The ultrastructure of the matrix of the sheep central incisor periodontium was quantitatively analysed. Ewes with healthy mouths were selected. The distribution of collagen fibril diameters was bimodal in the lower dental pad with a population standard deviation significantly larger than the unimodal distribution of fibril diameters found in the periodontal ligament. The lower dental pad connective tissue also contained significantly more oxytalan fibres, some of which resembled mature elastin. These results are consistent with the view that the sheep central incisor is supported primarily by tension within the fibres of the lower dental pad.  相似文献

ABSTRACT: Leishmania is inoculated, by the bite of an infected sandfly, into the skin of the host, where the promastigotes are phagocyted by dermal macrophages. The dermal region comprises cells and abundant extracellular matrix. Studies show that matrix metalloproteinases play an important role in host defense responses against pathogens in mammals and that their activities lead to the production of antimicrobial peptides. The aim of this study is to evaluate the changes in the distribution of fibronectin and laminin as well as in the elastic system fibres during the course of infection caused by Leishmania amazonensis in mice with distinct genetic backgrounds of susceptibility to this parasite. The results showed that BALB/c presented an enhancement of fibronectin during the course of infection when compared to their control group while the infected or non-infected C3H.He showed a decrease of this protein at end of the experiment. Laminin, on the other hand, remained unaltered in both strains. Also in both BALB/c and C3H.He mice the elastic and elaunin fibres remained unchanged while the oxytalan fibres decreased along the experiment. Ninety days after the infection C3H.He mice had recovered their capacity to produce oxytalan fibres.  相似文献

The extensor digitorum longus (EDL) and soleus (SOL) muscle fibres from albino rats submitted to experimental chronic alcoholism were evaluated in accordance with their metabolic and morphometric profiles. Twenty-seven male animals aged 4 months and weighing approximately 400 g were used. The animals were divided into three groups: control, isocaloric and alcoholic and sacrifices were carried out after 5, 10 and 15 months. The muscles were dissected, removed, cross-sectioned in a cryostat and submitted to the NADH (nicotinamide adenine dinucleotide) reaction. The SO (slow-twitch-oxidative), FG (fast-twitch-glycolytic) and FOG (fast-twitch-oxidative-glycolytic) muscle fibre types exhibited a polygonal, triangular or rounded shape and did not present noteworthy modifications in either muscles during the study. The cross-sectional areas of the fibres from the studied muscles did not present significant differences during the observations. Fibre area behaved similarly in the alcoholic animals up to the 10th month, i.e. it was decreased, as also observed in the other groups. At 15 months, however, all fibres were increased, with a predominance of FG fibres in the SOL muscle. Changes in fibre population were observed mainly in the SOL muscle of alcoholic animals: SO fibres were initially increased in number but decreased after the 10th month, and the opposite was observed for the population of FG fibres. FOG fibres increased linearly in number throughout the experiment. The statistical analysis showed nevertheless that the fibre population and cross-sectional area changes were not significant. In the alcoholic animals quantitative variations of muscle fibres were more evident in the SOL muscle, suggesting that the SOL muscle is more sensitive to the toxic action of ethanol. The results concerning the increased fibre diameter in alcoholic animals would be associated with muscle oedema induced directly or indirectly by the ethanol.  相似文献

Cruciate ligaments (CLs) are primary stabilisers of the knee joint and canine cranial cruciate ligament disease (CCLD) and rupture is a common injury. Elastin fibres, composed of an elastin core and fibrillin containing microfibrils, are traditionally considered minor components of the ligament extracellular matrix (ECM). However, their content and distribution in CLs is unknown. The purposes of this study were to determine the elastin content of canine CLs and to ascertain its relationship to other biochemical components and histological architecture.Macroscopically normal CLs were harvested from Greyhounds (n = 11), a breed with a low risk of CCLD. Elastin, collagen and sulfated glycosaminoglycan content were measured and histological scoring systems were developed to quantify ECM changes using a modified Vasseur score (mVS) and oxytalan fibre (bundles of microfibrils) staining. Elastin contents were 9.86 ± 3.97% dry weight in the cranial CL and 10.79 ± 4.37% in the caudal CL, respectively, and did not alter with advancing histological degeneration. All CLs demonstrated mild degenerative changes, with an average mVS score of 11.9 ± 3.3 (maximum 24). Increasing degeneration of the ligament ECM showed a positive correlation (r = 0.690, P < 0.001) with increased oxytalan fibre staining within the ECM.Elastin is an abundant protein in CLs forming a greater proportion of the ligament ECM than previously reported. The appearance of oxytalan fibres in degenerative CL ECM may reflect an adaptive or reparative response to normal or increased loads. This finding is important for future therapeutic or ligament replacement strategies associated with cranial CL injury.  相似文献

This study was undertaken to investigate insulin sensitivity and muscle characteristics in calves at different levels of physical activity. Ten male calves of the Swedish Holstein breed were allocated to either an exercise or a non-exercise group. The exercise group performed light work on a treadmill, 5-15 min at 10-12 occasions during 4-5 weeks. The treadmill speed varied from 0.6 to 1.5 m/s. At the beginning and end of the experimental period, the hyperinsulinemic euglycemic clamp technique was used to assess insulin sensitivity and gluteus medius muscle biopsies specimens were obtained for histochemical and biochemical analyses. There were no significant differences in insulin sensitivity, muscle fibre types, enzyme activities or glycogen content between the groups before or after the experimental period. The areas of type I, IIA, IIB fibres and mean muscle fibre area increased significantly in both groups during the experimental period. Muscle fibre characteristics and insulin sensitivity varied among the calves. Insulin sensitivity correlated positively to the percentage of type I fibres at the start of the experiment, and negatively to the mean muscle fibre area at the end of the experimental period. The results indicate that insulin sensitivity in young calves is associated with the change seen in muscle fibre areas with growth, but not by a short period of light physical activity.  相似文献

Quantitative measurements were made on cross-sectional preparations of the distal part of the recurrent laryngeal nerve (RLN) from nine young mixed-breed horses to establish reference values for the total number of myelinated fibres, mean fibre diameter and percentage of thickest fibres (over 9·5 gm) and to delineate diameter distribution curves. The total number of myelinated fibres, mean fibre diameter and percentage of thickest fibres for the left RLN were significantly lower than those of the right RLN (P<0–005). The distribution of fibres was unimodal. The fibre diameter ranged from 1 μm to 17 μm. Approximately, 95 per cent of fibres had a diameter larger than 5 μm.  相似文献

In order to obtain broader insights into the equine musculoskeletal system, we studied the fibre type composition of 2 locomotory muscles in biopsies from Dutch Warmblood foals taken at 3 different ages in the first year postpartum. The muscle fibre types were determined histochemically as well as immunohistochemically. ATPase-characterised IIB fibres appear to express either IId or type lIa plus IId myosin heavy chain (MHC). A high percentage of fibres classified as IIA with ATPase expressed both fast types of MHC. The type I classification by the 2 methods matched almost completely. There was an increase with age of fibres expressing I and IIa MHC in the gluteus medius. At the same time, there was a decrease of fibres expressing IId MHC and fibres co-expressing MHC IIa and IId. MHC expression of the semitendinosus muscle did not change over time at first, but from age 22-48 weeks there was a decrease in the percentage of type IId fibres. In general, the gluteus medius contained more type I fibres but fewer type IId fibres compared to the semitendinosus. At most ages the fibre type compositions of both muscles correlated with one another. To examine the effect of exercise, one-third of the foals were given box rest, one-third received training and one-third kept at pasture during the first 22 weeks of life. The 3 exercise groups differed in their fibre type composition; however, these differences could not be attributed to the effect of exercise.  相似文献

Fibre-type differentiation of lateral musculature has been studied in gilthead sea bream Sparus aurata (L.) and sea bass Dicentrarchus labrax (L.) during post-larval development using ultrastructural, histochemical and morphometric techniques. The study showed three muscle layers: red, intermediate (or pink) and white. Initially, most of the red muscle showed low myosin ATPase (m-ATPase) activity fibres, whereas near the transverse septum some small high m-ATPase activity fibres appeared and later acquired a rosette aspect. Afterwards, during adult growth the red muscle showed a histochemical mosaic appearance. The pink muscle in sea bass was observed at the beginning of juvenile development by the oxidative technique (NADH-RT reaction) whereas in gilthead sea bream it was also observed at the end of larval development. The pink layer consists of high m-ATPase activity fibres. However, along the muscle development other low and moderate m-ATPase activity fibres were observed close to the red and white muscles, respectively. The white muscle of juvenile fish showed a histochemical mosaic appearance near the pink muscle. In adult specimens the mosaic white muscle spread out occupying the whole of the myotome. Morphometric analysis shows a significant increase in mean fibre diameter during post-larval development, as shown by the Student's t-test (hypertrophic growth). Skewness and kurtosis values of fibre diameters point to the generation of a new fibres from the myosatellite cells (hyperplastic growth).  相似文献

Venous blood samples and middle gluteal muscle biopsies were obtained from 21 horses taking part in 100 km or 50 km endurance rides. Group A consisted of seven horses competing over 100 km (four horses completed the ride), Group B consisted of the six horses that were among the 10 best over 50 km while the other eight horses of Group C completed 50 km at a slower speed. Blood lactate, glycerol and creatine kinase increased in all groups while aspartate aminotransferase levels were higher only in Group A. No change was found in blood glucose in Groups B and C while horses in Group A had lower levels after the ride. Neither fibre composition, fibre areas nor enzyme activities differed between the groups. Intramuscular glycogen content was similar before the ride in all groups. After the ride glycogen had decreased only half as much in Group C as compared to Groups A and B. Group C had a higher intramuscular triglyceride content at rest than Group B. The greatest decrease in triglyceride content after the ride was found in Group C. In Groups A and B many Type I, IIA and IIB fibres were glycogen depleted after the ride while in Group C mainly Type I and some Type IIA fibres were depleted. The results of this study show that intramuscular carbohydrate and lipid stores are both important fuels during endurance rides.  相似文献

Two experiments were conducted to determine the influence of dietary fibres on small intestine histomorphology and lipid metabolism in broilers from 1 to 21 day of age. In experiment 1, diets containing insoluble [cellulose (CEL); 2% and 4%] or soluble [carboxymethyl cellulose (CMC); 2% and 4%] fibre were fed to broilers from day 1 to 21 post‐hatch and ileal tissue was collected at day 21 of age for histological evaluation. In experiment 2, broilers diet was supplemented with 0%, 1% or 2% insoluble fibre (Arbocel) during day 7 to 21 post‐hatch and plasma and liver lipid metabolism were evaluated at day 21. In experiment 1, inclusion of CMC reduced body weight gain (BWG) and feed intake (FI) and increased feed conversion ratio (FCR) compared with others. Intestinal histomorphology was unaffected by CEL, but CMC led to an increase in crypt depth (CD) and serosa thickness and a decrease in villus height (VH), villus width (VW), VH:CD ratio and villus surface area (VSA), rather than control and CEL groups. Treatment did not affect goblet cell type. Moreover, the CMC‐fed birds had greater total goblet cell count (GCC) as compared with others. In experiment 2, fibre inclusion was associated with increases in BWG from 7 to 14 day of age and an improvement in FCR, whereas FI was not influenced by treatments. Inclusion of fibre in the diet decreased the weight of the abdominal fat and cholesterol concentrations of liver and plasma. No significant effects on fatty acid composition of liver lipid were observed by fibre supplementation. These findings suggest dietary fibre affects performance, intestinal histomorphology and lipid metabolism in young chicks, which may directly affect poultry feeding strategies.  相似文献

The middle gluteal muscle of five, two-year-old untrained trotters was investigated by repeated needle biopsy sampling over a training period of six months. A second group of five, three-year-old untrained horses was included to examine the effect of growth. After the training period increases were found in the relative distribution of slow twitch (ST) fibres from 18 per cent to 25 per cent and fast twitch (FTa) fibres from 36 per cent to 45 per cent, and a decrease in FTb fibres from 46 per cent to 30 per cent. A proportionally equal reduction (approximately 18 per cent) in the cross sectional area of all fibre types was observed after the first two months of training succeeded by an increase to approximately pretraining levels at the end of the period. The number of capillaries per fibre was enhanced from 1.7 to 2.4. Proliferation of capillaries occurred around fibres of all types. The metabolic adaptations showed increases in the activities of 3-hydroxy-acyl-CoA dehydrogenase (HAD) (50 per cent) and citrate synthase (31 per cent). Growth had no effect on the relative fibre type distribution nor the capillary per fibre ratio, but as the mean fibre area increased 36 per cent (primarily because of increases in FT fibres) the number of capillaries/mm2 was lower in the older untrained horses (350 capillaries/mm2) compared with the younger untrained ones (460 capillaries/mm2). Increase with growth was found in the activity of phosphorylase and HAD and a decrease was seen in the activity of hexokinase. It is concluded that the training programme exclusively induced alterations which improved the aerobic capacity of the muscle.  相似文献

Fibre type composition, activities of enzymes such as citrate synthase (CS), 3-hydroxyacyl coenzyme A dehydrogenase (HAD), lactate dehydrogenase (LDH), as well as glycogen, lactate and pH levels were analysed in muscle biopsies (m. gluteus medius) obtained after bullfighting from 10 young and 10 old bulls. No changes were seen in fibre type composition between groups, but the older bulls had higher HAD and LDH activities. Low glycogen concentrations and low pH values were found in both groups, but the lactate concentration after bullfighting was higher in the older group of bulls. The histochemical stain for glycogen revealed that type IIB fibres in both young and old bulls contained more glycogen than seen in type IIA and type I fibres. These results show that young and old bulls have similar muscle fibre type composition, but the metabolic capacity differs, with a higher glycolytic capacity and lactate production in older bulls. Furthermore, it seems that the physical and emotional stress in connection with a bullfight causes a marked depletion of glycogen, especially of type I and IIA fibres.  相似文献

The elastic system fibres of the female urethra were investigated in seven dogs of different breeds. The fibres were stained and differentiated with orcein, Verhoeff's iron hematoxylin, and pararosanilin-based aldehyde fuchsin, with and without previous oxidation. Orcein and aldehyde fuchsin revealed some subepithelial longitudinally orientated delicate fibres (elaunin fibres) and numerous coarse longitudinal fibres (elastic fibres) in the deeper subepithelial connective tissue containing the vascular plexus, as well as a network of fibres of different calibers in the periurethral connective tissue. Elastic system fibres were also found in association with the sinusoids of the vascular plexus and the urethral smooth musculature. Verhoeff's iron hematoxylin only reacted distinctly with coarse (elastic) fibres. When applied following oxidation, aldehyde fuchsin disclosed an extensive meshwork of additional delicate fibres (oxytalan fibres) in the subepithelial connective tissue adjacent to the basement membrane. Oxytalan fibres were also discernible in the sinusoidal adventitia of the vascular plexus, as well as between smooth and striated muscle fibres. Due to the predominantly longitudinal orientation of the elastic system fibres and the low urethral resistance to manual expression of urine from the bladder post mortally, when the elasticity of the urethra is still intact and the activity of other continence factors can be excluded, the elastic tissue is not judged to be a major contributory factor to urinary continence.  相似文献

1. The incidence of microscopically detectable degenerative characteristics in 5 skeletal muscles (m. pectoralis thoracicus, m. supracoracoideus, m. biceps femoris, m. semitendinosus, m. femorotibialis medius) of turkeys was investigated. 2. Samples were obtained from 30 Large White turkey males 14, 16 and 18 weeks old. Hyaline degeneration, infiltration of mononuclear cells and necrotic fibres were observed. 3. Individual fibres varied greatly in size and muscle fibre nuclei were often shrunken and pyknotic. 4. Weak and/or uniform reaction for Ca++-ATPase and SDH in all types of muscle fibres and loss of alkaline phosphatase activity in cell membranes were noted. A positive reaction for acid phosphatase occurred in regions of perivascular infiltration and in necrotic muscle fibres. The majority of muscle fibres possessed high activity for phosphorylase a and b. 5. Based on the use of fluorescein alpha-bungarotoxin conjugate, motor end-plates appeared to be morphologically intact. Direct immunofluorescence with anti-chicken IgG showed positive reaction in muscle fibres undergoing necrosis and in the involved connective tissue. 6. Degenerative changes varied with age and were most marked in the oldest birds. 7. Because gross degenerative symptoms were absent from both the birds and the meat from them, the condition appears to be either different from or a precursor to the degenerative myopathy characterised by other authors.  相似文献

A technique for the evaluation of neuromuscular transmission using single fibre electromyography was developed for the peroneus longus muscle of dogs. Individual muscle fibre action potentials were recorded in 10 dogs, with a specialised needle electrode, during repetitive axon-al stimulation at the motor point of the right peroneus longus muscle. Variability in latency from stimulus artefact to action potential was evaluated for 50 to 100 consecutive discharges for several fibres in the muscle of each dog. Latency variability (jitter) was expressed as the mean of consecutive differences (MCD) in latency for each muscle fibre. A mean of MCDS was calculated for the muscle of each animal. The upper limits of normal for the peroneus longus suggested by this study are 30 us (for individual muscle fibres) and 19 μs (for mean of group of fibres).  相似文献

In this study, the effect of testosterone on gastrocnemius muscle fibres in growing and adult rats (male and female) was examined using histochemical, morphometric and ultrastructural techniques. After physiological saline (PS), olive oil (OvO) or olive oil + testosterone (OvOT) injections on 72 rats (growing and mature, 36 male and 36 female), the sample tissues of fibre types of the gastrocnemius muscle taken were examined by histochemical [alkaline adenosine triphosphatase (alk-ATPase), acid ATPase (ac-ATPase)], morphometric and ultrastructural techniques. In PS-injected control groups, the gastrocnemius muscle of both sexes contained all the fibre types studied [slow-oxidative muscle fibres (type I), fast-oxidative glycolytic muscle fibres (type IIA) and fast-glycolytic muscle fibres (type IIB)]. The type I fibres had the smallest diameter, type IIA had a medium diameter and type IIB fibres had the largest diameter. In OvO-injected groups, it was observed that the OvO had little effect on the gastrocnemius muscles of either sex, although there was significant enlargement of type IIB fibres. After the injection of OvOT, hypertrophy of muscle fibres was determined by morphometric study. The biggest increase in diameter was on type I fibres. In addition, degenerations on some mitochondria, accumulation of lipid droplets on type I and type II fibres, an increase in glycogen particles, bifurcation of myofibrils, an increase in the number and diameter of units resembling T tubules and an increase in ribosomal content were also observed in the same group by transmission electron microscope. Consequently, it was determined that testosterone can induce protein synthesis in gastrocnemius muscle fibres, and induces changes in shape and size, and also can change the appearance and the number of fibres.  相似文献

REASONS FOR PERFORMING STUDY: Most racehorses are trained regularly from about age 18 months; therefore, little information is available on the effect of training in Thoroughbred foals. HYPOTHESIS: Well-controlled exercise could improve muscle potential ability for endurance running. METHODS: Thoroughbred foals at age 2 months were separated into control and training (treadmill exercise) groups and samples obtained from the middle gluteal muscle at 2 and 12 months post partum. Muscle fibre compositions were determined by histochemical and electrophoretical techniques and succinic dehydrogenase (SDH) activity was analysed in each fibre type. RESULTS: All fibre types were hypertrophied with growth and type I and IIA fibres were significantly larger in the training than the control group at age 12 months. A significant increase of SDH activity was found in type IIX muscle fibres in the training group. CONCLUSIONS: Training in young Thoroughbred horses can facilitate muscle fibre hypertrophy and increase the oxidative capacity of type IIX fibres, which could potentially enhance stamina at high speeds. POTENTIAL RELEVANCE: To apply this result to practical training, further studies are needed to determine more effective and safe intensities of controlled exercise.  相似文献

1. Muscle fibre composition, capillary-fibre ratio and mitochondrial density of the Iliotibialis lateralis muscle were monitored during growth from the age of 8 weeks to sexual maturity (24 weeks) in sedentary birds and in birds subjected to regular treadmill exercise. 2. The Iliotibialis lateralis muscle consists almost exclusively of type IIA and IIB fibres with a small percentage of "unclassified" fibres. Over the 16-week period of growth there was no change in fibre composition but a marked fibre hypertrophy linked with a fall in the capillary-fibre ratio. Mitochondrial volume density increased with age but this was not significant. 3. Exercise significantly increased the IIA:IIB fibres ratio from 0.77 to 1.1 but had no significant effect on any other measured characteristics. 4. It is concluded that exercise at the intensity and duration employed (10-15 min daily, ca. 70-80% maximum oxygen consumption) has little effect on the growth and development of avian skeletal muscle although there was evidence of an increased oxidative capacity of the muscle.  相似文献

This study uses non-invasive evoked surface electromyography (SEMG) to investigate postnatal muscle development in pigs, and to assess any correlation between recorded signal parameters and muscle fibre types in two different skeletal muscles. Four litters (n = 43) of Large White × Landrace pigs were used. Evoked SEMG measurements were taken on days 2, 5, 14, 26, 60 and 151 post partum from m. Longissimus dorsi (LD) and on days 14, 26, 60 and 151 post partum from m. Biceps femoris (BF). A third of each litter was slaughtered at days 27, 61 and 153 post partum. Biopsy samples for LD and BF were taken to categorize muscle fibre types. For LD there was a significant increase in compound muscle action potential (CMAP) parameters from day 2 to day 5 post partum, whilst for BF significant increases occurred from days 14 to 26 post partum. Fibre type development in both muscles showed a significant decrease in type IIA fibre number (P < 0.001) and an increase in type IIB fibre number (P < 0.001). Analysis of CMAP parameters in relation to fibre type percentages showed significant positive correlations between percentage of type I fibres in BF and CMAP Area (r = 0.71; P = 0.05), SlopeL (r = 0.79; P = 0.02) and Corr Peak (r = 0.78; P = 0.02) and a negative correlation with SlopeT (r = 0.89; P = 0.003) at day 151. A greater CMAP Area and Corr Peak as well as steeper leading and trailing slopes were on the other hand correlated to fewer type IIB fibres.

It is concluded that 1) changes in the evoked CMAP are muscle-specific during early postnatal development in the pig, and 2) in spite of a small sample size, the correlations between CMAP signal parameters and fibre type percentages warrant further investigation.  相似文献

In eight specimens of Atlantic sea bass of commercial size (congruent with 350 g) muscle cellularity was studied at two selected sampling levels of the trunk axial musculature: caudal (anal opening) and cranial (fourth radius of the dorsal fin). The following parameters were quantified at both sampling levels: white muscle cross-sectional area, white muscle fibre diameter (900-1200 fibres), muscle fibre number and muscle fibre density. Results showed a higher total cross-sectional area at cranial than at caudal level (P < 0.05), what is related with their different gross morphology. However, the white muscle fibre size distribution, as well as the muscle fibre number and density did not show significant differences between them. This study contributes to typify muscle fibre sampling in sea bass of commercial size what is of great interest for morphometric studies where white muscle cellularity is commonly correlated with textural or organoleptic parameters.  相似文献
