Summary Tomato spotted wilt virus (TSWV) was obtained from infected tomatoes in commercial fields in Arkansas in 1985. A greenhouse screening procedure for identifying tomatoes resistant to TSWV was established using an enzyme-linked immunosorbent assay (ELISA) to detect infected plants. Symptom expression was variable and symptom expression was not reliable for identifying infected plants. Germplasm evaluated for resistance to one typical Arkansas isolate (85–9) of TSWV included: twenty cultivars and breeding lines of Lycopersicon esculentum Mill, 52 accessions of L. pimpinellifolium (Jusl.) Mill and 8 accessions of L. peruvianum (L.) Mill. All cultivated accessions and breeding lines evaluated were susceptible. Some individual plants in several accessions of L. pimpinellifolium were resistant and nearly all plants of the L. peruvianum accessions that were evaluated were resistant to isolate 85–9.Dept. of Plant Pathology  相似文献

Summary 1201 tomato cultivars, breeding lines and accessions of Lycopersicon species were screened for tomato leaf curl virus (TLCV) under field and laboratory conditions during summer seasons of 1986 to 1989. Two lines of L. hirsutum (PI 390658 and PI 390659) and 2 lines of L. peruvianum (PI 127830 and PI 127831) were resistant to TLCV infection. These accessions did not produce any leaf curl symptoms either in field or after inoculation by whitefly Bemisia tabaci with TLCV. Adult whiteflies died within 3 days after releasing on resistant accessions (PI 390658, PI 390659, PI 127830), whereas the whiteflies survived upto 25 days on susceptible tomato cultivars. Under field conditions 0–4 and 5–25 adult whiteflies were observed on resistant and susceptible cultivars respectively. Hybridization was effected using the popular tomato cultivars Arka Sourabh, and Arka Vikas, as the female parents and the resistant Lycopersicon wild species as the pollen parents, to incorporate the resistant gene(s) into the edible tomatoes.  相似文献

Summary The increasing popularity of organic agriculture and health food products has led to a renewed interest in hulled wheat species such as emmer (Triticum turgidum ssp. dicoccumSchubler). Knowledge on agronomic and quality traits is required for effective and efficient use of germplasm collections in breeding programs. The objective of this study was to estimate agronomic and grain quality traits of emmer wheat cultivated in Italy. A total of 20 emmer accessions consisting of landraces, breeding lines or cultivars selected from landraces and modern cultivars were examined under low input conditions. The study was conducted for three successive years (2002–2004) at one location of Southern Italy (Foggia). The entries were characterized for agronomic and qualitative traits [grain yield (t ha−1), thousand grain weight (g), test weight (kg hl−1), grain protein content (%), HMWG composition, dry gluten content (%), gluten index and yellow index, alveograph indices and Total Organic Matter (TOM) on cooked pasta]. The results showed a large genetic variability for most of the traits measured and, even if most of the accessions showed inferior bread- and pasta-making performance, modern cultivars exhibited improved quality traits with some potential to perform healthy and tasty food.  相似文献

Summary Wheat has traditionally been grown by the Beduin population in the semi-arid (150 to 200 mm, mean total annual rainfall) northern Negev region of Israel.A collection was made in this area (the size of which is 150 km2) from small (0.1 to 0.5ha) fields of mixed wheat, resulting in 1553 collected spikes. Each spike was planted in a 1 m row at Bet Dagan, and grown under favorable conditions. Qualitative and quantitative data were collected from each row. Qualitative data were submitted to hierarchial clustering and the results were compared with published information on the identification, classification and distribution of the land-races of wheat in the Middle East. Triticum durum was represented in 84% of the collection. It was clustered into 22 populations, identified as 11 known varietas of T. durum. They were aggregated into five groups, similar to groups of old varieties recognized by Jacubziner (1932). While 38.5% of the collection consisted of T. durum groups villosa and sinaica, aboriginal to the northern Negev, it included also forms similar to several land-races found in the past in other parts of the Middle East. Each of the populations, and the durum collection as a whole, was very diverse for the quantitatively measured plant attributes. Triticum aestivum was represented in 15.6% of the collection, clustered into six populations. Most of the common wheat accessions were analogous to the old locally grown variety Hirbawi. Summary A geographically representative selection of germplasm of Beta vulgaris, section Beta has been assessed for characteristics important in sugarbeet breeding, including downy mildew resistance, resistance to aphid colonisation and infection by the beet virus yellow complex. The occurrence of maintainer lines for cytoplasmic male-steriles was also investigated. Desirable qualities were found in some accessions, including nothern European wild vulgaris ssp. maritima and some old multigerm cultivars of fodder beets.  相似文献

A collection of 142 accessions of 23 Malus species, derived hybrids and cultivar accessions from the USDA-ARS Plant Genetic Resources Unit's core collection, which represents an extensive range of Malus species, was screened with a set of previously described SSR (simple sequence repeat) markers. The markers were used to determine genetic identities, estimate genetic diversity, identify genetic relationships among the accessions, and determine the utility of SSR primers developed from Malus ×domestica for making genetic assessments across the whole Malus genus. All eight primer pairs amplified multiple fragments when used in polymerase chain reactions with DNA from these accessions. High levels of variation were detected with a mean of 26.4 alleles per locus and a mean direct count heterozygosity across all eight loci equal to 0.623. The eight primer pairs used in this study unambiguously differentiated all but five pairs of accessions in this collection of 142 accessions of 23 Malus species, derived hybrids and cultivars. These SSR data were not useful in identifying genetic relationships among this diverse collection of accessions, with the majority of the accessions not clustering in ways concordant with taxonomic information and/or geographic origin. The resulting phenogram resolved only two meaningful clusters, for the taxonomically isolated Section Chloromeles and for M. fusca accessions, reflecting genetic relationships arising from geographic origin. The detection of identical accessions in the collection, which were previously considered to be unique, highlights the critical need to further bolster collections of certain Malus species. Summary A factorial mating design with six 4x cultivars as stylar parents mated to three groups of pollen parents (4x cultivars, 4x high protein selections, and 2x S. phureja) was used to study breeding behavior of yield, tuber set, marketable yield and hollow heart in potatoes. Mean yield for the 4x-2x hybrids was equivalent to that of their cultivar parents although yield components, tuber set and average tuber size, were greater and less, respectively, than the cultivar parents. High estimates of general combining ability were found in both pollen and stylar parents for yield, tuber set, marketable yield, number of marketable tubers and hollow heart in all three hybrid populations. Specific combining ability estimates were significant only for yield and hollow heart. Frequency of white-fleshed 4x-2x progeny of five heterozygous yellow-fleshed 2x phureja genotypes was used to confirm cytological identification of a first division restitution mechanism operative in the 2x parents. Summary Tomato accessions (Lycopersicon sp.), along with commercial cultivars and breeding lines were grown in a field infested with the brown root rot (BRR) organism, Pyrenochaeta lycopersici and evaluated for resistance. Three L. esculentum Mill. accessions, P.I. 260397, P.I. 262906 and P.I. 203231, were resistant and were used as male parents in crosses designed to transfer resistance to tomatoes of fresh market type. Through analysis of parental generations and F1 and F2 progenies from three crosses the heritability of resistance in the broad sense was estimated to range from 25 to 43 percent. Summary About 1000 Brassica oleracea accessions were evaluated in glasshouse tests for response to Plasmodiophora brassicae (clubroot). Resistance was confirmed in some north and west European kales and cabbage. A new source of resistance in cabbage, from Eire, is reported. Most other accessions were highly susceptible but lower levels of susceptibility were observed in open pollinated Brussels sprouts and forms of south European cabbage, cauliflower and broccoli. Modern breeding (as in the production of hybrid cultivars) appears to have resulted in increased susceptibility in several crop types. The implications of these results for the exploitation of germplasm are discussed.  相似文献

Bacterial soft rot caused by Erwiniacarotovora subsp. carotovora is amajor disease in Zantedeschia spp.,particularly in cultivars from the sectionAestivae. The disease can partiallybe controlled by cultivation measures, so acombination with resistant cultivars couldeffectively protect the crop. However,resistant commercial Aestivaecultivars are not available yet. By meansof a recently developed non-destructiveresistance test, variation inaggressiveness was observed among fiveisolates of Erwinia carotovora subsp.carotovora without interactionsbetween the isolates and three Zantedeschia accessions. Within elevenaccessions of Z. aethiopica,variation was observed from almost completeto moderate resistance, while theZ. odorata accession was susceptible.All 21 Aestivae cultivars weresusceptible. Within the Aestivaespecies, Z. elliotiana and Z.pentlandii were also susceptible, butwithin twelve accessions of Z.albomaculata, as well as in six accessionsof Z. rehmannii, variation was foundfrom susceptible to moderately resistant.Hence, new sources of resistance wereidentified that show good potentials forresistance breeding.  相似文献

Summary During the period 1988–90, several germplasm collecting trips were made to all republics of former Yugoslavia. A total of 56 old apple cultivars, many of which are represented in up to 5 types, 38 old pear cultivars and 367 genotypes of vineyard peaches were collected. The availability of so much genetic and genotypic wealth made it possible to start apple disease resistance breeding and peach cultivar and rootstock breeding programmes.  相似文献

Bananas cultivated for export all belong to Cavendish cultivars and are all recognized as very susceptible to nematodes, particularly to the burrowing nematode Radopholus similis and the lesion nematode Pratylenchus coffeae. Even if there have been many changes in the management of banana nematodes in large commercial banana plantations, chemical control still remains most often the last resort method to manage the nematodes, although the number of registered products is definitely declining. Therefore, nematode control though genetic improvement is gaining new interest worldwide. In this study, 55 banana accessions mostly diploids from the Musa acuminata genome group (AA) but including some triploid accessions (AAA), some diploids of the Musa balbisiana genome group (BB) and some interspecific hybrids (AAB, AB) were evaluated for resistance to four nematode species R. similis, P. coffeae, Meloidogyne incognita and M. arenaria. These experiments were conducted in a growth chamber under controlled conditions. All banana accessions were susceptible to nematode species, although many different levels of susceptibility were detected. This study confirmed the good resistance status to R. similis of some cultivars from the Pisang jari buaya and Pisang batuau subgroups and the partial resistance of 17 diploid accessions significantly different from the susceptible reference cv. Grande Naine. This study also showed that 12 diploid accessions exhibited a partial resistance to P. coffeae, including some usual or potential genitors belonging to the wild diploids subspecies burmannica (cvs. Long Tavoy 1 and 2) and burmannicoides (cv. Calcutta 4). No source of resistance to Meloidogyne spp. was found. These screening results, combining for the first time four nematode species, are discussed within the scope of banana breeding in order to produce parental diploid lines with single or combined nematode resistances and further develop triploids that can substitute existing susceptible commercial cultivars.  相似文献

In the present study diversity in a collection of the Garden Beet Group,consisting of 40 accessions from the BAZ Gene Bank and Polish cultivars, was evaluated in a two-year field trial. Thirteen morphological and yield traits were evaluated as well as content of betanin, vulgaxanthine, sugars, and nitrates in root tissue. The study revealed that accessions characterised by circular root shape and dark red skin, flesh and ring colour were the most common. It was also possible to identify several highly distinct accessions. The results allowed to assess genetic diversity and to make suggestions on rearranging of the Garden Beet Group core collection, which was previously set up a priori.Accessions showing a high nutritional value, potentially useful for quality breeding, were indicated. Broad genotypic variation in the response to low soilmoisture and reduced nitrogen supply was found amongthe wild Hordeum spontaneum accessions and thelandraces and modern cultivars of H. vulgare ofdifferent geographic origin. Measurements at the endof vegetative growth in plants grown in soil culturesrevealed genotypically specific responses to the usedenvironmental factors. Cultivars and breeding linesfrom Syria and Ethiopian landraces combined bothdrought resistance and tolerance to low nitrogen. TheSyrian barleys were also distinguished by a highnitrogen utilization efficiency (NUE) under low Nnutrition. European cultivars indicated a pooradaptation to N shortage, but some of them wereresistant to soil drought. No stress resistant barleyswere found among the wild accessions and Sardinianlandraces. Genotypic differences in the relativevalues of NUE and water use efficiency were associatedwith low N-tolerance. Some Syrian selections,Ethiopian landraces and the modern German cv. Maresiwere found to be most drought resistant. Maintenanceof a relatively high photosynthetic activity of theuppermost leaves was associated with droughtresistance. As far as concerning with the vegetativegrowth phase, the modern Syrian germplasm andEthiopian landraces may be recommended as donors ofadaptative characters for local barley breeding.  相似文献

Genetic diversity was studied using RAPD markers among119 coffee (Coffea arabica L.) individuals representing 88 accessions derived from spontaneous and subspontaneous trees in Ethiopia, the primary centre of species diversity, six cultivars grown locally in Ethiopia, and two accessions derived from the genetic populations Typica and Bourbon, spread in the 18th century, which gave rise to the most currently grown cultivars. Twenty-nine polymorphic fragments were used to calculate a similarity index and construct dendrograms. The Ethiopian material was separated from the Typica- and Bourbon-derived accessions and classified in four groups: one with most of the collected material from southwestern Ethiopia and three from southern and southeastern Ethiopia. Almost all detected diversity was found in the southwestern group while the southern and southeastern groups presented only 59% of identified markers. The genetic distances were low between the southwestern group and the southern and southeastern groups, and between the southwestern group and the Typica- and Bourbon-derived accessions. The cultivated coffee derived from the genetic populations Typica and Bourbon appeared little differentiated from wild coffee growing in the southwest. The results supported the hypothesis that southwestern Ethiopian coffee trees could have been introduced recently in the south and southeast. A separate analysis of the 80accessions classified in the southwestern group allowed identifying particular spontaneous- and subspontaneous-derived accessions and redundancies in the collected material from southwestern Ethiopia. RAPD markers did not detect any within-collection polymorphism except for two trees that were identified as off-types in the CATIE field genebank. Summary Soybean DNA fingerprints were analyzed by digoxigenin-labeled oligonucleotide probes complementary to simple repetitive sequences. The clearest and most polymorphic patterns were obtained with (AAT)6 as a probe, with which all 47 soybean cultivars tested could be distinguished. However, DNA fingerprints of individuals within cultivars showed the same pattern. Using (CT)8, (GAA)5 or (AAGG)4 as probes, clear polymorphic patterns among cultivars and accessions in the subgenus Soja (Glycine max and Glycine soja) were not observed, while quite different patterns were found in accessions in the subgenus Glycine. The results suggest that G. max and G. soja are closer in their genome structure. DNA fingerprints of reciprocal crosses between cultivars and accessions in the subgenus Soja were similar, and contained bands of both parents. In an F2 population from these crosses, such bands segregated in a Mendelian fashion.  相似文献
