The direct influence of intracaecal infusion of short-chain fatty acids (SCFA) and lactic acids (LA) on already established Oesophagostomum dentatum infection in cannulated pigs was investigated. We tested the hypothesis that the previously discovered anti-parasitic effect of inulin is mediated through its metabolic products SCFA and LA by infusing into cannulated pigs these compounds in amounts approximating to those produced in the pigs large intestine and caecum during the metabolism of inulin. The experiment comprised of 18 pigs--2 groups of 9 pigs in each. The normal diet used in the experiment was based on barley flour with insoluble fibre from oat husk with added soybean meal, vitamins and minerals. After 2 weeks of adaptation to the diet all the pigs were inoculated with 6,000 infective larvae of O. dentatum. Six weeks later, surgery on all pigs was performed to install cannulas into caeci. At 7 weeks post-infection (p.i.) the SCFA and LA infusion was initiated in Group 1 (experimental) pigs; at the same time pigs in Group 2 (controls) were infused with saline. At week 10 p.i., all pigs were killed and their worm burdens determined. SCFA and LA infused pigs exhibited markedly reduced fecal egg counts and worm recoveries (98 and 92% reduction, respectively, compared to saline controls). The results from this study demonstrate that SCFA and LA have a significant negative influence on established O. dentatum infection in growing pigs. The results also show that the type of dietary carbohydrates fed and its intestinal degradation can yield metabolic by products that profoundly influence helminth survival. 相似文献

The aim of this study was to examine interactions between Ascaris suum and Oesophagostomum dentatum infections in pigs with regard to population dynamics of the worms such as recovery, location and length; and host reactions such as weight gain, pathological changes in the liver and immune response. Seventy-two helminth-na?ve pigs were allocated into four groups. Group A was inoculated twice weekly with 10000 O. dentatum larvae for 8 weeks and subsequently challenge-infected with 1000 A. suum eggs, while Group B was infected with only 1000 A. suum eggs; Group C was inoculated twice weekly with 500 A. suum eggs for 8 weeks and subsequently challenge-infected with 5000 O. dentatum larvae, whereas Group D was given only 5000 O. dentatum larvae. All trickle infections continued until slaughter. Twelve pigs from Group A and B were slaughtered 10 days post challenge infection (p.c.i.) and the remaining 12 pigs from the each of the four groups were slaughtered 28 days p.c.i.. No clinical signs of parasitism were observed. The total worm burdens and the distributions of the challenge infection species were not influenced by previous primary trickle-infections with the heterologous species. Until day 10 p.c.i. the ELISA response between A. suum antigen and sera from the O. dentatum trickle infected pigs (Group A) pigs were significantly higher compared to the uninfected Group B. This was correlated with a significantly higher number of white spots on the liver surface both on Day 10 and 28 p.c.i. in Group A compared to Group B. The mean length of the adult O. dentatum worms was significantly reduced in the A. suum trickle infected group compared to the control group. These results indicate low level of interaction between the two parasite species investigated. 相似文献

Two strains of Oesophagostomum spp., consisting of both O. quadrispinulatum and O. dentatum, were subjected to a controlled in vivo assay for resistance to levamisole and pyrantel by comparison with susceptible isolates. One strain (LEM) was recently isolated from a commercial herd, where sows showed high numbers of strongyle eggs in faeces within 2 weeks of farrowing and following treatment with levamisole at the manufacturer's recommended dose rate 1 week before farrowing. Levamisole had been used as the sole anthelmintic for treatment for at least 7 years on this farm. Treatment with pyrantel in this herd also indicated cross-resistance to this drug. A mixed population of O. quadrispinulatum and O. dentatum of this strain was subjected to controlled in vivo assays. Faecal egg count reduction (FECR) was found to be -573.3% (P greater than 0.05) and worm count reductions (WCR) of O. quadrispinulatum and O. dentatum were estimated as 44.5% (P greater than 0.05) and 96.4% (P less than 0.001), respectively. Treatment with pyrantel showed that cross-resistance existed to this drug, with FECR of 10.4% (P greater than 0.05) and WCR of 64.5% (P greater than 0.05) and 90.7% (P less than 0.05) for O. quadrispinulatum and O. dentatum, respectively. Another strain (VJ) was isolated from another commercial pig herd, which was dosed with pyrantel citrate four times a year for at least 8 years. This strain showed resistance to pyrantel, with FECR of 43.8% (P greater than 0.05) and WCR of 65.9% (P greater than 0.05) and 49.4% (P greater than 0.05) for O. quadrispinulatum and O. dentatum, respectively. However, both species were susceptible to levamisole. Our results suggested that selection with levamisole gave rise to levamisole resistance and automatically conferred resistance to pyrantel, whereas selection with pyrantel only resulted in resistance to this drug alone. These findings are discussed in relation to the location of the two species of Oesophagostomum in the large intestine of pigs and the mode of action of this class of anthelmintics. 相似文献

The aim of the present study was to investigate the population dynamics and potential interactions between Trichuris suis and Oesophagostomum dentatum in experimentally co-infected pigs, by quantification of parasite parameters such as egg excretion, worm recovery and worm location. Forty-eight helminth naïve pigs were allocated into four groups. Group O was inoculated with 20 O. dentatum L3/kg/day and Group T with 10 T. suis eggs/kg/day. Group OT was inoculated with both 20 O. dentatum L3/kg/day and 10 T. suis eggs/kg/day, while Group C was kept as an uninfected control group. All inoculations were trickle infections administered twice weekly and were continued until slaughter. Faecal samples were collected from the rectum of all pigs at day 0, and twice weekly from 2 to 9 weeks post first infection (wpi). Six pigs from each group were necropsied 5 wpi and the remaining 6 pigs from each group were necropsied 10 wpi. The faecal egg counts (FEC) and total worm burdens of O. dentatum were dramatically influenced by the presence of T. suis, with significantly lower mean FECs and worm burdens at 5 and 10 wpi compared to single infected pigs. Furthermore, in the presence of T. suis we found that O. dentatum was located more posteriorly in the gut. The changes in the Trichuris population were less prominent, but faecal egg counts, worm counts 5 wpi (57% recovered vs. 39%) and the proportion of infected animals at 10 wpi were higher in Group OT compared to Group T. The location of T. suis was unaffected by the presence of O. dentatum. These results indicate an antagonistic interaction between T. suis and O. dentatum which is dominated by T. suis. 相似文献

The aim of the present study was to investigate parasite induced immune responses in pigs co-infected with Trichuris suis and Oesophagostomum dentatum as compared to mono-species infected pigs. T. suis is known to elicit a strong immune response leading to rapid expulsion, and a strong antagonistic effect on O. dentatum populations has been observed in co-infected pigs. Forty-eight helminth naïve pigs were allocated into 4 groups in a 2-factorial design. Two groups were trickle inoculated with either 10 T. suis eggs/kg/day (Group T) or 20 O. dentatum L3/kg/day (Group O). Group OT was infected with same levels of both T. suis and O. dentatum (Group OT) and Group C remained uninfected. In each group, six pigs were necropsied after 35 days and the remaining pigs after 71 days. Parasite E/S-antigen specific serum antibodies were quantified by an in-direct ELISA. qPCR was used to measure the expression of immune function related genes in the mucosa of proximal colon and the draining lymph node. Highly significant interactions were identified for O. dentatum specific IgG1 (p < 0.0001) and IgG2 (p < 0.0006) antibodies with a remarkable 2-fold higher antibody response in group OT pigs as compared to group O. These findings indicated that T. suis enhanced the antibody response against O. dentatum in Group OT. The gene expression data confirmed a strong Type 2 response to T. suis (e.g. marked increase in IL-13, ARG1 and CCL11) and clearly weaker in amplitude and/or delayed onset response to O. dentatum in the single infected group. Interactions were found between the two nematodes with regard to several cytokines, e.g. the increase in IL-13 observed in Group T was absent in Group OT (p = 0.06, proximal colon mucosa, 35 and 71 p.i.). Some of these immune response-related interactions may support, or even partially explain, the observed interactions between the two worm populations in co-infected pigs. 相似文献

The present work was undertaken to examine the effects of 3 different population densities of Oesophagostomum dentatum upon the development of worm induced mucosal changes in the colon following single infections. Groups of pigs were infected with single doses of 2000 (low dose), 20,000 (medium dose) or 200,000 (high dose) infective larvae, respectively. A total of 18 infected pigs (6 from each group) were examined for histopathological changes together with 3 helminth-free control pigs. There was a dose related difference in the intensity of colonic lesions; and using morphometry it was observed that the mucosal crypts of pigs in the high dose group were significantly longer than those in the 2 other groups. These differences disappeared by day 25 after infection despite the presence of larvae in the mucosa of the high dose group. This phenomenon may be related to inflammatory reactions in the colon, possibly in connection with the initiation of an immunological response in sites distant from the parasite larvae. 相似文献

The vertical migratory behavior of third-stage infective larvae (L3i) of Oesophagostomum dentatum was investigated using upright truncated agarose cones and equivalent conical depressions in agarose. Geotactic response varied with the age of the infective larvae. In this study, the effect of feeding different types and amounts of dietary fiber (DF) on luminal environment and morphology in the small and large intestine of sows was studied. Three diets, a low-fiber diet (LF) and 2 high-fiber diets (high fiber 1, HF1, and high fiber 2, HF2) were used. Diet LF (DF, 17%; soluble DF 4.6%) was based on wheat and barley, whereas the 2 high-fiber diets (HF1: DF, 43%; soluble DF, 11.0%; and HF2: DF, 45%; soluble DF, 7.6%) were based on wheat and barley supplemented with different coproducts from the vegetable food and agroindustry (HF1 and HF2: sugar beet pulp, potato pulp, and pectin residue; HF2: brewers spent grain, seed residue, and pea hull). The diets were fed for a 4-wk period to 12 sows (4 receiving each diet). Thereafter, the sows were killed 4 h postfeeding, and digesta and tissue samples were collected from various parts of the small and large intestine. The carbohydrates in the LF diet were well digested in the small intestine, resulting in less digesta in all segments of the intestinal tract. The fermentation of nonstarch polysaccharides in the large intestine was affected by the chemical composition and physicochemical properties. The digesta from pigs fed the LF diet provided low levels of fermentable carbohydrates that were depleted in proximal colon, whereas for pigs fed the 2 high-DF diets, the digesta was depleted of fermentable carbohydrates at more distal locations of the colon. The consequence was an increased retention time, greater DM percentage, decreased amount of material, and a decreased tissue weight after feeding the LF diet compared with the HF diets. The concentration of short-chain fatty acids was consistent with the fermentability of carbohydrates in the large intestine, but there was no effect of the dietary composition on the molar short-chain fatty acid proportions. It was further shown that feeding the diet providing the greatest amount of fermentable carbohydrates (diet HF1, which was high in soluble DF) resulted in significant morphological changes in the colon compared with the LF diet. 相似文献

Abstract An experiment was conducted to investigate changes in the lumen and wall of the intestinal tract of weaned pigs caused by fructo-oligosaccharides (FOS) and consequences for growth. Twelve male pigs weaned at 24 days were individually housed and fed either control, 0.25% FOS, or 3% FOS diets for 21 days after weaning. The pH of chyme in the caecum and proximal colon were lower (p<0.001) in 3% FOS compared to other treatments. Mean molar proportions (%) of total SCFA values along the large intestine were significantly affected by FOS diet. Acetic acid was significantly higher in control compared with other treatments. Butyric acid and isobutyric acid were significantly increased in 0.25 and 3% FOS compared to the control. Feed intake, empty body weight gain, disaccharidase activities and villous height in the small intestine were not significantly affected by FOS. It was concluded that dietary FOS stimulates fermentation in the large intestine, but does not significantly affect enzyme activity, ammonia content, villous height and growth. 相似文献

A 2 x 3 factorial experiment was conducted with 24 finishing pigs (Yorkshire x [Finnish Landrace x Dutch Landrace]) to determine the effects of dietary buffering capacity (BC) and carbohydrate sources on apparent total tract digestibility (TD), N retention, and manure characteristics. Twelve of these pigs were fitted with steered ileo-cecal value cannulas to measure the apparent ileal digestibility (ID) of N. Experimental variables were two levels of BC (High = 600 mEq/kg and Low = 530 mEq/kg) and three dietary carbohydrates (tapioca [28%], soybean hulls [25%], and sugar beet pulp [25%]). The two levels of BC were achieved by adjusting the amounts of alkalogenic limestone and acidogenic Ca sulfate in the diet. Pigs were fed twice daily at 2.4 x maintenance requirement for metabolizable energy (418 kJ ME/BW(0.75)). Chromic oxide was used as an indigestible marker. Feces and urine were collected over 5 d for nutrient balance and in vitro measurements of pH and ammonia emission over 7 d. No significant interactions between BC and dietary carbohydrates on the ID, TD, N retention, or manure characteristics were observed. Lowering BC with Ca sulfate did not affect digestibility of most nutrients, except for a negative impact on the TD of Ca (P = 0.015) and Mg (P = 0.003). Although all pigs receiving Ca sulfate had more acidic urine (by 0.49 pH units; P = 0.001), ammonia emission from manure was not lowered, irrespective of the carbohydrate source. Carbohydrates affected significantly the ID and TD of most nutrients, whereas N retention remained similar (P > 0.10). Urinary N:fecal N ratios in manure of pigs fed diets with tapioca, soybean hulls, and beet pulp were 2.09, 1.35, and 1.67, respectively. These ratios corresponded with in vitro ammonia emission (169, 125, and 148 mmol/7 d; P = 0.023). In conclusion, these results indicate that the acidification of urine in pigs fed Ca sulfate in the presence of NSP-rich carbohydrates was achieved. However, acidity of manure and ammonia emission were not affected. The addition of NSP from soybean hulls and(or) sugar beet pulp reduced ratios of urinary:fecal N, and thereby ammonia emission. 相似文献

A study was conducted to evaluate and compare the efficacy of two injectable formulations of ivermectin (IVM-1 and IVM-2) at a dose rate of 0.3 mg/kg bodyweight versus placebo in the treatment and control of larval and adult stages of Ascaris suum and Oesophagostomum spp. in experimentally infected pigs. Seventy helminth free pigs were allocated on a liveweight basis to 7 groups each comprising 10 pigs (A-G). Group A served as an untreated control group. Groups B and C were used to investigate the efficacy of both formulations against adult stages of A. suum and Oesophagostomum spp., Groups D and E for efficacy against larval stages of A. suum and Groups F and G for efficacy against larval stages of Oesophagostomum spp. Pigs of groups A, B, C, D and E were infected on Day-0 with 1000 infective A. suum eggs each. Infective larvae of Oesophagostomum spp. (10,000/pig) were given on Day-0 to pigs of Groups F and G and on Day-21 to pigs of Groups A, B and C. Treatment was given to pigs of Group A (saline as placebo) on Day-7 and -28, IVM-1 to pigs of Group F on Day-7, pigs of Group D on Day-14 and pigs of Group B on Day-49. IVM-2 was given to pigs of Group G on Day-7, Group E on Day-28 and Group C on Day-49. Pigs of Groups F and G were sacrificed on Day-28, pigs of Groups A, D and E on Day-49 and pigs of Groups B and C on Day-56. Post mortem worm counts showed the following efficacies: (IVM-1) against larval A. suum 100%, against adult A. suum 94.4%, against larval Oesophagostomum spp. 52.0% and against adult Oesophagostomum spp. 83.0%. The effects of two sources of dietary fiber (DF) on behavior and heat production (HP) in group-housed growing pigs were studied. Twenty clusters of 14 barrows (50 kg) were fed one of 10 diets. Diets differed mainly in type and content of fermentable DF (fDF) and in content of digestible starch. Five diets contained solvent-extracted coconut meal (SECM) and five diets contained soybean hulls (SBH) as the main fDF source. On an as-fed basis, pigs received 3.5, 13.2, 23.0, 32.7, or 42.4 g x kg(-0.75) x d(-1) of SECM or SBH. A total of 280 crossbred growing pigs were used, divided into clusters of 14 pigs each. Pigs were group-housed and fed at 2.5 times the assumed maintenance energy requirements. All clusters were fed similar amounts of NE, ileal-digestible protein and amino acids, vitamins, and minerals. Consequently, DMI differed among diets because NE content decreased with increasing DF content. After a 32-d preliminary period, HP was measured per cluster during a 7-d experimental period in environmentally controlled respiration chambers. Behavior of the pigs was recorded using time-lapse video recordings during two different days within the experimental period. Intake of digestible starch and fDF was different (P < 0.001) among diets, whereas intake of digestible CP was similar among diets. On average, pigs spent 153 min standing, 42 min sitting, 202 min lying on their chest, and 1,043 min lying on their flanks each day. Pigs fed SECM diets spent, on average, less time (P < 0.05) lying on their chest than pigs fed SBH diets. Total time spent on physical activity (i.e., standing plus sitting, 195 min/d) was not affected by diet. Total HP and resting HP were affected by diet and were on average lower (P < 0.01) for pigs fed SECM diets than for pigs fed SBH diets. Activity-related heat production (AHP) averaged 65 kJ x kg(-0.75) x d(-1) and was not affected by diet. There was a linear relationship (P < 0.001) between fDF intake and HP, but there was no relationship between fDF intake and AHP. During different parts of the day, fDF intake also affected HP. The saving effect of physical activity on the NE values of fDF from SECM and SBH were 0.56 and 0.84 kJ/g of fDF intake, respectively. Neither of these saving effects was significantly different from zero. We conclude that fDF from SECM and SBH did not affect energy expended on physical activity by growing pigs, and that the NE value of fDF from SECM and SBH was not affected by changes in physical activity. 相似文献

Two controlled tests were performed to investigate the benzimidazole resistance of a nodular worm isolate "GIBZ" from a pig breeding farm in Germany. In Trial I, groups of five pigs, artificially infected with Oesophagostomum larvae isolated from that farm were treated with flubendazole at a single dose of 5 mg kg(-1) bodyweight (BW) or remained untreated. In Trial II, three groups of three pigs each infected with larvae after a further laboratory passage of this isolate were treated with flubendazole either at a single dose of 5 mg kg(-1) BW or at a divided dose of 1.5 mg kg(-1) BW daily for 5 consecutive days, or with fenbendazole at a single dose of 5 mg kg(-1) BW, the fourth infected group remained untreated. The respective doses of anthelmintics were mixed with a small amount of feed and administered to individual pigs in both trials. Fecal egg counts before and after treatment and post-mortem worm burdens 7 days after (last) treatment were examined to assess the anthelmintic efficacies. Only infections with Oesophagostomum dentatum were found in both trials. In Trial I, the mean worm count reduction by flubendazole was 30% as compared to the untreated controls. In Trial II, flubendazole administered at a single or divided dose reduced the mean worm burden by 0 and 85%, respectively, whereas fenbendazole was 100% effective. These results establish resistance to flubendazole in the isolate "GIBZ" of O. dentatum. The failure to reveal side resistance to fenbendazole may be explained by that the currently recommended dose rate of this compound is supra-optimal for porcine nodular worms. 相似文献

This study was carried out to obtain basic information on the transmission of Oesophagostomum dentatum and Hyostrongylus rubidus in outdoor reared pigs in Denmark. Eighteen 10 weeks old worm-free pigs were allocated into 3 groups of 6 pigs each. In May, all pigs were turned out on the same parasitologi-cally naive pasture, and after 2 weeks the pigs in groups 2 and 3 were experimentally infected with 10,800 O. dentatum and 8,700 H. rubidus infective larvae, respectively. Pigs in group 1 served as non-infected controls. All pigs were reared together on the experimental pasture for further 134 days until slaughter in October. Strongyle egg counts, differentiation of infective larvae at species level, serum pepsinogen, and herbage larval infectivity were monitored at regular intervals throughout. Both strongyle species established in the originally parasite-free pigs (group 1) and cross infections were established in group 2 and 3. The pigs were exposed to steadily increasing herbage infectivity of both species of strongyles. At the end of the experiment, geometric mean worm burdens of O. dentatum in groups 1,2 and 3 were 1202, 6136 and 1431 respectively, the burden in group 2 being significantly higher (p<0.05) than that of the 2 other groups. The geometric mean worm burdens of H. rubidus in groups 1, 2 and 3 were 4907, 3679 and 5246 respectively, showing no significant differences between groups. 相似文献
