One kidney was taken from each of 100 pigs at slaughter; 50 had gross lesions of multifocal interstitial nephritis and 50 had no gross lesions. Forty-nine of the affected kidneys had lesions that were characterised by the presence of either a few randomly distributed or numerous widely disseminated pale foci, 1 to 3 mm in diameter, on the cortical surface (white-dotted kidneys). Microscopically, these focal inflammatory lesions often had a distinct lymphofollicular pattern (follicular nephritis). Lesions of chronic vasculitis were observed in 21 of the affected kidneys. Histologically, the control kidneys had only small and sparse inflammatory foci. Standard bacterial cultures of kidneys of both groups were not significant, and cultures for the isolation of leptospires were all negative. Virological examination of the kidney homogenates by PCR did not reveal any porcine reproductive and respiratory syndrome virus and only a few cases were positive for the porcine circovirus type 1. However, porcine parvovirus (PPV) and porcine circovirus type 2 (PCV-2) were detected in many kidneys of both groups but in a significantly higher proportion of the kidneys with interstitial nephritis. There was a significant association between the lesions and the presence of PPV and PCV-2 with odds ratios of 7.5 (P<0.0001) and 3.4 (P=0.0074), respectively, and the odds ratio increased to 22.7 (P<0.0001) when both viruses were identified in the same kidney. However, a subsample of kidneys taken from both groups were negative by immunohistochemistry for the presence of PPV and PCV-2 antigens.  相似文献

An abattoir survey was undertaken to determine the prevalence of leptospirosis and its association with lesions of multifocal interstitial nephritis (so-called "white spotted kidneys") in swine at slaughter. Both cross-sectional and case-control study designs were used. Of 197 kidneys from hogs randomly selected at slaughter, 11 (5.6%) had generalized grey-white foci typical of multifocal interstitial nephritis (MFIN). Antibody titers greater than or equal to 1:80 against Leptospira pomona were detected in nine (4.6%) hogs and against L. bratislava in 63 (32%) of these hogs. Leptospira pomona (kennewicki) was detected by immunofluorescence in 5/197 (2.5%) of randomly selected hogs. Leptospires identified as genotype kennewicki were isolated from six (9.8%) of 61 kidneys cultured. Leptospira bratislava was not detected by immunofluorescence or culture. There was a highly significant (p = 0.00) and strong association (odds ratio (OR) = 195) between high L. pomona titer (greater than or equal to 1:80) and the presence of leptospires in the kidneys, as detected by culture. There was also a significant (p = 0.046) and strong (OR = 8.10) association between multifocal interstitial nephritis and the presence of renal leptospires as detected by culture. The association between leptospiral titer and MFIN lesions in the prevalence survey group of animals was statistically significant (p = 0.031), but this association was not significant in the case-control study group (p = 0.071) The failure to identify L. bratislava despite serological evidence of infection suggests that some of these seropositive animals may have been transiently infected at an early age, that serological findings were falsely positive, or that immunofluorescence and isolation attempts failed to detect L. bratislava if they were indeed present in the kidneys.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The stomachs of pigs (n=15,741) originating from 136 herds from the Australian states of Queensland, Western Australia, Victoria and New South Wales were examined at slaughter for the presence of oesophago-gastric ulcers (OGUs). Stomachs were categorised as being normal, hyperkeratotic, eroded, ulcerated, or having strictures. A questionnaire was distributed to piggery owners to identify factors associated with an above-average herd prevalence of OGU. Thirty percent of all pigs examined had OGU (median within-herd prevalence of 17%). The median within-herd prevalence in Victoria (53%) was significantly higher than in Western Australia (30%) or Queensland (7%). The prevalence of OGU in culled breeding animals was significantly higher than in porkers or baconers from the same herds. There was no difference between the prevalence of OGU in male and female pigs sampled from the same Western Australian herds. The relationship between OGU and herd and pig risk factors was assessed by random effects logistic-regression analysis. Herds with a high prevalence of OGU were more likely to feed ad libitum (OR=13.7), use automated feeding systems (OR=7.8), feed a pelleted ration (OR=384) and get water from a dam rather than from a bore or river (OR=3.8). Pigs can harbour a variety of viruses in their gastrointestinal tract. Some of them are closely related to human viruses and are therefore suspected to have a zoonotic potential. Only little is known about the presence of those viruses in pigs at slaughter. However, by contamination of meat with zoonotic viruses during the slaughtering process, food-borne transmission to humans may be possible. Here we analyzed 120 faecal samples of pigs at slaughter from 3 different geographical regions of Germany for the presence of astrovirus (AstV), encephalomyocarditis virus (EMCV), hepatitis E virus (HEV), norovirus genogroup II (NoV GII) and group A rotavirus (GARV). Using real-time RT-PCR, the most frequently detected virus was AstV, which was present in 20.8% of the samples, followed by NoV GII with a detection rate of 14.2%. EMCV, HEV and GARV were found only occasionally with detection rates of 4.2%, 2.5% and 0.8%, respectively. Analyses of partial genome sequences of the viruses indicated that the detected AstV and NoV GII mainly represented typical pig virus strains, which have not been detected in humans so far. However, the GARV and HEV strains were more closely related to human strains. The results indicate that enteric viruses, some of them with zoonotic potential, are present in pig faeces at slaughter. Application of good hygiene practice is necessary to minimize the risk of introducing these viruses into the food and to prevent virus transmission to highly exposed persons such as slaughterers and veterinarians.  相似文献

A cross-sectional study was conducted with 617 finishing pigs aged 10–25 weeks in 11 commercial herds of different health statuses as defined by the Danish monitoring program for specific-pathogen-free (SPF) herds. A standard clinical examination was performed and a blood sample was obtained from each pig for determination of haptoglobin concentration in serum.

Pigs aged 10–14, 15–19 and 20–25 weeks in conventional herds had higher haptoglobin concentrations than high-health SPF (SPF-x) pigs of the same age. There was no significant difference between SPF-x pigs of different ages. Conventional pigs aged 15–19 and 20–25 weeks had higher haptoglobin concentrations than conventional pigs aged 10–14 weeks. Herd influenced the haptoglobin concentration. Lame pigs and pigs with tail or ear bite had elevated haptoglobin concentrations. No significant effect of respiratory symptoms or umbilical hernia was found.  相似文献

To establish the relationship between serum levels of three acute phase proteins, haptoglobin (Hp), C-reactive protein (CRP) and serum amyloid A (SAA), and the occurrence and severity of lesions at slaughter, a study was carried out using 70 fattening pigs from a finishing unit. Pigs were divided into three groups: Group 1 (25 pigs with clinical signs of disease), Group 2 (25 apparently healthy pigs with lesions at slaughter) and Group 3 (20 apparently healthy pigs with no lesions at slaughter). Serum levels of CRP, SAA and Hp were significantly higher in pigs with clinical signs of disease than in apparently healthy animals. Additionally, in apparently healthy pigs, serum levels of Hp and CRP were significantly higher in animals with lesions than those without lesions. The extent and severity of lung lesions were related to serum levels of Hp.  相似文献

Background

Tail biting is a common welfare problem in pig production and in addition to being a sign of underlying welfare problems, tail biting reduces welfare in itself. The aim of this study was to evaluate the effects of tail biting on different pre and post mortem indicators of stress in slaughter pigs and on carcass and meat characteristics. A total of 12 tail bitten (TB) and 13 control (C) pigs from a farm with a long-term tail biting problem were selected for salivary cortisol analyses before and after transport to the slaughterhouse. After stunning, samples were taken for the analysis of serum cortisol, blood lactate, intestinal heat shock protein 70 (HSP70), and meat quality characteristics. In addition, body temperature immediately after and muscle temperature 35 min after stunning were measured, as well as lean meat percentage and carcass weight.

Results

TB pigs showed a lower cortisol response to the transport-induced stress than C pigs and also had a lower serum cortisol concentration after stunning. HSP70 content in the small intestine was higher in the TB pigs than in C pigs. TB pigs had a considerably lower carcass weight therefore produced a lower total amount of lean meat per carcass than C pigs.

Conclusions

This study suggests that prolonged or repeated stress in the form of tail biting causes a blunted stress response, possibly a sign of hypocortisolism. In addition, it underlines the importance of reducing tail biting, both from an animal welfare and an economic point-of-view.  相似文献

The present study investigated the effect of outdoor rearing and genetic background on behaviour and meat quality traits in heavyweight pigs. Large White and Duroc-sired pigs were reared in groups of 3 castrated males and 3 females, conventionally or in fields, from April to October, in a study with two replicates. Compared to indoor pigs, outdoor pigs were more active and showed a larger range of behaviour suggesting improved welfare from a behavioural point of view. During 5 h of isolation, outdoor pigs were less active and had lower heart rates, and in another test, they reacted less to a non-familiar object (traffic cone). Pigs were slaughtered at 150 kg live weight, half of each treatment group after mixing, short transport, and overnight lairage and half immediately following short transport. Outdoor pigs were less aggressive during pre-slaughter mixing. Ante- and post-mortem glycogen content of the Longissimus lumborum (LL), Semimembranonus (SM) and Semispinalis capitis (SC) muscles depended on slaughter conditions, sire breed, rearing conditions, gender and year of experimentation, sometimes influencing ultimate pH. Effects of rearing on muscle glycogen content and post-mortem pH were stronger during the second year of rearing. Effect of slaughter conditions on glycogen content and ultimate pH depended on fighting levels during pre-slaughter mixing. Outdoor rearing increased muscle redness. Drip and cooking loss were higher in Large White than Duroc-sired pigs. Drip and cooking loss were correlated with early post-mortem and ultimate pH. Thawing loss was correlated with early post-mortem temperature and ultimate pH. Finally, reactivity to isolation had a predictive value as pigs more active during isolation had less skin damage due to fighting during pre-slaughter mixing. In conclusion, despite large effects of year of experimentation, outdoor rearing, sire breed, and slaughter conditions influenced behaviour and muscle characteristics.  相似文献

The study was performed to investigate changes in the behaviour of male and female pigs when one or more pigs were removed from the pen and sent to slaughter. Twelve pens were included, half of them housed six female pigs each, while the other half housed six male pigs each. Skin lesions and behaviour were recorded on two occasions, four days before and two days after one or more animals per pen were sent to slaughter. On the first occasion the male pigs fought on average about twice as often as the females (2.6 vs. 1.0 per animal per hour) and had twice as many skin lesions (6.7 vs. 3.2). On the second occasion, the frequency of fighting among both males (6.8) and females (1.8) had increased, however, more in males than in females. The number of skin lesions was not significantly higher on the second occasion. In the male group there was also a tendency towards increased mounting on the second occasion (from 2.3 to 3.3 per animal per hour). The study shows that the very common practice of slaughtering pigs from one pen over more than one occasion results in an increase in unwanted behaviour. The effect is more pronounced in groups of entire male pigs than in groups of females.  相似文献

As a part of a nationwide programme to survey and control salmonella in pig herds, a microbiological survey of 1363 pig herds was performed in Denmark. A total of 13 468 slaughter pigs were examined at slaughter by culture of 5 g of caecal contents. Overall, 30 different serotypes of Salmonella enterica were isolated from 832 pigs (6.2%). The predominant serotype was S. Typhimurium, comprising 536 (64.4%) of the isolates. Four hundred and forty-eight isolates of S. Typhimurium were examined by phage typing, resulting in detection of 17 different phage types (definitive types, DT) with DT12 being the most frequent (49.1%).

Salmonella enterica was found in 302 herds (22.2%), S. Typhimurium was found in 61.1% of these. 279 (23.1%) large herds (producing more than 2600 slaughter pigs per year) were found to be salmonella positive compared with 23 (14.7 %) small herds (annual production of 500 to 550 slaughter pigs). Practical constraints in the study design did not allow for a firm conclusion on the interplay among herd size, geographical location and occurrence of salmonella.

In 284 of 302 infected herds (94.0%) only one serotype was detected. Infections with two different serovars were seen in 18 herds (6.0%).  相似文献

Pleuritis in slaughter pigs has increased in recent years in the Netherlands. The aim of the present study was to determine what respiratory pathogens were involved in pleuritis.In total, lungs of 968 slaughter pigs from 10 herds with high prevalence of pleuritis were morphologically examined for size, location, and type of lesions. Moreover, histology and bacteriology were performed.Examination of gross lung lesions showed 45% pleuritis, 14% pleuropneumonia and 38% catarrhal pneumonia. Peribronchiolar cuffing was found in 61 of 142 samples. Actinobacillus pleuropneumoniae was cultured from 22 lung samples from four herds. Pasteurella multocida was cultured from 55 lung samples in eight herds. No specific pattern with respect to the causal pathogens was found.In conclusion, no single infectious cause of pleuritis was found. A variety of infectious agents combined with environmental factors should be considered as a cause of pleuritis.  相似文献

In order to evaluate the effect of slaughter weight and rearing system on the Chato Murciano pig carcass and meat quality traits, a total of 71 castrated male pigs were used. Forty-one pigs were reared in an outdoor system, divided in one heavy group (CHOHW) with an average live weight of 132.05 kg, and a second group with lower weights (CHOLW, 115.7 kg average). Simultaneously, 30 pigs were reared indoors and divided into one heavy group (CHIHW) with an average live weight of 144.3 kg, and a second group with lower weights (CHILW, 117.6 kg average).Heavier pigs showed higher values for several carcass parameters (hot carcass weight, carcass length, maximum perimeter of the ham, hand length, leg length, ham length, wrist perimeter) and meat cuts, higher Dorsal Fat Thickness (DFT) and Intramuscular Fat (IMF) levels, but no difference was found in the hot carcass yields (HCY). Differences in the colour parameters and several minerals (Ca, Mg, K and Na) were also due to the different slaughter weights. The rearing system had an influence on the HCY (it is higher in the outdoor system), on many carcass parameters, on meat the ultimate pH (it is higher for animals reared indoors) and on colour (this is variable). It also affected the levels of many minerals with the levels of Mg, P and K being higher for animals reared outdoors, whereas the Fe, Cu and Zn content were higher in the animals reared indoors. DFT and IMF levels were not influenced by the rearing system.  相似文献

The aim of the present study was to determine, in horses destined to slaughter, the effects of transport, lairage and stunning on some blood constituents related to stress. Twenty one horses culled from a race track due to poor athletic performance, transported weekly to a nearby slaughterhouse were used. Each horse was canulated through venipuncture of the right jugular vein with a central venous catheter which remained permanently until after the exsanguination. Through the catheter, blood samples were taken at 6 sampling times: one hour before loading of the horses, immediately after loading, at the end of the journey, immediately after unloading, after lairage, in the stunning box before stunning and during exsanguination. Blood samples were analyzed for the following variables: plasma lactate concentration, plasma creatine phosphokinase activity (CK), plasma glucose concentration, plasma cortisol concentration and packed cell volume (PCV). Time in the stunning box, number of stunning attempts, presence of return to consciousness signs and time between stunning and sticking were also recorded. The results for pre-mortem variables were: mean time in the stunning box 9 min 48 sec, 85.7% of the horses fell at the first stunning attempt, 57.2% of the horses showed signs of returning to consciousness after stunning and the most frequent interval between stunning and sticking was 1.01 to 2 min. Mean values for the blood variables showed a significant rise (P < 0.05) in the concentrations of lactate, glucose and cortisol, as well as CK activity and PCV during transport. The highest values for these blood constituents were seen before stunning and during exsanguination, except for cortisol concentration, which showed the highest values during the transport process.  相似文献

针对红河州猪繁殖障碍综合征危害日趋严重的现象,进行了该病的流行病学、病原学、人工感染及免疫试验。结果表明,种公猪发病率为23.77%、妊娠母猪发病率为41.38%,仔猪死亡率为19.94%、流产率11.19%、死胎率47.96%、弱仔率31.78%;血清学调查阳性检出率分别为猪瘟(CSF)1.1%、细小病毒(PPV)43.97%、鹦鹉热衣原体(CP)36.98%、伪狂犬病毒(PRV)24.6%、乙型脑炎(JEV)20.9%、弓形虫(Tg)14.66%、蓝耳病病毒(PRRSV)11.5%、猪圆环病毒(PCV)7.2%、布鲁氏菌(Br)2.44%。其中单一感染占39.6%,混合感染占35.6%。从死亡新生仔猪和流产胎儿的脑及内脏中分离获得PPV、PRV、CP各2株病原,经细胞传代,鸡胚传代,毒价测定、形态观察、动物感染试验和应用PCR技术等鉴定,确证为伪狂犬、细小病毒病、衣原体三种。应用猪伪狂犬、细小病毒病、衣原体三种疫苗免疫注射,使本地区猪群的受胎率由92.7%提高到99.5%,妊娠母猪发病率从31.43%下降到0.77%,初生乳猪发病死亡率从24.9%下降到0.27%。调查研究表明,近年来引起红河州地区猪繁殖障碍综合征广泛流行的主要病因是PPV、PRV、CP三种病原单一或混合感染所致,而且动物感染试验证实是PPV、PRV、CP是造成猪繁殖障碍病的主要病原。  相似文献

In order to study the early events associated with infection of swine by Yersinia enterocolitica, 42 five-week-old crossbred piglets were inoculated per os with approximately 10(8) Y. enterocolitica O:3. Groups of 5 animals (and one negative control) were euthanized 30 min, 3, 6, 12, 24, 48 and 72 h following the infection. Palatine tonsils, retropharyngeal and mesenteric lymph nodes, esophagus, duodenum, jejunum, ileum (and Peyer's patches), stomach, liver, spleen and feces (from colon) were collected and analyzed for the presence of Y. enterocolitica by standard bacteriological procedures. Natural infections were also analyzed, as a complementary study, by taking one-gram samples of fecal material and tonsils from 291 pig carcasses less than 3 h after slaughter and culturing them for Y. enterocolitica using a cold enrichment technique. Within 30 min, Yersinia enterocolitica O:3 was already present at most sites. The presence of Y. enterocolitica in the liver of 3 out of 10 animals and also in the spleen of 3 out of 10 piglets, within the first 3 h postinfection, but not at later times (with one exception), probably indicated a transient bacteremia accompanying the initial stages of infection. The tonsils were colonized in most animals (13/20) as the bacteria remained present from 12 to 72 h postinfection, while only 4 out of 20 fecal samples were found to be positive over the same period. Up to 10(4) colony-forming units of Y. enterocolitica per gram of tonsil and fecal material were recovered. Finally, among the 291 animals sampled at the abattoir, a total of 79 were found positive, 70 of the tonsils sampled were positive, and bacteria were recovered in 17 fecal samples. It is therefore suggested that palatine tonsils are the most reliable tissue for the indication of an infection/colonization by Y. enterocolitica O:3 in swine and that the removal of this tissue during the slaughter process should be considered in order to minimize the possibility of contamination of meat products.  相似文献

Methicillin-resistant Staphylococcus aureus (MRSA), especially CC398, have emerged in livestock worldwide. We investigated the occurrence of MRSA in pigs at slaughter and in retail meat. During 2009, nasal swabs (n=789) were taken from pigs at slaughter. Moreover, 866 meat samples [Danish: pork (153), broiler meat (121), beef (142) and; imported: pork (173), broiler meat (193), and beef (84)] were randomly collected in retail stores and outlets. MRSA was isolated from nasal swabs or from meat samples after preenrichment (Mueller Hinton broth with 6.5% NaCl), selective enrichment (tryptone soya broth with 4 mg/L cefoxitine and 75 mg/L aztreonam) and selective plating on Brilliance Chromogenic MRSA agar. The presence of mecA was confirmed by PCR and the MRSA isolates were spa typed. Novel MRSA spa types were characterized by MLST, PFGE and SCCmec typing. Thirteen percent (101/789) of the pigs had MRSA. Based on spa types 93% corresponded to CC398 (spa t011, t034, t1451, t2876, t2974), 4% to CC30 (t1333) and one isolate to CC1 (t0127). The spa type t1333 (CC30), which is common among methicillin susceptible S. aureus (MSSA) from pigs in Denmark, contained a SCCmec cassette type V and czrC zinc resistance gene. Imported broiler meat had the highest occurrence (18%) of MRSA, followed by imported pork (7.5%) and Danish pork (4.6%). MRSA ST398 was found for the first time in Danish beef (1.4%). The finding of MRSA CC30 (spa t1333) suggest possible spread of the SCCmec cassette normally associated with ST398 into another S. aureus lineage common in pigs.  相似文献

In Denmark, a serological Salmonella surveillance programme in finishing pig herds has been in place since 1995. The programme was founded on data from experimental studies, which demonstrated a strong association between Salmonella serology and the prevalence of these bacteria. The current study was carried out in three Danish abattoirs to evaluate the correlation under field conditions. A total of 160 Danish finishing pig herds were included. Seven out of these were examined twice, yielding a total of 167 observations. The herds were selected according to their herd serology based on data from the national surveillance. From each herd, samples were taken from 10 finishers at slaughter. The prevalence of Salmonella bacteria was measured at four sites: (1) caecal-content; (2) carcass surface; (3) pharynx; and (4) caecal lymph nodes. A logistic regression model was constructed for each sampling site. Abattoir, sanitary slaughter and herd seroprevalence were used as explanatory variables. The results demonstrated that there was a strong association between herd serology and the prevalence of Salmonella bacteria measured at three of the sampling sites: caecal-content, pharynx, and carcass surface. For these sites, the odds for being culture-positive for Salmonella varied from 1.3 to 1.5 for each increase of 10% in herd serology (P < 0.0001). Three commercial swine herds were selected for study, because pigs at slaughter consistently had lung lesions typical of bronchopneumonia and snout lesions consistent with atrophic rhinitis. Pigs were reared in the conventional system for each herd except that they were identified at birth and weighted at various intervals. At slaughter, individual pig lungs and snout were examined for lesions of pneumonia and atrophic rhinitis, respectively. Lesions were scored and correlated with growth indicators for each pig. Included in the growth indicators were: average daily gain (growing phase), average daily gain (finishing phase), average daily gain (total), and days to reach 104.5-kg body weight. Additionally, for each pig, scores for lung lesions were correlated to grades for snout lesions. Three correlation coefficients for measurements of pigs within herd B were significant and included days to 104.5-kg body weight and grades for snout lesions, -0.15 (P less than 0.02); average daily gain (finishing) and grades for snout lesions, 0.17 (P less than 0.01); and average daily gain (total) and grades for snout lesions, 0.16 (P less than 0.01). Contrary to findings in other investigations, pigs that attained market weight at the youngest age did not have the lowest score for lung lesions, the lowest grade for snout lesions, or the least extensive or severe lesions. Combining data from all 3 herds, the mean scores for lung lesions and mean grades for snout lesions decreased significantly (P less than 0.05) as the age of pigs at slaughter increased. Background

The use of secondary data is widespread in a range of surveillance and monitoring applications because of the low cost and high availability associated with this form of data. However, as they are often collected for quite unrelated purposes, they are not necessarily fit for the new purpose that is required of them. Routine meat inspection data were originally collected with the purpose of safeguarding food, but have been re-tasked to also include animal welfare assessment. The objective of the present study was to compare the recording of pericarditis, pleuritis and lungs with lesions at routine meat inspection (RMI) with those performed at systematic health monitoring (SHM) in Danish pigs at slaughter, in order to assess the usefulness of RMI for monitoring the prevalence of these diseases. Data originating from 165 Danish pig herds were collected in the period September 2011 to November 2013. From each herd, a batch consisting of all pigs slaughtered on a specific day from a specific farm were included as the RMI data, while lungs and hearts sampled from the batches were used for the SHM. The RMI data and SHM data included recordings related to a) chronic pericarditis, b) chronic pleuritis and c) lung lesions. The proportion of carcases with a specific disease recording was estimated for each batch of pigs, and linear regression was used to relate the RMI-proportion to the SHM-proportion for the conditions mentioned above.

Results

The coefficients of determination (R2) were estimated as R2,pericarditis = 0.16; R2,pleuritis = 0.67; R2,lungs with lesions = 0.40. R2,pericarditis changed to 0.42 when the regression analysis included inspection type at the abattoir (with purely visual inspection of the hearts versus traditional inspection including an incision into the heart).

Conclusions

Overall, the results suggest that the correlation between findings at RMI and SHM was moderate for pleuritis and lungs with lesions, but poor for pericarditis. The latter could partly be explained by the type of meat inspection conducted at the abattoir. We conclude that caution should be used whenever RMI data are used for purposes other than those for which they were originally intended.  相似文献
