Summary The main purpose of the research reported in this article was to see if the biomass maintenance of parental potato clones was correlated with the growth and yield of their progeny. Seedling populations with parents of European and of Andean origin were cultivated at a temperature of 10°C during the day and 4°C at night. The biomass maintenance of the parents was defined as the ratio between the total biomass yield at 20°C day/10°C night and the total biomass yield at 10° C day/4°C night. The Andean parents had good maintenance ability at the lower regime compared to the Eurpean parents.Biomass maintenance appears to be hereditary. Significant correlation was found between the dry matter maintenance of the parents and the growth rate of seedlings, especially during the second month after potting. In populations with only Andean or only European parents, differences in height increase and leaf formation rate between seedling populations were correlated to the dry matter maintenance of the parents cultivated at the temperature regime 10/4°C. In the hybrid population between an Andean and an European parent only differences in height increase during the second month after potting correlated well to the average dry matter maintenance of the parents. It appears therefore that the rate of height increase during the second month after potting can be used as a criterion for selection.  相似文献

Summary Pollen from four male pistachio (Pistacia vera L.) clones was stored at –196°C and –20°C for up to 12 months and tested for ability to germinate in vitro following a period of hydration at high humidity. Germination of fresh pollen was high (>80%) for each clone. At –196°C, pollen of cv. Peters survived freezing, storage and thawing with no loss of germinability; pollen of the other three clones had sharp declines in germination possibly attributable to cellular lesions incurred during freezing or thawing. When the relative humidity of the –20°C storage environment was maintained at or near 33%, Peters pollen had high rate of germination through 12 months storage. Without control of relative humidity, Peters pollen germination was high at 4 months, but declined at 12 months. Germination requirements became more exacting for pollen stored at –20°C for 12 months at suboptimal humidity conditions. Pollen of the other three clones did not tolerate storage at –20°C as well as Peters pollen regardless of the storage humidity environment.  相似文献

Summary Plants grown from seed derived by crossing conventional European S. tuberosum material were compared with plants grown from seed derived by crossing S. tuberosum with various, Andean frost resistant tuberbearing Solanum species. Biomass growth at optimal (20°/10°) and suboptimal (10°/4°) temperatures was studied.Differences in increase of fresh and dry matter were found between populations of Andean and European orgins at 10°/4°. At 20°/10° no such differences were found.At suboptimal temperature, Andean hybrids produced significantly more fresh and dry matter than European crosses in two harvests (64 and 178 days old plants). Statistically, Andean hybrids were found to produce the same amount of dry matter per day at both temperatures, over the complete growth period.Height increase and flower development were strongly depressed in European crosses under the suboptimal temperatures, but much less so in Andean crosses.The Andean material appears to be an under utilized resource in potato breeding for cool climates.  相似文献

Summary The influence of temperature on self-incompatibility of six S-homozygous clones of Brussels sprouts was studied. The clones were treated with constant temperatures of 14°, 17° and 20°C and alternating day and night temperatures of 17/14°, 20/14°, 23/14° and 26/14°C. To determine the degree of incompatibility the mean number of pollen tubes per style after selfing was calculated.The clone with the weak S-allele S-5 was less self-incompatible in the 23/14° and 26/14°C treatments than in the other treatments. The other clones with the weak S-alleles S-15 and S-45 remained sufficiently self-incompatible during all treatments. The clone with the rather strong S-39 showed the highest level of self-incompatibility at 26/14°C.In the two weakly self-incompatible clones with S-5 and S-39 a clear difference in self-incompatibility was found between young and old flowers. The strongly SI clones with S-39 showed no difference and in the two clones with S-15 the differences were small and significant in one case only.  相似文献

Summary For the production of inbred lines and F1 hybrids in witloof-chicory information is wanted on characteristics such as the incompatibility system. These characteristics can only be studied properly if the influence of temperature and physiological status of the plant on pollen germination and seed production is known. Investigations were carried out with 9 self-incompatible (SI) and 6 self-compatible (SC) clones in glasshouses of the IVT phytotron at constant temperatures of 10, 14, 17, 20, 23 and 26°C. In general, in vivo pollen germination percentages were rather low after self pollination with an optimum for germination around 17–20°C. No seeds were formed at the lowest temperature (10°C) while seed production for SC clones was usually (rather) good at higher temperatures. At 26°C seed production in some clones decreased. Both pollen germination and seed production decreased at the end of the flowering period. There was a rather positive relationship at e.g. 17 and 20°C between pollen germination after selfing and seed production. When no pollen germination was observed, no seed formation occurred. When pollen grains did germinate, seed development would not necessarily occur in all cases. So this relationship only enables negative mass selection for SC.  相似文献

Summary Interspecific crosses were made to introduce the disease resistance of Cyclamen graecum into C. persicum cultivars and the abortive hybrid embryos were rescued by ovule culture. Diploid and tetraploid cultivars of C. persicum (CPD, 2n=2x=48; CPT, 2n=4x=96) were the pistillate parents and wild form of C. graecum (CG, 2n=84) were the staminate parents. After pollination, crossed ovaries were periodically collected and examined using paraffin sections. Histological observations suggested that both hybrid ovules of CPD × CG and CPT × CG should be transferred to the culture medium 35 days after pollination. Based upon this observation, crossed ovaries were collected 35 days after pollination, then ovules with placenta were explanted on culture medium and cultured in the dark at 25°C. Plantlets were induced from ovules cultured in MS medium containing 3% sucrose and 10% coconut milk. The hybrids (2n=66) derived from CPD × CG failed to yield viable seeds by self-pollination, although they showed some pollen fertility. In contrast, the hybrids (2n=90) derived from CPT × CG showed high pollen fertility and yielded viable seeds by self-pollination. Furthermore, they were resistant to disease caused by Fusarium oxysporum f. sp. cyclaminis, Erwinia herbicola pv. cyclamenae and Pseudomonas marginalis pv. marginalis.Abbreviations CPD C. persicum diploid - CPT C. persicum tetraploid - CG C. graecum  相似文献

Self and cross-incompatibility determination by means of fruit and seed set experiments or pollen tube growth observations in the style has been frequently reported to be unclear in pear (Pyrus communis L.). Thus,in order to develop a reliable in vivo method to test pollen-pistil incompatibility in pear, pollen tube performance has been studied along the pistil following self and cross-pollinations. Results show that, while pollen tube growth in the style may be an unclear test, ovule observation at the microscope for the presence of pollen tube in the nucellus is a proper method to test incompatibility in this crop. With this analysis we could identify S-alleles of ‘Williams’ (S1S2) and ‘Coscia’(S3S4), and three of the four possible S-genotypes resulting from the ‘Williams’ × ‘Coscia’ cross, as represented by ‘Butirra Precoz Morettini’ (S1S3), ‘Santa Maria Morettini’ (S2S3)and ‘Tosca’ (S1S4). This result demonstrates that ‘Williams’ and ‘Coscia’ cultivars do not share any allele in common. We also established two new inter-incompatibility groups in pear. Furthermore, the presence of a common allele between ‘Williams’ and ‘Agua de Aranjuez’,and ‘Coscia’ and ‘Agua de Aranjuez’, three apparently unrelated old cultivars, may indicate a narrower genetic base than expected for European pear. This finding together with the fact that 40% of new released cultivars have direct or indirect parental relationship with the cultivars ‘Coscia’ and/or ‘Williams’, anticipates the possibility of new cases of cross-incompatibility for this crop in the future. Both the method described and the determination of the S-genotypes will facilitate the characterisation of self and cross-incompatibility relationships in this species. Cultivars of C. persicum diploid (CPD, 2n=2×=48) and C. persicum tetraploid (CPT, 2n=4×=96) were the pistillate parents and wild species of C. purpurascens (CP, 2n=34) were staminate parents. After pollination, crossed ovaries were collected periodically and examined using paraffin sections. Histological observations suggested that both hybrid ovules of CPD x CP and CPT x CP should be transferred to culture medium 35 days after pollination. Based up on this observation, crossed ovaries were collected 28 days after pollination and ovules with placenta were transferred to MS (1962) medium containing 3% sucrose. These ovules were cultured in the dark at 25° C. The hybrids (2n=41) derived from CPD x CP had the scent of C. purpurascens, whereas the hybrids (2n=65) derived from CPT x CP had the scent of C. persicum. Summary The maximum temperature (34°C) at which perennial ryegrass plants from one breeding population successfully shed pollen was determined and the seed set of ten clones taken from the same population and self-pollinated both in an unheated glasshouse and at a constant 34°C during anthesis was compared. Seed set by selfing of all ten clones was markedly increased by heat treatment, from a mean of 2.3% to 30.7%. Heat treatment of anthers, but not stigmas, greatly enhanced pollen tube growth on stigmas from the same genotype. Germination of progeny was not affected by the heat treatment. Heat treatment of ten clones from a breeding population with contrasting morphology and growth rate was not so successful. Only one clone shed pollen freely at 34°C and showed a marked improvement in subsequent seed set. The advantages to the breeder of routinely producing full-sib families of elite perennial ryegrass parents by selfing are outlined.  相似文献

Summary A quantitative examination of pollen grain germination and pollen tube growth within the overy wall was made in selected wheat × tetraploid H. bulbosum hybridizations, to investigate the effect of the wheat and the H. bulbosum genotype on these characters. As expected, variation at the known crossability loci had no effect on pollen grain germination. The frequency of pollen tube penetration of the ovary wall was, however, severely reduced when the dominant alleles were present. Pollen tube penetration was nevertheless observed in 3 of the 110 ovaries examined of the non-crossable cultivar Highbury. The H. bulbosum genotype had a much smaller effect on these characters, but significant differences between the clones were observed in the numbers of pollen tubes initially penetrating the ovary wall. Although two H. bulbosum genotypes showed no significant differences in the number of ovaries with pollen tubes at the base of the ovule, significant differences in the frequency of fertilization were observed. The possible cause of this discrepancy is discussed.The frequency of fertilization in crossable wheat × H. bulbosum hybridizations was improved by the application of gibberellic acid within 10 minutes of pollination, and reduced by an increase in the ambient temperature from 20°C to 26°C. Fertilization following the pollination of non-crossable wheat genotypes was not affected by either of these factors.  相似文献

Summary Four cultivars of Populus spp., compatible to varying degrees with four races of M. larici-populina Kleb., were raised in a controlled environment on a high (28°/20°C, day/night) and low (20°/10°C) temperature regime. Leaf discs cut from the plants were inoculated separately with four individual races of M. laricipopulina and subsequently incubated at either low (20°C) or high (25°C) temperature for 14 days when disease development on the discs was assessed using three parameters (Incubation period to flecking, uredia per leaf disc and uredospores per mm2). The degree of resistance in all cultivar/race combinations was high on cultivars cultured at a high temperature regime compared to those cultured on a low temperature regime. Analysis of variance demonstrated that the major components: pre-inoculation temperature regime, post-inoculation temperature regime, race and cultivar, and most second and third order interactions between these were highly significant (P<0.001) for most disease parameters. The variance of the temperature components and all interactions involving these were usually higher than those for the cultivar and race components and those interactions lacking temperature components.These results emphasize the importance of the temperature regime at which plants are raised and the temperature of incubation, following the inoculation in determining the relative degree of resistance of these cultivars of poplar to races of M. larici-populina. The implications of these results in the epidemiology of leaf rust and the stability of the host-parasite relationship are discussed.  相似文献

Summary The effect of short term high temperature exposure on the performance of five Phaseolus species and of long term (continuous) exposure on the performance of P. vulgaris was studied at three growth stages. Phaseolus species subjected to 26.7, 32.2 or 37.3°C for two days showed small differences in the number of pods produced and in visual leaf damage, but large differences in leaf heat killing time, as measured by conductivity. P. coccineus had the shortest heat killing time (20–60 minutes) and P. acutifolius and P. lunatus the longest times (180 and 153 minutes), respectively. The P. vulgaris genotypes were intermediate in killing times to P. acutifolius and P. coccineus. Species response was not consistent with temperature within developmental stage. On average, the number of pods decreased as temperature increased from 32.2 to 37.3°C. Heat killing time and leaf damage also increased with temperature. CO2 exchange rates of plants grown at prolonged high temperatures (30–40°C/20–30°C, day/night) decreased with the age of the plant. Shoot lenght was decreased as high temperature. P. vulgaris genotypes differed on the basis of either short term exposure or of continuous exposure. A total of 17 pollen incompatibility groups in sweet cherry (Prunusavium L.) were identified among 46 accessions by PCR based S-alleletyping analysis and by controlled test pollinations. Two putativeS-alleles different from S 1 to S 6,S z and S y were identified. Five S-genotypes, S 1 S 5, S 1 S 6,S 2 S 6, S 4 S 6, andS 5 S 6, combinations of S 1 toS 6 alleles that had not previously been identified from cultivars in NYSAES, were positively confirmed by PCR based S-genotyping analysis. Also, the S-genotypes of cultivars in some pollen incompatibility groups that had previously been incorrectly reported have been clarified. Several popular cultivars, which were previously used as testers for S-allele typing analysis, were found to have been inaccurately genotyped. In addition, the S-genotypes and self-incompatibility groups of some relatively recentlyintroduced cultivars were identified. The molecular typing system ofS-genotypes based on PCR is a useful and rapid method for identifying newS-alleles and incompatibility groups in sweet cherry.  相似文献

Summary Growth of four breeding lines and two cultivars of slicing cucumber with different growth at low temperature and light intensity was analysed in relation to night temperature. Plants were grown from 17 days after sowing at 20°C day temperature and 20, 15 or 10°C night temperature (NT), a light intensity of 24 W/m2 visible radiation and a daylength of 8 hours. Data from six successive harvests, ending 47 days after sowing, were analysed according to a MANOVA procedure.RGR, NAR, LAR, SLA and LWR generally decreased with time. Ln W and RGR were largely similar at 20 and 15°C NT, but reduced at 10°C NT. LAR and SLA decreased at each lower NT, whereas LWR increased. NAR was only marginally affected by NT. Significant genotypic differences occurred for 1n W, LAR, SLA, LWR, stem length and percentage of dry matter in leaves and stems at all NT's and for RGR and NAR only at 20°C. Genotypic differences for 1n W could not be explained by differences for RGR, but by differences for initial plant weight.Significant genotype × night temperature interactions were present for LWR. Breeding lines increased LWR in response to lower NT, while that of cultivars remained fairly constant. Summary An investigation was undertaken on the storage characteristics of pollen collected from two English rose cultivars. A rapid decline in viability was observed in pollen stored at +4° C and –20° C, whereas the viability of pollen, stored at ultra-low temperature (–196° C), remained constant. Cryopreserved pollen was shown to retain its ability for fertilisation. The effects of the stage of flower development and anther dehiscence were assessed on both pre-and post-cryopreservation viabilities. Successful long-term storage of pollen will facilitate hybridisation of rose species and cultivars that do not flower synchronously.  相似文献

Summary Pollen from eight clones of English walnut (Juglans regia L.) was stored in liquid nitrogen (LN2) at-196°C for one year. Pollen was monitored for percent germination in vitro before being frozen. Pollen was frozen within two hours of anther dehiscence and subsequently at 24 hr intervals until germination assays indicated that the ability to germinate was lost. Survival after one year was evaluated by determining percent germination in vitro. Freshly collected pollen of only five of the eight clones survived LN2. By contrast, when the same pollen was held for 24 and 72 hr before freezing, pollen of all eight clones survived. Survival is correlated with moisture content (MC) of the pollen. Pollen samples with MC greater than 7.5% were killed by freezing in LN2. All pollen samples with MC between 4 and 7.5% survived as did most of the samples with MC between 3.2 and 4%.  相似文献

Summary In vitro pollen germination at 10°C, 14°C and 22°C of four groups of two pure line tomato varieties was compared with their plant growth at 19°C D/10°C N under controlled environmental conditions. Generally, pollen germination was slow at 10°C but after 6 h the percentage of germination was similar to that at 22°C. Maximum germination was obtained at 14°C already after 4 h. The longest pollen tubes occurred at 22°C. The two varieties within each group differed significantly from each other for percentage of pollen germination. For one group, this difference was greater at 10°C than at 22°C. The varieties in two groups also differed significantly for pollen tube growth. These differences in pollen tube growth rate were greater at 22°C than at 10°C. There was no clear relationship between pollen germination, pollen tube growth and plant growth in any of the four pairs of varieties. The results are discussed with regard to the possibility of pollen selection at low temperature in order to improve the efficiency of breeding for growth at low temperature.  相似文献

Summary Six pear and five apple trials were carried out to ascertain the outcome of combinations of compatible pollen (C) with self (S) or incongruous pollen (I) as to the pollination index (PI=seeds/pollinated flower). The PI of the mixture C+I (1:5) was consistently depressed as compared to that of the control C. The results of the double pollinations S/C and I/C were affected by the temperature at pollination; their PI's at <15°C were twice as high as those at >15°C, being well above and below the PI of C in the former and latter case respectively. The opposite was true for the C/S combination, the PI of which increased with the pollination temperature; the PI of C/I did not differ much from the PI of C, irrespective of temperature. The conclusion was reached that the interaction previously and presently found between compatible and self-incompatible pollen also exists to a fair extent between compatible and incongruous pollen. However, in pear neither the mentor nor the pioneer pollen technique proved to aid its hybridization with apple, the formation of self seed was not observed either. In apple the production of apple × pear hybrids was likewise doubtful, but the double pollinations S/C and C/S formed 4–10% self seed.  相似文献

Summary To determine whether in potatoes the tetraploid level is preferable to the diploid level, especially regarding tuber yield, four diploid (2n=2×=24) Solanum phureja x dihaploid S. tuberosum hybrid parents and their vegetatively doubled, tetraploid (2n=4×=48) counterparts were intermated, which resulted in two F1 hybrid families at both levels of ploidy. The parents and clones of the F1 families and their offspring were used in crosses in such a way that in addition Sib1, Sib2, F1×Sib1, BC1 and Sib1×Sib1 families were produced. Of the first clonal generation of the 12 2 x families and their 12 counterpart 4 x families two tubers per clone were planted in three replications in a field experiment at Sturgeon Bay in 1969; of the parents six tubers were planted in each replication. Data were recorded on 16 characters, including plant height at four different times.The ANOVA's showed significant clone effects within each family for all characters. Computed from all family means as well as from the family means per ploidy level, differences due to family were also significant for all characters except one.As at the 2 x level and at the 4 x level of ploidy the mean phenotypic correlations between characters were of similar magnitude, it is concluded that they are independent of ploidy level.With the exception of eye depth, the mean coefficient of variation was greater at the 2 x than at the 4 x level of ploidy, indicating the greatest response to selection for those characters at the 2 x level. From the differences in family means between the 4 x and the 2 x level of ploidy it was apparent that the 4 x families generally had significantly taller plants, later maturity, fewer tubers, higher mean tuber weight, more tuber yield and more dry matter yield than their counterpart 2 x families. In contrast, the 4 x parents had on the average shorter plants, lower mean tuber weight, much lower tuber yield and lower dry matter yield than their 2 x counterparts.The phenotypic correlation and Spearman's rank correlation between the family means of the 2 x and the 4 x level of ploidy were positive for almost all characters and significant for nearly half of them.From the results it is concluded that 1. in potatoes the 4 x level of ploidy is preferable to the 2 x level, and 2. the performance of 4 x families is predictable from the performance of their counterpart 2 x families.Based on results mentioned in the literature and on the present results, a continued use of S. tuberosum dihaploids in potato breeding needs to be dissuaded.  相似文献

Summary Meiotic observations in PMCs were made in eight kinds of interspecific hybrids in Lilium. Three hybrids, 6134-S × L. cernuum. L. regale × L. leichtlinii maximowiczii and L. auratum platyphyllum × L. henryi showed respectively the mean chromosome association frequencies of 1.4II, 0.8II and 1.5II per cell at MI which were the lowest values hitherto reported in Lilium hybrids. In L. longiflorum × L. henryi, 33.3° of the cells had 1 or 2 multivalents of three to five chromosomes in addition to uni- and bivalents. 12° bivalents and 12 univalents were invariably observed in the triploid hybrid L. longiflorum × L. cernuum. The hybrids between the species belonging to the different sections of the genus generally showed high pollen sterility, with some exceptions. A more or less remote genomic homology was found between the different sections in Lilium.  相似文献
