旨在对甘肃某羊场小尾寒羊疑似绵羊痘进行确诊,并深入探讨其病理变化特点。对临诊疑似患绵羊痘的小尾寒羊进行了羊痘间接ELISA抗体检测和PCR检测确诊,并对6只病羊进行病理剖检、病理组织学和超微结构研究。结果如下:羊痘间接ELISA抗体检测显示6例病羊样品均为阳性;同时,对2例病羊样品进行PCR检测,均扩增出绵羊痘病毒(SPV)585 bp的目的基因片段,表明本次小尾寒羊所患疫病为绵羊痘。病理剖检显示,病羊皮肤无毛或少毛处充血,局部有红斑和坏死,尾部常有感染化脓形成溃疡;消化道黏膜,尤其舌和瘤胃黏膜有大小不等痘疹与坏死病灶,局部甚至形成糜烂或溃疡。肺膈叶以及肝、脾等常有痘疹和坏死病灶。病理组织学观察显示,肺痘疹病灶主要呈现渗出-增生结节和坏死-增生结节变化,病灶内血管、细支气管等周围常有大量绵羊痘细胞,胞质内有嗜酸性包涵体;病变消化道黏膜局部,上皮细胞程度不等增生、水泡变性,固有层及黏膜下层充血、水肿,有大量绵羊痘细胞,其细胞质内包涵体明显。肝、脾、肾也有程度不等坏死-增生结节变化。肺超微结构观察显示,肺泡上皮细胞肿大,细胞核浓缩、染色质聚集,线粒体程度不等肿胀、嵴断裂、空泡化,可见细胞质内包涵体。研究证明,除局部皮肤病变外,消化道黏膜和肺等实质器官的痘疹病变是患绵羊痘小尾寒羊具代表性的病理变化,尤其痘疹病灶中大量绵羊痘细胞的出现具有证病性意义。  相似文献

从山东东营疑似绵羊痘病羊的肺脏组织中分离到1株病毒。取疑似绵羊痘病羊组织病料研磨、冻融、离心后分别接种11日龄SPF鸡胚绒毛尿囊膜和牛睾丸继代细胞,鸡胚接种部位出现明显的痘斑;牛睾丸细胞出现聚集、变圆等明显的细胞病变。利用1对绵羊痘病毒T4肽基因特异性引物进行了PCR扩增,获得与预期大小一致的约300 bp的片段。将所得序列与GenBank收录的绵羊痘病毒、山羊痘病毒等毒株相关序列进行比较分析。结果表明,该分离株与国外其他绵羊痘病毒株具有较高的同源性,达97.4%~99.3%;与山羊痘病毒株同源性为96.4%。通过试验初步证明所分离的病毒为绵羊痘病毒,并将该毒株命名为绵羊痘病毒DY株。  相似文献

Goat pox virus was isolated during an outbreak of pox infection among goats in the Sersenk district, Iraq. The isolated virus grew on the chorioallantoic membranes of developing chick embryos and in primary lamb testis cell cultures. It was identified morphologically as a pox virus and serologically as a member of the Capripoxvirus group in the family Poxviridae. The isolated virus was designated the "Sersenk" strain.  相似文献

In recent years, some outbreaks of skin lesions suspected to be avian pox were observed in the backyard poultry in different parts of western areas in Iran. Consequently, 328 backyard poultries with suspected signs of avian pox virus infection were sampled. All birds showed nodular lesions on unfeathered head skin and/or fibronecrotic lesions on mucus membrane of the oral cavity and upper respiratory tract. For histopathological analysis, the sections of tissue samples from cutaneous lesions of examined birds were stained with H&E method. For PCR, after DNA extraction a 578-bp fragment of avian pox virus from 4b core protein gene was amplified. Results showed 217 and 265 out of 328 (66.1 and 80.7 %, respectively) samples were positive for avian pox virus on histopathological and PCR examination, respectively. In this study, the samples that had intracytoplasmic inclusion bodies on pathologic examination were PCR positive. This study revealed that PCR is a valuable tool for identification of an avian pox virus and that the frequency of pox infection in backyard poultry in western areas of Iran is high.  相似文献

The Sersenk strain of goat pox virus first isolated in Iraq, grew on chorioallantoic membranes (CAM) of developing chicken embryos producing generalised large pocks, 2 to 3 mm in diameter, on the third day post-inoculation. The virus killed the inoculated embryos. Replication of the virus in primary cultures of lamb testis cells induced a cytopathic effect (CPE) and plaque formation characteristic of pox viruses. It was antigenically related to reference strains of goat pox and sheep pox viruses. The virus was sensitive to ether and chloroform, failed to agglutinate erythrocytes and was strictly pathogenic to goats.  相似文献

本试验对送检的金丝雀经实验室诊断,确认为金丝雀痘,对分离的痘病毒进行动物致病性试验、病毒分离传代和致试验、致弱毒保护力试验和返强试验。结果表明:金丝雀痘病毒经鸡胚盲传3后能适鸡胚并致死金丝雀,8代后无致病性,13例毒性快速通过本动物3次无返强现象,金丝雀用第13代毒刺种30天能抵抗强毒攻击。  相似文献

对发生在贵州省的山羊痘进行了病理形态学观察.山羊痘以全身皮肤、呼吸道及消化道粘膜出现痘疹为特征.皮肤疹最初为红斑,红斑肿胀隆起形成结节状丘疹,然后丘疹发生凝固性坏死,脱落而形成痂皮.组织学检查早期皮肤病变可见充血,水肿和多量的巨噬细胞和中性白细胞浸润,随着病变的发展可见更多的巨噬细胞,淋巴细胞,浆细胞和一些体积较大的所谓"山羊痘细胞"出现.在真皮的病变细胞和痘细胞中可见嗜酸性胞浆内包涵体.引起水肿和坏死的血管炎常伴有血栓和梗死.上皮的变化包括棘细胞增生,空泡变性,不全角化,过度角化和坏死等.其他器官的病变也有相似的细胞浸润和血管炎变化.但发生在上呼吸道及上消化道时常出现溃疡.透射电镜观察在感染的皮肤和肺上皮细胞的胞浆内发现有大量不同成熟阶段的典型的山羊痘病毒颗粒.受感染的细胞除可见胞浆内散在的病毒颗粒或形成病毒包涵体外,通常可见细胞水肿、线粒体肿胀、内质网和高尔基复合体扩张.  相似文献

以贵州本地分离纯化的山羊痘病毒为试验材料,感染Vero细胞,通过台盼蓝、吖啶橙/溴乙锭荧光染色分析细胞的死亡及凋亡比例;经特异性PCR从山羊痘病毒基因组中分离出山羊痘病毒抗凋亡蛋白样基因,基因长531bp,含有完整的编码框,与绵羊痘病毒抗凋亡蛋白基因的相似性为97%,因此确定为山羊痘病毒抗凋亡蛋白基因。进一步研究山羊痘病毒抗凋亡蛋白基因在Vero细胞中的表达,结果显示,病毒感染Vero细胞24h时没有检测到凋亡现象,山羊痘病毒抗凋亡蛋白基因的mRNA水平最高;感染48h时,Vero细胞的凋亡率上升到13%,山羊痘病毒抗凋亡蛋白基因的表达量随之下降。结果表明,山羊痘病毒抗凋亡蛋白基因的表达量与Vero细胞的凋亡呈负相关,有助于病毒在细胞内的生存和侵染等过程。  相似文献

We report about the infection of an 18-year-old man with an orthopox virus (OPV) which was transmitted by a cat. The infectious route from cat to man could be proved by epidemiological, virological and serological methods. The corresponding techniques are described. The patient had not been vaccinated against smallpox and was intensively immunosuppressed by medication on account of a severe endogeneous eczema combined with an allergic asthma bronchiale. A cyclic poxvirus disease developed with a generalised, partly confluent pox virus exanthema disseminated over the body. The clinical symptoms were similar to a "variola pustulosa haemorrhagica". The young man died of a lung embolism in the course of the intensive medical therapy. The haemorrhagic character of the pox virus pustules with central necrosis (pox navel) could be reproduced in the rabbit skin and on chorioallantois membranes. The pox virus isolated from the patient could be differentiated from variola, vaccinia and monkeypox virus. It is a member of the group of "cowpox-like viruses". The environmental importance of these OPVs is discussed.  相似文献

绵羊痘是由绵羊痘病毒引起绵羊的疾病,其特征为发热、全身皮肤痘疹,内脏(特别是肺脏)黏膜结节,是危害养羊业最为严重的传染病之一。该病经常引起绵羊的大批感染或死亡,病死率可达50%以上,羔羊可达100%,不仅给养羊业带来巨大的经济损失,还严重阻碍了养羊业的发展。文章针对2011年1月份新疆某地区部分养羊户饲养的卡拉库尔羊发生的羊痘疫情进行了流行病学调查、临床症状、病理变化观察和实验室诊断,结果确诊为绵羊痘。  相似文献

从内蒙古牛场疑似山羊痘病毒感染的病羊中分离到1株病毒,命名为NM株。取病羊的皮肤痘疹和水疱作病料样品,分别接种BHK-21传代细胞和10日龄SPF鸡胚。结果表明,BHK-21细胞出现了明显的、规律的细胞病变,鸡胚接种部位出现了明显的痘斑;NM株细胞毒和胚毒均能够被山羊痘病毒阳性血清所抑制。利用1对山羊痘病毒特异性检测引物对NM株病毒进行PCR扩增,获得445 bp片段,与预期大小一致。将扩增产物提纯后克隆至pMD18-T载体,经酶切鉴定正确后,对阳性克隆株进行序列测定,结果表明,NM株的基因片段与已发表的山羊痘病毒毒株的相应基因片段具有高度同源性,同源性在99.2%~100%之间。  相似文献

A 4-month-old female domestic shorthair cat was infected by a virus of the Poxvirus family. The animal developed a severe pneumonia and generalized ulcerating lesions of the skin. Histologically, typical eosinophilic intracytoplasmic inclusion bodies indicative of an Orthopoxvirus (OPV) infection were present. The lung showed grey-white to haemorrhagic nodular lesions with a central zone of complete necrosis of alveolar and bronchial tissue. Electron microscopy from skin and lung nodules revealed typical square-shaped OPV particles. Cultivation of the virus on chorio-allantoic membranes of embryonated chicken eggs resulted in haemorrhagic plaques. Restriction enzyme analysis, PCR and sequencing of the D8L gene identified the OPV isolate as a typical Cowpox virus. It was transmitted by the cat to a human contact person who developed a local nodular dermatitis at the inoculation site in association with signs of general infection and had an increase of OPV-specific neutralizing antibodies in paired serum samples.  相似文献

山羊痘病毒的分离鉴定及生物学特性的研究   总被引:6,自引:0,他引:6  
本研究对2003年广西部分地区山羊群发生的疑似山羊痘进行了病毒分离鉴定及生物特性的研究。取疑似山羊痘病羊的皮肤丘疹、水泡或脓泡组织的病毒悬液,接种初生羔羊睾丸细胞观察到明显的细胞病变,免疫荧光试验结果显示,病毒能与山羊痘标准阳性血清反应,在感染的细胞浆内发出特异性的黄绿色荧光。病毒悬液接种乳鼠、小鼠、豚鼠、兔子都未发病,而接种3月龄山羊则出现典型的山羊痘症状和病理变化,接种9~10日龄鸡胚绒毛尿囊膜,未见出现痘斑,连传3代,均无异常变化。通过病理组织学观察可以看到在细胞浆内有大小不一圆形或椭圆形的包涵体,在电子显微镜下可以观察到150nm~300nm大小,卵圆形、砖形,有囊膜的病毒颗粒。利用一对山羊痘病毒P32基因引物进行了PCR扩增,将所得序列与GenBank收录的5株山羊痘病毒P32基因的核苷酸及氨基酸序列比较分析。结果与疫苗株的同源性分别为99.8%和99.4%。与国外其它毒株的同源性为99.6%和98、8%~99.4%。研究结果表明,所分离的病毒为山羊痘病毒,在生物学特性上与资料记载存在一定的差异,P32基因与疫苗毒和国外毒株之间同源性非常高。将该毒株命名为山羊痘病毒LiuJiang/2003株。  相似文献

Poxvirus has been identified morphologically from skin lesions in captive and free-ranging bottlenosed dolphins, Tursiops truncatus and a stranded Atlantic white-sided dolphin, Lagenorhynchus acutus. The lesions, commonly referred to as ring or pinhole lesions, appear as solitary or coalesced circular grey blemishes. Advanced ring lesions may take the form of black punctiform stippled patterns known as "tattoo". Histologically, the stratum externum is thickened, and there is ballooning degeneration and eosinophilic intractyoplasmic inclusions in the stratum intermedium. These includions contain virus particles which exhibit typical poxvirus morphology. Stress, environmental conditions and general health appear to play a major role in the clinical manifestation of dolphin pox.  相似文献

为探究猪痘病毒作为载体表达猪圆环病毒2型(PCV2)-Cap蛋白,插入外源基因P28-ORF2的拷贝数与PCV2-Cap蛋白表达量的关系。本试验以猪痘病毒为载体,通过双酶切连接及无缝克隆方法构建重组质粒,并纯化到了8株含不同拷贝数(1~8拷贝) P28-ORF2的重组猪痘病毒,基于荧光斑大小、电镜观察病毒粒子及Western blotting结果进行判断。纯化到的8株重组病毒的荧光斑直径为317.41~384.96 μm,大小无明显差异。重组猪痘病毒粒子形态与亲本病毒一致。Western blotting结果为插入1拷贝P28-ORF2的重组蛋白表达量最低;随着P28-ORF2拷贝数的增加,PCV2-Cap表达量也随之增加;至插入4拷贝P28-ORF2时,PCV2-Cap表达量达到峰值;随后减少。8株重组猪痘病毒荧光斑大小无明显差异,表明猪痘病毒基因组中插入不同拷贝数P28-ORF2对重组病毒在PK15细胞内的增殖无影响。重组猪痘病毒粒子的形态未发生变化说明多拷贝外源基因的插入并未对猪痘病毒的结构造成影响。本研究结果表明,猪痘病毒为载体表达外源蛋白时,单启动子启动多拷贝外源序列能明显提高外源蛋白的表达量,插入4拷贝的外源序列为较合适的拷贝数。  相似文献

对贵州省2002年以来发生的疑似山羊痘病例样品进行了病毒的分离与鉴定。取山羊痘病羊的皮肤痘疹和水疱作待检病料,接种BHK-21传代细胞盲传3代后,出现了明显的、规律的细胞病变(CPE),病毒细胞培养物在F4代以后,能与山羊痘标准阳性血清在琼脂扩散试验中出现白色沉淀线,而与正常细胞的培养物及PBS不出现沉淀线;参照GenBank上山羊痘病毒P32基因序列,设计了1对特异性引物,对现场分离毒株进行PCR扩增,可扩增出963bp特异性的DNA条带。用待检病料感染的BHK-21细胞培养物接种9日龄鸡胚绒毛尿囊膜,随着传代次数的增加,痘斑病变的出现率从13.3%~20%上升至33.3%~40%;用山羊痘病变皮肤、鸡胚绒毛尿囊膜和感染细胞进行超薄切片,在电子显微镜下可以观察到典型的山羊痘病毒粒子。  相似文献

Under conditions of a maximum security laboratory, four cross-bred sheep were inoculated intradermally only or intradermally and intratracheally with a West African isolate of sheep pox virus. All sheep had increased temperature and depression by the fourth or fifth day after infection. Nasal and lacrimal discharge and coughing occurred in all sheep but were more severe in sheep receiving the virus via the tracheal route. From the fifth day after infection, numerous papular erythematous skin lesions developed at the inoculation sites. These were 3-7 mm in diameter and gradually became nodular. Some of these lesions healed and others coalesced to form tumorlike masses. In one sheep, euthanized 14 days after intradermal and intratracheal inoculation, nodular lesions were found in the skin around the eyes, nostrils, oral and perianal regions, the mucosa of the rumen and throughout the lungs. Histologically, skin nodules were characterized by ischemic necrosis, vasculitis, microvesicualtion, eosinophilic cytoplasmic inclusions in the dermal epithelial cells and vacuolar nuclear degeneration. The pulmonary lesion was that of proliferative alveolitis with occasional cytoplasmic inclusions in the alveolar cells and macrophages. Ultrastructurally, large cuboidal virus particles were found both in the skin lesion and inoculated tissue cultures. The sheep pox virus structure was easily distinguished from contagious ecthyma virus, a parapoxvirus which causes sporadic disease in Canada. Serum neutralizing antibodies developed in all the sheep by 14 days postinfection.

The clinical and pathological characteristics of experimental sheep pox produced with this West African isolate were similar to those caused by Neethling virus of lumpy skin disease in cattle.

为了解贵州省山羊流产与山羊痘的相关性,采用琼脂扩散试验和PCR法对本省10个市(县)流产羊群的血清和病料样本进行山羊痘抗原抗体及病原核酸检测,同时血清进行布氏杆菌抗体检测,流产胎儿病料进行羊流产亲衣原体病原核酸检测。结果发现山羊痘羊群流产率达37.1%(4329/11660),山羊痘血清抗体阳性率为38.2%(34/89),抗原阳性率为72.7%(32/44),流产胎儿山羊痘病毒核酸检出率为83.3%(10/12),发病羊群未检出布氏杆菌和羊流产亲衣原体感染。结果表明,山羊流产与山羊痘感染有一定关系,提示在山羊养殖中应加强饲养管理,防止山羊痘感染引起孕羊流产。  相似文献

应用我们建立的绵羊痘病毒和山羊痘病毒通用RPA检测方法,通过对口蹄疫病毒、传染性脓疱皮炎病毒、鸡痘病毒、绵羊痘病毒、羊痘疫苗和羊痘重组质粒样本的检测,验证绵羊痘病毒和山羊痘病毒通用RPA检测方法对绵羊痘/山羊痘病毒检测的特异性。结果显示该方法对羊痘疫苗、羊痘重组质粒、绵羊痘病毒核酸和山羊痘病毒核酸阳性样品能够在试纸条上显示出阳性条带,而对其他样品不显示阳性条带。表明该方法具有良好的特异性,在绵羊痘/山羊痘病毒检测中应用前景广阔。  相似文献

The paraspecific effect of pox viruses i.e. stimulation of the nonspecific part of the complex immune system, was demonstrated in vitro and in vivo for 3 different types of virus (vaccinia virus, avipox virus, parapox virus). By means of purification, concentration and inactivation, it was possible to prove that the paraspecific effect is brought about by the pox virus particle per se. The effect is induced by certain epitopes in the complex structure of the virus and is independent of virus multiplication. Both purified pox viruses and inactivated, purified pox viruses stimulate NK cell activity, the CSA and the formation of interferon, and reach higher efficacy indices in a challenge test with baby mice using stomatitis vesicularis virus than non-purified and non-inactivated cell culture material. The degree of purity of the pox virus preparations was established using the electron microscope and monoclonal antibodies. The purified and concentrated pox virus preparations proved to be pyrogen-free; they were not toxic to baby mice in vivo. An endotoxin-induced effect could be ruled out. The broad paraspecific effect of pox viruses and their structural components lends itself to use as paramunity inducers in prophylaxis and therapy.  相似文献
