The aim of the study was to contribute to the discussion of overdosing vitamin E in laying hens. A total of 45 laying hens, divided into 5 groups were fed diets supplemented with either 0; 100; 1000; 10,000 or 20,000 mg dl-alpha-tocopheryl acetate/kg diet over a period of 10 weeks. Concentrations of vitamins A and E were measured in plasma, various tissues and egg yolk. Furthermore egg yolk colour and some carotinoids were measured in egg yolks. None of the vitamin E doses significantly influenced performance of the hens. As expected, vitamin E concentration in plasma, all tissue samples and egg yolk was significantly increased with increasing tocopherol content in the diet. The egg yolk showed the highest vitamin E concentration, followed by liver and muscles. Feeding 1000 mg alpha-tocopheryl acetate per kg diet resulted in an increase of vitamin A concentration in the liver. Very high doses (10,000 and 20,000 mg/kg diet) significantly decreased retinol concentration in the liver and egg yolk, as well as carotinoid concentration in the egg yolk. The lower carotinoid concentration in egg yolk resulted in a decreased intensity of egg yolk colour. A prooxidative and/or competitive effect of very high doses of vitamin E with other fat soluble substances has been discussed.

1. Three experiments were carried out with light strain laying hens to evaluate the effects of relatively high doses of dietary vitamin E (125 mg/kg food) or ethoxyquin (EQ) (250 mg/kg food) on their laying performance. The control diet contained 5 and 125 mg/kg vitamin E and EQ, respectively. The experimental diets were fed either from one or 32 weeks until 88 or 89 weeks of age. 2. The two antioxidants did not affect the growth of the pullets, age at first egg, final body weight, average egg weight or relative abdominal fat pad size and liver weight at the termination of the experiments. In two out of three experiments, vitamin E and EQ did not affect egg production, food efficiency or mortality; in the third experiment vitamin E significantly (P less than 0.05) improved egg production and food efficiency after an outbreak of Newcastle disease which occurred at 34 weeks of age. EQ significantly reduced mortality during the course of this experiment, but did affect the variables of performance. In two experiments vitamin E consistently improved shell density, although a significant effect was observed in only one of the eight determinations carried out. EQ did not affect this variable. 3. The uterine muscle was more susceptible to oxidation than the drumstick meat, as evaluated by TBA values. In both tissues, vitamin E significantly and consistently decreased TBA values and restricted their increase during incubation, while EQ was less effective, particularly in the drumstick meat. 4. It is concluded that increasing vitamin E and EQ concentrations in diets of laying hens have no effect on the decrease in egg production due to aging. However, vitamin E may minimize the decline in egg production and food efficiency following the outbreaks of some diseases and slightly improve--under certain yet undefined conditions--shell density.

1. The effects of different dietary concentrations of vitamin E (alpha-tocopherol acetate) were investigated in 2 experiments on laying hens exposed to chronic heat stress at 32C. 2. In the first experiment, egg production and plasma concentrations of calcium and egg yolk precursors were measured in 24 hens before, during and after a stress period of one week and fed on diets containing 10 or 500 mg vitamin E/kg. 3. In the second, larger experiment, egg production and food intake were measured in 300 hens housed in 2 temperature-controlled rooms and fed on diets containing 10, 125 or 500 mg vitamin E/kg. Birds in room 1 were stressed from 24 to 28 weeks of age and those in room 2 from 32 to 36 weeks. 4. In experiment 1, egg production and egg weight were significantly higher (72.6 vs 51.2%, P < 0.05 and 66.6 vs 63.1 g, P < 0.005 respectively) during and after the period of stress in the group given 500 mg vitamin E/kg. Plasma concentrations of calcium, vitellogenin (zinc) and VLDL (triglyceride) were also higher in this group. 5. In experiment 2, egg production was significantly higher (65.4 vs 56.2%, P < 0.05) during and after the period of heat stress in birds in room 1 fed on the diet containing 500 mg vitamin E/kg. Egg production was also higher (49.9% vs 44.7%) on this treatment during the stress period in room 2, though the difference was not significant (P < 0.10). Egg weight and food intake were unaffected by treatment in either room. 6. It is concluded that dietary supplementation with extra vitamin E can, at least in part, alleviate the adverse effects of chronic heat stress in laying hens, perhaps by maintaining the supply of egg precursors in plasma.

The marked increase observed in malondialdehyde concentration of the blood plasma and liver of laying hens during egg shell formation in a previous experiment (Mézes and Lencsés, 1985) were explained as a possible consequence of physiologically controlled lipid peroxidation. In this experiment, 54-week-old laying hens were treated intrauterinally with indomethacin (1.0 and 2.0 mg/kg body mass) after oviposition. Two other groups of hens were treated per os with excess amounts of vitamin E (100 and 200 mg/bird). The higher dose of indomethacin significantly decreased the PGF2 alpha production and malondialdehyde content of the sell gland, as well as the malondialdehyde concentration of the plasma. The excess amount of vitamin E had the same effect. The results suggest that during egg shell formation malondialdehyde is derived from prostanoid biosynthesis rather than from a free-radical initiated lipid peroxidative process. On the other hand, the excess amount of vitamin E inhibited prostaglandin biosynthesis as well as malondialdehyde production.

1. The influence of different forms of dietary selenium (Se) on vitamin E (alpha-tocopherol) and Se contents of egg yolk and chicken meat was investigated. 2. Eggs were collected from laying hens subjected to 4 different dietary treatments after 24 weeks of age. Treatments compared the effects of inorganic Se supplementation (selenite) to those of organic (Se-enriched yeast, Se-enriched alga Chlorella) supplements. In a second experiment the effect of the above organic dietary Se supplementation on the alpha-tocopherol contents of meat from broiler chickens was evaluated. 3. Dietary Se supplementation increased the alpha-tocopherol content of egg yolks from 297 mg/kg dry matter in treatment without supplementation to 311 mg/kg when selenium was supplemented as selenite, and to 370-375 mg/kg when organic supplements were used. The Se and alpha-tocopherol contents of breast and thigh meat in broilers were significantly increased by organic dietary Se supplementation. 4. The inclusion of organic dietary Se sources in the diets of laying hens and broilers would enhance the nutritional value (vitamin E and Se contents) of products (eggs and meat) for human consumption.

In two experiments with 1,250 laying hybrids the influence of the elimination of vitamin E and antioxidant supplements to the laying hen feed was tested. With mixed feed according to official quality requirements but without vitamin E and antioxidant supplements laying hens achieved the same laying performance and individual weight as the control group. There was no influence on feed consumption, feed expenditure per 100 g egg, mortality, development of body weight, the tensile strength of the eggs and the fat quality of the body either. Vitamin E content in the yolk increases with the growing vitamin E content in the feed. The vitamin E requirement norm of laying hens for consumer egg production is achieved at a native vitamin E content of 7 mg/kg laying hen feed. The supplement of synthetic antioxidant is unnecessary.

In 5 experiments with 1,296 laying hybrids at the age of 22-73 weeks producing eggs for consumption a varied supplementation of between 0 and 20 mg vitamin E per kg of a semisynthetic ration was tested. The vitamin E content of the basal ration was less than 1 mg per kg feed. The quotas of selenium and unsaturated fatty acids in the feed were taken into consideration. Vitamin E and Se supplements did not influence, feed intake, laying, performance, feed efficiency/100 g egg development of body weight and mortality. The supplementation of 14% vitamin-E-free sunflower oil caused a significant decrease of the laying performance by 15%. There was no relation between the humoral immune reaction and vitamin E supply. The peroxide value of the abdominal fat and the activities of the creatine phosphokinase, the aspartate and alanine amino transferase in the blood serum were not directionally changed by the vitamin E, Se and oil supplements. There were no significant relations between the vitamin E content of the feed and that of the yolk and the yolk sac of the chicken. A vitamin E requirement of less than 1 mg and a requirement norm, including a safety margin, of 5 mg per kg feed was derived from the results for laying hens for egg production. A supplement of synthetic vitamin E to laying hen feed is thus not necessary.

1. The effects of different dietary concentrations of vitamin E (alpha-tocopherol acetate) were investigated on laying hens exposed to chronic heat stress at 32 degrees C from 26 to 30 weeks of age. 2. Diets containing 5 dietary concentrations of vitamin E (a control diet containing 10 mg alpha-tocopherol/kg or this diet supplemented to contain 125, 250, 375 and 500 mg alpha-tocopherol/kg) were fed to 335 birds. Half of the birds received the supplemented diets for only 4 weeks before the heat stress period (short supplementation duration, SSD) and were fed on the control diet for a further 12 weeks. The remaining birds were fed on the supplemented diets throughout the experiment, 4 weeks before, 4 weeks during and 8 weeks after the heat stress period (long supplementation duration, LSD). 3. Egg production was significantly higher during (80-6 vs 68.9%, P<0.02) and after (75.3 vs 62.7%, P<0.02) the period of stress in the LSD group fed on the diet containing 250 mg vitamin E/kg compared with the group fed on the control diet. LSD birds given 375 and 500 mg vitamin E/kg also had higher egg production than control birds during heat stress but the differences failed to reach significance (74.6 vs 68.9% and 77.1 vs 68.9% respectively). In the SSD groups, mean egg production of the birds given the diets supplemented with 125 mg vitamin E/kg or more was significantly different from the control group after heat stress (70.3 vs 62.7%, P<0.05). Egg weight and food intake were similar in all the groups. 4. Plasma and liver vitamin E concentrations were proportional to the vitamin E intake before the stress period, dropped during heat stress in the SSD groups but were maintained at concentrations closer to those observed before heat stress in the LSD groups. 5. It is concluded that a dietary supplement of 250 mg vitamin E/kg provided before, during and after heat stress is optimum for alleviating, at least in part, the adverse effects of chronic heat stress in laying hens.

In 4 experiments with a total of 1296 laying hens it was ascertained that a laying hen requires 1 mg vitamin E/kg feed for egg production if the ration contains sufficient selenium and is not rich in multiply unsaturated fatty acids. For the fertilization of the eggs little more of this vitamin is required. There were close relations, however, between the amount of vitamin E supplement to the feed and the number of hatched chickens in % of the fertilized eggs. With the help of the experiments carried out so far the exact requirement could not yet be ascertained. Based on the results of our experiments we recommend 20 mg vitamin E/kg feed for breeding hens of the laying variant. There is a close relation between the vitamin E content of the feed and that of the yolk. A supplement of 7 or 14% resp. of vitamin-free sunflower oil decreases -in dependence on the vitamin E supplement-hatching in particular without diminishing the vitamin E content of the yolk. The vitamin E content of the yolk sac, however, was drastically reduced. The influence of polyene fatty acids in hatching is smaller when sunflower oil rich in vitamin E is supplemented. Vitamin-free sunflower oil has an influence on the vitamin content of the yolk, but it diminishes drastically the vitamin content of the yolk sac and, provided that vitamin E supply is low, prevents hatching.

Effect of T-2 toxin on egg production and hatchability in laying hens.
The effect of diets containing different levels of T-2 toxin on egg production and hatchability was studied in a four-week experiment using 100 laying hens of the SSL hybrid line and 10 cocks divided into 10 groups. Another aim of the experiment was to investigate how effectively the increased dietary vitamin E content neutralized the adverse effects of T-2 toxin. The diet of the control group (C) contained no mycotoxin, while those of the experimental groups included the following levels of T-2 toxin: groups 1, 2 and 3: 1 mg/kg, groups 4, 5 and 6: 5 mg/kg; groups 7, 8 and 9: 10 mg/kg. Vitamin E was added to the diet of groups C, 1, 4 and 7 at a rate of 50 mg/kg while to that of groups 2, 5 and 8 at a rate of 100 mg/kg. To the diet of groups 3, 6 and 9 no vitamin E was added. Contamination of the diet with T-2 toxin markedly decreased egg production and impaired hatchability. The production decrease was proportional to the T-2 toxin concentration of the diet. Increased dietary vitamin E concentration exerted no influence on egg production. However, during the first week of the experiment it significantly (P < 0.01) decreased the number of infertile eggs and significantly (P < 0.01) improved the hatching percentage. Dietary vitamin E concentration was in positive correlation with the hatching percentage; this correlation was rather close (r = 0.74) in the first week of the experiment.

1. The effects of vitamin E (DL-alpha-tocopheryl acetate) and vitamin C (ascorbic acid) supplementation on performance, some immune variables and antioxidant status of White Leghorn layers (aged 44-56 weeks) exposed to tropical summer conditions were investigated. 2. Both vitamins E and C significantly improved the egg production, food conversion efficiency, antibody responses to inoculated sheep red blood cells (SRBC) and Newcastle disease virus (NDV) vaccine, lymphocyte proliferation in response to mitogen and activities of antioxidant enzymes red blood cell catalase (RBCC) and glutathione peroxidase (GSH-PX) in layers. Vitamin C increased the inflammatory response to phytohaemagglutinin-P (PHA-P) inoculation. 3. Supplemental vitamin C at 200 mg/kg diet significantly increased eggshell weight, eggshell thickness, and shell breaking strength. However, no effect of supplemental vitamin E on eggshell quality was observed. 4. Vitamins E and C, as well as interactions between them, significantly influenced the activities of glutathione reductase and lipid peroxidase. A combination of vitamin E at 125 IU/kg with vitamin C at 200 mg/kg diet had an additive effect on reducing the activity of oxidative enzyme lipid peroxidase (LP) and increasing the activity of antioxidant enzyme glutathione reductase (GSHR). 5. It is concluded that vitamin E (125 IU/kg) and vitamin C (200 mg/kg) could independently alleviate the effects of heat stress on production performance and immunological variables of layers. However, combination of both the vitamins at the above-mentioned concentrations is beneficial in eliciting higher antioxidant status in laying hens exposed to tropical summer conditions.

1. One-day-old Taiwan Native Breeder female chicks were fed on maize/soybean growing diets without supplemental vitamin E from hatch to 17 weeks of age. After 17 weeks the birds (n = 300) were randomly assigned to 5 dietary treatments and fed on maize/soybean laying diets supplemented with 0, 40, 80, 120 and 160 mg/kg of vitamin E (dl-alpha-tocopherol acetate), respectively, until 46 weeks of age. The variates measured included: age at first egg, feed consumption (FC), feed efficiency (FE), egg production (EP), egg weight (EW), egg specific gravity (ESG), eggshell strength (ESS), fertility and hatchability. 2. The addition of 120mg/kg of vitamin E lowered the first EW (P<0.05); however, there was no significant difference in the age or body weight (BW) of pullets at first egg or mortality rate to 46 weeks of age among the treatments. FE and egg mass were improved (P<0.05) in pullets fed 80 mg/kg of supplemental vitamin E. A significant increase in EP was observed after peak EP in pullets given 80 mg/kg of supplemental vitamin E. However, this favourable effect decreased as supplemental vitamin E exceeded 80 mg/kg. 3. From 17 to 46 weeks of age, egg quality (ESG and ESS) decreased with age. However, there was no correlation between age and fertility or hatchability during the experimental period, suggesting that egg quality is more age-sensitive than reproductive performance for breeder pullets. 4. Compared with the control, fertility and hatchability of all eggs set for the treatment with 80 mg/kg supplemental vitamin E increased by 7.7 and 13.4%, respectively. There was no difference in the hatchability of fertile eggs. 5. These results suggest that using supplemental vitamin E during the laying period can improve the reproductive performance of breeder pullets. The addition of 80 mg/kg of vitamin E obtained the best performance in EP, egg mass, FE (feed/egg), hatchability and fertility.

1. The effects of selenium and vitamin E supplementation of the maternal diet on their transfer to the egg yolk and tissues of the newly hatched chick and on the development of the antioxidant system in the chick liver in early postnatal life were investigated. 2. One hundred Cobb broiler breeder hens were divided into 10 equal groups and housed in pens at 25 weeks of age. Each hen received 1 of the treatment diets which included 0.2 or 0.4 mg/kg selenium, 40, 100, 200 mg/kg vitamin E or their combination. After 6 weeks, the hens were artificially inseminated once per week. From week 8, eggs were collected and placed in an incubator. After hatching, chicks from each group were reared (under standard commercial conditions) to 10 d of age. The chicks were fed on a standard starter commercial broiler diet. At the time of hatching, and at 5 and 10 days old, 4 chicks from each group were sacrificed and blood, liver and brain were collected for the subsequent biochemical analyses. 3. The inclusion of organic selenium or vitamin E in the commercial diet significantly increased their concentration in the egg and in the liver of 1-d-old chicks obtained from the eggs enriched with these substances. A positive effect of such dietary supplementation was seen at d 5 and d 10 of postnatal development. 4. There was a positive effect of selenium supplementation of the maternal diet on glutathione concentration in the liver of 1-d-old and 5-d-old chicks. A combination of a dietary selenium supplementation with high vitamin E doses further increased glutathione concentration in the liver. Dietary selenium supplementation significantly increased selenium-dependent glutathione peroxidase (Se-GSH-Px) activity in the liver of the 1-d-old and 5-d-old chicks and decreased liver susceptibility to peroxidation. 6. It is concluded that the nutritional status of the laying hen determines the efficiency of the antioxidant system throughout embryonic and early postnatal development of the offspring.

This study was an attempt to find whether the reproductive performance of cocks would be influenced by oral administration of different amounts of dietary vitamin E over a long period of time. For that purpose 60 cocks were divided into five dietary groups of 12 animals each, and supplemented with 0 (control group), 100, 1,000, 10,000 or 20,000 IU alpha-tocopherol/kg diet, respectively, over a period of 12 months. The effect on semen parameters and biochemical parameters measured in pooled semen samples and the weight of the testes were determined. The weight of testes decreased with increasing amounts of supplemented vitamin E. Volume, pH, colour, consistency and motility were not influenced by the diets, but density of ejaculate (sperm/microl), total amount of spermatozoa and morphology of sperm were significantly lowered by increasing amounts of supplemented vitamin E. The alpha-tocopherol concentration in ejaculates increased significantly in relation to the diet whereas phospholipid content and thiobarbituric acid-reactive substances (TBARS) of the semen samples decreased significantly with increasing vitamin E supplementation. The reproductive performance of cocks was negatively influenced by high doses of vitamin E although decreased TBARS indicated rising oxidative defence.

An experiment was conducted to determine if vitamin E (alpha-tocopherol-acetate) and chromium (chromium picolinate, Cr Pic) supplementation attenuate the negative effects of cold stress on egg production, egg quality, serum metabolites, and antioxidant status in Japanese quails (Corurnix coturnix japonica). One hundred and fifty laying Japanese quails (50-day-old) were divided into five groups, 30 birds per group. The laying quails kept at 6 degrees C for 12 h/d (08.00 p.m. to 08.00 a.m.) were fed either a basal diet (low temperature-basal diet, CS group) or the basal diet supplemented with either 400 microg of Cr/kg of diet (Cr group), 250 mg of alpha-tocopherol-acetate per kg of diet (Vit. E group) or 400 microg of Cr plus 250 mg of alpha-tocopherol-acetate per kg of diet (Vit. E + Cr group) while quails kept at 18 degrees C were fed a basal diet (thermo-neutral-basal diet, TN group). Performance and egg quality were significantly reduced in CS group compared with TN group. Supplemental chromium and vitamin E significantly increased live weight change, egg production, and improved feed efficiency in cold-stressed laying hens compared with the group fed the basal diet at 6 degrees C. Egg production and egg weight were also greater (P < 0.05) in each supplemental group compared with the CS group. However, a combination of vitamin E and chromium, rather than each separately, provided the greatest performance. Supplemental vitamin E and chromium also increased serum vitamin C and E but, decreased malondialdehyde (MDA) concentrations (P < 0.05); the combination of vitamin E and chromium resulted in the highest levels of serum vitamin C and E within the cold-stressed quails. Results of the present study indicate that combined antioxidant supplements increased performance, egg quality and serum antioxidant levels while lowering MDA in cold-stressed quails.

The allocation of nutrients in the metabolism of laying hens favors the egg over the body. It is unclear whether this programming also includes micronutrients and antioxidants. This was tested with five by seven 76-wk-old Heisdorf & Nelson Brown Nick layers. They were fed a basal diet low in antioxidants either unchanged (control) or supplemented with 40 IU of α-tocopherylacetate/kg of diet (vitamin E) or 2.5% chokeberry pomace, rose hip, or sage in the last 4 wk before slaughter. The traits measured were subjected to ANOVA. The additives did not affect ADFI, performance, egg quality, carcass, or meat quality. Yolk tocopherol contents were higher with vitamin E and sage compared with control and rose hip treatments. Oxidative stability was more affected in egg yolk powder than in the meat. After 12 wk of storage, yolks from vitamin E-treated hens were lowest in TBA levels (5.0 mg of malondialdehyde/kg), followed by chokeberry (8.7) and sage (8.8). Rose hip (18.1) and control (18.9) treatments were similar. In meat, TBA was slightly decreased after 9 d of storage in meat from chokeberry- and sage-supplemented hens (contrast analysis). In conclusion, antioxidant deposition to the egg seems to be part of the genetic determination of the hen. Adding antioxidants is therefore interesting for layer nutrition in general, and herbal additives allow performing this in a natural way. The programming for partitioning, however, renders the strategic feeding of antioxidants before slaughter, with the goal to increase oxidative stability of spent hen meat rather inefficient.
