桃蚜高效氯氰菊酯抗药性与乙酰胆碱酯酶的关系 总被引:3,自引:0,他引:3
于室内对桃蚜进行高效氯氰菊酯抗药性筛选,选育至10代后抗性倍数增长到49.9倍。生化分析表明,抗性品系乙酰胆碱酯酶(AChE)活性均显著高于敏感品系。比较两个品系乙酰胆碱酶活性个体频率分布发现,更多的桃蚜个体向酶活性高的区域分布。酶动力学测定结果显示,抗性桃蚜酯酶对底物的Vmax、Km显著大于敏感品系。 相似文献

对桃蚜进行室内噻虫嗪抗性品系筛选,选育至15代后抗性倍数达到75.6倍。对噻虫嗪敏感品系(THI-S)和抗性品系(THI-R)桃蚜的谷胱甘肽S-转移酶(GSTs)、酸性磷酸酯酶(ACP)、碱性磷酸酯酶(ALP)、羧酸酯酶(CarE)、多功能氧化酶(MFO)O-脱甲基活性进行了比较,结果显示:敏感品系(THI-S)和抗性品系(THI-R)的谷胱甘肽S-转移酶比活力分别为3.127 5和3.215 9,差异不显著,桃蚜抗性品系体内酸性磷酸酯酶、碱性磷酸酯酶、羧酸酯酶和多功能氧化酶O-脱甲基活性均显著高于敏感品系,分别达到了1.57、2.10、6.12、2.03倍。表明桃蚜对噻虫嗪抗性的产生与酸性磷酸酯酶、碱性磷酸酯酶、羧酸酯酶和多功能氧化酶O-脱甲基的活性相关。 相似文献

The susceptibility of 88 and 38 field samples of Myzus persicae (Sulzer) to imidacloprid and deltamethrin respectively was examined using the FAO dip test bioassay. The field samples were collected from tobacco and peach from various regions of Greece in the period from 2004 to 2006. In addition, 497, 349 and 370 clones originating from peach and tobacco were screened for the three known resistance mechanisms, elevated esterases, modified acetylcholinesterase (MACE) and knockdown resistance (kdr) respectively, using biochemical and DNA diagnostics. Most of the samples assayed with imidacloprid showed low resistance factors (RFs)-39% below 5 and 21% between 5 and 10. However, 9% of the samples (all from tobacco) showed relatively high RF values (24-73). Differences were found between crops, with higher RF values recorded in samples from tobacco than in those from peach. Bioassays with deltamethrin revealed the development of strong resistance in the populations examined. The RFs were mostly higher than 23, and in 29% of the samples they were extremely high (152-436). Finally, the three known resistance mechanisms were found in high frequencies in the populations examined, although some differences between crops and years were detected. The implications of the study for management schemes against M. persicae are discussed. 相似文献

以甘肃省宕昌县桃蚜为敏感种群,测定了兰州、永登等地桃蚜对6种杀虫剂抗药性。结果表明兰州市安宁区桃蚜对溴氰菊酯已产生低水平抗性(6.84倍),对氰戊菊酯则处于耐药力增高或敏感性降低阶段,对灭多威、敌敌畏、氧乐果和乐果已显示出早期抗性;永登县桃蚜对溴氰菊酯和氰戊菊酯处于敏感性降低阶段,对灭多威、敌敌畏、氧乐果和乐果仍属敏感阶段;皋兰县和榆中县桃蚜对上述6种杀虫剂则处于敏感阶段。用酶抑制剂进行增效作用测定结果显示,兰州地区桃蚜的抗药性与羧酸酯酶和多功能氧化酶有关。针对兰州市桃蚜的抗药性现状和抗性机制,提出了以建立抗性监测制度、合理用药为主的抗性治理对策。 相似文献

Treatment of the lowermost leaf of flowering plants of oilseed rape cv. Linetta with 20 m m oxalic acid induced systemic disease resistance in the three next youngest leaves. Resistance was not caused by the translocation of fungitoxic concentrations of oxalic acid. Resistance was also expressed in the stem; in oxalic acid-treated plants the vertical spread of stem lesions was halted and the signal travelled both acropetally and basipetally. Expression of resistance in the leaves was detected 6 h after treatment and continued for at least 5 weeks. Twenty-four h after treatment, resistance was detectable in the leaves following the use of as little as 0.5 m m oxalic acid. Local resistance in the oxalic acid-sprayed leaf was detectable only at certain times (12 and 48 but not 24 h after treatment) and at very low (0.25 m m ) or high (≥40 m m ) oxalic acid concentrations. When oxalic acid was applied to a discrete area of the leaf, significant local resistance was expressed in the surrounding leaf tissue, maximum resistance being exhibited by the tissue closest to the site of petiole attachment. 相似文献

BACKGROUND: The accuracy of predicting the survival of insecticide‐resistant aphids following the application of commonly used insecticides from the carbamate, the pyrethroid, a mix of the two or the neonicotinoid chemical classes was evaluated in a potato field in Scotland. Equal proportions of five genotypes of the peach‐potato aphid, Myzus persicae (Sulzer), with none, resistance to dimethyl‐carbamates, resistance to pyrethroids or combinations conferring resistance to both chemical classes were released into potato field plots. The insecticides were sprayed separately onto these plots, the aphid populations were analysed after 6–8 days and the process repeated. RESULTS: For each assessment after the three separate spray events, plots treated with the carbamate had 48, 147 and 28%, those treated with pyrethroid 53, 210 and 89%, those treated with carbamate/pyrethroid 28, 108 and 64% and those treated with neonicotinoid 43, 55 and 11% of the numbers of M. persicae by comparison with untreated controls. Only the proportions of surviving aphids from the genotype containing no insecticide resistance traits and the genotype containing elevated carboxylesterases matched ratios predicted from the selective advantage afforded by the resistance traits alone. Survival of aphids from the other three genotypes that carried 1–3 of the insecticide resistance traits differed from expectations in all cases, possibly owing to physiological differences, including their vulnerability to predators and hymenopterous parasitoids present at the site and/or their carrying unknown insecticide resistance mechanisms. CONCLUSION: Control strategies based on knowledge of the genetically determined insecticide resistance profile of an M. persicae population alone are insufficient. Experiments were done on commercial potato crops in the UK to investigate the effect of different insecticide sequences on the control and insecticide resistance status of Myzus persicae (Sulzer). The work was done to provide field validation of similar laboratory studies done in 'field simulators'. To ensure adequate aphid populations and to influence the initial resistance status of the aphid population, cultured M. persicae from a clone of known resistance status (esterase R1, kdr heterozygote, non-MACE (modified acetylcholinesterase)) were inoculated into both experiments. Two-spray programmes starting with lambda-cyhalothrin (a pyrethroid insecticide) gave poor control in comparison with programmes starting with pirimicarb (a carbamate insecticide) or pirimicarb-containing mixtures. This concurred closely with the results obtained from single applications in field simulator studies. Treatment sequences containing pymetrozine (a pyridine azomethine insecticide) were also effective, though slower-acting. This again concurs with field simulator studies. The proportions of aphids carrying different resistance mechanisms were largely unaffected by treatment in these experiments. The peach-potato aphid Myzus persicae (Sulzer) has developed a number of insecticide resistance mechanisms owing to the high selective pressure produced by world-wide insecticide treatments. Knowledge of the geographical distribution and the temporal evolution of these resistant phenotypes helps to develop suitable pest-management programs. Current understanding of the major mechanisms of resistance at the molecular level makes it possible to diagnose the presence of modified acetylcholinesterase (MACE) or knockdown resistance (kdr). This paper describes a rapid method for the identification of both resistance mechanisms in a single molecular assay by using restriction fragment length polymorphism of PCR products (RFLP-PCR) in individual as well as pooled aphids. 相似文献

A range of insecticides was applied at recommended application rates against populations of Myzus persicae (Sulzer) carrying various combinations of three insecticide resistance mechanisms (carboxylesterase-based metabolic resistance and two target-site mechanisms, known as MACE and kdr), supported on either Chinese cabbage or potatoes in field simulator cages. Patterns of response were similar on both host species. MACE conferred extreme resistance to pirimicarb and triazamate (dimethylcarbamate insecticides). The kdr mechanism was associated with resistance to lambda-cyhalothrin, cypermethrin and deltamethrin (pyrethroid insecticides). A mixture of pirimicarb plus lambda-cyhalothrin was only effective against M persicae not carrying kdr or carrying kdr and low carboxylesterase-based resistance. None of the insecticides tested was effective against M persicae carrying both MACE and kdr resistance. The implications of these findings for the formulation of control strategies, based on regular monitoring of resistance genotype frequencies, are discussed. 相似文献

油菜根肿病在江西省多市均有分布, 尤以上饶婺源、吉安永新、九江瑞昌等地发生严重。为明确江西省油菜主产区根肿菌的生理小种, 采用SCD鉴定系统对江西油菜主产区根肿菌进行了生理小种鉴定, 结果共鉴定出3个生理小种类型:Pb1、Pb5和Pb12。为筛选出适于江西根肿病发病地区种植的油菜材料, 开展了油菜材料对根肿菌的抗性测定。室内盆栽抗性测定表明, 育种中间材料‘P360429001’‘2018362481’‘P360924009’‘P360281040’‘中油杂12’‘华油杂62R’和‘华双5R’的病情指数均低于或等于30.00, 其中‘华油杂62R’和‘华双5R’表现为中抗水平。田间抗性测定结果表明, ‘华油杂62R’‘秦优DK4’‘阳光50’和育种中间材料‘P360429001’‘ZS115R’的根肿病病情指数均低于30.00, 但只有‘华油杂62R’对Pb1生理小种表现出中抗水平。本研究针对性地筛选出适于江西油菜根肿病发病地区种植的油菜材料, 对江西油菜根肿病的防控和创制抗病性油菜资源提供了科学依据。 相似文献

采用毛细管点滴法测定了我国西南烟区云南和贵州省的10个桃蚜Myzus persicae(Sulzer)种群对氰戊菊酯、氧化乐果、灭多威和吡虫啉的抗性。结果表明,云南楚雄种群对氰戊菊酯的抗性高达26.87倍,对氧化乐果、灭多威、吡虫啉的抗性分别为敏感种群的10.02、6.00和5.83倍。云南大理、丽江、石林和曲靖4个种群对氰戊菊酯的抗性分别达24.98、14.94、11.11和10.33倍,抗性水平也较高,但对氧乐果、灭多威和吡虫啉仍较为敏感,应尽早实施抗性治理措施。贵州省的5个种群仍然处在敏感阶段。 相似文献

BACKGROUND: Pollen beetle, Meligethes aeneus F. (Coleoptera: Nitidulidae), is a major pest in European winter oilseed rape. Recently, control failures with pyrethroid insecticides commonly used to control this pest have been reported in many European countries. For resistance management purposes, the neonicotinoid insecticide thiacloprid was widely introduced as a new mode of action for pollen beetle control. RESULTS: A number of pollen beetle populations collected in Germany, France, Austria, Great Britain, Sweden, Denmark, Finland, Poland, Czech Republic and Ukraine were tested for pyrethroid resistance using lambda‐cyhalothrin‐coated glass vials (adult vial test). Most of the populations tested exhibited substantial levels of resistance to lambda‐cyhalothrin, and resistance ratios ranged from < 10 to > 2000. A similar resistance monitoring bioassay for the neonicotinoid insecticide thiacloprid was developed and validated by assessing baseline susceptibility data for 88 European pollen beetle populations. A variation of less than fivefold in response to thiacloprid was detected. The thiacloprid adult vial bioassay is based on glass vials coated with an oil‐dispersion‐based formulation of thiacloprid, resulting in a much better bioavailability compared with technical material. Analytical measurements revealed a > 56 and 28 day stability of thiacloprid and lambda‐cyhalothrin in coated glass vials at room temperature, respectively. No cross‐resistance between thiacloprid and lambda‐cyhalothrin based on log‐dose probit–mortality data was detected. CONCLUSION: Pyrethroid resistance in many European populations of M. aeneus was confirmed, whereas all populations are susceptible to thiacloprid when tested in a newly designed and validated monitoring bioassay based on glass vials coated with oil‐dispersion‐formulated thiacloprid. Based on the homogeneous results, it is concluded that thiacloprid could be an important chemical tool for pollen beetle resistance management strategies in European winter oilseed rape. 以地丁草的全草为试材,在室内测定了不同提取方法和不同溶剂提取的地丁草提取物对桃蚜的生物活性。结果表明:不同提取方法和不同溶剂的提取物对桃蚜的毒力有显著差异。组合中以石油醚为溶剂的地丁草索氏提取物对桃蚜触杀活性最高,在浓度为100 g/L时,12、24、48 h的校正死亡率分别达80.00%、96.67%、98.33%,24 h的致死中浓度LC50达22.50 g/L;忌避作用试验表明,处理24、48、72 h后,地丁草索氏提取物对桃蚜的忌避率分别为88.75%、83.33%、83.33%。表明以石油醚为溶剂的地丁草索氏提取物对桃蚜有较高的触杀和忌避活性。 相似文献

BACKGROUND: Failures in pollen beetle control using pyrethroids since 2005 indicated pyrethroid resistance in Germany. Therefore, resistance monitoring using bioassays was established in Germany for oilseed rape pest insects. RESULTS: The spread and intensity of pyrethroid resistance of Meligethes aeneus increased from 2005 onwards, with no sensitive samples left in any region of Germany in 2011. Sensitivity also declined for the newly introduced actives bifenthrin, etofenprox (both class‐I pyrethroids) and tau‐fluvalinate; all three claimed to be less affected by resistance, although there was no clear cross‐resistance to lambda‐cyhalothrin (class‐II pyrethroid). In the German region with the longest tradition and high intensity of oilseed rape production, pyrethroid resistance of Psylliodes chrysocephala and Ceutorhynchus obstrictus, with resistance factors of up to 81 and 140 respectively, was detected. CONCLUSION: The intensive use of only one mode of action for many years is risky, because even pest insects with a low intrinsic resistance risk may develop resistance.
