In vertebrates, chemosensitivity of nutrients occurs through the activation of taste receptors coupled with G-protein subunits, including α-transducin (Gαtran) and α-gustducin (Gαgust). This study was aimed at characterising the cells expressing Gαtran immunoreactivity throughout the mucosa of the sea bass gastrointestinal tract. Gαtran immunoreactive cells were mainly found in the stomach, and a lower number of immunopositive cells were detected in the intestine. Some Gαtran immunoreactive cells in the stomach contained Gαgust immunoreactivity. Gastric Gαtran immunoreactive cells co-expressed ghrelin, obestatin and 5-hydroxytryptamine immunoreactivity. In contrast, Gαtran immunopositive cells did not contain somatostatin, gastrin/cholecystokinin, glucagon-like peptide-1, substance P or calcitonin gene-related peptide immunoreactivity in any investigated segments of the sea bass gastrointestinal tract. Specificity of Gαtran and Gαgust antisera was determined by Western blot analysis, which identified two bands at the theoretical molecular weight of ~45 and ~40 kDa, respectively, in sea bass gut tissue as well as in positive tissue, and by immunoblocking with the respective peptide, which prevented immunostaining. The results of the present study provide a molecular and morphological basis for a role of taste-related molecules in chemosensing in the sea bass gastrointestinal tract. 相似文献
Under experimental challenges, the gastrointestinal (GI) tract of fish has been proposed as an infectious route of several pathogenic bacteria. Is this also the case for diseased fishponds? A field research was conducted to verify this hypothesis. A crucian carp (Carassius carassius) reared fishpond with motile Aeromonas septicaemia outbreak was sampled in this study. A total of 62 strains of Aeromonas hydrophila were isolated and identified. The clonal relationship among these strains was determined by sequencing the gyrB gene, ERIC‐PCR, RAPD‐PCR, and the presence of seven virulence genes. Strains with identical genotypes were further confirmed as the same clone by multilocus sequence typing analysis. Experimental infection assays were also conducted in zebrafish (Danio rerio). The results show that the same clone strains identical to those in the blood of diseased fish existed in the intestinal digesta of diseased and uninfected fish. Regardless of their origins, all these strains were highly pathogenic to zebrafish. The result indicates that pathogenic strains of A. hydrophila had existed in the GI tract of fish before the infection occurred. In this study, β-haemolysin gene (AHTPS30HEM) of Aeromonas hydrophila was cloned from diseased fish in mainland China. AHTPS30HEM gene (AB021152) resulted in a 1589 bp fragment which covers the open reading frame (ORF) in region 5–1486 coding for 493 amino acids. Multiple alignment of AHTPS30HEM with other β-haemolysin amino acid sequences showed 18 amino acid substitutions between AHTPS30HEM and A. hydrophila β-haemolysin. Although the ORF sizes between AHTPS30HEM and Aeromonas species are different, four cysteins and four potential N-glycosylation sites were conserved. To identify the β-haemolysin-producing virulent or pathogenic A. hydrophila, a specific PCR to amplify 208 bp target DNA of β-haemolysin gene was established. Twenty strains containing pathogenic A. hydrophila, Aeromonas sobria, Vibrio anguillarum, Cytophaga columnaris, Pseudomonas fluorescens, and Yersinia yuckeri were investigated by PCR. Based on the cloned β-haemolysin sequences, the specific PCR method for identification of the β-haemolysin gene of A. hydrophila was established, and surveyed on those samples. This study tested the hypothesis that the renal reabsorption of urea occurs via the glucose transport pathway in the freshwater rainbow trout (Oncorhynchus mykiss). The relationship between glucose transport and urea transport was examined by experimentally elevating the rate of renal glucose reabsorption via infusion of the fish with exogenous glucose, and by inactivating the glucose transporters via the administration of phlorizin. Under all treatments, urea was reabsorbed against a concentration gradient, with plasma levels of urea being higher than urine levels. Glucose was almost completely reabsorbed (88%) whereas urea was reabsorbed less efficiently (47%) but to a greater extent than water (22%). Glucose and urea reabsorption were both found to be correlated with Na+ and Cl– reabsorption, though the latter were 20 fold and 200–300 fold higher than glucose and urea transport rates, respectively. Glucose infusions greatly increased glucose reabsorption but urea reabsorption was unaffected. Phlorizin treatment completely blocked glucose reabsorption, but urea reabsorption was again unaffected. We conclude that there is no relationship between glucose and urea handling in the trout kidney, thus disproving the hypothesis. 相似文献
Abstract This paper describes a pilot culture system for the production of on‐grown Artemia in freshwater ornamental fish farms. The system had 21 culture units, each consisting essentially of three components: an oval‐shaped raceway, an air‐water lift system and two waste collectors. Using artificial seawater at 20 ppt for culture and at a mean production rate of 3 kg/m3 of water in a 12‐day cycle, the system had a production capacity of 8 metric tons of on‐grown Artemia a year. Biochemical analyses were preformed to evaluate the nutritional value of the on‐grown Artemia against three conventional live feeds, viz. live Artemia nauplii, live Moina and frozen bloodworms. Cost‐benefit analysis showed that with a capital investment of US$ 82,000 and an annual cost of production of US$ 81,000, the system achieved a high internal rate of return of 88% over a 10‐year period and a short payback period of 1.23 years. The availability of on‐grown Artemia would not only offer farmers and exporters a better alternative live food organism for feeding to their fish, but more importantly the possibility of enhancing the fish performance and quality through bioencapsulation. 相似文献
There has been a worldwide increase in consumption of fish from open waters and aquaculture during the last decades. In 2013, the Czech Republic was the sixth largest producer of fish within the EU, with annual production of 20,135 tonnes. However, during the period 2010–2015 import of fishery products to the Czech Republic increased from 39,098 to 43,399 tonnes. The monetary value of import increased by 81 % from 90.2 million EUR in 2010 to 169.9 million EUR in 2015. In 2010, the major imported fishery product was frozen fillets of sutchi catfish (Pangasianodon hypophthalmus) from Vietnam (8872 tonnes). In 2015, the most imported fishery product was whole chilled salmon (Salmo salar) from Norway (6897 tonnes). During 2010–2015, the export of fishery products increased from 18,167 to 22,134 tonnes, which is an increase by 21.8 %. The value of export has increased by 80 % from 54.5 million EUR in 2010 to 98.1 million EUR in 2015. The major exported fish was live common carp (Cyprinus carpio) at volume of 7545–9075 tonnes. Biological invasions are a major driver of global environmental change as invasive non-native species can exert severe environmental impacts on invaded ecosystems. Estuaries are especially vulnerable to biological invasions, which in highly urbanised areas are further facilitated by introduction pathways linked to commercial activities. This study provides a risk screening of non-native invasive species for the highly urbanised River Neretva Estuary (Eastern Adriatic Sea: Croatia and Bosnia–Herzegovina). In total, 12 species of non-native aquatic organisms were identified and screened for their invasiveness with the Aquatic Species Invasiveness Screening Kit. Of these species, eight were classified as carrying a high risk of invasiveness under current climate conditions and nine under future climate conditions. Amongst the high-risk species, blue crab Callinectes sapidus has already caused impacts in the risk assessment area, where it also represents an important economic resource. The “horizon” species Pacific oyster Crassostrea gigas and Manila clam Ruditapes philippinarum also carried a high risk of invasiveness, which for the latter species applied under predicted global warming. The present findings will contribute towards preventative management and control measures for the conservation of the natural ecosystem of the River Neretva Estuary whilst accounting for aquatic farming demands. 相似文献
Marine heatwaves are increasingly affecting marine ecosystems, with cascading impacts on coastal economies, communities, and food systems. Studies of heatwaves provide crucial insights into potential ecosystem shifts under future climate change and put fisheries social-ecological systems through “stress tests” that expose both vulnerabilities and resilience. The 2014–16 Northeast Pacific heatwave was the strongest and longest marine heatwave on record and resulted in profound ecological changes that impacted fisheries, fisheries management, and human livelihoods. Here, we synthesize the impacts of the 2014–2016 marine heatwave on US and Canada West Coast fisheries and extract key lessons for preparing global fisheries science, management, and industries for the future. We set the stage with a brief review of the impacts of the heatwave on marine ecosystems and the first systematic analysis of the economic impacts of these changes on commercial and recreational fisheries. We then examine ten key case studies that provide instructive examples of the complex and surprising challenges that heatwaves pose to fisheries social-ecological systems. These reveal important insights into improving the resilience of monitoring and management and increasing adaptive capacity to future stressors. Key recommendations include: (1) expanding monitoring to enhance mechanistic understanding, provide early warning signals, and improve predictions of impacts; (2) increasing the flexibility, adaptiveness, and inclusiveness of management where possible; (3) using simulation testing to help guide management decisions; and (4) enhancing the adaptive capacity of fishing communities by promoting engagement, flexibility, experimentation, and failsafes. The distribution and movement between habitats with growth of the hard clam Meretrix lusoria were examined by comparing the size–frequency distributions among 17 sites in the Shirakawa–Midorikawa Estuary of the Ariake Sea. This clam is a commercially important bivalve in Japan, and its annual catch is largest in the Ariake Sea. At the study sites, new recruits were abundant at river mouths, and density was strongly affected by the distance from coastal tidal rivers and streams. In contrast, adults were widely distributed on tidal flats, although density was low at many sites. At marine site Oda, the density of adults was higher than that of new recruits at all sites and higher than that of juveniles at most sites. These results indicate that this species moves from riverine sites to marine sites with growth. Moreover, the observed low density of adult clams may indicate overfishing. The active movement of M. lusoria across the fishing grounds complicates the sustainable resource management of this species. Therefore, further studies are needed for effective resource management of M. lusoria. 相似文献
1. The intensity of marine activities in the exclusive economic zone (EEZ) of British Columbia, Canada, was quantified.
2. Humans use the ocean for a multitude of purposes, many of which have a direct impact on marine life and habitat. Yet such uses are seldom assessed in an integrated fashion.
3. Using a GIS approach, spatial information for 39 marine activities was mapped, including commercial and recreational fishing areas, transportation and infrastructure uses, and terrestrial activities along the coast of British Columbia.
4. A relative scale was used to rank both the impact of marine activities and the extent of stressors beyond the location of occurrence. Limited information on the latter led to the application of three ranges of buffer distances to the data (0–1 km, 0–5 km, and 0–25 km).
5. The most conservative estimate (?1 km buffers) indicates at least 83% of the continental shelf and slope of British Columbia is currently being used by humans. The largest buffer assumption shows 98% of the continental shelf and slope being affected by stressors from anthropogenic activities.
6. This analysis provides a baseline for assessing future changes in the state of British Columbia's marine environment, and could assist in identifying areas of conservation potential.
The matrinch? Brycon amazonicus, a commercially important freshwater fish resource, has no heteromorphic sex chromosomes so far described. In the present study, we performed a screening of sex-associated DNA markers in this species, through the use of a random amplified polymorphic DNA (RAPD) assay and a genomic DNA restriction digestion analysis. DNA digestions evidenced no differences between sexes. Sixty-six random primers were used in pooled and individual DNA samples of males and females, and the analysis of the RAPD fingerprints revealed one female sex-associated band. Cloning and sequencing of this band led to the identification of two distinct DNA segments. While one of the isolated fragments showed a significant identity with a described protein gene (phosphatidylinositol glycan anchor biosynthesis, class W), the other fragment, composed of 535?bp, corresponds to a novel DNA marker. Further experiments were performed with this second DNA fragment in order to verify its sex-specificity. Data on dot blot hybridization, using total DNA of both sexes, confirmed its female-specificity in B. amazonicus. A primer set was designed based on its sequence data and used in PCR with DNA samples of this species, leading to diagnose the animals' sexes with a 100?% overall accuracy through a sequence characterized amplified region approach. No amplification results were found for two other species of the genus-B. orbignyanus and B. lundii. The obtained data can lead to the hypothesis that B. amazonicus may present heteromorphic sex chromosomes that should be in an early phase of differentiation. 相似文献
In this study, we investigated the bacterial communities in the shrimp and sea cucumber culture environment, including shrimp ponds (SP), sea cucumber ponds (SCP), mixed-culture ponds (MCP) and the effluent channel (EC) in Qingdao, China. Bacteria cultivation showed that the counts of heterotrophic, nitrate-reducing and sulfate-reducing bacteria in the sediment of SP were higher than that in the sediment of SCP and MCP, varying between 8.7 × 104 and 1.86 × 106, 2.1 × 104 and 1.1 × 105, and 9.3 × 101 and 1.1 × 104 CFU g?1, respectively. In contrast, the counts of ammonium-oxidizing and nitrifying bacteria in the sediment of SP was lower than that in the sediment of SCP and MCP. Denaturing gradient gel electrophoresis (DGGE) of 16S rDNA gene and dendrogram analyses showed that bacterial diversity in the mixed-culture environment was higher than that in the monocultures. The similarity of bacterial community between EC and SCP or MCP was higher than that between EC and SP. These results indicated that sea cucumber culture played a significant role in influencing the environmental bacterial communities that were composed mainly of Flavobacteriaceae (64.3%), Bacteriodetes (21%) and delta proteobacteria (14.7%), including the genera of Croceimarina, Lutibacter, Psychroserpens and so on. 1. Recently, the snails Hinia reticulatus, Cyclope neritea and Cerithium rupestre and the hermit crabs Diogenes pugilator and Clibanarius erythropus from warm European coastal waters have started to be commercially collected and traded.
2. The average number (± standard deviation) of specimens captured per hour, by collectors were 475±237 for H. reticulatus, 363±112 for C. neritea, 384±173 for C. rupestre, 302±121 for D. pugilator and 210±54 for C. erythropus.
3. All species survived well during transportation, were popular among aquarium hobbyists and attained high retail values, ranging from €0.90 to €4.00 per specimen.
4. The collection and trade of these novel commercial species can either become a sole economic activity or an additional one for traditional fishermen and aquaculturists.
5. Although no destructive collection techniques are employed, the lack of population biology studies and legislation regulating the capture of these species is a major problem in ensuring the sustainable use of these highly valuable marine resources.
6. These species are currently mainly collected from natural parks areas protected by Portuguese and European Union designations.
The understanding of the biological responses of copepods under crowding conditions contributes to establish their stable cultures at high densities for aquaculture industry, which are preferred live feeds for fish larvae. The present study investigated survivorship, fecundity, hatching success and respiration rate, of Acartia steueri raised under five densities, from 100 to 2,000 ind. L?1, to clarify the biological responses of the copepod under dense culture. There were no significant differences in survival, fecundity and hatching success among all density conditions, whereas the respiration rate at 2,000 ind. L?1 decreased by 80% as compared with a condition at 100 ind. L?1. The female copepods raised under a copepod density of >500 ind. L?1 probably invested a larger proportion of energy in reproduction in relation to total assimilated energy as compared with females under lower copepod densities. This change of energy allocation may allow A. steueri under high densities to maintain high fecundity. Acartia steueri might be a promising species for dense culture because its mortality and fecundity were independent of the effect of crowding, and the density‐dependent reduction in the metabolic rate might increase reproductive investment to maintain a constant rate of reproduction even under high densities. 相似文献

The food sources of the main economic animals and trophic levels of biotic communities from an optimized culture model (Rhopilema–Sinonovacula constricta–Paralichthys olivaceus–Penaeus chinensis) in a sea water pond in Donggang, Liaoning province were studied using the stable carbon and nitrogen isotopes technique. The results indicated that the values of δ13C range from (?27.28 ± 0.35) ‰ to (?16.65 ± 0.20) ‰ and the values of δ15N range from (3.68 ± 0.23) ‰ to (13.91 ± 0.26) ‰, both of which exhibited significant fluctuations. The δ13C values of P. chinensis, Macrobrachium mipponensis and P. olivaceus were comparatively higher than those of other aquaculture animals, and the δ15N values of P. olivaceus was also comparatively higher. The contribution to the food sources of aquaculture animals (Rhopilema, S. constricta, P. olivaceus and P. chinensis) was analyzed by using the IsoSource software. The results indicated that fish meat had the greatest contribution to Rhopilema, S. constricta and P. chinensis; P. chinensis had the greatest contribution to P. olivaceus, followed by the M. mipponensis. The trophic level of the biotic communities under that optimized culture model in a sea water pond was 3.54, in which P. olivaceus was in the fourth level (3.54); P. chinensis, Synechogobius hasta, Ablennes anastomella, M. mipponensis, Ditrema temmincki Bleeker, Chelon haematocheilus were in the third trophic levels; Rhopilema, Engraulis japonius, S. constricta and zooplankton (1.00) in the second trophic levels, suspended matters (0.15) including phytoplankton, bacteria, humus, etc., were in the primary trophic levels. 相似文献
In order to understand the processes affecting early survival of the Japanese anchovy Engraulis japonicus population in the Seto Inland Sea, we examined the monthly fluctuations in the abundance and survival rates from eggs to recruits (15-day-old larvae, 13?mm in standard length fish, defined as 1-month-old fish) during their main spawning season from 1980 to 2007. The abundance of the yolksac larvae positively correlated with the abundance of eggs, but the abundance of recruits did not significantly correlate with the abundance of yolksac larvae. These results imply that the survival rate during the egg stage is relatively stable, but that the rate during the larval stage is variable. Of the three most recent decades (1980s, 1990s and 2000s), the abundance of recruits was highest in the 1980s. The decrease in the abundance of recruits after the 1990s can be considered to have occurred through a decline in survival rate during the larval stage because there were no significant differences in egg abundance, yolksac larval abundance, and survival rates during the egg stage in the three decades of the study period. 相似文献
In the present study, the tilapia Oreochromis mossambicus were exposed to food deprivation for a period of 6 or 12 days and changes in the luteinizing hormone (LH) immunoreactivity in the proximal pars distalis (PPD) of the pituitary gland and the testicular activity were examined. Intensely immunoreactive LH content was noticed in the PPD of the pituitary gland in the initial controls, controls on days 6 and 12, and fasting fish on day 6, whereas the LH immunoreactivity was moderate or weak in fasting fish on day 12. In addition, although the mean gonadosomatic and hepatosomatic indices among different experimental groups did not show any statistically significant difference, the mean numbers of spermatogonia, spermatocytes, and spermatids were significantly lower in food-deprived fish on days 6 or 12 compared to those of controls. The inhibition of the spermatogenesis was accompanied by the presence of abundant spermatozoa in the lumen of seminiferous tubules of the testis in food-deprived fish, whereas the occurrence of spermatozoa was relatively infrequent in initial controls and controls. Furthermore, refeeding to food-deprived fish on day 6 onwards resulted in occurrence of few intensely stained LH secreting cells and significantly higher numbers of spermatocytes and spermatids concomitant with sparse spermatozoa in majority of tubules compared to those of food-deprived fish. These results suggest that prolonged exposure to food-deprivation causes suppression of the LH secretory activity in the pituitary gland and disruption in the spermatogenesis in O. mossambicus.
