Populations of Choristoneura rosaceana (Harris) from orchards in Ontario were shown to be resistant to azinphos-methyl and to other types of organophosphorus insecticides. Resistance extended to methyl carbamates and to methomyl. The laboratory population used for these assays and selected with azinphosmethyl was also resistant to the pyrethroid, cypermethrin. Resistance was associated with increased esterase activity and was reduced by the addition of the synergist DEF. IEF studies of esterases also indicated increased activity in resistant populations, but did not identify any unique esterases associated with the resistance. Resistance was highly correlated (r = 0.78) with elevated esterases but not with increased glutathione-S transferase activity (r = 0.13). Other mechanisms did not appear to be related. 相似文献

The interactions between six insecticides (indoxacarb, cypermethrin, chlorpyrifos, azinphosmethyl, tebufenozide and chlorfenapyr) and three potential synergists, (piperonyl butoxide (PBO), S,S,S-tributyl phosphorotrithioate (DEF) and diethyl maleate (DEM)) were studied by dietary exposure in a multi-resistant and a susceptible strain of the obliquebanded leafroller, Choristoneura rosaceana (Harris). The synergists did not produce appreciable synergism with most of the insecticides in the susceptible strain. Except for tebufenozide, PBO synergized all the insecticides to varying degrees in the resistant strain. A very high level of synergism by PBO was found with indoxacarb, which reduced the resistance level from 705- to 20-fold when PBO was administered alone and to around 10-fold when used in combination with DEF. DEF also synergized indoxacarb, cypermethrin, chlorpyrifos, azinphosmethyl and tebufenozide in the resistant strain. DEM produced synergism of indoxacarb, chlorpyrifos, azinphos-methyl and chlorfenapyr in the resistant strain. DEM was highly synergistic to cypermethrin, and to some extent to tebufenozide in both the susceptible and resistant strains equally, implying that detoxification by glutathione S-transferases was not a mechanism of resistance for these insecticides. The high level of synergism seen with DEM in the case of cypermethrin may be due to an increase in oxidative stress resulting from the removal of the antioxidant, glutathione. These studies indicate that enhanced detoxification, often mediated by cytochrome P-450 monooxygenases, but with probable esterase and glutathione S-transferase contributions in some cases, is the major mechanism imparting resistance to different insecticides in C. rosaceana. 相似文献

The effects of sub-lethal residues of azinphos-methyl on pheromone production, calling, female attractiveness and the ability of males to locate sources of natural and synthetic pheromone were compared in azinphos-methyl-susceptible (susceptible) and azinphos-methyl-resistant (resistant) obliquebanded leafrollers, Choristoneura rosaceana (Harris). The amount of pheromone in susceptible females was reduced by 29-33% after exposure to azinphos-methyl; this treatment did not affect the pheromone content of resistant females. Azinphos-methyl-treated resistant females contained 39-43% less pheromone than azinphos-methyl-treated susceptible females. Resistant females that were not treated with azinphos-methyl contained 35-56% less pheromone than susceptible females that were not treated with insecticide. The incidence of calling was reduced by 67-100% in azinphos-methyl-treated susceptible females; the incidence of calling by resistant females was not affected by exposure to azinphos-methyl. The incidence of calling by azinphos-methyl-treated susceptible females was 58-100% lower than that of azinphos-methyl-treated resistant females. There was no difference in the incidence of calling between susceptible and resistant females that had not been treated with insecticide. In a flight tunnel, treatment with insecticide reduced the attractiveness of susceptible females by 38%; treatment with insecticide did not affect the attractiveness of resistant females. There was no difference in the proportion of males attracted to susceptible and resistant females that had, or had not been treated with insecticide. In an apple orchard, the attractiveness of susceptible and resistant females treated with azinphos-methyl was reduced by 84 and 12%, respectively. The proportion of males attracted to azinphos-methyl-treated susceptible females was 58% lower than the proportion attracted to azinphos-methyl-treated resistant females, whereas, if females were not treated with insecticide, the proportion attracted to resistant females was 57% lower than the proportion attracted to susceptible females. In a flight tunnel, azinphos-methyl did not affect the ability of susceptible or resistant males to locate a source of pheromone gland extract. Likewise, in an apple orchard, the insecticide treatment had no effect on the ability of susceptible or resistant males to locate a source of synthetic pheromone. In a flight tunnel, there was no difference in the proportion of azinphos-methyl-treated susceptible and resistant males locating a source of pheromone gland extract; however, in the orchard, 39% fewer azinphos-methyl-treated resistant males located a source of synthetic pheromone than azinphos-methyl-treated susceptible males. A similar proportion of susceptible and resistant males that had not been treated with insecticide located a source of pheromone gland extract in the flight tunnel, but in the orchard, the proportion of resistant males not treated with azinphos-methyl that located the source of synthetic pheromone was 32% lower than the proportion of susceptible males not treated with this insecticide. The implications of the differences in the effect of sub-lethal residues of azinphos-methyl on the pheromone communication system of susceptible and resistant moths are discussed in relation to the theory of the development of insecticide resistance, the detection of resistance in feral populations of moths using sex pheromone-baited traps, and the control of moths using sex pheromone-mediated mating disruption. 相似文献

BACKGROUND: Obliquebanded leafroller, Choristoneura rosaceana (Harris), is an economic pest of pome fruits throughout North America. Repeated applications of broad‐spectrum insecticides have resulted in the development of resistance, cross‐resistance and multiple resistance in this pest. Studies were conducted to determine the possibility of resistance evolution, stability of resistance and the effect of metabolic synergists on toxicity of new reduced‐risk insecticides chlorantraniliprole and spinetoram in C. rosaceana. RESULTS: Larvae of C. rosaceana were selected for resistance to chlorantraniliprole and spinetoram in the laboratory. Significant levels of resistance to each insecticide were observed after 12 generations of selection. In the absence of selection pressure, susceptibility of a subset of larvae from both chlorantraniliprole‐ and spinetoram‐selected populations reverted to preselection levels after five and six generations respectively, indicating that resistance to both chlorantraniliprole and spinetoram was unstable in C. rosaceana. In synergist bioassays performed after 12 generations of selection, S,S,S‐tributylphosphoro trithioate (DEF) and piperonyl butoxide (PBO) synergized the toxicity of chlorantraniliprole and spinetoram respectively, suggesting the involvement of esterases in chlorantraniliprole resistance and the involvement of mixed‐function oxidases in spinetoram resistance. CONCLUSION: These findings suggest that chlorantraniliprole and spinetoram could be incorporated into C. rosaceana resistance management programs by using rotational strategies. BACKGROUND: Studies were designed to assess baseline toxicity of the two recently developed reduced‐risk insecticides chlorantraniliprole and spinetoram to obliquebanded leafroller (OBLR), Choristoneura rosaceana (Harris), and to determine stage‐specific effects of these products on OBLR, so that these new chemicals could be judiciously incorporated into IPM programs by targeting only the most susceptible stages of OBLR. RESULTS: Both chlorantraniliprole and spinetoram were highly effective against neonate OBLR larvae at much lower doses than conventional standard azinphosmethyl. Most of the mortality caused by spinetoram occurred during the 4 day exposure period, while significant delayed mortality was observed as a result of chlorantraniliprole treatment during the 7 day recovery period, indicating that chlorantraniliprole is a slower‐acting compound compared with spinetoram. Unlike broad‐spectrum predecessors, the toxicity of chlorantraniliprole significantly increased with larval age. Similarly, the toxicity of spinetoram increased as larvae grew from neonates to third instars. CONCLUSION: Chlorantraniliprole and spinetoram are highly effective against OBLR larvae, regardless of larval age. Incorporation of these reduced‐risk chemistries into IPM programs for OBLR would lead to effective management of this pest in tree fruit systems. Copyright © 2011 Society of Chemical Industry 相似文献

Neonate larvae of obliquebanded leafroller, Choristoneura rosaceana, from a laboratory colony were exposed to two reduced-risk insecticides, chlorantraniliprole and spinetoram. After nine generations of selection, significant levels of resistance to each insecticide were observed. Biochemical assays were performed on third instars to determine potential resistance mechanisms. Enzyme assays indicated that esterase activity was significantly increased in the chlorantraniliprole-selected colony, whereas mixed-function oxidase levels were elevated in the spinetoram-selected colony as compared to the unselected colony. No difference in glutathione-S-transferase activity was seen in either of the insecticide-selected colonies. These results indicate the potential involvement of esterases and mixed-function oxidases as detoxification mechanisms responsible for resistance to chlorantraniliprole and spinetoram, respectively. Furthermore, the results of this study suggest that chlorantraniliprole and spinetoram are not detoxified by similar mechanisms and could therefore be incorporated into resistance management programs in tree fruit leading to sustainable management of C. rosaceana. 相似文献

A laboratory study conducted in 2003 determined the influence of a kaolin-based particle film (Surround WP Crop Protectant) on spruce budworm (Choristoneura fumiferana (Clem)) oviposition. Three concentrations (15, 30 and 60 g litre(-1) spray carrier) were applied to white spruce (Picea glauca (Moench) Voss). When presented with different paired choices of oviposition surfaces, spruce budworms laid very few egg masses overall and showed no significant (P < 0.05) preference between kaolin-coated and untreated foliage. During no-choice tests, spruce budworms were significantly (P < 0.05) less inclined to oviposit on the 60 g kaolin litre(-1) treated foliage than on the controls, but no kaolin treatment completely inhibited spruce budworm oviposition. Egg mass size and percentage hatch were unaffected by the kaolin treatments, and overall the percentage of egg masses laid on foliage was inversely proportional to treatment concentration. It is unlikely that kaolin-based particle film would be practical for wide use in natural forests without significant adaptations to current pesticide application equipment and methods. The development of resistance in the Oriental fruit moth, Grapholita molesta (Busck) to organophosphorus (OP) insecticides (azinphos-methyl and phosmet) is a serious threat to the tender fruit industry in Ontario (50% crop losses in 1994). Resistance to carbamate insecticides and increased survival of field-collected moths at diagnostic concentrations of pyrethroids were widespread. As a result, four different treatment regimes were tested to manage resistance in G molesta, and the changes in resistance frequencies under each treatment regime were monitored from 1996 to 1999. The data indicated that the levels of resistance were significantly influenced by the various treatment regimes. The seasonal pattern of resistance was similar for all treatment regimes, in that resistance peaked in mid-season and declined in the late season. Levels of resistance in G molesta to OPs decreased from 55% to 14% and that to pyrethroids declined from 30% to 10% from 1996 to 1999 under a treatment regime consisting of endosulfan-organophosphate-pyrethroid rotation. Similarly, under a treatment regime implemented in commercial orchards (organophosphate-pyrethroid rotation), resistance to OP insecticides declined from 50% to 12% and resistance to pyrethroids evolved to around 16%. The overall data indicated that resistance was unstable; a strategy based on rotation of insecticides by class for each generation of G molesta was successful in managing resistance to both OP and pyrethroid insecticides. Spanish Cydia pomonella (L.) field populations have developed resistance to several insecticide groups. Diagnostic concentrations were established as the LC90 calculated on a susceptible strain (S_Spain) for five and seven insecticides and tested on eggs and neonate larvae field populations, respectively. The three most relevant enzymatic detoxification systems (mixed-function oxidases (MFO), glutathione S-tranferases (GST) and esterases (EST)) were studied for neonate larvae.In eggs, 96% of the field populations showed a significantly lower efficacy when compared with the susceptible strain (S_Spain) and the most effective insecticides were fenoxycarb and thiacloprid. In neonate larvae, a significant loss of susceptibility to the insecticides was detected. Flufenoxuron, azinphos-methyl and phosmet showed the lowest efficacy, while lambda-cyhalothrin, alpha-cypermethrin and chlorpyrifos-ethyl showed the highest. Biochemical assays showed that the most important enzymatic system involved in insecticide detoxification was MFO, with highest enzymatic activity ratios (5.1-16.6 for neonates from nine field populations). An enhanced GST and EST activities was detected in one field population, with enzymatic activity ratios of threefold and fivefold for GST and EST, respectively, when compared with the susceptible strain. The insecticide bioassays showed that the LC90 used were effective as diagnostic concentrations. Measures of MFO activity alongside bioassays with insecticide diagnostic concentrations could be used as tools for monitoring insecticide resistance in neonate larvae of C. pomonella. 相似文献

A continuous programme for monitoring resistance of Helicoverpa armigera (Hubner) to commonly used insecticides was undertaken for assessing the impact of Bt cotton planting on the evolution of the pest resistance from 1994 to 2002 in China. The bioassay results showed that the resistance levels per year in field populations of H armigera to lambda-cyhalothrin, phoxim and endosulfan decreased, respectively, to 9-15-fold, 11-14-fold and 6-8-fold in 2001-2002 from 197-262-fold, 52-74-fold and 18-38-fold before the cultivation of Bt cotton in 1997. This significant increase in susceptibility to insecticides is expected to result in a reduction in insecticide application for H armigera control in Bt cotton. It is concluded that Bt cotton is playing an important role in the long-term management of H armigera by increasing the potential for natural and chemical control of the pest. 相似文献

Two field strains of the fall armyworm, Spodoptera frugiperda (JE Smith), collected from corn in north Florida showed high resistance to carbaryl (626- and 1159-fold) and moderate resistance to parathion-methyl (30- and 39-fold) as compared with a laboratory susceptible strain. A field strain of the diamondback moth, Plutella xylostella (L.) collected from cabbage in north Florida and selected for 20 generations with permethrin showed high resistance to permethrin (987-fold) as compared with a susceptible strain. However, in all instances, no cross-resistance to indoxacarb, a novel oxidiazine insecticide, was observed in these two species. Biochemical studies revealed that, in S. frugiperda, activities of detoxification enzymes (microsomal oxidase, glutathione S-transferase and general esterase) were significantly higher in the field strains than in the susceptible strain, indicating that these detoxification enzymes were not actively involved in the resistance to indoxacarb. The lack of cross-resistance between indoxacarb and permethrin in P. xylostella further supports the notion that the mode of action of these insecticides on the insect sodium channel is different. 相似文献

The damson hop aphid, Phorodon humuli (Schrank) (Hemiptera: Aphididae), is one of the most important sucking pests of many hop-growing areas world-wide. In this study we determined the efficacy of several insecticides against strains collected throughout the year 2001. All strains were collected in different hop gardens in the Hallertau (Bavaria), Germany, the largest hop-growing area of the world. First of all we established a leaf dip bioassay, carried out using six-well tissue culture plates and appropriate for monitoring susceptibility against imidacloprid, oxydemeton-methyl, cyfluthrin, amitraz, pymetrozine and pirimicarb. Four of these compounds, imidacloprid, cyfluthrin, pymetrozine and amitraz, are currently registered for the control of sucking pests in German hop gardens and are useful against P. humuli. The leaf-dip bioassay system turned out to be very reliable and robust. Ten P. humuli strains were collected in May 2001 and maintained in the laboratory to assess their resistance to the different insecticides in comparison with two laboratory reference strains (H2 and H5). Using diagnostic concentrations, resistance monitoring for imidacloprid and cyfluthrin was investigated during July and August 2001 on 53 populations from 30 sites around the Hallertau, an area of ca 2500 km2. Resistance to diagnostic concentrations (LC95 for reference strains) of imidacloprid, amitraz and pymetrozine was not detected in any strain received in 2001, but late-season (August) populations seemed to respond more heterogeneously than those collected mid-season (July). Overall composite mean mortalities to diagnostic concentrations of imidacloprid (13 mg litre(-1)) in collections from May, July and August were 95 (+/-2.5), 98 (+/-2.3) and 87 (+/-5.9)%, respectively. Moderate resistance to pyrethroids was observed in all strains collected in May and August using a diagnostic concentrations of 10 mg litre(-1) cyfluthrin (LC95 of the susceptible reference strain H5). Slight to moderate resistance to diagnostic concentrations of oxydemeton-methyl and pirimicarb was observed in some, but not all, strains collected early season. BACKGROUND: Resistance to numerous insecticide classes in Bemisia tabaci Gennadius has impaired field control efficacy in south‐eastern China. The biotype and resistance status of B. tabaci collected from these areas was investigated. RESULTS: Two different biotypes of B. tabaci (B‐biotype and Q‐biotype) were detected in south‐eastern China, and the samples collected from geographical regions showed a prevalence of the Q‐biotype and the coexistence of B‐ and Q‐biotypes in some regions. Moderate to high levels of resistance to two neonicotinoids were established in both biotypes (28–1900‐fold to imidacloprid, 29–1200‐fold to thiamethoxam). Medium to high levels of resistance to alpha‐cypermethrin (22–610‐fold) were also detected in both biotypes. Four out of 12 populations had low to medium levels of resistance to fipronil (10–25‐fold). Four out of 12 populations showed low levels of resistance to spinosad (5.7–6.4‐fold). All populations tested were susceptible to abamectin. CONCLUSION: The Q‐biotype B. tabaci is supplanting the B‐biotype which used to be ubiquitous in China. Field populations of both B‐ and Q‐biotypes of B. tabaci have developed high levels of resistance to imidacloprid and thiamethoxam. In the susceptible strain PBO produced appreciable synergism only of formetanate, methiocarb and methamidophos. Except for endosulfan, PBO synergized all the insecticides to varying degrees in the resistant strains. A very high level of synergism by PBO was found with acrinathrin, which reduced the resistance level from 3344- to 36-fold. PBO slightly synergized the carbamates formetanate (4.6-fold) and methiocarb (3.3-fold). PBO also produced a high synergism of deltamethrin (12.5-fold) and methamidophos (14.3-fold) and completely restored susceptibility to both insecticides. DEF did not produce synergism with any insecticide in the resistant strains and DEM was slightly synergistic to endosulfan (3-fold). These studies indicate that an enhanced detoxification, mediated by cytochrome P-450 monooxygenases, is the major mechanism imparting resistance to different insecticides in F occidentalis. Implications of different mechanisms in insecticide resistance in F occidentalis are discussed. 相似文献
