The ontogeny of hepatic growth hormone (GH) receptors (GHR), as measured by responses of both plasma insulin-like growth factor-I (IGF-I) and hepatic GHR to an exogenous bGH stimulus, was examined using sheep of different ages (Days 1-7, 14-21, 28-35, and 56-63 of life, and yearlings). The IGF-I response to bGH was first examined in yearling sheep using two doses of bGH (0.1 and 0.2 mg/kg LW/d). Based on these results, lambs in four groups up to Day 63 of life were treated for 5 d with bGH (n = 10) at a dose of 0.15 mg/kg LW/d or with saline (n = 10). Jugular blood samples were taken once daily on Days - 1, 4, and 5 of treatment. bGH treatment in lambs up to Day 63 of life had little effect on plasma concentrations of GH, insulin, glucose or urea, but significantly (P < 0.05) increased circulating concentrations of IGF-I at all ages and of NEFA at Day 62/63 of life. In contrast, bGH treatment at either dose in yearlings significantly increased these parameters, except for plasma urea concentrations which were decreased in bGH-treated yearlings. However, the responses of plasma IGF-I concentration to bGH stimulus in lambs up to Day 63 of life were small compared to those in yearling sheep. Consistent with this, bGH treatment failed to affect hepatic GH binding in young lambs, but up-regulated it in yearling sheep. Furthermore, basal (unstimulated) GH binding did not differ between sheep of 7 vs. 63 vs. 365 d of age, despite the greater IGF-I responses to bGH in the latter group. It is suggested that hepatic GHR in lambs up to Day 63 of life are not fully functional compared to the situation in yearlings.  相似文献

Renal disorders in lambs may be congenital, infectious, toxic, immunologically mediated or secondary to urethral obstruction. Congenital abnormalities are uncommon. Kidney infections often are only one aspect of more widespread diseases, for example, tick pyaemia or salmonellosis. Toxic diseases, which affect mainly the renal tubules, may be caused by bacterial, chemical or plant toxins, although in lamb nephrosis the cause is unknown. Immunecomplex glomerulonephritis is uncommon, and one form, mesangiocapillary glomerulonephritis of Finnish landrace lambs, is under genetic control. Urolithiasis may have secondary effects on the kidneys.  相似文献

Several parameters of immune competence were measured in lambs with genetic resistance to Haemonchus contortus and in lambs with average susceptibility to H contortus (random-bred). Increase in skinfold thickness following intradermal injection was greater in resistant lambs for concanavalin A and to a lesser extent for phytohaemagglutinin. Both mitogens induced infiltration of superficial and deep dermis by eosinophils, CD4+, CD8+ and T19+ lymphocytes, though the responses did not differ between groups. Primary but not secondary antibody responses to the T cell-dependent antigen, ovalbumin was greater in resistant lambs. In contrast, antibody responses to the T cell-independent antigen, Brucella abortus lipopolysaccharide, did not differ between groups. Taken together the data suggest that enhanced T lymphocyte function may contribute to the greater resistance to H contortus of genetically resistant lambs.  相似文献

Lambs were inoculated with Ehrlichia phagocytophila infected blood at the age of either two or six weeks. The temperature response and degree of parasitaemia were both most pronounced in the lambs inoculated at six weeks old. The half-life of antibodies to E phagocytophila was estimated to be 17.5 days. Lambs inoculated with tick-borne fever (TBF) had lower liveweights in the autumn than TBF-free controls.  相似文献

Acute renal failure was diagnosed by clinical, necropsy and histological criteria in 39 flocks (20 low ground, 13 hill and six marginal upland) in areas served by six veterinary investigation centres. Forty-eight lambs of 12 different breeds or crosses were investigated. The mean age of affected lambs was 38 days (range seven to 84 days); 21 lambs (44 per cent) were aged seven to 28 days, while only eight (17 per cent) were older than two months. Mortality in clinically affected lambs was almost 100 per cent, with no response to various treatments. Histological examination showed that 40 lambs (83 per cent) had nephrosis, while the rest had toxic tubular necrosis, interstitial nephritis or tubular damage associated with oxalate crystal deposits. Only about half of the lambs had any evidence of enteric infections or enteropathy. Acutely ill lambs had azotaemia, haemoconcentration and proteinuria; some lambs had glycosuria or haematuria. Samples of plasma from 22 lambs with nephrosis were compared with similar samples from 82 incontact but asymptomatic lambs. The clinically affected group had significantly elevated plasma urea, creatinine, total protein, globulin, phosphorus and chloride concentrations and significantly reduced plasma calcium concentrations compared with healthy lambs. Affected lambs had a significant reduction also in the calcium:phosphorus ratio. No significant differences between groups was found in plasma concentrations of albumin, glucose, lactate, glycerol, creatine kinase, alkaline phosphatase, sodium, potassium or magnesium.  相似文献

The effects of cold stress were studied in pregnant ewes during the last three weeks of gestation and in their progeny during the first three days of life. In general, ewes were unaffected by treatment whereas changes were observed in the cold-stressed lambs. Cold-induced changes in lambs included physical weakness, depression, and poor nursing response. Serum concentrations of glucose and insulin were lowered whereas concentrations of blood urea nitrogen, alkaline phosphatase, aspartate aminotransferase, lactate dehydrogenase, triglycerides, and cortisol tended to be higher in cold exposed lambs. The mortality rate was higher (40%) in cold-stressed lambs than in lambs kept at warmer temperatures (10%). At necropsy, cold-exposed lambs had reduced amounts of adipose tissue in perirenal areas, and extensive subcutaneous hemorrhages and edema in the distal portions of the thoracic and pelvic limbs.  相似文献

Pain and distress following castration were assessed in lambs using three indicators: behaviour, plasma cortisol and mechanical nociceptive thresholds. Three castration methods: rubber ring (RR), combined ring and Burdizzo clamp (CM) and surgery (SU) were compared. The effects of castration following local anaesthetic pre-treatment (LA) and castration performed under general anaesthetic (GA) were compared to castration performed with no anaesthetic. This gave a 4 x 3 x 3 block design i.e. 36 experimental treatments. Six lambs were allocated to each treatment i.e. 216 lambs were used in all. SU produced the greatest response, followed by RR then CM castration. LA abolished the responses to RR and CM castration but had no effect on the response to SU castration. GA did not reduce the responses to RR and SU but abolished the rise in mechanical nociceptive thresholds and markedly attenuated active pain behaviours in lambs CM castrated without anaesthesia. This suggests that the clamping procedure itself rather than post-castration pain and distress is responsible for the rise in nociceptive thresholds and active pain behaviours in CM castrated lambs.  相似文献

Abomasal bloat in young lambs in clinically characterized by a distension of the abdomen, especially on the right side. The significant finding at necropsy is a gas-filled abomasum.

Abomasal bloat was studied over a 2-year period in 28 flocks located in Alvdal and Rendalen, two mountain valley communities in south-east Norway. The herd incidence rate varied from 0 to 20%. Abomasal bloat was most often seen 15–30 days after birth, and most cases occurred in the period just before and after the lambs were turned out to spring pasture.

There was no significant difference in the incidence rate of abomasal bloatin single, twin and triplet lambs. The following factors were found to have a significant effect on the incidence rate: geographical area, type of floor, and interaction between the type of floor and the % grass silage in the roughage. Highest rates were found in flocks with built up litter floors and more than 25% silage. Substituting perforated floors for built up litter floors in 3 sheep houses reduced the incidence rate markedly.

The aetiology of the disease is discussed. It is suggested that young lambs housed on built up litter with a wet consistency nibble hay contaminated with faeces. This can lead to an excessive gas-producing flora in the abomasum.  相似文献

Five out of 60 purebred Suffolk lambs died when four to six weeks old with a nephrosis characterised by heavy deposits of crystals resembling calcium oxalate in the tubules. The source of oxalate was not clearly identified but may have resulted from oxalate-forming mould contamination of concentrates fed to both ewes and lambs.  相似文献

Lambs at 4-5 weeks of age were studied during the first 4 hours after castration and/or tailing using three methods in various commonly used combinations. The methods were cutting with a knife, application of constricting rubber rings and using a heated docking iron (tailing only). The integrated cortisol response (area under the cortisol curve) was determined for each lamb during the first 4 hours after treatment and was considered to reflect the overall magnitude of the lamb's acute distress response. On that basis, cut lambs (tailing only, castration only, castration plus tailing) experienced more distress than any other groups. Also the distress response (indicated by elevated plasma cortisol concentrations) lasted longer than 4 hours in cut lambs, unlike all other groups. The use of rings apparently caused similar distress when lambs were castrated only, were castrated plus tailed, or were short-scrotumed (testes pressed against the abdominal wall by a distally located scrotal ring) plus tailed. The magnitudes of distress apparently caused by tailing alone with a ring or a docking iron were similar and were lower than the distress caused by any other castration and/or tailing procedure. It is concluded that acute distress responses to these husbandry procedures would be minimised in lambs of this age if rings and/or a docking iron were used in preference to a knife.  相似文献
