In the present study, fetuses were hypophysectomized (hypox) in utero on d 72 to 74 of gestation with an electrical cauterizing needle. One to six successfully hypox fetuses were removed on d 110 of gestation from each of five gilts. Subcutaneous adipose tissue samples and semitendinosus muscles were obtained from the hypox fetuses and an equal number of control fetuses. Body weights of control fetuses (n = 15; mean +/- SE, 1,195 +/- 33 g) were similar to weights of hypox fetuses (n = 15; 1,179 +/- 67 g). Fat cell size in the middle subcutaneous layer of adipose tissue was increased in hypox fetuses (P less than .01) compared with control fetuses. The number of obvious fat cell clusters (outer layer) in lipid stained sections was reduced (P less than .01) by 50% in hypox fetuses. Histochemical reactions for glucose-6-phosphate dehydrogenase, esterase and lipoprotein lipase (LPL) activities in middle layer cell clusters were considerably enhanced in sections from hypox fetuses compared with sections from controls. Quantitative analysis of percent light transmittance (Zeiss photometer) through LPL-stained cell clusters indicated an increase (P less than .001) in LPL staining in sections from hypox fetuses when compared with sections from control fetuses. Transverse muscle sections (cryostat) from hypox fetuses failed to show normal patterns (as seen in control muscles) of reactions for acid ATPase, malate dehydrogenase (NAD-dependent), NADH-TR and alpha-glycerol phosphate dehydrogenase (without NAD). The number of muscle fibers that were stained for these enzymes was greatly reduced in hypox fetuses compared with control fetuses. Fetuses were decapitated in one uterine horn in each of 14 sows at 45 d of gestation. Control (C) and decapitated (D) fetuses were removed by Caesarean section from three sows at 65 d of gestation (total of 10 D and 10 C fetuses), two sows at 85 d (six D and six C fetuses) and nine sows at 110 d (nine C and nine D fetuses) of gestation (Exp. 1). In Exp. 2, four to six fetuses were removed from each of two Ossabaw (O) gilts and three crossbred (C, Landrace X Yorkshire) gilts at 70 d of gestation, from three C and O gilts at 90 d of gestation and from three C and two O gilts at 110 d of gestation. In Exp. 1, one semitendinosis muscle was removed for histochemistry, whereas the contralateral muscle was removed and weighed. A medial portion of biceps femoris muscle was removed and used for histochemistry in Exp. 2. In both experiments, transverse sections (cryostat) of muscle were stained for lipid, glycogen (PAS) and the following enzymes: acid ATPase, NADH-TR, NADPH-TR, malate dehydrogenase (NAD- and NADP-dependent reactions; MDH), succinate dehydrogenase (SDH), alpha-glycerol phosphate dehydrogenase (with and without NAD; alpha-GPDH), isocitrate dehydrogenase (NAD dependent; ICDH), esterase, lipoprotein lipase and lipase. The development of the innermost layer of backfat was studied histologically and histochemically in fetuses from Yorkshire sows at 45, 60, 65, 75, 85, 90, 105, 110, and 112 d of gestation. Samples were also obtained from fetuses at 110 d of gestation from Ossabaw sows and from sows selected for high (obese) or low (lead) backfat thickness. Fetuses from some sows were decapitated at 45 d of gestation and samples were obtained from these fetuses at 110 d of gestation. The inner layer develops before 75 d of gestation and has stromal-vascular characteristics that clearly distinguish it from the middle layer of backfat. For instance, a well organized and dense collagen matrix immediately surrounds small fat cell clusters in the inner layer. Most fat cell cluster capillaries (in the inner layer) reacted strongly for ATPase histochemically and are histologically immature. The inner layer morphology and cell size was similar in Ossabaw, obese and lean fetuses. The number of PAS (Periodic Acid Schiff) positive cells were dependent on fetal strain. The number was highest in obese fetuses and lowest in Ossabaws. Comparison of decapitated fetuses and weight-matched controls indicated that the inner layer was most affected by decapitation. A decreased organization and amount of collagen was associated with larger fat cell clusters in decapitated fetuses, and most fat cell cluster capillaries reacted little for ATPase histochemically and were histologically mature. The range in inner-layer fat cell diameter was wider in decapitated fetuses (11 to 55 micron) than in control fetuses (11 to 30 micron). These results demonstrate that the characteristics of the inner layer are clearly different from the other layers and that the amount and organization of collagen was inversely associated with the size of fat cell clusters and fat cells and with the maturity of the adipocyte-associated capillaries.  相似文献

At 110 d of gestation, fetuses were removed from Ossabaw, Yorkshire and crossbred sows and from sows selected for high (obese) or for low (lean) backfat thickness. Ossabaw and obese fetuses were smaller than lean, Yorkshire and crossbred fetuses (595 +/- 32 and 863 +/- 44 g vs 1,030 +/- 77, 1,380 +/- 1,144 +/- 80 g; means +/- SE), respectively. Minimum fiber diameters in the semitendinosus muscle were larger in obese, lean and Ossabaw fetuses than in Yorkshire and crossbred fetuses (12.9 +/- .3, 12.5 +/- .2 and 11.8 +/- .2 micron vs 10.2 +/- .2 and 11.1 +/- .8 micron), respectively. Histochemical analysis for NADH-tetrazoleum reductase (NADH-TR) and esterase activities indicated no fiber type differentiation and no strain differences. Fiber type differentiation was obvious with acid ATPase histochemistry in muscles from all fetuses. The white portion of the semitendinosus from Ossabaw, obese and lean fetuses had many fibers that contained no histochemically detectable lipid (oil red O staining). The unstained fibers (oil red O) were always the most peripherally located fibers in a fasciculi. In some instances, 50% of the fibers in a fasciculi were not stained for lipid. All the fibers in the red portion of muscle from Ossabaw, obese and lean fetuses contained lipid. All the fibers in the red and white portions of muscles from crossbred and Yorkshire fetuses contained lipid. Muscles from young (1 to 2 d old) Ossabaw, Yorkshire and crossbred pigs were also histochemically analyzed. Analysis for NADH-TR, esterase and alkaline phosphatase (capillary staining) activities indicated no fiber type differentiation and no strain differences. As in the fetuses, the white portion of muscle from Ossabaw pigs had many fibers with no lipid (oil red O). Lipid was present in all fibers in the deep portions of muscle from Ossabaws and in all fibers in both portions of muscle from crossbred and Yorkshire pigs. These results indicate that when lipid staining is used as the criterion, fiber type differentiation is evident in muscle from fetuses and young pigs from strains not genetically selected for muscling (Ossabaw, obese and lean strains). Furthermore, fiber type differentiation is not evident in muscle from strains of pigs genetically selected for greater muscling (crossbred and Yorkshires).  相似文献

Fetuses from linebred lean (L) and linebred obese (O) and reciprocal crossmatings were examined at 110 d of gestation for line, maternal and heterotic effects. There was no significant heterotic effect for any trait measured. A significant maternal effect was observed for adipose tissue lipoprotein lipase (LPL) activity and for serum triglycerides. The enzyme activity and triglycerides concentration were higher in fetuses from O dams than in fetuses from L dams. In a lipid clearance test, no maternal effect was observed for changes in serum concentrations of triglycerides and free fatty acids or in optical density (associated with the disappearance of injected Liposyn). Linebred O fetuses exhibited higher LPL activity in both the biceps femoris muscle and sc adipose tissue compared with linebred L fetuses. The LPL activity of the adipose tissue was higher than that of the skeletal muscle. The percentage of dry matter, percentage of triglycerides and protein/DNA were higher in the muscle of linebred O fetuses than in that of linebred L fetuses. Based on tissue LPL activity and on muscle compositional traits, linebred O fetuses were more mature at 110 d of gestation than were the linebred L fetuses. Maternal obesity had little detectable influence on fetal development of the pig when measured at 110 d of gestation.  相似文献

Body composition, liver and adipose lipogenesis, and pancreatic insulin release were examined in intact and decapitated fetal pigs on d 110 of gestation. Decapitation was on d 45 of gestation. Decapitated fetuses deposited more body lipid and less body ash compared with intact fetuses. Body weight, water, dry matter and protein remained similar in intact and decapitated fetuses. Hepatic fatty acid esterification and synthesis were two- and threefold greater, respectively, in decapitated than in intact fetuses. Fatty acid synthesis in subcutaneous adipose tissue of decapitated fetuses was three times greater than values obtained in intact fetuses. The data supported the concept that substrate availability from the dam was not the rate-limiting step in fetal pig lipid synthesis and storage. High growth hormone levels in normal fetal pigs may be responsible for inhibiting lipogenesis, while fetal decapitation would remove this inhibition and be associated with greater lipid deposition. However, pancreatic insulin release was greater in decapitated than in intact fetuses; an indication that elevated lipid deposition may also be due to greater fetal insulin secretion.  相似文献

Maternal diet influences fetal growth and postnatal development. We hypothesized that conceptuses gestated in sows provided ad libitum vs. restricted feed intake would differ in the milieu of hormones, growth factors, nutrients, and metabolites associated with growth and metabolism. This hypothesis was tested in two experiments by providing fourth-parity sows (Pig Improvement Co. C15 bred to Line 326 boars) with either 1.81 kg/d (as-fed basis; control) or ad libitum access to gestation diet. In Exp. 1, control (n = 6) or ad libitum (6.4 +/- 0.11 kg/d; n = 9) treatments were provided from d 29 to 45 (onset of estrus is d 0), and sows were slaughtered on d 46. Ad libitum sows gained more weight from d 29 to 45 than controls (34.0 vs. 4.32 kg, respectively; P < 0.01). No differences were observed on d 46 for the number of fetuses, conceptus attachment length, allantoic + amniotic fluid volume, placental weight, fetal weight, and fetal crown-to-rump length. Variation in fetal crown-to-rump length was less (P < 0.03) in sows fed ad libitum. Sows fed ad libitum had greater (P < 0.01) IGF-I and insulin concentrations in plasma than controls on d 43. In Exp. 2, sows were fed 1.81 kg/d (n = 6) or ad libitum (7.0 +/- 0.11 kg/d; n = 4) from d 30 to 56 of gestation, when sows were anesthetized and samples were collected surgically from their gravid uteri. Sows fed ad libitum gained more weight (P < 0.01) than did controls and had more (P < 0.06) IGF-I in their plasma and the plasma collected from umbilical veins of their fetuses. No differences were found for concentrations of insulin or glucose in plasma of sows or fetuses, but urea N concentrations were greater (P < 0.05) in maternal plasma and in the plasma, and allantoic and amniotic fluids of conceptuses from sows fed ad libitum. Combined data from Exp. 1 and 2 revealed a treatment x fetal number interaction (P < 0.05) for average fetal weight. The expected negative relationship between within-litter average fetal weight and the number of fetuses per uterus was observed for control sows (y = 115.4 -1.75 x fetal number; P < 0.05), but litters of ad libitum sows did not show this effect. The hypothesis that providing feed in excess of established requirements in early gestation affects the in utero milieu is supported by these results. Data further reveal that, at least at mid-gestation, the restraint to fetal growth that is exhibited when fetal number increases in control sows is not exhibited when sows are fed ad libitum.  相似文献

At 110 d of gestation, fetuses were removed from sows selected for high (obese) or for low (lean) backfat thickness. The body weights of lean (1,031 +/- 64 g) and obese (864 +/- 55 g) fetuses were not significantly different. Analysis of muscle composition and of in vitro metabolic characteristics was conducted on the biceps femoris muscle. The percentage of dry weight, protein and glycogen was greater in the muscle of obese fetuses than in the muscle of lean fetuses (P less than .01, P less than .05, and P less than .05, respectively). Percentage of muscle triglyceride was similar (P greater than .05) between the two phenotypes. The rate of glucose oxidation to CO2 tended to be greater (P less than .07) and the rate of lactate production was lower (P less than .05) in the muscle from obese fetuses than in the muscle from lean fetuses. The rates of leucine oxidation to CO2 and of palmitate oxidation to CO2 did not differ between phenotypes. The rate of alpha-ketoisocaproate release from the muscle of obese fetuses was greater (P less than .05) than from that of the lean fetuses. The rate of release of alanine and of glutamine plus glutamate did not differ between phenotypes. The rate of esterification of palmitate did not differ between phenotypes. It was concluded that abnormalities in glucose metabolism and in the partitioning of leucine between oxidation and release as the keto acid existed at 110 d of gestation in the muscle of obese fetuses. Any relation between these differences and ultimate differences in carcass composition were not evident.  相似文献

Fetal intrauterine position relative to the sex of adjacent fetuses has an effect on reproductive performance in rodents. An experiment was conducted to determine whether sex of adjacent fetuses in utero has an influence on fetal and placental weights and whether the hormonal mechanisms documented in rodents are similar in fetal pigs. Sows were slaughtered at 70.1 +/- 1.7 d (n = 123) and 104.5 +/- .05 d (n = 135) of gestation. The fetuses and placentas were removed from the uterus and the position and sex of each fetus was recorded to indicate whether the fetus was between two males, two females, or a male and a female. Fetal blood was sampled for later hormonal analysis. At 70 d of gestation, male fetal and placental weights were heavier than those of females (P less than .05), but no differences were detected relative to the sex of adjacent fetuses. At 104 d of gestation, a fetus surrounded on each side in utero by fetuses of the opposite sex (two males or two females) was lighter in weight than a fetus surrounded by fetuses of the same sex (P less than .01). Differences in fetal weight due to the sex of adjacent fetuses were not related to placental function because placental weights were generally not different at 104 d of gestation. By 104 d of gestation, most placentas were not separated by necrotic regions and were in close apposition with surrounding placentas. Uteroferrin, an Fe-containing, progesterone-induced glycoprotein is involved in maternal to fetal Fe transport in swine. These studies examined the effect of im Fe injection of dam on conceptus and piglet Fe stores. In Exp. I, eight gilts were bred and assigned to either treatment I (no Fe injections) or treatment II (total of 22 mg iron-dextran/kg body weight on d 40, 45, 50, 55 and 60 of gestation) and hysterectomized on d 90 to determine whether Fe injections increased Fe stores in the conceptus. Total Fe in allantoic fluid (P less than .10) as well as uteroferrin concentration (P less than .05) and total uteroferrin (P less than .05) in placentae were greater for gilts in treatment II. In Exp. II, 19 cross-bred sows were bred and assigned to treatments I and II (d 40, 50 and 60 of gestation), as in Exp. I, and treatment III (total of 22 mg iron-dextran/kg body weight on d 90, 100 and 110 of gestation) to determine effects of treatment on hemoglobin (Hb) values of the piglets at 8 +/- 1 h and d 4 postpartum. Piglets from treatment II had higher (P less than .01) Hb at 8 +/- 1 h, but not on d 4 postpartum. Experiment III was a replication of Exp. II except that Hb values were determined at 8 +/- 1 h, d 4 and d 7 postpartum. The objective was to estimate breed, heterosis, and recombination effects on pig reproductive traits in two different four-breed composite populations. Breeds included Yorkshire, Landrace, Large White, and Chester White in Exp. 1 and Duroc, Hampshire, Pietrain, and Spot in Exp. 2. Data were recorded on purebred pigs, two-breed cross pigs, and pigs from generations F1 through F6, where F1 pigs were the first generation of a four-breed cross. Litter traits were considered a trait of the gilt. There were 868 first parity litters in Exp. 1 and 865 in Exp. 2. Direct heterosis significantly increased sow weight at 110 d of gestation and litter weight at 14 and 28 d (weaning) in both experiments. Direct heterosis significantly increased number of nipples, weight at puberty, lactation weight loss, litter size, and litter birth weight in Exp. 2. Gestation length in Exp. 1 and age at puberty in Exp. 1 and Exp. 2 were significantly decreased by direct heterosis. Maternal heterosis significantly increased age at puberty in Exp. 2 and decreased sow weight at 110 d of gestation in Exp. 1. Recombination significantly increased sow weight at 110 d of gestation and tended to increase total number born and litter birth weight in Exp. 1. Recombination significantly decreased age at puberty in Exp. 2. Litter heterosis significantly increased number of pigs at 14 and 28 d; litter weights at birth, 14, and 28 d; and tended to increase lactation weight loss in Exp. 1. Litter heterosis decreased litter size in Exp. 2. Maternal heterosis and recombination effects had a sampling correlation of -0.97 in Exp. 1 and -0.91 in Exp. 2 for number of fully formed pigs. Therefore, maternal heterosis and recombination effects were summed, and their net effect was tested. This net effect tended to increase number of nipples, lactation weight loss, and litter birth weight and significantly increased number of fully formed pigs in Exp. 1. Direct, maternal, and litter heterosis and recombination effects significantly influenced reproductive traits.  相似文献

The development of obesity in porcine fetuses was investigated using a lean and obese strain of pigs at 80, 90, 100 and 110 d of gestation. In absolute terms, fetuses of obese gilts (FO) generally had lower carcass weight and contained less total protein, dry matter and ash than fetuses of lean gilts (FL). In relative terms (percentage of wet carcass weight) FO, compared with FL, generally had decreased percentages of water and increased percentages of protein and lipid. Comparisons based on absolute terms revealed body composition of the strains to be different at 90 d, indicating that factors responsible for obese-type growth were active before that time. Both body composition and hormone concentration differences were most pronounced at later gestation ages. Depressed growth hormone, elevated cortisol, and a tendency toward elevated insulin concentrations in fetal plasma were apparent in late gestation for FO compared with FL. These hormonal patterns are consistent with onset of obesity in FO in late gestation. Greater weights of semitendinosus and longissimus muscles were observed in FL vs FO at 90, 100 and 110 d of gestation (P less than .05). These greater muscle weights were generally accompanied by greater contents of RNA, DNA and protein in FL muscles at these same ages. However, at 80 d, FL had greater absolute DNA content in semitendinosus muscle whereas muscle weight was similar between the strains. This suggests that greater muscle weights for FL than FO were caused by more nuclei in muscle of FL. In general, indices of hypertrophy (protein/DNA) and protein synthetic capacity (RNA/DNA) of muscle were usually similar for both strains at all gestation ages. It is concluded that decreased muscle growth in late gestation of FO compared with FL is more related to fewer total nuclei and perhaps fewer myofibers than to an impaired cellular capacity for protein synthesis.  相似文献

Objectives of this study were to evaluate maternal and fetal influences on development of gravid uterine tissues of cows. Brahman cows with Brahman or Charolais fetuses and Charolais cows with Brahman or Charolais fetuses were used. Cows were killed 232 +/- .5 or 271 +/- .7 d after mating. The gravid uterus of each cow was weighed and dissected into its component parts. Weights of the fetus, fetal membranes, cotyledons, caruncles, and uterus were recorded as were weights of the fetal liver, heart, kidneys, spleen, lungs, stomach complex, intestines, and semitendinosus muscle. Ribonucleic acid, DNA, and protein concentrations in caruncles, cotyledons, liver, heart, kidney, and semitendinosus muscle were determined. Data were analyzed by analysis of variance; breed of cow (C), breed of fetus (F), day of gestation (D), and all interactions were included in the model as fixed effects. Fetal weights were influenced (P less than .003) by C, F, D, and C x D and tended (P = .07) to be influenced by C X F X D. Weight, RNA, DNA, and protein contents of selected fetal tissues followed similar patterns of significance. Thus, both maternal and fetal genotype influenced fetal growth. Greater influences of the maternal system and interrelationships between maternal and fetal systems were observed at the latter stage of gestation. Placentomal (caruncle + cotyledon) weights were greater for Charolais than for Brahman cows (P less than .02) or fetuses (P less than .001) and were greater (P less than .01) at 271 than at 232 d. An ontogeny study of the porcine semitendinosus muscle was conducted to meet two objectives: 1) to evaluate enzyme histochemistry, lectin cytochemistry and actin staining for usefulness as quantitative markers of muscle capillaries and 2) to describe the ontogeny of capillary density changes in developing porcine muscle. Muscle samples were obtained from three to eight crossbred pigs at each of the following ages: newborn, 1 to 2 d and 1, 2, 3, 4.5 and 24 wk. Cryostat sections were stained or reacted for alkaline and neutral phosphatase, dehydrogenase, actin, a panel of nine plant lectins (fluorescein isothiocyanate conjugated) and routine cytochemical tests for muscle fiber typing. Capillaries were quantitatively marked by reactions for dehydrogenase activity in the young pigs but not in the oldest animals. The lectins, soybean agglutinin, Bauhinia purpuria agglutinin, Ulex europeus agglutinin-I and Griffonia simplicifolia agglutinin-I (GS-I) quantitatively stained capillaries at all ages. Histological observations of thin sections of epon-embedded tissues served to validate the lectin and cytochemical capillary staining. Micrographs of sections stained with the lectin GS-I were used to count capillaries and muscle fibers so the capillary:fiber ratio (C/F) could be calculated. Deep (red) and superficial (white) aspects of muscle sections had different C/F at birth, 2, 4 and 24 wk. The deep aspects (DA) had higher C/F than superficial (SA) aspects (at all four ages), and there were age-dependent increases (P less than .001) in C/F of DA and SA. The effects of adrenergic agonists and antagonists on blood pressure and heart rate were investigated in 18 chronically catheterised pig fetuses aged between 102 and 109 days of gestation (term is 114 days). One fetus had been decapitated in utero at 42 days of gestation. The alpha adrenergic agonist methoxamine produced a small but dose dependent hypertension and a dose related slowing in heart rate. The beta adrenergic agonist isoprenaline decreased mean arterial pressure in a dose related manner and produced tachycardia. Propranolol, a beta adrenergic blocker, increased mean arterial pressure and decreased heart rate. The response to subsequent alpha adrenergic blockade with phentolamine was hypotension and a slight bradycardia. Decapitation at 42 days of gestation did not seem to change the fetal responsiveness to adrenergic agonists but removed the blood pressure response to beta blockade. These observations indicate that the heart and circulation of the pig fetus are under adrenergic vasomotor control during late gestation.  相似文献

Influence of initial length of uterus available to each embryo on its subsequent survival and development was determined by systematic restriction of the length available to each potential embryo. Fifty-seven pregnant crossbred gilts were laparotomized at d 3 of gestation, length of uterine horns was measured in situ and corpora lutea (CL) were counted. In Exp. 1, uterine space available to each potential embryo was restricted by ligating one uterine horn 5 cm from the tip per CL. Uteri were examined at d 20, 25 or 50. In Exp. 2, one uterine horn was ligated on d 3 at 10, 20 or 30 cm from the tip per CL and uteri were examined at d 50. Embryos in the restricted section (RS) had a specific mean uterine length available to each potential embryo of 5, 10, 20 or 30 cm. Embryos in the nonrestricted section (NRS) had a variable mean uterine length available to each potential embryo of 44 +/- 4 cm. When embryos were restricted to 5 cm, the proportion of surviving fetuses at d 20, 25 and 50 was 61, 12 and 8%, respectively, whereas in combined NRS it was 82%. When the uterus was examined at d 50 after restricting embryos to 10, 20 or 30 cm/CL, 25, 33 and 52% of fetuses survived; in combined NRS survival was 71%. Each fetus surviving to d 50 in RS was associated with 36 cm of initial uterine length but fetal survival was not associated with number of CL. Two anti-adipocyte monoclonal antibodies (MAbs: AD-1 and AD-2) have been used to study the development of dorsal s.c. adipose tissue in fetuses from 50 to 110 d of gestation. Immunofluorescent staining of cryostat sections with each antibody revealed antigen-positive cells in fetal s.c. mesenchyme prior to lipid deposition. Lipid droplets as well as AD-1 and AD-2 positive cells were detected within the underlying muscle at 50 d. From 70 to 110 d of development, the AD-1 and AD-2 MAbs each detected all adipocytes examined, as well as capillaries associated with fat cell clusters in s.c. tissues. Reactivity toward both antibodies, as well as lipid deposition, also was detectable in the muscle underlying the s.c. mesenchyme from 70 d onward. Each MAb possessed a distinct pattern of reactivity. The AD-2 MAb stained arrector pili muscles and vessels in the s.c. mesenchyme and vessels in the underlying muscles, whereas the AD-1 did not. No reactivity using either MAb was detectable toward any other cell types within s.c. tissues. These results established the presence of cells expressing surface determinants found on mature adipocytes and associated capillaries prior to adipogenesis. A lineage relationship between adipocytes and capillary endothelial cells is suggested.  相似文献

The objective of these studies was to evaluate the effects of dietary restriction and Se on maternal and fetal metabolic hormones. In Exp. 1, pregnant ewe lambs (n = 32; BW = 45.6 +/- 2.3 kg) were allotted randomly to 1 of 4 treatments. Diets contained (DM basis) either no added Se (control), or supranutritional Se added as high-Se wheat at 3.0 mg/kg (Se-wheat), or sodium selenate at 3 (Se3) and 15 (Se15) mg/kg of Se. Diets (DM basis) were similar in CP (15.5%) and ME (2.68 Mcal/kg). Treatments were initiated at 50 +/- 5 d of gestation. The control, Se-wheat, Se3, and Se15 treatments provided 2.5, 75, 75, and 375 microg/kg of BW of Se, respectively. Ewe jugular blood samples were collected at 50, 64, 78, 92, 106, 120, and 134 d of gestation. Fetal serum samples were collected at necropsy on d 134. In Exp. 2, pregnant ewe lambs (n = 36; BW 53.8 +/- 1.3 kg) were allotted randomly to treatments in a 2 x 2 factorial arrangement. Factors were nutrition (control, 100% of requirements vs. restricted nutrition, 60% of control) and dietary Se (adequate Se, 6 microg/kg of BW vs. high Se, 80 microg/kg of BW). Selenium treatments were initiated 21 d before breeding, and nutritional treatments were initiated on d 64 of gestation. Diets were 16% CP and 2.12 Mcal/kg of ME (DM basis). Blood samples were collected from the ewes at 62, 76, 90, 104, 118, 132, and 135 d of gestation. Fetal blood was collected at necropsy on d 135. In Exp.1, dietary Se source and concentration had no effect (P > 0.17) on maternal and fetal serum IGF-I, triiodothyronine (T(3)), or thyroxine (T(4)) concentrations. Selenium supplementation increased (P = 0.06) the T(4):T(3) ratio vs. controls. In Exp. 2, dietary Se had no impact (P > 0.33) on main effect means for maternal and fetal serum IGF-I, T(3), or T(4) concentrations from d 62 to 132; however, at d 135, high-Se ewes had lower (P = 0.01) serum T(4) concentrations than adequate-Se ewes. A nutrition by Se interaction (P = 0.06) was detected for the T(4):T(3) ratios; ewes fed restricted and adequate-Se diets had greater (P = 0.10) T(4):T(3) ratios compared with the other treatments. Nutrient-restricted ewes had lower (P < 0.05) serum IGF-I, T(3), and T(4) concentrations. Fetal serum IGF-I concentrations were lower (P = 0.01) in restricted-vs. control-fed ewes; however, fetal T(3) and T(4) concentrations were unaffected (P > 0.13) by dietary Se or maternal plane of nutrition. These data indicate that dietary Se may alter maternal T(4):T(3) ratios. In addition, nutrient restriction during gestation reduces maternal IGF-I, T(3), and T(4) and fetal IGF-I concentrations.  相似文献

Five experiments were conducted to test the effects of various dietary humic substances (HS; HS1, 2, 3, and 4, each with different fulvic and humic acid contents) on pig growth, carcass characteristics, and ammonia emission from manure. In Exp. 1, 120 pigs were allotted to 3 dietary treatments without HS (control) or with HS1 at 0.5 and 1.0% (8 pens/treatment and 5 pigs/pen) and fed diets, based on a 5-phase feeding program, from weaning (d 21.3 +/- 0.3 of age) to 60 kg of BW. In Exp. 2 and 3, 384 pigs (192 for each experiment) were allotted to 3 dietary treatments without HS, with HS1, or with HS2 (0.5%) for Exp. 2 and without HS, or with HS3 or HS4 (0.5%) for Exp. 3 (8 pens/treatment and 8 pigs/pen in each experiment). Pigs were fed diets, based on a 6-phase feeding program, from weaning (25.4 +/-0.2 and 23.6 +/-0.3 d of age for Exp. 2 and 3, respectively) to 110 kg of BW. In Exp. 4, 96 pigs were weaned at 22.1 +/-0.2 d of age and allotted to 2 treatments without or with HS1 at 0.5% (6 pens/treatment and 8 pigs/pen), and in Exp. 5 96 pigs were weaned at 20.9 +/-0.3 d of age and allotted to 3 treatments without HS, or with HS3 or HS4 (0.5%; 4 pens/treatment and 8 pigs/pen). Pigs were fed the diets for at least 2 wk before they were moved to an environmental chamber to measure aerial ammonia and hydrogen sulfide for 48 h at 5-min intervals. In Exp. 1, pigs fed diets with HS1 at 0.5% had greater (P < 0.05) ADG during phase 3 and greater (P < 0.05) G:F during phases 3 and 5 than control pigs. In Exp. 2, pigs fed diets with HS1 or HS2 at 0.5% had greater (P < 0.05) ADG and G:F than control pigs during the entire feeding period, whereas in Exp. 3 HS3 or HS4 did not improve pig growth performance. Ammonia emission from manure was reduced by 18 or 16% when pigs were fed diets with HS1 (P = 0.067) or HS4 (P = 0.054), respectively. The results of this study indicate that the effects of dietary HS are variable but may improve growth performance of pigs and reduce ammonia emission from manure. Further research is needed to clarify these effects and the mechanisms by which HS may cause them.  相似文献

Fetal pigs in one uterine horn of each of five gilts were hypophysectomized (HX) in utero by electrical cauterization at 72-74 days of gestation and sera collected at 110 days of gestation. Sera from HX fetuses had lower levels of insulin-like growth factor-1 compared to control littermates (P less than .05). Sera were tested for their effects on primary cultures of stromal-vascular cells from adipose tissue. The soluble protein concentration/dish was lower when pig cells were cultured in sera from HX fetuses compared to sera from control fetuses (P less than .01). Sera from HX fetuses inadequately supported growth of stromal-vascular cells so subsequent experiments utilized pooled sera from normal and HX adult pigs. Sera from HX and control fetuses were mixed with sera from the two adult pools and tested for incorporation of tritiated thymidine into rat preadipocytes and the appearance of adipocytes (determined histochemically) in pig stromal-vascular cultures. In cultures fed sera from HX fetuses there was a lower (P less than .05) number of pig fat cells/culture and a lower level (P less than .06) of preadipocyte proliferation in rat cell cultures when compared to control fetal sera. Fetal pig serum contains factors (adipogenic) which promote the proliferation and differentiation of adipocytes in culture. Serum from HX fetuses has a lower level of adipogenic factors.  相似文献
