The present study evaluated somatotrophic gene expression in liver, muscle and adipose tissue 4 d after weaning, a time point corresponding to greatly reduced serum concentrations of insulin-like growth factor (IGF)-1 and IGF-2 in pigs. Two-week-old barrows were either cross-fostered to a sow (SOW, N = 8) or weaned and fed a phase 1 diet containing either 0 or 7% spray-dried plasma (NP, N = 8 and SDP, N = 8; respectively). Piglets were allocated such that two size groups were equivalently represented in each experimental group (small, 3.5–4.3 kg and large, 4.6–5.7 kg). Animals were weighed daily and sacrificed 4 d after weaning for blood and tissue collection. Daily gains of the SOW piglets were significantly greater than those of the weaned pigs for the first 3 d of the experiment (P < 0.0001). Weight gains in the SOW and SDP pigs between d 3 and 4 were equivalently elevated relative to the NP pigs (P < 0.0001). Serum IGF-1 and IGF-2 concentrations were decreased in both NP and SDP compared to SOW (P < 0.0001). Serum IGF-2 levels were significantly lower in small piglets (P = 0.006). A Weaning Group X Size interaction was noted for liver IGF-2 mRNA (P < 0.03), reflecting a higher level of expression in large SOW piglets relative to small SOW piglets. Weaning did not affect IGF-1, IGF-2, or growth hormone (GH) receptor mRNA levels in liver, muscle, or fat (P > 0.05). Liver IGF-binding protein (IGFBP)-3 and acid-labile subunit (ALS) mRNA levels also were unaffected by weaning. Small pigs had lower levels of liver ALS (P = 0.0003), muscle IGF-2 (P = 0.02), and muscle GH receptor (P = 0.006) mRNAs. In contrast, adipose tissue IGF-1 and IGF-2 mRNA levels were greatest in the small piglets (P = 0.001 and 0.029, respectively).  相似文献

An acute stress response can be provoked by abrupt social, nutritional, and environmental changes associated with weaning, and this may disrupt homeostasis and thus compromise well-being. Manipulating environmental factors, such as photoperiod, might provide a simple way to reduce the physiological consequences that piglets experience due to weaning stress. The objective of this study was to evaluate the impacts of photoperiod manipulation across various weaning ages on leukocyte populations, lymphocyte proliferation, natural killer cytotoxicity (NK), chemotaxis, phagocytosis, and immunoglobulin G, cortisol, and BW of piglets during the nursery phase. Sixty-eight crossbred piglets were obtained from sows kept on a short-day (8 h of light/d) photoperiod from d 90 of gestation until weaning. Piglets were weaned at 14, 21, or 28 d of age and kept on a short or long (16 h of light/d) photoperiod until 10 wk of age. Piglet BW and blood samples were collected at weaning and at 6, 8, and 10 wk of age. Pigs weaned at 28 d had reduced neutrophil counts (P < 0.001), phagocytosis (P < 0.001), and lymphocyte proliferation (P < 0.05) at weaning compared with those weaned at 14 and 21 d. Pigs weaned at 21 d tended to have lower (P = 0.08) lymphocyte counts than did pigs weaned at 14 or 28 d. Pigs weaned at 14 d had reduced (P < 0.01) NK relative to those weaned at 21 or 28 d. Photoperiod also influenced pig BW and immune status. Generally, those pigs on the long-day photoperiod and weaned at 28 d were heavier (P < 0.001) than their counterparts weaned at 14 or 21 d. At 6 wk of age, NK was greater (P = 0.002) in pigs kept on a long day and weaned at 14 or 21 d than in pigs weaned at 28 d. Phagocytosis was less (P = 0.005) at 6 wk of age, but was greater at 8 wk, in piglets kept on the long day and weaned at 28 d than in long-day pigs weaned at 14 or 21 d. These results suggest that photoperiod differentially influences immune responses in piglets weaned at different ages and indicate an inverse relationship between growth and immune status. Here, weaning at 28 d and a long-day photoperiod was the treatment combination that was most physiologically beneficial to piglets, whereas a 14-d weaning and short-day photoperiod was least physiologically beneficial.  相似文献

Two experiments were conducted that demonstrated that a single injection of hydrocortisone 21-acetate (HYD, 25 mg/kg BW) administered to 6-d-old nursing piglets resulted in a twofold elevation (P less than .02) of pancreatic amylase within 2 d; activity was unaffected by an injection of 15 IU adrenocorticotropic hormone (ACTH)/kg BW (P greater than .20). Intestinal sucrase and maltase activity tended to be elevated (P less than .20) 2 and 4 d postinjection with HYD but returned to normal (uninjected) levels by 14 d of age. The normal decline of intestinal lactase activity was delayed by at least 4 d in response to both hormones (P less than .10). Organ weights were not affected by either hormone. In a separate experiment, postweaning mortality was reduced (12 vs 27%) and growth rate was substantially improved by administration of HYD to piglets 4 and 2 d prior to weaning at 14 d of age. Hydrocortisone resulted in a faster rate of gain the 1st wk postweaning for pigs weaned at 21 or 28 d. Subsequent gain by control and HYD piglets weaned on d 21 was similar, but HYD subsequently impaired growth rate of piglets weaned at 28 d of age. Growth rates of control and ACTH piglets were similar at each postweaning period regardless of weaning age (weaning age [lin.] x week postweaning [quad.] x treatment, P less than .07). This differential treatment response of daily gain may be due in part to effects on feed intake (weaning age [lin.] x week postweaning [lin.] x treatment, P less than .10). We conclude that a single injection of HYD to 6-d-old piglets precociously induces pancreatic amylase and that the sensitivity of piglets to HYD is age-dependent.  相似文献

Four experiments involving 265, 410, 894, and 554 sows (Exp. 1 to 4, respectively) were conducted to determine the effect of spray-dried plasma (SDP) at 0 or 0.25% (Exp. 1 and 2) and 0 or 0.50% (Exp. 3 and 4) in lactation diets on average daily feed disappearance (FD), sum of sow BW, fetal and placental loss from d 110 gestation to weaning (SWL), litter size at weaning, litter weight at weaning, and average days from weaning to first estrus (WEI). Experiments 1, 3, and 4 were conducted during summer months, and Exp. 2 was conducted during fall to winter months. Experiment 1 used only parity 1 and parity 2 sows and Exp. 4 used only mature (>2 parities) sows, whereas Exp. 2 and 3 used all parity groups. Sows fed SDP in Exp. 1 had increased (P < 0.01) FD and a tendency for reduced (P = 0.06) SWL and WEI (P = 0.06). Sows fed SDP in Exp. 2 had a tendency for increased (P = 0.09) sow BW at weaning and reduced (P = 0.09) SWL, whereas other variables were not different between diets. Parity 1 and 2 sows fed SDP in Exp. 3 had increased (P < 0.01) FD, but mature sows fed SDP had reduced (P = 0.02) FD. Pig survival and litter size at weaning for all parity groups was not different between diets. The WEI for parity 1 sows fed SDP was reduced (P = 0.02) and tended to be reduced (P = 0.10) for mature sows fed SDP, but was not different between diets for parity 2 sows. More parity 1 sows fed SDP were detected (P = 0.01) in estrus 4 to 6 d after weaning, and fewer were detected (P < 0.01) in estrus 6 d after weaning compared with control parity 1 sows. In Exp. 4, FD was reduced (P < 0.01) for mature sows fed SDP; however, litter weight and average pig BW at weaning was increased (P < 0.01) with more (P < 0.01) marketable pigs (pig BW > 3.6 kg) weaned per litter. Relatively low dietary levels of SDP (0.25 to 0.50%) fed to parity 1 sows farrowed during summer months increased lactation FD and reduced WEI. Mature sows fed SDP during summer months consumed less lactation feed without compromising WEI, but had an increased litter weight, average pig BW, and number of marketable pigs at weaning.  相似文献

Two experiments were conducted to evaluate the effects of Fe injection timing after birth on suckling and subsequent nursery and growing-finishing pig performance. The injectable Fe source used in both experiments was GleptoForte (Ceva Animal Health, LLC., Lenexa, KS). GleptoForte contains gleptoferron which is a Fe macromolecule complex. In Exp. 1, a total of 324 newborn pigs (DNA 241 × 600, initially 1.6 ± 0.04 kg body weight [BW]) within 27 litters were used. Two days after birth, all piglets were weighed, and six barrows and six gilts per litter were allotted to 1 of 6 treatments consisting of no Fe injection or 200 mg of injectable Fe provided in a single injection on d 2, 4, 6, 8, or 10 of age. Pigs were weaned (~21 d of age) and allotted to nursery pens with all pigs in each pen having received the same Fe treatment. In Exp. 2, a total of 1,892 newborn pigs (PIC 359 × C40; initially 1.5 ± 0.02 kg BW) within 172 litters were used. One day after birth, piglets were weighed, and 11 pigs within each litter were allotted to 1 of 6 treatments consisting of no Fe injection or 200 mg of injectable Fe provided on d 1, 3, 5, or 7 of age, or 200 mg on d 1 plus 200 mg on d 12 of age. Pigs were weaned (19 d of age) and placed in a commercial wean-to-finish facility in a total of 15 pens with equal representation of treatments in each pen. In both experiments, not providing an Fe injection after birth decreased (P < 0.05) preweaning average daily gain (ADG), weaning weight, and hemoglobin and hematocrit values compared with all other treatments. In Exp. 1, increasing the age that piglets received an Fe injection until 4 or 6 d after birth provided marginal evidence for an improvement (quadratic; P = 0.070) in preweaning ADG. For the nursery period, increasing the age that piglets received an Fe injection improved (quadratic; P = 0.013) d 80 BW, but there was no evidence of a difference (P > 0.10) in d 173 BW at the end of the grow-finish period. In Exp. 2, increasing the age that piglets received a 200 mg Fe injection showed no evidence of difference (P > 0.10) for subsequent nursery and growing-finishing ADG. In both experiments, hemoglobin and hematocrit values were decreased (linear; P < 0.05) at weaning with increasing age when pigs received an Fe injection. These experiments suggest that providing a 200 mg Fe injection within 7 d after farrowing is sufficient for optimizing preweaning and subsequent growth performance.  相似文献

This study evaluated how socializing piglets before weaning affects behavior of lactating sows and the pre- and postweaning behavior and performance of piglets. Two farrowing rooms, each with 6 pens, and 1 nursery with 4 pens were used. In total, data were obtained from 24 sows and their litters. In each farrowing room, the solid barriers between 3 farrowing pens were removed on d 12 after farrowing, and the sows remained confined in their crates (experimental group). In the other 3 farrowing pens of each farrowing room, sows and their litters were kept under conventional conditions until weaning (control group). All piglets were weaned 28 d after birth. After weaning, piglets from each group remained together in 1 pen of the nursery. The behavior of sows (lying, standing, sitting, nursing) and piglets (lying, active, suckling) in the farrowing rooms was observed for 24 h before and for 48 h after removal of the barriers between the pens. In addition, behavior (active, lying, feeding, agonistic behavior) of piglets was observed in the nursery during the initial 48-h period after weaning. Each piglet was weighed on d 5, 12, and 28 after birth and thereafter weekly until the fifth week of rearing. In the farrowing room, mixing of litters did not influence behavior of piglets and sows. Preweaning weight gain of the piglets did not differ (P = 0.60) between the treatments. In the initial 48 h after weaning, less agonistic behavior (P < 0.001) was observed in piglets belonging to the experimental group. During 5 wk of rearing, piglets in the experimental group gained more weight compared with the control group (P = 0.05). The advantage shown by the experimental group became especially conspicuous in the first week after weaning (P = 0.05). By socializing unfamiliar piglets before weaning, stress due to mixing could at least be distanced in time from the other burdens of weaning, thereby improving performance.  相似文献

The influence of heated mash on growth and feeding behavior of newly weaned piglets was investigated. An automatically ventilated nursery with 4 identical pens was used. Twenty piglets weaned at 21 d were housed in each pen. The experiment was repeated 3 times. In total, data were obtained from 240 piglets of 12 pens. The pens were provided with a sensor-controlled, automatic feeding device, which dosed a ready-mixed mash in a trough. In each of 2 of the pens, the feed was mixed with warm water at 36 degrees C, during the first week of weaning. This heated mash had a temperature of 34 degrees C at the outlet of the automatic feeding device (experimental group). In the 2 control groups, the water was not heated and the temperature of the mash was 14 degrees C at the outlet of the automatic feeding device. From the second week of weaning, the mash had a temperature of 14 degrees C at the outlet of the automatic feeding device in all 4 pens. Piglets were weighed at weaning, at weekly intervals through 49 d after weaning, and on d 139 after weaning. Behavior of the whole group, as well as behavior of selected focal animals, was evaluated for the first 48 h after weaning. In addition, skin condition of piglets was assessed on day of weaning and on d 7, 14, and 21 after weaning. The amount of feed consumed by the piglets was recorded on a daily basis throughout the whole period of nursery. Over the total period of the study, piglets in the experimental group gained 3.98 +/- 1.66 kg (P = 0.047) more than the control group. The difference was particularly clear during the nursery period (49 d) when the experimental group gained 0.89 +/- 0.23 kg more than the control group (P = 0.03). Although piglets in the control group consumed 37.15 +/- 0.15 kg of feed over the complete nursery period, the experimental group consumed 42.56 +/- 0.15 kg per piglet (P = 0.023). By heating the mash feed in the first week after weaning, both growth performance as well as feed consumption of piglets could be increased. No difference in feed conversion and feeding behavior was found between groups.  相似文献

Syndyphalin-33 (SD-33) increases feed intake in sheep and recently weaned pigs. To assess the effects of SD-33 on hypothalamic gene expression, hypothalami were collected from unweaned pigs (n=19; 21±3 d of age) on day 0. Remaining pigs received an intramuscular injection of 0.5 μmole/kg SD-33 (SD) or saline (VEH) and weaned into individual pens. On days 1, 4, and 7 after weaning, hypothalami were collected from subsets of pigs (n=8 or 9) within each treatment group. Expression of μ-opioid receptor (MOR) was less in SD pigs than in VEH pigs on day 1 and day 4, suggesting down-regulation of the receptor by SD-33. Expression of hypothalamic melanocortin 4 receptor (MC4R) at 1 d after weaning was increased in VEH pigs (but not SD pigs) relative to levels before weaning. Expression of AGRP was not significantly altered by weaning or treatment at 1 d after weaning. At 4 d after weaning, expression of AGRP was greater in SD pigs than in VEH pigs, but at day 7 expression was less in SD pigs than in VEH pigs. A strong positive correlation was noted between expression levels of MOR and MC4R across treatment and time. Treatment with SD-33 appeared to partially abrogate the effects of weaning on expression of two key appetite-regulating genes within 24 h. Effects of SD-33 appear to be mediated at least in part by the μ-opioid receptor and include actions on the melanocortinergic pathway.  相似文献

The aim of this study was to determine the effect of dietary Forsythia suspensa extract (FSE) supplementation to lactating sows and nursery pigs on post-weaning performance, antioxidant capacity, immunoglobulins, and intestinal health. Based on backfat, body weight (BW), and parity, 24 gestating sows (Landrace × Yorkshire) with average parity of 3.38 ± 0.61 and BW of 234 ± 6.81 kg were allotted into two dietary treatments (control vs. 100 mg/kg FSE) with 12 sows per treatment from day 107 of gestation to day 21 of lactation. After weaning, based on the initial BW and source litter, 192 nursery pigs (Duroc × [Landrace × Yorkshire], average BW of 6.98 ± 0.32 kg, weaned at day 21) were allotted into four dietary treatments with eight replicate pens per treatment, six pigs per pen for a 4-wk study. The treatments included the following: 1) CC (sows and their piglets both fed control diet); 2) CF (sows fed control diet and their piglets fed FSE diet [containing 100 mg/kg FSE]); 3) FC (sows fed FSE diet and their piglets fed control diet); and 4) FF (sows and their piglets both fed FSE diet). The MIXED procedures of SAS for a split-plot arrangement with sow diet as the whole plot and nursery diet as split plot were used to analyze the data. After weaning, piglets from FSE-fed sows had improved (P < 0.05) average daily gain and feed efficiency, and lower (P < 0.05) diarrhea rate in overall (day 1 to 28) compared with those from sows fed control diet. Piglets from FSE-fed sows also had higher (P < 0.05) contents of immunoglobulin G (IgG), growth hormone, superoxide dismutase (SOD), total antioxidant capacity in serum, villus height in ileum, and villus height to crypt depth ratio in jejunum, as well as lower (P < 0.05) content of malondialdehyde (MDA) in serum and crypt depth in ileum compared with those from sows fed control diet. Piglets fed FSE during nursery had increased (P < 0.05) concentrations of IgG, SOD, and catalase, and decreased (P < 0.05) MDA and tumor nuclear factor-α levels in serum compared with those fed control diet during nursery. Piglets from FC group had increased (P < 0.05) protein expression of occludin in jejunal mucosa and relative abundance of Lactobacillus on genus level in colon compared with those from CC group. In conclusion, for the performance and intestinal health, diets supplemented with FSE during lactation phase seemed more efficient to alleviate weaning stress than the nursery phase. In terms of the antioxidant status and immunoglobulins, FSE supplemented in both phases were efficient for nursery pigs.  相似文献

In pig (Sus Scrofa) production, within-batch variation in bw gain of piglets during the nursery period (up to 10 wk of age) can be high and is of high economic importance. Homogeneity of BW within batches of animals is important as it influences the efficiency of use of the grower and finisher facilities, and provides an extra value for the fattening farms. In the current study, factors for a light BW at the end of the nursery period of pigs were determined by analyzing datasets from 3 different swine research centers in the Netherlands and France. The entire dataset contained information on 77,868 individual piglets born between 2005 and 2010. Body weight was determined at different time points over the pre- and post-weaning phase, and sex, season of birth, litter information (litter size at day of birth and after cross-fostering, number of piglets born alive per litter, number of total born littermates, sow parity number), cross-fostered animals (yes or no), and pen group size over the post-weaning period were recorded. A risk factor analysis approach was used to analyze the datasets to determine factors that predict piglet bw at the end of the nursery period. Body weight at the end of the nursery period corrected for age was mainly determined by season (P < 0.001), birth weight (BiW, P < 0.001), weaning weight (WW, P < 0.001), and BW at 6 wk of age (P < 0.001). These variables were consistent among datasets and explained approximately 70% of the overall variation in BW at the end of the nursery period. Litter information did not significantly (P > 0.05) contribute to explaining the BW at the end of the nursery period. To discard the possibility of intrauterine growth retarded piglets (IUGR) being the reason for the influence of BiW as an explanatory factor in the regression model, a further analysis was performed on the effect of this category of piglets on the results of the regression analysis. Overall, it was concluded that the bw of piglets at the end of the nursery phase is mainly determined by season, sex, birth, WW, and BW at 6 wk of age. Piglets with a BiW greater than the mean biw minus 2.5 times the sd have the potential to compensate during the subsequent phases of growth.  相似文献

Selection was applied from 1964 to 1978 for increased weaning weight (WWL) or yearling weight (YWL) in two Hereford lines. An Angus line was maintained as an unselected control line (CL). Each line was maintained with 50 cows and four sires each year (two sires selected each year and used for 2 yr). Primary traits measured in the lines were birth weight (BW), preweaning daily gain (WDG), weaning weight (WW), weaning conformation grade (WG), weaning condition score (WC), weaning to yearling daily gain (YDG), yearling weight (YW), yearling conformation grade (YG) and yearling condition score (YC). Averaged over two methods, estimated genetic responses/generation (in standard deviation units) in WWL and YWL were: BW, .29, .26; WDG, .17, .15; WW, .22, .19; WG, .19, .26; WC, .12, .12; YDG, -.02, .04; YW, .08, .14; YG, .19, .16; YC, -.13, -.03. The realized heritability estimates were .23 and .15 for WW and YW, respectively. The realized genetic correlation between WW and YW was .69. Progeny from crosses of selected WWL and YWL sires to Angus cows had similar feedlot and carcass performance. At the end of the study, milk yield and composition were similar for mature cows in WWL and YWL.  相似文献

A study was conducted with 20 weaned barrows (14 d, 4.98 +/- 0.21 kg) to determine the effect of feeding spray-dried plasma (SDP) after weaning on the pig's stress response to a lipopolysaccharide (LPS) challenge. After weaning, pigs were fed a diet containing 0 or 7% SDP for 7 d. On d 6 after weaning, all pigs were nonsurgically fitted with a jugular catheter. On d 7 after weaning, the pigs were given i.p. injections of either saline or LPS (150 microg/kg BW) followed by serial blood collection every 15 min for a 3-h period. Following the 3-h blood collection, all pigs were killed and tissue was collected for mRNA analysis. Pig weight on d 7 after weaning was not affected by dietary treatment (P > 0.21). Pigs fed the diet with SDP had lower (P < 0.05) levels of hypothalamic corticotropin-releasing hormone (CRH) mRNA, pituitary gland CRH receptor mRNA, and adrenal gland adrenocorticotropin-releasing hormone (ACTH) receptor mRNA. Dietary treatment did not affect pituitary gland proopiomelanocortin (POMC) mRNA. No effect of LPS treatment was observed in any of the mRNA levels examined. For both serum ACTH and cortisol, there was a significant diet x LPS treatment interaction (P < 0.01) such that both the ACTH and cortisol responses to the LPS challenge were greater in the pigs fed the diet with SDP than in the pigs fed the diet without SDP. For pigs given the saline injection, diet did not affect basal serum cortisol concentration; however, basal serum ACTH concentration was lower in those pigs fed the diet with SDP (P < 0.0001). A diet x LPS treatment interaction (P < 0.024) was observed for adrenal gland mRNA expression for steroidogenic acute regulatory (StAR) protein such that the LPS-induced increase in StAR mRNA was greater in the pigs fed SDP than in pigs fed the diet without SDP. These results demonstrate that pigs fed a diet with SDP have an increased activation of the pituitary-adrenal axis following an LPS challenge compared to pigs fed a diet without SDP.  相似文献

The effects of piglet birth weight and liquid milk replacer supplementation of piglets during lactation on growth performance to slaughter weight was evaluated in a study carried out with 32 sows (PIC C-22) and their piglets (n = 384; progeny of PIC Line 337 sires). A randomized block design with a 2 x 2 factorial arrangement of treatments was used. Treatments were birth weight (Heavy vs Light) and liquid milk replacer (Supplemented vs Unsupplemented). The study was divided into two periods. At the start of period 1 (birth to weaning), pigs were assigned to either Heavy or Light (1.8 [SD = 0.09] vs 1.3 kg [SD = 0.07] BW, respectively, P < 0.001) litters of 12 pigs and half of the litters were given ad libitum access to supplemental milk replacer from d 3 of lactation to weaning (21 +/- 0.2 d). In period 2 (weaning to 110 kg BW), a total of 308 pigs were randomly selected from within previous treatment and sex subclasses and placed in pens of four pigs. Pigs were given ad libitum access to diets that met or exceeded nutrient requirements. Pigs in heavy litters were heavier at weaning (6.6 vs 5.7 kg BW; SE = 0.14; P < 0.001) and tended to have more pigs weaned (11.4 vs 10.9 pigs/litter; SE = 0.21; P = 0.10). After weaning, pigs in the Heavy litter had greater ADG (851 vs 796 g; SE = 6.7; P < 0.001) and ADFI (1,866 vs 1,783 g; SE = 17.6; P < 0.001), similar gain:feed (0.46 vs 0.45; SE = 0.003; P > 0.05), and required seven fewer days (P < 0.001) to reach slaughter weight compared to pigs in the Light treatment. Feeding supplemental milk replacer during lactation produced heavier pigs at weaning (6.6 vs 5.7 kg BW; SE = 0.14; P < 0.001) and tended to increase the number of pigs weaned (11.4 vs 10.9 pigs/litter; SE = 0.21; P = 0.10) but had no effect (P > 0.05) on growth performance from weaning to slaughter. However, pigs fed milk replacer required three fewer days (P < 0.01) to reach 110 kg BW. Sow feed intake and BW loss during lactation were not affected (P > 0.05) by either birth weight or milk replacer treatment. In conclusion, birth weight has a substantially greater impact on pig growth performance after weaning than increasing nutrient intake during lactation.  相似文献

The aim of the present study was to investigate the effect of cysteamine on growth performance of preweaning piglets and gastric expression of ghrelin mRNA in vivo and in vitro. Twelve litters of newborn piglets were allocated randomly to control and treatment groups. From 15 d of age, piglets in the control group were fed basal creep diet, whereas the treatment group received basal diet supplemented with 120 mg cysteamine per kg of diet until weaning on 35 d of age. Body weight gain, creep feed consumption, and diarrhea rates were recorded, and gastric mucosal tissues were collected for quantifying mRNA expression. To evaluate the direct effect of cysteamine on gastric ghrelin expression, primary cultures of gastric mucosal cells isolated from 35-d-old piglets were exposed to cysteamine for 20 h at 0, 1, 10, and 100 μg/mL, respectively. Dietary cysteamine increased (P < 0.05) average daily creep feed consumption and BW gain in preweaning pigs, which was accompanied by reduction in diarrhea rates. At 35 d of age, piglets treated with cysteamine showed increased (P < 0.05) ghrelin and gastrin and decreased (P < 0.05) somatostatin mRNA expression in gastric mucosa. Moreover, dietary cysteamine treatment increased serum concentration of gastrin (P < 0.05). In vitro, cysteamine significantly increased ghrelin mRNA expression in gastric mucosal cells at the concentration of 10 μg/mL. In conclusion, dietary cysteamine is effective in improving the growth performance and health condition of preweaning piglets, which is associated with its stimulatory effects on gastric ghrelin mRNA expression both in vivo and in vitro.  相似文献

The objectives of this study were to determine the effects of oral Gln supplementation on growth performance, intestinal morphology, and expression of heat shock protein (Hsp) 70 in weaning piglets. A total of 65 piglets after weaning at 21 d of age (d 0) were used in this experiment. Five piglets were randomly selected and euthanized initially at d 0 to determine baseline values for the expression of Hsp70 in the small intestine. The remaining piglets were randomly assigned to 1 of 2 treatments and received 0 or 1 g of oral Gln/kg of BW every 12 h. After piglets were humanely killed at d 3, 7, and 14 postweaning, the duodenum, jejunum, and ileum of piglets were sampled to evaluate intestinal morphology and the expression and localization of Hsp70. The results indicated that oral Gln supplementation increased plasma concentrations of Gln compared with those in control piglets (P < 0.05). Average daily gain and ADFI were greater in piglets orally supplemented with Gln than in control piglets during the whole period (P < 0.05). The incidence of diarrhea in piglets orally supplemented with Gln was 24% less than (P = 0.064) that in control piglets at 8 to 14 d after weaning. The weights of the jejunum and ileum were greater in piglets orally supplemented with Gln compared with those of control piglets relative to BW on d 14 postweaning (P < 0.05). The villus height and the villus height:crypt depth ratio in the jejunum and the ileum were greater in piglets receiving oral Gln on d 14 postweaning (P < 0.05) than in control piglets. These results indicate that Gln supplementation can influence the intestinal morphology of weaned piglets. The expression of hsp70 mRNA and Hsp70 proteins in the duodenum and jejunum was greater in piglets supplemented with Gln than in control piglets (P < 0.05). However, Gln supplementation had no effect on the expression of hsp70 mRNA and Hsp70 proteins in the ileum. Moreover, the localization of Hsp70 in the cytoplasm indicated that Hsp70 has a cytoprotective role in epithelial cell function and structure. These results indicate that Gln supplementation may be beneficial for intestinal health and development and may thus mitigate diarrhea and improve growth performance. The protective mechanisms of Gln in the intestine may be associated with the increase in Hsp70 expression.  相似文献

This study was conducted to examine the effects of weaning stress on gene expression of specific markers in hypothalamus‐pituitary‐adrenal (HPA) axis and neuronal activity in the newly weaned piglets. Twelve 28‐days‐old, newly weaned crossbred (Landrace × Yorkshire × Duroc) male piglets from 6 l (2 piglets/l) were randomly categorized into two groups: (a) weaning stress: piglets were separated from their dams, relocated and mixed with the unacquainted domestic piglets for 2 hr (stress, n = 6); (b) no‐stress: piglets stayed with their dams in the farrowing house (NS; n = 6). After weaning stress, all piglets were electrically euthanized and the blood samples/HPA tissues were collected for subsequent analysis, including plasma cortisol and mRNA expression of c‐fos, c‐jun, corticotropin‐releasing hormone (CRH), CRH receptor 1 (CRHR‐1) and adrenocorticotropin hormone receptor (MC2R). Results: Weaning stress significantly (p < 0.05) increased the plasma cortisol level and suppressed the expression of c‐fos and CRH in hypothalamus. In addition, weaning stress enhanced the mRNA abundance of c‐jun and CRHR‐1 in the pituitary gland. No significant differences in the gene expression of MC2R and CRHR‐1 were observed in the adrenal gland between treatment groups. Taken together, HPA involved in weaning stress and CRHR‐1 and c‐jun could be potential markers to evaluate the activation of HPA axis.  相似文献

Data consisting of 948 calf records collected from 1978 to 1982 were analyzed to determine the effects of breeding methods used to improve commercial herds genetically on birth and weaning traits. Four distinct groups were used in the project: Group 1 (G1), an unselected, random mating Hereford control line; Group 2 (G2), a Hereford group using sires selected for yearling growth; Group 3 (G3), a rotational cross with Angus, Hereford, Charolais and Simmental breeds; and Group 4 (G4), a rotational cross with Angus, Hereford, Simmental and Holstein-Friesian breeds. Traits analyzed were birth weight (BW), calving difficulty (CD), percent assisted births (%AB), percent born alive (%BL), preweaning average daily gain (PWDG), relative growth rate (RGR), weaning weight (WWT) and percent weaned (%WND). The use of high yearling weight sires in G2 increased calf size (P less than .01) at birth and weaning by 8.9 and 28.1 kg, respectively, along with increased CD (P less than .01). Use of rotational crossbreeding systems increased calf size and growth from birth to weaning (P less than .01), but decreased CD and %AB (P less than .01) by .17 units and 13.5%, respectively. Including Holstein-Friesian in G4 resulted in further increases in preweaning growth (P less than .01) and calving ease was improved without affecting BWT compared with G3.  相似文献

Orexin is a hypothalamic neuropeptide that regulates feeding behavior in rats. Orexin-B has recently been cloned in pigs and was shown to stimulate food intake after intramuscular injection. This study was designed to determine whether intracerebroventricular (ICV) and intravenous injections of orexin could regulate appetite in sheep. Suffolk wethers were moved to indoor facilities, adapted to diets for 6 wk, and trained to stand in stanchions for 3 to 6 h each day for 2 wk before indwelling ICV cannulas were installed. These sheep were provided water and they consumed feed ad libitum. On the day before an experiment, each sheep was cannulated in a jugular vein. On the day of an experiment, sheep were placed in stanchions and allowed to stand for 1 h before use. Sheep were then monitored over a 2-h control period before i.v. injection with saline or porcine orexin-B (3 micrograms/kg BW) or ICV injection with artificial cerebrospinal fluid (CSF), orexin (0.03, 0.3, or 3 micrograms/kg BW) or in a second experiment with either orexin B (0.03, 0.3, 3 micrograms/kg BW), neuropeptide-Y (NPY; 0.3 microgram/kg BW), or orexin plus NPY. Food intake was monitored for consecutive 2-h periods. The i.v. injections of orexin did not affect food intake or metabolite or hormone concentrations. In ICV sheep, orexin increased food intake at 2 (P < 0.04) and at 4 h (P < 0.02). Food intake was greatest with the 0.3 microgram/kg BW dosage of orexin (P < 0.05). In the first 2 h after injection, orexin had an effect similar to that of NPY (0.23 kg for orexin and 0.2 kg for NPY). The combination of NPY and orexin had a greater effect on food intake (to 0.34 kg) than did either orexin (P < 0.05) or NPY (P < 0.008) alone. Differences were not apparent in the subsequent 2-h interval. No differences were noted in free fatty acid, glucose, growth hormone, luteinizing hormone, or insulin concentrations following orexin injection. There was an effect of ICV orexin treatment on plasma cortisol concentrations (P < 0.002). Cortisol was increased by orexin at the 0- to 2-h (P < 0.008) and in the 2- to 4-h (P < 0.009) intervals after orexin injection. These data indicate that central administration of orexin stimulates feed intake in sheep.  相似文献
