The presence of ESBL/AmpC-producing Escherichia coli in livestock such as pigs has been known for some time. However, to date there is little information about the transmission of these resistant bacteria between pig farms and their surroundings. Thus, the aim of this study was to explore this topic by investigating seven German pig fattening farms. Samples from outside (including ground surfaces, ambient air, slurry and digestate from biogas plants) and, in parallel, from inside the pig barns (including pig feces, dust, barn air, flies and mice feces) were examined for ESBL/AmpC-producing E. coli and selected isolates were compared by pulsed-field gel electrophoresis (PFGE) analysis. 14/17 (82.4%) slurry samples and three of four samples of digestate from biogas plants tested positive for ESBL/AmpC-producing E. coli. In the vicinity of the pig barns these resistant bacteria were detected in 14/87 (16.1%) boot swabs taken from various ground surfaces and in 2/36 (6%) ambient air samples. Inside the pig barns, 6/63 (9.5%) barn air samples and a small proportion of flies and mice feces samples were ESBL/AmpC-positive. PFGE analysis proved fecal emission as well as a possible spread via flies, as identical ESBL-E. coli isolates were detected in slurry and on fertilized fields, as well as in flies and pooled feces from inside the barn and slurry. Contaminated slurry presented the major emission source for ESBL/AmpC-producing E. coli in the pig fattening farms, but a spread via the airborne route or via different vectors also seems possible.  相似文献

A high prevalence of MRSA among farm animals, especially pigs, has been observed for some time. However, knowledge on transmission routes of MRSA in livestock production is still scarce. Therefore, the aim of this study was to determine the occurrence of MRSA in pig house air as well as in samples from pigs and their housing environment in 27 MRSA positive pig barns of different sizes and production types. In 85.2% of all barns MRSA was detected in the animal house air. Impingement turned out to be a more sensitive sampling technique than filtration. Other environmental samples such as boot swabs or faeces showed prevalences of MRSA from 55.6% to 85.2% at sample level. The level of MRSA was 88.3% for pooled and 82.1% for single nasal swabs, in skin swabs the one was 87.7%, the others was 78.7%. Spa typing of isolates from air and nasal swabs showed predominantly spa types t011 and t034. MRSA prevalences in pigs as well as in various environmental samples were significantly higher in fattening farms than in breeding farms. This study provides good reference that there could be an airborne transmission of MRSA within pig herds indicating a potential contamination of the environment of barns.  相似文献

In this work, faecal samples were collected from 15 pig farms to determine the Campylobacter prevalences at different times during the rearing period and to visualize the exchange of strains among the pig population by genotyping specific isolates. All isolated strains were identified as C. coli. Whereas no Campylobacter were detectable in the faeces of piglets at the day of birth, the Campylobacter incidence rose within days to 32.8%. After transfer to the nursery unit the prevalence increased to 56.6%. Approximately two-thirds of the pigs remained C. coli shedders in the fattening unit. In contrast to most farms, one farm expressed a very low Campylobacter incidence during the whole rearing period. Amplified fragment length polymorphism (AFLP) analysis was performed on all C. coli isolates of one farm. Clonal strains were identified from the brood sows and their offsprings or neighbouring piglets. After moving to the nursery unit, new genotypes appeared in that pig group but the original C. coli strains largely remained within that group. C. coli genotypes, identified during the fattening period, replaced the previously isolated genotypes. Transportation to the abattoir had no significant influence on the shedding rate of C. coli. The detection rate before transportation was 79.1% and decreased slightly to 78.2% (n=330). Additionally, eleven of 1474 environmental samples from different sources of the pig farms were positive for C. coli. This study demonstrates the importance of pigs as a reservoir for C. coli. Maternal C. coli strains are the primary source of infection but non-related genotypes from different sources appear during the rearing period and these latter strains constitute largely the final C. coli flora.  相似文献

A total of 142 stool specimens from pigs on 24 farms from the province of Zaragoza (northeastern Spain) were screened for Cryptosporidium spp. Samples were first analysed by routine techniques (formalin-ethyl acetate sedimentation method and modified Ziehl-Neelsen stain) selecting those microscopically positive for genetic characterization. Cryptosporidium species and genotypes were determined by a nested PCR-RFLP technique at the 18S ribosomal DNA locus and sequencing of the PCR-positive secondary products. Cryptosporidium oocysts were microscopically identified in the faeces of 32 pigs (22.5%) from 15 farms (62.5%). Infected animals included 23 weaned piglets (30.7%), 5 fattening pigs (11.9%) and 4 sows (16%). Diarrhoea was not detected in any of the infected pigs. The molecular characterization was successfully performed in 26 samples from 14 farms. Cryptosporidium suis was found in 10 specimens from 7 farms (nine weaned piglets and one sow) and the Cryptosporidium pig genotype II in 16 samples from 10 farms (13 weaned piglets and 3 fattening pigs). Various methods have been described in the literature for the detection of virulent Yersinia enterocolitica in pigs. The risk factors for pig herd contamination have yet to be determined. The objective of this study was to validate a sensitive method for the detection of Y. enterocolitica and to describe the distribution of the bacteria in pigs at slaughter from conventional and alternative ("organic") housing systems. First, samples were collected from tonsils, caecum with caecal contents, and the caecal lymph nodes of 60 slaughter pigs. These samples were used to compare the sensitivity of six different laboratory culture methods either in common use or described in the literature with that of a polymerase chain reaction with two primer pairs (multiplex PCR). Then, only PCR was used to examine tonsils, caecum and caecal lymph nodes from two groups of slaughter pigs: 210 from six conventional fattening farms and 200 from three with alternative housing. The results of the multiplex PCR were positive in 28 cases. All culture methods proved inferior to PCR in sensitivity. In the second part of the study, PCR detected 36 (18%) positive pigs from alternative housing and 60 (29%) from conventional housing (p<0.05). The highest rate of Y. enterocolitica contamination was found in tonsils (11% alternative, 22% conventional; p<0.05), followed by caecum (5%, 11%) and lymph nodes (2%, 7%). The housing system appears to be one important factor in the prevalence of this common pathogen in pig herds, as we found important differences between the two systems studied here. In the conventional system, the main risk factors appeared to be sourcing pigs from different pig suppliers, use of commercial feed and transportation to slaughter.  相似文献

Verocytotoxin-producing Escherichia coli O157:H7 (VTEC O157:H7) was detected in two of 2446 individual faecal samples collected from pigs slaughtered at five Swedish slaughterhouses, indicating a prevalence of 0.08 per cent, with a 95 per cent confidence interval from 0 to 0.16 per cent Four Swedish VTEC O157:H7-positive farms which kept ruminants and pigs were studied by repeated faecal sampling; VTEC O157:H7 was isolated from the ruminants and pigs on all the farms and the same strains were present in the pigs and the ruminants. On one of the farms, the organism persisted in the pig population for 11 months. On all four farms, management practices which might have influenced the isolation rate in pigs were identified. A group of young VTEC O157:H7-positive pigs was moved from one of the VTEC O157:H7-positive farms to a fattening herd where there were no ruminants. The number of VTEC O157:H7-positive faecal samples decreased gradually and after nine weeks the pigs were all negative; at slaughter none of the pigs was VTEC O157:H7-positive.  相似文献

为了解黑龙江省东部地区猪蓝耳病(PRRS)的流行情况,从黑龙江省东部地区的牡丹江市、宁安市、海林市、绥芬河市、鸡西市、鸡东市、佳木斯市、密山市、虎林等地,共采集猪血清样本663份。采样猪群包括公猪、母猪、仔猪、育肥猪。采集的猪场包括规模化猪场、小规模的猪场及散养户。应用ELISA方法对该省东部地区猪蓝耳病的流行情况进行血清学调查,结果显示:663份血清中,其中,134头为阳性,阳性率为20.21%;529头为阴性,阴性率为79.79%;S/P值大于2.0的26头,占总阳性比率的19.40%。  相似文献

The helminth infections on 13 pig fattening farms with different management systems (complete or partial all-in-all-out system or continuous fattening) in North-Western Germany were investigated over at least three fattening periods. Pooled faecal samples were taken from pens once before and three times after anthelmintic treatment. At the beginning of fattening 34.9% of the samples contained helminth eggs, mainly from Oesophagostomum spp. (27.5%). Ascaris suum eggs were found in 10.5% of the samples, while other parasites were only rarely found. The number of pig-supplying farms was positively correlated with the helminth infection prevalence. Immediately after deworming, all pen samples were free of helminth eggs. However, the prevalences increased again, and by the end of fattening A. suum was found in 33.0% and strongylids in 6.0% of the samples. Pens harbouring A. suum-excreting pigs at the beginning of fattening had higher infection levels at the end, and this was also the case for nodular worms. The final prevalence of Ascaris was higher in partial exchange systems than in complete all-in-all-out systems and in old pig houses compared to new ones. Transmission of both Ascaris and Oesophagostomum was highest in autumn and winter. Thus, a single anthelmintic treatment at the beginning of fattening could not prevent infection during fattening, and the state of infection at the beginning was associated with the helminth burden at slaughter. Therefore, the purchase of parasite-free pigs in combination with appropriate hygiene management may minimise the initial infection pressure and keep subsequent infection of the herd at a minimum.  相似文献

为了解2016年1月~2018年12月期间江苏省规模猪场猪繁殖与呼吸障碍综合征(porcine reproductive and respiratory syndrome,PRRS)流行动态,本研究对该省27家规模猪场不同批次送检的2062份未免疫PRRS疫苗血清样本,采用ELISA方法进行了猪繁殖与呼吸障碍综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)抗体检测,并对抗体阳性率≥80%的猪群阶段,依据PRRSV抗体S/P分布频段选取135份血清,采用RT-qPCR方法检测并统计PRRSV阳性率,进行血清学调查。结果显示,27家未免疫PRRS疫苗猪场抗体总阳性率为68.82%。不同地区抗体阳性率较高的为宿迁和南通,分别为72.04%和71.09%,连云港地区最低为58.82%。2016~2018年呈现逐年下降的趋势,从76.41%下降至57.72%。不同猪群阶段中以公猪、后备母猪、12~16周龄育肥猪和18~25周龄育肥猪抗体阳性率较高,分别为80.95%、92.86%、86.84%和98.09%。在上述4个抗体阳性率≥80%的猪群阶段,RT-qPCR检测结果显示,24份PRRSV阳性血清样本C t值与抗体S/P值高低无相关性。血清中PRRSV阳性率以后备母猪最高为20.58%,在PRRSV抗体S/P值<0.4和S/P值≥2.5异常分布频段,血清PRRSV阳性率均≥15.38%,而0.4≤S/P值<2.5正常范围内PRRSV阳性率均≤10%,差异显著,具有统计学意义(P<0.05)。表明江苏省规模化猪场不同地区、不同规模和不同阶段猪群普遍存在 PRRSV 感染,该研究结果为规模猪场PRRS的防控提供了参考。  相似文献

This article presents the results of a study on the occurrence of two bacteria that cause zoonoses, Campylobacter spp. and Yersinia enterocolitica. The study was carried out in 30 fattening herds in Lower Saxony, Germany, in 2004 and compares the results of bacteriological and serological methods of detection. Bacteriological findings of Campylobacter spp. in the faeces indicated that 69.7% of the fattening pigs were positive, but 81.2% tested positive serologically. All herds tested here were both bacteriologically and serologically positive for Campylobacter spp. Furthermore, only 8.4% tested positive for Yersinia enterocolitica in the faecal samples, but 66.8% of the animals were serologically positive for that bacterium. While bacteriological examination did not detect Yersinia enterocolitica in 56.7% of the herds tested, serological testing showed that only 16.7% of the units were without reacting animals. The great difference between the results of bacteriological and serological testing, especially in the case of Yersinia enterocolitica, can be explained by the intermittent intestinal excretion and predominance of this bacterium in the animals' tonsils. Low faecal excretion is also the reason for the low detection rate of 3.4% of Yersinia enterocolitica in the environmental samples, while that of Campylobacter spp. was 33.3%. These results indicate that the environment plays only a secondary role in the distribution of Yersinia enterocolitica in pig herds.  相似文献

We investigated characteristics of Yersinia enterocolitica infection in Ontario finisher pig herds. Our specific objectives were to estimate or test: prevalence of Y. enterocolitica shedding in finisher pigs, bioserotype distribution, agreement between the herd-level tests based on sampling pig and pooled fecal samples, whether bioserotypes cluster by farms, and whether Y. enterocolitica-positive herds cluster spatially. In total, 3747 fecal samples were collected from 100 farms over the years 2001, 2002, and 2004 (250 total herd visits). Fecal samples were tested by culture and positive isolates were biotyped and serotyped.Apparent pig-level prevalence of Y. enterocolitica was 1.8%, 3.2%, and 12.5% in 2001, 2002, and 2004, respectively. Estimated true pig-level prevalence of Y. enterocolitica was 5.1%, 9.1%, and 35.1% in 2001, 2002, and 2004, respectively. Herd-level prevalence was 16.3%, 17.9%, and 37.5% in 2001, 2002, and 2004, respectively. In all years, the most common bioserotype was 4, O:3, followed by bioserotype 2, O:5,27. Kappa between herd-level status based on pig and pooled samples ranged between 0.51 and 0.68 for biotype 1A and bioserotype 4, O:3, respectively. For 4, O:3, a significant bias in discordant pairs was detected, indicating that pig samples were more sensitive than pooled samples in declaring a herd as positive. Farms tended to be repeatedly positive with the same bioserotype, but positive study farms did not cluster spatially (suggesting lack of between herd transmission and lack of a common geographic risk factor).  相似文献

The present study is the first conducted in Spain to estimate the bacteriological herd prevalence of Salmonella enterica in fattening units and to describe the Salmonella serovar diversity on these farms using a sample representative of the entire swine population. For this purpose, 10 faecal samples were collected from 10 different pens containing pigs close to market weight in a total of 232 fattening units. Total sample size was proportionally distributed according to the fattener census in each of the regions of the country and all the samples were examined by culture of 25 g of faecal material. One hundred (43.1%) farms had at least one Salmonella-positive sample (95% CI: 37-49.1%). Salmonella enterica was detected in 290 (12.5%) pooled faecal floor samples (95% CI: 11.2-13.8%). The apparent herd prevalence of salmonellosis was similar among multi-site, finishing and farrow to finish farms. Overall, 24 different serovars were identified, with S. Typhimurium, S. Rissen and S. Derby being the most common both at herd and sample level. Results of phage typing were available for the 91 isolates of S. Typhimurium. A total number of 10 different phage types were identified, with DT 193 being the most frequent. Phage types DT 104, DT 104b and DT U302, which have been associated with several multi-resistant patterns, accounted for 23% and 29% of the Typhimurium total isolates or Typhimurium infected farms respectively.  相似文献

The objective of this study was to describe the distribution of Salmonella spp. on Ontario grower-finisher pig farms. Eighty swine farms were visited from January through July 2004. On each farm, fecal samples were collected from 5 pens, 2 rectal samples and 1 pooled sample from fresh manure on the floor per pen. Salmonella was isolated from 91 (11%) of the 800 rectal samples and 73 (18%) of the 397 pooled samples. Overall, Salmonella was recovered from 37 (46%) of the 80 farms. On each positive farm, Salmonella was cultured from 1 to 7 pigs or 1 to 5 pens. Of the 37 farms, 18, 13, 5, and 1 yielded 1, 2, 3, and 4 serovars, respectively. The most common serovars were S. Typhimurium var. Copenhagen, S. Infantis, S. Typhimurium, S. Derby, S. Agona, S. Havana, and S. enterica subsp. I:Rough-O. The 3 most frequent phage types were PT 104, PT 104a, and PT 104b. There was a statistically fair agreement between samples collected directly from pigs and pooled pen samples in determining the Salmonella status at the pen and farm level (kappa = 0.6, P < 0.0001). However, in 62 pens, Salmonella status, serovars, or phage types differed between the pig and pooled pen samples. The distribution of Salmonella on the swine farms in this study indicates that, in developing an intervention strategy, priority should be given to farms positive for S. Typhimurium var. Copenhagen. Also, the variation in Salmonella status between pig and pooled pen samples deserves consideration in a sampling strategy.  相似文献

A survey of Salmonella was carried out in fecal samples of 887 pigs with diarrhea collected from 235 pig farms between April 1996 and March 2001. Salmonella was isolated from 84 feces (9.5%) of 887 pigs and from 45 (19.1%) of 235 farms. The higher prevalence was found in weaned pigs (12.4%) and fattening pigs (17.3%) than in sows (4.2%) and suckling pigs (4.5%). Isolation rates of S. Typhimurium were higher from weaned and fattening pigs than from the others. Therefore, risk of horizontal infection of S. Typhimurium will increase, if no adequate health managements are practiced when weaned and fattening pigs have diarrhea.  相似文献

During the last few years, methicillin‐resistant Staphylococcus aureus (MRSA) ST398 has been isolated frequently from livestock, especially from pigs and to a lesser extent from cattle and poultry. To gain insight into the distribution of this bacterium in pig farms versus multispecies farms, 30 Belgian farms (10 pig, 10 pig/poultry and 10 pig/cattle farms) were screened for the presence of MRSA. On each farm, 10 nasal swabs were taken from pigs. When present, cattle (n = 10) were sampled in the nares and poultry (n = 10) in the nares, earlobes and cloaca. A selection of the obtained isolates were further characterized using multilocus sequence typing (MLST), spa typing, SCCmec typing, pulsed field gel electrophoresis (PFGE), multiple‐locus variable‐number tandem repeat analysis (MLVA) and antimicrobial susceptibility testing. On 26 of 30 farms, MRSA was isolated from pigs. Furthermore, MRSA was also isolated from poultry and cattle on one pig/poultry and five pig/cattle farms, respectively. All tested MRSA isolates belonged to ST398. Eight spa types (t011, t034, t567, t571, t1451, t2974, t3423 and t5943) were detected, among which t011 predominated. SCCmec cassettes type IVa and V were present in 20% and 72% of the isolates, respectively. When combining the results of the two remaining typing methods, PFGE and MLVA, eighteen genotypes were obtained of which one genotype predominated (56% of the positive farms). All MRSA isolates were resistant to tetracycline. Resistance to trimethoprim, aminoglycosides, macrolides, lincosamides, fluoroquinolones and chloramphenicol was also observed. In conclusion, there was no effect of the farm type on the MRSA status of the pigs. A statistically significant difference was observed when comparing the pig/poultry or the pig/cattle MRSA status on the multispecies farms. Additionally, a wide variety of MRSA ST398 strains was found within certain farms when combining different typing methods.  相似文献

为了解华东地区近三年来猪圆环病毒病(porcine circovirus disease,PCVD)的流行状况,以便更好地控制该病的发生,2008年1月~2010年9月在华东地区5个省、直辖市及河南省的68个规模化猪场(均未免疫商品化猪圆环病毒疫苗)共收集1017份猪血清样品,应用商品化猪圆环病毒(Porcine circovirus,PCV)抗体诊断试剂盒(PCV2-ELISA),对送检血清样品进行PCV2抗体水平检测。结果显示:送检猪血清中抗体阳性总数为609份,总阳性率为59.88%;2008、2009和2010年送检样品的阳性率分别为53.4%、35.33%和80.63%,PCV血清抗体阳性率呈先下降后上升的趋势;成年猪血清阳性率最高为81.40%,保育猪血清阳性率最低为41.48%,仔猪和育肥猪血清阳性率相近,分别为57.95%和52.20%,说明仔猪断奶后随母源抗体下降其抗体阳性率亦下降,但随猪龄的增长而感染逐渐加重后其抗体阳性率又升高,且不同生长阶段PCV毒抗体阳性率有明显差异。  相似文献
