In a continuous biogas plant, receiving manure from 200 dairy cows and 400 calves and young stock, survival of salmonellas and Ascaris suum eggs was studied. The bacteria and parasite eggs were kept in filter sacs in the manure that had a temperature of 55°C. No viable salmonellas or Ascaris suum eggs could be found after 24h in the digester.Survival of salmonellas and Ascaris suum eggs was also studied in the manure pit where the manure was stored after digestion. The temperature in the manure pit varied between 22–27°C. Salmonellas survived 35 but not 42 days. On day 56, when the experiments had to be stopped, 60% of the Ascaris eggs were viable.  相似文献

Naturally occuring outbreaks of Ascaris suum infection in calves have usually beeen found in animals nine to 12 months of age. The circumstances surrounding these outbreaks suggest that yearling calves are either particularly susceptible to a primary exposure to A. suum or react strongly to A. summ after sensitization early in life to this or some related ascarid. To determine the effect of reinfection with A. suum nine to 12 months after varying levels of exposure to this nematode, six calves were inoculated with 200,000 to 9,000,000 eggs. Neither death nor, in general, severe clinical signs resulted from reinfection. All calves were examined 15 days after reinfection with pathological changes noted only in the lungs and consisting of emphysema, alveolar wall thickening as well as accumulations of fibrin, eosinophils and hemorrhage in the lumina of alveoli. The findings suggested that exposure to A. suum early in life is not a factor in the development of disease in calves infected at one year of age. It was also found that the eosinophilia that develops following a primary infection with A. suum evidently persists for at least one year.  相似文献

A worm-control program utilising treatment of young grazing cattle with fenbendazole on two occasions during summer was tested in the Mediterranean-type climatic environment of south-west Western Australia. The grazing system aimed to produce steers by introducing three-month-old weaned calves to pasture in mid-winter until they were sold in early summer. Comparisons were made of the numbers of worm eggs passed on to plots by treated and untreated animals during autumn, the performances of treated and untreated cattle and the performances of calves introduced to the plots in mid-winter. The “tracer” calf technique was used to determine the availability of infective larvae on one untreated and one treated plot for each of the two years of the experiment.Treated animals deposited less Ostertagia spp. eggs on to pasture during autumn than did untreated animals in one of the two years. In both years they deposited less eggs of worm species other than Ostertagia spp. Less intestinal worms were acquired by “tracer” calves grazing treatment plots than those grazing no-treatment plots in both years, but there were no differences in the number of abomasal worms acquired.The reduction in availability of infective larvae of intestinal worms was insufficient to prevent the occurrence of parasitic gastroenteritis in calves introduced to the plots in mid-winter. The fenbendazole treatments did not confer any immediate body-weight advantage on treated animals.On both treatment and no-treatment plots, there were few infective larvae available to grazing cattle during early autumn, there was a rapid attainment of peak availability in winter and then a decline to low availability by mid-spring. In one year, infective larvae of intestinal worms (almost exclusively Cooperia spp.) increased in availability again in late spring and early summer. A high proportion of retarded worms was never a feature of the worm counts of “tracer” calves.It was concluded that the treatments may have been more effective had they been given during autumn.  相似文献

Gross changes in the lungs of Ascaris suum- infected calves consisted of atelectasis and hemorrhagic foci, edema and emphysema, frequently with bullae. Prominent microscopic lung lesions were edema and emphysema of the interlobular septa with large numbers of eosinophils within and around lymphatics, peribronchiolar lymphoid nodules and parasitic granulomas. Many of the microscopic features were consistent with those found in atypical interstitial pneumonia. Changes in the alveoli were atelectasis, the exudation of plasma proteins, mononuclear cells and eosinophils, and alveolar wall thickening. Lesions found later included fibrosis and fetalization of the alveolar walls. Plasma cells and neutrophils were not common. Challenge with Toxocara canis after sensitization with A. suum resulted in the lungs developing a few areas of atelectasis. Migration of T. canis to lungs of calves is slower than A. suum. A. suum larvae were always found in bronchi, bronchioles and alveoli of calves that died. Lesions were observed in the liver but not the kidney of A. suum infected calves; both lung and liver lesions tended to resolve with time.  相似文献

Clinical signs consistent with those of atypical interstitial pneumonia (AIP) were induced in calves sensitized with infective Ascaris suum eggs at seven to 20 weeks of age and challenged at three-week intervals one or more times. These signs usually appeared on the sixth or seventh day postinfection and reached maximum severity between the tenth and 13th days following infection. Prominent signs were: dyspnea, often with expiratory grunt, coughing, mouth breathing and emphysema as well as increased respiration and heart rates. In general, the intensity of signs was dependent upon dose size, although a single small dose resulted in acute signs and death in one calf. Intermittent coughing and vesicular sounds were induced in calves given A. suum eggs continually over prolonged periods. No respiratory abnormalities resulted from challenge with Toxocara canis after sensitization with A. suum. Antihistamine therapy did not alter the clinical signs in A. suum infected calves.  相似文献

Groups of piglets 20 to 28 days of age were dosed with 50 or 10 000 Ascaris suum eggs. The pigs were sacrificed from day 16 post infection onwards and worms present in the intestine were counted. Large numbers of immature parasites were recovered initially following the 10 000 egg dose, but the numbers decreased rapidly with the and worms were not found in pigs examined in the late prepatent phase. All pigletts infected with 50 eggs were found to harbour worms and the infection reached patency. The high level infections resulted in an eosinophil response which reached maximum values during the second week after infection. The low level infections which resulted in patency were not followed by any significant eosinophil response.  相似文献

Calves were immunized with 10 000 infective larvae each of Haemonchus contortus, Ostertagia ostertagi, and Trichostrongylus axei and challenged with 100 000 of each species. When compared with nonimmunized controls, the immunized calves had 95% fewer H. contortus, 10% fewer O. ostertagi, and 20% fewer T. axei; the latter two reductions were not significant. In four other experiments single or multiple immunizing inoculations of T. axei followed by challenge at various intervals after immunization failed to result in significant resistance measured by the number of worms of the challenge inoculation that became established, by post-challenge fecal egg count, and by gross pathologic alterations of abomasa at necropsy.  相似文献

In a survey of 2500 market weight pigs in a Saskatchewan abattoir, 37% were infected with adult Ascaris suum and in 46% there were milkspot hepatic lesions. A total of 60% of the pigs examined had some evidence of A. suum infection. Most infected pigs contained less than 50 adult ascarids.

In a second abattoir survey of a further 2500 market weight pigs, 44% of the animals had milk-spot hepatic lesions. In approximately 12% of the affected pigs these lesions were very severe and a very similar proportion of pigs' livers were condemned or sent for use as animal feed.

The results of these surveys are discussed with particular reference to the significance of A. suum infection in growing pigs in Saskatchewan.

Under adverse obstetrical conditions, appropriate supervision and assistance during the immediate neonatal period are of the utmost importance, especially for weak calves. The aim of this study was to establish the effects of dystocia and oxytocin infusion on neonatal vitality, acid-base balance, and electrolyte and haematological homeostasis of dairy calves. Data were collected for 30 Holstein calves which were allocated to three groups: normal calving (n = 10); dystocia with mild to severe obstetric assistance (n = 10); and uterine inertia treated with oxytocin (n = 10). All 30 calves exhibited normothermia at birth, but had a significant decrease in body temperature after 60 min. Dystocic calves had lower Apgar scores than calves in the other two groups, and had respiratory and metabolic acidosis. Calves from normal calvings had normal blood pH, but base excess below the reference range. The mean partial pressure (Pa) of oxygen of calves whose dam had been treated with oxytocin was lower than that of calves from normal calvings. In all experimental groups, there was improvement in metabolic status in the first 60 min postpartum as PaCO2 values significantly decreased. All calves had normonatraemia, normokalaemia and normochloridaemia during the study period, but calves born to dams treated with oxytocin had a higher sodium concentration than those in the two other groups.  相似文献

Pilobolus kleinii spores isolated from cattle faeces were administered daily by the oral route to two donor calves excreting Dictyocaulus viviparus larvae in their faeces.Faeces were collected at daily intervals from the donor calves, and on each day a pooled sample of 500 g was placed outside a pen containing two parasite-free experimental calves. A wire screen prevented the experimental calves from coming into contact with the artificially deposited faecal portions.Pilobolus kleinii fruit bodies were observed on the faecal surface in large numbers 5–10 days after deposition.Sporangial discharge was observed to be directed against a window which served as the only light source. Sporangia were found up to 110 cm away from the faeces, and it was estimated that at least 29% of them had been transmitted to a feeding trough placed close to the wire screen. Infective D. viviparus larvae were observed on some of the discharged sporangia.The experimental calves started excreting low numbers of D. viviparus larvae in their faeces 40–49 days after the first portions were deposited. These results indicate that P. kleinii was reponsible for the infection of the experimental calves with D. viviparus, through the discharge of sporangia carrying infective lungworm larvae.  相似文献

By grazing parasite-free susceptible calves on naturally infected paddocks for eighteen day periods, Nematodirus helvetianus was shown to survive over two winters on marshland pastures under Maritime climatic conditions in three successive years although the numbers surviving the second winter in two of the three years were low. Climatic conditions of below normal temperatures and little snowfall would appear to have a detrimental effect on the survival of Nematodirus over a second winter although large numbers did survive one winter of such adverse weather. However, under certain conditions, large numbers of Nematodirus can overwinter two years on pastures since over 12,000 worms were picked up by individual calves grazing paddocks in 1969 which had been vacant since 1967.

The findings indicated that few, if any, Ostertagia ostertagi and Cooperia oncophora survive over a second winter under Maritime conditions. The sharp decline in numbers of infective Ostertagia and Cooperia on pastures during the first grazing season following pasture contamination observed in this and previous investigations indicate that few worms of these species survive longer than a year under Maritime conditions.

An increment in the number of circulating eosinophils occurred between the 11th and 14th days after infection with Ascaris suum and this increase was generally greater after a challenge infection. Continual infection with small numbers of A. suum eggs over prolonged periods resulted in circulating eosinophil levels which fluctuated and were dose-dependent. The per cent marrow eosinophils always increased after a primary infection and a greater increase usually followed a challenge infection. The maximum increment of marrow eosinophils occurred between the tenth and 12th days and preceded the rise in circulating eosinophils by 36 hours. Antihistamine therapy did not alter eosinophil responses to A. suum. Circulating eosinophilia was not usually reflected by drastic changes in differential white cell counts. However, an increase in total white cell count often followed infection with A. suum and frequently parallelled changes in eosinophil counts. Hemoglobin and P.C.V. values remained within normal limits in A. summ infected calves.  相似文献

The isolation of Oesophagostomum dentatum from pig is described, and several morphological characteristics of the parasite and some special features about its life-cycle are illustrated and discussed.Ninety-eight female worms with a tail-length of about 350 μm were isolated from one sow. The eggs were separated from the uterus and transferred to a culture medium. The infective larvae obtained were fed to a Vietnamese miniature pig. As a control for the species isolation, a Belgian landrace pig was infected with 2000 L3, which had originated from the Vietnamese pig. Eight weeks later the control animal was slaughtered; the tail-length of 200 female worms isolated was measured. The measurements showed a symmetric and normal distribution with a mean of 313.23 μm.Five three-month-old sows were each infected orally with 50 000 O. dentatum larvae. The prepatent period was found to be between 26 and 28 days. The egg-counts fluctuated widely and after killing the pigs only a small number of mature worms were found to be present. Histological examination of the mucosa of the intestines did not show a prolonged histotropic phase. Two hundred worms from each pig were measured, and the lengths were 8.72 to 10.83 mm for females, 7.70 to 8.74 mm for males. The length of the spicules ranged from 1004 to 1043 μm, and the tail-length of the females from 299 to 18.75 μm. Only the measurements of the tail-length belonged to a symmetric population. The adult worms had a buccal capsule with parallel sides and a club-shaped oesophagus.  相似文献

Summary

The activity of albendazole against gastrointestinal helminths in naturally infected calves in the Netherlands was tested. The calves were in their fist grazing season and kept in two groups of ten. One of these groups was grazed alternately with sheep. Five out of each group were drenched with albendazole (7.5 mg/kg) on the day they were housed (November 1). Before and 2, 14, and 28 days after treatment individual faecal samples were taken from all calves and larval cultures were made. Ten calves, six treated and four untreated, were killed for post mortem studies 14 days after treatment The remaining calves were slaughtered 14 days later.

The drug was highly effective in reducing the egg output, measured as the number of larvae cultured per gram of faeces. Compared with the untreated calves, the reduction was more than 99% two days after treatment, 100% at 14 days, and 99% after 28 days. It was shown that egg output 28 days after treatment came from worms which had developed from arrested larvae of Ostertagia ostertagi that had survived treatment. Post mortem results showed an efficacy of 100% against adult O. ostertagi, of almost 100% against Trichostrongylus axei, and 100% against adult and larval Cooperia oncophora.

Twenty‐eight days after treatment, the reduction of arrested early fourth stages of O. ostertagi was 85% in comparison with the untreated calves. Apparently less effect was found against Trichuris ovis at the given dose rate.  相似文献

The aim of this study was to examine whether 2.16% hypertonic saline solution (HSS) is useful for the treatment of diarrheic calves with hyponatremia. Eleven of 13 female Holstein calves exhibiting moderate diarrhea and hyponatremia received 1,250 ml of 2.16% HSS over 15 min regardless of body weight. The remaining two calves that were unable to stand and had severe hyponatremia received 2,500 ml of 2.16% HSS intravenously over 30 min. As a result, hyponatremia in all diarrheic calves was significantly improved by the administration of 2.16% HSS from 122.2 ± 7.0 mEq/l at pre to 134.8 ± 3.7 mEq/l at post, which was above the threshold of 132 mEq/l for hyponatremia. Therefore, 2.16% HSS may be useful for hyponatremia in calves with diarrhea.  相似文献

This study examined known or possible virulence-associated genes in type A Clostridium perfringens from cases of both bovine clostridial abomasitis (BCA) and jejunal hemorrhage syndrome (JHS) and compared these to isolates from calves that were healthy or had undifferentiated diarrheal illness. A real-time polymerase chain reaction (PCR) assay was used to genotype the 218 C. perfringens isolates. Isolates were sourced from healthy and diarrheic young and mature cattle (n = 191), from calves with confirmed or suspected BCA (n = 22), and from mature cattle with JHS (n = 5). Of 216 isolates (96%), 208 were positive for the cpa gene and 13% (29/218) were positive for atypical cpb2. Three of 8 (37.5%) confirmed BCA isolates, 2 of 13 (15.4%) suspected BCA isolates, and no JHS isolates tested positive for atypical cpb2. As all isolates were negative for cpb, cpb2, cpe, etx, netB, and tpeL, the results of the present study do not support a role for these genes in BCA or JHS. A subset of unique genes identified in 1 bovine clostridial abomasitis isolate (F262), for which a genome sequence is available, was searched for in 8 BCA isolates by PCR. None of the 10 genes was consistently present in all or even in a majority of BCA isolates. Many of these genes were also variably and inconsistently present in type A isolates from calves that did not have BCA. Although a virulence signature to aid in the diagnosis of BCA caused by C. perfringens type A was not identified, further work may discover a gene or group of genes that would constitute such a signature.  相似文献

The ELISA technique was found to be very useful for the detection and monitoring of anti-Cooperia and anti-Ostertagia antibodies in calves. Six differens antigens were used; saline extracts from third- and fourth-stage larvae and from adult worms of both genera.Some degree of genus specificity was found when using L4 or adult antigens but not when L3 antigens were used. Stage-specificity could be observed for Cooperia L4 antigen for a limited period after primary single infection. These findings were supported by the results of immuno-electrophoresis.  相似文献

The impact of the laboratory induced Schistosoma mansoni with decreased PZQ sensitivity on the biological performance of its different developmental stages and the concomitant structural changes of adult worms’ total proteins were investigated. PZQ exposed snails showed stoppage of cercarial shedding for eight weeks followed by progressive significant reduction of cercarial production along four successive weeks. In the vertebrate host, in comparison to Schistosoma mansoni susceptible isolate, inoculated cercariae with decreased PZQ sensitivity led to an evident decrease in male to female ratio associated with significant reduction in tissue egg counts and significant increase in dead egg percentage. Significant reduction in the fecundity was also determined. Interestingly, eggs from adult worms with decreased PZQ sensitivity showed two unique features as they found to be smaller and more spherical in addition to the observation of hourglass shaped miracidium in about 10% of the detected mature eggs. Proteomic analysis of adult worms with decreased sensitivity to PZQ using mass spectrometry revealed up-regulation of Ca2+ ATPase 2 and Hsp70. This study can point to the increase incidence of the neuroschistosomiasis due to the small size eggs of Schistosoma mansoni with reduced PZQ sensitivity. These worms can also impact the epidemiology in the field. The study can also provide help to elucidate underlying potential molecular mechanisms of resistance that could lead to possible strategies to reverse drug resistance.  相似文献

This study aimed to evaluate the sampling method for the detection of Ascaris suum larval DNA in chicken livers using real-time PCR. Chickens were inoculated with A. suum eggs of a single dose (Group A) or repeatedly low doses (Group B). White spots (WSs) were continuously observed on liver from day 3 after the last infection in Group B and day 14 in Group A. In Group A, larval DNA was detected in WS lesions (78.6%) at a significantly higher rate than in the remaining tissue samples (31.3%). In conclusion, applying WS lesions to the assay improved the detection rate of A. suum DNA in chicken livers, especially in the case of a single infection.  相似文献

Ivermectin, either as an injectable or topical formulation, is used widely to control parasites in New Zealand cattle. These formulations have been well documented as having high efficacy against the major nematode species which infect tattle(1)(2)(3)(4), although the efficacy against adult Cooperia oncophora is reported to be 95%(5). This is considered to be one of the doselimiting species. There have been two reports of lower than expected efficacy of the topical formulation against Cooperia spp. in cattle. In a slaughter trial, Bisset et al. (6) found that ivermectin applied topically at the recommended dose rate of 500 𝛍g/kg failed to achieve a statistically significant reduction in the burden of Cooperia spp. worms compared with untreated calves. The geometric mean of adult Cooperia spp. worms present in the treated calves was 5788 compared with 7524 present in the untreated calves. This reduced efficacy was not attributed to acquired anthelmintic resistance because ivermectin had not been used on the farm previously. In Australia, Eagleson and Allertonc(7) found in controlled field trials that ivermectin applied topically at 500 𝛍g/kg ranged in efficiency in reducing faecal egg counts from 80.7% to 100% ; four out of 13 trials achieving less than 90% and a further four were in the range of 90–95% reduction. Larvae obtained from the faeces of ivermectin-treated cattle on days 11–14 after treatment were predominantly Cooperia spp.  相似文献
