A cross-sectional survey was performed to estimate the prevalences of antibodies to Toxoplasma gondii (ELISA, IFAT), Sarcocystis spp. (ELISA, using S. miescheriana as antigen) and Neospora caninum (ELISA, immunoblotting) in sows from breeding farms in southern Hesse, Germany. A total of 2041 plasma samples of sows from 94 randomly selected farms was examined. Data on farm profiles, husbandry management and sows were collected by a questionnaire and exploratively analysed. For T. gondii the ELISA results agreed well with the results obtained by IFAT (kappa=0.71). Antibodies to T. gondii were detected by ELISA in 19% of the sows. Sixty-nine percent of the farms had at least one seropositive sow, and a within-farm seroprevalence of >or=50% was observed in 14% of all farms. The prevalence of anti-T. gondii antibodies was positively correlated with the age of sows. The within-herd seroprevalence was significantly higher in farms with reproductive disorders than in those without such problems. On the farm level, the farm type 'piglet production' (versus 'pedigree breeding' or 'farrow-to-finish') was the only risk factor associated with the presence of T. gondii-seropositive sows. Antibodies to Sarcocystis spp. were found in 29% of the sows. Seventy-two percent of the farms harboured at least one seropositive sow, and a within-farm seroprevalence of >or=50% was detected in 23% of all farms. The seroprevalence increased significantly with the age of sows. On the farm level, only the farm type 'piglet production' (versus 'pedigree breeding') and the replacement of sows by purchasing (versus raising on the own farm) were identified as risk factors for seropositivity. Antibodies to N. caninum were detected in one sow using both the screening ELISA and the confirmatory immunoblotting technique. This may indicate the first natural N. caninum infection in pigs.  相似文献

Toxoplasmosis in pigs is a large threat to pig industry as well as pork consumers. Most pigs become infected by ingestion of oocysts from contaminated environment (soil, water and feed) or infected animal tissues postnatally. In the present study, field studies were conducted to evaluate the relationship between soil contamination status of Toxoplasma gondii oocysts and T. gondii infection in pigs in 12 pig farms with different density of cats in central China. The presence of T. gondii oocysts in soil were determined by PCR and loop-mediated isothermal amplification (LAMP). T. gondii DNA was found in 11 farms with different cat density excepting one farm exposed to low cat density. 自2017年1月—2018年10月,在河南省不同地区不同规模的143个猪场采集4 203份血清样品,使用ELISA试剂盒检测弓形虫抗体。结果显示,河南省猪弓形虫抗体阳性率达38.7%。按不同季节汇总分析,冬季阳性率最高(45.7%),其次为秋季和春季,夏季最低(31.9%)(P0.01)。按不同地区汇总分析,豫中阳性率最高(53.6%),其次为豫西、豫东和豫南,豫北最低(30.4%)(P0.01)。按猪场不同规模汇总分析,小型规模猪场阳性率最高(46.0%),其次为中型规模猪场,大型规模猪场最低(32.9%)(P0.01)。按猪群不同年龄汇总分析,母猪群阳性率最高(57.8%),其次为后备母猪和育肥猪,保育猪最低(12.2%)(P0.01)。按繁殖母猪不同胎次汇总分析,6胎及以上母猪阳性率最高(73.8%),其次为3~5胎母猪和2胎母猪,1胎母猪最低(57.3%)(P0.05)。同时,本试验也调查了猪繁殖与呼吸综合征(PRRS)和猪伪狂犬病(PR)对弓形虫抗体阳性率的影响。结果显示,PRRS发病场弓形虫抗体阳性率最高(49.4%),其次为PRRS阳性稳定场,最低为PRRS阴性净化场(9.2%),三者之间差异极显著(P0.01)。在PR阳性带毒场和PR阴性净化场,弓形虫抗体阳性率差异不显著(P0.05)。在24个PR阳性带毒场内,gE抗体阳性母猪和gE抗体阴性母猪的弓形虫抗体阳性率差异也不显著。结果表明,在河南省猪群中普遍存在弓形虫感染情况。猪场通过完善生物安全体系和提高饲养管理水平,可以减少弓形虫的感染。  相似文献

A serological survey of Toxoplasma gondii infection in adult breeding sheep in Great Britain was conducted using surplus sera taken during a seroprevalence study of Brucella melitensis in 2009. Of the 3539 sera collected from 227 flocks, 2619 (74 per cent) were found to be positive for T gondii specific antibody when tested using latex agglutination. Multilevel logistic modelling suggested that the likelihood of infection increased with age and this effect appeared to be amplified in animals vaccinated against T gondii. The model also indicated that the odds of sheep being seropositive were increased on premises where cattle were also kept. These results suggest a high level of Toxoplasma infection in breeding sheep in Great Britain and provide further evidence to suggest that postnatal infection is more common than congenital infection in sheep.  相似文献

In a random sample of 103 sheep farms in Ontario, 99% of the farms had some sheep serologically positive for Toxoplasma gondii, based on an enzymelinked immunosorbent assay (ELISA). The percent of sheep affected within farms ranged from 3.8% to 97.8%, with an average flock prevalence of 57.6%. When farm management variables were considered in a multivariate analysis, significantly lower rates of serologically positive sheep were associated with neutering of female cats and clipping of ewes' perineums before lambing; significantly higher prevalence rates were found on farms where sheep were purchased from other flocks, pigs were raised on the same farm, sheep shared pasture with other animals, flowing water was available at pasture, and pastured replacements had access to housing. As well, in univariate analyses, higher prevalence was positively associated with an increasing number of cat litters born over the previous two years and offering creep feed or forage to lambs, and inversely with the amount of labor expended on sheep rearing.  相似文献

Data on the genotypes of Toxoplasma gondii circulating in wildlife are scarce. In the present study, foxes and rodents from two Federal States in Central or Eastern Germany were examined for T. gondii infections. Body fluids were collected at necropsy or fluids were obtained from frozen tissues of naturally exposed red foxes (Vulpes vulpes), voles (Microtus arvalis), shrews (Neomys anomalus) and a striped field mouse (Apodemus agrarius) and tested for T. gondii by serology. DNA isolated from tissues of seropositive foxes and all the rodents was examined by PCR. In the German Federal States of Brandenburg and Saxony-Anhalt 152/204 (74.5%) and 149/176 (84.7%) of foxes, respectively, but none of the rodents (0/72) had antibodies to T. gondii. Only 28/152 (18.4%) and 20/149 (13.4%) of seropositive foxes from Brandenburg and Saxony-Anhalt, respectively, but none of the rodents tested PCR-positive for T. gondii. The complete T. gondii genotype could be determined for twelve samples using nine PCR-restriction fragment length polymorphism (PCR-RFLP) markers (newSAG2, SAG3, BTUB, GRA6, c22-8, c29-2, PK1, L358 and Apico). In addition to T. gondii clonal type II (Apico II) and type II (Apico I), type III and T. gondii genotypes showing non-canonical allele patterns were observed in foxes. This suggests that, while T. gondii type II prevails in foxes, other genotypes circulate in wildlife. The population structure of T. gondii in Germany may be more diverse than previously thought.  相似文献

ABSTRACT: A seroepizootiological study of Toxoplasma gondii infection involving a total of 488 slaughter pigs (468 market-weight pigs and 20 sows) in the Belgrade area, also included examination of the presence of T. gondii in the blood. Blood sampled at the slaughter line was examined for specific antibodies by modified direct agglutination, and blood clots of those seropositive at titres of 1:50-1:12800 were bioassayed in mice. The overall seroprevalence was 9.2%, significantly higher (p = 0.0063) in sows (30.0%) than in market-weight pigs (8.3%). Amongst the 22 bioassays performed, a total of 16 (72.7%) were positive, by observation of T. gondii cysts (12), seropositivity (7, including 3 in which cysts were not detected), and/or detection of T. gondii DNA by real-time PCR (12, including one otherwise negative). The positive bioassays originated from the blood of 12 market-weight pigs and 4 sows. Despite a general increase in the rate of demonstration of T. gondii with the increase in the specific antibody level, the association was not significant (p = 0.101). The risk of infection was 41-fold increased in sows vs market-weight pigs, and 15-fold in pigs from smallholders' finishing type farms vs those from large farrow-to-finish farms. The presence of viable T. gondii in a proportion of the samples indicates that some of the pigs had an active parasitaemia at the time of slaughter, which, along with the seroprevalence established, points to a potential source of human infection in Serbia. This is the first report on parasitaemia in naturally infected swine.  相似文献

Toxoplasmosis gondii causes one of the most common zoonoses worldwide. The rate in Panama is one of the highest in the world. Infections are primarily asymptomatic in immune competent individuals; however, in immunocompromised patient and congenital cases can be lethal. Exposure to the pathogen is hypothesized to occur when handling or consuming infected food such as swine meat. In this study, we analyzed 290 swine sera collected from six provinces in Panama by Indirect Immunofluorescence antibody test (IFAT) against T. gondii. Toxoplasma-specific IgG were found in 32.1% of the samples. The highest seroprevalence was found in the province of Panama.  相似文献

为了进一步了解甘肃省W市种羊场羊布鲁菌病血清流行率情况及空间分布,为当地种羊场羊布鲁菌病的防控提供参考。本研究采用血清学检测以及群间、空间分析的方法,对甘肃省W市种羊场进行羊布鲁菌病血清流行病学调查。结果表明,W市2016年种羊场布病表观血清流行率为38.10%,真实血清流行率为45.69%,个体表观血清流行率为6.61%,真实血清流行率为5.84%;2017年种羊场布病表观血清流行率为47.62%,真实血清流行率为57.75%,个体表观血清流行率为4.79%,真实血清流行率为3.53%;2次检出的阳性种羊场主要分布在农区,牧区种羊场未检出布病阳性羊。表明W市阳性种羊场数量有所增加,布病表现出明显的区域性特征。  相似文献

Sera from 456 wild rabbits (Oryctolagus cuniculus) collected between 1992 and 2003 from five geographical regions of Spain were examined for antibodies to Toxoplasma gondii by the modified agglutination test. Antibodies to T. gondii were found in 65 (14.2%) wild rabbits. Prevalence of infection was significantly higher in samples collected from wild rabbits from Catalonia, northeast Spain (53.8%), where rabbits lived in forest, compared to other areas (Huelva and Cádiz, Andalucía, south Spain; Toledo, Castilla-La Mancha, central Spain; and Zaragoza, Aragón, northeast Spain) with more dry conditions, where prevalence ranged from 6.1 to 14.6%. No differences were observed on prevalence and age (young animals <7 months of age compared to older animals), sex, date of samples collection or season of samples collection. The results indicate that prevalence of T. gondii in some areas of Spain is high, and this finding could have environmental and/or public health implications if wild rabbits are to be used as a source of food.  相似文献

Toxoplasmosis is a globally distributed zoonosis with a clinical impact in the unborn fetus and in the immunosuppressed individual. In Serbia, studies of risk factors for Toxoplasma gondii infection in humans have shown that the relatively high prevalence is associated mainly with consumption of undercooked meat and/or meat products. However, data on T. gondii infection in domestic animals mostly used for human consumption are scarce. We thus conducted a cross-sectional survey on the seroprevalence of T. gondii infection in a representative sample of cattle, sheep and pigs from different regions of Serbia between June 2002 and June 2003, and analyzed the main risk factors associated with the infection. Sera from 611 cattle (yearlings and adults of both sexes), 511 ewes, and 605 pigs (market-weight and sows), were examined for T. gondii antibodies by the modified agglutination test. The seroprevalences determined were 76.3% in cattle, 84.5% in sheep and 28.9% in pigs. The antibody levels ranged from 1:25 to 1:400 in cattle, and up to 1:25,600 in sheep and to 1:12,800 in pigs. Among the seropositive, the proportion of high antibody levels (> or =1:1600), suggestive of acute infection, was 10% in sheep, and 4% in pigs. Possible association of the infection with biologically plausible risk factors including gender, age, herd size/farm type, type of housing, feeding practices and region, was analyzed by univariate analysis, and variables significant at P< or =0.1 were included in multivariate logistic regression models. The results showed that risk factors for cattle were small herd size (odds ratio, OR=2.19, 95% confidence interval, CI=1.28-3.75, P=0.004) and farm location in Western Serbia (OR=2.04, 95% CI=1.10-3.79, P=0.024), while housing in stables with access to outside pens was protective (OR=0.37, 95% CI=0.21-0.67, P=0.001). In sheep, an increased risk of infection was found in ewes from state-owned flocks (OR=4.18, 95% CI=2.18-8.00, P<0.001) vs. private flocks, and, interestingly, also in those from Western Serbia (OR=4.66, 95% CI=1.18-18.32, P=0.028). In pigs, the risk of infection was highly increased in adult animals (OR=3.87, 95% CI=2.6-5.76, P<0.001), as well as in those from finishing type farms (OR=3.96, 95% CI=1.97-7.94, P<0.001). In addition to providing data on the current T. gondii seroprevalence in meat animals in Serbia, the results of this study show the main risk factors associated with infection, thereby pointing to the type of preventive measures to reduce T. gondii infection.  相似文献

Ingesting meat of free-range livestock, mainly sheep, is associated with human toxoplasmosis in European countries. Data on Toxoplasma gondii infection in French ovine livestock are relatively scarce. Sera from 164 lambs and 93 ewes slaughtered in Haute-Vienne district, France, were tested by a direct agglutination test. Antibodies to T. gondii were found in 36 (22.0%) lambs and in 61 (65.6%) ewes. In addition, to attempt parasite isolation for genotyping, hearts from 50 other ewes were obtained from a local slaughterhouse, and were screened by a direct agglutination test. T. gondii was isolated in 8 of 30 seropositive hearts bioassayed in mice. All isolates were type II by genetic characterization at five microsatellite loci (TUB2, TgM-A, W35, B17, B18). These results indicate that bovines slaughtered in France may be highly infected by T. gondii with a potential risk of parasite transmission to humans by consumption of undercooked meat. Multilocus microsatellite analysis shows the predominance of type II in sheep as previously described in humans.  相似文献

OBJECTIVE: To provide an epidemiologic investigation of the seroprevalence of antibodies to Toxoplasma gondii in populations of cats and wild rodents in Rhode Island and to address the possible epidemiologic role of wild rodents in the spread of toxoplasmosis. ANIMALS: 200 cats and 756 small wild rodents. PROCEDURE: Serum samples were obtained from 84 cats in animal shelters and 116 cats in veterinary hospitals. Serum samples were also obtained from 756 small wild rodents from multiple sites in Rhode Island. Sera from rodents and cats were assayed for antibodies to Tgondii by use of the modified agglutination test RESULTS: Overall, 42% (84/200) of cats had serum antibodies to Tgondii. Seroprevalence was not significantly different between stray (50%; 42/84) versus client-owned (36%; 42/116) cats, between male (43%; 40/94) versus female (42%; 39/93) cats, or between indoor (26%; 7/27) versus outdoor (39%; 35/89) cats. Seroprevalence rate of trapped rodents was 0.8% (6/756). Six rodents captured in Washington County accounted for of the seropositive rodents. Four of 6 of the seropositive rodents were trapped at a single site in Washington County (an abandoned barn). Five stray cats, known to have resided at the same site in Washington County as 4 of the seropositive rodents, were also found to be seropositive for antibodies to T gondii. CONCLUSIONS AND CLINICAL RELEVANCE: Seroprevalence rate in rodents was not correlated with the seroprevalence rate in cats. Stray cats, especially those known to be feral, may be more likely to perpetuate the cat-mouse cycle of T gondii than client-owned cats.  相似文献

The seroprevalence of Toxoplasma gondii was examined in adult women and domestic animals used for meat products from the Mymensingh District, Bangladesh. Cattle, goats and sheep showed a high seroprevalence (12, 32 and 40%, respectively), while the sera from all fifteen women examined in the same area were seronegative. Considering that primary infection in women during pregnancy can cause abortion and congenital defects, accidental ingestion of T. gondii infected meat products from domestic animals represents a risk factor for adult women living in the same area.  相似文献

At the end of the national eradication program for Enzootic Pneumonia (EP) and Porcine Actinobacillosis (APP) in Switzerland (2003), A. pleuropneumoniae serotype 2 is considered to have been eradicated. There is no current information about the distribution of the other serotypes available. The ApxIV ELISA detects antibodies against all serotypes of A. pleuropneumoniae, without cross-reaction with other bacterial species. The aim of this study was to achieve actual data concerning the seroprevalence of A. pleuropneumoniae in breeding-herds and to validate the ApxIV ELISA under field conditions, especially for the diagnosis of latently infected breeding-herds without clinical signs, and to achieve more information about the role of herd book farms for the spread of the infectious agent. A total of 2068 serum samples from 96 pig herds in Switzerland were examinated. Over half of the examinated herd book farms showed positive results in this ELISA. 93% of the breeding herds were positive. On single animal level sensitivity was 96% and specifity 100%. Herd sensitivity ranged between 67% and 99%. Herd specifity was 100%. The results show that the ApxIV ELISA is a valuable tool for the detection of latently infected herds.  相似文献
