选择犬的三个STR基因座,复合扩增后,使用PED377自动测序仪及Genescan3.1.2软件进行电泳及数据分析,共检测了57例无新缘关系的犬,得到三个基因座的个体识别率(DP)分别为0.4217、0.8963、0.8095,多太信息含量(PIC)分别为:0.3339、0.7381、0.5979,及三个位点的联合个体识别率(TDP)为0.9886。这为犬的群体遗传学研究,司法鉴定等方面提供了方向。  相似文献

Although 36 DLA-DRB1 and 10 DLA-DQA1 allele sequences have been published to date, no data on individual allele frequencies exists, either for specific breeds or cross breeds, and the full extent of the polymorphism at each of these loci is still not known. We have used sequence-specific oligonucleotide probing (SSOP) to characterise a series of 367 dogs for their DRB1 and DQA1 alleles. These included individual animals from over 60 different breeds, with numbers per breed ranging from 1 to 39. DLA types were generated from 218 dogs for DRB1 and from 330 dogs for DQA1, while 181 dogs were characterised for both these loci. The frequency of individual DRB1 and DQA1 alleles showed considerable interbreed variation, e.g. 83% of West Highland White Terriers were DRB1*01 as opposed to 9% of Collies. No breed had >9 of the 22 DRB1 types defined in this study; several breeds had only two DRB1 types. DLA-DQA1 showed less variation in allele numbers per breed, but also showed considerable interbreed frequency variation. Haplotype analysis revealed over 44 different DRB1/DQA1 combinations. Of these, 25 were in a number of animals, and also in an animal that was homozygous for one or both of these loci. Some DRB1 alleles could be found in combination with several different DQA1 alleles, while others were only present in one haplotypic combination. DLA allele frequency data in normal dogs will be critical for disease association studies. It may also be possible to use haplotype data to establish the genetic relationships between different dog breeds.  相似文献

Genetic diversity was compared among eight dog breeds selected primarily for conformation (Standard Poodle, Italian Greyhound and show English Setter), conformation and performance (Brittany), predominantly performance (German Shorthaired and Wirehaired Pointers) or solely performance (field English Setter and Red Setter). Modern village dogs, which better reflect ancestral genetic diversity, were used as the standard. Four to seven maternal and one to two Y haplotypes were found per breed, with one usually dominant. Diversity of maternal haplotypes was greatest in village dogs, intermediate in performance breeds and lowest in conformation breeds. Maternal haplotype sharing occurred across all breeds, while Y haplotypes were more breed specific. Almost all paternal haplotypes were identified among village dogs, with the exception of the dominant Y haplotype in Brittanys, which has not been identified heretofore. The highest heterozygosity based on 24 autosomal microsatellites was found in village dogs and the lowest in conformation (show) breeds. Principal coordinate analysis indicated that conformation‐type breeds were distinct from breeds heavily used for performance, the latter clustering more closely with village dogs. The Brittany, a well‐established dual show and field breed, was also genetically intermediate between the conformation and performance breeds. The number of DLA‐DRB1 alleles varied from 3 to 10 per breed with extensive sharing. SNPs across the wider DLA region were more frequently homozygous in all pure breeds than in village dogs. Compared with their village dog relatives, all modern breed dogs exhibit reduced genetic diversity. Genetic diversity was even more reduced among breeds under selection for show/conformation.  相似文献

Primary ciliary dyskinesia was diagnosed in three Newfoundland dogs with histories of chronic rhinitis and bronchopneumonia from an early age. Thoracic radiographs of two of them showed severe, dependent bronchopneumonia and right displacement of the cardiac apex but normal positioning of other organs. Histopathological examination of sections of lung from the other dog showed severe bronchopneumonia. A semen sample from one dog had a high percentage of spermatozoa with abnormal tails and poor progressive motility. Transmission electron microscopy of nasal brushings from all three dogs showed consistent ultrastructural defects in the cilia, including an absence of outer and inner dynein arms, disorganisation of peripheral doublets, occasional supernumerary doublets and singlets, and consistently disorganised basal bodies and foot processes; sections of trachea from one dog also had disorganised basal bodies. Pedigree analysis was consistent with a monogenic autosomal recessive pattern of inheritance for the defect. One dog is still alive, one dog died aged five years two months, and one dog was euthanased aged nine months. This is the first time primary ciliary dyskinesia has been reported in Newfoundland dogs.  相似文献

From May 2007 to March 2008, blood samples were collected from 92 healthy dogs living in 21 households (17 farms in rural area, and 4 homes in urban area) in 6 counties of the State of Espírito Santo, southeastern Brazil. In addition, ticks were collected from these dogs. A mean of 4.4 ± 3.0 dogs (range: 1–12) were sampled per household; 78 and 14 dogs were from rural and urban areas, respectively. Polymerase chain reaction (PCR) designed to amplify fragments of the 18S rDNA gene of Babesia spp or Hepatozoon spp revealed amplicons of the expected size in 20 (21.7%) dogs for Babesia, and 54 (58.7%) dogs for Hepatozoon. All Babesia-positive dogs were also Hepatozoon-positive. Among the 21 households, 15 (71.4%) from 3 counties had at least one PCR-positive dog, including 13 farms (rural area) and 2 homes (urban area). A total of 40 PCR products from the Hepatozoon-PCR, and 19 products from the Babesia-PCR were submitted to DNA sequencing. All generated sequences from Hepatozoon-PCR were identical to each other, and to corresponding 18S rDNA sequences of H. canis in GenBank. Surprisingly, all generated sequences from the Babesia PCR were also identical to corresponding 18S rDNA sequences of H. canis in GenBank. Dogs from 10 rural and 2 urban households were found infested by Rhipicephalus sanguineus ticks. Immature of Amblyomma cajennense ticks were found in dogs from only 4 rural households (also infested by R. sanguineus). All but one household with R. sanguineus-infested dogs had at least one Hepatozoon-infected dog. Statistical analysis showed that the presence of ticks (i.e. R. sanguineus) infesting dogs in the households was significantly (P < 0.05) associated with at least one PCR-positive dog. There was no significant association (P > 0.05) between PCR-positive dogs and urban or rural households. Canine hepatozoonosis caused by H. canis is a high frequent infection in Espírito Santo, Brazil, where it is possibly vectored by R. sanguineus. Since all infected dogs were found apparently healthy, the pathogenicity of H. canis for dogs in Espírito Santo is yet to be elucidated.  相似文献

OBJECTIVE: To evaluate the breed distribution of the ABCB1-1Delta polymorphism in a large number of dogs in North America, including dogs of several herding breeds in which this polymorphism has been detected and other breeds in which this polymorphism has not yet been identified. DESIGN: Cross-sectional study. ANIMALS: 5,368 dogs from which buccal swab samples were collected for purposes of ABCB1 genotyping. PROCEDURES: From May 1, 2004, to September 30, 2007, DNA specimens derived from buccal swab samples collected from 5,368 dogs underwent ABCB1 genotyping. These data were reviewed, and results for each dog were recorded in a spreadsheet, along with the dog's breed. The genotypes for each breed were tallied by use of a sorting function. RESULTS: The ABCB1-1Delta allele was identified in 9 breeds of dogs and in many mixed-breed dogs. Breeds that had the ABCB1-1Delta allele included Collie, Longhaired Whippet, Australian Shepherd (standard and miniature), Shetland Sheepdog, Old English Sheepdog, Border Collie, Silken Windhound, and German Shepherd Dog (a breed in which this mutation had not been detected previously). CONCLUSIONS AND CLINICAL RELEVANCE: The ABCB1-1Delta polymorphism is associated with increased susceptibility to many adverse drug reactions and with suppression of the hypothalamic-pituitary-adrenal axis and is present in many herding breeds of dog. Veterinarians should be familiar with the breeds that have the ABCB1-1Delta polymorphism to make appropriate pharmacologic choices for these patients.  相似文献

: Infection with Angiostrongylus vasorum was diagnosed at necropsy on a dog that died from acute pulmonary haemorrhage, and on recovery of L1 larvae by Baermann examination of faeces from two dogs, one of which had abdominal pain and retroperitoneal haemorrhage, while the other had right-sided heart failure due to cor pulmonale. The presenting signs included syncope (one dog), exercise intolerance (two dogs), cough (two dogs), abdominal pain (one dog) and depression (one dog). One-stage prothrombin time and activated partial thromboplastin time were prolonged in two dogs, buccal mucosal bleeding time was prolonged in one dog and globulin was elevated in all three dogs. In this study the pedigrees of 42 German Shepherd dogs with German Shepherd dog Pyoderma (GSP) were analysed. Parents, littermates and offspring of the affected dogs were traced and their owners were questioned on characteristic skin lesions. Evidence suggesting an autosomal recessive trait was found. The purpose of this study was to investigate the diversity of yeast associated with the degree of canine seborrheic dermatitis (SD) by anatomical sites. Fifty-seven samples were divided as 17 healthy skin, 20 with primary seborrheic dermatitis (PSD), and 20 with secondary seborrheic dermatitis (SSD). Yeast isolation and characterization were carried out based on microscopical features and biochemical properties. DNA analysis at the internal transcribed spacer I of 26S rDNA region was utilized for species confirmation. Four species of yeast consisting Malassezia pachydermatis, Malassezia furfur, Candida parapsilosis and Candida tropicalis recovered from examined dogs. M. pachydermatis and C. parapsilosis were isolated from all dogs, but C. tropicalis and M. furfur were recovered from 3 healthy dogs and one diseased dog, respectively. The number of M. pachydermatis and C. parapsilosis in diseased dogs was higher than that of healthy specimens (P<0.01). High frequency and population size of C. parapsilosis were closely associated to PSD, while those of M. pachydermatis were associated with both PSD and SSD (P<0.01). C. parapsilosis were predominant at the perianal area. This study demonstrated the co-colonization of M. pachydermatis and C. parapsilosis in large amounts and frequency associated with stage of disease and anatomical site.  相似文献

OBJECTIVE: Testing of the cataract-causing insertion/deletion mutation in the canine HSF4 gene for its linkage and association with primary cataracts (CAT) in Dachshunds and Entlebucher Mountain dogs. MATERIALS: Exon 9 with flanking intronic regions of the canine HSF4 gene was sequenced in 24 Dachshunds and 20 Entlebucher Mountain dogs. The HSF4 cDNA sequence of lens tissue was analyzed in a CAT-unaffected mixed-breed dog and in three CAT-affected dogs of different breeds, including a Wire-haired Dachshund, a Dachshund-mix and a German Shepherd dog. RESULTS: In all dogs investigated here, the previously reported CAT-causing mutation did not exist. We found a single nucleotide polymorphism (SNP) in intron 9, which was neither associated nor linked with the CAT phenotype in the two dog breeds. CONCLUSION: The CAT phenotype in the two dog breeds investigated here was not caused by the same mutation found to be associated with early-onset CAT in the Staffordshire Bull Terrier and Boston Terrier. The intronic SNP may be useful to test HSF4 for linkage with CAT in further dog breeds.  相似文献

Dilated cardiomyopathy (DCM) is a heart disease which is often found in humans and animals. The age of onset of this progressive disease varies between 3 and 7 years of age. A juvenile form of DCM has been found in Portuguese Water Dogs and Doberman Pinscher Dogs. Some breeds such as Doberman pinscher, Newfoundland, Portuguese Water dog, Boxer, Great Dane, Cocker Spaniel and Irish Wolfhound exhibit a higher prevalence to DCM. There also seems to be a sex predisposition as male dogs are affected more often than female dogs and in Great Danes an X-linked recessive inheritance is likely. In Newfoundland and Boxer an autosomal dominant inheritance was found whereas an autosomal recessive inheritance was described in Portuguese Water Dogs. Atrial fibrillation as a cause or consequence of DCM is assumed for certain breeds. The causes of DCM are widely unknown in dogs. A genetic basis for this heart disease seems to exist. Apart from a few exceptions the mode of inheritance and the possible underlying gene mutations are not known for DCM in dogs. In humans mutations in several genes responsible for DCM have been identified. Comparative genetic analyses in dogs using genes causing DCM in men and a genome-wide scan with anonymus markers were not able to detect causative mutations or genomic regions harboring gene loci linked to DCM. The investigation of the genetic basis of canine DCM may lead to new insights into the pathogenesis of DCM and may result in new therapeutic approaches and breeding strategies.  相似文献

The incidence of pancreatic degenerative atrophy (PDA) was investigated in 59 German shepherd dogs from two kindred. The male progenitors were the same in both kindred. In the four litters of the first kindred the incidence of PDA was 24 % (10 dogs out of 41), and there was at least one affected dog in each litter. When one of the litter bitches, later affected with PDA, was mated with one of the obligate carriers of PDA, one of the resultant seven offspring has so far been found to suffer from PDA. In the second kindred when a PDA-affected bitch and a clinically healthy male (heterozygote) were mated, two of the resultant six offspring were found to suffer from PDA.These results indicate that PDA is a disease inherited as an autosomal recessive trait, although the possibility of dominant inheritance with incomplete penetrance cannot be excluded.  相似文献

Ribosomal RNA gene (rDNA) loci of Russian Mus musculus musculus and of Japanese Mus musuculus molossinus were mapped by double color FISH. The total number of rDNA loci was varied from 5 to 12, although the loci on chromosomes 12, 15, 16, 18, and 19 were common to all mice examined. Instead, polymorphisms of the rDNA loci were found on chromosomes 1, 5, 8, 9, 10, 11, 13 and 17. The novel rDNA loci of M. m. musculus were found in Nov/TUA strain on chromosomes 8 and 17. These observations, together with those of previous reports, suggest that the rDNA loci of Mus musculus species are in the evolutionary process of further translocation to other chromosomes.  相似文献

The issues surrounding dog bites are a major public health concern, particularly in areas of low income where accessibility to adequate health care, veterinary medicine and sufficient management of canine population control is low. An understanding of the risk factors associated with human-dog conflicts may be important when establishing dog bite and disease prevention strategies. In May 2008, a census of 12 consociated neighbourhoods in Todos Santos, Guatemala was conducted to investigate dog bite incidences and the public perception of free-roaming dog populations. Approximately 16.5% (78/472) of households reported at least one dog bite between May 2006 and May 2008. In total, 85 incidents occurred: 49.4% (42/85) with adults (≥18 years) and 50.6% (43/85) children (<18 years). However, there was no significant difference in cumulative incidence of dog bites by victim gender or among age categories, there was a non-significant trend of higher cumulative incidence of dog bites in children aged six to 17 years compared to other age categories. The anatomical location of the bite varied, but bites to the legs were the most common (73/85; 85.9%). Of the 85 reported dog bites, 5.9% (5/85) were from dogs from the victims' own households, 48.2% (41/85) were from a neighbour's dog, 9.4% (8/85) were from dogs regularly seen in the community, and 15.3% (13/85) were from dogs not regularly seen in the community; the ownership status of the latter two categories of dogs could not be determined. Approximately 21% (18/85) of respondents did not know the type of dog that bit. Residents were asked for their opinions on potential problems associated with dogs in the community. The majority of respondents strongly agreed that dogs posed physical risks (78.8%; 372/472), could transmit infections to people (88.6%; 418/472), scared the family (82.4%; 389/472) and were too high in number (82.6%; 390/472). There were significant but weak correlations between owning a dog and expressing negative perceptions of community dogs (Spearman rho<0.13). Reporting of a dog bite was not significantly correlated to expressing fear or negative perceptions towards community dogs. A further understanding of current programs directed at the prevention of dog bites and means of dog population control may help determine appropriate future steps for canine management. Conflicts between free roaming dogs and people are a pressing issue worldwide, and a focus on prevention strategies through education, rather than rapid extermination, would be of benefit.  相似文献

To identify the incidence of Babesia gibsoni (B. gibsoni) in Aomori Prefecture, northeastern Japan, dogs with acute B. gibsoni infection were investigated at the Animal Teaching Hospital, Kitasato University, between April 2002 and March 2003. Eighteen dogs with acute B. gibsoni infection were recognized; they were all male dogs of the fighting dog breed Tosa. Their platelet counts were below normal and their packed cell volumes (PCVs) were at various levels. We collected blood samples from 141 Tosa dogs from Aomori Prefecture and used polymerase chain reaction assay to investigate the incidence of subclinical B. gibsoni infection. We also looked into the serological abnormalities associated with thrombocytopenia or anemia in subclinical infection. Forty-one of 87 dogs (47.1%) with histories of dog fighting, and one dog of 54 without a history of dog fighting were positive for B. gibsoni; that is, 42 of 141 dogs (29.8%) showed a positive result. The mean platelet counts of dogs with subclinical infection were significantly lower and levels of anti-platelet IgG were significantly higher than levels for dogs without infection. Anti-erythrocyte membrane IgG levels were significantly higher in dogs with subclinical infections, although mean PCVs were not significantly different. Tosa dogs from Aomori Prefecture, Japan, were highly infected with B. gibsoni subclinically and this pathogen might be successfully transmitted during dog fighting. Dogs with subclinical infections were at risk of chronic thrombocytopenia, which may be due to autoimmune mechanisms.  相似文献

A novel surgical approach, using portal venotomy during total hepatic vascular occlusion, was used to locate and attenuate congenital intrahepatic portosystemic shunts in nine dogs. Shunt location was consistent with a persistent ductus venosus in only two dogs. In the remaining seven dogs the shunts were window-like orifices arising from either the left (two dogs) or right portal vein branch (five dogs) and communicating with the ipsilateral hepatic vein or caudal vena cava. The transportal approach using total hepatic vascular occlusion consistently provided good access to the portosystemic shunts, including those with window-like communications. A 7 to 16 minute period of total vascular occlusion was well-tolerated hemodynamically, with few intraoperative complications. Intrahepatic shunts were successfully attenuated in eight dogs, while one dog with portal atresia was euthanatized. The postoperative course was complicated by high protein pulmonary edema (one dog), an encapsulated biliary pseudocyst (one dog) and uncontrollable hemorrhage caused by an uncharacterized coagulopathy (one dog). Three dogs required a second operation to further attenuate their shunts. The clinical condition of all seven surviving dogs was improved after surgery.  相似文献

OBJECTIVE: To characterise Australian domestic dogkeeping practices relevant to canine health and welfare. PROCEDURE: A quantitative questionnaire was distributed nationally through Dog's Life magazine. Data on management practices, household and dog demographics from 690 Australian dogs in 485 self-selected respondent households were analysed using ordinal and binary regression. RESULTS: Several variables, relating to dog, owner and environment were associated with management practices. The size of the dog correlated positively with being fed outdoors (P < 0.001), gameplaying (P = 0.016) and being trained (P = 0.001) and negatively with time spent indoors (P < 0.001). Neutered dogs were more likely to be walked every day (P = 0.027) and have had prior treatment for unwelcome behaviours (P < 0.001) but less likely to be fed outdoors (P < 0.013). The number of children in the household correlated with the dog being fed outdoors (P < 0.001). The number of adult female humans in the household correlated with dog being neutered (P < 0.001) whereas the reverse was linked to number of adult male humans in the household (P < 0.013). Larger numbers of adult female humans in the household were associated with a higher incidence of prior treatment for unwelcome behaviours (P = 0.020). The number of dogs in the household correlated negatively with dogs being walked every day (P < 0.004) and gameplaying (P = 0.001). Pure bred dogs were more likely to have attended puppy preschool than cross breeds (P = 0.002). CONCLUSION: Reference to previous studies of Australian dog owners suggests that the current data are representative in terms of location of households, sex and age of dogs, however, the proportion of pure bred dogs in the current survey was surprisingly high. This was perhaps because respondents were from a particular sector of the Australian dog-owning population. It must be acknowledged that Dog's Life magazine readers may be atypical of Australian dog owners, nevertheless, the study demonstrates that variables relating to the dog, owner or environment can be used to characterise management styles.  相似文献

Microscopic examination of Giemsa-stained peripheral blood smears collected from three naturally infected dogs originating from Turkey revealed the presence of large (around 4.5-5.0 microm) intraerythrocytic Babesia parasites in all dogs. DNA was extracted from the three infected blood samples and an around 410 bp portion of the 18S rDNA gene of Babesia species was PCR amplified for subsequent molecular characterization. RFLP analysis of the PCR products suggested the presence of the species B. vogeli in all infected dogs and sequencing of the PCR products from two of the three samples revealed 100% identity among the two Turkish isolates. Comparisons with the equivalent 410 bp portions of the 18S rDNA gene of Babesia species confirmed the affiliation of these isolates to the B. vogeli species. This is the first report and molecular characterization of dog infection with a large Babesia species in Turkey.  相似文献

The medical records of seven dogs with severe, grade 3, open shearing wounds of the carpus or tarsus that were treated with an immediate arthrodesis were reviewed. Six dogs were managed with a transarticular external skeletal fixator (ESF), and one dog was treated with plate fixation. The soft tissues were managed simultaneously along with the definitive joint stabilization in all cases. Minor complications occurred in four dogs: one dog that required a skin graft, one dog in which a skin graft was recommended but not performed, one dog in which a secondary skin closure was performed, and one dog in which a delayed cancellous bone graft was placed. Major complications occurred in three dogs: two dogs that required restabilization of the arthrodesis and one dog that required implant (i.e., plate) removal due to infection. All dogs healed with acceptable functional and cosmetic results. Further long-term evaluation of five dogs revealed that all but one dog had either excellent or good functional outcomes, and the remaining dog had a fair outcome. Similarly, five dogs had either excellent or good cosmetic outcomes, with the remaining dog having a fair outcome. All owners were very satisfied with the overall results. This clinical study demonstrated that an immediate, definitive management technique, in which bone fusion and wound management are undertaken as simultaneous objectives, is a viable technique of managing severe periarticular shearing injuries. Transarticular stabilization with an ESF is the technique recommended.  相似文献
