Transportation causes stress in cattle that may alter numerous physiological variables with a negative impact on production and health. The objectives of the current study were to investigate the physiological effects of truck transportation and to characterize a pattern of phenotypes in the circulation that may aid in the early identification of stress-susceptible animals that often succumb to severe respiratory disease. Thirty-six young beef bulls (Aberdeen Angus, n = 12; Friesian, n = 12; and Belgian Blue x Friesian, n = 12) were subjected to a 9-h truck transportation by road. Blood (10 mL) was collected at -24, 0, 4.5, 9.75, 14.25, 24, and 48 h relative to the initiation of transportation (0 h). Plasma was collected for the assay of various metabolic, inflammatory, and steroid variables, and total leukocyte counts were determined in whole blood at each time point. Body weight and rectal temperature were recorded at -24, 9.75, and 48 h. Transportation decreased measures of protein metabolism in the plasma, including albumin (P = 0.002), globulin (P < 0.001), urea (P = 0.006), and total protein (P < 0.001), and increased creatine kinase (P < 0.001). The energy substrate beta-hydroxybutyrate was not changed (P = 0.27). Acute phase proteins haptoglobin and fibrinogen were both decreased (P < 0.001), whereas total leukocyte counts were elevated (P = 0.002). Circulating steroid concentrations were altered, because a classical acute increase in plasma cortisol was observed with the onset of transit (P < 0.001), in association with a decrease in dehydroepiandrosterone (P = 0.07), resulting in a profound increase in cortisol:dehydroepiandrosterone ratio (P < 0.001). Plasma testosterone was decreased, whereas plasma progesterone was increased (P < 0.001) in association with the increase in cortisol (P < 0.001). There was also an effect of breed for all variables except plasma urea, creatine kinase, and testosterone, perhaps indicating that a genetic component contributed to the physiological response to transportation stress, although without any clear trend. Taken together, this profile of physiological variables in the circulation of transportation-stressed bulls may aid in the future detection of disease-susceptible cattle after transportation. Further research to validate these potential biomarkers is necessary.  相似文献

The transportation of beef cattle results in a stress response that is associated with increased susceptibility and severity of respiratory diseases, presumably due to an alteration in immune function. Neutrophils are phagocytic immune cells important in lung defense and are also targets of the stress response. The objective of this study was to determine if a 9h transportation of young bulls by road induced changes in the expression of candidate genes known to be important in neutrophil-mediated defense and inflammation in the lung. These neutrophil genes encompassed functions of apoptosis (A1 and Fas), tissue remodeling (MMP-9), vascular margination (L-selectin), bacterial killing (BPI), and wound healing (betaglycan), as well as responsiveness of the cells to stress-induced increases in glucocorticoid hormones (GRalpha). To explore gene expression changes, blood was collected, plasma harvested, and neutrophils isolated from six Belgian Blue x Friesian bulls (231+/-7.0 kg in weight; 282+/-4 days of age) at -24, 0, 4.5, 9.75, 14.25, 24, and 48h relative to commencement of a 9h road transportation by truck. Plasma cortisol concentrations were elevated at 4.5 and 9.75h, peaking at 50.64+/-4.46 ng/mL (P<0.0001) and confirming that the animals experienced stress. Blood neutrophil count was elevated between 4.5 and 14.25h (P<0.0001), reaching a peak that was over 3-fold higher than the -24h concentration. Neutrophil Fas gene expression was acutely down-regulated (P=0.02) by transportation stress, while expressions of MMP-9, l-selectin, and BPI were profoundly up-regulated (P=0.003, 0.002, and <0.001 respectively). However, no changes in neutrophil expressions of betaglycan, GRalpha, and A1 were detected. It is concluded that a 9h transportation of young bulls induces a gene expression signature in blood neutrophils that increases their circulating numbers and may enhance their pro-inflammatory and anti-bacterial potential.  相似文献

This study investigated the effects of transportation stress on blood concentrations of the main pro-inflammatory cytokines (interleukins IL-1β, IL-2 and IL-6; tumour necrosis factor-α) and anti-inflammatory cytokines (IL-4 and IL-10) and the expression of these cytokines and their receptors in the thymus. Pigs were assessed after 1, 2 and 4 h of transportation (n = 5 per group), with normal housing conditions as a control (n = 4). Serum concentrations of IL-2, IL-6 and IL-10 were highest at 1 h, whereas concentrations of IL-6 and IL-10 were significantly decreased at 4 h. Expression of these three cytokines and their receptors was also significantly altered in the thymus during transportation stress. Serum IL-10 concentrations and thymus IL-10 mRNA expression were significantly correlated. The thymus may contribute towards the regulation of cytokines in pigs during transportation.  相似文献

Twenty-one Simmental crossbred bulls (311 +/- 11 kg, 9 mo of age) were used to determine the effect of feeding 10, 12 or 14% CP on concentration of hormones in blood and the relationship of these hormones to composition of gain. Six bulls were slaughtered on d 0 to provide an estimate of initial carcass composition (9-11 rib section). Remaining bulls were assigned to dietary treatments. Blood samples were collected every 30 min from 0800 to 2000 on d 0, 66, 136 and 202 of treatment; bulls were slaughtered on d 203. Across all treatments, growth hormone (GH) declined (P less than .05) from d 0 to d 202. Free insulin-like growth factor I (IGF-I) was lowest (P less than .05) on d 0. In four randomly selected bulls, IGF-I fluctuated during the 12-h sampling periods. Within each treatment group, insulin was greatest on d 202 (P less than .05). Testosterone (T) increased from d 0 to d 66, then declined. Cortisol (C) was lowest on d 66. Thyroid hormones increased (P less than .05) after d 0. Growth hormone and IGF-I were correlated negatively with carcass fat percentage, fat accretion rate and fat thickness. IGF-I concentrations were correlated positively with percentage of carcass protein. Testosterone:cortisol ratio was not related to composition, but high T coupled with low C may be related to carcass leanness (mean carcass fat = 24.4%). These data suggest that GH and IGF-I are the hormones most related to composition of gain in growing beef bulls.  相似文献

Premature spiral deviation of the penis was investigated in 1083 British breed beef bulls in 42 herds in Victoria (35) and Western Australia (7) by means of the serving capacity test. The herd prevalence was 60%. The prevalence of the defect at the initial test for all bulls was 10%. The prevalence in 635 polled bulls was 16% compared to a prevalence of 1 % in 448 horned bulls. The breed prevalence and the number of bulls tested per breed was Red Poll 27% (15), Murray Grey 25% (4), Poll Shorthorn 21 % (34), Angus 17% (415), Poll Hereford 10% (167) and Hereford 1 % (448). The difference between the last 2 breed types was highly significant (P<0.001). The prevalence of the defect increased from 0 to 30% over 9 years in one Victorian herd. The age of affected bulls varied from 2 to 11 years with most affected bulls aged from 3 to 6 years. Common ancestry was found between affected bulls in Western Australia, providing circumstantial evidence that it may be an inherited defect. None of the 103 affected bulls examined in this study had suffered obvious injury to the penis or prepuce. Eighty per cent of 56 affected bulls from Victoria had low serving capacity. Four bulls (2 from each state) with moderate to severe spiral deviation of the penis attained pregnancy rates of 43%, 33%, 3% and 3% respectively, in single sire mating trials. Recommendations for culling defective bulls are presented, based on the herd prevalence and severity of the defect in individual bulls.
It is concluded that bulls with moderate to severe spiral deviation of the penis have reduced fertility and remain permanently affected. The serving capacity test is a valuable diagnostic test for this serving defect in unhandled beef bulls. It is a common, although frequently undiagnosed disorder of polled beef breed bulls in Australia.  相似文献

Performance testing started after it was recognized that growth traits were heritable. In the early years of performance testing there was a tendency to feed higher levels of energy for longer periods of time. More recently, the trend has been to feed lower levels of energy for shorter periods. There are still differences in opinion as to the appropriate level of energy to use. Although it is important that the level of energy fed is adequate to correctly establish a bull's ability to gain, it is essential to know that it will pose no risk of impaired spermatogenesis or cause any degree of laminitis. Clinical observations and research on overfeeding clearly show that both libido and spermatogenesis can be impaired by excess energy intake. The damage in 2-year-old bulls can be very extensive and in some animals it may not be reversible. The scant amount of research in yearling bulls indicates that there is considerable potential danger from overfeeding energy as well. Test stations are under used in regard to performing research that would help identify heritable defects that would interfere with the productive and reproductive efficiency of beef cattle. The first performance testing programs emphasized average daily gain from weaning to 1 year of age, so "performance" has traditionally meant rate of gain to most cattle raisers. The term "performance" is now starting to acquire a broader and more inclusive definition. For many breeders, it now includes weight per day of age, which is in part a maternal trait, and some kind of male evaluation for reproductive potential that can also be extrapolated to the female side. One of the first breakthroughs in this regard was to recognize the heritability of testicular size, and that testicular size could be fairly accurately determined by scrotal circumference measurement. It was also found that there was a favorable relationship between larger testicle size and the ability to produce high quality semen. As a result, it became a common practice to include scrotal circumference measurements in the published bull test results. However, many test station patrons were, and still are, content to consider the scrotal circumference measurement alone as an evaluation of a bull's breeding potential. Blood samples were taken from bull calves at two Meat and Livestock Commission performance testing centres, just after weaning at six months of age and at six weekly intervals until the end of the performance test seven months later. Sera were assayed by specific ELISAS for antibodies to bovine herpes virus 1 (BHV1), respiratory syncytial virus, parainfluenza 3 (Pi3) and adenoviruses A and B. Seroconversions between each sampling were related to the occurrence of clinical respiratory disease using chi-squared (chi 2) and relative risk (RR) analyses. In 294 bulls there were 123 cases of respiratory disease. Seroconversion to bovine respiratory syncytial virus (RR = 4.7, chi 2 = 96.3, P less than 0.001) and adenovirus A (RR = 1.8, chi 2 = 8.9, P less than 0.001) and adenovirus B (RR = 1.9, chi 2 = 5.6, P less than 0.05) were significantly associated with clinical respiratory disease. There was evidence that prior exposure to respiratory syncytial virus (RR = 0.4, chi 2 = 9.8, P less than 0.01) Pi3 (RR = 0.4, chi 2 = 12.8, P less than 0.01) and adenovirus A (RR = 0.7, chi 2 = 7.5, P less than 0.01) conferred some protection against respiratory disease after arrival at the centre. It is concluded that vaccination before weaning, at least against bovine respiratory syncytial virus, would be beneficial.  相似文献

Scrotal circumference measurements were analyzed to develop appropriate adjustment factors for age and body weight differences in beef bulls. Age, body weight, and scrotal circumference measurements were collected on 863 Angus, 753 Polled Hereford, and 302 Simmental bulls from the Missouri tested bull sale, 1169 Polled Hereford bulls from the American Polled Hereford Association, and 465 Polled Hereford and 264 Simmental bulls from Nichols Farms in Bridgewater, Iowa. Within sources, data were subdivided into appropriate subsets and then each subset was statistically analyzed. Adjustment factors to remove differences in scrotal circumference due to age and body weight were developed for bulls of each of the 3 breeds studied.  相似文献

For 28 weeks a feeding experiment was conducted to determine the effects of dietary flavomycin on performance of beef bulls. In the first and second half of the trial, 40 animals (average initial weight 352 kg) had free access to corn silage or beet pulp, respectively, both supplemented with an appropriate concentrate containing 0 or 10 mg/kg of the antibiotic. Daily flavomycin intake was 39 to 46 mg when the animals were on the corn silage diet and 51 to 54 mg when they were fed the beet pulp diet. The animals fed the corn silage-based diet supplemented with flavomycin showed a non-significant improvement of daily gain (1.4%) and feed conversion (2.3%). When the beet pulp-based diet was fed, flavomycin significantly increased daily gain (15.2%, P less than 0.002) and feed conversion efficiency (9.1%, P less than 0.03). For the overall feeding experiment the positive effect of flavomycin feeding was 6.7% on body weight gain (P less than 0.09) and 5.2% on feed conversion (P less than 0.13). Flavomycin did not affect carcass yield of beef bulls. Also the effect of flavomycin on diet digestibility was studied in wethers fed a mixture of beet pulp and concentrate, containing 0 or 10 ppm flavomycin. No significant effect from the antibiotic on digestion of dry matter, crude protein, fibre, fat, ash and N-free extract was observed.  相似文献

This study tested the hypothesis that the increased glucose requirement of lactation had effects that were independent of the suckling-dependent inhibition of postpartum endocrine function in beef cows. Mature Hereford cows were either suckled ad libitum and infused with saline iv (n = 9) from d 2 through 4 (d 0 = jugular catherization on d 32 +/- 3 postpartum); were nonsuckled and infused with saline from d 2 through 4 (n = 10); or were nonsuckled and infused with phlorizin (3 g/d) from d 2 through 4 (n = 10). Nonsuckled cows infused with phlorizin had lower (P less than .05) plasma concentrations of glucose and amino acid nitrogen (AAN) on d 2 compared with pre-infusion levels (d 1), but their metabolic profile returned to levels similar to the suckled cows by d 3 and 4. Nonsuckled cows infused with saline had elevated glucose and insulin and lower AAN and free fatty acids (FFA) on d 3 and 4 compared with pre-weaning (d 1) levels (P less than .05). Nonsuckled cows infused with phlorizin did not show this weaning-induced elevation in glucose and insulin. The number of luteinizing hormone (LH) pulses was not affected by treatment. However, in contrast to the large LH pulses observed in the nonsuckled cows infused with saline, both the suckled cows and the nonsuckled cows treated with phlorizin had more small and fewer large amplitude pulses (P less than .01). Treatment did not affect serum concentrations of follicle stimulating hormone, gonadotropin release in response to gonadotropin releasing hormone (25 micrograms) or the number of cows ovulating by 55 d after calving. We conclude that the increased glucose clearance caused by phlorizin infusion or lactation results in depression of LH pulse amplitude in suckled postpartum beef cows.  相似文献

To determine the effects of Follicle Stimulating Hormone (FSH) treatment and subsequent withdrawal on uterine proliferation and estrogen receptor (ESR), Brahman crossbred heifers (n = 12) were twice daily injected with FSH (4, 3, and 2 mg/injection) on Days 17–19 of the estrous cycle (FSH 3 days) and (4 and 3 mg/injection) on Days 17–18 (FSH 2 days) and withdrawal with saline on Day 19 and (4 mg/injection) on Day 17 (FSH 1 day) and withdrawal with saline on Days 18–19. Uterine tissue was subjectively collected on Day 20 and microscopically classified to four regions: endometrial stroma (ES), surface endometrial gland (EG), deep endometrial gland (DG), and myometrium (Myo). The cell proliferation marker, Ki-67, was quantified as labeling index (LI) in uterine regions, and tissues were immunostained to detect ESR2 followed by image analysis. The LI of ES, EG, and DG was greater (P = 0.0018, P = 0.0005, and P = 0.0103; respectively) in heifers received FSH for 3 days. The expression of ESR2 protein on ES and EG was greatest (P < 0.0001 and P = 0.0036, respectively) in FSH 3 days-treated group. Thus, FSH administration during proestrus stimulates uterine cell proliferation, and ESR2 expressions are affected by FSH during proestrus and differentially distributed in the uterine regions.  相似文献

We tested the hypothesis that expression of sexual behavior in bulls is affected by the manner in which they are exposed to unrestrained, sexually receptive females. Twelve Angus bulls were used in a crossover design involving two treatments, each tested four times for a total of eight tests for each bull. Sexual interactions were quantified for each of four, 30-min periods under the following treatments: 1) exposure to each of four estrual females in sequence (SEQ); or 2) exposure to four estrual females as a group (GRP). Bulls were blocked into three testing groups, the order of which was stratified across eight test days. The order in which bulls were tested on a particular day had no effect on bulls' expression of mount interactions, or flehmen responses, suggesting that each group of bulls had similar sexual motivation at the beginning of each test. However, the bull testing order x treatment x time interaction influenced mounting interval (P = 0.08), copulation frequency (P <0.05), and copulation success ratio (P <0.05). When bulls were in GRP and tested first on test days, more (P <0.05) copulations were distributed to the first three females encountered compared with either the fourth female (P <0.05) or to each of the other females in SEQ (P <0.05). During later tests, other bulls in GRP were not able to copulate as frequently (P <0.05) with each female, displayed lower (P <0.05) copulation success ratios, and were allowed copulations by fewer (P <0.05) females during each 30-min test. When bulls were in SEQ, they displayed similar numbers of copulations regardless of the order in which they were tested, and had stable mounting intervals; however, copulation success ratio decreased (P <0.05) more rapidly during subsequent tests. Flehmen responses were initially displayed more frequently (P <0.05) when bulls were in GRP, but this effect diminished during subsequent 30-min tests. In conclusion, exposure of bulls to GRP induced greater sexual responsiveness than SEQ; however, this effect was due to enhanced sexual activity during the early stages of sexual encounters and with females that were not recently mated. Interestingly, bulls seem to repeatedly copulate with each individual female until, apparently, female sexual receptivity became attenuated. Thereafter, recently mated females allowed fewer episodes of repeated copulations, but they did not completely cease copulating with novel bulls.  相似文献

Accuracy of culture for diagnosis of Tritrichomonas foetus was investigated in 2832 naturally exposed range beef bulls from 124 herds. Preputial fluid samples were inoculated into the culture medium, incubated at 37 degrees C, and daily examined. Diagnostic test was evaluated using Bayesian techniques to estimate sensitivity and specificity without a gold standard. Median posterior test sensitivity was 72.04% (95% probability interval: 58.07-86.38%) and specificity was 95.37% (95% probability interval: 94.07-96.65%). Low diagnostic test accuracy may have resulted from host and/or diagnostic test procedure related factors. Under natural range conditions, more accurate methods for T. foetus diagnostic and repeated preputial samplings of bulls may be necessary on trichomonosis control programs.  相似文献

Three experiments were conducted with 5 mg Mestranol per animal and die over 70 to 80 days and resulted in daily surplus gains of twelve, 14, and 18 per cent in terminal fattening of Frisian bulls. Also achieved were fodder savings by 1 kg dry matter and energy savings by 0.7 kEFr for each kilogram of gain as well as increase in slaughter weight by 8-9 kg per animal. Those favourable effects were attributed to anabolic and sexual-inhibiting action. A sedative effect on the nervous system is assumed. Combination with CAP failed to give any advantage regarding sexual inhibition and fattening result, but it rather entailed certain shortcomings, such as lower slaughter weight and stronger fat development. The development of an anabolic medicament for veterinary bio-engineering in terms of growth stimulation is suggested.  相似文献
