The combination of specific probes and advanced optical microscopy now allows quantitative probing of biochemical reactions in living cells. On selected systems, one can detect and track a particular protein with single-molecule sensitivity, nanometer spatial precision, and millisecond time resolution. Metabolites, usually difficult to detect, can be imaged and monitored in living cells with coherent anti-Stokes Raman scattering microscopy. The mechanism of formation of polymer molecules in the gas phase is difficult to study because the involatile polymers tend to condense out of that phase. However, new techniques, involving the use of cloud chambers, have enabled workers to use the nucleation of liquid drops in supersaturated monomer vapors to detect single polymer molecules and therefore to work with so few simultaneously growing polymers that aggregation and condensation are avoided. Chain polymerization in which the chain carriers are either radicals or ions can therefore be studied in the vapor. Furthermore, the ability to work with such small concentrations of growing polymeric radicals, for example, makes it possible to avoid encounters between them that lead to recombination and the formation of "dead" polymers that are incapable of further growth. Many aspects of gas-phase polymerization can be studied including, besides radical and ion chains, ring-opening polymerization, initiation, radiation-induced polymerization, and especially "ultraslow" chemistry.  相似文献

F-actin is the major component of muscle thin filaments and, more generally, of the microfilaments of the dynamic, multifunctional cytoskeletal systems of nonmuscle eukaryotic cells. Polymeric F-actin is formed by reversible noncovalent self-association of monomeric G-actin. To understand the dynamics of microfilament systems in cells, the dynamics of polymerization of pure actin must be understood. The following model has emerged from recent work. During the polymerization process, adenosine 5'-triphosphate (ATP) that is bound to G-actin is hydrolyzed to adenosine 5'-diphosphate (ADP) that is bound to F-actin. The hydrolysis reaction occurs on the F-actin subsequent to the polymerization reaction in two steps: cleavage of ATP followed by the slower release of inorganic phosphate (Pi). As a result, at high rates of filament growth a transient cap of ATP-actin subunits exists at the ends of elongating filaments, and at steady state a stabilizing cap of ADP.Pi-actin subunits exists at the barbed ends of filaments. Cleavage of ATP results in a highly stable filament with bound ADP.Pi, and release of Pi destabilizes the filament. Thus these two steps of the hydrolytic reaction provide potential mechanisms for regulating the monomer-polymer transition.  相似文献

A living, ammonia-obligate, umbellate form, similar to the Precambrian microfossil Kakabekia umbellata Barghoorn, has been isolated from two soil specimens collected at Harlech, Wales. This organism is amenable to culture on agar and in broth. The two soil specimens are similar in that they differ from a typical clay loam in high content of carbon, hydrogen, and organic nitrogen and low levels of sodium, potassium, and titanium. In all other constituents, such as calcium, magnesium, and iron, they are quite dissimilar. Kakabekia-like forms can be grown in glucose-anmonia media with the latter as the sole source of nitrogen, but they can also be grown on peptone and silicate in glucose-free media. Ammonia is necessary, and growth is always slow without glucose. The fission process was not observed, but the enlargement and differentiation of a preumbellate structure into its "mature" form, followed by disintegration (senescence) of this stage, was seem. An ontogeny is proposed in which the stalk and basal bulb of the complete umbellate structure are assumed to be part of the reproductive appartus.  相似文献
