Bacillus amyloliquefaciens PPCB004 was selected as a potential antagonist to control Botrytis cinerea, Penicillium expansum and Rhizopus stolonifer on peach fruit. The HPLC data of PPCB004 indicated the lipopeptides iturin A, fengycin and surfactin as secondary metabolites. The GC/MS analysis of PPCB004 showed 3-hydroxy-2-butanone as the dominant compound (97.52% of relative peak area). Thyme (TO) and lemongrass (LO) oils showed over 50% and 25% inhibition of radial mycelial growth respectively with 8 μl oil per plate for all pathogens. Combination treatment with both oils failed to increase the percentage inhibition of radial mycelial growth of the pathogens. Combined application of PPCB004 with TO or LO was tested to assess the effectiveness in the control of these pathogens during postharvest storage. The biofilm formation of PPCB004 was significantly higher in LO than TO. LO (6 μl plate−1) and PPCB004 completely inhibited the mycelial growth of the pathogens. Fruit inoculation trials with PPCB004 + LO in NatureFlex™ modified atmosphere packaging (MAP), showed lower disease incidence and severity at 25 °C for 5 d than treatments with PPCB004 + MAP, PPCB004 + TO + MAP, LO + MAP, TO + MAP or stand-alone MAP. On naturally infected fruit, PPCB004 + LO + MAP and LO + MAP treatments retained the total soluble solids/titratable acidity ratio and flesh firmness but failed to stimulate the levels of total phenolic content, phenylalanine ammonia-lyase, β-1,3-glucanase and chitinase activities. Combination of PPCB004 (spray treatment) + LO (in pad delivery system) in NatureFlex™ MAP showed absence of disease and off-flavour development, retained the overall appearance and increased the overall acceptance at market shelf conditions (20 °C for 2 d) after cold storage at 4 °C for 14 d.  相似文献

The development of natural crop protective products as alternatives to synthetic fungicides is currently in the spotlight. In vitro experiments are valuable precursors to more costly in vivo trials, allowing the identification of effective essential oils and establishing the concentrations required for inhibition of a specific, or spectrum of decay pathogens. In this study, the antifungal properties of eighteen essential oils were evaluated in vitro by addition to the fungal growth medium of five pathogens (Lasiodiplodia theobromae, Colletotrichum gloeosporioides, Alternaria citrii, Botrytis cinerea and Penicillium digitatum) isolated from mango, avocado, citrus, grapes and cactus pear. The inhibitory properties of some of the major compounds of the oils, identified and quantified by gas chromatography-mass spectroscopy and gas chromatography-flame ionization detection were also determined. Most of the oils were selected on the basis of commercial availability and for containing a predominant compound. Visual inspection of fungal growth was done and the lowest concentration where fungal growth was completely inhibited on all replicates was recorded. Thyme oil proved to be the most effective inhibitor, totally inhibiting all of the pathogens tested at concentrations of 1000 μl/l and lower, with the exception of a resistant Penicillium strain. Cinnamon oil, rich in eugenol (81.2%), demonstrated good fungicide potential, while the carvone-rich oils displayed promising activity against the citrus pathogens. Oils characterized by high concentrations of S-carvone were less effective than those containing the R-enantiomer. Essential oil of Lippia citriodora was active against all of the pathogens, excluding L. theobromae from avocado. These essential oils, applied alone or in combination, are good candidates for further in vivo testing and for investigations concerning their modes of action.  相似文献

Requirements of consumers for products with low residues of pesticides have increased the need for alternative disease management practices. The concentration of boron in fruit affects its quality, shelf life and the development of physiological disorders. However, the effect of boron on the susceptibility of peach to fruit rots has not been reported. This study investigated the effect of boron (Power B and Borax) on the development of Monilinia laxa on peaches (cv Andross). Mycelial growth of M. laxa was inhibited on potato dextrose agar supplemented with 750 μg ml−1 of Borax or 1000 μg ml−1 of Power B. The EC 50 values were 107.9 and 522.4 for Borax and Power B respectively. Field investigations showed that the incidence of peach infections by M. laxa was negatively correlated with the content of Boron in the leaves. Post-harvest dipping of peaches in Power B or Borax solution, at concentrations recommended by manufacturer (2 μg ml−1 for Power B and 1 mg ml−1 for Borax), significantly reduced the development of M. laxa. Power B, at rates of 6 μg ml−1, and Borax at rates of 3 mg ml−1 were the most effective in reducing infections by M. laxa. Finally, post-harvest dipping of fruit in Power B or Borax reduced losses of fruit weight and improved fruit firmness one month after storage, showing that boron increased the maintainability of peaches in cold storage. Peaches treated with 6 μg ml−1 Power B or 3 mg ml−1 Borax had the highest flesh firmness and the lowest water losses, while untreated control peaches were the least firm. Generally, Borax was significantly less effective than Power B, but better than the control treatment.  相似文献

Transgenic corn hybrids that express toxins from Bacillus thuringiensis (Bt) are highly effective against the European corn borer, Ostrinia nubilalis (Hübner), and the closely related Asian corn borer, Ostrinia furnacalis (Guenée). Since the registration of Bt corn hybrids in the U.S. in 1996, there has been a great deal of information generated on O. nubilalis. However, relatively little information exists for O. furnacalis. To help determine whether the information generated for O. nubilalis can be leveraged for decisions regarding the use of transgenic Bt corn against O. furnacalis, experiments were designed to determine whether the pattern of sensitivity to various Bt Cry1 toxins is similar between the two species. Test insects included laboratory-reared O. furnacalis originating from Malaysia, a Bt-susceptible laboratory colony of O. nubilalis maintained at the University of Nebraska-Lincoln (UNL) and an out-group consisting of the sugarcane borer, Diatraea saccharalis (F.), from Louisiana which represents a different genus from the same family. O. furnacalis and O. nubilalis exhibited a similar pattern of susceptibility to all the Cry1 toxins and were highly susceptible to the range of Bt toxins tested including Cry1Aa, Cry1Ab, Cry1Ac and Cry1F. Both of the Ostrinia species were more tolerant to Cry1Ba compared with D. saccharalis, although sensitivity of O. furnacalis was intermediate and did not differ significantly from that of O. nubilalis and D. saccharalis. D. saccharalis was also susceptible to the range of toxins tested but unlike the two Ostrinia species, was more tolerant to Cry1F and more susceptible to Cry1Ba. These results indicate that both of the Ostrinia corn borer species are similar in sensitivity to the Cry1Aa, Cry1Ab, Cry1Ac, Cry1Ba and Cry1F toxins, thus suggesting shared toxin receptors and mechanisms of toxicity for the two species.  相似文献

Phytophthora fruit rot of watermelon, caused by Phytophthora capsici, is an important and emerging disease in Southeastern U.S.A. The effects of two cultural practices (raised bare ground and raised plastic mulched beds) used for growing watermelon and different fungicide treatments on development of Phytophthora fruit rot were evaluated. The experiments were conducted over three years (2005-2008) at research stations in North Carolina and South Carolina, U.S.A. Fungicides were applied at weekly intervals on the diploid cv. Mickey Lee for an average of five applications. Fruit rot incidence was recorded at the end of each experiment. Fruit rot incidence in the non-treated plots was 66% across two states and six trials. Overall, the levels of fruit rot on the raised bare ground and raised plastic mulched beds were not significantly different. Based on percent disease reduction relative to the non-treated check plots, the fungicide Captan was the most effective across years and locations (range = 23-70%, mean = 57%), followed by mandipropamid (25-65%, mean = 50%), fluopicolide (24-65%, mean = 43%) and cyazofamid (0-48%, mean = 31%). Mefenoxam, the current standard treatment reduced fruit rot by 8-28% (mean = 18%). The addition of copper hydroxide to the spray mix did not significantly enhance effectiveness of Captan or mandipropamid. The variability in fungicide efficacy observed in these experiments across locations and years demonstrates the importance of environmental conditions in disease development and management. Even when the most effective fungicides are used, heavy losses may occur when conditions are highly favorable for disease development. Ultimately, effective control of Phytophthora fruit rot of watermelon will require an integrated management strategy that includes well-drained fields, water management and crop rotation in addition to fungicides.  相似文献

Trichoderma viride was proved as an effective biocontrol agent against two fungal pathogens, Fusarium oxysporum f. sp. adzuki and Pythium arrhenomanes, infecting soybean. During an in vitro biocontrol test, Trichoderma showed mycoparasitism and destructive control against the tested fungal pathogens. Both the pathogens significantly influence the germination and P. arrhenomanes had a severe effect (only 5% germination). The root system of the soybean plant was poorly developed due to the infection and it exerted a negative influence on the nodulation and further growth phases of the plant. During pot assay along with biocontrol activity, Trichoderma showed growth promoting action on the soybean plant. Trichoderma enhanced growth of shoot and root systems and fruit yield after 12 weeks of growth. Pythium and Fusarium infected plants treated with Trichoderma had ∼194% and 141% more height than pathogens alone. The fruit yield treated with Trichoderma was ∼66 per plant whereas the yield was only 41 for a control plant. The plants infected with Pythium and Fusarium and treated with Trichoderma had fruit yields of 43 and 53 respectively and those were 5 and 1.6 times higher than plants infected with pathogens.  相似文献

The nematicidal effect of Pseudomonas fluorescens, Paecilomyces lilacinus, Pichia guilliermondii and Calothrix parietina singly or in combination was tested against root-knot nematode, Meloidogyne incognita. Treatments with P. fluorescens and P. lilacinus caused mortality of M. incognita as 45% and 30% of juveniles after 48 h of exposures, respectively compared to water control in vitro. Under greenhouse conditions, all treatments reduced the disease severity and enhanced plant growth compared to untreated control. Application of P. fluorescens, P. lilacinus and P. guilliermondii Moh 10 was more effective compared to C. parietina. There was a negative interaction between C. parietina and either P. lilacinus or P. guilliermondii. Fresh and dry weight of shoots and roots of plants were significantly reduced as a result of infection with M. incognita, however application of biocontrol agents singly or in mix recovered this reduction. Moreover, they enhanced the growth parameters compared with the control. Our results proved that application of different biocontrol agents (P. fluorescens, P. lilacinus and P. guilliermondii) not only has a lethal effect on nematode, but also enhances the plant growth, supplying many nutritional elements and induction the systemic resistance in plants. Presence of C. parietina as a soil inhabitant cyanobacterium could antagonize biocontrol agents leading to the reduction of their practical efficiency in soil.  相似文献

Field corn, Zea mays L., plants expressing Cry1Ab and Cry1F insecticidal crystal (Cry) proteins of Bacillus thuringiensis (Bt) Berliner are planted on considerable acreage across the Southern region of the United States. The fall armyworm, Spodoptera frugiperda (J.E. Smith), is an economically important pest during the mid-to-late season on non-Bt and some commercial Bt corn hybrids. The objective of this study was to quantify foliar injury and survivorship of fall armyworm on transgenic corn lines expressing Cry1Ab or Cry1F Bt proteins. Corn lines/hybrids expressing Cry1Ab, Cry1F, and a conventional non-Bt cultivar were evaluated against artificial infestations of fall armyworm in field trials. Larvae (second instars) of fall armyworm were placed on corn plants (V8-V10 stages). Leaf injury ratings were recorded 14 d after infestation. Hybrids expressing Cry1F had significantly lower feeding injury ratings than non-Bt corn plants. Development and survivorship of fall armyworm on Bt corn lines/hybrids were also evaluated in no-choice laboratory assays by offering freshly harvested corn leaf tissue to third instars. Transgenic corn hybrids expressing Cry1Ab or Cry1F significantly reduced growth, development, and survivorship of fall armyworm compared to those offered non-Bt corn tissue. However, 25-76% of third instars offered Bt corn leaf tissues successfully pupated and emerged as adults. These results suggest Cry1Ab has limited effects on fall armyworm; whereas Cry1F demonstrated significant reductions in foliar injury and lower survivorship compared to that on non-Bt corn tissues. Although fall armyworm is not considered a primary target for insect resistance management by the U.S. Environmental Protection Agency, these levels of survivorship could impact selection pressures across the farmscape, especially when considering that transgenic Bt cotton cultivars express similar Cry (Cry1Ac or Cry1F) proteins.  相似文献

Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) is a serious pest of cotton and many other crops in northern China. To evaluate the contribution of alternative hosts as an effective refuge for transgenic cotton expressing the Bacillus thuringiensis (Bt) Cry1Ac toxin, the susceptibility to this toxin was measured in progeny derived from field-collected H. armigera larvae and pupae from different hosts in the Xiajin’s region of the Shandong Province in northern China. During 2008-2010, progeny from a total of 258,56,184 and 160 single-pair crosses derived from wheat (first-generation), Bt cotton (second-generation), Bt cotton (third-generation), and corn (third-generation) were screened on Cry1Ac diets, respectively. Based on relative average development rates (RADR) of H. armigera larvae in these F1 tests, the second and third-generation moths emerging from Bt cotton fields were more tolerant to the Bt toxin than the first and third-generation moths emerging from wheat and corn each year. These results suggest that there is significant variation in susceptibility to Bt toxins among H. armigera populations derived from different host crops. Alternate crops, such as corn, that maintain Bt susceptible populations of H. armigera could be used as refugia to minimize the evolution of resistance to Bt cotton.  相似文献

Grain protein concentration (GPC) affects wheat nutritional value and several critical parameters for bread and pasta quality. A gene designated Gpc-B1, which is not functional in common and durum wheat cultivars, was recently identified in Triticum turgidum ssp. dicoccoides. The functional allele of Gpc-B1 improves nitrogen remobilization from the straw increasing GPC, but also shortens the grain filling period resulting in reduced grain weight in some genetic backgrounds. We developed isogenic lines for the Gpc-B1 introgression in six hexaploid and two tetraploid wheat genotypes to evaluate its effects on bread-making and pasta quality. In common wheat, the functional Gpc-B1 introgression was associated with significantly higher GPC, water absorption, mixing time and loaf volume, whereas in durum wheat, the introgression resulted in significant increases in GPC, wet gluten, mixing time, and spaghetti firmness, as well as a decrease in cooking loss. On the negative side, the functional Gpc-B1 introgression was associated in some varieties with a significant reduction in grain weight, test weight, and flour yield and significant increases in ash concentration. Significant gene × environment and gene × genotype interactions for most traits stress the need for evaluating the effect of this introgression in particular genotypes and environments.  相似文献

The legume pod borer Helicoverpa armigera (Hubner) (Lepidoptera: Noctuidae) has developed high levels of resistance to conventional insecticides, and therefore, efforts are being made to develop transgenic chickpea expressing toxin genes from the bacterium Bacillus thuringiensis (Bt) for controlling this pest. However, there is an apprehension that acid exudates in chickpea might interfere with the biological activity of Bt. Therefore, we studied the biological activity of Bt (BiolepR) on four chickpea genotypes with different levels of resistance to H. armigera under field conditions, and by incorporating lyophilized leaf and pod tissue into the artificial diet with and without Bt. The pH of the acid exudates varied from 2.1 to 2.9, and malic and oxalic acids were the major components of the acid exudates in different chickpea genotypes. There was no survival of H. armigera larvae in chickpea plants sprayed with 0.1, 0.2 and 0.5% Bt. There was a significant reduction in larval survival, larval and pupal weights and fecundity, and prolongation of larval and pupal periods in chickpea plots sprayed with Bt (0.05%) as compared to the unsprayed plots. Biological activity of Bt was lower on artificial diets with leaf or pod powder of chickpea genotypes, which might be because of a low intake of Bt toxins due to the antifeedant effects of acid exudates in the chickpea or reduction in biological activity of Bt due to the interaction of biochemical constituents in chickpea with the Bt toxins. Larval survival, larval and pupal weights, pupation and adult emergence were significantly lower on diets with leaf or pod powder of the H. armigera-resistant genotypes than on the susceptible check. Chickpea genotypes with resistance to H. armigera acted in concert with Bt to cause adverse effects on the survival and development of this insect. The results suggested that development of transgenic chickpeas expressing toxin genes form Bt will be quite effective for controlling of the pod borer, H. armigera.  相似文献

Heat treatments affect the germination and development of pathogens and enhance the ability of fruits to resist infection after harvest. In the present work, the effects of heat, in combination with other alternative decay control methods, were studied on ‘Montenegrina’ tangerines along with the effects on the cuticular surface of the fruit. Different treatment combinations using hot water (60 °C), brushing, and immersion in chloride dioxide, imazalil, sodium bicarbonate, and hand-applied carnauba wax were used. The tangerines were then put in cold storage for 20 days at 5 °C and retrieved to ambient conditions for 7 more days. Fruit samples were analyzed for total soluble solids, titratable acidity, epidermal color, weight loss, and decay incidence; an analysis of fruit surface was done through scanning electron microscopy. Heat treatments significantly reduced the number of tangerines with decay symptoms and enhanced the efficacy of the tested products. Carnauba wax significantly increased the number of rotten fruits, exerting a protective effect on the fungi by covering their structure. Sodium bicarbonate in combination with heat exerted a good control on decay incidence. The heat treatments partially removed the hyphae and spores of pathogens on the surface and also melted the cuticular waxes that cover stomata and cracks, reducing possible entry points for pathogens.  相似文献

Transgenically expressed Bacillus thuringiensis insecticidal-protein Cry1Ab was quantified in target tissues of insect feeding of several YieldGard® corn hybrids. The Cry1Ab protein is intended to protect corn plants from two economically important stem borers, Chilo partellus and Sesamia inferens. A total of seven YieldGard hybrids, all with MON810 event, were field-tested in a total of fourteen locations during the dry season (October–March) of 2005/2006 and wet season (May–October) of 2006. S. inferens and C. partellus oviposit on leaves of young corn plants, 15–60 days after emergence (DAE). The neonates initially feed by scraping the leaf lamina before migrating to bore into the stem. Thus high concentrations of Cry1Ab in whorl leaf and stem tissues would ensure effective control of the borers. The mean tissue Cry1Ab concentrations during the oviposition window of the borers (15–60 DAE), ranged from 50.05 to 21.01 ppm in whorl leaf, and between 9.26 and 3.47 ppm in stem tissue during the same period in the dry season of 2005/06. Similarly, Cry1Ab concentrations in whorl leaf and stem between 15 and 60 DAE during the wet season of 2006 ranged between 19.30 to 11.08 and 14.28 to 4.69 ppm, respectively. The baseline-sensitivity data of these insects to Cry1Ab in laboratory assays was determined. The concentrations of Cry1Ab in the target tissues as studied in seven YieldGard hybrids tested suggest effective management of the two borers. This paper also provides a summary of the expression of the Cry1Ab gene in various genetic backgrounds.  相似文献

Multiple independent trials were conducted to evaluate the performance of Cry1Ab-susceptible (Cry1Ab-SS), -heterozygous (Cry1Ab-RS), and -resistant (Cry1Ab-RR) genotypes of the sugarcane borer, Diatraea saccharalis (F.), on eight commercial hybrids and six experimental corn lines. The commercial varieties included two non-Bt and six Bt corn hybrids that expressed a single Bt protein (either Cry1Ab or Cry1F) targeting above-ground lepidopteran pests. The six experimental lines consisted of two non-Bt and four Bt corn lines, two expressing just the Cry1Ab protein and two containing the pyramided-genes Cry1A.105 and Cry2Ab2 (event MON 89034). Larval mortality on non-Bt corn leaf tissue ranged from 6 to 45% after 12 d across insect genotypes. The 12 d mortality of Cry1Ab-SS on leaf tissue of commercial Cry1Ab or Cry1F corn was 96-100%, whereas it was 80-96% for Cry1Ab-RS and 68-78% for Cry1Ab-RR. On intact plants, 39-64% of larvae survived on non-Bt corn plants after 21-25 d. Larval survivorship on intact plants of commercial Cry1Ab or Cry1F corn was 0-8.1% for Cry1Ab-SS, 1.3-34% for Cry1Ab-RS, and 19-51% for Cry1Ab-RR. Larvae of Cry1Ab-RR and -RS also caused significant plant injury to most of the commercial Bt corn hybrids, especially to the Cry1Ab corn. Cry1Ab resistance in D. saccharalis was incompletely dominant on commercial Bt corn hybrids. However, both experimental lines with pyramided genes of Cry1A.105 and Cry2Ab2 provided complete control of all three insect genotypes in both leaf tissue and intact plant tests. Results of this study suggest that MON 89034 should offer a means for Bt resistance management in D. saccharalis.  相似文献

Talaromyces flavus a fungal antagonist, was isolated from soil samples collected from potato fields in Varamin and Karaj districts, Tehran province, Iran. Antagonistic effects of T. flavus isolates against Verticillium albo-atrum, the causal agent of potato wilt disease were investigated in the laboratory and greenhouse conditions. T. flavus colonies were recovered after three weeks from soil samples cultured on selective medium. Antagonistic effects of volatile and non-volatile extracts of T. flavus isolates on V. albo-atrum growth were investigated in the laboratory and five that caused higher growth inhibition of V. albo-atrum, were selected for greenhouse experiments. Infection index was compared in the greenhouse in a split plot trial with five isolates applied to soil, seed, or both arranged in a randomized complete block design with four replications. The minimum infection index was observed when seed were treated with T. flavus with the most effective isolate being Tf-Po-V-52. On seed, the minimum infection index was observed with Tf-Po-V-50. The most effective T. flavus isolate was also evaluated in a field experiment. Results indicated that infection index in seed treatment contained this isolate was 0.15 whereas that of control was 3.5. The overall results of this study showed that it may be possible to manage potato Verticillium wilt disease effectively by using T. flavus, a biocontrol fungus.  相似文献

Ten strains of Pseudomonas aeruginosa (PN1 ˜ PN10) isolated from rhizosphere of chir-pine were tested for their plant growth promontory properties and antagonistic activities against Macrophomina phaseolina in vitro and in vivo. P. aeruginosa PN1 produced siderophore, IAA, cyanogen and solubilized phosphorus, besides producing chitinase and β-1,3-glucanase. In dual culture, P. aeruginosa PN1 caused 69% colony growth inhibition. However, cell free culture filtrate also posed inhibitory effect but to a lesser extent. After 90 days, P. aeruginosa PN1 increased plant growth and biomass in pots trial containing M. phaseolina-infested soil. PN1 showed the strong chemotaxis toward root exudates resulting in effective root colonization. Moreover, increased population in rhizosphere of these bacteria was also recorded after 90 days of treatment. Thus, chemotactic fluorescent P. aeruginosa PN1 exhibited strong antagonistic property against M. phaseolina, suppressed the disease and improved plant growth of the seedlings of chir-pine proving potential biocontrol agent.  相似文献

Field experiments were conducted to evaluate cover crop (none, winter wheat, or winter rye) and pre-emergence (PRE) applications of clomazone plus ethalfluralin alone or with halosulfuron [PRE or post-emergence (POST)] for smooth crabgrass [Digitaria ischaemum (Schreb. ex Schweig) Schreb. ex Muhl.] and redroot pigweed (Amaranthus retroflexus L.) control in no-tillage ‘Aspen’ jack-o-lantern pumpkin (Cucurbita pepo L.). At pumpkin harvest, cover crops had reduced smooth crabgrass density, but not redroot pigweed. Although PRE or POST applications of halosulfuron alone were more effective at reducing redroot pigweed density than clomazone plus ethalfluralin PRE, the combination of halosulfuron plus clomazone and ethalfluralin PRE reduced redroot pigweed density to the greatest extent. Pumpkin yields were not affected by cover crop, although average pumpkin sizes were greater with the inclusion of cover crop residues. Pumpkin fruit sizes and yields were the greatest with clomazone and ethalfluralin PRE in combination with halosulfuron applied PRE or POST. Overall, cover crop had relatively little influence on pumpkin yields compared with herbicide treatments. The addition of halosulfuron to clomazone and ethalfluralin provided greater broadleaf weed control resulting in greater jack-o-lantern pumpkin yields.  相似文献

Strain 3A25 was isolated from Indian soil during a screening study for new plant protecting candidates. The present study further characterized this strain regarding its species identity, cellular ultrastructure and potential for protecting Brassica juncea against exposure to ozone. The strain was identified as Bacillus subtilis by 16S rDNA sequencing. Transmission electron microscopy revealed that strain 3A25 had intact bacterial flagella, endospores were found only occasionally. The growth-promoting capacity of strain 3A25 on B. juncea seedlings was investigated in a series of treatments with or without ozone stress in open top chambers. Chlorophyll content, shoot length, root length and leaf area were significantly increased by the treatment with B. subtilis 3A25. The ozone treatment (150 pbb) had a significant inhibiting effect on the development of B. juncea, however, the number of secondary roots was increased. Application of B. subtilis 3A25 either as foliar spray or directly on the soil significantly reduced the negative ozone effect, indicating that this strain has capacities in plant protection against environmental stress.  相似文献

A local isolate of Trichoderma asperellum was tested for its antagonistic activity against Thielaviopsis paradoxa (telemorph = Ceratocyctis paradoxa). The highest antagonistic activity was achieved when the concentration of T. asperellum conidia was 1 × 107 conidia/mL. The highest biomass and number of colony forming unit/mL of the T. asperellum peaked at 144 h after incubation in yeast waste residue medium. The minimum inhibition concentration value of the formulation was observed as 1% on growth of Th. paradoxa incubated at 28 ± 2 °C for 10 d. In the soil fungicide-screening test, the effect of concentrations 100-1600 μg/mL on mycelia growth was not significant (P < 0.05). Complete mycelial growth inhibition occurred at concentration above 52,600 μg/mL. Results of the fruit application tests clearly showed that all treated fruits were free of disease at the end of the incubation period. No significant differences (P > 0.05) in pH, total soluble solids and titratable acidity were observed between fruits treated with formulation of T. asperellum and the control formulation treated pineapples.  相似文献

In a mature Vitis labrusca L. “Niagara” vineyard, we compared an airblast sprayer and an air-assisted rotary atomizer (AARA) low-volume sprayer for coverage of grape clusters and control of grape berry moth, Paralobesia viteana. In addition, the effect of spray volume on control of fungal diseases was evaluated using the fungicides ziram and azoxystrobin applied with an airblast sprayer. For evaluation of coverage, sprayers applied kaolin clay at 28 kg/ha to grapevines while operating at low, medium and high water volumes. Kaolin residues on grape clusters were analyzed to compare the total amount of spray material deposited, percent of fruit surface covered, number of deposits, size of deposits and distance between deposits. There were no significant differences between sprayers or water volumes in the total amount of kaolin deposited on clusters, but the percent surface coverage was much greater on outside-facing berry surfaces (facing the sprayer) than inside-facing surfaces (facing the rachis). On the outside-facing berry surfaces, the airblast sprayer at 468 L/ha of water (medium volume) provided the highest percent coverage, the greatest density of deposits, the largest deposit diameter, and the smallest distance between deposits. On the inside-facing berry surfaces, the same sprayer–volume combination provided the highest percent coverage and greatest deposit diameter, but deposits from the airblast sprayer operating at 935 L/ha (high volume) had the smallest distance between deposits. No significant differences between sprayers or among water volumes were detected in the deposit density on the inside-facing berry surfaces. Bioassays of grape clusters sprayed with fenpropathrin or methoxyfenozide using the airblast or AARA sprayers at two water volumes revealed the greatest fruit protection from P. viteana at the higher volume for the airblast sprayer, but at the lower volume for the AARA sprayer. Spray volume of the airblast sprayer also affected disease control by the protectant fungicide ziram more than by the systemic fungicide azoxystrobin, with 468 L/ha providing better control than 187 L/ha. However, for most diseases, fungicide type was more influential than spray volume in determining the disease control outcome. The results of this study emphasize the need for appropriate water volumes relative to the sprayer and pesticide being used to optimize pest and disease control in juice grape vineyards.  相似文献
