Summary Plants grown from seed derived by crossing conventional European S. tuberosum material were compared with plants grown from seed derived by crossing S. tuberosum with various, Andean frost resistant tuberbearing Solanum species. Biomass growth at optimal (20°/10°) and suboptimal (10°/4°) temperatures was studied.Differences in increase of fresh and dry matter were found between populations of Andean and European orgins at 10°/4°. At 20°/10° no such differences were found.At suboptimal temperature, Andean hybrids produced significantly more fresh and dry matter than European crosses in two harvests (64 and 178 days old plants). Statistically, Andean hybrids were found to produce the same amount of dry matter per day at both temperatures, over the complete growth period.Height increase and flower development were strongly depressed in European crosses under the suboptimal temperatures, but much less so in Andean crosses.The Andean material appears to be an under utilized resource in potato breeding for cool climates.  相似文献

Summary The main obstacle in breeding potato (Solanum tuberosum L.) dihaploids is the severe limitation of male fertility. To determine pollen viability assays that correlate well to fertility in crosses, results of five different pollen viability assays were compared by correlation analysis with fruit and seed set characters in test crosses, and to pollen tube growth in situ (PL-test). The methods used were: staining the pollen cells with carmino acetic acid (CAA-test); in vitro pollen germination (PG-test); and detection of pollen staining rates after incubation with fluoresceine diacetate (FDA-test), 2-3-5-triphenyle tetrazolium chloride (TTC-test), and 5-bromo-4-chloro-3-indolyle--galactoside (X-Gal-test).The results of test crosses and pollen tube growth in situ correlated with the results of all other assays with the following ranking, from highest to lowest: enzyme activity assays (X-Gal-test, FDA-test, TTC-test), in vitro pollen germination (PG-test), and pollen staining by CAA. The newly developed X-Gal-test for monitoring -galactosidase activity showed the least variation of all assays investigated. A mutation breeding method was developed to select clones of Lemhi Russet that have resistance to blackspot bruise and low temperature sweetening. Following irradiation with gamma rays from a Cobalt60 source, over 2000 tuber eye pieces were planted directly to the field and tubers from the resulting crop were individually evaluated for blackspot bruise potential. Selection for blackspot bruise resistance continued for five clonal cycles. Selection for low temperature sweetening began in the M3 and continued for three cycles. Ten clones were selected, eight with significantly (p=0.05) better blackspot bruise resistance, and two with increased resistance to low temperature sweetening. The results confirm the possibility for the development of a system to improve single selectable quality characteristics in potatoes.  相似文献

Summary Pollen grains from selected cutivars of almond [Prunus dulcis (Mill.) D. A. Webb] and peach (Prunus persica Batsch L.) were exposed to a range of temperatures between 1°C and 34°C on a thermogradient plate. Pollen germination at temperatures below 9°C was conspicuously greater in almond than peach. Miximal germination percentages were attained at about 16°C (almond) and 23°C (peach). The two species did not differ in their capacity for pollen tube elongation over a broad range of temperatures. Maximal pollen tube elongation occurred at temperatures 5°C to 8°C higher than maximal pollen germination. Species affiliation appeared to be of much greater consequence than chilling requirement or bloom date of the sporophyte in predicting gametophytic response to temperature.  相似文献

Summary Low temperature germination responses were evaluated for 18 high altitude accessions representing five wild Lycopersicon species and 19 accessions of L. esculentum which have reputed ability to germinate in the cold. Survival analysis indicated that one accession of L. chilense germinates better at 10°C than PI 120256, the fastest-germinating L. esculentum genotype, and that PI 120256 germinates as well as PI 126435 (L. peruvianum). Additional wild ecotypes exhibiting rapid germination at 10°C were identified from L. peruvianum and L. hirsutum. These ecotypes may possess genetic potential for introgressing cold germination ability into L. esculentum cultivars.  相似文献

Summary A number of previously selected hydroxyproline (hyp) resistant cell lines of a diploid potato (Solanum tuberosum L., clone H2578, 2n=2x=24) could be regenerated into plants which were further analysed. Hyp resistance, although lower than in the originally selected calli, was still present in regenerated shoots and in callus initiated from these shoots and it was not lost upon (mini)tuber propagation. Regenerated shoots showed a wide range of phenotypic variation. The chromosome number, analysed in 4 clones, appeared to be hypotetraploid (44 or 45). Tuber-propagated regenerants generally showed increased frost tolerance both at the plant and the cell level. In leaves this seemed to be associated with increased levels of both proline and total amino acid content. However, in callus only the total amino acid content but not proline was still elevated. In the wild type the frost tolerance of the tubers appeared to be lower than that of the leaves. Between tubers of wild type and the hyp resistant regenerants no differences in frost tolerance were found, although proline and total amino acid content tended to be higher in tubers from the regenerants.  相似文献

Summary In vitro pollen germination at 10°C, 14°C and 22°C of four groups of two pure line tomato varieties was compared with their plant growth at 19°C D/10°C N under controlled environmental conditions. Generally, pollen germination was slow at 10°C but after 6 h the percentage of germination was similar to that at 22°C. Maximum germination was obtained at 14°C already after 4 h. The longest pollen tubes occurred at 22°C. The two varieties within each group differed significantly from each other for percentage of pollen germination. For one group, this difference was greater at 10°C than at 22°C. The varieties in two groups also differed significantly for pollen tube growth. These differences in pollen tube growth rate were greater at 22°C than at 10°C. There was no clear relationship between pollen germination, pollen tube growth and plant growth in any of the four pairs of varieties. The results are discussed with regard to the possibility of pollen selection at low temperature in order to improve the efficiency of breeding for growth at low temperature.  相似文献

Haploid selection for traits related to pollen cold tolerance in tomato was performed in segregating populations derived from a Lycopersicon esculentum × L. pennellii hybrid. BC1 populations were obtained by combining normal and low temperature treatments on two stages of pollen development: pollen formation, and germination and pollen tube growth. F1 hybrids were cultivated under low and normal temperatures and their pollen was used to pollinate L. esculentum plants at low and normal temperatures. The four BC1 populations obtained were tested for the quality and quantity of pollen produced at low temperatures. The population obtained by cold treatment at both stages had a significantly improved pollen germination ability at low temperatures. The two other coldselected BC1 populations showed no differences compared with the unselected BC population. A second cycle of pollen selection, corresponding to BC2, was applied in order to test its persistence in the subsequent generations and the possibility to further improve the character. This second cycle showed no improvement although some plants retained the high pollen germination ability at low temperatures that was observed in the first cycle. Hence, gametophytic selection of some characters related with tomato pollen performance may be feasible, at least for the first cycle of selection.  相似文献

Summary Three staining methods (acetocarmine, fuchsin and oxidation of benzidine) and germination in vitro and in vivo were applied to estimate pollen fertility in Solanum species and dihaploids. Pollen was divided into six classes based on shape and contents of the grains. With acetocarmine, fuchsin, peroxidase and germination in vitro 4, 3, 2 and 1 classes, respectively, are supposed to be included in the percentage of good pollen as measured by these methods. This percentage therefore, in more than 96% of the cases studied, shows a decrease in the order indicated. Neither aging of pollen at room conditions nor collecting pollen from flowers on 1–9 days after anthesis does influence the percentage of good pollen with acetocarmine and fuchsin, whereas this percentage drops sharply to zero with peroxidase and germination in vitro. The latter two methods apparently measure as good pollen only the grains with living cytoplasm. When pollen is collected at three successive dates from the same flowers the percentage of good pollen drops sharply with all methods used. There is a relation between quantity of pollen per flower and pollen quality (% good), low-quantity pollen containing significantly lower percentages of good pollen than medium- and high-quantity pollen. The latter two are not significantly different in this respect.From calculations of correlation coefficients it is concluded that only germination of pollen in vitro is significantly correlated with berry and seed set and thus gives a reliable estimate of male fertility. This does not hold true for the two staining methods without due reserve. The peroxidase method is not useful for the Solanum material studied.After standardized pollination the average number of haploid pollen grains on diploid stigmata was found to be 1625±127, that of diploid pollen on tetraploid stigmata 2863±98.A. Summary The development of potato cultivars having acceptable chipping quality after cold storage is important because it reduces costs to growers while ensuring continuity of supply to processors throughout the year. Over 5100 single-hill progenies were planted to select for cold chippers. A breeding procedure for selecting and evaluating these progenies for cold chipping ability was used, and 38 desirable genotypes were identified. These selections were increased in the second clonal generation and evaluated for yield, specific gravity, and seven chipping treatments of varying storage duration/temperature/reconditioning duration and were compared to standard chipping cultivars Atlantic, Monona, Norchip, and Snowden. Twenty-two selections yielded Norchip the highest yielding cultivar, while 15 selections had a specific gravity Atlantic, the highest specific gravity cultivar. Snowden was the best chipping cultivar overall and some selections were not significantly different than Snowden. Overall, nine selections combined high yield and specific gravity with the ability to produce attractive chips from 4° C. Single-hill selection for cold chipping could potentially save four years in the breeding process by the identification of good parents, the recycling of good parental cross combinations, and the evaluation of progeny for chipping earlier in the breeding program.  相似文献

Summary Salt-tolerance in potato (Solanum tuberosum L.) was selected on the basis of germination and survival of seed sown in trays of perlite suspended in either 75 or 150 mM NaCl. Salinity reduced the germination of seed. Genetic differences in salt-tolerance were apparent with salt reducing germination more in seed collected from cv. Cara than in that collected from cv. Maris Piper. Progeny from the seedling selection were then grown to maturity to produce tubers.The relative tolerance of the parental cultivars and of unselected and selected progeny to long-term exposure to salinity was examined in a pot experiment in which plants were irrigated with either fresh water or 50 mM NaCl solution from one week after plant emergence. In this experiment, salinity significantly reduced leaf conductance, total dry matter production and partitioning of assimilate to tubers. Salinity reduced dry matter production and assimilate partitioning to tubers to a greater extent in Cara than in M. Piper. Progeny selected for short-term salt-tolerance did not exhibit greater long-term salt-tolerance than unselected progeny, and both were more sensitive than M. Piper.These results demonstrate genetic variation in salt-tolerance in potato. However, although there was a correlation between the performance of the parent to long-term salinity and survival of progeny in the seedling selection, there was no correlation between short- and long-term salt-tolerance. This suggests that characters underlying short-term tolerance may contribute to long-term tolerance but do not of themselves confer long-term tolerance. Future progress in selecting for improved salt-tolerance depends on understanding the effects of salinity on the physiological processes underlying growth and carbon partitioning.  相似文献

Summary For the production of inbred lines and F1 hybrids in witloof-chicory information is wanted on characteristics such as the incompatibility system. These characteristics can only be studied properly if the influence of temperature and physiological status of the plant on pollen germination and seed production is known. Investigations were carried out with 9 self-incompatible (SI) and 6 self-compatible (SC) clones in glasshouses of the IVT phytotron at constant temperatures of 10, 14, 17, 20, 23 and 26°C. In general, in vivo pollen germination percentages were rather low after self pollination with an optimum for germination around 17–20°C. No seeds were formed at the lowest temperature (10°C) while seed production for SC clones was usually (rather) good at higher temperatures. At 26°C seed production in some clones decreased. Both pollen germination and seed production decreased at the end of the flowering period. There was a rather positive relationship at e.g. 17 and 20°C between pollen germination after selfing and seed production. When no pollen germination was observed, no seed formation occurred. When pollen grains did germinate, seed development would not necessarily occur in all cases. So this relationship only enables negative mass selection for SC.  相似文献

Low-temperature adapted Andean potato clones and low-temperature sensitive European clones cultivated at 20/10°C were compared during stress occuring as the result of low-temperature treatments. The stress response of leaf tissue was assessed by measuring the decrease in chlorophyll fluorescence (Fv/Fm) in detached leaves during a period of six days. After temperature stress combined with light Fv/Fm decreased only slightly in the Andean clones, but was markedly lower than the initial values in the European clones, However, after a 2°C treatment in darkness no difference between clones of different origins was detected. In general, chlorophyll and carotenoid contents of the Andean clones were higher than those of the European clones. They were markedly lower in all clones in treatments where low temperature was combined with light, than at 20/10°C and at 2°C in the dark. The contents of carotenoids and total chlorophylls decreased more in the European than in the Andean clones when leaves were transferred to a higher light intensity and then the lowest temperature used was combined with light. Thus carotenoids and chlorophylls in the Europeans clones were more sensitive to photo-oxidation at low temperatures than in the Andean clones. The changes in chlorophyll fluorescence in illuminated, detached leaves induced by a decrease in temperature were found to correlate with the low temperature performance of the genotypes. The correlation between low temperature performance and photosynthetic activity was highly significant and could be used by plant breeders to identify plants that perform well at low temperatures.  相似文献

Summary Tuber characteristics of potato clones (Solanum tuberosum L.) regenerated from cultured leaf, rachis or stem pieces of cv. Desiree were assessed in field trails over three years. In the first two seasons, when the crop was grown for seed potato production, tuber numbers and the tuber yields were normally distributed and the population means of the clones were about the same as those of the controls. When 78 of the clones were grown as a ware crop in a replicated field trial there were statistically significant differences between most clones and the controls in total yield (generally lower) and ware yield (generally higher), brought about by alterations of the size distributions of tubers. In all years the population of clones had fewer scab (Streptomyces scabies) lesions than the controls. Clones with consistent differences in flesh colour and eye depth were also observed. Two out of 78 clones, in 1984, had significantly higher dry matter content. No stable decrease in susceptibility to the cyst nematode Globodera pallida was observed amongst 167 clones tested. These potato plants regenerated from explants clearly show that somaclonal variation can include agronomically relevant changes.  相似文献

Summary Germination trials of pollen grains of the tea hybrid rose cultivars: Ferry Porche, Bronze Masterpiece, Queen Elizabeth, John F. Kennedy and Lady X, were carried outin vitro, to determine whether these cultivars are suitable as pollen donors in a breeding programme.Pollen grains of all five cultivars germinated poorly in a medium containing sucrose only. Addition of boric acid in the medium improved the germination percentages. The highest germinability was observed in Ferry Porche and Lady X on 15% sucrose+50 ppm boric acid, and 20% sucrose+100 ppm boric acid, respectively. Addition of calcium nitrate in the medium reduced germination percentages in all cultivars.The effect of pH of the medium on pollen grain germinability depended on the cultivar and the composition of the medium. On a medium free of boric acid, the pH values between 5.5 and 7.0 caused a significant increase in the germination percentages of pollen grains of Lady X and Queen Elizabeth. The addition of boric acid decreased the responsiveness of the pollen of all cultivars to the pH changes of the medium.The germination capacity of pollen grains of Lady X and Ferry Porche was not affected by incubation temperatures ranging between 20°C and 30°C.On the basis of the above results Lady X and Ferry Porche are considered the most suitable for pollen donors.  相似文献

Summary A quantitative examination of pollen grain germination and pollen tube growth within the overy wall was made in selected wheat × tetraploid H. bulbosum hybridizations, to investigate the effect of the wheat and the H. bulbosum genotype on these characters. As expected, variation at the known crossability loci had no effect on pollen grain germination. The frequency of pollen tube penetration of the ovary wall was, however, severely reduced when the dominant alleles were present. Pollen tube penetration was nevertheless observed in 3 of the 110 ovaries examined of the non-crossable cultivar Highbury. The H. bulbosum genotype had a much smaller effect on these characters, but significant differences between the clones were observed in the numbers of pollen tubes initially penetrating the ovary wall. Although two H. bulbosum genotypes showed no significant differences in the number of ovaries with pollen tubes at the base of the ovule, significant differences in the frequency of fertilization were observed. The possible cause of this discrepancy is discussed.The frequency of fertilization in crossable wheat × H. bulbosum hybridizations was improved by the application of gibberellic acid within 10 minutes of pollination, and reduced by an increase in the ambient temperature from 20°C to 26°C. Fertilization following the pollination of non-crossable wheat genotypes was not affected by either of these factors.  相似文献

Summary In studies to determine the inheritance of response-time traits, such as time to seed germination, some viable individuals may fail to respond during an experiment. If these right-censored observations are ignored, sample means and variances will be underestimated. This is illustrated using data from time to seed germination at 9°C for Lycopersicon esculentum (Mill.) fast germinating PI 120256, slow-germinating T3 and their reciprocal F1, F2 and backcross progeny. This paper presents methods to detect and to accommodate right-censored data in generation means analysis. Genetic interpretations derived from corrected and uncorrected estimates of generation means and variances are compared. Correction for right-censoring increased estimates of environmental and phenotypic variances, and decreased heritability estimates.  相似文献

Summary The main objective was to determine the reliability of selection in early generations of a potato breeding programme. To that end data were recorded in the seedling generation, an early and a late harvested experiment of the first clonal generation and the late harvested second clonal generation on up to 30 genotypes of 55 populations from crosses of three Solanum tuberosum ssp. tuberosum cultivars and seven selected S. tuberosum ssp. andigena clones, adapted to the northern temperate zone. Phenotypic correlations were computed per population in almost all possible combinations within and between characters between and within generations. The mean across 55 comparable correlation coefficients was considered to be the best estimate of the relationship concerned.In all four experiments data were collected on number of tubers, tuber yield and mean tuber weight; in the three experiments of the two clonal generations in addition on date of emergence, number of stems, plant height, haulm type, and general impression of the underground plant parts and in the two late harvested clonal generations on date of maturity and under water weight.The results confirmed that general impression which is largely decisive for the selection in early generations, is a little reliable measure. However, from the magnitude of the mean correlations between experiments within characters it must be concluded that selection for most of these separate characters is hardly more reliable. An exception was date of maturity for which a mean correlation was determined of r=0.78 between the two clonal generations. The second strongest mean correlations, perhaps surprisingly, were for tuber yield, viz. r=0.52 between the seedling generation and the late harvested first as well as the second clonal generation and r=0.63 between the late harvested clonal generations. From the correlations between characters it appeared that for a high tuber yield, besides its components, in particular an early emergence, a large plant height, an attractive haulm type and a late maturity are of importance.The main conclusion is that, as long as not enough seed tubers are available for the layout of statistically sound experiments, a negative instead of a positive selection should be applied to avoid the rejection of most of the valuable genotypes in an early stage. On the basis of the results directives for the application of such a negative selection are given.  相似文献

Summary Cultivars tolerant to low temperature during the germination and emergence stages and carriers of the grain quality standards demanded by consumers are needed to increase the success of irrigated common bean in Southern Minas Gerais State. To study the genetic mechanisms controlling these traits and assess the possibility of generating the desirable materials, a diallel cross involving ten cultivars including introductions and pure lines from the breeding program of ESAL was carried out. Speed of germination of the F2 generation from the crosses was assessed under laboratory conditions at 12°C. The diallel analysis was carried out using Griffing's method IV (1956) and predictions of the cross potential were made by the methodology developed by Jinks & Pooni (1986). The results indicated that the parents differed in germination speed at low temperatures with Small White, A-488 and Rio Vermelho being the most tolerant and Carioca, ESAL 591 the most sensitive. No effect of the reciprocal crosses was observed either for germination percentage or germination speed. The parents A-488, Small White and Rio Vermelho showed the greatest general combining ability. Additive genetic effects predominated for both traits. Our results suggest that cold tolerance can be bred successfully into commercially acceptable cultivars.  相似文献

Summary The inheritance of potato tuber yield was studied considering the seasonal course of harvest index. Harvests of several progenies and their parents were performed at intervals and fresh weight of foliage, underground stems, tubers and stolons was assessed.Tuber yield depends on the time of tuber initiation and the proportion of the assimilates allocated to the tubers, as reflected in the increase rate of harvest index. The time of tuber initiation of the progenies was associated with the time of tuber initiation of the earliest tuberizing parent. The mean increase rate of the harvest index of the progenies was in most cases equal to the mean values of parents, however in crosses combining early with late maturing parents the rate equaled the value of the maternal parent. Using these relations, differences in harvest index between progenies can be predicted with a high degree of precision by using the harvest index parameters of the parents. This estimated harvest index showed a fairly good correlation to tuber yield at mid-September and can be used in the prediction of the yield of the progeny. The use of the mid-parent value for this purpose is not suitable.  相似文献
