通过对桑叶褐斑病病原菌在不同种类培养基培养、不同氮源与碳源、不同温度等6种条件下培养6天后,测定菌落直径,观察菌丝颜色和菌落形态,分析了桑褐斑病病原菌的生物学特性。结果表明,在室内培养条件下,不同的营养成分、 pH值、温度、光照都对菌丝的生长有影响。病原菌菌丝生长的最适宜培养基是燕麦片培养基,适宜生长温度范围是20~30℃,最适生长温度为25℃,光照对菌丝生长无显著影响,最适pH值为9~10,最适碳源为α-乳糖,最适氮源为尿素,菌丝在55℃水浴10 min后则不能生长。 相似文献

橡胶树炭疽病病原菌Colletotrichum laticiphilum生物学特性研究及防治药剂筛选 总被引:1,自引:0,他引:1
对橡胶树新发现病害病原菌Colletotrichum laticiphilum的生物学特性进行初步研究并开展室内筛选防治药剂,旨在为该病害的防治提供理论依据。通过单因子变量试验,研究病原菌最适生长条件。菌丝最适生长培养基为土豆琼脂培养基,最适生长温度为26℃,最适合生长pH值为6,菌丝致死温度为50℃、10 min,分生孢子致死温度为51℃、10 min。该病原菌能利用多种碳源和氮源,碳源以可溶性淀粉的利用率最高,牛肉浸膏为最适氮源,在24 h光照条件下更有利于菌丝生长。采用菌丝生长速率法测定室内毒力,结果表明:4种杀菌剂对病原菌的毒力存在明显差异,其中咪鲜胺和丙环唑的毒力最强,EC_(50)分别为0.1286 mg/L、0.5208 mg/L。 相似文献

黄伞是一种食药兼优且具有较高商品价值的珍稀食用菌,研究黄伞的生物学特性,对于开展黄伞栽培具有重要意义。该文通过传统形态学和分子生物学相结合的方法对在沈阳柳树树干基部采集到的真菌新鲜子实体进行鉴定,并在室内纯培养条件下,采用生长速率法对该菌在不同碳源、温度、pH值、光照条件下菌丝生长和菌落形态进行研究。鉴定表明该菌为黄伞Pholiota adipose。测试结果为:该菌生长最适碳源为葡萄糖;菌丝生长适宜温度范围为20~30℃,最适生长温度为25℃;适宜pH值为5.5~6.5;最佳光照条件为光暗交替,但是在培养初期,光暗交替条件下菌丝生长速度较慢。 相似文献

通过对从不同林分的油茶根系分离的3种高效固氮菌(N7-3、B7-7、N5-18)的适宜生长环境以及回接效应的研究,结果表明:菌株N5-18适宜生长条件为温度20℃,pH为6,适宜碳源为蔗糖,耐高渗透压;菌株B7-7适宜生长条件为温度20℃,pH为5,适宜碳源是柠檬酸,渗透压对菌株生长无影响;菌株N7-3适宜生长条件为温度30℃,pH为6,适宜碳源是柠檬酸,高渗透压抑制生长。接种结果表明接种3种固氮菌能明显促进油茶生长,菌株N7-3对油茶的苗高、地茎、生物量、叶绿素含量的增益效果最好,B7-7对硝酸还原酶活性增幅最大,N5-18对叶片全氮和粗蛋白含量增幅最大。 相似文献

研究了碳源和氮源对白腐菌Sarcodon asparatus合成脂肪酶的影响。橄榄油是脂肪酶合成的最佳碳源,脂肪酶活力达0.65U/mL。蛋白胨、尿素和氯化铵对脂肪酶的合成有促进作用,它们的酶活力分别为0.65、0.67和0.62U/mL,而硫酸铵抑制脂肪酶的合成,其酶活力仅为0.08U/mL。C/N比对细胞生长和酶的合成影响很大,当C/N比从2增加到10时,脂肪酶活力从0.65下降到0.14U/mL,而细胞浓度从3.08升高到4.58mg/mL。在培养基初始pH值5.5、28℃下培养,S.asparatus合成的脂肪酶活力最高,脂肪酶活力和细胞浓度分别为0.65U/mL和3.08mg/mL。 相似文献

2012年6~8月,通过对桑树轮纹病病原菌分离、田间致病性测定、病原菌形态观察,将该病原菌鉴定为桑膝节霉,并对此病病原菌的生物学特性进行了研究。结果表明,不同培养基配方、温度、酸碱度、碳源、氮源、光照对桑树轮纹病病原菌菌丝的生长有一定影响,菌丝生长的最适宜培养基是理查培养基,最适温度是25℃,最适pH值是7.0,最适碳源为葡萄糖,最适氮源为蛋白胨,光照对菌丝生长无显著性差异,菌丝的致死温度和时间分别为85℃和10 min。对该病病原菌生物学特性研究为云南首次报道,对桑树轮纹病的防治具有一定的参考价值。 相似文献

通过P.t.优良菌株9109在不同培养基上生长速度、培养特性及其生长规律的研究,筛选出生长最佳培养基作为基础培养基,进行C、N营养研究,试验结果表明:P.t.9109不仅能利用单糖、低聚糖和无机N等速效C、N源,而且能利用多糖、多元醇和有机N等长效C、N源。其菌丝生长最适C源是玉米粉和甘露糖;最适N源是蛋白胨和氯化铵。在无机N利用中,对硝态N和铵态N的利用无显著差异。应用L16(45)正交设计确定了P.t.9109菌丝生长的最佳营养配方,菌丝增殖高达860倍,不但提高菌丝生物产量,而且节省了原料,降低了成本,为改进P.t.菌剂生产配方和研制新型菌剂提供了最佳营养条件。 相似文献

系统研究了碳源、氮源、温度、起始pH值、培养基装量和接种量等因素对绿粘帚霉(Gliocladium virens)F051产几丁质酶的影响。结果表明,碳、氮源分别为麦芽糖和酵母粉最好,并通过正交试验法优化了发酵条件。优化后的产酶条件为:温度为28℃,pH值7,瓶装量为100 ml,每100 ml接种1 ml孢悬液,且碳源和氮源的量分别为1.5%和0.2%。从时间对菌丝生长和几丁质酶活的影响来看,培养第4天酶活便达到最高。在此基础上探索了几丁质对病原菌细胞壁的作用,结果显示,该几丁质酶粗酶液对3种病原真菌细胞壁有一定的降解效果,使病原真菌出现细胞壁变薄、断裂,质壁分离,原生质空泡等现象。 相似文献

Forest floor materials from a 45-year-old natural stand of Pinus palustris—P. elliottii growing on two coastal flatwood Spodosols (Electra and Wauchula fine sands), differing mainly in drainage characteristics, were similar except for total and organic matter weights and Al and Fe concentrations. The floors averaged 9.8 and 28.0 t/ha total dry weight in the 01(L) and 02(F + H) layers, respectively. Mineral soil represented 12 and 28% of the weights of these two layers. The 01 layer was equivalent to about 1.5 times and the 02 about 3.7 times the annual litter fall of the stands. The generally low nutrient concentrations, particularly of N (0.58 and 1.02% in 01 and 02 layers) and P (0.025 and 0.045%) are believed to influence decomposition rate of floor materials and result in a relatively heavy accumulation. This conclusion was supported by the wide C:N ratios of 92 (01 layer) and 53 (02 layer) and C:P ratios of 2116 and 1200 in these layers. Cation exchange capacity (CEC) at field pH averaged 32 and 45 meq/100 g of 01 and 02 material, on an ash-free basis. Grinding to pass a 20-mesh sieve significantly increased CEC of 01, but not of 02 materials. CEC increased linearly with decreasing acidity of the saturating solutions over the range of pH 4 to 7. The increase was from 35 to 75 meq/100 g in 01 materials and from 62 to 128 meq/100 g in 02 materials, over this pH range. The correlation between ash-free total N and CEC (pH 7) was highly significant (r2 = 0.87), with 1.65 meq/100 g CEC associated with each meq/100 g N. The nutritional and physical requirements for the efficient mycelial colonization ofVolvariella volvacea (Bull. ex. Fr.). Singer were elucidated with the percentage mycelial colonization and density as references. This investigation was limited to the evaluation of two commercial strains (designated Vvc1 and Vvc2) and two wild strains (designated EAAC-0001 and EAAC-0002) ofV. volvacea from the Philippines with the aim of providing baseline data on their physiological requirements. The four strains ofV. volvacea had varying preferences for carbon. Vve1 preferred polysaccharides (starch and cellulose), whereas Vvc2 grew luxuriantly at a relatively rapid rate in sugar alcohol (sorbitol). The two wild strains preferred starch as a carbon source. In terms of nitrogen utilization, soytone, peptone, and glycine supported efficient mycelial colonization of the four strains. The vitamin utilization test revealed that ascorbic acid, calcium pantothenate, and biotin are good sources. The mycelial growth performances of the strains were also evaluated on six dehydrated mycological media. Efficient colonization of Vvc1, Vvc2, and EAAC-0002 with dense mycelial growth was noted in mycological agar. EAAC-0001, on the other hand, grew more efficiently in malt extract agar. The Philippine strains ofV. volvacea grew luxuriantly when incubated at 35°C and pH 8.0 under dark and sealed conditions. Moreover, the relatively higher moisture content (70%) of the oolong tea leaf formulation favorably stimulated efficient mycelial colonization. Under optimum physiological conditions, Vvc1, Vvc2, and EAAC-0002 were fast-growing strains, whereas EAAC-0001 was a moderately growing type. 相似文献
