Three groups of ewes were vaccinated with formalin inactivated, whole cell, aluminum hydroxide adjuvanted bacterins prepared from capsulated enteropathogenic Escherichia coli (EEC). Lambs born to and suckling these ewes, compared with lambs of nonvaccinated control ewes, were highly resistant to homologous EEC challenge exposure. Lambs of ewes vaccinated with products prepared from K99 antigen-positive, noncapsulated E coli were highly resistant to heterologous EEC challenge exposure. In both cases, lambs of vaccinated ewes had significantly (P less than 0.005) less morbidity and mortality, fewer challenge inoculum-type E coli per rectal swab evaluation, and had superior weight gains over a 4-day period. Immunoglobulin assay of 122 lamb sera (collected 12 hours after birth) failed to reveal any correlation between serum immunoglobulin values and morbidity or mortality. When tested by plate agglutination technique, using whole cell antigens, or by reverse radial immunodiffusion, using purified capsular antigens, colostral whey samples of vaccinated ewes did not have increased capsular antibody titers. The K99 serum antibody values of K99 antigen-vaccinated ewes were markedly higher than were those of ewes vaccinated with other bacterins or of control ewes.  相似文献

Pregnant ewes were vaccinated twice, seven weeks and three weeks before lambing, with a multivalent formalin-killed Escherichia coli vaccine containing an added K99, F41 antigen preparation. Lambs born to vaccinated and unvaccinated ewes were exposed to oral infection with E coli B44 (09:K30, K99, F41). All 10 lambs from vaccinated ewes were protected whereas all 10 control lambs developed severe diarrhoea and five died or were killed in extremis. In the following year, previously immunised ewes were given a single dose of the vaccine two weeks before lambing. Eleven of their 12 lambs were protected against a similar challenge, which caused the death of six of eight control lambs and severe diarrhoea in the two survivors. Higher levels of antibody to the K99, F41 preparation were detected by enzyme-linked immunosorbent assay in the serum and colostrum from vaccinated ewes and in the serum of their lambs when compared with similar samples from control ewes and lambs.  相似文献

Breeding ewes were immunised with clostridial vaccine using different inoculation schedules. Results, showing differences in the class of antibody produced, were heavily dependent on the vaccination regime used. Immunoglobulin M (IgM) levels were significantly lower in ewes given double doses of vaccine compared to ewes given a single inoculation or no treatment at all (P less than 0.01). Neonatal lambs showed significant de novo IgM production with interference in this antibody production in the lambs of ewes vaccinated with the clostridial vaccine (P less than 0.05). Immunoglobulin G1 (IgG1) levels were significantly increased in all lambs which had mothers vaccinated with the clostridial vaccine prior to or during pregnancy (P less than 0.025). The greatest quantity of IgG1 was transferred to lambs when their mothers were given a double injection with primary inoculation prior to conception and booster prepartum (P less than 0.025). No antigen specific immunoglobulin G2 (IgG2) was detected in the lambs. Ewes were also immunised with BSA and their isotype specific serum antibody response was compared with their respective lambs. There was no detectable anti-BSA IgM in the lambs of all groups of ewes though specific IgG1 antibodies could be readily detected in the lambs of hyperimmunised ewes. The efficiency of transfer was related directly to the ability of the adjuvant to maximise IgG1 production in the ewe. Although immunised ewes produced high levels of IgG2, this was not transferred passively to the lamb.  相似文献

Fifty-two Targhee twin-bearing ewes were used in a factorial arrangement of treatments to investigate the role of supplemental vitamin E (vit E); 0 (NE) vs 400 IU of vit E x ewe x (-1)d(-1) (E) and parainfluenza type 3 (PI3) vaccination; none (NP) vs PI3 vaccination (P) in immune function. Parainfluenza type 3 vaccination was used to evoke an immune response. Ewes receiving PI3 were vaccinated at 49 and 21 d before the expected lambing date. Ewes receiving vit E were orally dosed daily, 32 to 0 d before lambing. Blood was collected from ewes at the time of the initial PI3 vaccination and 4 h postpartum. Blood was collected from lambs (n = 104) at 3 d postpartum. Ewe and lamb sera were analyzed for anti-PI3 antibody titers, immunoglobulin G (IgG) titers, and vit E concentrations. Colostrum was collected 4 h postpartum and analyzed for IgG. The model for ewe and lamb analysis included the main effects of vit E and PI3, sex (lambs model only), and their interactions. No interactions were detected (P > 0.20) for any ewe or lamb variables. Serum anti-PI3 titers were greater (P < 0.01) in P ewes and their lambs than NP ewes and their lambs. Serum vit E concentrations were greater (P < 0.01) in E ewes and their lambs than NE ewes and their lambs. Colostral IgG titers and serum anti-PI3 titers did not differ (P > 0.20) between E and NE ewes. Serum IgG titers in E ewes and their lambs did not differ (P > 0.15) from IgG titers in NE ewes and their lambs. Lamb anti-PI3 titers did not differ (P = 0.76) between lambs reared by E and NE ewes. These results indicate that, although supplemental vit E to the ewe increased lamb serum vit E concentration, it had no effect on measures used in this study to assess humoral immunity in the ewe or passive immunity to the lamb.  相似文献

In 30 female goats hyperimmunized with bovine insulin, antibody concentrations in serum were determined by radioimmunoassay. Blood samples were taken from the newborn lambs immediately after birth and during the 12 weeks thereafter. Twin lambs were separated and reared in either a colostrum-fed or a colostrum-deprived group. Bovine insulin was added to the milk rations of colostrum-deprived lambs. At the age of 10 weeks, these kids were given a single dose of 1 IU of insulin in combination with complete Freund's adjuvant. Colostrum from nanny goats was also secured for further investigations. Most of the hyperimmunized adult goats responded with the development of insulin antibodies. Their kids were born without these specific immunoglobulins, but several hours after they were allowed to suckle colostrum, the antibody concentration reached values comparable with the values measured in the nanny's blood. Colostrum-deprived kids did not show an increase in binding capacity. The addition of insulin to the milk rations of bottle-fed kids did not act as a primer in antibody production. Immunoglobulins in blood serum of nanny goats and kids and in colostral whey belong mainly to the IgG class. These results indicate that in goats, maternal insulin antibodies are transferred from nanny to offspring only by colostrum.  相似文献

The transfer of immunoglobulin G (IgG) and immunoglobulin M (IgM) from ewe to lamb was quantitated to determine the occurrence of failure in passive transfer. Concentrations of IgG and IgM in ewe serum did not correlate with those in the colostrum. Colostrum from all ewes contained abundant amounts of immunoglobulins when compared with serum values, with IgG being selectively concentrated over IgM. Absorption through the intestinal tract of the lamb appeared to be a nonselective process, lacking predilection for IgG and IgM. All lambs tested 24 hours after birth absorbed colostral immunoglobulins to some extent; however, 13 (14%) of 91 clinically normal lambs demonstrated some failure of passive transfer. In contrast, failure of passive transfer was found in 27 (46%) of 59 lambs dying of natural causes between 24 hours and 5 weeks of age. Evidence presented emphasizes the importance of absorption of adequate amounts of immunoglobulins to enable the newborn lamb to survive the first few weeks of life.  相似文献

The effect of colostral antibodies on the immune response in lambs following adenovirus vaccination was studied. Young lambs of different ages, born from vaccinated ewes, were vaccinated with an inactivated and aluminium hydroxide-adsorbed sheep adenovirus vaccine. In a group of lambs vaccinated at 5–7 days of age, the titre of humoral antibodies declined in parallel to unvaccinated controls. In lambs vaccinated at 24–35 days of age, antibody titres to adenovirus stabilised and persisted after an initial fall. Cell mediated immunity, as measured by blastogenic responses of circulating lymphocytes, was stimulated in both groups, but higher numbers of IgG-producing cells became evident in the group of lambs vaccinated at 24–35 days of age.  相似文献

A 2 x 2 factorial arrangement of treatments with 78 mature ewes was used to evaluate the effects of supplementing the pregnant ewe's diet with high levels of minerals and vitamin E on immunoglobulin G (IgG) absorption by the lamb and whether any altered efficacy of IgG absorption was due to the colostrum or to the lamb. The ewes were estrus-synchronized in October and housed in wk 10 of gestation. In the final 7 wk of gestation, a grass silage-based diet, offered ad libitum, was supplemented with 500 g of a 19% CP concentrate, and from 1 wk later until lambing, half the ewes was offered 48 g of a mineral/vitamin supplement containing 6.5 g of Ca, 4.9 g of P, 5.9 g of Mg, 4.0 g of Na, 790 mg of Zn, 3.5 mg of Se, 40 mg of I, 200 mg of Mn, 20 mg of Co, and 40 IU of vitamin E. At birth, the lambs were allocated to one of four treatments in a 2 x 2 factorial arrangement, with lamb origin and colostrum origin as the two factors. The lambs born to ewes not offered the mineral supplement were fed colostrum obtained from their own dams or from ewes in the mineral-supplemented treatment, whereas lambs born to ewes given supplemental minerals were fed colostrum obtained either from their dams or from ewes in the control treatment. The ewes were milked at 1, 10, and 18 h postpartum and the lambs were fed using a stomach tube. A 5-mL blood sample was taken from each lamb at 24 h postpartum for IgG analysis. The level of fecal adhesion to the upper tail/rump area of the lamb was subjectively scored at 72 h postpartum. There was no difference in gestation length, lamb birth weight, colostrum yield, or IgG production (P = 0.16 to 0.82). When ewes were fed supplemental minerals, the serum IgG content of the progeny was lower than in their control counterparts (6.8 vs. 16.1 g/L; P < 0.001), regardless of whether the lamb received colostrum from ewes with or without access to supplementary minerals. The difference in serum IgG concentrations at 24 h postpartum was a direct reflection of a compromised efficiency in IgG absorption. The progeny of ewes with access to minerals had higher (P < 0.05) levels of fecal adhesion, which was not related to the origin of the colostrum, indicating altered digestive function in these lambs. We conclude, using the sheep as a model, that high mineral intakes in late pregnancy not only lower serum IgG concentrations in the lamb, but also that high mineral intakes result in the neonate being preprogrammed at birth so that it is born with a compromised ability to absorb colostral IgG.  相似文献

Subclinical mastitis was induced by inoculating one mammary gland in dairy ewes (n=8) with a Staphylococcus simulans isolate; control ewes (n=4) were included. The milk yield of inoculated glands decreased (P<0.001), the California Mastitis Test (CMT) score increased and the organism could be recovered from the inoculated glands. With time, there was significantly increased frequency of "hindering sucking" (P=0.016) and "head up posture" (P<0.001) in the control ewes. Infected ewes had a significantly increased frequency of "vocalisation" (P=0.013) compared to controls. There was a significant difference in the frequency of "sucking attempt" and "successful suck" (P<0.05) behaviours between lambs of the two groups. Lambs of the challenged ewes also showed significantly increased frequency and duration of these behaviours towards the uninoculated glands of their dams, rather than to the challenged glands (P<0.05); no such difference was evident for the lambs of control ewes. It was concluded that subclinical mastitis alters the sucking behaviour of both ewes and lambs.  相似文献

Influence of selenium on antibody production in sheep   总被引:3,自引:0,他引:3  
Three experiments were carried out, using sheep fed a marginally low selenium diet, to study the effect of selenium supplementation on the antibody response to tetanus toxoid and on the serum IgG concentration. Six groups of three six-month-old lambs were fed a basal diet containing 0.13 mg Se kg-1 supplemented with either 0.1, 0.5 or 1.0 mg Se kg-1, as sodium selenite or as selenomethionine. These animals generally showed enhanced antibody response to tetanus toxoid, parainfluenza-3 virus and Corynebacterium pseudotuberculosis, and their total serum IgG concentrations were higher than in unsupplemented control animals although few responses were statistically significant. In two field studies significantly higher titres to tetanus toxoid were detected in ewes injected with 100 mg selenium as barium selenate, although no influence on serum IgG concentrations was detected. Lambs from selenium supplemented ewes had significantly higher titres to tetanus toxoid than lambs from ewes in the control group. Dietary vitamin E supplementation had a similar effect on the antibody response to tetanus toxoid in ewes, though no additive effect was seen when vitamin E was given together with selenium.  相似文献

Polymorphs collected from ewes which had been infected with live staphylococci and immunised with a killed staphylococcal vacccine, had significantly enhanced capacity for phagocytosis of staphylococci compared with polymorphs from ewes which received only a killed vaccine. The numbers of viable bacteria were greatly reduced when blood serum or whey were incorporated in incubation mixtures compared to incubation mixtures containing only polymorphs and staphylococci. When serum was used in the incubation mixtures, the numbers of staphylococci which survived were significantly less than when colostral or milk whey was used. No significant difference was observed between whey collected from locally immunised mammary glands and that from non-immunised glands within each group.  相似文献

Ewes carrying twin fetuses were maintained during late pregnancy on a uniformly high plane (n = 22), a uniformly low plane (LP) (n = 24) or a low plane increasing to a high plane (n = 14) of nutrition. Seven ewes at each nutritional level were killed at 142 days of gestation and the liver and muscle glycogen and body lipid concentrations of the fetuses were determined. The rest of the ewes lambed naturally at about 145 days and the colostrum yields were obtained by hand milking after oxytocin injections during the first 18 hours after birth. The lactose, lipid and protein concentrations of colostrum were determined. Undernutrition in the LP group did not apparently affect the body concentrations of available glycogen, reduced available body lipid by about 47 per cent and reduced the lactose, lipid and protein available in colostrum during the first 18 hours by about 50 per cent. Refeeding previously underfed ewes to a high plane during the last five to 10 days of pregnancy did not improve the available reserves of glycogen or lipid in the lambs but did increase the yields of colostral constituents by about 30 per cent. The relative contributions of body reserves and colostral constituents to the maintenance of high, medium and low metabolic rates in lambs from different groups was assessed. It was concluded that under normal field conditions refeeding undernourished ewes during the last weeks of pregnancy would improve only marginally the survival potential of the lambs and that most lambs would be compelled to draw on their body glycogen reserves in order to maintain heat production during the first 18 hours after birth, even when they consumed all the available colostrum.  相似文献

Colostral and milk whey rotavirus (RV) and coronavirus (CV) antibody titers stimulated in 15 beef heifers by vaccination with a modified live-RV-CV vaccine were compared with titers in 15 nonvaccinated heifers. Geometric mean antibody titers to RV in colostral and 3-day milk whey from vaccinated heifers were 2,807 and 92, respectively, and in control heifers were 1,613 and 71, respectively. Geometric mean antibody titers to CV in colostral and 3-day milk whey of vaccinated heifers were 877 and 13, respectively, compared with titers were 877 and 13, respectively, compared with titers in nonvaccinated heifers of 731 and 7, respectively. Differences in antibody titers between vaccinated and nonvaccinated heifers were not significantly different.  相似文献

Lambs born to 5 ewes in 3 successive years were studied for presence of maedi virus and its antibodies. In the middle of the first-year pregnancies the ewes and the only ram of the colony were inoculated with maedi virus. No antibodies or viraemia could be detected in the lambs at birth. After sucking colostrum, antibodies appeared in the lambs of the ewes which themselves were seropositive, and reached their peak in a few days. Maternal antibodies disappeared within 12 weeks in all the lambs. Neutralizing antibodies were demonstrated in the colostrum and their content declined rapidly after lambing. Virus was isolated from the milk of 2 ewes in the third year of the studyIn the first year the spread of maedi virus was demonstrated to only 1 of the lambs, but in the other 2 years maedi virus was detected in tissues of half of the lambs sacrificed at 3–12 weeks of age. It was concluded that lambs born to chronically infected ewes are readily infected, indicating excretion of virus by ewes. The study yielded no information on the specific routes of transmission, except for the finding of the virus in milk of 2 ewes in the third year of the study. No evidence was obtained of transplacental transmission of maedi.  相似文献

New-born calves, artificially fed colostrum or native colostral whey, either dried or preserved by another method, had good health and good weight gains (between 0.05 and 0.60 kg). No greater differences were observed between the groups of calves given three times the colostrum of their mothers, calves given mixed colostrum, and calves fed colostral whey powder. In all groups only individual differences in IgG content in the blood serum were observed after 48 hours from birth. Hypogammaglobulinaemia occurred in individual cases both in calves given small amounts of colostrum or colostral whey and in calves given sufficient quantities. The time that had elapsed from birth to the first drinking did not exert any greater influence upon the IgG level in the blood; the decisive factor was the amount of colostrum taken in by the calf in the first dose. The rate of the absorption of IgG1, IgG2, IgA, and IgM from colostrum and the concentration of the immunoglobulins in the serum depended on the quantity of colostrum in the first dose and were not influenced to any greater degree by the amount of colostrum given to the calves in further doses. The amount of IgG in the blood serum of calves corresponded approximately to the level of colostral antibodies to the virus PI-3. The antibodies to the virus PI-3 and small quantities of IgG were observed also in the serum to new-born calves before drinking colostrum.  相似文献

Lambs sucking non-immunised ewes or ewes immunised 4-5 weeks before lambing with live attenuated, aromatic-dependent (aroA) Salmonella typhimurium (strain CS 332) were challenged orally at either 2, 4 or 7 days of age with virulent S. typhimurium (strain CS 94) at doses ranging from 109 to 1013 colony forming units. No lambs displayed signs of clinical salmonellosis and all survived challenge but those sucking immunised ewes had organisms of the challenge strain in their faeces for much shorter periods of time than lambs of the control ewes. High titres of specific antibodies were measured in colostrum and milk of immunised ewes in comparison with very low titres measured in samples from control ewes; these differences were reflected by the titres of antibodies in the sera of corresponding lambs. At 2 days after lambing, the major antibody isotype in the colostrum of immunised ewes and sera of their lambs was IgM whereas at 7 days IgG1 was the predominant isotype. While it was clear that vaccination of pregnant ewes with the live attenuated vaccination conferred protection against experimentally-induced salmonellosis in their lambs, considerable protection was observed in control lambs in spite of there being very low titres of antibodies in the mammary secretion of their dams. The latter observation could be related to the presence of contain non-antibody potent bactericidal factors previously described in colostrum and milk.

Des agneaux qui tètent des brebis non immunisées, ou bien immunisées, 4 à 5 semaines avant la mise bas avec un vaccin atténué constitué d'une souche de Salmonelle typhimurium dépendente pour sa croissance de la presence de composés aromatiques (souche aro A CS 332) ont reçu, à l'âge de 2, 4 ou 7 jours, par voie orale, des doses allant de 109 à 1013 Salmonella (souche virulente CS 94). Aucun agneau n'a présenté de signes cliniques et tous ont survécu à cette administration, mais les agneaux des brebis immunisées ont excrété la souche d'épreuve dans leur fécès pendent moins longtemps que les agneaux des brebis non vaccinées. Des titres plus élevés d'anticorps sont détectés dans le colostrum et le lait des brebis vaccinées et comparés aux brebis témoins. Cette différence est également notée pour les titres d'anticorps présents dans le sérum des agneaux issus de brebis vaccinées ou non. Deux jours après la mise bas, la classe des anticorps majoritaires, tant au niveau du colostrum des brebis vaccinées que du sérum de leurs agneaux, est la classe des IgM, alors qu'après 7 jours, les IgG1 prédominent. Bien qu'il soit clair que des brebis pleines vaccinées à l'aide d'un vaccin vivant atténué, apportent à leurs agneaux une protection vis à vis d'une Salmonella expérimentale, une réelle protection est également observée chez les agneaux témoins, malgré les titres faibles d'anticorps présente dans les sécrétions mammaires de leur mères. Cette dernière observation peut être mise en rapport avec la présence d'agents bactériodes ne contenant pas d'anticorps, déjà décrits dans le colostrum et le lait.  相似文献

To investigate the influence of maternal Se supply and plane of nutrition on lamb morbidity, mortality, and passive transfer of IgG, pregnant ewe lambs were used in 2 experiments with 2 × 3 factorial treatment arrangements. Supplementation of Se began at breeding and was either adequate Se (ASe, 9.5 μg/kg of BW) or high Se (HSe, 81.8 μg/kg of BW) in Exp. 1 or ASe (11.5 μg/kg of BW) or HSe (77.0 μg/kg of BW) in Exp. 2. On d 50 or 40 of gestation for Exp. 1 or 2, respectively, ewes were assigned randomly to 1 of 3 nutritional planes: 60% (RES), 100% (control, CON), or 140% (HI) of NRC requirements. This resulted in the following treatments: ASe-RES, ASe-CON, ASe-HI, HSe-RES, HSe-CON, and HSe-HI. Upon parturition, lambs were separated from their dams and serum samples obtained. Lambs were fed artificial colostrum for the first 20 h and then placed on milk replacer and grain pellets until completion of the study (Exp. 1, 57 d; Exp. 2, 21 d). Twenty-four hours after parturition, lamb serum samples were collected for IgG analysis. All lambs were reared similarly and morbidity and mortality assessed. Main effects were considered significant when P ≤ 0.05. In Exp. 1, there was a Se × plane of nutrition interaction (P ≤ 0.01) for lamb morbidity from birth to weaning and for 24-h IgG concentration. Lambs from ASe-RES and HSe-HI ewes were treated more frequently (P < 0.01) for respiratory and gastrointestinal disease, and lambs from HSe-HI ewes had the smallest (P < 0.01) 24-h serum IgG concentration. In Exp. 1, lambs from HI ewes also had the greatest (P < 0.01) mortality rates from birth to weaning compared with lambs from CON and RES ewes. In Exp. 2, there was an effect (P < 0.01) of maternal plane of nutrition with lambs from RES ewes having increased 24-h IgG compared with lambs from CON and HI ewes. There was no effect of maternal Se supplementation on lamb 24-h IgG in Exp. 2; however, there was a Se × plane of nutrition interaction (P < 0.01) for morbidity. From birth to 21 d of age, lambs from ASe-CON ewes had fewer (P < 0.01) treatment days compared with lambs from any of the other treatment groups. There also tended (P = 0.08) to be an effect of maternal Se supplementation on lamb mortality with increased mortality observed in lambs from HSe ewes. Results from the studies show a restricted maternal plane of nutrition can increase lamb serum IgG concentration. Selenium results were not consistent between the 2 experiments and may be due to differences in maternal Se.  相似文献

Oral inoculation of approximately 1.2 x 10(9) viable Escherichia coli to pregnant cows resulted in increased blood serum and colostral whey titers to the "O" antigen. The antibody titers were more pronounced in colostral whey and were correlated with the inoculum strain of Escherichia coli. There was no correlation between antibody titers of the colostrum ingested and the resulting serum antibody titers of the calves. The incidence of diarrhea in calves did not correlate with the antibody titer in the colostrum. The occurrence of diarrhea was significantly greater in calves that did not ingest colostrum until they were 12 hours old, compared with calves that had free access to their dams and suckled within an hour of birth.  相似文献

Neonatal lamb mortality is a major factor affecting profitability in the sheep industry, and lamb thermogenesis is a key element in neonatal lamb survival. Increased lamb vigor has been reported when ewes were supplemented during late gestation with algae-derived docosahexaenoic acid (DHA); however, the effects of DHA on lamb thermogenesis and immunocompetence have not been investigated. Eighty twin-bearing Targhee ewes (ages 2 to 5 yr; 68.5 ± 3 kg) were assigned randomly to 1 of 2 supplement treatments to determine the effects of feeding DHA to ewes during late gestation and early lactation on lamb thermogenesis, serum metabolites and hormones, and lamb growth. Supplement treatments were 12 g·ewe(-1)·d(-1) of algae-derived DHA (DHA Gold Advanced Bionutrition Corp., Columbia, MD; algae-derived DHA); and no algae-derived DHA (control). Supplements were individually fed daily during the last 30 d (±7 d) of gestation and pen fed (6 pens/treatment with 6 or 7 ewes/pen) during the first 38 d (±7 d) of lactation. One hour after lambing and before nursing, twin-born lambs were weighed, blood sampled via jugular puncture, and placed in a dry cold chamber for 30 min (0°C), and rectal temperatures were recorded every minute for 30 min. Lambs were removed from the cold chamber, blood sampled, warmed for 15 min, and returned to their dam. Ewes were blood sampled, and colostrum samples were collected 1 h postpartum. Ewe and lamb sera were assayed for glucose, NEFA, cortisol, and leptin. Lamb rectal temperature, glucose, NEFA, cortisol, leptin, and birth weights did not differ between treatments. The BW at 38 d was greater (P = 0.03) for lambs born to control ewes than for lambs born to algae-derived DHA-supplemented ewes; however, the colostrum of algae-derived DHA-supplemented ewes had a greater specific gravity (P = 0.05) than for control ewes. Overall, despite a potentially positive effect on ewe colostral IgG concentrations, supplementation of algae-derived DHA during late gestation and early lactation had a negative effect on lamb BW and did not affect indices of lamb thermogenesis.  相似文献

OBJECTIVE: To evaluate the immunologic response of a killed tachyzoite vaccine against Neospora caninum and its effectiveness in preventing vertical transmission of N caninum in sheep. ANIMALS: 40 Dorset ewes seronegative for N caninum. PROCEDURE: Group-A ewes (n = 20) were vaccinated on days 1 and 126 with a killed N caninum tachyzoite preparation in a commercially available adjuvant. Group-B ewes (n = 20) were sham vaccinated. Blood samples were collected from ewes every 2 weeks and a recombinant ELISA (rELISA) was used to determine serum antibody titers against N caninum. During pregnancy, ewes were challenged with live N caninum tachyzoites. Precolostral serum was collected from lambs and tested for antibodies against N caninum by use of an indirect fluorescence antibody test and the rELISA. Tissue specimens from stillborn lambs or lambs that died within 2 weeks of birth were collected and examined for N caninum antigen and DNA by use of immunohistochemistry and polymerase chain reaction assay, respectively. RESULTS: Serum antibody titers against N caninum were significantly higher in group-A ewes, compared with group B ewes, following vaccination. Serum antibodies against N caninum were detected in 100% (33/33) of group-B lambs and 75% (18/24) of group-A lambs. In tissue specimens, N caninum DNA was detected in 9 of 11 group-B lambs and 0 of 10 group-A lambs. Histologically, N caninum tachyzoites were observed in 4 group-A lambs and 3 group-B lambs. CONCLUSIONS AND CLINICAL RELEVANCE: The killed tachyzoite vaccine against N caninum stimulated a humoral immune response in sheep and provided partial protection against vertical transmission.  相似文献
