Four groups of cattle were artificially infested with 50 first instar Hypoderma lineatum after either a primary natural infestation or in the absence of a primary infestation. In two groups the primary infestation had been terminated by the application of either an organophosphate insecticide or a macrocyclic lactone parasiticide. Circulating hypodermin C and specific antibodies were measured for 40 weeks after the artificial infestation. Stage specific mortality of the larvae was also monitored. Previously uninfested cattle exhibited typical antibody and antigen profiles during the infestations. Antibodies were first detected on Week 7 p.i., they rose to maximum values between Weeks 24 and 25, then declined as larvae reached the back. A second peak occurred at Week 32 and antibody remained more less constant thereafter. Previously infested groups exhibited a dramatic anamnestic response by Week 3 p.i. with antibody levels peaking at Week 8. A second peak was noted between Weeks 24 and 26 p.i. after which antibody levels declined and then remained relatively stable. The dynamics of circulating hypodermin C in the previously infested cattle resembled those in the previously uninfested cattle. Mortality of first instars did not differ among the four groups. Similarly mortality of second and third instars, in the warble, did not differ although there was a tendency for higher mortality in the previously infested, untreated group. These results reinforce previous work demonstrating the development of a significant immune response during the primary infestation that is reflected in the rapid and substantial production of antibodies upon re-infestation. It is significant that a challenge model using subcutaneous injection of newly hatched first instars avoids host immune responses in the skin that result in substantial larval mortality. The current data also support the concept that migrating first instars induce significant reduction in host immune response. There is a peak of antibody production as antigen levels peak during first instar migration to the back. As larvae molt to the second instar and antigen production ceases there is a persistence of antibody which suggests release of the immune response from the suppression induced by the first instar secretory antigens.  相似文献

In France, autumn and spring treatments are used for the control of cattle grubs. The autumn treatment consists of pour-on application of one of the following systemic organophosphorus insecticides: 4-tert-butyl-2-chlorophenylmemthyl methylphoporamidate fenthion, O,O-dimethyl phosphorodithioate S-ester with N-(mecaptomethyl)-phthalimide, and trichlorfon. The spring treatment consists of an aqueous preparation of trichlorfon or derris applied to the backs of infested cattle and cattle not treated in the autumn.The autumn treatment is not compulsory, although the Veterinary Services encourage it by providing technical and financial assistance. It was applied to 900 000 cattle in 1973. The spring treatment is compulsory, and the owner pays for it. It was applied to 3 × 106 cattle in 1973.The infestation rate seems to remain at 8–10% in areas where spring treatments have been used for a long time. The rate for the rest of the country is 20% and even there only 8% of the animals have more than five grubs per head.  相似文献

The activity of moxidectin was evaluated in 1988 and 1989 against natural infections of internal parasites in 20 critical tests (n = 20 equids) and three controlled tests (n = 20 equids). Two formulations, injectable administered intramuscularly (i.m.) or intraorally (i.o.) and gel i.o., were given at dose rates of 0.2, 0.3 or 0.4 mg kg-1 body weight. For the critical tests (all three dose rates evaluated), removals of second instar Gasterophilus intestinalis were 93-100%, except (89%) for the injectable formulation (i.m.) at 0.2 mg kg-1. Removals of third instar G. intestinalis were 88-100% for the injectable formulation given i.m. or i.o. and 93-100% for the gel formulation, except (53%) for one batch (0.4 mg kg-1). Activity was 100% for third instar Gasterophilus nasalis, Parascaris equorum, Strongylus vulgaris and Strongylus edentatus. For Oxyuris equi, removals were 91-100%, except (27%) for one batch of the injectable formulation given i.o. at 0.3 mg kg-1. There was apparent activity against migrating S. vulgaris and S. edentatus at various dose rates and routes of administration for both formulations. At necropsy, there were local reactions observed at the injection site of three equids. In the controlled tests, dose rates were 0.2 or 0.4 mg kg-1. Removal of third instar G. intestinalis was highest for the injectable formulation given i.m. All formulations and dose rates were highly effective against S. vulgaris and S. edentatus, but variable and incomplete against O. equi. Removal was excellent on Habronema muscae and on migrating S. vulgaris and S. edentatus, although incomplete on S. vulgaris. Gasterophilus nasalis third instars and P. equorum were present in low numbers in some non-treated equids, but none were recovered from treated equids. Toxicosis was not evident.  相似文献

In controlled experiments utilizing artificial infestations, a topical formulation of ivermectin (IVOMEC Pour-On for Cattle) was 100% effective against migrating first instar Hypoderma lineatum for 3 weeks following treatment. Larvae were apparently killed early in the infestation as no significant level of specific antibodies was found in the treated calves. At 4 weeks post-treatment the efficacy remained high at 96%; mortality of larvae in the one calf in which warbles were observed and from which mature third instars were collected, was higher than that seen in the untreated calves, indicating some level of treatment induced effect. No specific antibodies were detected in calves that did not develop palpable warbles. Antibody kinetics in those calves from which viable larvae emerged were typical. The length of activity of this product against early stages of the cattle grub makes it practical to apply treatment up to 3 weeks before the end of fly activity.  相似文献

For decades, about 30% of the young cattle in the Steinfurt district of the North German Basin were infested with Hypoderma bovis. Compulsory treatment with rotenone between 1934 and 1963 failed to control infestation. When systemic organophosphorous insecticides became available a 3-year scheme was instituted in which all the 6–24-month-old cattle were treated annually, between 15 November and 15 December, with trichlorfon spot-on.At first, the treatment was not compulsory and warble infestation was occasionally found in cattle purchased outside the district. Later, the treatment was made compulsory and since 1966 warble infestation has not been detected in the district.  相似文献

Two studies were conducted with doramectin topically administered at 500 microg/kg body weight to assess retention of therapeutic efficacy against nematode infections of cattle before, and after, simulated rainfall. In the first study, 50 heifers, with patent nematode infections, were allocated to one of five treatment groups. An untreated control group and one doramectin-treated group were not exposed to simulated rainfall. Simulated rainfall was applied at a rate of 25.4 mm of water in 35 min to three of the five groups: one group immediately before treatment, the second group 90 min after treatment, and the third group 24 h after treatment. Fecal samples were collected for determining egg counts 14 days after treatment. Percentage efficacy ranged from 97.3% to 100% in all treated calves, regardless of exposure to simulated rainfall. The second study involved 40 mixed-sex cattle that were allocated to one of four treatment groups (one saline control and three doramectin-treated groups). All cattle were confirmed to be free of nematode infections prior to oral gavage with infective larvae of Dictyocaulus viviparus, Cooperia oncophora, and Ostertagia ostertagi. Twenty-six days after infection, three groups were treated with doramectin pour-on and exposed to 20 mm of simulated rainfall over 40 min: one group 60 min before treatment, the second 20 min after treatment, and the third 40 min after treatment. Approximately two weeks after treatment, all cattle were necropsied for worm counts. In all treated groups, the percentage efficacy against O. ostertagi and D. viviparus was >99% to 100%. Percentage efficacy against Cooperia ranged from 97% to 98%. Results indicated that doramectin pour-on remains efficacious against nematodes of cattle when administered immediately before or after rainfall.  相似文献

OBJECTIVE: To assess the efficacy of zeta-cypermethrin pour-on to control cattle lice. DESIGN: Five field trials in south-eastern Australia. PROCEDURE: Zeta-cypermethrin pour-on, deltamethrin pour-on and pour-on vehicle were applied to groups of 10 cattle. Lice were counted before treatment and 14, 28, 42 and 56 days after treatment. RESULTS: Zeta-cypermethrin pour-on given at 2.5 mg/kg was equivalent to, or marginally more effective than a deltamethrin pour-on at 0.75 mg/kg. It eliminated B bovis and H eurysternus and gave good control of L vituli and S capillatus. Zeta-cypermethrin at 1 mg/kg gave good control of B bovis and H eurysternus but was not satisfactory against L vituli and S capillatus. CONCLUSION: Zeta-cypermethrin pour-on, given at 2.5 mg/kg, is an effective treatment for cattle lice control. Zeta-cypermethrin, and other synthetic pyrethroid pour-ons, are the treatment of choice to control B bovis.  相似文献

AIM: To investigate the efficacy of pour-on anthelmintics against field strains of parasitic nematodes in young cattle on five farms in New Zealand. METHODS: Faecal nematode egg count (FEC) reduction (FECR) tests were carried out on five calf-rearing farms using pour-on formulations of levamisole, ivermectin, eprinomectin, and the simultaneous administration of levamisole and ivermectin. Faecal samples were collected per rectum before treatment and about 7, 14, 21 and 28 days after treatment, for FEC and faecal nematode larval culture. RESULTS: Resistance (i.e. <95% reduction in FEC) of Cooperia oncophora to ivermectin and eprinomectin was identified on all five farms. There was limited evidence of possible emerging resistance in Ostertagia spp to ivermectin but not eprinomectin, in short-tailed larvae of Cooperia spp to ivermectin and eprinomectin, and in Trichostrongylus spp to ivermectin, eprinomectin and levamisole used separately. Levamisole was effective against C. oncophora, but had variable efficacy against Ostertagia spp in the calves in this study. Simultaneous treatment with levamisole and ivermectin pour-on formulations were effective against all genera on all farms. CONCLUSIONS: To effectively manage roundworm parasites in their calves farmers need to be aware of the resistance status of the parasites on their farms. Levamisole is likely to be an effective anthelmintic on most farms at times of the year when the impact of Ostertagia spp is not high. Simultaneous administration of levamisole and ivermectin pour-on anthelmintics to cattle is likely to control both ML-resistant C. oncophora and stages of Ostertagia spp that are not controlled by levamisole alone.  相似文献

为了解龟纹瓢虫(Propylaea japonica)控制豌豆蚜(Acyrthosiphon pisum)的能力,在实验室内采用圆盘法测定了不同龄期龟纹瓢虫对豌豆蚜的捕食功能反应,以期掌握龟纹瓢虫控制豌豆蚜的能力。结果表明:龟纹瓢虫4个龄期的幼虫和雌雄成虫捕食豌豆蚜的数量与豌豆蚜密度呈负加速曲线关系,符合Honing-Ⅱ型功能反应模型。龟纹瓢虫雌成虫的日捕食量最大,其次是第4龄幼虫和雄虫。模型预测了龟纹瓢虫1,2,3,4龄幼虫及雄、雌成虫对豌豆蚜的日最大捕食量分别是6.5,20.4,32.3,71.4,58.8,80.0头。综上所述,龟纹瓢虫对豌豆蚜有较强的控制能力。  相似文献

Goats naturally infested with Damalinia caprae lice were used to evaluate the efficacy of flumethrin pour-on. The maximum load of lice infestation was detected in the neck region. The LC50 value of flumethrin pour-on against D. caprae was found to be 119.35 ppm and the 95% confidence limits were calculated to be 104.10 and 136.85 ppm. Flumethrin at the rate of 1 mg/kg body weight by pour-on application along the mid-dorsal line was found to be 100% effective in treating lice-infested goats compared with the control goats. The residual action of flumethrin was found to be at least 42 days during which period no apparent signs of any local or general adverse effects were observed. Based on these studies, it is suggested that flumethrin pour-on may be successfully used for both the prophylaxis and treatment of D. caprae infestation in goats and it is necessary to repeat the treatment after 42 days. The method of application is highly advantageous as it can be conveniently applied, even in animals with thick hair coat and during the winter months.  相似文献

Several insecticides were applied to cattle at various rates, mixtures, application methods and numbers of treatments and evaluated for control of cattle lice. The insecticides included permethrin at various rates as a pour-on with and without the synergist piperonyl butoxide (PBO), and also at various rates as a spray with and without PBO. Pirimiphos-methyl was evaluated as a pour-on at various rates and with one or two applications. Lambda cyhalothrin in microencapsulated formulation was compared to a non-microencapsulated formulation. Rabon(R) was applied as a pour-on at two rates and at one or two treatments for each rate. Dimilin(R) was applied as a pour-on at two rates and mixed with permethrin and applied at two rates. Five endectocides, Eprinex(R), Ivomec(R), Dectomax(R), Cydectin(R) and Phoenectin(R) were all applied at the same rate. The cattle utilized in this research were all infested with a mixture of lice species. The species included: Bovicola (Damalinia) bovis (L.), Haematopinus eurysternus (Nitzsch), Linognthus vituli (L.) and Solenoptes capillatus (Enderlein). Most of the cattle were infested with at least two species and some had three or all four species present. All of the treatments except permethrin 1.0%+5% PBO, at a rate of 3ml/45.4kgwt. provided acceptable lice control with one application. Data indicated that applying the insecticides in early January should provide enough control to render the need for a second treatment unnecessary.  相似文献

Four studies were conducted to determine the efficacy of a 1% lambdacyhalothrin pour-on (Saber Pour-on, Schering-Plough Animal Health) for control of sucking lice (Linognathus vituli, Haematopinus eurysternus, Solenopotes capillatus) and biting lice (Damalinia bovis) on beef cattle. Seventy-four mixed-breed cattle naturally infested with one or more species of lice at locations in Wisconsin, Nebraska, Oklahoma, and North Dakota were included in the study. Pretreatment lice samples were taken and identified by genus with the exception that sucking lice were not identified by genus at the North Dakota site. In January or February, half of the cattle at each location received a single application of 1% lambdacyhalothrin pour-on at 10 ml per head for cattle weighing less than 273 kg (600 lb) or 15 ml per head for cattle weighing 273 kg or more. The other cattle at each site served as untreated controls. Lice on designated body areas were counted 2, 4, 6, and 8 weeks after treatment, and the sum of all lice observed on each counting area was reported for each animal. The collective efficacy 6 weeks after treatment against three species of sucking lice (L. vituli, H. eurysternus, S. capillatus) was 88.4% at the Wisconsin site, 92.0% at the North Dakota site, and 100% at the Nebraska and Oklahoma sites. The 1% lambdacyhalothrin pour-on eliminated all biting lice within 2 weeks after treatment, and no biting lice were detected 8 weeks after treatment. A single treatment of 1% lambdacyhalothrin pour-on administered when lice populations were highest (January or February) provided effective season-long control of both biting and sucking lice on cattle.  相似文献

The efficacy of minidose of pour-on ivermectin and eprinomectin formulations against first instar larvae of Przhevalskiana silenus was observed in naturally infested goats in the Jammu region, North India. The study was performed in mid August 2011. A total of 280 goats were randomly divided in to 7 groups of 40 each. Goats of the first three groups were treated with pour-on ivermectin at dosage of 2, 5, and 200 μg/kg body weight, respectively, whereas animals of the fourth to sixth groups were treated with pour-on eprinomectin at 25, 50, and 500 μg/kg body weight, respectively. Group VII animals were kept as untreated control. The results indicated that no warbles were recorded between December 2011 and March 2012 on back of animals treated with pour-on preparations of ivermectin at dosage of 5 and 200 μg/kg body weight, respectively, and eprinomectin at dosage of 50 and 500 μg/kg body weight, respectively. Thus, it is concluded that administration of minidose of pour-on ivermectin (5 μg/kg body weight) and eprinomectin (50 μg/kg body weight) is cost effective and so can be used for warble fly control campaign in Jammu region.  相似文献

A pilot survey using a structured questionnaire was conducted in Tororo and Busia districts of Uganda on the knowledge and attitudes of cattle owners regarding tsetse fly and trypanosomosis control, in order to understand factors that hindered their full participation. A total of 81 cattle owners was randomly selected and interviewed, of which 92.5% were aware of tsetse flies and trypanosomosis and 87.6% recognised animal trypanosomosis as a problem in the area. Most cattle owners were aware of tsetse fly trapping (76.5%), isometamidium chloride use (55.5%), diminazene aceturate use (48%) and pour-on applications (18.5%). However, knowledge did not coincide with the application of control measures. Despite the widespread awareness, tsetse fly trapping and pour-on applications were used by only a small percentage of cattle owners (7.5% applied tsetse fly trapping while 76.5% were aware of it; 1.2% applied pour-on insecticides while 18.5% were aware of them). Differences between awareness and application were highly significant for tsetse fly trapping (chi2 = 67.8, d.f. = 1, P < 0.001) and pour-on applications (chi2 = 10.8, d.f. = 1, P < 0.05), but not for isometamidium chloride use (chi2 = 0.08, d.f. = 1, P = 0.77) and diminazene aceturate use (chi2 = 0.00, d.f. = 1, P = 1.00). Most cattle owners (97.5%) were willing to participate in future control programmes, but preferred participating on a group basis (85.2%) rather than individually (14.8%). The 4 most favoured control options in order of importance were: fly traps supplied by the government and maintained by cattle owners; contribution of labour by cattle owners for trap deployment; self-financing of trypanocidal drugs and self-financing of pour-on insecticide. The control options that should be selected in order to elicit full participation by cattle owners are discussed.  相似文献

Soluble fractions of Hypoderma lineatum third instar fat body, haemocytes and haemolymph were formulated with Quil A and used to immunize four groups of calves while a fifth group remained untreated. Calves received two subcutaneous injections of the soluble fractions, or adjuvant only delivered two weeks apart. Two weeks after the last injection the calves were exposed to 50 newly hatched larvae of H. lineatum which were placed on the skin and allowed to penetrate. Survival of larval stages was monitored by weekly palpation and collection of emergent third instars. Antibody responses to the immunogens were evaluated by immunoblots and following infestation antibody responses to first instar antigens were evaluated by an ELISA. Non-immunized calves and calves injected with adjuvant were all palpation positive for cattle grubs. In groups immunized with fat body, haemocyte and haemolymph components 100%, 33% and 33% were palpation positive for grubs respectively. First instar mortality, as reflected in palpable grubs, was high in the groups receiving injections with tissue components (99.3%, 95.1%, 95.8%, 83.9 and 80.4% mortality for those groups receiving fat body, haemocyte, haemolymph, adjuvant or control respectively). Second and third instar mortality was also higher in the immunized groups (100.0%, 91.7%, 91.7% for fat body, haemocyte, and haemolymph respectively) in comparison to the adjuvant only (14.0%) and unvaccinated (33.3%) groups. No viable flies emerged from pupae originating from larvae emergent from any of the immunized groups. Calves receiving the tissue extracts developed antibodies to several protein components following the second immunization which were still present 13 weeks post-infestation. Several proteins appeared to be common among the three tissue extracts and were recognized by antibodies from the immunized calves. All groups of calves became positive for antibodies to first instar antigens, although in some immunized calves the antibodies were transient.  相似文献

Anthelminthic resistance in nematodes of beef cattle is an emerging issue globally with implications for effective parasite control. The prevalence of resistance in beef cattle in the Mediterranean-style climatic zone of south-west Western Australia was assessed on 19 farms, using faecal egg count reduction tests. Pre-treatment faecal worm egg counts were compared with counts at 14 days after treatments with ivermectin (injectable), fenbendazole (oral), or levamisole (oral). A separately grazed group treated with topical ivermectin (pour-on) and sampled at 28 days was included as a comparison against injectable ivermectin. The results demonstrate that resistance is common, with failure of at least one anthelmintic (<95% reduction for each species, by arithmetic means) for either of the major species Cooperia oncophora or Ostertagia ostertagi on 17 of the 19 properties. Resistance to ivermectin (injectable) was demonstrated in C. oncophora in 59% of tests, but ivermectin was fully effective against O. ostertagi by this route. Conversely, O. ostertagi resistant to fenbendazole and levamisole were present on 50% and 67% of farms respectively, with both fully effective against C. oncophora. The finding of Haemonchus placei on several properties was unexpected but the egg counts were low and there is no suggestion of pathogenic effects. An indication of reduced efficacy of the pour-on ivermectin formulation compared to the injectable was apparent against both C. oncophora and O. ostertagi, and this may have implications for resistance development, given the widespread use of topical treatments reported in this region. This survey confirms that anthelminthic resistance in nematodes of beef cattle is common in Western Australia and the pattern of occurrence is in general agreement with surveys elsewhere in Australia and in other countries.  相似文献

Thirty-five castrate or intact Holstein male calves, ranging in weight from 122 to 210kg, were used in the study. On study Days -15 and -14, all study calves were treated with fenbendazole 10% paste at 10mg/kg to remove existing nematode infections. All cattle had zero egg counts on Day -1. Experimental infections of a mixed species inoculum were administered to all calves on Day 0. The inoculum consisted of Cooperia spp. (primarily C. punctata, but also C. pectinata, C. spatulata, C. oncophora and C. surnabada-total 40,961); Ostertagia ostertagi-1550; Trichostrongylus colubriformis-4996; and Oesophagostomum radiatum-38. Necropsy results from two of three monitor calves slaughtered on Day 6 after infection indicated that Day 6 was an appropriate time to evaluate efficacy of moxidectin against fourth larval stages (L(4)). The remaining 32 calves were randomly allotted to four groups of eight based on body weights. Eight calves (Group 1) were treated with moxidectin 0.5% pour-on at 0.5mg/kg on Day 6 to evaluate efficacy against nematode larval stages; eight control calves (Group 2) were matched with these principals. Eight calves (Group 3) were also treated with moxidectin pour-on at the same dosage on Day 23 in order to determine efficacy against adult nematodes; eight control calves (Group 4) were matched with these principals. In both cases, principals and controls of the Days 6 and 23 treatments were necropsied at 14-15 days after treatment. C. punctata was the only species found in a sufficient number of controls to evaluate moxidectin efficacy against the L(4). Moxidectin pour-on was not effective (P<0.05) against C. punctata L(4) by arithmetic means, but was highly effective based on geometric means. Regardless of whether arithmetic or geometric means were used for percent efficacy calculations, moxidectin pour-on was demonstrated to be highly effective (>99%) against the adult stages of C. oncophora, C. punctata, C. spatulata, C. surnabada, Cooperia spp. adult females and T. colubriformis adults.  相似文献

Ivermectin is a worldwide-used antiparasitic drug largely administered to cattle as a topical formulation (pour-on). The actual plasma and faecal disposition of pour-on ivermectin in cattle was documented using an original pharmacokinetic model, and taking into account the oral ingestion of the topical drug following physiological licking as a secondary route of exposure. Six pairs of monozygotic twin cattle received successively one i.v. and two pour-on administrations of ivermectin at a 3-5-month interval. For one pour-on administration, the twins were separated into an unrestrained group and a group where self- and allo-licking were prevented. Ivermectin concentrations in the plasma and faeces were determined by HPLC. Licking resulted in a high intra-and inter-individual variability of systemic exposure after topical application. By the means of pharmacokinetic modelling, we showed that 58-87% of the pour-on dose was ingested, while only 10% was absorbed percutaneously. Approximately 72% of the ingested ivermectin transited directly into the faeces, resulting in a 7-fold higher faecal excretion of the parent drug than in the non-lickers. We conclude that topical administration does not guarantee a controlled drug delivery in cattle. More importantly, the simulations revealed that non-treated cattle could get easily contaminated by allo-licking, raising the public health problem of unexpected drug residues in edible tissues.  相似文献

AIMS: To measure the efficacy of a pour-on formulation of moxidectin against lungworm and abomasal parasites in weaner wapiti x red deer and to compare this with its efficacy in weaner red deer. METHODS: Six red and six wapiti hybrid deer, naturally infected with lungworm and gastro-intestinal parasites, were treated with pour-on moxidectin at 500 microg/kg body weight and slaughtered 14 or 16 days later, along with six red and six wapiti hybrid untreated control deer. Total worm counts were performed on the lungs, abomasum and abomasal digest of each deer. RESULTS: The efficacy of moxidectin pour-on was 100% against adult and immature lungworms (Dictyocaulus viviparus) in red deer, and 100% and 99.7% effective against adult and immature lungworm in wapiti hybrid deer. The efficacy of moxidectin pour-on was 100, 100, 99.9 and 99.9% respectively against adult, fifth stage, late fourth stage and early fourth stage larvae of Ostertagia-type nematodes (assumed to be Ostertagia, Spiculopteragia, Skrjabinagia and Apteragia spp.) in both red and wapiti hybrid deer. CONCLUSIONS: The pour-on formulation of moxidectin, at 500 microg/kg body weight, is highly effective against mature and immature lungworms and abomasal nematodes in wapiti hybrid deer and equally effective in red deer.  相似文献
