The impact of the laboratory induced Schistosoma mansoni with decreased PZQ sensitivity on the biological performance of its different developmental stages and the concomitant structural changes of adult worms’ total proteins were investigated. PZQ exposed snails showed stoppage of cercarial shedding for eight weeks followed by progressive significant reduction of cercarial production along four successive weeks. In the vertebrate host, in comparison to Schistosoma mansoni susceptible isolate, inoculated cercariae with decreased PZQ sensitivity led to an evident decrease in male to female ratio associated with significant reduction in tissue egg counts and significant increase in dead egg percentage. Significant reduction in the fecundity was also determined. Interestingly, eggs from adult worms with decreased PZQ sensitivity showed two unique features as they found to be smaller and more spherical in addition to the observation of hourglass shaped miracidium in about 10% of the detected mature eggs. Proteomic analysis of adult worms with decreased sensitivity to PZQ using mass spectrometry revealed up-regulation of Ca2+ ATPase 2 and Hsp70. This study can point to the increase incidence of the neuroschistosomiasis due to the small size eggs of Schistosoma mansoni with reduced PZQ sensitivity. These worms can also impact the epidemiology in the field. The study can also provide help to elucidate underlying potential molecular mechanisms of resistance that could lead to possible strategies to reverse drug resistance.  相似文献

The morphology of Cephalochlamys namaquensis (Cohn 1906) Blanchard (1908), a diphyllida cestode from the African clawed toad, Xenopus laevis, is examined with respect to variability reported in the literature. The parasite has a high prevalence of infection (68,9%) in the X. laevis population studied and shows a preference for attachment in the anterior duodenum. Approximately 76% of the worms recovered were juveniles. Coracidia had an LT50 of 6 h and transmission is thought to take place in darkness at the sediment/water interface. The copepod Eucydops gibsoni serves as an intermediate host and it is suggested that chaoborid larvae may act as paratenic hosts.  相似文献

SUMMARY: The efficiency of the broad spectrum anthelmintic nitroscanate against tapeworm and nematode infections of dogs was tested in artificially or naturally infected dogs. The safety of the compound was also evaluated in acute, subacute and chronic toxicity tests. At the recommended single dose rate of 50 mg/kg given to the dogs with food, nitroscanate was 98% efficient against Taenia hydatigena, T. ovis and T. pisiformis. The drug was highly efficient against Echinococcus granulosus only at the dose rate of 200 mg/kg given twice but total elimination of worms was not achieved. Natural infections of Dipylidium caninum, Ancylostoma spp and Uncinaria stenocephala were totally eliminated from all dogs at the dose rate of 25 mg/kg or higher. Nitroscanate was 97% efficient against adult Toxocara canis at a single dose of 50 mg/kg. When the dose was repeated 24 hours later total elimination of both mature worms and immature worms in puppies aged 2 weeks was achieved. The repeated dose of 100 mg/kg removed 98% of early immature worms from puppies aged 3 days. The drug was 100% efficient against adult Toxascaris leonina at a single dose of 50 mg/kg and 90.5% efficiency was achieved against early 4th stage larvae by two doses of 50 mg/kg. Nitroscanate was not efficient against Trichuris vulpis. The drug was efficient for the removal of Toxocara cati and Ancylostoma tubaeforme from cats. Nitroscanate caused no serious symptoms of toxicity at dose rates up to 10,000 mg/kg in single or repeated doses in young or older adult dogs. The drug was safely given to dogs during pregnancy, to young puppies and to cats. The regular use of nitroscanate as a broad spectrum anthelmintic for the prevention and control of parasitic infections of dogs is discussed.  相似文献

Mediorhynchus gallinarum was recovered from the small intestines of 36 of 50 Helmeted guineafowls sampled from August 1988 to May 1989. The intensity of infection ranged from 1-141 worms per host, with a mean intensity of 23.2 (+/- 34) and a median intensity of 5. The Wilcoxon-Mann-Whitney test revealed no significant differences between the mean worm burdens of male and female birds at the 5% level (P > 0.05). Slightly more female than male acanthocephalans were collected. The majority (63.4%) of females had eggs with fully-developed embryos, 9% had immature eggs, 21.2% had no eggs and the egg status of 6.4% could not be determined. No seasonal pattern of intensity of infection emerged from the data, but worm burdens were markedly higher after good rains in February 1989. South Africa constitutes a new geographic record for M. gallinarum.  相似文献

Eggs from the pig roundworm Ascaris suum are easily obtainable in large numbers from uterus of adult worms. It is therefore natural that eggs isolated from that organ have been used almost exclusively in experimental ascariasis, both in the natural host (Kelley & Nayak 1965, Gaafar & Keittevuti 1972, Andersen et al. 1973, Jørgensen et al. 1975 and others) and in small laboratory animals (Jeska et al. 1969, Berger 1971 and others). In some cases no details are given on the origin and preparation of the infective eggs or the eggs may originate both from uteri of adult worms and from pig faeces (Kelley et al. 1957).  相似文献

Following subcutaneous injection, a proportion of the larvae of Strongyloides papillosus became arrested in the musculature of rabbits and sheep. By the anthelmintic removal of the adult worms from the small intestine, it has been shown in both host species that arrested larvae are capable of continuing their migration and replacing the adult population. While arrested larvae seldom remained longer than 4–5 weeks in the muscles of rabbits, and most migrated earlier, they remained in excess of 12 weeks in sheep. It is suggested that these arrested larvae have the function of maintaining the intestinal population of adult worms.  相似文献

An experimental infection of freshly cultured Haemonchus contortus larvae was given to housed, non-pregnant sheep in October. Phenothiazine was administered to all animals shortly after patency, but the egg output remained negative until mid-April indicating that removal of adult worms from the abomasum did not stimulate the resumption of development of arrested larvae. Worm burdens at post mortem suggest that adults were not being expelled to any great degree during the winter from the host, but this information is clouded by the difference in numbers of both adult worms and arrested larvae in sheep of different haemoglobin type. The results suggest that haemoglobin type may also be a factor in arrest of larvae as it is in the resistance of sheep to adult H. contortus.  相似文献

Adult worms from a levamisole resistant population ofOstertagia consisting ofO. circumcincta andO. trifurcata showed no dose response to morantel tartrate. The lowest dose rate of 2.2 mg/kg removed 30.8% of worms while the highest dose rate of 17.6 mg/kg removed 39.2%. A dose response was obtained against naphthalophos and the LD50 and LD95 were determined as 12.54 mg/kg and 23.25 mg/kg respectively. Clarias gariepinus were collected from Lake Chivero, Zimbabwe, and examined for nematode parasites from November 2000 to May 2002. Of the 202 specimens collected, 42.6% were infected with third-stage larvae of Contracaecum sp. in the body cavity. The intensity of the infection was 1-7 worms per fish (mean intensity = 2.2). Seasonal variation in the prevalence of the parasite was not obvious and there was no significant difference in the prevalence of infection between males and females (chi 2 = 2.228; P > 0.05). No significant relationship between host size and prevalence was established. There was also no significant relationship between intensity and the body condition factor (r = 0.11; P > 0.05). The low parasite prevalence may have been caused by the disruption of the infection cycle since piscivorous birds, which are the final hosts of the parasite, do not feed on C. garieplnus in Lake Chivero.  相似文献

Radiolabelled fenbendazole (FBZ) and thiabendazole (TBZ) were administered into the rumen or abomassum of sheep previously infected with benzimidazole-susceptible or resistant Haemonchus contortus and Trichostrongylus colubriformis. Plasma radioactivity was determined for 12 hours following anthelmintic administration and at the end of this time the incorporation of radiolabel into the parasites was determined.Incorporation of radiolabel from FBZ was significantly greater in susceptible than in resistant worms. However, with TBZ there was no significant difference. There were no significant differences in the incorporation between H. contortus and T. colubriformis. Incorporation of TBZ was greater than FBZ in the worms except after ruminal administration to sheep showing clinical parasitism resulting from infection with benzimidazole- susceptible worms. TBZ was administered at five times the dose rate as FBZ, and thus a lower proportion of the administered TBZ than FBZ was incorporated.With the exception of incorporation of TBZ by resistant worms, the route of administration had a major effect on incorporation by the parasites and on plasma levels. Based on plasma levels, TBZ appeared to be rapidly absorbed from both the rumen and abomasum, whereas FBZ was less readily absorbed from the rumen with the rate of absorption being markedly greater if FBZ was administered directly into the abomasum. The findings suggest that the rumen may act as a reservoir of FBZ prolonging the period of high anthelmintic concentration in the host. This may contribute to the high efficacy of this anthelmintic.  相似文献

Background: Capillariasis, an important parasitic disease of birds is caused at least by seven different genera of trichurid nematodes with clinical outcome ranging from mild enteritis to high mortality.

Objective: This study was aimed to investigate the causative agent involved in high mortality associated with severe enteric illness among turkey flocks in an organized commercial poultry farm at Bareilly, India.

Materials and Methods: Turkey carcasses (n = 119) and fecal samples from the affected deep litter pen constituted as the study materials. The disease was investigated by systematic necropsy, direct microscopy and histopathology. Representative samples were screened for other enteric pathogens.

Results: Microscopic examination of mucosal scraping revealed capillarid worms and their eggs in all the samples. The morphological features of adult worms were typically consistent to Baruscapillaria obsignata. Histopathology exhibited thickened muscular and mucosal layers, mononuclear and heterophilic infiltration in the lamina propria, blunting and clubbing of villi, epithelial denudation and sections of capillarid worms. Administration of levamisole at 80 ppm in drinking water reduced the mortality, clinical illness and worm load after three days of therapy.

Conclusions: The capillarid worms in different avian hosts can cause different clinical manifestations and outcomes. From India, this is the first report describing intestinal pathology caused by B. obsignata in turkeys. We conclude that the B. obsignata infection is capable of causing life threatening enteropathy in turkeys and, hence, routine screening, scheduled deworming and good litter management are crucial to control the infection and its associated loss.  相似文献

Heartworms, Dirofilaria immitis, were recovered from 17 out of 24 (71%) coyotes, Canis latrans, 38 out of 46 (83%) coyote × red wolf hybrids and all of 8 (100%) red wolves, Canis rufus gregoryi, collected from the Gulf coastal prairies of southeast Texas and southwest Louisiana. Intensities of infection ranged from 1 to 176 (x=25) worms per host. There was a significantly (P<0.05) higher intensity of infection in red wolves. Prevalence of heartworms increased significantly with increasing age. There were no significant differences between coyotes, hybrids and red wolves or between different host sexes in terms of prevalence. The female to male ratio of heartworms was close to unity (1.2:1) and was not correlated with worm burdens. Nematodes were primarily localized in the right heart, frequently extending into the pulmonary artery and the pulmonary arterial tree in the lungs. In 13 instances, 1–4 adult heartworms were recovered from the venae cavae. Pathological responses in the right heart were variable, depending on the intensity of infection. In severe infections, there were small areas of infarctive necrosis with mild to severe interstitial edema in the myocardium. Lesions in the pulmonary artery and pulmonary arterial trunk varied from mild focal hyperplastic intimal changes to extensive exudative villous endarteritis. The latter was characterized by a hyperplastic collagenous stroma with numerous histiocytes, plasma cells and eosinophils. Lung pathology varied from patchy to extensive areas of congestion, edema, hemorrhage, interstitial pneumonitis and infarction. In cases with heartworms in the venae cavae, hepatic changes were minimal and associated with liver changes such as passive congestion and centrolobular necrosis seen in cases without adult worms in the venae cavae. In heavily infected animals, hemosiderosis of the liver, spleen and kidneys was pronounced. A microfilaremia was noted in 46% of the infected wild canids. Microfilariae were observed in tissue sections of the myocardium, lungs, liver, kidney, spleen, lymph nodes, pancreas and appendix. Wild canids from this area are regarded as natural definitive hosts and primary reservoirs for heartworms and it appears that this infection is an important factor in the morbidity and mortality of these hosts.  相似文献

Distal ileum, caecum and proximal colon of 470 horses were examined for helminths during 1 year at an abattoir in central Sweden. The infection levels of the horse tapeworm Anoplocephala perfoliata, their stage of development, site of attachment and gross pathological lesions caused by the worm were recorded. Faecal samples from 395 of the horses were examined specifically for tapeworm segments and eggs in order to correlate these findings with the numbers in the alimentary canal. In total 65% of the horses were infected with A. perfoliata and the mean intensity of infection was 79 worms per infected horse with a maximum of 912. The level of infection was significantly higher in (1) 3rd and 4th than in 1st and 2nd quarter of the year; (2) older horses than in yearlings; (3) females than in males and geldings; (4) thoroughbreed and cold-blooded horses than in Swedish standardbreeds and ponies. The level of infection was unaffected by the usage of anthelminthics against nematodes. Of the horses examined 51% had 1-100 worms whereas 14% were infected with more than 100 worms. Of the tapeworm positive horses 72% had mixed infections with both adult and juvenile worms, 20% solely juveniles, and 8% solely adults. The severity of intestinal lesions exacerbated by increasing numbers of A. perfoliata. About 11% of the intestines examined had severe lesions, but there was no history of acute abdominal distress in any of the horses included in this study. Although the number of detectable eggs was significantly higher for horses heavily infected with A. perfoliata, the egg recovery among infected horses was only 35%. An additional field survey comprising 218 horses on 88 premises in central and southern parts of Sweden showed that the prevalence of A. perfoliata egg positive horses was the same as found on faecal examination during the abattoir survey.  相似文献

Fifty six dogs of mixed age and sex were acquired from farms in the Otago/Southland region, and maintained at the Hydatid Research Unit, Taieri, where 43 were each fed two Taenia ovis cysts. All were bled fortnightly for six or 12 weeks. Coded sera were sent to Wallaceville Animal Research Centre for testing using ELISA, with antigen from T. ovis scoleces. Dog treatments were identified after all tests were complete. A discriminant level was derived from the mean absorbance value plus three standard deviations of 56 sera taken at time zero and 78 sera from serially bled uninfected dogs. None of these 134 sera registered as a false positive using this discriminant level. The data showed no significant deviation from normality, and the expected frequency of the occurrence of false positives is therefore less than 0.14%. Four weeks after infection 63% of dogs proved to be infected were serologically positive, rising to 78% after 6 weeks. When worms were removed by anthelmintic treatment, ELISA absorbance levels decreased. Four weeks after removal 70% of previously infected dogs remained positive, decreasing to 30% after 6 weeks.

Six weeks after infection the sensitivity of the test was 78%, and the specificity 63%. However, if dogs with positive ELISA absorbance levels, but which did not purge worms, were regarded as having had worms, the respective figures would be 82% and 100%. The latter figures are similar to our previously published laboratory results. The test is of comparable efficiency to arecoline purgation for surveillance, and has the additional advantage of detecting infection in the majority of those dogs that have been infected for three weeks or more but fail to pass worms on purgation, and a substantial proportion of those infected dogs that were treated by their owners prior to presenting them for purgation in order to avoid detection of infection.  相似文献

The importance of preparation technique, culture media and incubation time in the embryonation of the infective egg stages of the intestinal nematode parasite Heterakis gallinarum was studied. Mature H. gallinarum worms were isolated from the caeca of infected chickens and separated by sex. In a first experiment intact female worms were kept for the development of their eggs in four different media (0.5% formalin, 2% formalin, 0.1 N sulphuric acid, 0.1% potassium dichromate) and incubated under constant temperature (20-22 degrees C) for 2, 4, 6 or 8 weeks. Afterwards the body of the worms were ruptured and the numbers of unembryonated and embryonated eggs were determined using a McMaster egg counting chamber, and the percentage of embryonated eggs was calculated. After 8 weeks of incubation in 0.5% formalin, 0.1 N sulphuric acid or 0.1% potassium dichromate 27.6%, 26.7% and 29.4% of the eggs, respectively, embryonated into third stage larvae (p > 0.05). In contrast, incubation in 2% formalin resulted in an embryonation of 18.6% only (p < 0.05). In a second experiment H. gallinarum eggs were directly harvested from worm uteri and cultivated afterwards in different media (2% formalin, 0.1 N sulphuric acid, 0.1% potassium dichromate) at 20 to 22 degrees C for 6 weeks. An incubation of isolated eggs in 2.0% formalin or 0.1% potassium dichromate during 6 weeks resulted in a significantly higher percentage of embryonation in comparison to the incubation of intact worms (first experiment). The results suggest that preparation technique, media and time of incubation has an essential influence on the development rate of H. gallinarum eggs.  相似文献

Sheep aged 9–10 months were vaccinated with three doses of 20 000 irradiated Trichostrongylus colubriformis larvae at fortnightly intervals. This gave 97–99% protection against subsequent “impulse” challenge with 40 000 normal larvae or “sequential” challenge with 2 000 larvae/week-day for 4 weeks.The sheep were protected against both types of challenge when worms from the immunizing infections were present at the time of challenge, and when these worms were first removed by anthelmintic treatment.  相似文献

The whipworms Trichuris trichiura and Trichuris suis in humans and pigs, respectively, are believed to be two different species yet closely related. Morphologically, adult worms, eggs and larvae of the two species are indistinguishable. The aim of this study was to examine the genetic variation of Trichuris sp. mainly recovered from natural infected pigs and humans. Worm material isolated from humans and pigs living in the same geographical region in Uganda were analyzed by PCR, cloning and sequencing. Measurements of morphometric characters were also performed. The analysis of the ITS-2 (internal transcribed spacer) region showed a high genetic variation in the human-derived worms with two sequence types, designated type 1 and type 2, differing with up to 45%, the type 2 being identical to the sequence found in pig-derived worms. A single human-derived worm showed exclusively the type 2-genotype (T. suis-type) and three cases of 'heterozygote' worms in humans were identified. However, the analysis showed that sympatric Trichuris primarily assorted with host origin. Sequence analysis of a part of the genetically conserved β-tubulin gene confirmed two separate populations/species but also showed that the 'heterozygote' worms had a T. suis-like β-tubulin gene. A PCR-RFLP on the ITS-2 region was developed, that could distinguish between worms of the pig, human and 'heterozygote' type. The data suggest that Trichuris in pigs and humans belong to two different populations (i.e. are two different species). However, the data presented also suggest that cross-infections of humans with T. suis takes place. Further studies on sympatric Trichuris populations are highly warranted in order to explore transmission dynamics and unravel the zoonotic potential of T. suis.  相似文献

Tests were carried out on the efficacy of Cambendazole (5-isopropoxy carbonylamino-2-(4-thiazolyl) benzimidazole) against lungworm and gastrointestinal parasites of calves and lambs. Against Dictyocaulus viviparus the compound was highly effective (80%) against mature worms at levels of 35 mg/kg and over and immature “arrested” worms at 60 mg/kg (71% efective).

In the Calves the compound was very effective (90 to 99%) against adult Haemonchus placei, Ostertagia spp., Trichostrongylus axei and Cooperia oncophora at all levels tested (15, 20, 25, 30, 40 and 60 mg/kg). Against adult Nematodirus spp. dosages of 30, 40 and 60 mg/kg were 81%, 94% and 99% effective respectively

Against arrested Ostertagia spp. larvae, a dosage of 60 mg/kg was 90% effective. Immature stages of Cooperia spp. were susceptible at all levels used but those of Nematodirus spp. required levels of 60 mg/kg for 99% removal.

In the lambs the compound was highly effective against immature and mature nematodes when given orally at all levels tested from 15 mg/kg to 30 mg/kg. Significant reductions (81%) in Moniezia expansa scolices were also observed at dosages from 15 mg/kg to 30 mg/kg.

Metacestodes of Neogryporhynchus cheilancristrotus (Wedl, 1855) were found in the gut of some gibel carp (Carassius gibelio) specimens from a Hungarian water reservoir. Location of metacestodes in the freshly opened gut was marked with disseminated, red-coloured, pinhead-sized nodules in the anterior part of the intestine. In histological sections, metacestodes were found in a hole inside the propria layer of the intestinal folds. The worms were in direct contact with the host tissue without being encapsulated as a result of host reaction. In some specimens with extruded rostellum the rostellar hooks were bored into the host tissue and suckers grabbed pieces of the surrounding connective tissue. Around the worms, congested capillaries and formation of macrophages were seen in the lysed connective tissue.  相似文献
