In 1990, 1443 sera randomly selected by computer generated random numbers from a bank of greater than 15,000 sera collected earlier from sows at slaughter in various geographic regions of Canada were tested for anti-Toxoplasma immunoglobin G (IgG) by the modified direct agglutination test. One hundred and thirty-six (9.4%) samples were positive for anti-Toxoplasma IgG while 36 (2.5%) of the sera gave borderline reactions. The highest prevalence of anti-Toxoplasma IgG was in swine originating in the Atlantic provinces and Ontario while the lowest was in swine from Manitoba and Saskatchewan.  相似文献

The objective of the present study was to determine the prevalence of Toxoplasma gondii antibodies from slaughter sows and from pigs raised at an indoor and an outdoor swine farm. Serum samples were obtained from 230 slaughter sows belonging to 83 farms distributed in 5 provinces. Blood samples were collected monthly from pigs of different ages from an intensive management indoor farm (farm 1). A cross-sectional study was carried-out from an outdoor farm (farm 2). All sera were tested for T. gondii antibodies by the modified agglutination test (MAT), using formalin-fixed tachyzoites as antigen. An antibody titer > or =1:25 was considered positive. Antibodies to T. gondii were detected in 87 (37.8%) of 230 sows sera. Distribution among provinces was: 37.1% from Santa Fe, 62.8% from Buenos Aires, 3.3% from San Luis, 58.7% from La Pampa and 24% from Córdoba. Four of 88 (4.5%) serum samples from farm 1 had antibodies to T. gondii and none of the negative pigs seroconverted. However, 45 of 112 samples from farm 2 were positive (40.2%) with the following distribution: sows 100%; nursery 40%; growers 13.8% and fatteners 20%. It is concluded that the prevalence of T.gondii antibodies among sows seems to be quite variable. T. gondii prevalence was related to the facilities and management of the farm.  相似文献

为调查输血传播病毒(TTV)在我国猪群中的感染状况,本研究对2009年采自29个省市的1990份猪血清样品进行了TTV1、TTV2的双重PCR方法检测,并对结果进行了统计分析.结果表明,我国猪群中TTV总的感染率为63.37%(1261/1990),其中TTV1感染率为55.88%(1112/1990),TTV2为32.91% (655/1990),TTV1和TTV2双重感染率为25.43%(506/1990).进而对样品的地区性分布特征、样品来源等影响因素进行分析,表明我国猪群中TTV感染率以东北地区最高,西北地区最低.样品来源不是影响感染率的关键因素.  相似文献

Porcine circovirus type 2 (PCV2) can be vertically transmitted resulting in fetal infection with or without clinical signs and lesions. The primary objective of this study was to assess the prevalence of intrauterine PCV2 infection in clinically normal newborn piglets in conventional pork production facilities. Five commercial breeding herds located in the U.S. and Mexico were included in the study. A total of 125 sows and 3-5 neonatal piglets per sow were arbitrarily selected. Blood and colostrum samples were collected from sows. Blood was collected from piglets prior to suckling. All samples were analyzed for the presence of anti-PCV2 IgG antibodies and presence and amount of PCV2 DNA. In addition, PCV2 DNA positive samples were further subtyped into PCV2a and PCV2b. All (125/125) sow colostrum samples and 96.8% (121/125) of the sow serum samples and 21.4% (107/499) of the piglet pre-suckle serum samples were positive for anti-PCV2 IgG antibody. The overall PCV2 DNA prevalence was 47.2% (59/125) in sow serum, 40.8% (51/125) in sow colostrum, and 39.9% (199/499) in pre-suckle piglet serum. In the PCV2 DNA positive samples, PCV2b was detected at a higher frequency (69.5% for sow serum, 84.3% for sow colostrum, and 74.4% for piglet serum) compared to PCV2a (18.6% for sow serum, 9.8% for sow colostrum, and 15.6% for piglet serum). Concurrent PCV2a and PCV2b infection was detected in 11.9% of the sow serum, in 5.9% of the colostrum samples, and in 10.0% of the piglet serum samples. In conclusion, an unexpectedly high prevalence of PCV2 viremia was detected in healthy sows (serum and colostrum) and their pre-suckle piglets in the five breeding herds investigated and PCV2b was more prevalent than PCV2a. This information adds to the knowledge of PCV2 infection in breeding herds.  相似文献

A total of 2,614 swine from 104 herds located throughout Iowa were tested for antibodies against encephalomyocarditis virus (EMCV) by use of the microtitration serum neutralization test. The sample was composed of 587 sows and gilts and 2,027 finishing swine. A statistically significant (P less than 0.002) difference was observed between prevalence in sows and gilts (17.2%) and that in finishing swine (12.2%). Breeding swine maintained in total confinement (20.5%) had significantly (P = 0.04) higher prevalence than did breeders maintained in other types of housing (12.1%), whereas prevalence in finishing swine raised in total confinement (6.4%) was significantly (P = 0.02) lower that in finishers not raised in total confinement (13.6%). Association was not detected between prevalence and herd size or between prevalence and season of the year. Adjusting for test specificity and sensitivity, the true prevalence of EMCV infection in swine in Iowa was estimated to be 13.8% in breeding stock and 8.5% in finishing swine. On a herd basis, 89.4% (93/104) of the herds had one or more EMCV-positive swine.  相似文献

This research included 2 prevalence studies and a risk-factor investigation conducted in 2001 at 93 sites with sows only, finishers only, or both. In 2001, 1300 serum samples from sows in 65 herds and 720 serum samples from finisher pigs in 72 herds were tested for antibodies to swine influenzavirus (SIV) of H1N1 subtype with an enzyme-linked immunosorbent assay (ELISA). In 2003, 1140 serum samples from sows in 76 herds were tested for antibodies to SIV of H3N2 subtype with a hemagglutination-inhibition assay based on A/Swine/Colorado/1/77 and A/Swine/Texas/4199-2/98 isolates. The apparent pig-level H1N1 seroprevalence in 2001 was 61.1% and 24.3% in sows and finishers, respectively. The apparent pig-level seroprevalence in 2003 for H3N2 A/Sw/CO/1/77 and A/Sw/TX/4199-2/98 in sows was 0.6% and 0.7%, respectively. The factors associated with sow-herd H1N1 positivity included pig or farm density at different geographic levels, an external source of breeding pigs, number of animals on site, and decreasing proximity to other barns. Higher-parity sows had higher odds of seropositivity, but there was significant random variability in this association among herds. The odds of finisher-herd SIV positivity were higher with large herd size, high pig farm density, and farrow-to-finish type of farm. Finisher herds were SIV-positive only if source sow herds were positive. Simultaneously, 45% of finisher herds were SIV-negative although sow source herds were positive.  相似文献

An indirect fluorescent antibody (IFA) test was developed and standardized to detect and quantitate antibody for swine infertility and respiratory syndrome (SIRS) virus in swine sera. Test results were evaluated using sera of pigs infected both experimentally and naturally with SIRS virus. The IFA test used swine alveolar macrophage (SAM) monolayers prepared in 96-well microplates and infected with SIRS virus. The monolayers were incubated with test sera, washed, and stained with fluorescein isothiocyanate-labeled rabbit anti-swine IgG. After another wash step, the monolayers were examined under a fluorescent microscope. A noninfected SAM control well was included for each sample. The antibody titers for each serum sample were recorded as the highest serum dilutions with specific cytoplasmic fluorescence but no fluorescence in the control wells. To evaluate the test, sera of 4 6-week-old pigs that had been infected with SIRS virus, 2 contact pigs, and 13 experimentally infected sows were used. In the experimentally infected pigs, antibody was first detected at 7 days postexposure (PE) and peaked (1:256-1,024) between 11 and 21 days PE. All 13 sow sera were negative at time of infection but were positive (1:64- greater than or equal to 1:1,024) at 14-26 days PE. Seven hundred twenty sera collected from 25 different swine farms with or without a history of SIRS were also tested. Of 344 sera from 15 swine farms with a clinical history of SIRS, 257 (74.7%) sera had IFA titers greater than or equal to 1:4, whereas 371 (98.7%) of 376 sera from herds with no history of SIRS were negative.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Sera were collected from 6 large farrow-to-finish swine herds infected with pseudorabies virus (PRV) in Illinois. All herds were participating in the Large Herd Cleanup Study, a USDA-initiated project to evaluate the feasibility of eradicating pseudorabies from large farms (greater than 400 sows) by use of a combination of vaccination and management changes. Herd size ranged between 425 and 1,500 breeding females. Between April and July 1990, sera for measurement of PRV antibodies were obtained from 113 to 156 sows and 112 to 162 finishing pigs (body weight greater than 70 kg)/herd. Duplicate sera from 30 sows and 30 market-weight pigs/herd were obtained for measurement of serum antibodies to the following associated organisms: swine influenza virus, transmissible gastroenteritis virus, encephalomyocarditis virus, Actinobacillus pleuropneumoniae, Eperythrozoon suis, and 6 serovars of Leptospira interrogans. Prevalence of PRV antibodies attributable to field virus infection ranged between 53.8 and 100% for sows and between 0.7 and 97.3% for finishing pigs, as determined by the appropriate differential test for the vaccine being used on each farm. In only 1 herd, PRV seroprevalence was increased with higher sow parity. For associated infections, the risk of seropositivity attributable to PRV was not significant (for most infections) on all farms and varied among farms. Thus, pseudorabies did not appear, in general, to increase susceptibility to infection with other disease agents.  相似文献

Sera of pigs from élite breeding herds, of boars and sows collected at slaughter-houses, and of pigs from herds known to be infected, were examined for antibody to Aujeszky virus. The conventional and a modified virus-neutralizing antibody (VNA) test and a modified direct complement fixation (CF) test were employed. In simultaneous titrations of positive sera the modified VNA test gave titers approx. 4 log2 units above the titers obtained by the conventional test. The conventional VNA test was found insufficiently sensitive. Unspecific neutralization in the modified VNA test was infrequent in serum dilution 1/2 and rare in dilution 1/4. The GF tests on sera of slaughter sows and animals from known infected herds showed a remarkable consistency with the VNA tests. Inconsistent results were obtained with but few sera. Abt. 5 % of the sera could not be examined because of complement fixation with control antigen.  相似文献

Multiparous sows from 19 central Iowa swine farms were tested for antibodies against Toxoplasma gondii by the modified direct agglutination test. Antibody titers of 1:32 or greater were considered positive. Rodents, domestic cats, opossums (Didelphis virginiana), raccoons (Procyon lotor) and striped skunks (Mephitis mephitis) were live-trapped on each farm and similarly tested for antibodies. The overall prevalence of T. gondii antibodies in the species tested was 39/273 (14.3%) swine, 31/74 (41.9%) cats, 2/588 (0.3%) house mice (Mus musculus), 0/21 mice of the genus Peromyscus, 0/9 Norway rats (Rattus norvegicus), 1/34 (2.9%) opossums, 4/14 (28.6%) raccoons and 2/7 (28.6%) striped skunks. The overall prevalence was significantly greater in adult cats versus juvenile cats, adult male cats versus adult female cats, and adult raccoons versus juvenile raccoons. The prevalence of T. gondii antibodies in sows was compared with the prevalence in each non-swine species on a farm basis in order to identify existing associations. The prevalence in sows (and each of the non-swine species) was also analyzed on a farm basis for association with farm characteristics or swine management practices, including the degree of confinement of swine, population size and average parity of breeding female swine, estimated cat population size, and estimated mouse and rat abundance. Average titers of seropositive animals were compared on a species basis. The prevalence in sows which were totally and continuously confined was lower than that in sows which were not totally and continuously confined. The prevalence in sows from farms with an average parity of less than 2.0 was significantly lower than that in sows from farms with an average parity of 2.0 or greater. These results suggested that the prevalence of T. gondii antibodies in swine increased with age and that prevalence in swine could be reduced through total confinement. No associations could be established between prevalence in sows and prevalence in non-swine species or other farm characteristics/swine management practices. However, the high prevalence of T. gondii antibodies in cats suggested that fecal contamination of the environment by cats may be the most significant source of toxoplasmosis for swine. The extremely low prevalence of T. gondii antibodies in house mice suggested that this species was not an important source of T. gondii for swine in Iowa.  相似文献

Swine and free-living nonporcine species from 20 Iowa swine farms were surveyed for antibodies against encephalomyocarditis virus (EMCV). The microtitration serum neutralization test was used, and antibody titers ≥ 1:8 were considered positive. The overall prevalence in 267 sows in various groups was 37.8% (range 20-86%). The prevalence in sows maintained in total confinement was significantly lower than in sows not maintained in total confinement (p = 0.01). Prevalence in sows was not associated with other herd characteristics or with abundance estimates of rats and/or mice.

Free-living animals tested included 74 domestic cats (Felis domestica), 203 house mice (Mus musculus), 15 mice of the genus Peromyscus, nine Norway rats (Rattus norvegicus), 34 opossums (Didelphis virginiana), 14 raccoons (Procyon lotor), and seven striped skunks (Mephitis mephitis). Of these, the only seropositive animals were two domestic cats.

The results of this study failed to implicate the free-living species surveyed as important reservoirs of EMCV for swine, and suggested that swine themselves and/or some species not included in this survey are the main reservoir of EMCV for swine in Iowa.

Severe clinical signs of swine infertility and respiratory syndrome (SIRS) of unknown cause were observed in several Minnesota swine farms between November 1990 and March 1991. Forty-five lung samples of weak pigs were collected from 13 swine farms, and virus isolation was attempted using swine alveolar macrophage (SAM) cultures. A cytopathic virus was isolated from 19 lung samples collected from 6 different farms. Four pregnant sows were infected intranasally with a tissue suspension from which virus was isolated, and 4 6-week-old pigs and 2 contact pigs were infected intranasally with 1 of the isolates. The 4 sows farrowed 12 stillborn and 32 normal pigs. Virus was recovered from 10 of 19 pigs examined. Infected 6-week-old pigs were clinically normal except for slightly elevated rectal temperatures and mild respiratory signs. No or mild interstitial pneumonic lesions were observed in inoculated pigs, but the lesion was obvious in the 2 contact pigs. Seroconversion was observed in sows and pigs as measured by indirect fluorescent antibody (IFA). Serologic identification of the isolates was carried out by IFA using reference serum prepared from an experimentally infected sow. A cytoplasmic fluorescence was observed on the SAM monolayers infected with each of the 19 different isolates. Fluorescence was also observed when the monolayers were tested with SIRS virus ATCC VR-2332-infected sow sera. Replication of the isolates was not affected in the medium containing 5-iodo-2'-deoxyuridine but was inhibited by treatment with ether. Swine are one of significant reservoirs and sources of Leptospira infections for man. Serological screenings help to effectively control the epidemiological situation in swine herds and to prevent transmission of Leptospira from animals to man. The purpose of this study was to investigate, by the use of serological methods, the prevalence of infections caused by selected Leptospira serogroups in swine population in Poland. A total of 7112 swine serum samples were examined. The samples were collected from January to October 2008 and came from 280 counties situated in all 16 provinces of Poland. All sera were examined preliminary by enzyme-linked immunosorbent assay (ELISA) using heat-stable antigenic preparation. The samples positive or doubtful in ELISA were investigated by microscopic agglutination test (MAT) with use of serovars Icterohaemorrhagiae, Pomona, Canicola, Sejroe, Tarassovi and Of the collected sera examined by ELISA 73 (1.02%) samples were positive, 85 (1.20%)--doubtful and 6954--negative. Among ELISA-positive and doubtful sera 64 samples (coming from 14 provinces) were recognized in MAT as positive. Among MAT positive samples 42.19% of sera demonstrated titres with serovar Pomona, 32.81%--with Sejroe, 14.06%--with Icterohaemorrhagiae, 6.25%--with Tarassovi, 3.13%--with Grippotyphosa and 1.56% with Canicola.  相似文献

African swine fever among slaughter pigs in Mubende district, Uganda   总被引:1,自引:1,他引:0  
Owing to frequent reports of suspected outbreaks and the presence of reservoir hosts and vectors (warthogs, bushpigs and O. moubata ticks), African swine fever (ASF) is believed to be an endemic disease in Uganda. There have, however, been very few studies carried out to confirm its existence in Uganda. This study was carried out to describe the prevalence of ASF based on pathologic lesions and analysis of serum samples from slaughtered pigs during a suspected outbreak in the Mubende district of Uganda. The study was based on visits to 22 slaughterhouses where individual pigs were randomly selected for a detailed ante-mortem and post-mortem inspections. Sera were also collected for laboratory analysis. A total of 997 pigs (53.7% male and 46.3% female) were examined for lesions suggestive of ASF and sero-positivity of sera for ASF antibodies. The sera were tested using enzyme-linked immunosorbent assay (ELISA) and positive samples were further confirmed with an immunoblot assay. The results showed that 3.8% (38/997) of the pigs examined had clinical signs and post-mortem lesions suggestive of ASF. Two of 997 (0.2%) sera analysed were positive for ASF antibodies. Of the sub-counties investigated, Bagezza (12%) and Kiyuni (11%) had the highest prevalence of lesions suggestive of ASF based on ante- and post-mortem examination results, while Mubende town council (1.7%) had the lowest. This study found a low number of pigs (3.8%) with lesions suggestive of ASF at slaughter and an even lower number of pigs (0.2%) that were seropositive at slaughter, however a significantly higher number of pigs were slaughtered during the outbreak as a strategy for farmers to avoid losses associated with mortality.  相似文献

Competitive ELISA was applied to detect antibodies against bluetongue virus in sheep sera collected from different agro-climatic areas in Ethiopia. A total of 90 serum samples were tested and 42 (46.67%) were positive for bluetongue virus antibodies. A prevalence rate ranging from 9.67% for sheep sampled in the highland to 92.85% for sheep sampled in the lowland was recorded. The prevalence correlated with the probable distribution of the Culicoides vector. This is the first report indicating the presence of bluetongue virus infection in animals from Ethiopia.  相似文献

Encephalomyocarditis virus (EMCV) has been found on pig farms worldwide and can cause myocarditis in young pigs and reproduction disorders in sows. So far, clinical signs of EMCV have not been reported in the Netherlands. The aim of this study was to estimate the seroprevalence of EMCV infection in Dutch sow herds. A total of 277 Dutch sow herds were randomly selected, from which 3237 serum samples were collected. These samples were tested for EMCV antibodies using the virus neutralization test (VN test). The apparent prevalence of EMCV antibodies was 9.3% in the total sow population, and the apparent herd prevalence was 58.8%. An exact determination of the prevalence of EMCV infections in the Dutch sow population was not possible because the characteristics of the VN test under field circumstances were not known. However, Dutch sow herds seem to be infected with EMCV because the distribution of positive blood samples in the tested sow population was significantly different from that expected if random false-positive reactions had occurred.  相似文献

A survey for West Nile Virus (WNV) haemagglutination-inhibition (HI) antibody was carried out in humans and domestic animals. Human sera were collected from Ibadan, while the animal sera were collected from both Ibadan and Maiduguri. Out of 304 human sera tested, 123 were positive (40%). There was a higher prevalence of HI antibody in adults than children. Sex distribution of positive sera showed that 37% of males and 43% of females had WNV HI antibody. There was no significant difference in the prevalence of HI antibody in both sexes. On the 123 WNV HI positive sera tested, 104 (85%) and 78 (75%) had yellow fever and Potiskum HI antibody respectively. Monotypic WNV virus reactions were frequently found in children while polytypic reactions were frequently found in adults. A total of 200 animal sera were examined, 50 camels, 50 goats, 49 cattle and 51 sheep. The highest prevalence of HI antibody was found in camels (26%), followed by sheep (20%). Percentage of positive sera in other species were: goat (18%) and cattle (6%). Of the 35 WNV HI positive animal sera, 26 and 20% reacted with Yellow fever and Potiskum virus antigens respectively.  相似文献

Serological survey of bovine herpesvirus type 1 infection in China   总被引:2,自引:0,他引:2  
To understand the nationwide seroprevalence of bovine herpesvirus type 1 (BoHV-1) infection of cows in China, 1344 sera of dairy cows from 29 provinces and 765 sera from 6 herds in Hubei province were collected with stratified random sampling. Another 483 sera from imported cows were included. The serum antibody was tested by BoHV-1 gG ELISA. The results demonstrated that the overall nationwide seroprevalence was 35.8% (481/1344), while the prevalence for individual province ranged from 12.1% to 77.8%. Although each province had positive samples, the prevalence was clustered in areas based on the cow population size. In Hubei Province, the overall seroprevalence was 22.2% (170/765) while the prevalence for individual farms varied greatly from 0.0% to 41.5%. The sera from imported cows had a moderate prevalence of 21.7% (105/483).  相似文献
