Mature seeds of H. salicifolia, collected from five provenances (i.e. Hanuman Chatti, Helang, Lata, Rambara and Janggal Chatti) in west Himalaya, India, were treated with stratification (at 4°C for 15, 30 and 60 days) and in different concentrations of GA3 (5, 10, 20 mM), KNO3 (50, 100, 200 mM) and Thiourea (50, 100, 200 mM) solution to determine the variations in seed germination. Results reveal that the germination rates of seeds from different provenances under different pre-sowing treatments are significantly increased compared to those in control (24%–30%). The seeds treated with Thiourea (100 mM) have highest germination rate (76%–83% for different seed sources), followed by those (63%–71% for different seed sources) pretreated with stratification (4°C, 30 days). GA3 treatment significantly shortens the mean germination time (MGT) and improves seed germination percentage. Considering the practical applicability and cost effectiveness, thiourea (100 mM) and stratification (at 4°C) treatments for seed germination are recommended for mass multiplication through seeds of H. salicifolia in village/forest nurseries of the west Himalaya, India.  相似文献

The effects of seed moisture content (MC), gibberellic acid (GA3) concentration, chilling and priming pretreatments on the germination of common alder (Alnus glutinosa) and downey birch (Betula pubescens) seeds were examined. After treatment, the seeds were allowed to germinate for 42 days at 15 °C or 20 °C (dark)/30 °C (light). Treatment responses were similar at both temperatures and in both species. GA3 treatment of seeds in fully imbibed (FI) state for 30 days, or at the lower, target moisture content (TMC) for 30–90 days, significantly improved germination, but longer treatment periods reduced it (FI seeds) or had no effect (TMC seeds). Priming for two days improved germination in the FI seeds, but more than 4 days reduced it. Priming for up to 14 days had little effect on the germination of the TMC seeds.  相似文献

We characterized the effects of KNO3 pretreatment and germination temperature on dormancy breaking and germination of mature mountain ash seeds. Seeds treated with KNO3 and germinated at 25 ℃ followed by 5 ℃ had significantly higher germination percentages and germination potentials (51% and 49%, respectively), compared with controls. These treated seeds also exhibited reduced germination initiation times (minimum of 48 days), and elevated germination rate indices (up to 97). The germination of seeds subjected to long-term cold storage (2 years at 0-5 ℃) was also significantly improved by 3 days of 4% KNO3 pretreatment before germinating under a variable temperature regimen (5 ℃ followed by 25 ℃, and followed by 5 ℃). Germination percentages and germination potentials for these cold-stored seeds reached 67% and 54%, respectively, and the germination rate index increased to 126.99. Pretreatment of mountain ash seeds with KNO3 represents a practical, effective, and pollution-free method for improving germination, and can be implemented easily within a variety of nursery settings.  相似文献

The present study was conducted to evaluate the effect of different concentrations of salts on seed germination and seedling growth of Prosopis juliflora. The experiment was carried out in Forestry Laboratory at Sari Agricultural Sciences and Natural Resources University, Iran. The collected seeds were treated with boiling water and H2SO4 then left to germinate in controlled growth chamber. Seed were grown under salinity levels 100, 200 and 300 mM of CaCl2, NaH2PO4, KNO3 and KH2PO4. Germination parameters and various seedling growth variables such as root and stem length, seedlings fresh and dry weight were measured after 56 days. Results showed that germination percentage significantly decreased as the salinity level of medium increased. Salinity treatments at 100 mM enhanced the percentage of seeds germination and significantly improved the germination index and seeds stamina. In this study, the best treatment was 100 mM salinity for biomass production of seedlings. For Prosopis juliflora, 100 mM salinity significantly enhanced both germination and biomass production.  相似文献

Natural forest succession takes a long time to accumulate sufficient nutrients to support plant growth and enhance soil microbial activity. Human intervention in selecting native pioneer plant species is therefore required to accelerate sustainable restoration. Trema orientalis (L.) Blume, a fast growing pioneer plant species, has the ability to grow in nutrient deficient soils and proves to have reclamation potential in mine wastelands. However, its use has been limited due to low germination percentages and nonsynchronous seed germination. In the present study we tested the effect of sulphuric acid (H2SO4), hydrochloric acid (HCl), gibberellic acid (GA3), and potassium nitrate (KNO3) in varying concentrations and time durations on germination percentages and seed germination synchrony. We found that all treatments had a significant effect in predicting seed germination probabilities. Logistic regression analysis revealed that treatment solution and concentration had a significant effect on seed germination. Treatment with concentrated H2SO4 for 15 min increased germination up to 92% within 20 days with the least imbibition time (8 days) and highest Seedling Vigor Index (491). The scanning electron microscope images of seeds treated with H2SO4 showed complete dissolution of the honeycomb-like network of deposits on the seed coat surface removing the physical barrier and enhancing germination.  相似文献

The effects of exogenous gibberellic acid (GA3) and abscisic acid (ABA) on the total respiratory rate, percentages of total respiratory rate contributed by respiratory pathways [Embden-Meyerhof- Parnas Pathway (EMP), Pentose Phosphate Pathway (PPP), and Tricarboxylic Acid Cycle (TCA)], and conversion of starch to soluble sugars in the buds of black currants during secondary bud burst were investigated to determine the relationship between respiratory rates and secondary bud burst. ‘Adelinia’, a black currant cultivar that is prone to secondary bud burst after the first harvest, was used in this study. Mature bushes of Adelinia were sprayed with 30 mg/L GA3 and 50 mg/L ABA to manipulate bud burst. The results showed that exogenous applications of GA3 and ABA had opposite effects on bud respiratory rate. Generally, GA3 treatment increased the total respiratory rate and respiratory rate of the TCA and PPP, and the respiratory rates after GA3 treatment were higher than those of control. While ABA treatment mostly decreased the total respiratory rate and the respiratory rate of TCA and PPP in buds in comparison to control. In terms of the percentage of the three respiratory rates in comparison to the total respiratory rate, GA3 treatment significantly increased the percentage of TCA and PPP respiratory rate in comparison to the control (P < 0.01), whereas ABA decreased the rates. GA3 significantly increased the content of soluble sugars and decreased the starch content, while the starch content in buds after ABA treatment was significantly higher than that of the control. All results showed that PPP is a critical process for the second bud burst in black currants. While the EMP–TCA pathway is still dominant in bud respiration, provides a series of basic materials and energy (ATP). The conversion of starch to soluble sugars is essential for bud burst. Thus, we conclude that an energy shortage is a main contributor in ABA inhibition of the secondary bud burst of black currants.  相似文献

Five chemical and four non-chemical presowing treatments were tested on seeds of three conifers (Larix principis-rupprechtii Mayr, Pinus sylvestris L. var. mongolica Litv, and Pinus tabulaeformis Carr.). By itself, cold stratification with snow for 60 days did not significantly improve the germination percentage over that of soaking the seed in warm water (45 °C) for 12 hours. However, including either one or two dehydration-rehydration cycles after the cold stratification did improve both the percentage and speed of germination of seed from all three species. Soaking seed for 12 hours in various chemicals (KNO3, CaCl2, CuSO4, KMnO4 and GA3) before germination was less effective than the snow stratification followed by double dehydration-rehydration treatments (Chang and Lin, 1989).  相似文献

对喜马拉雅长叶松种子进行24小时GA3和H2O2浸泡预处理和15d的2-3℃低温预处理后,研究了在20℃,25℃和30℃萌发条件下,21个不同种源的喜马拉雅长叶松种子的萌发情况。结果表明,H2O2(1%v/v)和GA3(10mg/L)浸泡预处理,种子萌发率分别是82.39%和78.19%,而未经预处理的种子平均萌发率为70.79%。GA3和H2O2浸泡预处理分别使种子萌发时间缩短了8d和10d。在超过21天的20℃萌发条件下,湿冷处理提高了种子萌发率和缩短了萌发时间 而在25℃和30℃萌发条件下,总的萌发率未受到影响。喜马拉雅长叶松种子因缺少休眠而表现出很好的萌发,但是因为越来越多的造林项目需要大量的种子,播种预处理有利于提高种子萌发率和萌发速率,有助于满足种子需求。  相似文献

The effects of an exogenous nitric oxide donor (sodium nitroprusside, SNP), a NO scavenger 2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxode (PTIO) and carboxy-PTIO potassium salt (cPTIO) on the embryo germination of Sorbus pohuashanensis were studied in a petri dish test. SNP at 0.5–5 mmol L?1 increased germination percentage, mean time to germination, germination index and germination energy compared with the control to different degrees. Treatment with 2 mmol L?1 SNP improved germination most significantly; embryo germination percentage for mother tree 1 (91.11%) and mother tree 2 (64.44%) were much higher than the control. In addition, excessive SNP levels did not enhance embryo germination. Combined treatment with SNP and an NO scavenger delayed embryo germination. Treatment with cPTIO inhibited embryo germination; germination percentage was 42.22% and was lower than that of the control. These results show that low concentrations of exogenous NO can enhance the embryo germination of S. pohuashanensis, providing a simple, effective way for promoting germination of S. pohuashanensis.  相似文献

This study aimed to understand the interaction between temperature and genotype in terms of the effect on early seedling development of European aspen (Populus tremula) and various F2-aspen hybrids. We evaluated the response of 16 different European aspen and F2-hybrid families on seed germination, survival rate and seedling height in one- and two-family trials under three different temperature regimes. In one-family trials, higher germination and higher survival rates were observed in higher temperature (C1700), leading to taller seedlings. European aspen × hybrid aspen individuals (Asp × Hyb) had a higher survival rate and taller seedlings than Asp, Hyb × Asp or Hyb × Hyb individuals. The difference between Asp × Hyb and Hyb × Asp was pronounced. Both growth conditions and genotype (i.e., hybrid cross) had strong effects on germination, survival rate and height of European aspen and F2-hybrid seedlings. However, the interaction of genotype and growth conditions also had a significant influence on survival rate and seedling height, but not on germination. Two-family trials involving European aspen and F2-hybrids led to significant negative effects on germination and survival rate and also facilitation effects on seedling height. Similarly, genotype had strong effects on germination and survival rate. Different genotypes and traits affected survival rate and seedling height differently in different growth conditions. These differences were more pronounced in the warmest environment, i.e., 1,700 degree days. We conclude that introgression between F2-hybrids and local European aspen is likely in the current climate, and any warming will likely favor certain F2-hybrids, especially the most probable types (P. tremula × (P. tremula × P. tremuloides)).  相似文献

To facilitate restoration of the lava field forests surrounding Mexico City, we developed methods to improve the germination and field seedling performance of Quercus rugosa using hydropriming (regulated hydration of seeds in water), and we used special watering regimes to improve seedling acclimation. The size, dry mass, fresh mass and water content of seeds were measured, and curves were generated to evaluate acorn hydration and dehydration. The effects of stratification (5°C), heat shock (50°C) and scarification on germination were tested. All treated seeds and controls were germinated in control chambers at 21°C. One hydropriming cycle (PC) consisted of two hydration days followed by two dehydration days; treatments of 1, 2 and 3 PCs were tested. Seedlings from 1PC to 2PC were acclimated in a shade house under high and low watering regimes (400 and 200 mL week?1, respectively). In the shade house and field, the effects of hydropriming and watering treatments were evaluated by measuring length, basal diameter, crown cover, number of leaves and branches and leaf area of seedlings. Dry and fresh mass were used to calculate acorn water content. Dehydration and hydration curves displayed hysteresis. Acorns exhibited physiological dormancy, which could be overcome by stratification or by 1 month of storage. 1PC led to increased germination rates and final germination. In both the shade house and field, 1PC showed a positive effect on all seedling growth parameters except branch number. Field survival was not affected. Generally, 1PC favoured efficient seed germination, seedling vigour and homogenous plant production.  相似文献

Studies on the germination characteristics of various seed groups, extent of seedling and wildling establishment, as well as measurements on early seedling growth, were conducted on Prunus africana (Hook. f.) Kalkm. (syn. Pygeum africanum Hook. f.) (Rosaceae). Germination of seeds gathered en masse (together with soil and litter) from beneath Prunus trees was significantly greater (P < 0.01) compared to those collected in pure form from the ground (with or without fruit flesh) or those picked from the trees. Treating seeds with 10 or 100 µmol gibberellic acid (GA3) showed better germination than the control, but the difference was not statistically significant. Gibberellic acid at 1000 µmol (10–3 M) inhibited germination significantly (P = 0.02). Seedlings survived better in the glasshouse (90%) than in the nursery (87%). Whereas 60% of the 280 wildlings between 2.5 and 4.0 cm successfully established, only 10% of the 220 wildlings between 8.0 and 13.0 cm survived 3 mo after these had been maintained in the glasshouse. Seedlings derived from the GA3-treated seeds showed higher initial growth than the control owing to rapid hypocotyl elongation. Transferring seedlings to larger pots enhanced growth significantly after a lag period of about two weeks. Seedlings maintained in the glasshouse grew better than those in the nursery (P < 0.01). The study found that germination of seeds/fruits (gathered en masse from beneath trees) on a seedbed is convenient, technically less demanding, and is cost-effective.  相似文献

Jatropha curcas L. is being promoted by governments for greening marginal areas for rural development and biodiesel production in sub-Saharan Africa, however large gaps in knowledge on its agronomy and yield potential exist. The effect of duration of storage at room temperature, pre-sowing seed treatment of Jatropha seed and fruit colour harvest index on germination and seedling growth parameters was investigated. There was a significant negative correlation between the duration of storage of Jatropha seed and seed oil content (P < 0.001, r = ?0.99), viability (P < 0.01, r = ?0.93), percent germination (P < 0.05, r = ?0.90) and germination index (P < 0.05, r = ?0.89). The relationship between duration of seed storage and seed oil content and seed germination parameters was best described by quadratic and exponential functions which showed rapid declines in seed oil content and germination parameters in the first 3 years of storage and a slower decline between 3 and 8 years of storage. There was a significant negative correlation between duration of storage and seedling height (P < 0.01, r = ?0.97), stem diameter (P < 0.01, r = ?0.93) and dry weight (P < 0.05, r = 0.91) that was best described by quadratic functions. Seed oil content was significantly and positively correlated to seed viability (P < 0.01, r = 0.96), germination per cent (P < 0.01, r = 0.94) and germination index (P < 0.01, r = 0.93). The relationship between seed oil content and seed germination parameters was best described by exponential functions which showed an exponential increase in seed germination parameters as oil content increased above 30% and smaller increases below 30% oil content. Progressive peroxidation of lipids as seed aged explains the loss of oil from the seed. Peroxidation of lipids produced highly reactive radicals that damaged membranes and proteins reducing seed germination and seedling growth parameters as seed aged. Pre-sowing seed treatments (seed soaking and scarification) produced marginal gains in germination and seedling growth parameters in Jatropha seed stored for 3 months to 8 years. Seeds harvested at the yellow fruit maturity index, 60 days after anthesis (DAA), showed the highest seed germination percent while seed harvested at the black fruit maturity index, 75 DAA, had the highest oil content. Harvesting of Jatropha seed for oil expression is therefore recommended at the black maturity index while harvesting for seed is recommended at the yellow maturity stage. The results of this study indicate that Jatropha seed should be stored for as short a period of time as possible to maximize seed germination, seedling growth and oil yield.  相似文献

Nine seed lots each of Norway spruce (Picea abies L.), and Scots pine (Pinus sylvestris L.) were treated with combinations of the growth regulators: Indole‐3‐acetic acid, indole‐3‐butyric acid, etephone, kinetin, fusicoccin, and the gibberellins A1 A3 A4/7, and A9. GA9 GA4/7 and fusicoccin significantly promoted the germination of five Norway spruce, and three Scots pine, seed batches. Independent of species, the germinability of seeds treated with auxins, etephone, kinetin, GA1 and GA3 were unaffected or reduced. The rate of germination was stimulated for both species when treated with GA9 or GA4/7. The germination percentage for Scots pine seeds was enhanced by GA9 but not by GA4/7. Neither of the two GAs enhanced the germination percentage of Norway spruce seeds.  相似文献

The combination of gibberellin acid (GA) soaking with moist chilling plays a pivotal role in seed dormancy breaking. However, knowledge involving physiological and biochemical mechanisms for such a response is limited. This work focused on the effects of GA3 and moist chilling on overcoming seed dormancy as well as the response of endogenous hormones and nutrient reserves to dormancy release in Chinese dogwood (Cornus kousa var. chinensis). Seeds subjected to GA3 soaking and moist chilling exhibited a high germination percentage (average 97%), and the combination of 500 mg·L?1 GA3 soaking for 3 days followed with moist chilling for 50 days was more effective than other treatments. For GA3 treatments, lipids were initially hydrolyzed during the first stage of moist chilling (0–20 days), resulting in the increase of starch and soluble sugars, while proteins were kept relatively stable. During the second stage (20–50 days), contents of lipids, starch, soluble sugars, and proteins decreased by 46%, 82%, 61% and 59%, respectively. Abscisic acid (ABA) contents and ratios of ABA to GA3 in GA3 treatments decreased by nearly 70% and over 90%, respectively, when dormancy was terminated. However, endogenous GA3 contents in all treatments significantly increased with prolonged moist chilling. The present results provide insight into the metabolic mechanism involving hormonal regulation and mobilization of reserves during the release of seed dormancy in Chinese dogwood.  相似文献

Seeds of Strychnos nux-vomica have slow and erratic germination; thus different presowing treatments were applied to enhance the germination of its seeds collected from Tamaraikulam, Tamil Nadu, India. In addition, the effects of desiccation and different storage conditions on the germination of S. nux-vomica seeds were investigated. The results show that soaking in 500 ppm gibberellic acid (GA3) for 24 h, incubation of seeds at 40 °C for 3 days and alternate water soaking (16 h) and drying (8 h) for 14 days significantly increased the percentage germination compared to the control. Desiccation of seeds down to 10% moisture content resulted in better germination. Germination of S. nux-vomica seeds differed significantly between different storage periods, moisture contents of the seed and for first and second order interactions (p<0.001). The highest germination (92%) was achieved when seeds with 10% moisture content were stored at ambient temperature for 30 weeks. Evidence from the present study indicates that S. nux-vomica seeds possess physiological dormancy that can be broken effectively by after-ripening. As seeds of S. nux-vomica are found to be desiccation tolerant, dry seed (10% moisture content) can be hermitically stored at ambient temperature for 30 weeks without losing their viability.  相似文献

Pre-sowing treatments are expected to increase seed germination. This evaluates response to pre-sowing treatments and the growth performance of two native and rare tree species, Garuga pinnata Roxb. and Vitex glabrata R. Br. The hard seed coats were treated by rubbing with sand paper, nail clipping, and immersion in water and acid (H2SO4). Results indicate that G. pinnata showed a 90% germination rate and 30% germination energy when seed coats were nicked with a nail clipper. Rubbing with sand paper was the best pre-sowing treatment for V. glabrata, resulting a 80% germination rate and 30% germination energy. G. pinnata and V. glabrata seedlings from seeds soaked 24 h in water resulted in maximum heights and collar diameters. These were significantly higher (at p < 0.05) than those of other treatments. The results indicate that scarification or nicking of seeds may have some negative impacts on seedling growth. Therefore, it is difficult to recommend a pre-sowing treatment of seeds for achieving both maximum germination and good early seedling growth.  相似文献

We attempted to develop a method for the regeneration of plantlets from mature seeds of medically important Magnolia obovata via the induction of somatic embryogenesis in vitro. We initially cultured halves of mature seeds on either Murashige and Skoog (MS) medium or B5 medium that contained 0, 1, 5 or 10 μM gibberellic acid (GA3) for 1 month and then transferred the half-seeds to half-strength MS basal medium or B5 basal medium for further culture in the absence of GA3. Proembryogenic masses (PEMs) were observed 1 month after the transfer of the halved mature seeds to the medium without GA3. The frequency of formation of PEMs was higher (28%) after initial culture in MS basal medium plus 1 μM GA3 than in other tested media (0 or 4%). Somatic embryos that had been developed from PEMs were cultured on half-strength MS basal medium or B5 basal medium for completion of maturation and then transferred to fresh aliquots of the same medium for initiation of germination. The frequency of germination, with the formation of normal primary leaves and roots, was above 80%. We transferred the somatic embryo-derived plantlets to soil for acclimatization and the plantlets continued to thrive.  相似文献

As very little is known about the impact of cold pre-treatments on insecticidal toxicity to the surviving stored-product insects, we examined the effects of cooling (?5 °C) on the toxicity of five contact insecticides to Sitophilus granarius adults from three populations (laboratory, field and selected). We determined: (a) weevil lethal time after exposure to ?5 °C, (b) the effects of two cold pre-treatments (LT20 and LT50—lethal time for 20 and 50 % of exposed adults) on 24 and 72 h recovery rates of laboratory adults after exposure to five insecticides and (c) deltamethrin, dichlorvos and malathion toxicity to two weevil populations with altered insecticide susceptibility after exposure to the LT20 and LT50 pre-treatments. The tested S. granarius populations showed no significant differences in their susceptibility to cooling. All insecticides except dichlorvos were more toxic to the laboratory weevils after 24 h than after 72 h recovery from the LT20 pre-treatment. Dichlorvos and deltamethrin were more toxic to the other two populations after 72 h of recovery. Comparing the effects of cold pre-treatment and non-treatment on the laboratory strain, no significant increase in the toxicity of insecticides was detected, while only deltamethrin was significantly more toxic to the field and selected populations recovering for 24 h (12.1 and 11.0 times, respectively) and 72 h (6.9 and 36.6 times) from the LT20 pre-treatment. In conclusion, only the shorter of the two cold pre-treatments was found effective in terms of increasing the insecticidal toxicity, especially against the populations with altered susceptibility to insecticides.  相似文献
