Summary Studies were made of days to ear emergence under the constant temperatures of 9, 14, 19 and 25°C and 16 h photoperiod in three sets of wheat lines each possessing genotypes differing for developmental responses.Days to ear emergence in three near-isogenic lines of the wheat cultivar Triple Dirk, which differed for vernalization response, increased as the strength of the response increased. At the four temperatures Triple Dirk D (Vrn 1 vrn 2) was not significantly different from normal Triple Dirk (Vrn 1 Vrn 2) but Triple Dirk B (vrn 1 Vrn 2) was significantly (P=0.01) later than normal Triple Dirk at each temperature. This indicates that the vrn 1 allele confers stronger vernalization response than vrn 2 over the range of temperatures (9–24°C). However, Triple Dirk C (vrn 1 vrn 2) failed to head after 120 days at each temperature indicating strong interaction between vrn 1 and vrn 2 with each other (and possibly the Triple Dirk back-ground) to give a much stronger vernalization response than predictions from additivity of their individual effects.The second set comprised the four Chinese Spring/Thatcher chromosome substitution lines CS/T 3B, 6B, 7B and 5D, plus Chinese Spring and Thatcher, and were grown in the unvernalized condition. CS/T 5D was similar in days to ear emergence as Chinese Spring at all four temperatures but the other three lines were earlier to ear emergence, particularly as the temperature increased. Days to ear emergence was fastest at 14°C in all lines, except CS/T 3B, in which it decreased progressively from 9 to 24°C.The third set of Chinese Spring and Thatcher and the homoeologous group 2 chromosomes of Thatcher substituted in Chinese Spring, the group which is considered to be involved in the control of photoperiod sensitivity. The three substitution lines responded differently to temperature compared with Chinese Spring and with each other, with chromosome 2D being the least, and chromosome 2B the most, responsive to temperature.  相似文献

Summary Selection for resistance to Plasmodiophora brassicae Wor. in oriental groups of Brassica rapa L.Two hundred and sixty-five cultivars of leafy, oriental bassicas were tested for resistance to 18 collections of Plasmodiophora brassicae, the causal agent of clubroot. The tests were conducted in the greenhouse at low and high level inoculum concentrations. Eleven cultivars of B. rapa pe-tsai, five cultivars of B. rapa pak-choy and three cultivars of B. rapa choy-sum consistently segregated for resistance at the lower concentration of inoculum (1000 spores/ml). All 265 cultivars were susceptible at the higher concentration (1 000 000 spores/ml). Three cultivars were used in pedigree and recurrent selection schemes for increased resistance. After three cycles of selfing resistant individuals, significantly more resistant S3 lines were derived from each cultivar. Lines derived from two cultivars. Chinese White and PI 257236, continued to improve with each cycle of selection and demonstrated increased resistance to higher levels of inoculum (up to 1 000 000 spores/ml) New cultivars based on intercrosses of S2 resistant individuals also had significantly better resistance than the original cultivar. After two cycles of selection in the third cultivar, PI 419007, resistance did not increase and its S2 mass did not differ significantly from the original cultivar. Evidence that indicates resistance is pathotype-non-differential and offers an alternative to major gene, pathotype-differential types of resistance currently being introduced to the leafy oriental brassicas from other Brassica rapa groups.  相似文献

This paper reports the waxy proteins and apparent amylose contents of the tetraploid species Triticum dicoccum, Triticum polonicum and Triticum durum. Three waxy proteins were found in the three species; two showed the same electrophoretic mobility as the alleles Wx-A1a and Wx-B1a of the hexaploid variety ‘Chinese Spring’, while the third showed the same mobility as the allele Wx-B1e belonging to the hexaploid wheat variety ‘Bai Huo’. In apparent amylose content no significant differences between the alleles Wx-B1a and the Wx-B1e were found for each species. However, the mean amylose contents of T. durum and T. polonicum were significantly greater than that of T. dicoccum, regardless of which allele was present. Success in obtaining an interspecific hybrid between B. pekinensis and B. oleracea var. acephala was possible through the use of the Chinese cabbage as the seed parent and the use of the growth regulator, N-m-tolylphthalamic acid. The hybrids appeared vegetatively intermediate between the parents except for the inflorescence which appeared more like the kale parent. The F1 was backcrossed with ease to the Chinese cabbage parent using the hybrid either as the pollen or seed parent. No backcross was made to the kale parent. Summary Appropriate definitions of, and methods of measuring, resistance are sought using Phaseolus vulgaris, a major source of protein in Latin America, and its most important pest, Empoasca kraemeri. The concepts of general vigour and narrow-sense resistance are defined: these two components make up broad-sense resistance. The validity of these definitions, which depend on the inclusion of tolerance as a resistance mechanism, is discussed. In order to distinguish the components, 38 genotypes of P. vulgaris were grown with and without insecticidal proctection, in three replications in a split-plot design, with three planting dates. Seed yield per plant, and a visual score of damage symptoms or vigour, were measured. Three methods of estimating narrow sense resistance are considered: 1) the genotype × insecticide interaction effect in a split-plots analysis, 2) the yield of each genotype unprotected (Yu) divided by its yield protected (Yp). and 3) the deviation of each genotype from a regression of Yu on Yp. The third method overcomes some objections to the first two, and can be applied to damage and vigour scores as well as to yields. The 38 genotypes differed in narrow-sense resistance according to all three methods, and it appears that in practice Methods 2 and 3 were both valid. They also differed in general vigour. Breeders working with many pests and crops have found that low damage and high yield in the presence of the pest are effective selection criteria. The work reported here supports this view.  相似文献

应用SRAP分子标记技术对云南、广西、福建、海南等地收集的16个栽培肾茶种质进行遗传多样性分析。15对引物经PCR扩增后共检测到124个条带,其中多态性条带80个,多态性条带百分率(PPL)为66.7%;肾茶种质间的观测等位基因数(Na)为1.669 4、有效等位基因数(Ne)为1.434 0,Nei’s基因多样性指数(H)为0.252 9,Shannon信息指数(I)为0.374 7,遗传相似度介于0.566 9到0.991 7之间,说明肾茶的遗传多样性水平较低。UPGMA聚类结果将肾茶种质分为2类,与根据花冠颜色对肾茶类型进行划分的结果对应,而遗传特性与地理分布没有明显相关性。实验结果可为肾茶遗传研究、品种选育与资源保护提供科学依据。  相似文献

Yellow mosaic disease, caused by wheat yellow mosaic virus (WYMV), is one of the most serious diseases of winter wheat in Japan and China. A single major QTL for WYMV resistance in the Japanese wheat variety 'Yumechikara', designated Q.Ymym, has been mapped on a 43.6 cM linkage block between the two markers Xcfd233 and Xgwm349 on chromosome 2D. We were able to obtain two recombinants within the block, which facilitated reducing the size of the linkage block. The pseudomolecule sequence of 'Chinese Spring' (CS) indicated that the original Q.Ymym region of 43.6 cM corresponded to 68.5 Mb and the narrowed Q.Ymym region represents a size of 27.3 Mb. The sequence features of the Q.Ymym region were unique in comparison with CS sequences, which may have led to the low recombination rate within the block. The Q.Ymym haplotype block was detected in other WYMV-resistant varieties but not in the susceptible varieties used in this study. The unique sequence structure of the Q.Ymym region allowed the development of co-dominant markers for use in marker-assisted selection.  相似文献

研究和了解植物内多倍化现象的发生规律和特点对于有效调控器官分化过程, 加速有用器官形成具有重要意义。本文分别以白菜及其近缘属种为材料, 运用流式DNA分析技术, 系统检测了不同发育阶段、不同组织器官以及不同种属植物叶片的内多倍化水平。结果表明, 白菜的内多倍化特点同样具有组织、发育阶段和种属特异性。一些木质化程度较高的输导组织如幼根、胚轴、茎和叶柄比其他非输导组织的叶片、子叶、苞叶具有更高的内多倍化程度; 在给定的组织器官中内多倍化水平随该器官的发育进程而提高, 当器官成熟时, 内多倍化水平也趋于稳定; 显微观察结果表明, 不同倍性的细胞核大小差异悬殊, 呈无规律状态散布于白菜表皮组织相邻的细胞内; 最高倍性水平的细胞核, 在白菜叶片中为64C, 而在甘蓝、芥菜和拟南芥中分别为32C、16C、16C, 在同科的不同植物中显示了不同的内多倍化水平。  相似文献

SDS-PAGE analysis of seed proteins of the cultivar‘Red River 68’showed a considerably higher staining intensity of the band corresponding to HMW-GS Bx7 relative to the equivalent band in the cultivars‘Chinese Spring’and‘Cheyenne’. Southern blots of restriction enzyme fragments from DNA of these three cultivars were analyzed densitometrically to reveal that the band corresponding to the Bx7 gene of‘Red River 68’had a double staining intensity compared to the equivalent bands from the other two cultivars, which indicates that in‘Red River 68’a duplication of the Bx7 gene has occurred. Although the possibility of the gene copy being a pseudogene was not ruled out, the greater amount of protein corresponding to Bx7 in‘Red River 68’most likely is in accord with an increase in active gene number. SDSPAGE analysis of the proteins showed also that the mobility of Bx7 in‘Cheyenne’was slightly different from the mobilities of the Bx7 subunits of‘Red River 68’and‘Chinese Spring’. The same difference was observed at the gene level by PCR amplification of the genes encoding these subunits.  相似文献

Cultivated peanut, the second most economically important legume crop throughout the United States and the third most important oilseed in the world, is consistently threatened by various diseases and pests. Sclerotinia minor Jagger (S. minor), the causal agent of Sclerotinia blight, is a major threat to peanut production in the Southwestern U.S., Virginia, and North Carolina and can reduce yield by up to 50% in severely infested fields. Although host plant resistance would provide the most effective solution to managing Sclerotinia blight, limited sources of resistance to the disease are available for use in breeding programs. Peanut germplasm collections are available for exploration and identification of new sources of resistance, but traditionally the process is lengthy, requiring years of field testing before those potential sources can be identified. Molecular markers associated with phenotypic traits can speed up the screening of germplasm accessions, but until recently none were available for Sclerotinia blight resistance in peanut. This study objective of this study was to characterize the US peanut mini-core collection with regards to a recently discovered molecular marker associated with Sclerotinia blight resistance. Ninety-six accessions from the collection were available and genotyped using the SSR marker and 39 total accessions from spanish, valencia, runner market types were identified as new potential sources of resistance and targeted for further evaluation in field tests for Sclerotinia blight resistance.  相似文献

Summary An investigation was made of factors affecting the frequency of development of fertilized ovules following wheat x Hordeum bulbosum hybridizations. Seed survival, a term used here to represent the relationship between seed set and fertilization, was shown to vary between the wheat genotypes tested, but was unrelated to the crossability genes present within the wheat genotype. A high seed survival was obtained in crosses involving the variety TH3929 and the single chromosome substitution line, Chinese Spring (Hope 5B). Chinese Spring showed a poor seed survival. Some influence of the H. bulbosum genotype on seed survival was detected. The two environmental factors studied, namely the post-pollination application of gibberellic acid and the ambient temperature, were both shown to affect seed survival. The application of gibberellic acid immediately after pollination or 1 and 2 days after pollination improved seed survival, although the extent of the response was dependent upon the number and timing of the application(s). A genotype-dependent response to the ambient temperature was observed, TH3929 showing a slight but insignificant reduction and Chinese Spring a significant improvement in seed survival with an increase in temperature from 20°C to 26°C. The physiological processes which may be influenced by GA and the ambient temperature and may thereby affect seed survival are discussed.  相似文献

Most genes for resistance to barley leaf scald map either to the Rrs1 locus on the long arm of chromosome 3H, or the Rrs2 locus on the short arm of chromosome 7H. Other loci containing scald resistance genes have previously been identified using lines derived from wild barley, Hordeum vulgare ssp. spontaneum. A single dominant gene conditioning resistance to scald was identified in a third backcross (BC3F3) line derived from an Israeli accession of wild barley. The resistance gene is linked to three microsatellite markers that map to the long arm of chromosome 7H; the closest of these loci, HVM49, maps 11.5 cM from the resistance gene. As no other scald resistance genes have been mapped to this chromosome arm, it is considered to be a novel scald resistance locus. As the Acp2 isozyme locus is linked to this scald resistance locus, at 17.7 cM, Acp2 is assigned to chromosome 7H. Molecular markers linked to the novel scald resistance gene, designated Rrs15, can be used in breeding for scald resistance.  相似文献

Summary An efficient transformation system for Chinese cabbage cotyledon explants was developed using Agrobacterium tumefaciens strains LBA4404 harbouring the plasmid pMOG 411 and the plasmid pBinΩSCK respectively. Various factors affecting the transformation efficiency and subsequent regeneration were identified. The age of seedlings, growth conditions and status of Agrobacterium suspension, preculture of explants, cocultivation time, ratio between Agrobacterium and explants, acetosyringone and concentration of kanamycin had a significant influence on transformation frequency and plant regeneration. The presences of the antibacterial peptide gene and the cowpea trypsin inhibitor gene in those selected shoots in kanamycin medium were confirmed by PCR, Southern blotting and Northern blotting, The frequency of regeneration from cotyledons was analyzed in eight parental lines of Chinese cabbage and five lines were suitable for obtaining transformed plants, among which Qingmaye C and Qingmaye D were more responsive than other lines.  相似文献

The Chinese wheat landrace, Xinjiang rice wheat (T. petropavlovskyi Udacz. et Migusch., 2n = 42), known as ‘Daosuimai’ or rice-head wheat is characterized by long glumes, and was found in the agricultural areas in the west part of Talimu basin, Xinjiang, China in 1948. The gene for long glume from T. petropavlovskyi was introduced into a line of spring durum wheat, LD222. The gene for long glume is located approximately46.8 cm from the cn-A1 locus, which controls the chlorinatrait. Significant deviation from a 3:1 in the F2 of LDN7D(7A)/ANW5C confirmed that the long glume of T. petropavlovskyi can be controlled by a gene located on chromosome 7A. The gene locates approximately 12.4 ± 0.5 cM from the centromere on the long arm of 7A. It is considered that the gene for long glume from T. petropavlovskyi is an allele on the P 1 locus, and it should be designated as P 1a. It is suggested that T. petropavlovskyi originated from either the natural hybrid between T. aestivum that has an awn-like appendage on the glume and T. polonicum or a natural point mutation of T. aestivum. A genome specific DNA sequence that detects Secale africanum chromatin incorporated into wheat was developed in this study. Random amplified polymorphic DNA (RAPD) analysis was used to search for genome specific DNA sequences of S. africanum in lines, R111, “mianyang11” (MY11) and wheat-rye 1RS/1BL translocations R25 and R57. A high copy rye-specific DNA segment pSaD15940 of the S. africanum genome was obtained. The sequence of pSaD15 did not show any significant homology to other reported sequences in databases and it is therefore a new repetitive sequence of Secale. PCR primers were designed for pSaD15940, which amplify a clear 887 bp fragment in S. africanum but not in any wheat. The primers also amplified an 887 bp fragment in other accessions of rye, Chinese Spring-Imperial rye chromosome additions and a diverse range of material carrying different rye chromosomes or chromosomal segments. In situ hybridization showed that probe pSaD15940 was specifically hybridized throughout all rye chromosomes arms except for the terminal regions. The advantage of the rye-specific probe developed herein compared to those of previous reports is that it has been shown to be widely applicable to other Secale species. The probe will be useful as a molecular marker for the introgression of S. africanum and other rye chromosome segments into the wheat genome.  相似文献

Summary In vitro selection of salt tolerant plants of Brassica juncea L. (Indian mustard) cv. Prakash has been accomplished by screening highly morphogenic cotyledon explant cultures on high NaCl media. Out of a total of 2,620 cotyledons cultured on high salt medium, 3 survived, showed sustained growth and regenerated shoots. They were multiplied by axillary bud culture on NaCl free medium. The salt-selected shoots retained salt tolerance following 3 month of growth and multiplication on control medium. While two of these somaclones flowered and set seeds, third one grew slowly, had abnormal leaf morphology and was sterile. The seed of the two fertile plants were sown in the field to raise R1 segregating generation. Data were recorded for field, other agronomic components and oil content. The somaclonal lines, both selected salt-tolerant and non-selected, showed tremendous amount of variation for all the characters studied. One of the two tolerant somaclones invariably showed reduced height, longer reproductive phase and higher 1000 seed weight. Based on the agronomic performance of R1 plants of these somaclones, some plants were selected and their progeny were evaluated for agronomic performance under standard field conditions and salt-tolerance in the greenhouse using sand pot culture method. Most of the lines bred true for their specific characteristics. In the greenhouse, selected salt-tolerant somaclones (SR-2 and SR-3) performed better for plant growth, yield and other agronomic traits at higher salt treatments, indicating thereby that salt-tolerance trait selected in vitro was expressed in the whole plants and is genetically stable and transmitted onto the progeny. The two tolerant lines, however, differed in their salt-tolerance during vegetative and reproductive phases as indicated by their salt-tolerance and stress susceptibility indices. The mechanism of salt-tolerance is not clear and needs to be further investigated.  相似文献

The Chinese native rice cultivar ‘Maowangu’ expresses a high level of resistance to many races of rice blast (Pyricularia grisea) collected from North China and Japan. ‘Maowangu’ was crossed with 10 Japanese differential cultivars and the susceptible Chinese cultivar ‘Lijiangxintuanheigu’ (LTH). Allelism tests were conducted in the F2 populations with rice blast races. The resistance of ‘Maowangu’ was governed by two dominant genes which were non-allelic to the resistance genes at seven loci: Pi-a, Pi-i, Pi-k, Pi-z, Pi-ta, Pi-b, and Pi-t. To identify the two resistance genes, two F3 lines of ‘Shin 2/Maowangu’ segregating 3R:1S were selected for linkage tests in 1994. One was linked to marker genes C and Amp-3 on chromosome 6 with recombination frequencies of 35.8 ± 6.4% and 42.1 ± 6.2%, respectively, and the other to Amp-1 on chromosome 2 with a recombination frequency of 37.6 ± 6.0%. To confirm these results, two F3 lines of ‘LTH/Maowangu’ were selected for linkage tests in 1995. The one was linked to Amp-3, and other was linked to Amp-1, with recombination frequencies of 36.9 ± 3.1% and 34.3 ± 3.2%, respectively. The two genes on chromosomes 6 and 2 were designated Pi13(t) and Pi14(t), respectively.  相似文献

法尼基焦磷酸(farnesyl diphosphate, FPP)是植物萜类化合物合成途径的重要前体之一,经不同的酶催化可形成各类萜类化合物,Vetispiradiene合酶可催化FPP形成Solavetivone和Lubimin的前体Vetispiradiene。本研究利用白木香转录组数据,首次克隆了编码白木香Vetispiradiene合酶的基因(命名为As VS, Genbank登录号为MH378283),AsVS基因序列全长为1 632 bp,编码543个氨基酸,该氨基酸序列与马来沉香倍半萜合酶聚类于同一分支,具有较近的亲缘关系,该蛋白植物倍半萜的保守性结构域DDXXD,NST/DTE和R(R)X8W,相对分子量为62.73 kD,理论等电点为5.51,包含40个磷酸化位点;二级结构以α螺旋和无规则卷曲为主;不含跨膜结构域。RT-qPCR分析结果表明AsVS基因在结香剂处理过程中于白木香茎干样品中持续上调表达,其第6天及第9天的表达量分别为对照处理的2.36和5.02倍。此结果为进一步研究As VS基因在白木香萜类化合物的合成及沉香致香成分富集中的功能提供了理论基础。  相似文献

Quantitative trait loci (QTL) for three traits related to awning (awn length at the base, the middle and the top of the ear) in wheat were mapped in a doubled‐haploid line (DH) population derived from the cross between the cultivars ‘Courtot’ (awned) and ‘Chinese Spring’ (awnless) and grown in Clermont‐Ferrand, France, under natural field conditions. A molecular marker linkage map of this cross that was previously constructed based on 187 DH lines and 550 markers was used for the QTL mapping. The genome was well covered (more than 95%) and a set of anchor loci regularly spaced (one marker every 20.8 cM) was chosen for marker regression analysis. For each trait, only two consistent QTL were identified with individual effects ranging from 8.5 to 45.9% of the total phenotypic variation. These two QTL cosegregated with the genes Hd on chromosome 4A and B2 on chromosome 6B, which are known to inhibit awning. The results were confirmed using ‘Chinese Spring’ deletion lines of these two chromosomes, which have awned spikes, while ‘Chinese Spring’ is usually awnless. No quantitative trait locus was detected on chromosome 5A where the B1 awn‐inhibitor gene is located, suggesting that both ‘Courtot’ and ‘Chinese Spring’ have the same allelic constitution at this locus. The occurrence of awned speltoid spikes on the deletion lines of this chromosome suggests that ‘Chinese Spring’ and ‘Courtot’ have the dominant B1 allele, indicating that B1 alone has insufficient effect to induce complete awn inhibition.  相似文献
