The Orchidaceae contains a large number of threatened species that could benefit from seed banking. However, successful seed storage requires a greater understanding of the enigmatic seed biology of orchids. In particular, there is little documentation of the presence or timing of the maturation drying that has been shown to be critical for longevity of agricultural seeds in storage. The aims of this work were to document seed water content, germinability and desiccation tolerance as a function of maturation in developing capsules of hand-pollinated flowers of Phalaenopsis hybrids. Lengthening of capsules was complete by 50 DAP, but capsule diameter continued to increase up to a maximum at 70 DAP. Relative water content rapidly declined from 75% moisture content (fresh weight basis) at 150 DAP to 50% moisture content at 165 DAP where it remained until dehiscence. Seed germinability remained high (75–99.5%) throughout the harvesting period (90–216 DAP), but desiccation tolerance only developed 165 days after pollination. Therefore, if Phalaenopsis seeds are to be stored, we propose that their harvest should be delayed until moisture content drops below 50%.  相似文献

The effects of different types of chitosan on seed germination and protocorm development were determined for two orchid species, Dendrobium bigibbum var. compactum and Dendrobium formosum. Six chitosan types derived from polymer or oligomer chitosan each with 70, 80 or 90% levels of deacetylation (P70, P80, P90, O70, O80 and O90, respectively), were evaluated as direct medium supplements at 0, 10, 20, 40 or 80 mg/L in modified VW medium by following seed germination and protocorm growth for 12 weeks. Chitosan of all six tested types and four concentrations were found to significantly enhance the proportion of D. formosum seeds that germinated, when compared to these germinated without chitosan. In contrast, chitosan caused no enhanced germination rate was noted for D. bigibbum var. compactum with all tested chitosans and doses tested. However, almost all types of chitosan at 10 mg/L, except O90, were able to significantly improve the growth of D. bigibbum var. compactum protocorms, whilst 10 or 20 mg/L of P70 chitosan was the best formula to enhance the growth of D. formosum protocorms. It is concluded that chitosan responses in seed germination and protocorm development were somewhat species and developmental stage dependent. Therefore, the appropriate chitosan application for each plant species should be evaluated first before use.  相似文献

To develop efficient seedling production methods for Laccosperma secundiflorum and Eremospatha macrocarpa, a study was conducted to examine regeneration using offsets combined with several physical and chemical treatments of seeds. Offsets categorized into small, medium and large diameters, were planted in three conditions: shaded and open nursery, and greenhouse. We tested sucker from E. macrocarpa, and sucker and rhizome from L. secundiflorum. For both species, high viability percentage (ranging from 55% to 100%) were observed for small and medium suckers planted in shaded nursery and greenhouse, against less than 49% for sucker planted in open nursery. The mean seedling emergence times were estimated to 84, 77 and 75 days after planting (DAP) for small, medium and large sucker of L. secundiflorum, respectively under open nursery condition, and 76, 75, 95 DAP for small, medium and large suckers of the same species, respectively in shaded condition. Greenhouse has a significant positive effect on E. macrocarpa seedlings emergence time. For this species, the mean seedling emergence times were estimated to 43 DAP for small sucker and 76, 93 DAP for medium and large suckers. No seedling was obtained from rhizome planted in all the growing conditions tested. Concerning seed dormancy breaking, germination percentages and rates were determined for 13 treatments. The best treatments were pre-soaking unscarified seeds for 4 days in 1.01 g l−1 and 0.10 g l−1 KNO3, with 79% and 68% of germination, respectively and in 3.46 × 10−3 g l−1 GA3 for 68% of germination. These methods are suggested to improve germination of L. secundiflorum seeds. Successful and recommended methods for E. macrocarpa are pre-soaking scarified seeds in 3.46 × 10−3 g l−1 and 3.46 × 10−4 g l−1 GA3, 96% and 94% of germination, respectively. Dormancy, probably a combination of mechanical and chemical dormancy, is present in the two species.  相似文献

Efficient protocols were established for in vitro seed germination, neo-formation of secondary (2°) protocorms from primary (1°) protocorms and multiple shoot buds and protocorm-like body (PLB) induction from pseudo-stem segments of in vitro-raised seedlings of Cymbidium giganteum. Four nutrient media, namely Murashige and Skoog (MS), Phytamax (PM), Mitra et al. (M), and Knudson ‘C’ (KC) were evaluated for seed germination and early protocorm development. In addition, the effects of peptone, activated charcoal (AC) and two plant growth regulators [6-benzylaminopurine (BAP) and 2,4-dichlorophenoxyacetic acid (2,4-D)] were also studied. Both M and PM supplemented with 2.0 g l−1 peptone or 1.0 mg l−1 BAP resulted in ∼100% seed germination. Media supplemented with 2.0 g l−1 AC could effectively induce large protocorms (1.6 ± 0.1 mm in diameter). Neo-formation of 2° protocorms from 1° protocorms was achieved in liquid and agar-solidified PM medium fortified with different concentrations and combinations of auxins (α-naphthalene acetic acid (NAA) and 2,4-D) and cytokinins [BAP and kinetin (KN)]. The highest number of 2° protocorms was obtained in liquid medium (10.7 ± 0.9/1° protocorm) supplemented with 2.0 mg l−1 BAP + 1.0 mg l−1 NAA. Although protocorms proliferated profusely in liquid medium, these did not develop further unless transferred to agar-solidified medium within 6–8 weeks. Multiple shoot buds and PLBs were induced from pseudo-stem segments on agar-solidified PM medium fortified with different concentrations and combinations of BAP and NAA and the maximum number of PLBs (6.00 ± 0.20) was recorded when BAP and NAA were applied at 2.0 mg l−1 each. A solid root system was induced from PLBs and shoot buds when these were transferred to half-strength PM or M media fortified with 0.5 mg l−1 indole-3-acetic acid. Well-rooted plants were transferred to the greenhouse with 95% survival.  相似文献

This study was conducted to determine the effects of coconut water (CW) and activated charcoal (AC) on multiplication of Phalaenopsis gigantea protocorms. The protocorms used for this study were obtained by germinating seeds in vitro. Protocorms with trimmed and untrimmed bases were cultured on XER basal medium containing 0, 10, 15 or 20% (v/v) CW; and 0, 1, 2 or 2.5 g AC l−1. Trimmed protocorms exhibited the highest percentage of proliferation on a medium containing 15% (v/v) CW and 2.5 g AC l−1 (56.82 ± 38.86%) with an average of 4.24 ± 2.89 protocorms formed per protocorm. Untrimmed protocorms cultured on a medium containing 20% (v/v) CW without AC produced the highest percentage of new protocorms (6.93 ± 6.28%) with an average of 0.72 ± 0.57 per protocorm. When CW was added to a medium singly, 10% (v/v) CW induced a higher degree of proliferation on trimmed protocorms (5.68 ± 10.14%) with an average 0.50 ± 0.84 new protocorms per protocorm. Untrimmed protocorms proliferate to a much lower extent (2.57 ± 2.74%) with an average of 0.72 ± 0.57 protocorms per protocorm when cultured on a similar medium. A high concentration of CW enhanced proliferation on untrimmed protocorms, but increased mortality of trimmed protocorms. The addition of CW with AC to media increased protocorm proliferation and survival of both trimmed and untrimmed protocorms. When cultured on all media, trimmed protocorms produced a higher number of new protocorms (an average 0.5–7.0) as compared to untrimmed protocorms (0.3–1.9). Comparative studies showed that trimmed protocorms produced up to 10 times more new protocorms than untrimmed ones. Altogether this study showed that trimmed protocorms cultured on a medium containing CW and AC can be used for high-frequency multiplication of P. gigantea seedlings.  相似文献

Vanilla (Vanilla planifolia) is a crop of great commercial importance as the source of natural vanillin, a major component of flavor industry. The primary gene pool of V. planifolia is narrow and is evidently threatened due to destruction of its natural habitats making the secondary gene pool important as a source of desirable traits especially for resistance to diseases. Many species of vanilla are considered rare and endangered hence an urgent need to conserve them, arises. Effective procedures for micropropagation and in vitro conservation by slow growth in selected species of vanilla, are described. Synthetic seed technology was standardized by encapsulating 3–5 mm in vitro regenerated shoot buds and protocorms in 4% sodium alginate, which could be stored up to 10 months with 80% germination in sterile water at 22 ± 2 °C. In vitro conservation technology of Vanilla was standardized and shoot cultures could be maintained for more than 1 year without subculture, on slow growth medium, i.e. Murashige and Skoog medium supplemented with 15 g l−1 each of sucrose and mannitol in sealed culture vessels at 22 ± 2 °C. These cultures were maintained in vitro for more than 7 years with yearly subculture. The conserved material could be retrieved and multiplied normally in MS medium with 1.0 mg l−1 BA and 0.5 mgl −1 IBA. The in vitro conserved plants showed good growth and developed into normal plants. This synseed and in vitro conservation system can be utilized for conservation and exchange of vanilla genetic resources.  相似文献

The effects of sodium hypochlorite treatment and growth medium on in vitro seed germination and seedling growth of the leopard orchid (Ansellia africana Lindl.) were investigated. Forty minutes treatment of seeds with 1.75% sodium hypochlorite stimulated germination (70.6%) and seedling development when measured at Stage 5 (emergence of first leaf) on P668 medium after 8 weeks of culture. The growth medium played a significant role in determining the germination response of A. africana seeds. Dark pretreatment of seed cultures significantly enhanced the germination percentage and the growth of rhizoids on the protocorms. Leaf growth in terms of length was substantially enhanced on P668 medium. It was significantly reduced in modified Knudson C medium after 16 weeks of culture. Seedlings developed on P668 medium showed a significantly better growth performance in relation to leaf length, leaf number, root length, fresh and dry weights per plant in vermiculite after 12 weeks of ex vitro growth in a mist house with 90% relative humidity. Further studies developing a suitable method for in vitro symbiotic germination could assist in reintroduction of this threatened orchid species into the wild.  相似文献

Seeds of wild asparagus (Asparagus acutifolius L.) were treated and compared in this research to investigate seed dormancy class and level involved in this species. Four seed lots were compared: (i) freshly harvested seeds in 2007 (07Fr); (ii) freshly harvested seeds in 2008 (08Fr); (iii) after-ripened (AR) 2007 seeds dry stored in glass jars (ARg); (iv) AR 2007 seeds dry stored in paper bags (ARp). The 07Fr seeds were exposed to (1) chemical scarification combined with gibberellic acid (GA3) levels (0, 200, 400, and 600 mg L−1) and to (2) 28-day moist stratification at 5 and 23 °C, and two sequences of 5/23 °C combined with 0 and 400 GA3 mg L−1 levels, and (3) together to the 08Fr and AR seeds were exposed to 56-day moist stratification at 5, 23, or 5/23 °C. With the 08Fr and AR seed lots this last stratification treatment was combined with 0 or 800 GA3 mg L−1 levels. The dormancy depth of 08Fr (32% germination) was less than 07Fr seeds (2%). The latter after-ripened during dry storage and when stored in glass germinated more (47.5%) than in paper (12%). Stratification for 4 weeks was ineffective in improving germination of 07Fr seeds; when chemically scarified they did not germinate at all. The highest (nearly 70%) and the most rapid and uniform germination were observed for all the lots when they were warm stratified for 56 days. Warm stratification improved germination more than alternate temperature stratification, while cold stratification inhibited germination especially for the 08Fr and ARg lots, thus seeds seem not to have a morphological component to their dormancy. GA3 only improved germination of 07Fr seeds, at a low rate. A. acutifolius seeds fit the characteristics of a non-deep physiological dormancy.  相似文献

The application of modern biotechnology for conservation of any endangered species requires an efficient in vitro regeneration protocol. In this study a reliable protocol was developed for in vitro seed germination, protocorm multiplication and subsequent plantlet regeneration of Vanda coerulea, an endangered orchid species. Among the four basal media evaluated for asymbiotic seed germination, Phytamax was found to be the best followed by Murashige and Skoog (MS). Phytamax was also found good for protocorm development. For protocorm like body (PLB) regeneration, protocorms were then further cultured on Phytamax media fortified with different phytohormones either individually or in combinations. The frequency of protocorm like body (PLB) regeneration significantly relied on kinds and concentrations of plant growth regulators used. A combination of 1-naphthaleneacetic acid (NAA) (5.36 μM) and 6-benzyle amino purine (BAP) (3.80 μM) was found to be suitable for maximum PLB regeneration. Healthy plantlets were induced from PLBs when cultured on same basal medium supplemented with activated charcoal (AC – 3.0 g/l). Plantlets with well developed leaves and roots were transplanted to pots filled with a mixture of charcoal, brick pieces and sphagnum moss and transferred to the greenhouse. This protocol will enable mass propagation and conservation of this exquisite orchid.  相似文献

The bitter gourd seed has a thick, hard seed coat. Mildew often occurs during germination and causes uneven and low rates of seed germination. However, the problems caused by mildew can be overcome by treating seeds with hot water, by soaking in water, or by using microorganisms. Seeds of the ‘Ching Pi’ bitter gourd were treated in a water bath at 60 °C for 10 min and then soaked in tap water at 25 °C for 24 h. The resulting germination percentage was 86.7%, and the resulting percentage of mildewed seeds was 10%. The biocontrol potential of three commercially available Bacillus subtilis solutions was examined. For seeds primed with Huodijun B. subtilis solution, the germination percentage was 73.3% and the mildewed percentage 6%. In dual cultures, the antibiotic content in the Huodijun B. subtilis solution was significantly greater than in Yunghsing and Huolibao, the other B. subtilis solutions examined. B. subtilis effectively inhibited the growth of Rhizoctonia solani mycelium and caused abnormal mycelial growth.  相似文献

The influence of seed maturity, seed storage and germination pre-treatments on seed germination of cleome (Cleome gynandra L.) were investigated. Seed maturation studies showed that capsules harvested at 18 days after anthesis possessed the highest dry weight with 19.2% moisture and 1% germination. Development of fresh-ungerminated seed was observed with increasing maturity of fruit, suggesting that cleome exhibited forms of seed dormancy. Storing mature seed at 15 °C and at room temperature for 5 months showed that seed dormancy was broken after 3 months under both storage regimes. When mature seeds were subjected to different treatments including various levels of GA3, KNO3, leaching, pre-chilling, soaking and pre-heating at different temperatures, it was found that pre-heating at 40 °C for period of 1–5 days was the most effective method in breaking dormancy in cleome.  相似文献

Passiflora mollissima, Passiflora tricuspis and Passiflora nov sp. are three passion fruit species occurring in Bolivia. Germination percentages and rates were determined for 11 different treatments. Per species, germination of 100 seeds was monitored every 3 days, during 90 days. Germination started after 9 days and 50% of final germination was reached within a month or less. Successful, recommended methods for P. mollissima are removing the basal point of seeds (27% germination) or removing the basal point in combination with pre-soaking seeds for 48 h in 50 ppm GA3 (18%). Pre-soaking seeds for 24 h in 400 ppm GA3 (42%) and removal of the basal point in combination with pre-soaking seeds for 48 h in 50 ppm GA3 (36%) are suggested methods to improve germination of P. nov sp. Removing the apical point of P. tricuspis seeds resulted in maximal germination (57%). No unique treatment gave satisfactory results for the three species tested. Exogenous dormancy, probably a combination of mechanical and chemical dormancy is present in the three species studied. Presence of physical dormancy was found in P. mollissima.  相似文献

Wild asparagus (Asparagus acutifolius L.) is a widespread species found in all the Mediterranean areas. The spears are highly valued by consumers and owing to its frugality, this species is a feasible new crop with high income potential, especially for Mediterranean marginal areas. Currently, the cultivation of this species is limited because of its low and erratic seed germination that makes difficult the production of seedlings for plant propagation. In this research, non-after-ripened (1 month-old) and after-ripened seeds (dry stored at room temperature for 13 months) were exposed for 30 days in the dark to three moist stratification treatments: cold (5 °C), warm (23 °C) or no stratification; subsequently they were soaked for 12 h in warm water (35 °C) or not soaked. The effect of these pre-germination treatments on three germination parameters (germination percentage, time to 50% of final germination – T50 – and germination pattern) was studied, as well as some possible seed dormancy forms involved therein. The 1-year dry storage period proved to be effective in after-ripened seeds by enhancing seed sensitivity to the subsequent pre-germination treatments. After-ripened seeds exhibited higher and more rapid germination compared to non-after-ripened seeds. Soaking, cold or warm moist stratification had similar single effect on non-after-ripened seeds (27% germination). With after-ripened seeds, only soaking or warm stratification were effective (47% germination) when singularly applied, while cold stratification did not improve germination. By combining stratification and soaking treatments, a higher germination for both non-after-ripened and after-ripened seed-lots was achieved. The highest germination was obtained when after-ripened seeds were stratified and soaked (76%), without any significant difference between cold or warm stratification. Single or combined application of moist stratification (regardless of the temperature used) and soaking resulted always in a faster germination compared to that of no-treated seeds and especially with after-ripened seeds (T50 = 6 days). A non-deep type 1 physiological dormancy can be hypothesized for the seeds of this species. Low stratification temperature induce secondary dormancy in after-ripened seeds that can be removed by soaking them at 35 °C for 12 h.  相似文献

Aqueous slurries of commercial preparations of Trichoderma harzianum Rifai strain KRL-AG2 G41 (Th), T. virens Strain G-41 (Tv), or their combination (ThTv, at half rates each of the single application rate) were applied to ‘Marktetmore 76’ cucumber seeds (Cucumis sativus L.) that were non-primed or primed for 3 days at 25 °C either osmotically (−2.5 MPa from 0.337 molal Ca(NO3)2) or osmomatrically (−1.0 MPa from 0.135 molal Ca(NO3)2 plus −1.5 MPa from 50% water in exfoliated grade 5 vermiculite). Slurries were applied to seeds (1 mg per seed) either before or after priming. Seeds were sown in soilless, peat-based media with or without inoculation with Pythium aphanidermatum (Pa). Protection against damping-off caused by high pressures of Pa (16% emergence in non-coated, non-primed seeds) was increased by slurry coating Th on non-primed (76.4% emergence) or on osmotically primed seeds, with coating either before or after priming having no effect on efficacy (average 62.6% emergence). Slurry coating Th on osmomatrically primed seeds failed to increase final emergence percentage (FEP). Colony forming units per three seeds (CFU, all 103) was 2.8 for non-primed seeds, and 3.2 and 2.6, respectively, when osmotically and osmomatrically primed seeds were coated after priming. In a second study with lower disease pressure (58.1 FEP from non-coated, non-primed seeds), slurry coating of non-primed or osmotically primed seeds with Th, Tv or ThTv reduced percentage damping-off and increased FEP. The combination coating eliminated damping-off only in non-primed seeds, and tended to reduce percentage damping-off in osmotically or osmomatrically primed seeds compared to coating with Th or Tv alone. In a third study using only non-primed seeds, slurry coatings with mefenoxam fungicide, Th, Tv, or ThTv decreased total damping-off to 2.6%, 7.4%, 2.0%, and 0%, respectively, from the 30.1% occurring in non-coated seeds. Th, Tv or ThTv applied to growth media at the same rate as the seed coating (1 mg per seed) were generally as effective as the seed coatings, and only the ThTv growth medium application eliminated damping-off. A fourth experiment revealed that Th, Tv or ThTv remained viable on non-primed seeds for up to 4 weeks (the longest storage duration) at 21 or 4 °C, but 21 °C storage resulted in faster seed germination by week 3 and higher CFU per three seeds by week 4. In summary, coating of non-primed seeds with Th, Tv or ThTv was more effective than coating primed seeds in reducing percentage damping-off. While priming treatments generally led to faster seedling emergence and greater seedling shoot fresh weight than was achieved with non-primed seeds, only for non-primed seeds was damping-off eliminated by the ThTv seed coating or growth medium application.  相似文献

In vitro symbiotic seed germination is an important tool not only for the study of orchid-fungus specificity but also for the production of mycobiont-infected healthy seedlings that could be valuable for both horticultural and conservation purposes. The current study compared effectiveness of eight putative orchid mycorrhizal fungi obtained from mature orchids in the genera Paphiopedilum, Cymbidium and Dendrobium, in promoting in vitro seed germination and protocorm development of Grammatophyllum speciosum Blume and Dendrobium draconis Rchb. f., native Thai orchids. The developmental stages of seeds and protocorms cultured on Murashige and Skoog (MS) medium, oat meal agar (OMA), or OMA inoculated with one of the eight fungal isolates were evaluated weekly. Two isolates of Epulorhiza repens (Bernard) Moore (=anamorphic species of Tulasnella calospora (Boud.) Juel), Da-KP-0-1 and Pv-PC-1-1, were found to be the most effective fungi in promoting protocorm development of G. speciosum. At week 13, protocorms co-cultured with either one of these two fungal isolates, on the average, were significantly more advanced than those sown on OMA. Protocorms co-cultured with isolate Pv-PC-1-1 were also significantly more advanced than those cultured on MS medium. For D. draconis seed germination, three fungal isolates of different anamorphic species of Tulasnella, C1-DT-TC-1, Pv-PC-1-1, and C3-DT-TC-2, were found to be the most effective fungi in promoting protocorm development. However, none of these fungal isolates outperformed MS medium. Additionally, the compatibility between the fungal isolates tested and the two orchid species was discussed.  相似文献

This study was initiated to investigate the differences in germination percentages and rates between Corylopsis coreana Uyeki and Corylopsis sinensis var. calvescens Rehder & E.H. Wilson following a warm stratification (WS) and cold stratification (CS), and to study the effect of different WS temperatures interacting with different durations of CS. Warm stratification at 10 °C, 15 °C, 20 °C, and 25 °C was given for 1 month (1 M 10 °C, 15 °C, 20 °C, and 25 °C WS) followed by 0 M, 1 M, 2 M, and 3 M of CS at 5 °C (0 M, 1 M, 2 M, 3 M CS) and seeds were germinated in an air conditioned greenhouse maintained at 18.5 °C/18 °C. On average, less than 1% of C. coreana seeds germinated when sown without any WS and CS or with 1 M 15 °C, 20 °C, and 25 °C WS without CS treatment. However, 26% C. coreana seeds germinated after 1 M 10 °C WS without any CS treatment. Germination was not affected by WS temperatures when followed by 2 M 5 °C CS. It is concluded that C. coreana exhibited low seed germination at 10 °C and that this temperature could be considered the upper limit of CS for C. coreana. Only 2 M CS was required for more than 90% seeds to germinate. However, C. sinensis var. calvescens required longer than 3 M CS for more than 29% seeds to germinate. This clearly shows that there is an interspecific variation in optimum dormancy-breaking requirements.  相似文献

Michelia yunnanensis Franch. is a Chinese endemic ornamental shrub with potential for greater utilization as a landscape and medicinal plant if propagation was less difficult. Seed development and breaking of seed dormancy were investigated to improve propagation of M. yunnanensis. No fresh seeds germinated when tested at the time of dispersal. Newly matured seeds of M. yunnanensis contained differentiated linear underdeveloped embryos that were physiologically dormant. The embryo/seed length ratio of M. yunnanensis was 0.15. Warm stratification did not break seed dormancy. Dormancy was broken by cold stratification at 4 °C but not by flowing water or nitrate. Embryos developed grew inside seeds during cold stratification at 4 °C. In newly harvested dormant seeds, embryos were 0.94 mm long and increased in length 139% before radicle emergence (germination). GA3 substituted for cold stratification to break dormancy in seeds of M. yunnanensis incubated at 25 °C or 20/25 °C. Mature M. yunnanensis seeds exhibited intermediate complex morphophysiological dormancy. Optimal germination of non-dormant seed in terms of both germination percentage and rate occurred at 20/25 °C.  相似文献

A study was carried out to investigate the influence of desiccation and freezing followed by various presowing rehydration procedures on the desiccation sensitivity of the seed of Citrus suhuiensis cv. limau madu. The freshly harvested seeds of limau madu were desiccated under a broad range of relative humidity (RH) to various equilibrium water contents (g H2O g−1 dw). The desiccated and desiccated–frozen seeds were either directly sown under germination conditions or subjected to presowing rehydration procedures: seed preheating, prehumidification and osmoconditioning. The hydrated and desiccated seeds were sown in controlled germination conditions and the survival was evaluated 4–6 weeks after sowing. The results showed that desiccation progressively reduced the percentage of normal seedling of the seeds of limau madu and the viability is almost lost at water contents below 0.08 g H2O g−1 dw. The estimated desiccation sensitivity was substantially high (WC50 = 0.143 g H2O g−1 dw) when the desiccated seeds were rapidly rehydrated (uncontrolled rehydration). In contrast, seed prehumidification, preheating and osmoconditioning (controlled rehydration procedures) markedly enhanced normal seedling percentages decreasing the estimated values of WC50 (between 0.08 and 0.127 g H2O g−1 dw). While the rapidly rehydrated desiccated–frozen seeds were almost killed at water content of 0.15 g H2O g−1 dw, prehumidification and preheating have noticeably increased percentage of frozen seeds survival at the same water content. However, at water content of 0.21 g H2O g−1 dw preheating significantly (P < 0.05) increased percentage of normal seedling of the frozen seeds. Seed desiccation markedly reduced the percentages of germinated seeds with multiple seedlings. Seed controlled rehydration remarkably increased the survival of polyembryos. The beneficial effect of seed controlled rehydration on the survival of the desiccated seeds was pronounced at medium water contents (0.08–0.25 g H2O g−1 dw).  相似文献

Dessert quality of guava fruit is considerably reduced by high seed content. Number of seeds in guava is associated with different seed and fruit characters. Influence of different seed characters on number of seeds per fruit (NSPF) was studied by analysing character association and direct and indirect effects on NSPF and seed related traits for 68 genotypes of guava collected from diverse sources and conserved at National Active Germplasm Site, CISH, Lucknow. Seed related characters were studied at the colour break stage of the fruits using image analysis software for counting number of seeds. The data was subjected to path analysis to find out direct and indirect effects of different characters on number of seeds in the fruits. At genotypic and phenotypic levels, NSPF was significantly and positively associated with seed weight per fruit (SWPF), number of seeds 100 g−1 pulp (NSPHP) and fruit weight (FW). The genotypic (0.0029) and phenotypic (0.0563) residual values were fairly very low, which revealed that variables included in this study had significant contribution in determining NSPF. The NSPHP exhibited high positive direct effect on NSPF. The maximum direct response (P = 0.737, G = 1.004) of this component was attributed to the indirect positive effects of the SWPF and fruit:seed weight proportion (FSWP). SWPF also had a very high positive direct effect (P = 0.521, G = 0.694) on the NSPF. Correlation and path coefficient analysis revealed that SWPF, NSPHP and 100-seed weight (100SW) were deciding factors for realizing improvement for NSPF. The importance of small seeded genotypes in selection of less seeded varieties was emphasized.  相似文献

Effects of banana extract (BE) and 6-benzylaminopurine (BAP) were evaluated on asymbiotic seed germination and an early differentiation of protocorms and plant regeneration of Dendrobium lituiflorum Lindl. High percentage germination was achieved by culturing seeds on modified Knudson C medium supplemented with 10% (v/v) BE. Rapid regeneration was observed within 60 days of culture on 10% (v/v) BE supplemented KC medium where maximum percentage propagules showed development of leaves and root formation. Propagules on BAP supplemented KC medium showed no further development beyond one leaf stage. In another experiment, culture of shoots on 12.5% (v/v) BE supplemented KC medium led to multiplication, shoot elongation as well as vigorous rooting. Shoots cultured on 10 μM BAP supplemented MS medium showed maximum multiplication but these were stunted. Plants with well expanded deep green leaves and elongated roots from BE media were first hardened in vitro followed by ex vitro hardening on cocopeat:perlite (9:1) in the greenhouse conditions and exhibited 90% survival. The study emphasizes the role of BE as a natural additive at different stages of development from seed germination to plant regeneration.  相似文献
