Abstract

Biofertilizers are an alternative to mineral fertilizers for increasing soil productivity and plant growth in sustainable agriculture. The objective of this study was to evaluate possible effects of three mineral fertilizers and four plant growth promoting rhizobacteria (PGPR) strains as biofertilizer on soil properties and seedling growth of barley (Hordeum vulgare) at three different soil bulk densities, and in three harvest periods. The application treatments included the control (without bacteria inoculation and mineral fertilizers), mineral fertilizers (N, NP and P) and plant growth promoting rhizobacteria species (Bacillus licheniformis RC04, Paenibacillus polymyxa RC05, Pseudomonas putida RC06, and Bacillus OSU-142) in sterilized soil. The PGPR, fungi, seedling growth, soil pH, organic matter content, available P and mineral nitrogen were determined in soil compacted artificially to three bulk density levels (1.1, 1.25 and 1.40 Mg m?3) at 15, 30, and 45 days of plant harvest. The results showed that all the inoculated bacteria contributed to the amount of mineral nitrogen. Seed inoculation significantly increased the count of bacteria and fungi. Data suggest that seed inoculation of barley with PGPR strains tested increased root weight by 9–12.2%, and shoot weight by 29.7–43.3% compared with control. The N, NP and P application, however, increased root weight up to 18.2, 25.0 and 7.4% and shoot weight by 31.6, 43.4 and 26.4%, respectively. Our data show that PGPR stimulate barley growth and could be used as an alternative to chemical fertilizer. Soil compaction hampers the beneficial plant growth promoting properties of PGPR and should be avoided.

The interactions between soil P availability and mycorrhizal fungi could potentially impact the activity of soil microorganisms and enzymes involved in nutrient turnover and cycling, and subsequent plant growth. However, much remains to be known of the possible interactions among phosphorus availability and mycorrhizal fungi in the rhizosphere of berseem clover (Trifolium alexandrinum L.) grown in calcareous soils deficient in available P. The primary purpose of this study was to look at the interaction between P availability and an arbuscular mycorrhizal (AM) fungus (Glomus intraradices) on the growth of berseem clover and on soil microbial activity associated with plant growth. Berseem clover was grown in P unfertilized soil (−P) and P fertilized soil (+P), inoculated (+M) and non-inoculated (−M) with the mycorrhizal fungus for 70 days under greenhouse conditions. We found an increased biomass production of shoot and root for AM fungus-inoculated berseem relative to uninoculated berseem grown at low P levels. AM fungus inoculation led to an improvement of P and N uptake. Soil respiration (SR) responded positively to P addition, but negatively to AM fungus inoculation, suggesting that P limitation may be responsible for stimulating effects on microbial activity by P fertilization. Results showed decreases in microbial respiration and biomass C in mycorrhizal treatments, implying that reduced availability of C may account for the suppressive effects of AM fungus inoculation on microbial activity. However, both AM fungus inoculation and P fertilization affected neither substrate-induced respiration (SIR) nor microbial metabolic quotients (qCO2). So, both P and C availability may concurrently limit the microbial activity in these calcareous P-fixing soils. On the contrary, the activities of alkaline phosphatase (ALP) and acid phosphatase (ACP) enzymes responded negatively to P addition, but positively to AM fungus inoculation, indicating that AM fungus may only contribute to plant P nutrition without a significant contribution from the total microbial activity in the rhizosphere. Therefore, the contrasting effects of P and AM fungus on the soil microbial activity and biomass C and enzymes may have a positive or negative feedback to C dynamics and decomposition, and subsequently to nutrient cycling in these calcareous soils. In conclusion, soil microbial activity depended on the addition of P and/or the presence of AM fungus, which could affect either P or C availability.

Introducing specific microorganisms into the soil ecological system is an important strategy for improving nutrient use efficiency. Two pot experiments were conducted in the greenhouse from December 3, 2012 to January 25, 2013 (Experiment 1) and March 11 to April 23, 2013 (Experiment 2) to evaluate the effect of nitrogen (N) source and inoculation with plant growth-promoting rhizobacteria (PGPR) on plant growth and N and phosphorus (P) uptake in tomato (Lycopersicon esculentum Mill.) grown on calcareous soils from South Florida, USA. Treatments included urea, controlled release urea (a controlled release fertilizer, CRF) each at low and high N rates and with or without inoculation of PGPR. A mixture of PGPR strains Bacillus amyloliquefaciens IN937a and Bacillus pumilus T4 was applied to the soil during growing periods of tomato. Treatments with PGPR inoculation increased plant height compared to treatments without PGPR in both experiments. Inoculation with PGPR increased shoot dry weight and shoot N uptake for the same N rate and N source. In both experiments, only at high N rate, CRF and urea treatments with PGPR had significantly (P < 0.05) greater shoot biomass than those without PGPR. Only at high N rate, CRF treatment with PGPR significantly increased shoot N uptake by 39.0% and 10.3% compared to that without PGPR in Experiments 1 and 2, respectively. Meanwhile, presence of PGPR in the soil increased shoot P uptake for all treatments in Experiment 1 and for most treatments in Experiment 2. In Experiment 1, only at low N rate, CRF treatment with PGPR significantly increased shoot P uptake compared with that without PGPR. In Experiment 2, a significant increase in shoot P uptake by inoculation of PGPR was only observed in CRF treatment at high N rate. Results from this study indicate that inoculation with PGPR may increase plant growth and N and P uptake by tomato grown on calcareous soils. However, the effect of PGPR varied and was influenced by many factors such as N source, N rate, and soil fertility. Further investigations are warranted to confirm the effect of PGPR under different soil conditions.

Summary The relationship between the microbial activity in the soil and the effect of seed inoculation with the rhizopseudomonad strain 7NSK2 was evaluated in a series of pot experiments under greenhouse conditions. The microbial activity in plain soil, as measured by the respiratory activity, was significantly increased by the growth of the plants. Both the respiration rate of the microorganisms and the density of the bacteria and fungi in the bulk soil increased with increasing duration of the plant growth. Upon repeated short-term growth of plants on the same soil, a similar stimulation was noticed. The effect of seed inoculation on the growth of the maize cultivar Beaupré and the barley cultivar Iban was most pronounced in the microbiologically more active soils. The results suggest that the increase of the plant growth by seed inoculation is probably due to the inhibition of deleterious root microorganisms.

Field experiments were conducted to evaluate the impact of seed and seedling inoculation by plant growth promoting rhizobacteria (PGPR) on nitrogen use efficiency, growth, yield, and chemical composition of cabbage at varying levels of nitrogen (N). Data revealed that N alone or in combination with PGPR either as seed or seedling inoculation significantly improved growth, yield, and nutrients of cabbage. PGPR inoculations were more efficient than non-inoculated controls in terms of yield and yield parameters. This study showed that seed and seedling inoculation increased yield and yield parameters as well as chlorophyll reading value and stomatal conductance versus controls. Applications of mineral fertilizers with microbial applications save 25% of mineral fertilizers yet give 33% more yield versus full doses of mineral application without microbial applications. Both seed and seedling treatments increased the nitrogen use efficiency (NUE) rates by 45, 53, 58, 45, and 40%, and 49, 59, 68, 69, 60, and 55%, respectively.

Plant growth–promoting rhizobacteria (PGPR) have been reported to stimulate the growth and yield of grain crops, particularly when nutrient supply is poor. However, the mechanisms underlying stimulation of plant growth may vary depending not only on growth conditions and crop management but also on plant and bacterial species. The present study assessed the effect of an inoculation with single or multiple PGPR strains on phosphorus (P)‐solubilization processes in the soil and on grain yield in wheat. Single inoculation with Bacillus subtilis OSU‐142, Bacillus megaterium M3, or Azospirillum brasilense Sp245 increased grain yield by 24%, 19%, and 19%, respectively, while a mixed inoculation with OSU‐142, M3, and Sp245 increased grain yield by 33% relative to noninoculated plants. Single inoculations with Paenibacillus polymyxa RC05 or Bacillus megaterium RC07 were less effective. Single or mixed treatments with OSU‐142, M3, and Sp245 increased the concentrations of the labile and moderately labile P fractions in rhizosphere soil. The growth‐stimulating effect of OSU‐142, M3, and Sp245 was also reflected by higher P concentrations in most plant organs. Among all inocula tested, the highest plant P acquisition was obtained in the presence of M3 and accompanied by the highest microbial P levels and the highest phosphatase activities in the rhizosphere soil. In conclusion, seed inoculation with mixed PGPR strains may effectively substitute for a part of P‐fertilizer application in extensive wheat production, and in particular M3 appears to improve the solubilization of inorganic soil P.

The influence of plant-growth-promoting rhizobacteria (PGPR) and spruce seedlings on the composition and activity of forest soil microbial communities was studied in a microcosm experiment in which sterile, sand-filled 25mm×150mm glass tubes were treated with a forest soil suspension containing Bacillus or Pseudomonas PGPR and 2-week-old spruce seedlings. Eighteen weeks after treatments were established, bacterial, actinomycete and fungal population sizes were determined by dilution plating, as were seedling dry weights and soil carbon substrate utilization profiles using Biolog plates. PGPR inoculation had little influence on the population sizes of actinomycetes or fungi. However, significant effects were detected on the total bacterial population size, primarily in microcosms without seedlings. Euclidean distances between treatments plotted on two dimensions by multidimensional scaling showed that the introduction of PGPR strains changed the type of microbial community, particularly when inoculated into soil without seedlings. Significant changes were also detected in one soil type in the presence of seedlings. Our results suggest that the type of soil community and the presence of seedlings are significant factors influencing the responses of soil communities to bacterial inoculation, and that for some soil communities, the presence of seedlings may mitigate perturbations caused by the introduction of PGPR. In order to study the effect of plant growth promoting rhizobacteria (PGPR), Azolla compost and Azolla biochar on some soil quality indicators and rosemary growth, a greenhouse experiment was conducted in a completely randomized design with six replications. Treatments consisted of T1 (control), T2 (1% Azolla compost), T3 (1% Azolla biochar), T4 (PGPR (P. fluorescens), T5 (1% compost + PGPR) and T6 (1% biochar + PGPR). Rosemary growth parameters and nutrients concentration increased in all treatments compared to control. Treatments increased soil nutrient concentrations, soil microbial respiration (SMR) and microbial biomass C (MBC) but decreased soil metabolic quotient (qCO2) compared to control treatment. A significant enhancement in rosemary growth occurred due to the improved soil quality as a result of organic fertilizers application, particularly by co-application of P. fluorescens and compost or biochar of Azolla.

Plant growth-promoting rhizobacteria (PGPR) have been widely recognized as an important agent, especially as a biofertilizer,in agricultural systems. The objectives of this study were to select efective PGPR for Chinese kale (Brassica oleracea var.alboglabra) cultivation and to investigate the efect of their inoculation on indigenous microbial community structure. The Bacillus sp.SUT1 and Pseudomonas sp.SUT19 were selected for determining the efficiency in promoting Chinese kale growth in both pot and field experiments. In the field experiment,PGPR amended with compost gave the highest yields among all treatments. The Chinese kale growth promotion may be directly afected by PGPR inoculation. The changes of microbial community structure in the rhizosphere of Chinese kale following PGPR inoculation were examined by denaturing gradient gel electrophoresis (DGGE) and principal coordinate analysis. The DGGE fingerprints of 16S rDNA amplified from total community DNA in the rhizosphere confirmed that our isolates were established in the rhizosphere throughout this study. The microbial community structures were slightly diferent among all the treatments, and the major changes depended on stages of plant growth.DNA sequencing of excised DGGE bands showed that the dominant species in microbial community structure in the rhizosphere were not mainly interfered by PGPR,but strongly influenced by plant development. The microbial diversity as revealed by diversity indices was not diferent between the PGPR-inoculated and uninoculated treatments. In addition, the rhizosphere soil had more influence on eubacterial diversity, whereas it did not afect archaebacterial and fungal diversities.

There is an increasing concern that the continuous use of chemical fertilizers might lead to harmful effects on soil ecosystem. Accordingly, a biocompatible approach involving inoculation of beneficial microorganisms is presented to promote plant growth and simultaneously minimize the negative effect of chemical fertilizers. In this study, Rhodopseudomonas palustris, a plant growth-promoting rhizobacterium (PGPR), was inoculated into both fertilized and unfertilized soils to assess its influence on Stevia rebaudiana plant growth and microbial community in rhizosphere soils in a 122-d field experiment. Soil enzyme assays (dehydrogenase, urease, invertase, and phosphomonoesterase), real-time quantitative polymerase chain reaction (RT-qPCR), and a high-throughput sequencing technique were employed to determine the microbial activity and characterize the bacterial community. Results showed that the R. palustris inoculation did not significantly influence Stevia yields and root biomass in either the fertilized or unfertilized soil. Chemical fertilization had strong negative effects on soil bacterial community properties, especially on dehydrogenase and urease activities. However, R. palustris inoculation counteracted the effect of chemical fertilizer on dehydrogenase and urease activities, and increased the abundances of some bacterial lineages (including Bacteroidia, Nitrospirae, Planctomycetacia, Myxococcales, and Legionellales). In contrast, inoculation into the unfertilized soil did not significantly change the soil enzyme activities or the soil bacterial community structure. For both the fertilized and unfertilized soils, R. palustris inoculation decreased the relative abundances of some bacterial lineages possessing photosynthetic ability, such as Cyanobacteria, Rhodobacter, Sphingomonadales, and Burkholderiales. Taken together, our observations stress the potential utilization of R. palustris as PGPR in agriculture, which might further ameliorate the soil microbial properties in the long run.

Responses of the tropical legumes, Desmodium heterocarpon and Galactia elliottii, to inoculation with rhizobia and fertilization with N and P were evaluated in greenhouse experiments. Although inoculation with rhizobia was effective in some instances, improvements in plant growth were not obtained in the presence of naturally occurring microbial populations on the Florida Spodosol soil. Phosphorus at 50 kg P/ha increased herbage and root growth of G. elliottii, but not of D. heterocarpon. Seedling growth responses to N indicate that high rates of N fertilization (in combination with heavy grazing) might be effectively used to enhance seedling vigor of D. heterocarpon oversown on bahiagrass (Paspalum notatum) pastures.

The effects of inoculating field peas (Pisum sativum L.) with Rhizobium leguminosarum and field beans (Phaseolus vulgaris L.) with R. phaseoli, alone or in combination with Pseudomonas syringae R25 and/or P. putida R105, were assessed under gnotobiotic conditions in growth pouches and in potted soil in a growth chamber. Inoculation of peas with P. syringae R25 or P. putida R105 alone had no effect on plant growth in pouches. In soil, however, the isolate R25 inhibited nitrogenase activity (as assessed by acetylene reduction assay) of nodules formed by indigenous rhizobia; strain R105 stimulated pea seedling emergence and nodulation. P. syringae R25 inhibited the growth of beans in either plant-growth system. P. putida R105, however, had no effects on beans in pouches, but reduced plant root biomass and nodulation by indigenous rhizobia in soil. Coinoculation of pea seeds with R. leguminosarum and either of the pseudomonads significantly (P<0.01) increased shoot, root, and total plant weight in growth pouches, but had no effect in soil. Co-inoculation of field beans with R. phaseoli and P. putida R105 had no effects on plant biomass in growth pouches or in soil, but the number of nodules and the acetylene reduction activity was significantly (P<0.01) increased in the soil. In contrast, co-inoculation of beans with rhizobia and P. syringae R25 had severe, deleterious effects on seedling mergence, plant biomass, and nodulation in soil and growth pouches. Isolate R25 was responsible for the deleterious effects observed. Although plant growth-promoting rhizobacteria may interact synergistically with root-nodulating rhizobia, the PGPR selected for one crop should be assessed for potential hazardous effects on other crops before being used as inoculants.

Sewage sludge (SS) can be used as an alternative fertilizer in agriculture. It is normally broadcasted and plowed into soil, but it is not clear if it has a potential as a placement fertilizer. A rhizobox experiment was conducted to investigate the placement effect of SS and mineral nitrogen (N) fertilizer on shoot and root growth as well as nutrient uptake of spring wheat (Triticum aestivum L.). The treatments included localized SS, mixed SS, localized SS and ammonium, localized ammonium, and a control without addition of SS and ammonium to examine the effect of SS placement and, further, if ammonium co‐localization would enhance the placement effect. The results show that SS fertilization improved soil N and P availability, which significantly increased plant N and P uptake and enhanced shoot growth, while root length was significantly reduced compared to the control. Localized SS increased root proliferation in the placement region, resulting in enhanced uptake of P from the SS patch compared to homogenous application. However, co‐localized application of ammonium with SS significantly depressed plant shoot and root growth. Localized ammonium markedly restricted root proliferation in the placement region and reduced soil pH in both bulk soil and placement region, contributing to decreased nutrient uptake and plant growth.

Plants link atmospheric and soil carbon pools through CO2 fixation, carbon translocation, respiration and rhizodeposition. Within soil, microbial communities both mediate carbon-sequestration and return to the atmosphere through respiration. The balance of microbial use of plant-derived and soil organic matter (SOM) carbon sources and the influence of plant-derived inputs on microbial activity are key determinants of soil carbon-balance, but are difficult to quantify. In this study we applied continuous 13C-labelling to soil-grown Lolium perenne, imposing atmospheric CO2 concentrations and nutrient additions as experimental treatments. The relative use of plant- and SOM-carbon by microbial communities was quantified by compound-specific 13C-analysis of phospholipid fatty acids (PLFAs). An isotopic mass-balance approach was applied to partition the substrate sources to soil respiration (i.e. plant- and SOM-derived), allowing direct quantification of SOM-mineralisation. Increased CO2 concentration and nutrient amendment each increased plant growth and rhizodeposition, but did not greatly alter microbial substrate use in soil. However, the increased root growth and rhizosphere volume with elevated CO2 and nutrient amendment resulted in increased rates of SOM-mineralisation per experimental unit. As rhizosphere microbial communities utilise both plant- and SOM C-sources, the results demonstrate that plant-induced priming of SOM-mineralisation can be driven by factors increasing plant growth. That the balance of microbial C-use was not affected on a specific basis may suggest that the treatments did not affect soil C-balance in this study.

Inoculants are of great importance in sustainable and/or organic agriculture. In the present study, plant growth of barley (Hordeum vulgare) has been studied in sterile soil inoculated with four plant growth-promoting bacteria and mineral fertilizers at three different soil bulk densities and in three harvests of plants. Three bacterial species were isolated from the rhizosphere of barley and wheat. These bacteria fixed N2, dissolved P and significantly increased growth of barley seedlings. Available phosphate in soil was significantly increased by seed inoculation of Bacillus M-13 and Bacillus RC01. Total culturable bacteria, fungi and P-solubilizing bacteria count increased with time. Data suggest that seed inoculation of barley with Bacillus RC01, Bacillus RC02, Bacillus RC03 and Bacillus M-13 increased root weight by 16.7, 12.5, 8.9 and 12.5% as compared to the control (without bacteria inoculation and mineral fertilizers) and shoot weight by 34.7, 34.7, 28.6 and 32.7%, respectively. Bacterial inoculation gave increases of 20.3–25.7% over the control as compared with 18.9 and 35.1% total biomass weight increases by P and NP application. The concentration of N and P in soil was decreased by increasing soil compaction. In contrast to macronutrients, the concentration of Fe, Cu and Mn was lower in plants grown in the loosest soil. Soil compaction induced a limitation in root and shoot growth that was reflected by a decrease in the microbial population and activity. Our results show that bacterial population was stimulated by the decrease in soil bulk density. The results suggest that the N2-fixing and P-solubilizing bacterial strains tested have a potential on plant growth activity of barley.

A greenhouse study was conducted to evaluate the effects of plant growth-promoting rhizobacteria (PGPR) on root establishment and biomass production of corn (Zea mays L.) using three fertility sources (poultry litter (PL), biosolids, and urea). Applying PL significantly improved root morphological parameters and increased plant biomass at the V4, V6, and VT growth stages when compared to the other fertility sources. At the V4 stage, PGPR stimulated root growth and enhanced aboveground biomass with urea and PL, while no differences were observed with biosolids. At the V6 stage, PL, biosolids, and urea with PGPR significantly increased some growth parameters (e.g., plant height, leaf area, and root morphology). However, at the VT stage, PGPR’s influence on plant growth was minimal regardless of fertility source. Applying the fertility sources at 135 kg N ha?1 may have masked PGPR’s influence on corn growth as the plants reached their later vegetative growth stages.
