Field trials were conducted to establish the effect of the use of an inactivated oil emulsion vaccine against Infectious Bursal Disease (IBD OEV) in broiler breeder hens, and its effects on their progeny. The performance of 18 broiler flocks, which were the progeny of the IBD OEV vaccinated breeder hens, but which were not vaccinated with a live vaccine against IBD, was equal to that of broiler flocks which were vaccinated with a live IBD vaccine and originated from parent stock that had been vaccinated only against IBD with a live vaccine. In none of the 18 flocks, progeny of IBD OEV vaccinated parents, was IBD diagnosed. In a second stage, 15 broiler flocks were included in the trial: these were derived partly from IBD OEV vaccinated parents, and partly from parents that received only live IBD vaccine at 8-10 days of age. No cases of IBD occurred and all flocks were positive for IBD precipitins at slaughter age. Vaccination with a live vaccine against IBD at the age of 8-10 days had no influence on NCD antibody development after a NCD vaccination at 7 days. No immunosuppressive effect from this type of live live IBD vaccine could be determined under field conditions.  相似文献

A comparison was made between four inactivated infectious bursal disease (IBD) oil emulsion vaccines. Vaccine A, which was prepared from antigen derived from bursae of Fabricius, stimulated higher levels of IBD gel diffusion titres than vaccines B, C and D, which were prepared from antigens derived from embryo fluids. The progeny of parents vaccinated with vaccine A resisted IBD challenge for eight days longer than the progeny of any of the other vaccinated parents.  相似文献

Serum samples and bodyweights were taken at regular intervals for six to seven months from piglets, born to foot-and-mouth disease (FMD) vaccinated or unvaccinated sows, which had been vaccinated at one, two, four or eight weeks old. Young pigs, devoid of maternally derived antibodies (MDA), were capable of responding to FMD vaccination at one week old, with no deleterious effects on their growth rate. However, their immunity to experimental infection at six to seven months old was poor (33.3 per cent). Vaccination of four or eight-week-old piglets, born to unvaccinated sows, protected 87.5 per cent from a similar challenge. In piglets, born to FMD vaccinated sows, the MDA had a suppressive effect on the early vaccination response. This suppression, which was affected by the titre of MDA present in the piglets at the time of vaccination, was complete in one-, two- and four-week-old piglets and partial in eight-week-old piglets. Furthermore, none of these piglets were immune to experimental infection at six to seven months old.  相似文献

A flock of 294 Swaledale sheep was injected with brain tissue from different sources so as to include strains of natural scrapie currently affecting the Swaledale breed. Discounting intercurrent deaths, 85 per cent of the flock developed clinical scrapie within an observation period of 2557 days (seven years). Scrapie cases fell into distinct early and late groups, occurring at (mean +/- sem) 287 +/- 7 days and 1207 +/- 38 days after injection, respectively. Breeding groups were formed from the survivors after the early cases had occurred and successive lamb crops were produced. Two groups of progeny and two groups of controls were injected with scrapie. The incidence of scrapie in the progeny (27 per cent) was much lower than in the controls (64 per cent). The incidence in the progeny was also assessed in relation to the response of their parents: it was high (about 40 per cent) when one or both parents developed late scrapie but otherwise very low (about 5 per cent). Hence the progeny of surviving parents had a greatly reduced susceptibility to the strains of scrapie used. Seventy-two of these selected progeny formed a nucleus flock which is being bred at Redesdale. The next stage of the experiment is to assess the effect of selected rams in recorded flocks with a high incidence of natural scrapie.  相似文献

Bivalent Newcastle disease (ND)/infectious bursal disease (IBD) and trivalent ND/IBD/infectious bronchitis (IB) inactivated oil emulsion vaccines were prepared in the laboratory and evaluated under field conditions. Broiler breeder parent chickens previously vaccinated with live vaccines were inoculated with commercial monovalent ND and experimental bivalent or trivalent oil emulsion vaccines. The commercial vaccine induced a higher initial ND haemagglutination inhibition (HI) response than the experimental vaccines but, by 34 weeks after vaccination, the mean ND HI levels were not significantly different in any of the three flocks. All three vaccines provided sufficient ND immunity to protect against the clinical disease and egg production losses. The IBD responses of both flocks vaccinated with oil emulsion vaccine were similar to each other and only slightly lower than those flocks vaccinated with monovalent IBD oil emulsion vaccine in earlier experiments. Six weeks after vaccination, sufficient immunity was transferred to protect all the progeny against IBD challenge up to 33 days of age and some of them up to 45 days of age. Thirty-four weeks after vaccination of the parents with oil emulsion vaccine, the progeny were totally immune up to 27 days of age and some of them were immune until 37 days. Application of oil emulsion vaccines in bivalent or trivalent form did not impair the responses of the chickens to the monovalent components.  相似文献

Pasteurella multocida toxin was purified by affinity chromatography and inactivated by treatment with formaldehyde before use as a single component vaccine against progressive atrophic rhinitis in pigs. Twenty pregnant gilts which were vaccinated twice before farrowing with either low or high doses of the purified toxoid, developed dose-dependent positive serum and colostrum titres to the toxin and, unlike the progeny of 10 untreated control gilts, the offspring of the vaccinated gilts also had serum titres. These titres could be measured in blood samples taken for more than eight weeks from birth for most pigs born to gilts vaccinated with low doses and more than 12 weeks for pigs born to gilts vaccinated with high doses of the vaccine. All the piglets were inoculated intranasally with Bordetella bronchiseptica and toxigenic P multocida. The clinical and post mortem examinations of snouts revealed a significant reduction in the frequency and degree of conchal atrophy in the two groups of pigs from the vaccinated gilts compared with the pigs from control gilts. Clinically 90 per cent of the snouts of pigs born to vaccinated gilts appeared normal whereas only 28 per cent of the snouts of control pigs were not shortened or deviated at eight weeks of age. Vaccination of parent chickens with a commercial live infectious bursal disease (IBD) vaccine under field conditions at varying ages and by different routes resulted in variable susceptibility to the disease in their chicks. There was little correlation between the methods of vaccination and the levels of immunity in the chicks. There was some evidence that levels of transferred immunity decreased with the age of the parents. Of five flocks examined, the onset of susceptibility to IBD occurred at two days of age in two flocks and about one week of age in the other three. Four flocks were completely susceptible by the 27th day, the other at 31 days. Precipitins were demonstrated at day old in 100 per cent of chicks in three flocks and in 80 per cent in the other two. The precipitins had disappeared in all flocks by 17 days of age.  相似文献

A vaccine containing inactivated cultures of Bordetella bronchiseptica, toxigenic Pasteurella multocida type D and dermonecrotic P multocida type D toxoid in an oil-in-water adjuvant was given to seven sows, with seven others acting as controls. Half the piglets in each litter were exposed intranasally when four days old to B bronchiseptica and when eight days old to toxigenic P multocida type D. There was considerably less sneezing in the litters of the vaccinated sows and when the piglets were 10 weeks old, only 18 per cent had deformed snouts compared with 74 per cent in the litters of the control sows. The average liveweight gain of the piglets born to vaccinated sows was significantly better (P less than 0.05) between two and 10 weeks of age than that of the piglets born to unvaccinated sows, although there were no significant lower respiratory tract lesions in either group. The conchal atrophy scores were significantly lower (P less than 0.001) in the piglets from the vaccinated sows and were negatively correlated (r = -0.37) with increasing liveweight gain. In the liters of the vaccinated sows, P multocida was not isolated from the nasal passages of the in-contact piglets and from only 7 per cent of those deliberately exposed compared with 65 per cent and 79 per cent, respectively, in the litters of the control sows. P multocida was isolated post mortem from the tonsils of 23 per cent of the piglets of vaccinated sows and from 87 per cent of those from unvaccinated sows.  相似文献

Five days old progeny chicks from breeders which have received primary and booster doses of live infectious bursal disease vaccine, demonstrated precipitating antibodies unlike progenies from single dose vaccinated breeders. All chicks from the two different groups of breeders were however seronegative at 22 days of age, despite vaccination at 7 or 14 days of age. Post vaccination seroconversion was first noticeable at 32 days in the group of chicks vaccinated at 7 days. Post challenge mortalities was significantly lower (P less than 0.05) and organ lesions very significantly minimized (P less than 0.01) in 7 day old vaccinated group than in 14 or 28 days old vaccinated chicks. These results showed that early (7 days) IBD Vaccination was superior to vaccination at 14 or 28 days, in terms of antibody response and protectivity against mortalities and bursal lesions.  相似文献

Presence and levels of maternal antibody (MA) in broiler chicks derived from hens vaccinated with a live infectious bursal disease (IBD) vaccine were investigated by a quantitative agar-gel precipitin test. At day old 100% of the chicks tested had MA; by 17 days of age it was present in only 10%. The mean MA level at day old was 337.5 UK units/ml but decreased to 6.3 UK units/ml at 17 days of age. Randomly selected chicks from the pool studied were challenged at weekly intervals from day old for 29 days with an IBD virus obtained from a natural outbreak. Subclinical and clinical disease were observed in chicks challenged at eight and 29 days of age respectively.  相似文献

Sixty-four ewes were vaccinated with tachyzoites of an incomplete strain (S48) of Toxoplasma gondii grown either in the peritoneal cavity of mice (group 1) or vero cell culture (group 2) and 30 ewes (group 3) were not vaccinated. All the ewes were mated 77 days later and challenged orally with 2000 sporulated oocysts at 89 to 90 days of gestation. Ten additional unvaccinated (group 4) and 10 vaccinated (group 5) control ewes were not challenged. The unvaccinated ewes developed a characteristic febrile response to challenge while in the vaccinated ewes the fever commenced earlier but was less severe and of shorter duration. After challenge, the antibody titres against T gondii rose rapidly to high values in the vaccinated ewes while the ewes in group 3 responded more slowly. Only eight of the 45 fetuses/lambs (17.8 per cent) from group 3 were viable compared with 72.3 per cent of those in group 1 and 80.8 per cent of those in group 2. Gestation in the unvaccinated challenged ewes was shortened and the mean birthweight of their single, viable offspring was significantly lower than the weight of single lambs from the vaccinated (groups 1 and 2) and control ewes (groups 4 and 5). Examination of precolostral sera showed that almost two-thirds of the lambs from the vaccinated ewes were infected in utero. The 20 control ewes appeared clinically normal at all times and lambed normally. The two vaccine preparations were equally effective.  相似文献

To establish for how long protective antibody levels may be maintained, lambs were vaccinated with the gut membrane antigen H11 and challenged with Haemonchus contortus 14, 84, 126 or 168 days later. Compared to controls, mean faecal egg counts of vaccinated lambs were reduced by 97 per cent, 99 per cent, 92 per cent and 86 per cent respectively. Total worm burdens at postmortem five weeks after infection were reduced by 87 per cent, 94 per cent, 92 per cent and 62 per cent respectively. In vaccinated lambs, antibody levels to H11 peaked at about 60 days after the first vaccination and were maintained for the duration of the experiment. There was evidence of secondary antibody responses to H11 following challenge.  相似文献

Cows were vaccinated with formalin-killed Salmonella typhimurium approximately seven weeks and two weeks before parturition to investigate whether passive immunity could protect their calves against experimental S typhimurium infection. After birth the calves were left with their dam for 48 hours and then separated and fed cold, stored colostrum from their own dam for a further eight days. Oral challenge five days after birth with 10(8) S typhimurium did not result in the death of these calves even when they had absorbed little colostrum. Mortality was reduced to 22 per cent in calves which sucked from vaccinated dams and were then fed colostrum from unvaccinated cows and to 50 per cent in calves born to unvaccinated cows and later fed colostrum from vaccinated animals. Calves which sucked from a vaccinated dam and then received stored colostrum from the same cow excreted salmonellas for significantly shorter periods after challenge and were less often infected at necropsy 28 days after inoculation. Protection was not correlated with the levels of O or H agglutinating antibodies in serum, which were at a maximum 24 hours after sucking and then slowly declined. There was no evidence of an active antibody response in the serum. Measurement of the O and H response of cows after vaccination indicated that the vaccination schedule could be improved. The highest levels of agglutinating antibody were measured between two and three weeks after the first vaccination and there was only a minimal response to the second vaccination before parturition.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Following the first diagnosis of campylobacteriosis in Jamaican cattle a field study was undertaken to determine the pathogenicity of Campylobacter fetus subspecies venerealis Jam (Jamaican strain) and to evaluate the effectiveness of vaccination in controlling the disease. A total of 46 nonpregnant yearling heifers and four two-year-old bulls were used in two separate experiments. The results showed that C fetus subspecies venerealis Jam readily colonised the reproductive tract of susceptible heifers and persisted in some animals (68 per cent of unvaccinated and 33 per cent of vaccinated animals) for the duration of the experiment. Pregnancy was confirmed in 13 of 18 (72 per cent) culture-negative heifers but in only eight of 28 (29 per cent) of the heifers with two or more positive cultures. Vaccination appeared to be curative because 44 per cent of vaccinated heifers were cleared of infection whereas 85 per cent of unvaccinated, inoculated heifers remained infected for at least 17 weeks. Vaccination improved the fertility level of the infected heifers threefold. Infection was not established in vaccinated bulls used for breeding infected heifers.  相似文献

This study evaluated the use of an avirulent live Salmonella Choleraesuis vaccine to reduce the seroprevalence and number of Salmonella carrier pigs at slaughter. Seven batches of 500 pigs were included in each of the two study groups: the vaccinated group (VG) that was orally vaccinated and the control group (CG) that received a placebo on the first day of life. The groups were managed in a three-site system and followed up from birth to slaughter. Blood samples (n=378) were collected from each VG and CG to monitor the on-farm seroprevalence in both groups. Mesenteric lymph nodes and blood from animals (n=390) belonging to each group were collected at slaughter. At the first day of life, the seroprevalence in control batches ranged from 77.9 to 96.3 per cent, while in vaccinated batches, it ranged from 66.6 to 92.6 per cent. At weaning (21 days of age), the number of seropositives decreased in both groups (mean of 12 and 3.7 per cent for CG and VG, respectively). At slaughter, batches of VG had a significantly (P<0.0001) lower seroprevalence (46.6±5 per cent) and isolation of Salmonella from lymph nodes (33.1±5 per cent) compared with CG batches (79.7±4 per cent and 59.5±5 per cent, respectively). The results indicate that administration of a Salmonella choleraesuis-attenuated vaccine on the first day of life decreases Salmonella isolation and seroprevalence in pigs at slaughter.  相似文献

Serum antibody titres to canine distemper virus (CDV), canine parvovirus (CPV) and rabies were measured in dogs that had not been revaccinated annually and compared with the titres in a control group of regularly vaccinated animals; 83 per cent (171 of 207) of the dogs vaccinated against CDV one or more years earlier had serum neutralising antibody titres equal to or greater than 16; 64 per cent (136 of 213) of the dogs vaccinated against CPV one or more years earlier had haemagglutination inhibiting titres equal to or greater than 80; and 59 per cent (46 of 78) of the dogs vaccinated against rabies two or more years earlier had serum neutralising antibody titres equal to or greater than 0.5 iu/ml. Three weeks after a single booster vaccination the dogs' antibody titres against CDV had increased above the threshold level in 94 per cent of the dogs, against CPV in 68 per cent, and against rabies in 100 per cent.  相似文献

The case records of 80 dogs in which idiopathic inflammatory bowel disease (IBD) had been diagnosed were reviewed, and owners were contacted for follow-up information using a telephone questionnaire. The types of IBD encountered were lymphocytic (n=6), lymphocytic-plasmacytic (n=38), eosinophilic (n=6) and mixed inflammation (n=30). Prednisolone, sulphasalazine, metronidazole and tylosin were the most frequently prescribed medications. At follow-up, 21 dogs (26 per cent) were classified as being in remission (for a median of 14 months), 40 dogs (50 per cent) had intermittent clinical signs (for a median of 17 months) and three dogs (4 per cent) had uncontrolled disease (for a median of 19 months). Ten dogs (13 per cent) had been euthanased due to refractory IBD and four of these had entered remission for a median of 21 months prior to developing severe relapse and refractoriness to further treatment. Six dogs (8 per cent) had been euthanased or had died for reasons unrelated to IBD. Hypoalbuminaemia at the time of diagnosis was significantly associated with a negative outcome (P=0.0007). No association was found between the site (P=0.75), type (P=0.44) and severity (P=0.75) of disease. Dietary change to single protein and carbohydrate commercial diets had no association with outcome (P=0.12). Owner assessment of quality of life at follow-up was significantly associated with outcome (P=0.006).  相似文献

A field study was designed to determine the prevalence of subclinical infectious bursal disease (IBD) in broiler chickens from a commercial poultry company. Bursae of Fabricius (BF) from two vaccinated and three nonvaccinated broiler flocks were evaluated histologically, and antibody profiles of these broiler and matched parent breeder flocks were established. Lesions of IBD, including lymphoid necrosis, stromal edema, and infiltrates of heterophils and macrophages, were first detected in BF at 24 days of age in both vaccinated and nonvaccinated chickens. At 41 days, all BF had lesions characteristic of IBD, including severe lymphoid depletion, proliferation of epithelial cells, and mild fibroplasia. Although mean maternal antibody levels (measured by enzyme-linked immunosorbent assay) in broilers were apparently protective through day 12, IBD antibodies decreased to nonprotective levels (below 1,000) by day 16 or 20. Titers began to increase by day 28 or 32 because of field exposure. Sentinel birds, placed with broiler flocks, also developed IBD antibody titers. Broiler breeders had low and nonuniform antibody titers. Prevalence of field IBD exposure was high, and existing vaccination programs were not effective.  相似文献

Canine distemper virus (CDV) antigen was detected in the serum of dogs by an ELISA and the results of this assay were compared with an anti-CDV immunoglobulin M (IgM) antibody test. In paired sera from 26 naturally infected dogs, the antigen-positive rate was 26.9 per cent at the first examination and 11.5 per cent at the second examination two to three weeks later. The antigen was detected in three of the 10 dogs which were negative for anti-CDV IgM antibody at the first examination. It could also be detected in the serum of between eight and two of 40 specific pathogen-free dogs vaccinated against CDV, for up to four weeks after they were vaccinated.  相似文献

Five groups of Tswana-cross castrated male cattle between 20 and 30 months of age (a total of 158 animals) were transported from a ranch in a heartwater-free area of south Botswana to a feedlot near Gaborone in the east of Botswana where heartwater is endemic. On arrival, one group was vaccinated intravenously with the Onderstepoort sheep blood heartwater vaccine, one group was vaccinated intravenously with the new Onderstepoort tick-derived heartwater vaccine and a third group was vaccinated subcutaneously with this tick-derived vaccine. Vaccine reactions were blocked with long acting oxytetracycline on the first day of fever. A fourth group had a series of injections of long acting oxytetracycline on days 0, 7, 14 and 21 after arrival, and a fifth served as untreated controls. The animals remained at the feedlot for 65 days during which time they faced a low level of challenge by Amblyomma hebraeum ticks. None contracted heartwater and so they were then challenged, together with a further group of control cattle, with a dose of the sheep blood vaccine. Some animals in all groups had severe heartwater reactions and died despite therapy, but 76.7 per cent, 64.5 per cent and 74.3 per cent of the cattle in the blood vaccine, intravenous tick vaccine and long acting oxytetracycline groups respectively were resistant to challenge, compared with 48.3 per cent of the subcutaneous tick vaccine group and 36.4 per cent of the controls. It was concluded that intravenous vaccination of susceptible adult cattle with either the blood or the tick-derived vaccine needs careful monitoring in the month after vaccination and does not necessarily result in immune animals.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
