-MeV linear accelerator was used to provide sprout-inhibiting irradiation to various lots of ‘Russet Burbank’ potatoes. Successful sprout suppression was achieved with doses of 40 to 50 Gy (0 g sprout weight after 9 months of storage at 7.2 C) while higher doses caused undesirable increases in reducing sugars in the tubers. Glucose concentrations of treated tubers (0.25% fresh tuber weight) were higher after treatment than the untreated controls (0.08%), but returned to control levels after 2 to 6 months in storage. Glucose concentrations in treated tubers were significantly reduced with storage management for reconditioning by increasing the storage temperature for one month (from 7.2 C to 14.4 C) resulting in glucose concentrations near control concentrations. By increasing the dose-rate from the accelerator, recovery of tuber glucose concentration due to radiation treatments was significantly faster than when the dose was given more slowly. Potato tubers irradiated with high rates (100 Gy) of high-energy electrons exhibited an increase in soft rot and dry rot incidence in storage. Disease development in potatoes treated with sprout inhibiting doses of radiation (50 Gy) increased in lots with a high rot potential. Treated lots with low rot potential were successfully stored for 8 months or more without an increase in rot potential or shrinkage. Selection of lots for irradiation treatment should include some preliminary evaluation for development of disease in long-term storage.  相似文献
