We analysed the kinetics of virus distribution in Tobacco ringspot virus (TRSV)-infected tobacco. Infected plants developed asymptomatic leaves above the symptomatic leaves that exhibited ringspots. We detected virus in the shoot apical meristem (SAM). Time-course immunohistochemical microscopy showed that TRSV continuously distributed in the centres of the SAM and in leaf primordia. In contrast, TRSV infection was transient in the root meristem. In western blot and RT-PCR analyses, TRSV infection occurred only within and around the ringspot tissues of the symptomatic leaf; however, TRSV was not or hardly detected in the asymptomatic leaves of inoculated tobacco plants. These results indicate that TRSV persists in the SAM, but that TRSV infection is transient in the root meristem and asymptomatic leaves.  相似文献

Cucurbit chlorotic yellows virus (CCYV), a whitefly-transmitted virus, is a new member of the Crinivirus genus, that causes major economic losses in cucumber, melon and watermelon crops. To develop immunodiagnostic methods for CCYV, we produced an antiserum by immunizing rabbits with bacterially expressed recombinant coat protein of CCYV and detected CCYV from CCYV-infected plants with western blotting and immunoelectron microscopy. CCYV in extracts from infected plants was also readily detected by double antibody sandwich enzyme-linked immunosorbent assay with high sensitivity and specificity. A tissue blot immunoassay indicated that CCYV localizes in the phloem tissues of the petiole and is distributed in tiny spots within the leaf lamina.  相似文献

In this study,the causal agents were identified from Canna indica viral diseased plants in Yunnan Province.The diseased C.indica plants mainly exhibited the symptoms like veinal chlorosis and yellowing,streak mosaic or interveinal chlorosis,while older leaves always showed veinal necrosis as well as chlorosis.Viral pathogens were detected by RT-PCR/PCR in 24 diseased C.indica samples collected from Kunming and Yuxi City in Yunnan Province.The results indicated that the main C.indica-infecting viruses were canna yellow mottle virus (CaYMV),bean yellow mosaic virus (BYMV),sugarcane mosaic virus (SCMV).CaYMV showed the highest detection rate of 87.5 %,whereas,the BYMV had the lowest rate of 16.7% in the 24 samples.Co-infections of CaYMV+SCMV,CaYMV+BYMV and CaYMV+SCMV+BYMV were also detected in the diseased samples.However,cucumber mosaic virus (CMV),tobamovirus,luteovirus,orthotospovirus,begomovirus and umbravirus were not detected in these samples.This is the first report of CaYMV and SCMV infecting C.indica in Yunnan province.  相似文献

Expression of many host genes can be altered during virus infection. In a previous study of sugarcane mosaic virus (SCMV) infection in maize (Zea mays), we observed that expression of ZmTrm2, a gene encoding thioredoxin m, was up-regulated at about 10 days post-inoculation (dpi). In this present study we determined that ZmTrm2 silencing in maize by virus-induced gene silencing significantly enhanced systemic SCMV infection. In contrast transient over-expression of ZmTrm2 in maize protoplasts inhibited accumulation of SCMV viral RNA. Furthermore, we found that in inoculated Nicotiana tabacum leaves transient expression of ZmTrm2 inhibited accumulation of the RNA of tobacco vein-banding mosaic virus (TVBMV), a potyvirus infecting dicotyledonous plants. Interestingly in ZmTrm2 transiently expressed N. tabacum leaves, we detected by semi-quantitative RT-PCR a reduced level of the mRNA of class I beta-1, 3-glucanase (GluI), a protein known to have a role in cell wall callose deposition and viral movement. Our data indicate that the maize ZmTrm2 plays an inhibitory role during infection of plants by SCMV and TVBMV.  相似文献

Sugarcane yellow leaf virus (SCYLV), a member of the Luteoviridae , is implicated in the sugarcane disease known as yellow leaf syndrome (YLS), which is characterized by yellowing of the leaf midrib followed by leaf necrosis and possible growth suppression. YLS is distributed worldwide and susceptible cultivars are commonly infected with SCYLV. However, not all cultivars infected with SCYLV show symptoms of YLS and some cultivars that show symptoms do so sporadically. Since it is difficult to obtain virus-free plants of susceptible cultivars, it has not been possible to study the factors involved in SCYLV infection nor the effects of infection on plant growth and yield. A tissue blot immunoassay was used to visualize in vivo presence of the virus so that virus-infected and virus-free plants could be distinguished. Meristem tip cultures were used to produce virus-free plantings of six SCYLV-susceptible sugarcane cultivars. Nearly all of the regenerated sugarcane lines remained virus-free over a period of up to 4 years, whether grown in isolated fields or in the glasshouse. Experimental re-infection of the virus-free plants by viruliferous aphids demonstrated that meristem tip culture did not affect susceptibility of sugarcane to SCYLV. Improved diagnosis and production of virus-free plants of SCYLV-susceptible cultivars will facilitate research to quantify the effect of the virus on yield and to analyse the processes involved in disease development.  相似文献

甘蔗是最重要的糖料作物,由于其栽培过程中采用种茎无性繁殖,病毒病发生逐年加重.已知侵染甘蔗的病毒种类有甘蔗花叶病毒(Sugarcane mosaic virus,SCMV)、高粱花叶病毒(Sorghummosaic virus,SrMV)、甘蔗线条花叶病毒(Sugarcane streakmosaic virus,SCSMV)、甘蔗黄叶病毒(Sugarcane yellow leaf virus,SCYLV)、甘蔗斐济病病毒(Sugarcane Fiji disease virus,SFDV)、甘蔗线.条病毒(Sugarcane streak virus,SSV)和甘蔗杆状病毒(Sugarcane bacilliform virus,SCBV).文中简要介绍上述几种病毒的基本特性及其所致病害的发生特点,对目前甘蔗病毒病防治技术进行了评述,提出了我国甘蔗病毒研究中需要关注的若干问题.  相似文献

果蔗脱毒种苗甘蔗花叶病、黄叶病和宿根矮化病分子检测   总被引:1,自引:0,他引:1  
为监测2016-2017年种植的果蔗脱毒种苗脱毒效果,分别采集广州市南沙区和增城区、湛江市麻章区及华南农业大学甘蔗育种基地共83份果蔗脱毒种苗样本,进行甘蔗花叶病毒(SCMV)、高粱花叶病毒(SrMV)和甘蔗黄叶病毒(SCYLV)RT-PCR检测。结果表明SCMV的阳性样本数为3个,阳性检出率3.61%;SrMV的阳性样本数为0;SCYLV的阳性样本数为78个,阳性检出率93.98%。采用常规PCR和巢式PCR技术对采集于广州市增城区和华南农业大学甘蔗育种基地的30份果蔗脱毒种苗样本进行宿根矮化病菌(Lxx)检测,常规PCR检测阳性样本数为0,巢式PCR检测疑似阳性样本数为8,疑似阳性检出率26.67%。本研究采用茎尖组织培养脱毒技术培育的果蔗脱毒种苗能有效脱除果蔗种苗内的SCMV、SrMV和Lxx,但SCYLV的脱除效果有待进一步研究。  相似文献

玉米致死性坏死病是由玉米褪绿斑驳病毒(Maize chlorotic mottle virus,MCMV)和一种或多种马铃薯Y病毒科病毒复合侵染引起的。2014年1月在对云南省玉溪市玉米病毒病害的调查中发现了一些表现严重花叶、矮化叶片甚至整个植株坏死症状的玉米。对采集样品进行RT-PCR检测,所有样品中都同时检测到了MCMV和甘蔗花叶病毒(Sugarcane mosaic virus,SCMV),在一个样品中同时检测到了MCMV、SCMV和南方水稻黑条矮缩病毒(Southern rice black-streaked dwarf virus)。  相似文献

Symptomatic and asymptomatic plantain and banana (Musa spp.) leaf samples were collected in 1998, 1999, 2000 and 2004 to determine the frequency of infection by Cucumber mosaic virus (CMV) in 11 major plantain- and banana-growing states of southern Nigeria in which virus symptoms had been reported. Of 996 Musa plants sampled, 641 (66.4%) were infected with CMV, as determined by dot blot immunoassay and ELISA. CMV occurred in 64 out of 84 villages and in all the states sampled. Incidences of CMV in the leaf samples were variable in survey years, ranging between 16.7 and 100%. Major virus-induced symptoms were interveinal chlorosis and chlorotic streaks, observed in 29.3 and 26.7% of the total number of surveyed Musa plants. Low incidences of general leaf chlorosis, leaf crinkling/puckering, vein thickening and chlorotic flecking between 1.4 and 10.5% were also observed. Multiple viral infections in the total number of surveyed Musa plants accounted for 5.4% while 7.9% were asymptomatic. CMV induced some of the viral infections on Musa plants in southern Nigeria.  相似文献

永安地区发病辣椒植株表现小叶、黄化、丛枝、簇芽等症状。利用植原体16SrDNA基因的通用引物R16mF2/R16mR2和R16F2n/R16R2,对发病辣椒植株总DNA进行巢式PCR检测,获得约1.2kb的特异性DNA片段。经测序并在GenBank数据库进行比对分析,共获得4条植原体特定的16SrDNA基因序列(CHY-C4-1、CHYY1-1、CHY-Y7-1、CHY-G1-1)。将测得的4条序列与已报道的植原体序列进行同源性比对,并构建系统进化树,结果显示获得的4条植原体序列均聚类到16SrI组,其中CHY-Y1-1、CHY-Y7-1、CHY-G1-1与16SrI-B亚组植原体聚类到同一支,而CHY-C4-1与已报道的16SrI组内的6个亚组均未聚类到一支,因此建议将CHY-C4-1命名为新的亚组。利用iPhyClassifier在线分析软件对获得的4条植原体序列进行虚拟RFLP分析,结果与进化树获得的结果一致。  相似文献

 Sugarcane mosaic disease, caused by Sugarcane mosaic virus (SCMV), Sorghum mosaic virus (SrMV), Maize dwarf mosaic virus (MDMV) or Johnsongrass mosaic virus (JGMV) in Potyvirus, is one of the most important viral diseases of sugarcane. In the study, four primer pairs specific to SCMV, SrMV, MDMV and JGMV, respectively, were designed and used to detect 29 sugarcane leaf mosaic samples collected from 9 locations in Jiangxi province. The representative RT-PCR products were sequenced. The results showed that 22 samples were infected by SCMV, three by SrMV, and four were mix-infected by SCMV and SrMV. MDMV or JGMV were not identified in all samples. The result indicates that SCMV is the major pathogen of sugarcane mosaic disease in Jiangxi province, and SrMV is also a pathogen for the disease.  相似文献

Two sugarcane plots were set up in Guadeloupe with disease-free tissue cultured plants in a banana growing location distant from sugarcane fields. Thirteen weeks after planting sugarcane in the field, a Xanthomonas albilineans strain belonging to serotype 3 (strain XaS3) was detected in water sampled at sunrise on the leaves in the first plot. This strain randomly invaded the sugarcane canopy. Seven weeks later, a new strain belonging to serotype 1 (strain XaS1) appeared on leaves and populations of strain XaS1 progressively increased on the leaf surface, whereas populations of strain XaS3 progressively decreased. Leaf scald symptoms were first noted 26 weeks after sugarcane planting. However, only strain XaS1 was isolated from leaves and a few sugarcane stalks showing symptoms. Both strains also colonized the second field plot, which was studied at the end of the experiment to avoid human interference of aerial contamination of sugarcane. After inoculation of three sugarcane cultivars by the decapitation technique, strain XaS1 was as virulent or more virulent than five other strains of X. albilineans isolated from diseased sugarcane plants in Guadeloupe. Although strain XaS3 colonized a few stalks, it failed to produce any symptoms and was the least virulent strain. Leaf surface colonization by X. albilineans was reproduced in a greenhouse trial by spraying the pathogen on sugarcane foliage. After 8 weeks, the pathogen was isolated from disinfected leaf blades. Although the leaf scald pathogen is thought to be mainly transmitted by infected cuttings, aerial transmission of X. albilineans is also known to occur. These results indicate the importance of sugarcane phyllosphere colonization by virulent strains in the epidemiological cycle of leaf scald disease in Guadeloupe.  相似文献

Since 2003 the presence of a new syndrome characterized by symptoms of stunting, chlorotic mottling, leaf deformation, reduced yields and quality has been reported in some white berry varieties of Vitis vinifera in Trentino‐Alto Adige and Friuli Venezia Giulia vineyards. The identification of a new virus, provisionally called Grapevine Pinot gris virus (GPGV), in a cv. Pinot gris vine suggested an association between this new syndrome and the virus presence (Giampetruzzi et al., 2012), however the contemporary presence of GPGV in both symptomatic and asymptomatic plants has still to be explained. In this work, a large‐scale monitoring over a 3‐year period (2012–14) of Friuli Venezia Giulia vineyards and nurseries has shown a widespread presence of GPGV in symptomatic plants and also in asymptomatic vines, even if at a slightly lower percentage. Quantitative analyses of the virus titer revealed a great variability in the viral content of both symptomatic and asymptomatic plants but the mean GPGV quantity in symptomatic vines was significantly higher than in asymptomatic plants.  相似文献

A survey revealed that Sugarcane yellow leaf virus (SCYLV) is found on all Hawaiian sugarcane plantations including those where no yellow leaf symptoms were observed. In a comparison of growth and yield between SCYLV-infected and SCYLV-free plants of the cultivar H87-4094, germination and early shoot growth of infected plants were retarded. The number of stalks per stool was reduced by 30%, biomass was reduced by 29%, and sugar yield by 26% when plants were harvested after 11 months. Yields did not decrease when plants were harvested after 2 years. Thus, SCYLV could reduce yield, even when the plants were asymptomatic. In a field test of SCYLV-susceptible (infected) and -resistant cultivars to compare growth and yield, 10 commercial cultivars (six susceptible and four resistant to SCYLV) were grown in eight fields with different climates and soils. Primary stalk length, biomass and sugar yield did not differ between susceptible and resistant cultivars under any field conditions. Thus, harmful effects of SCYLV on yield cannot be deduced by comparing different cultivars.  相似文献
