This study was undertaken to evaluate the milk protein response when cows were supplied a balanced AA profile and to determine whether a deficiency of Leucine (Leu) or Arginine (Arg) had a negative effect on milk protein. Eight mid‐lactation Holstein cows were randomly assigned to 5‐day continuous jugular infusions of saline (CTL), EAA mixture prepared on the profile of casein and supplied (in % of lysine (Lys)) 100% of Lys, 33.3% of methionine (Met), 110.2% of Leu, 43.6% of Arg, 50.8% of threonine (Thr), 81.6% of valine (Val), 69.7% of isoleucine (Ile), 61.4% of phenylalanine (Phe) and 34.2% of histidine (His) (Casein, 160 g/d), EAA mixture excluding Leu (?Leu, 163 g/d) or EAA mixture excluding Arg (?Arg, 158 g/d) in a duplicated 4 × 4 Latin square design with four infusion periods separated by 7‐day interval period. The basal diet supplied 1.6 Mcal NEL and 94.4 g MP per 1 kg DM to meet requirements for lactation. The Casein treatment provided a balanced supply (in % of MP) of 10.3% Leu and 5.3% Arg, whereas in the two subsequent ?Leu and ?Arg treatments, the concentration of Leu and Arg was reduced to 8.4 and 4.6% respectively. Dry matter intake (15.4 kg/day) was not affected by treatments. The Casein treatment increased milk yield (14.9%, p < 0.001), milk protein yield (120 g, p < 0.001) and milk protein efficiency (0.03, p = 0.099) than CTL treatment. However, the ?Leu treatment decreased the responses of above‐measured parameters by 6.25%, 70 g, 0.05 (p < 0.06) (compared with Casein). These effects of Leu were related to decreased Leu concentration and improved concentration of Ile and Val in plasma. The ?Arg treatment decreased the plasma Arg concentration than the Casein treatment, whereby resulted in the decrease of milk yield (5.7%, p = 0.073), milk protein yield (60 g, p = 0.011) and milk protein efficiency (0.04, p = 0.037). In conclusion, supply of EAA profile of casein can increase the lactation production in dairy cows, and 8.6% of Leu in MP partly limits the milk protein response when the requirements of Lys, Met and His were met. The level of Arg at 4.6% MP is not deemed to an ideal profile, as evidenced by decreased milk protein efficiency.  相似文献

1. The dietary limitation of isoleucine (Ile) and valine (Val) was evaluated in broilers. Common diets were given to all broilers from 0 to 28 d of age. Experimental diets were provided from 28 to 42 d of age.

2. The experimental test diet was based primarily on maize, soybean meal, and meat and bone meal, and formulated to satisfy all nutrient recommendations with the exception of Ile and Val.

3. Diets were supplemented with L-Ile and L-Val at either 0·5 or 1·0 g kg?1 of diet, alone or in combination. A diet composed only of commercially available amino acids and formulated to satisfy all nutrient requirements served as a positive control.

4. Body weight gain was depressed by the test diet, but recovered to a weight similar to birds fed on the positive control when Val was added to the test diet either alone or in combination with Ile. Feed efficiency improved when Val and Ile were simultaneously supplemented.

5. Abdominal fat yield was reduced with the combined supplementation of Val and Ile. Breast meat yield appeared to be more sensitive to Ile supplementation than to Val.

6. The results suggest that live performance may be more sensitive to Val, while breast meat yield was maximised with Ile supplementation. However, their combination is required for optimised growth and meat yield.  相似文献

The effects of Ile and Val supplementation of a low-CP, corn-wheat-soybean meal-based piglet diet on growth performance, incidence of diarrhea, and N balance were studied using 60 Landrace x Duroc male piglets in a 4-wk experiment. The 60 individually caged piglets were divided into 5 dietary treatments, each consisting of 12 piglets. Diet 1 was a positive control diet (20% CP); diet 2 was a low-CP negative control diet (17% CP); diets 3, 4, and 5 were low-CP diets to which Ile, Val, or the combination of Ile and Val were added, respectively. All diets were supplemented with Lys, Met, Thr, and Trp to provide the required concentrations of these AA according to the 1998 NRC. Average daily gain and ADFI were similar among pigs fed the positive control, Val-added, and the Val plus Ile-added diets. On wk-2 and wk-4, fecal score was greater (softer feces) in piglets fed the 20% CP level compared with the remaining treatments (P < 0.01). Nitrogen intake was decreased (P < 0.0001) in pigs fed diets containing low levels of CP compared with pigs fed the 20% CP diet. Fecal N excretion (g/d) was decreased (P < 0.05) in piglets fed low-CP diets at wk 1 and wk 4 of feeding, and in urine at wk 4 of feeding. Crude protein levels or AA supplementation had no effect on N retention efficiencies. These results indicate that the supplementation of Val alone, or in combination with Ile, to a low-CP piglet diet with adequate levels of Lys, Met, Thr, and Trp is necessary to achieve maximum performance in pigs consuming corn-wheat-soybean meal-based diets.  相似文献

An experiment was conducted to evaluate the effects of B. subtilis RX7 and B. subtilis B2A on growth performance, blood profiles, intestinal Salmonella population, noxious gas emission, organ weight and breast meat quality of broilers under S. typhimurium challenge. A total of 120, one‐day‐old Ross 308 male broiler chicks were assigned to four dietary treatments, composed of six replications, with five birds per replication, for 10 day. The dietary treatment groups were negative control (NC; no antibiotic, no B. subtilis), positive control (PC; NC + 0.1% virginiamycin), B. subtilis RX7 (NC + 0.1% B. subtilis RX7 1.0 × 10cfu/g) and B. subtilis B2A (NC + 0.1% B. subtilis 1.0 × 109 cfu/g). All birds were orally challenged with 2 ml suspension, containing 104 cfu/ml of S. typhimurium KCCM 40253. Results indicated that the body weight gain, feed intake and feed conversion did not differ, among all comparative treatments. Serum haptoglobin concentration was lower in Bacillus treatments (RX7 + B2A) than the NC treatment (p < 0.05). Intestinal and excreta Salmonella number, and excreta ammonia gas emission in the PC treatment or Bacillus treatments, was lower than the NC treatment (p < 0.05). Breast pH, colour and water‐holding capacity were not affected by supplementation of B. subtilis RX7 and B2A. However, drip loss at 1 day post‐slaughter from birds fed with B. subtilis RX7 and B2A decreased, compared with the positive control birds (p < 0.05). Relative gizzard weights of birds fed B. subtilis RX7 and B2A were significantly higher than the NC birds under S. typhimurium challenge. It is concluded from the results that B. subtilis RX7 and B2A increased the gizzard weight and decreased the intestinal and excreta Salmonella population and excreta ammonia gas, and drip loss of breast meat after being stored for 1 day, under stress caused by the S. typhimurium challenge.  相似文献

The present study investigated the influence of Bacillus subtilis GCB‐13‐001 on growth performance, nutrient digestibility, blood characteristics, faecal microbiota and faecal score in weanling pigs. A total of 120 weaning pigs [(Landrace × Yorkshire) × Duroc; 7.73 ± 0.75 kg (28 days of age)] were randomly allotted into three treatments according to their initial body weight (BW) and gender in a 6‐week experiment. There were 8 replication pens in each treatment, with five pigs/pen. Dietary treatment groups were as follows: (a) basal diet (CON), (b) CON + 0.1% Bacillus subtilis GCB‐13‐001 1 × 108 CFU/kg (T1) and (c) CON + 0.1% Bacillus subtilis GCB‐13‐001 1 × 109 CFU/kg (T2). Days 1 to 7, the BW and ADG with T2 treatment were higher (p < .05) than CON treatment, as well as F:G showed trends in linear reduction (p < .1). Days 8 to 21, the BW and ADG were improved (p < .05) in pigs offered T1 and T2 diets compared with CON diet. Days 22 to 42, BW and ADG were higher (p < .05) in pigs fed T2 diet than CON and T1 diets, and the pigs fed T1 diet had higher BW than CON treatment. Overall, the ADG with the T2 treatment was higher (p < .05) than that with the T1 and CON treatments, and pigs offered T1 treatment had higher (p < .05) ADG than CON treatment. Moreover, F:G ratio were significantly decreased (p < .05) by T2 treatment compared with CON treatment. The faecal Lactobacillus counts were improved, and E. coli counts were reduced (p < .05) in pigs fed T2 diet compared with CON at the end of the experiment. In conclusion, supplementation of 0.1% Bacillus subtilis GCB‐13‐001 1 × 109 CFU/kg has shown a beneficial effect in improving BW, increase ADG, decrease F:G ratio.  相似文献

This study aimed to investigate the effect of dietary supplementation with Lactobacillus plantarum and Bacillus subtilis on growth performance, apparent nutrient digestibility and stress‐related indicators in dairy calves. Twenty‐four neonatal Holstein calves were randomly allocated to three treatments: a basal diet with no supplementation (control), the basal diet supplemented with 1.7 × 1010 CFU per head per day (CFU/h.d) of L. plantarum GF103 (LB group) or the basal diet supplemented with a mixture of L. plantarum GF103 (1.7 × 1010 CFU/h.d) and B. subtilis B27 (1.7 × 108 CFU/h.d) (LBS group). Dry matter intake (DMI), average daily gain (ADG), feed conversation ratio (FCR), apparent digestibility of nutrients and stress‐related indicators were measured in this trail. The result indicated that no significant differences were observed in DMI or ADG (p > 0.05), but the FCR was improved in the LB group over the first 12 weeks (p > 0.05). The apparent digestibility of nutrients was not altered by probiotics in week 6 (p > 0.05), but the apparent digestibility of total phosphorus was significantly greater in the LB and LBS groups in week 8 (p > 0.05); additionally, an increase in the apparent digestibility of crude protein was detected in the LBS group (p > 0.05). Oral administration of L. plantarum alone improved the T‐lymphocyte transformation rate on days 58 and 62 (p > 0.05), while adding the mixture of L. plantarum and B. subtilis increased the T‐lymphocyte transformation rate (p > 0.05) but decreased the content of cortisol on day 58 (p > 0.05). No significant differences were detected between the LB and LBS groups in growth performance, apparent digestibility of nutrients and stress‐related indicators (p > 0.05). The results suggested that oral administration of L. plantarum improved growth performance, nutrient digestibility and relieved weaning stress in calves, but no additional effect was obtained by supplementation with B. subtilis.  相似文献

Based on results of a recent meta-analysis, we hypothesized that increased dietary Val, Ile, or Trp could correct possible amino acid interactions because of excess Leu in diets containing high levels of corn protein, namely dried distiller’s grains with solubles (DDGS). A total of 1,200 pigs (PIC TR4 × (Fast LW × PIC L02); initially 33.6 ± 0.6 kg) were used in a 103-d study. The 6 dietary treatments were corn–soybean meal (SBM)-DDGS-based as follows: (1) high SBM and low level of l-Lys HCl (HSBM), (2) high l-Lys HCl and moderate Ile, Val, Trp (AA above NRC 2012 estimates; NC), (3) moderate l-Lys HCl and high Ile, Val, and Trp (PC), and PC with either increased (4) L-Val (PC+Val), (5) L-Ile (PC+Ile), or (6) L-Trp (PC+Trp). Pigs fed the NC diet were predicted to have the poorest average daily gain (ADG), the PC diet to be intermediate, and pigs fed the HSBM, PC+Val, PC+Ile, and PC+Trp have the same and highest predicted ADG. In the grower period (34 to 90 kg), ADG was greater (Ρ < 0.05) for the pigs fed HSBM and PC+Val diets than the NC with pigs fed other diets intermediate. Pigs fed HSBM were more (Ρ < 0.05) efficient (G:F) than the NC and PC with pigs fed other diets intermediate. In the finisher period (90 to 136 kg), ADG was greater (Ρ < 0.05) for pigs fed PC+Ile than that of the NC with pigs fed other diets intermediate. Pigs fed PC+Val had greater (Ρ < 0.05) average daily feed intake (ADFI) than the NC with pigs fed other diets intermediate. However, PC+Ile pigs were more (Ρ < 0.05) efficient than PC+Val with pigs fed other diets intermediate. Overall, ADG was greater (Ρ < 0.05) for pigs fed HSBM, PC+Val, and PC+Ile diets than the NC with pigs fed other diets intermediate. Pigs fed the PC+Val diet had greater (Ρ < 0.05) ADFI than the NC with pigs fed other diets intermediate. No differences were detected between treatments for overall G:F or other carcass characteristics. In conclusion, increasing Val or Ile in high l-Lys-HCl-DDGS-based diets improved growth performance compared with pigs fed diets containing high levels of l-Lys HCl without added Val and Ile. These results present evidence that the recently developed meta-analysis can predict the relative differences in overall ADG for pigs fed the NC, PC, PC+Val, and PC+Ile diets; however, the predicted G:F was less accurate. The data demonstrate that the negative effects of high Leu concentrations in corn-DDGS-based diets can be reversed by increasing the ratios of Val and Ile relative to Lys.  相似文献

We conducted a meta‐data analysis to develop prediction equations to estimate enteric methane (CH4) emission from beef cattle in Southeast Asia. The dataset was obtained from 25 studies, which included 332 individual observations on nutrient intakes, digestibilities, and CH4 emissions. Cattle were provided tropical forage or rice straw, with or without concentrates in individual pens equipped with indirect open‐circuit head hood apparatus. The simplest and best equation to predict daily CH4 emission was CH4 (g/day) = 22.71 (±1.008) × dry matter intake (DMI, kg/day) + 8.91 (±10.896) [R2 = 0.77; root mean square error (RMSE) = 19.363 g/day]. The best equation to predict CH4 energy as a proportion of gross energy intake (CH4‐E/GEI, J/100 J) was obtained using DMI per body weight (DMIBW, kg/100 kg), content (g/100 g DM) of ether extract (EE) and crude protein (CP), and DM digestibility (DMD, g/100 g); CH4‐E/GEI = ?0.782 (±0.2526) DMIBW ? 0.436 (±0.0548) EE ? 0.073 (±0.0218) CP + 0.049 (±0.0097) DMD + 8.654 (±0.6517) (R2 = 0.39; RMSE = 1.3479 J/100 J GEI). It was indicated that CH4 emissions from beef cattle in Southeast Asia are predictable using present developed models including simple indices.  相似文献

Three experiments were conducted to determine the Val and Ile requirements in low-CP, corn-soybean meal (C-SBM) AA-supplemented diets for 20- to 45-kg pigs. All experiments were conducted for 26 to 27 d with purebred or crossbred barrows and gilts, which were blocked by initial BW. Treatments were replicated with 5 or 6 pens of 3 or 4 pigs per pen. At the beginning of Exp. 1 and the end of all experiments, blood samples were obtained from all pigs to determine plasma urea N (PUN) concentrations. All diets were C-SBM with 0.335% supplemental Lys to achieve 0.83% standardized ileal digestible (SID) Lys, which is the Lys requirement of these pigs. In Exp. 1, 0, 0.02, 0.04, 0.06, 0.08, or 0.10% L-Val was supplemented to achieve 0.51, 0.53, 0.55, 0.57, 0.59, or 0.61% dietary SID Val, and Thr, Trp, Met, and Ile were supplemented to maintain Thr:Lys, Trp:Lys, TSAA:Lys, and Ile:Lys ratios of 0.71, 0.20, 0.62, and 0.60, respectively. Also, supplemental Gly and Glu were added to all diets to achieve 1.66% Gly + Ser and 3.28% Glu, which is equal to the Gly + Ser and Glu content of a previously validated positive control diet that contained no supplemental AA. Treatment differences were considered significant at P < 0.10. Valine addition increased ADG, ADFI, and G:F in pigs fed 0.51 to 0.59% SID Val (linear, P < 0.08), but ADG and ADFI were decreased at 0.61% SID Val (quadratic, P ≤ 0.10). On the basis of ADG and G:F, the SID Val requirement is between 0.56 and 0.58% in a C-SBM diet supplemented with AA. In Exp. 2 and 3, 0, 0.02, 0.04, 0.06, or 0.08% L-Ile was supplemented to achieve 0.43, 0.45, 0.47, 0.49, or 0.51% dietary SID Ile, and Thr, Trp, Met, and Ile were supplemented to maintain Thr:Lys, Trp:Lys, TSAA:Lys, and Val:Lys ratios of 0.71, 0.20, 0.62, and 0.74, respectively. Also, supplemental Gly and Glu were added to achieve 1.66% Gly + Ser and 3.28% Glu as in Exp. 1. Data from Exp. 2 and 3 were combined and analyzed as 1 data set. Daily BW gain, ADFI, and G:F were not affected by Ile additions to the diet; however, ADFI was decreased among pigs fed the diet with 0.45% SID Ile (P < 0.10) compared with pigs fed the 0.43% SID Ile diet. Broken-line analysis requirements could not be estimated for the combined data from Exp. 2 or 3. The results of this research indicate that the SID Val requirement is between 0.56 to 0.58% (0.67 to 0.70 SID Val:Lys), and the Ile requirement is adequate at 0.43% SID Ile (0.52 SID Ile:Lys) for 20- to 45-kg pigs.  相似文献

To develop an ultrasonographic assay for determining plasma progesterone concentration (P4) as < 1 ng/ml or ≥ 1 ng/ml, the corpus luteum (CL) area and P4 were measured in 1094 multiparous Holstein cows. The area‐measuring function and frozen images were used to outline and measure CL imaged via ultrasonography, and CL area was estimated as a polygon of a continuation straight line. A significant correlation was found between CL area and P4 (p < 0.001), and this analysis resulted in the following correlation equation: y = ?0.35 + 1.02x (r = 0.81). According to the correlation equation, a CL area of 1.3 cm2 indicated a P4 of 1 ng/ml. Based on this relationship, each animal was categorized into one of six groups, groups differed based on CL area, and the area ranges were as follows: < 1.3 cm2 (Group A), 1.3–2.2 cm2 (Group B), 2.3–3.2 cm2 (Group C), 3.3–4.2 cm2 (Group D), 4.3–5.2 cm2 (Group E) and > 5.2 cm2 (Group F). For each group, the proportion of cows whose P4 was 1 ng/ml or more was 1.5% in Group A, 83.3% in Group B, 76.6% in Group C, 96.6% in Group D, 99.2% in Group E and 100% in Group F. There was a significant difference between Group A and the other five groups, and between Groups B or C and Groups D, E or F (p < 0.005). These results indicate that a functional CL does not exist when the CL area is less than 1.3 cm2 and that it exists when the CL area is 3.3 cm2 or more.  相似文献

Three experiments were conducted to evaluate the order of limitation for Lys, Thr, and Val in corn-soybean meal diets of lactating sows experiencing high BW loss during lactation. Plasma urea N (PUN) was used as the response criterion. Experimental diets used in all 3 experiments were derived from the basal diet (BSL) formulated to 0.51% Lys using corn and soybean meal as the only sources of AA. The BSL diet was formulated by diluting a reference corn-soybean meal diet (17.2% CP, 0.90% Lys, 0.65% Thr, 0.82% Val, and 0.28% Met) with cornstarch, sucrose, and soybean oil to maintain the same ratio of corn to soybean meal. Experiment 1 was conducted to determine whether PUN is sensitive to dietary Thr and Val levels and whether a 4-d feeding period was adequate to observe a PUN response. The results demonstrated that PUN could be used as the response criterion and that a 4-d feeding period was adequate. Experiment 2 was conducted to determine whether Thr or Val was most limiting in the BSL diet. Dietary treatments used in Exp. 2 consisted of 1) BSL supplemented with 0.09% L-Lys and 0.02% DL-Met, which served as the negative control (NC2); 2) NC2 plus 0.14% L-Thr (NC2 + T); 3) NC2 plus 0.17% L-Val (NC2 + V); and 4) NC2 plus 0.14% L-Thr and 0.17% L-Val (NC2 + T + V). The value for PUN was lower for the NC2 + T and NC + T + V treatments compared with the NC2 and NC2 + V treatments (5.18 and 5.33 vs. 6.43 and 6.62; P < 0.01), indicating that Thr was most limiting in the NC2 diet. Experiment 3 was conducted to determine whether Thr or Lys was most limiting in the BSL diet. Dietary treatments used in Exp. 3 consisted of 1) the BSL diet supplemented with 0.02% DL-Met only, which served as the negative control diet (NC3); 2) the NC3 diet supplemented with 0.20% L-Lys (NC3 + L); 3) the NC3 diet supplemented with 0.14% L-Thr (NC3 + T); and 4) the NC3 diet supplemented with 0.20% L-Lys and 0.14% L-Thr (NC3 + L + T). The value for PUN was lower for NC3 + L compared with NC3 and NC3 + T (7.45 vs. 9.11 and 8.45 mg/dL; P < 0.01), and PUN for NC3 + L + T was lower than the PUN measured for NC3 + L (5.94 vs. 7.45; P < 0.01). The results of Exp. 2 and 3 indicate that Lys was first-limiting and Thr second-limiting in the BSL diet.  相似文献

Three experiments were conducted to determine the optimum standardized ileal digestible Val-to-Lys (SID Val:Lys) ratio for 13- to 32-kg pigs. In Exp. 1, 162 pigs weaned at 17 d of age (8 pens/treatment) were used, and a Val-deficient basal diet containing 0.60% l-Lys·HCl, 1.21% SID Lys, and 0.68% SID Val was developed (0.56 SID Val:Lys). Performance of pigs fed the basal diet was inferior to a corn-soybean meal control containing only 0.06% l-Lys·HCl, but was fully restored with the addition of 0.146% l-Val to the basal diet (68% SID Val:Lys). In Exp. 2, 54 individually housed barrows (21.4 kg) were utilized in a 14-d growth assay. Pigs were offered a similar basal diet (1.10% SID Lys), ensuring Lys was marginally limiting with no supplemental l-Val (55% SID Val:Lys). The basal diet was fortified with 4 graded levels of l-Val (0.055% increments) up to a ratio of 75% SID Val:Lys. In Exp. 3, 147 barrows (13.5 kg) were fed identical diets, only with 1 additional level at a SID Val:Lys of 80% and fed for 21 d. In Exp. 2 and 3, a high protein, control diet was formulated to contain 1.10% SID Lys and 0.20% l-Lys·HCl. In Exp. 2, linear effects on ADG (713, 750, 800, 796, and 785 g/d; P = 0.05) and G:F (P = 0.07) were observed with increasing SID Val:Lys, characterized by improvements to a ratio of 65% and a plateau thereafter. In Exp. 3, quadratic improvements in ADG (600, 629, 652, 641, 630, and 642 g/d; P = 0.08) and G:F (P = 0.07) were observed with increasing SID Val:Lys, as performance increased to a ratio of 65% but no further improvement to a ratio of 80%. Pigs fed the control diet did not differ from those fed a ratio of 65% SID Val:Lys in Exp. 2, but did have improved G:F in Exp. 3 (P = 0.03). To provide a more accurate estimate of the optimum SID Val:Lys, data from Exp. 2 and 3 were combined. With single-slope broken-line methodology, the minimum ratio estimate was 64 and 65% SID Val:Lys for ADG and G:F, respectively. With combined requirement estimates, the data indicate that a SID Val:Lys of 65% seems adequate in maintaining performance for pigs from 13 to 32 kg.  相似文献

Zinc is effective in the prevention and treatment of post‐weaning diarrhoea and in promoting piglet growth. Its effects on the absorption of nutrients and the secretory capacity of the intestinal epithelium are controversial. We investigated the effects of age, dietary pharmacological zinc supplementation and acute zinc exposure in vitro on small‐intestinal transport properties of weaned piglets. We further examined whether the effect of zinc on secretory responses depended on the pathway by which chloride secretion is activated. A total of 96 piglets were weaned at 26 days of age and allocated to diets containing three different levels of zinc oxide (50, 150 and 2500 ppm). At the age of 32, 39, 46 and 53 days, piglets were killed, and isolated epithelia from the mid‐jejunum were used for intestinal transport studies in conventional Ussing chambers, with 23 μm ZnSO4 being added to the serosal side for testing acute effects. Absorptive transport was stimulated by mucosal addition of d ‐glucose or l ‐glutamine. Secretion was activated by serosal addition of prostaglandin E2, carbachol or by mucosal application of Escherichia coli heat‐stable enterotoxin (Stp). Jejunal transport properties showed significant age‐dependent alterations (p < 0.03). Both absorptive and secretory responses were highest in the youngest piglets (32 d). The dietary zinc supplementation had no significant influence on jejunal absorptive and secretory responses. However, the pre‐treatment of epithelia with ZnSO4 in vitro led to a small but significant decrease in both absorptive and secretory capacities (p < 0.05), with an exception for carbachol (p = 0.07). The results showed that, in piglets, chronic supplementation with zinc did not sustainably influence the jejunal transport properties in the post‐weaning phase. Because transport properties are influenced by the addition of zinc in vitro, we suggest that possible epithelial effects of zinc depend on the acute presence of this ion.  相似文献

Lysine (Lys) is the first limiting amino acid (AA) in most feed formulations for pigs and most abundant, along with leucine, in muscle proteins. An experiment was conducted with 17 pigs (17.7 ± 0.05 kg initial BW) to identify a role of dietary Lys in the control of protein synthesis in pigs. Fourteen pigs were randomly assigned to one of the two wheat‐based dietary treatments: Lys‐deficient, 3.0 g/kg (DEF) and Lys‐adequate, 10.8 g/kg (ADE). Samples from jejunum mucosa, liver, Longissumus and Semitendinosus muscles, and blood were collected. The other three pigs were sacrificed at the beginning of the trial to measure basal carcass composition. Weight gain, gain:feed ratio, Lys intake and loin eye area were greater in ADE than in DEF pigs (p < 0.01). Muscle‐related carcass characteristics were better, and myosin heavy chain IIb expression (MyHC IIb) in Semitendinosus was higher in ADE than in DEF pigs. Expression of AA transporters CAT‐1 was lower (p < 0.05), serum Lys was higher and serum Val was lower in pigs fed the ADE diet. The higher muscularity, MyHC IIb expression in Semitendinosus muscle and Lys serum of pigs fed the ADE diet suggest that Lys increases growth rate not only by functioning as protein construction unit but also as potential control of the protein synthesis process.  相似文献

A key concern with the flooding dose technique for measuring protein synthesis is that a large dose of amino acid (AA) can potentially change the animals’ hormonal and nutritional status, which in turn can influence protein synthesis. Among stable isotope tracers, 1‐[13C]‐valine is the preferred AA for measuring protein synthesis in gut tissue and mucins. A study was conducted to determine the impact of a flooding dose of valine on the metabolic status of pigs. Six barrows [16.5 kg body weight (BW)] were randomly assigned to intravenous infusions of either 150 mm valine (1.5 mmol/kg BW) or physiological saline, following a crossover design. Blood samples were taken 10 min prior to infusion, at the end of infusion, at 10‐min intervals for 60 min post‐infusion, and at 90 and 120 min post‐infusion. Plasma concentrations of insulin, glucose, AA, urea nitrogen and packed cell volume (PCV) were measured. Infusion of valine increased plasma valine concentrations (4129 vs. 582 μm ;< 0.05) but had no influence on PCV (26.4% vs. 27.2%) and plasma concentrations of glucose (6.0 vs. 5.8 mm ) and insulin (8.2 vs. 8.5 μU/ml; > 0.10). Plasma urea nitrogen concentration was reduced with valine infusion (8.5 vs. 7.8 mg/dl; < 0.05). A flooding dose of valine had no impact on plasma concentrations of AA, and specifically branched‐chain AA such as leucine (240 vs. 231 μm ) and isoleucine (310 vs. 331 μm ;> 0.10). There was, however, a slight increase in the plasma concentrations of threonine (224 vs. 263 μm ;< 0.05) and a tendency towards reduced glycine (1387 vs. 1313 μm ;< 0.10). The results indicate that a flooding dose of valine does not cause a substantial change in the metabolic status of growing pigs and is therefore suitable for measuring protein synthesis rates.  相似文献

We investigated effects of Rendement Napole (RN ) genotype on metabolic markers in Ossabaw pigs fed diets with different levels of dietary fat. Thirty‐two pigs, belonging to either the wild‐type (WT , rn+/rn+) or carrier (CAR , RN ?/rn+) genotypes (n  = 16/genotype), were divided into two dietary groups, (high fat [HF ] or low fat [LF ]) diets, for 12 weeks (n  = 8 pigs/genotype/diet) after which pigs were killed for gene expression analysis by RT ‐PCR . Feeding HF diet caused increased daily gain (ADG ,<  .05) and final body weight (BW ) (<  .05) in comparison with the LF diet (<  .05). Feed efficiency (gain:feed) was higher (<  .05) in pigs on the HF and was higher (<  .05) in CAR pigs compared to WT . There was genotype × diet interaction (=  .05) on final BW such that CAR animals on LF diet had the same final BW as animals of both genotypes on HF diet. Carrier pigs on LF diet had higher (<  .05) average daily gain and gain:feed than WT pigs. There was a trend (<  .08) for a higher feed consumption in pigs on the LF diet. Backfat thickness was higher (<  .01) in pigs on the HF diet. Serum triglyceride was higher (0.62 vs. 0.33 mg/dl, <  .01) in pigs on HF diet. Serum insulin was higher (<  .05) in CAR versus WT pigs (0.40 vs. 0.015 μg/ml). Pigs on the HF diet had a higher (<  .05) serum insulin compared to those on the LF diet (0.032 vs. 0.023 μg/ml). Carnitine palmitoyl transferase 1‐alpha was higher (<  .05) in the longissimus dorsi and semitendinosus muscles of pigs on HF diet. Acyl‐CoA oxidase I was elevated (<  .05) in the liver of pigs on HF diet. Fatty acid synthase was lower in the longissimus dorsi muscle, liver and mesenteric fat (<  .05) of carrier pigs. Abstract

Supplementing Bacillus (B.) subtilis mutants selected to overproduce a specific amino acid (AA) may be an alternative method to provide essential AA in pig diets. Two experiments on a B. subtilis strain selected to overproduce Trp were conducted using 8-kg pigs fed Trp-deficient diets for 20 d. B. subtilis were supplied in a low or high dose in Experiments 1 and 2, respectively. The Trp-deficient diet (0.15 SID Trp:Lys) reduced (p?B. subtilis strain was not able to counterbalance the Trp deficiency in any of the two experiments. No effect of B. subtilis supplementation to piglet diets was observed on the plasma AA profile. In conclusion, this mutant strain of B. subtilis was not able to compensate a Trp deficiency in the tested doses.  相似文献

1. The aim of the study was to determine the ideal ratio of the essential amino acids lysine (Lys), methionine (Met), threonine (Thr), tryptophan (Trp), arginine (Arg), valine (Val) and isoleucine (Ile) and to assess the required dietary lysine concentration for optimum performance in broiler chickens of 2 commercial strains from 20 to 40 d of age. 2. An identical basal diet was used throughout all experiments. It consisted mainly of maize and soyabean meal and contained 172 g crude protein and 13.2 MJ AME(N) per kg. For each experiment, the basal diet was adequately supplemented with all essential amino acids except the one to be tested, which was supplemented in 6 graded concentrations in exchange for maize starch. One (Met, Trp, Arg, Val, Ile) or 2 (Lys, Thr) growth trials were conducted per amino acid tested and the response in weight gain, food: gain ratio, breast meat yield and abdominal fat were examined. 3. The ideal amino acid ratio relative to Lys (expressed as a percentage) was calculated to be 75% Met+Cys, 63% Thr, 19% Trp, 112% Arg, 71% Ile and 81% Val on a true faecal digestible basis when the data were subjected to broken-line regression analysis. From both lysine studies the concentration for optimum food: gain ratio was calculated, by exponential regression analysis, to be 11.5 g true faecal digestible lys per kg diet.  相似文献

Valine is likely the fourth limiting amino acid in most diets based on corn and soybean meal (C/SBM). However, the exact needs for Val are not well known, and information regarding it is sparse. A series of studies was conducted to validate the limitation of Val in all-vegetable diets fed to broilers, and subsequently to quantify an adequate ratio to Lys in high-yield late-developing broilers (Ross × Ross 708) from 21 to 42 d. A preliminary study was designed to evaluate the supplementation of different amino acids likely to be fourth limiting on a C/SBM-based diet, where Lys, TSAA, and Thr were supplemented but no other critical amino acids were given minimums in the formulation. Results obtained for BW gain, abdominal fat weight, and abdominal fat percentage showed that birds were more responsive (P < 0.05) to l-Val supplementation. A follow-up study using a corn-peanut meal (C/PM)-based diet formulated to be deficient in Val validated (P < 0.05) a Val deficiency based on poor growth performance and resulted in an immediate return to good performance when this amino acid was supplemented. Furthermore, the C/PM diet was compared with a nutritionally similar C/SBM diet, and no difference was observed in the performance of broilers fed these diets, thus validating the ability of the C/PM-based diet to support adequate growth of these broilers. This C/PM-based diet was used to feed gradual concentrations of Val, from levels that would be considered deficient (0.59% digestible) up to adequate (0.84% digestible Val). Results indicate that a ratio of Val to Lys of 78, or a minimum dietary value of 0.74% digestible Val (0.82% total Val), should be adequate for this high-yield broiler grown from 21 to 42 d of age.  相似文献

Myostatin is a negative regulator of muscle growth and development in mammals, and variation in ovine myostatin gene (MSTN) has been demonstrated to be associated with variation in the muscularity of sheep. Polymerase chain reaction–single‐stranded conformational polymorphism (PCR–SSCP) was used to look for single nucleotide polymorphisms (SNPs) in a 304‐bp amplicon from the promoter region of ovine MSTN. Sequence analyses revealed two previously identified SNPs (c.?2449G/C and c. ?2379T/C) that resulted in three haplotypes (H1 (c.[?2449G; ?2379C]), H2 (c.[?2449C; ?2379C]) and H3 (c.[?2449G; ?2379T]). The effect of these SNPs on growth and carcass traits was investigated in 357 NZ Romney lambs. General linear mixed‐effect models revealed that sheep with the genotype c.?2449GC had a higher loin meat yield (p = 0.032) and proportion loin yield (p = 0.028), than those with the genotype c.?2449GG. The genotype c.?2379CC was associated with an increase in three weight traits: birthweight (p = 0.003), tailing weight (p = 0.009) and weaning weight (p = 0.028), when compared with the genotype c.?2379TC, but it was not found to have an association with growth rate. This suggests that c.?2379T/C has an effect that originates at, or before birth. Haplotype H3 was associated with a decrease in birthweight (p = 0.002), tailing weight (p = 0.003) and weaning weight (p = 0.011). Haplotype H2 was associated with increased loin yield (p = 0.012) and proportion loin yield (p = 0.002). The SNPs may have value as genetic markers for improved Romney breeding.  相似文献
