The objective of this study was to determine the effects of castration on short-term growth performance, hormone profiles, and behavior in pigs at 3, 6, 9, or 12 d of age. Ninety intact male pigs were assigned randomly to a treatment age by litter [3, 6, 9, or 12 d of age; n = 9 to 13 pigs per treatment (age) group]. Pigs within a single litter were then assigned to noncastrated (NC) or castrated (CAS) treatment groups according to BW. Pigs were nonsurgically fitted with jugular catheters, and blood samples were drawn immediately before castration (0 h) and at 0.5, 1, 1.5, 2, 24, and 48 h after castration. Body weights were obtained when pigs were catheterized and again at 24 and 48 h after castration. Serum samples were analyzed for cortisol, porcine corticosteroid-binding globulin, and dehydroepiandrosterone sulfate (DHEA-S). No differences were detected in initial BW of pigs, and there was no overall treatment effect on growth performance of pigs at 24 or 48 h posttreatment. A time x treatment interaction was detected (P < 0.01) for serum cortisol concentrations, such that cortisol was greater in CAS pigs than in NC pigs. No overall effect of age at castration was observed on cortisol concentrations. At 24 h after castration, serum cortisol concentrations returned to baseline in all treatment groups; however, at 48 h after castration, overall cortisol concentrations were elevated (P < 0.01) in the 6-, 9-, and 12-d-old pigs in both the CAS and NC groups compared with baseline concentrations. Total cortisol and porcine corticosteroid-binding globulin were used to calculate the free cortisol index (FCI). A time x treatment interaction was observed (P < 0.01) for FCI, such that FCI was greater in CAS males than in NC males. The FCI was also affected by age (P < 0.01). There was a time x treatment x age interaction (P < 0.01) for serum DHEA-S, such that DHEA-S concentrations decreased in CAS animals but increased in NC animals, and DHEA-S concentrations increased with age. During the first 2 h after castration, there was an overall age effect (P = 0.01) on the time that pigs spent standing, such that 3-d-old pigs stood more than 6-, 9-, or 12-d-old pigs. Treatment did not influence the time that pigs spent nursing, lying, standing, or sitting, although there was a trend (P = 0.08) for CAS pigs to be less active than NC pigs. These data indicate that castration is stressful regardless of age; however, the stress associated with handling seems to increase as pigs age.  相似文献

The cortisol response of calves to different methods of castration (ring, band, surgical, clamp) with or without local anaesthetic, or local anaesthetic plus a non-steroidal anti-inflammatory drug were recorded. All methods of castration caused a significant cortisol response and by inference pain and distress. Band castration caused a greater cortisol response than ring castration but the responses were eliminated by local anaesthetic. The cortisol response to surgical castration, by traction on the spermatic cords or by cutting across them with an emasculator, was not diminished by local anaesthetic but when ketoprofen was given with local anaesthetic the cortisol response was eliminated. Local anaesthetic did reduce the behavioural response to cutting the scrotum and handling the testes. Clamp castration caused the smallest cortisol response which was reduced or eliminated by local anaesthetic or local anesthetic plus ketoprofen respectively, but this method of castration was not always successful.  相似文献

The S(+) enantiomer of ketoprofen is predominant in the plasma of pigs after administration of racemic ketoprofen, although the occurrence and extent of R(-)-to-S(+) inversion is uncertain. Plasma concentrations of both enantiomers were measured and percentages of S(+) ketoprofen were calculated at different time points after intravenous and oral dosing of pigs with 1.5mg/kg R(-) ketoprofen. S(+) ketoprofen was formed immediately after administration and concentrations exceeded R(-) concentrations after 1h. Absence of pre-systemic inversion was deduced from the lower S(+) percentages after oral administration. A rapid and increasing inversion, reaching a maximum of about 70%, occurred and appeared to be responsible for the predominance of S(+) ketoprofen in pig plasma after administration of the racemate.  相似文献

In order to determine the effective dose, the effects of orally administered ketoprofen were evaluated in pigs following intravenous challenge with Escherichia coli endotoxin. One hour after the challenge, five groups of pigs were treated with either tap water or ketoprofen (0.5 mg/kg, 1 mg/kg, 2 mg/kg or 4 mg/kg). The body temperature was measured and a total clinical score was calculated after assessing the general behaviour, respiratory rate and locomotion of the pigs. Thromboxane B(2) and ketoprofen concentrations were analysed from blood samples. Ketoprofen treatment significantly reduced the rectal temperature and total clinical scores, and lowered blood thromboxane B(2) concentrations when compared with the control group. Ketoprofen plasma concentrations were lower than previously reported in healthy pigs after similar doses. The appropriate dose of orally administered ketoprofen in pigs in this model is 2 mg/kg, as the higher dose of 4 mg/kg failed to provide an additional benefit.  相似文献

Surgical castration is a common management practice performed on male pigs to prevent the occurrence of boar taint. Surgical castration is known to cause physiological and behavioral changes in pigs indicative of pain-induced distress; however, it is commonly performed without pain relief. The objective of this study was to evaluate the effectiveness of carbon dioxide gas (CO(2)) anesthesia and a non-steroidal anti-inflammatory drug (NSAID) to alleviate the pain caused by castration. At 3 d of age, male pigs were either control handled (CON), castrated without pain relief (CAS), given an NSAID and then immediately castrated (CAS+NSAID), anesthetized with CO(2) and then castrated (CAS+CO2), or anesthetized with CO(2) and given an NSAID at the time of castration (CAS+BOTH). Blood samples were collected before castration, and at 30, 60, 120, and 180 min, 24 h, and 3 d after castration or handling for analysis of cortisol, C-Reactive protein (CRP), and substance-P (SP) concentrations. This study was then repeated using the same treatment groups, and the behavioral response to castration and handling were measured using a 1-min scan sampling procedure. The percentage of stress vocalizations was recorded during the administration of all treatments. Anesthesia and analgesia did not effectively reduce (P > 0.05) the cortisol response to surgical castration. Overall, CRP concentrations were greater (P < 0.05) in CAS+CO2 pigs as compared with CON pigs. Sixty minutes after castration or handling, SP concentrations were greater (P < 0.08) in pigs given CO(2) anesthesia (CO2, CAS+CO2, and CAS+BOTH) than CON, CAS, and CAS+NSAID pigs. Pigs castrated without pain relief spent more (P < 0.001) time lying without contact than all other treatments during the first 30 min after castration, but thereafter CAS+CO2 pigs spent more (P < 0.001) time lying without contact than other treatments. During the first 30 min after the treatments were applied, CAS+CO2 pigs spent more (P < 0.01) time displaying pain-like behaviors than CON, CAS, CAS+NSAID, and CAS+BOTH pigs. The percentage of stress vocalizations was greater (P < 0.05) in CAS and CAS+NSAID pigs than all other treatments. Neither CO(2) anesthesia nor a NSAID, given separately or combined, markedly reduced the pain-induced distress caused by castration in pigs. More research is needed to evaluate practical methods of on-farm pain relief for pigs.  相似文献

To determine the effects of the anti-inflammatory ketoprofen, alone or with local anesthesia (LA) during castration on cortisol, immune, and acute phase responses, 40 Friesian calves (215 +/- 3.5 kg) were assigned as follows: 1) control, 2) surgical castration (SURG), 3) SURG following ketoprofen (3 mg/kg BW i.v.; SURG + K), 4) SURG following LA (9 mL of 2% lidocaine hydrochloride to each testis; SURG + LA), or 5) SURG following LA and K (SURG + LA + K). Total cortisol response was greater (P < 0.05) in SURG, SURG + LA, and SURG + K + LA calves than in control calves and was not different between control and SURG + K calves. The interval to peak cortisol was longer (P < 0.05) for SURG + K + LA calves than for either SURG or SURG + K calves. On d 3, KLH-induced interferon-gamma production was lower (P < 0.05) in SURG calves than in control calves, whereas concanavalin A-induced interferon-gamma production was lower (P < 0.05) in all castration groups than in control. On d 1 after surgery, fibrinogen was higher (P < 0.05) in SURG and SURG + LA calves than in control calves, whereas SURG + LA + K calves had lower (P < 0.05) fibrinogen than did SURG calves. Haptoglobin was higher (P < 0.05) in SURG calves on d 1, 3, and 7 than in control calves. On d 1 after surgery, SURG + K and SURG + LA + K calves had lower (P < 0.05) haptoglobin concentrations than SURG calves, whereas SURG + K calves had lower (P < 0.05) levels than SURG calves on d 3. In conclusion, surgical castration induced a significant elevation in cortisol secretion; the rise in cortisol was reduced to control levels by the administration of ketoprofen but not local anaesthetic. Thus, systemic analgesia using ketoprofen is more effective than local anesthesia during castration to alleviate the associated stress response.  相似文献

OBJECTIVE: To assess bioequivalence after oral, IM, and IV administration of racemic ketoprofen in pigs and to investigate the bioavailability after oral and IM administration. ANIMALS: 8 crossbred pigs. PROCEDURES: Each pig received 4 treatments in a randomized crossover design, with a 6-day washout period. Ketoprofen was administered at 3 and 6 mg/kg, PO; 3 mg/kg, IM; and 3 mg/kg, IV. Plasma ketoprofen concentrations were measured by use of high-performance liquid chromatography for up to 48 hours. To assess bioequivalence, a 90% confidence interval was calculated for the area under the time-concentration curve (AUC) and maximum plasma concentration (C(max)). RESULTS: Equivalence was not detected in the AUCs among the various routes of administration nor in C(max) between oral and IM administration of 3 mg/kg. The bioavailability of ketoprofen was almost complete after each oral or IM administration. Mean +/- SD C(max) was 5.09 +/- 1.41 microg/mL and 7.62 +/- 1.22 microg/mL after oral and IM doses of 3 mg/kg, respectively. Mean elimination half-life varied from 3.52 +/- 0.90 hours after oral administration of 3 mg/kg to 2.66 +/- 0.50 hours after IV administration. Time to peak C(max) after administration of all treatments was approximately 1 hour. Increases in AUC and C(max) were proportional when the orally administered dose was increased from 3 to 6 mg/kg. Conclusions and Clinical Relevance: Orally administered ketoprofen was absorbed well in pigs, although bioequivalence with IM administration of ketoprofen was not detected. Orally administered ketoprofen may have potential for use in treating pigs.  相似文献

The plasma cortisol response to castration using Burdizzo clamps and, by inference, the acute pain experienced, is less than that caused by surgical, rubber-ring or latex-band castration. The cortisol response may be influenced by the age of the animal castrated, but this has not been well defined. Local anaesthesia virtually eliminates the cortisol response, and thus the acute pain, caused by rubber-ring or latex-band castration, but needs to be combined with a systemic analgesic such as the non-steroidal anti-inflammatory drug ketoprofen to eliminate the cortisol response to Burdizzo or surgical castration. When used alone, ketoprofen sometimes reduces the cortisol response to Burdizzo or surgical castration but may need to be accompanied by local anaesthesia to eliminate the pain-induced behaviour seen during the castration process itself. Thus, pharmacological methods are available to virtually eliminate the acute pain experienced by calves during the 12 h following castration. The use of these methods is an additional cost for farmers and may be limited by the availability of drugs for farmers to use and the scarcity of veterinarians in farm animal practice.  相似文献

To determine the effects of burdizzo castration alone or in combination with ketoprofen (K), local anesthesia (LA), or caudal epidural anesthesia (EPI) on plasma cortisol, acute-phase proteins, interferon-gamma production, growth, and behavior of beef cattle, 50 Holstein x Friesian bulls (13 mo old, 307 +/- 5.3 kg) were assigned to (n = 10/treatment): 1) control (handled; C); 2) burdizzo castration (B); 3) B following K (3 mg/ kg of BW i.v.; BK); 4) B following LA (8 mL into each testis and 3 mL s.c. along the line where the jaws of the burdizzo were applied with 2% lidocaine HCl; BLA); and 5) B following EPI (0.05 mg/kg of BW of xylazine HCl and 0.4 mg/kg of BW of lidocaine HCl as caudal epidural; BEPI). The area under the cortisol curve against time was lower (P < 0.05) in BK than in B, BLA, or BEPI animals. On d 1 after treatment, plasma haptoglobin concentrations were higher (P < 0.05) in B, BLA, and BEPI than in BK animals. On d 3, haptoglobin and plasma fibrinogen concentrations were higher (P < 0.05) in all castration groups than in C. On d 7, haptoglobin and fibrinogen concentrations remained higher (P < 0.05) in BLA than in B and C animals. On d 1, concanavalin A-induced interferon-gamma production was lower (P < 0.05) in B, BLA, and BEPI than in C, but there was no difference between BK and C animals. From d -1 to 35, ADG was lower (P < 0.05) in B, BLA, and BEPI animals, but not in BK compared with C animals. Overall, there was a higher (P < 0.05) incidence of combined abnormal postures in B than in C, BK and BEPI animals. Although the use of K and EPI decreased (P < 0.05) these postures compared with B alone or B with LA, there was no difference between the K and EPI treatment. In conclusion, burdizzo castration increased plasma cortisol and acute-phase proteins, and suppressed immune function and growth rates. Local anesthesia prolonged the increase in acute-phase proteins. Ketoprofen was more effective than LA or EPI in decreasing cortisol and partially reversed the reduction in ADG following castration. The use of K or EPI was more effective than LA in decreasing pain-associated behavioral responses observed during the first 6 h after treatment. Systemic analgesia with ketoprofen, a non-steroidal antiinflammatory drug, was more effective in reducing inflammatory responses associated with castration than LA or EPI.  相似文献

The efficacy of ketoprofen in the treatment of non-infectious lameness in sows was examined in a double-blinded study. Two dose rates of oral ketoprofen were compared to placebo treatment over five consecutive days. Lameness was assessed with a five-grade scoring system prior to and on the last day of the treatment. The rate of treatment success was 54.3% for the ketoprofen 4mg/kg group (n=46), 53.2% for the ketoprofen 2mg/kg group (n=47) and 20.8% for the pigs in the placebo group (n=48). The difference between both ketoprofen groups and the placebo group was significant (P=0.001), but there was no difference between the two ketoprofen groups (P=0.78). Oral ketoprofen was well tolerated and no adverse events were observed. As lameness is a very common problem in sows, oral ketoprofen appeared to be a practical way to alleviate pain and improve the welfare of sows.  相似文献

Crossbred pigs (n = 240) from Pietrain x Large White sires mated to Landrace x Large White dams, with an average age of 100 d (60.5 +/- 2.3 kg of BW), were used to investigate the effects of sex and slaughter weight (SW) on growth performance and on carcass and meat quality characteristics. There were 6 treatments arranged factorially, with 3 classes (intact females, IF; castrated females, CF; and castrated males, CM) and 2 slaughter weights (114 and 122 kg of BW). Each of the 6 combinations of treatments was replicated 4 times, and the experimental unit was a pen with 10 pigs. Castrated males and CF ate more feed, grew faster, and had more carcass backfat depth and fat thickness at the gluteus medius muscle but lower loin yield than IF (P < 0.05). In addition, CF and CM had more intramuscular fat (P < 0.05) and less linoleic acid content in the subcutaneous fat (P < 0.01) than IF. Pigs slaughtered at 122 kg of BW had lower ADG (P < 0.05), decreased G:F (P < 0.05), and more gluteus medius fat than pigs slaughtered at 114 kg of BW (P < 0.05). It was concluded that CF and CM had similar productive performance and meat quality characteristics when slaughtered at the same age, and that castration of females improved ADG and increased weight and fat content of primal cuts with respect to IF. Therefore, castration of females is recommended in pigs destined for the dry-cured industry because of the beneficial effects on quality of the primal cuts.  相似文献

Changes in the activity of the sympathetic nervous system or in the hypothalamo-pituitary-adrenal axis have been extensively used to evaluate pain induced by castration or tail docking in numerous species. Such data are missing in pigs. Therefore, three experiments were conducted to determine the effects of castration, tail docking, or tooth resection on stress hormones. Glucose and lactate also were measured because catecholamines stimulate mobilization of glycogen, which results in glucose and lactate release. In Exp.1, 18 male pigs from seven litters (two or three pigs per litter) were catheterized surgically into one jugular vein, under general anesthesia, at 5 or 6 d of age. Two days later, they were submitted either to bilateral castration, control handling, or no handling (n = 6 per group). Blood samples were collected before (- 15 and -2 min) and after (5, 15, 30, 60, 90, 180 min) the experimental treatment. In Exp. 2, 27 female pigs from 12 litters (one to four pigs per litter) were submitted either to tooth clipping with pliers, tooth resection with a grinder apparatus, control handling, or no handling (n = 6 or 7 per group) at 1 d of age. In Exp. 3, 17 female pigs from nine litters (one to three pigs per litter) were submitted to one of the following treatments: 1) tail docking with an electric-heated scissor docking iron, 2) control handling, and 3) no handling (n = 5 or 6 per group) at 1 d of age. Castration induced significant (P < 0.05) increases in adrenocorticotropin hormone (ACTH; from 5 to 60 min), cortisol (from 15 to 90 min), and lactate (from 5 to 30 min). These variations are indicative of stress and tissue damage following castration. In contrast, neither tail docking nor tooth resection had marked effects on plasma cortisol, ACTH, glucose, and lactate. Measurements of plasma cortisol, lactate, and ACTH could be useful for validating treatments designed to relieve the distress reaction induced by castration in pigs.  相似文献

To determine the effect of repeated ketoprofen (K) administration to surgically castrated bulls on cortisol, acute-phase proteins, immune function, feed intake, growth and behavior, 50 Holstein x Friesian bulls (11 mo old; 300 +/- 3.3 kg) were assigned to one of five treatments: 1) untreated control (C); 2) surgical castration at 0 min (S); 3) S following an i.v. injection of 3 mg/kg of BW of K at -20 min (SK1); 4) S following 1.5 mg/kg of BW of K at -20 and 0 min (SK2); or 5) S following 1.5 mg/kg of BW of K at -20 and 0 min and 3 mg/kg of BW of K at 24 h (SK3). Castration acutely increased plasma cortisol concentrations in S- and K-treated animals compared with C, with no differences in peak and interval to peak cortisol responses among the castration groups. Overall, the integrated cortisol response was greater (P < 0.05) in the castrates than in C, whereas K treatments decreased (P < 0.05) this response compared with S alone, with no differences between K treatments. Plasma haptoglobin and fibrinogen concentrations were increased (P < 0.05) on d 3 in the castration groups compared with C as the result of tissue trauma induced by castration, whereas SK1 and SK2 had lower (P < 0.05) haptoglobin concentrations than S animals. On d 1, concanavalin A-induced interferon-gamma production was suppressed (P < 0.05) in S and SK3 compared with C, SK1, and SK2 animals. Overall from d 1 to 33, DMI were lower (P < 0.05) in S, SK1, and SK3 than in C animals. From d -1 to 35, ADG were lower (P < 0.05) in S, SK2, and SK3 compared with C animals. A higher (P < 0.05) incidence of standing postures and lower incidence of lying postures was observed in S compared with C during the first 6 h after treatment. However, the higher (P = 0.02) incidence of abnormal standing activities observed for S was reversed (P < 0.05) by the K treatments. In conclusion, surgical castration increased plasma cortisol and acute-phase proteins and decreased immune function, feed intake, and growth rate. Ketoprofen effectively reduced the cortisol response to castration, but there was no advantage in treating with two split doses of K (1.5 mg/kg of BW per dose). A repeated K dose 24 h after treatment (3 mg/kg of BW) had no influence on changes in acute-phase proteins and immune response. Systemic analgesia with K is an effective method for alleviating acute inflammatory stress associated with castration.  相似文献

In Canada, piglets receive analgesia to control pain after surgical castration. There is interest in examining the potential to mix non-steroidal anti-inflammatory drugs with iron dextran prior to injection to minimize piglet handling and labor. The objective of this study was to compare pharmacokinetics and the relative bioavailability of ketoprofen given alone (3.0 mg/kg IM) versus the same dose of ketoprofen mixed with iron dextran (52.8 mg/kg IM) (ketoprofen + iron dextran) before injection in piglets. Piglets 8 to 11 d old were allocated into 2 treatment groups (n = 8/group). Plasma drug concentrations were measured using mass spectrometry at 13 time points after injection. No significant differences were detected between the 2 groups when examining pharmacokinetic parameters (e.g., Cmax, Tmax, AUC) or relative bioavailability for either S- or R-ketoprofen enantiomers (P > 0.05). However, pain control efficacy and food safety studies of these formulations are required to further examine this practice.  相似文献

As a part of ongoing research to further elucidate frequent and species-specific causes of differences in oral bioavailability, a 3 mg/kg dose of racemic ketoprofen, a high permeability/low solubility compound in the human biopharmaceutics classification system, was administered intravenously and orally to different species. Due to possible enantioselective disposition kinetics and inversion, enantiomers were quantitated separately using a stereospecific HPLC assay. The absolute bioavailability of R(−) and S(+) ketoprofen in chickens, turkeys, dogs and pigs was 31.5% and 52.6%, 42.6% and 32.5%, 33.6% and 89.1%, and 85.9% and 83.5% respectively. Incomplete bioavailability in poultry is probably due to incomplete absorption in addition to first-pass elimination. Low bioavailability of R(−) ketoprofen in dogs, strongly indicates first-pass metabolism. High bioavailability of S(+) ketoprofen in dogs and both enantiomers in pigs confirms that absorption of these substances is complete and controlled by gastric emptying rather than dissolution.  相似文献

The absolute bioavailability of R(−) and S(+) ketoprofen in chickens, turkeys, dogs and pigs was 31.5% and 52.6%, 42.6% and 32.5%, 33.6% and 89.1%, and 85.9% and 83.5% respectively. Incomplete bioavailability in poultry is probably due to incomplete absorption in addition to first-pass elimination. Low bioavailability of R(−) ketoprofen in dogs, strongly indicates first-pass metabolism. High bioavailability of S(+) ketoprofen in dogs and both enantiomers in pigs confirms that absorption of these substances is complete and controlled by gastric emptying rather than dissolution.  相似文献

The purpose of this field trial was to determine if maximal care of pigs from birth until 16 d of age would result in a significant alteration in the survivorship and growth performance of the pigs compared with control pigs born in the same time period. Sows were randomly assigned to treatment group prior to farrowing. Control pigs received the standard, commercial farm care. In maximal care litters, pigs were dried off at farrowing, given a rubber mat in the creep area, and given electrolytes, chilled pigs were warmed and given colostrum or glucose, surgical instruments used for processing were dipped into an antiseptic between pigs, the castration wounds were sprayed with iodine, and sows were fed 3 times rather than twice a day. Pigs that received the maximal care weighed 170 g (+/− 80 g) more at 16 d of age than standard care pigs. Factors that reduced weight at 16 d included having a low birth weight, nursing a gilt or a parity 5–6 sow, nursing in a large litter, being clinically ill or being lame after 3 d of age, being cross-fostered and nursing an ill sow. In general, maximal care did not reduce mortality. Providing maximal care did improve weaning weights and enabled small birth weight pigs to reach 3.7 kg at 16 d of age.  相似文献
