Effects of the commercial product TrichoFlow WP™ (Agrimm Technologies Ltd., New Zealand), based on the fungus Trichoderma harzianum, on quality characteristics and yield of bulb onion was investigated. Bulb sets of the local cultivar Kantartopu was planted in soil with in and between row distances of 0.15 m and 0.40 m, respectively. The product, at considerably high dosages of 5 g m−2, 10 g m−2 and 15 g m−2, was mixed with water and sprinkled once to the plots at planting. Analyses of data at harvest did not show statistical significance for Trichoderma effect on total bulb yield, bulb diameter, leaf length, number of shoot apex, %titratable acidity, number of internal (fleshy) leaves, number of external (papery) leaves, %soluble solids and %bulbs with diameters of 20–39 mm, 40–69 mm and ≥70 mm. The yields obtained from the plots treated with the dosages of 5 g m−2, 10 g m−2 and 15 g m−2 and the control plots were 1063.7 kg da−1, 1051.0 kg da−1, 1066.5 kg da−1 and 985.0 kg da−1, respectively. Our results showed that high dosages of the Trichoderma product were not effective in enhancing onion bulb and yield characteristics under the given conditions.  相似文献

In the warm and wet north Florida climate, growing Italian parsley (Petroselinum crispum [Mill.]) is gaining popularity with small producers as a short duration crop on sandy soil. Application of compost to agricultural land can benefit the low fertile sandy soils in Florida and subsequent crop production, while providing an outlet for recycling municipal solid wastes (MSW) and biosolids. A field study was laid out in a randomized complete block design with four replications to evaluate the effects of compost (75% MSW:25% biosolids) application in comparison to fertilizer, fertilizer + compost (50:50), and control treatments on: (a) parsley fresh weight (FW), (b) soil and plant tissue nutrient concentrations, and (c) soil bulk density and moisture retention in winter and spring seasons. Soil amended with fertilizer or compost + fertilizer doubled parsley FW from 15.02 Mg ha−1 in the non-amended control plot to 30.75 and 32.67 Mg ha−1 in soils that received fertilizer + compost or fertilizer alone, respectively. Significantly higher total soil carbon (C) levels of 2.16% and 1.95% and nitrogen (N) levels of 0.19% and 0.16% were recorded in compost and fertilizer + compost treatments, respectively. Addition of compost reduced soil bulk density significantly to 1.03 Mg m−3 and increased soil moisture retention in simulated drier conditions at 1500 kPa to 0.12 m3 m−3 in plots that received only compost at the end of winter growing season. Overall, addition of compost resulted in improvement of both physical and chemical properties as well as increased parsley yields.  相似文献

The effect of enhanced ultraviolet-B on allelopathic potential of Zanthoxylum bungeanum was investigated. A significant inhibitory effect on germination rate of crop seeds under bioassay was observed at 25 g l−1 and 50 g l−1 by extracts from Zanthoxylum leaf both treated with enhanced ultraviolet-B radiation and untreated control. Medicago sativa and lettuce were more sensitive than radish to the extract from Zanthoxylum leaf treated with enhanced UV-B radiation, as the germination rates of M. sativa and lettuce were significantly reduced compared to control at 25 g l−1 and 50 g l−1, and so did alfalfa at 12.5 g l−1. However, as for radish (Raphanus sativus) there was no significant reduction in germination rate at any concentration under bioassay compared to control. Content of UV-B absorbing compounds and total phenols in Zanthoxylum seedlings responded positively to enhanced UV-B radiation. The results suggest that the allelopathic potential of Z. bungeanum was generally improved under enhanced UV-B radiation.  相似文献

Seed balls of ‘Early Wonder Tall Top’ table beet (Beta vulgaris L.) were incubated for 2 days at 21 °C in moist exfoliated grade 3 vermiculite (3 g seed balls [168 seed balls] and 1.1 g vermiculite) containing equal weights of Trichoderma harzianum Rifai strain KRL-AG2 G41 and T. virens strain G-41 (ThTv) at 0, 0.25, 0.50, 0.75 or 1.00 mg ThTv per seed ball. ThTv also was applied to the peat-lite growth medium 14 days before planting, at the same rates per seed ball as the seed ball treatments. Four days before planting, the peat-lite was inoculated with Pythium aphanidermatum (Edson) Fitzp. (Pa) at 0, 0.5 and 1.0× the rate that resulted in 96% damping-off when non-ThTv-treated dry seed balls were sown in peat-lite containing 1.0 Pa. Increasing ThTv level per seed ball decreased percentage damping-off, with 1.0 Pa giving greater percentage damping-off than 0.5 Pa. At 1 mg ThTv per seed ball, damping-off was 5% and 19% at 0.5 and 1.0 Pa, respectively. Including Agro-Lig UF (mostly humic acids) in the incubating seed-ball-ThTv mixture further decreased damping-off by an average 13 and 10 percentage points in 0.5 and 1.0 Pa, respectively. Increasing ThTv per seed ball in growth media decreased percentage damping-off, but not to the extent achieved with seed ball treatment, with 1 mg ThTv per seed ball giving 20% and 55% damping-off in 0.5 and 1.0 Pa, respectively. Decreasing incidence of damping-off with increasing ThTv application to seed balls or growth media was associated with increasing shoot fresh weight m−2 at 14 days after planting, a response attributable to increased percentage plant survival and not to a ThTv growth-promoting effect.  相似文献

Inter-section hybrids were obtained in the reciprocal crosses between Primula filchnerae (2n = 2x = 24) of Sect. Pinnatae and P. sinensis ‘Fanfare’ (2n = 2x = 24) of Sect. Auganthus by rescuing ovules on half-strength (1/2) Murashige and Skoog's (MS) medium supplemented with 50 g l−1 sucrose, 2.5 g l−1 gellan gum, 0.1 mg l−1 α-naphthaleneacetic acid (NAA), 0.1 mg l−1 6-benzyladenine (BA) and 50 mg l−1 gibberellic acid (GA3). In ovule culture, germination occurred with radicle elongation but no plumule was observed. The radicle kept on the initial medium showed root proliferation with callus formation. When the calluses were transferred to (1/2)MS media containing 30 g l−1 sucrose and 3 g l−1 gellan gum, without plant growth regulators (PGRs) or with 1 mg l−1 zeatin and 0.1 mg l−1 NAA, plantlets were regenerated. The plants thus obtained were confirmed to be hybrids through flow cytometry (FCM) and random amplified polymorphic DNA (RAPD) analyses. The hybrid obtained when P. filchnerae was used as the maternal parent was diploid, whereas hexaploid hybrid was obtained when using P. sinensis as the maternal parent. The hexaploid hybrid might be produced through chromosome doubling of a triploid originated from the fertilization of P. sinensis with unreduced pollen of P. filchnerae.  相似文献

Aqueous slurries of commercial preparations of Trichoderma harzianum Rifai strain KRL-AG2 G41 (Th), T. virens Strain G-41 (Tv), or their combination (ThTv, at half rates each of the single application rate) were applied to ‘Marktetmore 76’ cucumber seeds (Cucumis sativus L.) that were non-primed or primed for 3 days at 25 °C either osmotically (−2.5 MPa from 0.337 molal Ca(NO3)2) or osmomatrically (−1.0 MPa from 0.135 molal Ca(NO3)2 plus −1.5 MPa from 50% water in exfoliated grade 5 vermiculite). Slurries were applied to seeds (1 mg per seed) either before or after priming. Seeds were sown in soilless, peat-based media with or without inoculation with Pythium aphanidermatum (Pa). Protection against damping-off caused by high pressures of Pa (16% emergence in non-coated, non-primed seeds) was increased by slurry coating Th on non-primed (76.4% emergence) or on osmotically primed seeds, with coating either before or after priming having no effect on efficacy (average 62.6% emergence). Slurry coating Th on osmomatrically primed seeds failed to increase final emergence percentage (FEP). Colony forming units per three seeds (CFU, all 103) was 2.8 for non-primed seeds, and 3.2 and 2.6, respectively, when osmotically and osmomatrically primed seeds were coated after priming. In a second study with lower disease pressure (58.1 FEP from non-coated, non-primed seeds), slurry coating of non-primed or osmotically primed seeds with Th, Tv or ThTv reduced percentage damping-off and increased FEP. The combination coating eliminated damping-off only in non-primed seeds, and tended to reduce percentage damping-off in osmotically or osmomatrically primed seeds compared to coating with Th or Tv alone. In a third study using only non-primed seeds, slurry coatings with mefenoxam fungicide, Th, Tv, or ThTv decreased total damping-off to 2.6%, 7.4%, 2.0%, and 0%, respectively, from the 30.1% occurring in non-coated seeds. Th, Tv or ThTv applied to growth media at the same rate as the seed coating (1 mg per seed) were generally as effective as the seed coatings, and only the ThTv growth medium application eliminated damping-off. A fourth experiment revealed that Th, Tv or ThTv remained viable on non-primed seeds for up to 4 weeks (the longest storage duration) at 21 or 4 °C, but 21 °C storage resulted in faster seed germination by week 3 and higher CFU per three seeds by week 4. In summary, coating of non-primed seeds with Th, Tv or ThTv was more effective than coating primed seeds in reducing percentage damping-off. While priming treatments generally led to faster seedling emergence and greater seedling shoot fresh weight than was achieved with non-primed seeds, only for non-primed seeds was damping-off eliminated by the ThTv seed coating or growth medium application.  相似文献

An unique procedure for the mass shoot propagation of Gerbera using receptacle transverse thin cell layer (tTCL) culture procedure was developed. Genotype, flower bud age, explant size, position of receptacle tTCLs and culture media were found to affect the success of culture. Ten interspecific crosses of Gerbera showed different shoot regeneration rates and callus induction via receptacle tTCL culture, all of which had shoot regeneration rates higher than 57%. Flower buds collected on the 10th day resulted in 91% shoot regeneration after 6 weeks of culture on basal MS medium [Murashige, T., Skoog, F., 1962. A revised medium for rapid growth and bioassay with tobacco tissue cultures. Physiol. Plant. 15, 475–497] supplemented with 0.02 mg l−1 thidiazuron (TDZ), 0.8 mg l−1 adenine and 10% (v/v) coconut water (CW). This was significantly higher than those from flower buds on the 7th and 14th days (22% and 54%), respectively. Shoot regeneration rate was the highest (94–100%) in the middle layers of the receptacle. For mass shoot propagation, shoot clusters were subcultured on half-strength MS medium supplemented with 0.5 mg l−1 indole-3-butyric acid (IBA), 0.5 mg l−1 6-benzyladenine (BA) and 2.0 mg l−1 kinetin after every 4 weeks. Plantlets formed when single shoots were cultured on half-strength MS medium containing 1 mg l−1 IBA. All plantlets acclimatized well in the greenhouse.  相似文献

Very little information is available on organic specialty cut flower production, especially fertilization requirements. In order to better understand organic fertilization requirements of two specialty cut flower crops, Limonium sinuatum and Celosia argentea, we initiated a field and greenhouse experiment to study the effect of compost (organic) and conventional (inorganic) fertilization treatments on the growth and productivity of these crops. Optimum yields in the field, expressed as fresh weight per plot, were achieved at compost applications of 98.8 t ha−1 for both Limonium and Celosia. However, when number of stems, height of the stems, and the environmental impacts of such compost applications are considered we concluded that the optimal organic fertilizer amounts were 12.4 and 24.7 t ha−1, for Limonium and Celosia, respectively. Limonium and Celosia plants in the greenhouse experiment were fertilized with 100, 200, 300, and 400 mg L−1 nitrogen, combined with 0, 5, 10, 20, 40, and 60 mg L−1 phosphorus. The generation of response surfaces for total weight per pot, number of stems per pot, average weight per stem, and average stem length were attempted for each species. The results showed that nitrogen did not significantly contribute to any of the models, except for Celosia average weight and length per stem models. Total weights per pot on the other hand showed both a linear and quadratic relationship over the range of phosphorus applications we tested. Maximum number of stems and total weight per pot were observed between 30 and 46 mg L−1 P in both Limonium and Celosia. Our results suggest that organic fertilizer recommendations, in the form of animal manure composts should be based on phosphorus content of the compost rather than nitrogen content especially for soils high in initial phosphorous content.  相似文献

Magnetic fields (MFs) have been applied for the first time in orchid micropropagation. Protocorm-like bodies (PLBs) – approximately 3 mm in diameter – first derived from leaf segment culture of Phalaenopsis Gallant Beau ‘George Vazquez’, were subcultured every 2 months, and served as initial explants. The proliferation of Phalaenopsis PLBs in liquid medium in the Miracle Pack® culture system was affected by the action of different intensities and polarities of MFs: 0.1, 0.15 and 0.2 Tesla (T) at North (N) and South (S) poles. The MF of 0.1 T – S resulted in the greatest fresh weight of regenerated PLBs. The average number of neo-PLBs formed per clusters in the PLB treated by MF: 0.1–0.2 T was decreased compared to the control exposed to natural MF (5 × 10−6 T). The proliferation of PLBs under 0.15 T – MF at both N and S poles for 2 and 7 weeks demonstrated that a longer duration of exposure to an MF of 0.15 T, regardless of the polarity, resulted in greater biomass of newly formed PLBs and smaller average number of newly formed PLBs. The S pole of MF had stronger effects on Phalaenopsis PLBs proliferation than the N pole did in all treatments.  相似文献

In banana on-farm studies in the East African Highlands, quantification of production has been difficult because plants are at different stages of development at any given time. Production is continuous and quantification would therefore require permanent presence of an observer. Hence, most on-farm surveys have resorted to estimations from farmers or ‘experts’ through recall or visual qualitative observations. These methods are highly inaccurate. This study aims to develop and validate allometric relationships for quantitative estimation of bunch weights, based on rapid, inexpensive and non-destructive methods of data collection that take into consideration genotypic, spatial and developmental variability. The study was conducted in 179 farmer fields in central, south, southwest, and east Uganda. Bunch weights were estimated through linear regression with log-transformed girth of pseudostem at base and 1m, number of hands, and number of fingers in the lower row of the second lowest hand. The number of hands and fingers relate to the potential sink size (i.e. bunch), and the girth at base and 1m were used as a proxy for pseudostem volume, which relates to the potential of the plant to fill the sink. Bunch weight was significantly (P ≤ 0.001) and positively (R2 = 0.73) related to log-transformed pseudostem volume, and number of hands and fingers. When data were partitioned and regressed, regression coefficients did not differ significantly for cultivars (i.e. Enyeru, Kibuzi, Nakabululu and Nakitembe), developmental stages (flowering, early fruiting, late fruiting and full maturity), regions (central, south, southwest and east Uganda), foliar and soil N, P, K, Ca and Mg concentrations. Data partitioning improved the accuracy of prediction significantly for different cultivars, regions, and foliar Ca, but not for bunch developmental stages, foliar N, P, K and Mg and soil N, P, K, Ca and Mg concentrations. However, the residuals of all the regressions of partitioned data correlated highly (R2 between 0.92 and 1.00) to those of the regression based on pooled data. On validation of the regression based on pooled data, the bias (−9.97%) and modeling efficiency statistic (0.64), suggested that predictions were not always accurate. Still, the total predicted bunch weights were higher than the observed bunch weight by only 2%. This study therefore concludes that the regression derived using pooled data is suitable for on-farm prediction of bunch weight, for the East African Highland cooking banana, regardless of genotypic, developmental and spatial variations.  相似文献

Twelve commercially produced Dutch green waste composts were evaluated for their suitability to replace 20% (v/v) peat substrate in the cultivation of ornamentals. Salt concentrations were determined in water extracts of the composts and disease suppressive effects were assessed against various soilborne diseases. The Cl-concentration of the compost extract appeared to be the limiting factor for use of the composts in potting mixtures. The Cl-concentrations in 7 and 1 composts, respectively, were too high to replace 20% of peat for growing salt sensitive and moderately salt sensitive plants, according to guidelines set for these groups of plants. The suppressive effects of the composts were tested in peat-based potting mixtures using three bioassays: Phytophthora cinnamomi—lupin, Cylindrocladium spathiphylliSpathiphyllum and Rhizoctonia solani AG2-1—cauliflower. Disease levels in compost-amended mixtures were compared with the non-amended controls. None of the composts induced suppressiveness against P. cinnamomi; 3 and 9 composts significantly induced suppressiveness against C. spathiphylli and R. solani, respectively. No significant disease enhancement was observed in any of the bioassays. The pH of the potting mixture showed a negative correlation with suppression of the Rhizoctonia disease (R2 = 0.56). The effect of pH (pH 4–6) on suppression of R. solani and P. cinnamomi was further studied in non-amended peat. Disease suppression of R. solani in cauliflower decreased with increasing pH in two different kinds of peat, while there was no effect on P. cinnamomi. The suppressive effect of 3 composts was assessed in two experiments against Fusarium wilt in Cyclamen persicum (caused by F. oxysporum f. sp. cyclamini) and Begonia eliator (caused by F. foetens) under near-commercial conditions. None of the composts had a significant effect on Fusarium wilt in Cyclamen. Two and 3 composts significantly induced suppressiveness against Fusarium wilt in Begonia in the first and second experiments, respectively. No significant differences were observed in growth characteristics between Begonia plants grown in compost-amended and non-amended potting mixture in both experiments. In the second experiment, Cyclamen plants grown in compost-amended potting mixture had significant lower number of flowers than plants grown in non-amended potting mixture, which may have been due to lower concentrations of N in the compost-amended potting mixtures. In conclusion, most composts of the origin and composition tested can replace 20% peat in potting mixtures for moderately salt sensitive and salt tolerant plants. Amendment of these composts can contribute to control of Fusarium wilt in Begonia plants.  相似文献

The regenerability of three ornamental species—Lysimachia christinae, Lysimachia rubinervis and Lysimachia nummularia ‘Aurea’, were investigated using in vitro leaves and shoot tips. 6-Benzylaminopurine (BAP) and α-naphthalene acetic acid (NAA) added to Murashige and Skoog (MS) medium were tested for their effect on organogenesis. On the medium, shoot regeneration occurred directly without callus formation. In these species, L. christinae developed the highest regeneration rate and numbers of shoots/explant from shoot tips (100%, 12.25) and leaf bases (100%, 13.01) on the MS medium containing 3.0 mg l−1 BAP and 0.1 mg l−1 NAA. For L. rubinervis, the highest shoot induction rate and number of shoots/explant were obtained from shoot tip (100%, 16.87–17.20) on the MS medium with 0.1 mg l−1 NAA and 3.0–5.0 mg l−1 BAP. L. nummularia ‘Aurea’, however, showed the highest regeneration rate and number of shoots/explant (100%, 12.73) from leaf bases on MS medium supplemented with 1.0 mg l−1 BAP and 0.1 mg l−1 NAA. All in vitro shoots rooted well on half macronutrient MS medium containing 0.1 mg l−1 NAA. After acclimatization, transplanted plantlets grew normally and flowered in the field.  相似文献

Since cucumber plants are mostly discarded as large waste after crop harvesting, allelopathy of cucumber plants was investigated for possible weed management options and utilization of the waste. Two potent growth inhibitory substances were isolated from an aqueous methanol extract of cucumber (Cucumis sativus L. cv. Phung Tuong) plants. These substances were determined as 9-hydroxy-4,7-megastigmadien-9-one (HMO) and (6S,7E,9S)-6,9,10-trihydroxy-4,7-megastigmadien-3-one (THMO) by the analysis of MS, 1H NMR spectra and optical rotation. HMO inhibited the growth of cress (Lepidium sativum L.) and Echinochloa crus-galli (L.) Beauv seedlings at concentrations greater than 0.3 and 1 μM, respectively. THMO inhibited the growth of cress and E. crus-galli seedlings at concentrations greater than 1 and 3 μM, respectively. The concentrations required for 50% growth inhibition on roots and shoots of cress and E. crus-galli were 2.4–29.3 μM for HMO and 8.1–52.2 μM for THMO. The endogenous levels of HMO and THMO in cucumber plants were 31.8 and 43.5 μg g−1 dry weight, respectively. These results suggest that HMO and THMO may be the causal factors for the growth inhibitory effect of cucumber plants. Therefore, cucumber plants may be potentially useful for weed management options in an agricultural setting, such as a cover crop and soil admixture, which should be investigated further in the field.  相似文献

We assessed the effect of soil-applied derivatives of melia (Melia azedarach L.) and neem (Azadirachta indica A. Juss) on nitrogen (N) soil availability, root uptake and peach (Prunus persica L.) growth. First we evaluated the effectiveness of experimentally prepared amendments made with fresh ground melia leaves or commercial neem cake incorporated into the soil as nitrification inhibitors, then we evaluated the effect of fresh ground melia fruits and neem cake on growth and N root uptake of potted peach trees, and on soil microbial respiration. Soil-applied fresh ground melia leaves at 10 and 20 g kg−1 of soil as well as commercial neem cake (10 g kg−1) were ineffective in decreasing the level of mineral N after soil application of urea-N as a source of mineral N, rather they increased soil concentration of nitric N and ammonium N. The incorporation into the soil of fresh ground melia fruits (at 20 and 40 g kg−1) and neem cake (at 10 and 20 g kg−1) increased N concentration in leaves of GF677 peach × almond (Prunus amygdalus) hybrid rootstock alone or grafted with one-year-old variety Rome Star peach trees. An increase in microbial respiration, leaf green color and plant biomass compared to the control trees were also observed. The Meliaceae derivatives did not affect, in the short term (7 days), N root uptake efficiency, as demonstrated by the use of stable isotope 15N, rather they promoted in the long term an increase of soil N availability, N leaf concentration and plant growth.  相似文献

Most of the Japanese plum (Prunus salicina) cultivars grown in Israel produce relatively small fruit. Application of 2 l solution tree−1 of 25 mg l−1 2,4-dichlorophenoxypropionic acid (2,4-DP) as butoxyethyl ester (Power™), 15 mg l−1 3,5,6-trichloro-2-pyridyloxyacetic acid (3,5,6-TPA) as free acid (Maxim®), or 25 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) + 30 mg l−1 naphthaleneacetic acid (NAA) (0.3% Amigo™) at the beginning of pit-hardening, when fruitlet diameter was ca. 22 mm, caused an appreciable and significant increase in fruit size. The yield of large fruit per cv.: ‘Kesselmen’ (100% increase), ‘Songold’ (100%), ‘Black Diamond’ (800%), ‘Royal Diamond’ (160%) and ‘Royal Zee’ (100%). As a result, the total yield of all five cultivars was also increased dramatically. Anatomical studies with ‘Songold’ revealed that the main effect of these synthetic auxins was via direct stimulation of fruit cell enlargement. The above auxins had no negative effect either on fruit quality at harvest (and after 1 week in shelf-life), or on return yield in the following year.  相似文献

Aqueous (10, 20, 30 and 40 g/L) and organic (hexane, chloroform and methanol, at 3000 and 6000 ppm) extracts of Inula crithmoïdes L. (roots, stems, leaves and flowers) were evaluated for their allelopathic activities on radish, lettuce, peganum and thistle. Pot cultures were conducted by incorporation of I. crithmoïdes leaves and flowers powder (1.25 and 2.5 g/kg) in soil and by irrigation with their aqueous extracts at 40 g/L. PEG (polyethylene glycol) solutions, with similar osmotic potentials of aqueous extracts, at the highest concentration, were without effect on target species, this eliminates the extracts osmotic effect. Germination index was not affected by root and stem extracts and significantly decreased by the two other ones. For growth, leaf and flower extracts had the most significant toxicity at 40 g/L, inducing up to a total inhibition. For organic extracts, a strong toxicity was recorded for the three fractions at 6000 ppm, thistle seedling growth was more sensitive, especially to chloroform fractions. Soil incorporation of residues (at 2.5 g/kg) significantly decreased root (35–100%) and shoot (0–100%) length of target species. Irrigation soil with leaves and flower aqueous extracts was harmful for peganum and thistle, respective inhibition percentages were 100% and 66.5%. I. crithmoïdes may be favorably used for incorporating in agricultural systems for sustainable weed management.  相似文献

The present study reports that protoplasts isolated from stoloniferous shoots (SS) of potato represent an efficient system for somatic cell genetic manipulations. SS were established from single-node cuttings on MS medium supplemented with either 0.1 or 0.2 M sucrose (Suc), and protoplasts were isolated and cultured within the alginate strip, following an improved method. SS induced by 0.1 M Suc yielded 8–22 × 105 protoplasts g−1 fresh mass, with a high morphogenic competence. However, 0.2 M Suc-induced SS yielded protoplasts that contained large amounts of starch grains, resulting in their high degree of fragility, delayed cell division and poor morphogenic competence. For symmetric somatic hybridization (electrofusion) between Solanum tuberosum Gp. Tuberosum androgenic (di)haploid (2n = 2x = 24) ‘C-13’ and diploid (2n = 2x = 24) wild species S. pinnatisectum, protoplasts isolated from 0.1 M Suc-induced SS were also found to be most responsive. Out of several putative somatic hybrids, there were two tetraploids and five diploids, with 48 and 24 chromosomes, respectively at all the three shoot layers (L1–L3). This precluded the occurrence of mixoploidy vis-à-vis chimaerism in regenerants, as common in somatic fusion involving mesophyll protoplasts of S. pinnatisectum. Nuclear microsatellite analyses based on the two single-locus nSSR loci (STM0037 and STM2030) confirmed that one of the tetraploids was a true nuclear hybrid (heterokaryon), while the other a homokaryon of the Tuberosum parent ‘C-13’. The use of 0.2 M Suc-induced SS protoplasts for fundamental studies on tissue- and/or cell type-specific transient gene expression underlying tuberization has been discussed.  相似文献

Kosteletzkya virginica, a perennial dicot halophytic species of the Malvaceae, is native to American salt marsh. It was introduced into China as a potential species to improve coastal wetlands and to develop ecologically sound saline agriculture. K. virginica adapts excellently to the tidal-flat habitats in China's east coast, with multiple eco-benefits; in particular, its seed oil could be used to produce biodiesel. The purpose of this study was thus to develop a standardized protocol to induce a high frequency of callus and subsequent plantlet regeneration system for a K. virginica breeding program with the final objective of applying transgenic techniques to improve seed oil yield. The embryonic axes of K. virginica were used as explants for callus induction, shoot induction from the callus and then adventitious root induction from the shoots on nine culture media with different hormone combinations. The best results were achieved on the following media: (1) 93.94% callus induction on MS medium supplemented with 1.0 mg L−1 indole-3-acetic acid (IAA), 0.3 mg L−1 kinetin, 30 g L−1 sucrose and 8 g L−1 agar; (2) 65.83% shoot induction on 1/2MS medium supplemented with 0.1 mg L−1 IAA, 0.5 mg L−1 zeatin, 30 g L−1 sucrose and 8 g L−1 agar; (3) 96.67% rooting on MS medium containing 30 g L−1 sucrose and 8 g L−1 agar. The survival rate of plantlets by organogenic regeneration was 85% after being transplanted into potting soil in flowerpots and placed in the greenhouse. This experiment indicates that we established successful callus induction and plant regeneration protocols for K. virginica.  相似文献

The influence of IAA (1.0 mg dm−3), and IBA (1.16 mg dm−3), on the development of highbush blueberry (Vaccinium × covilleanum But. et Pl.) ‘Herbert’ in vitro shoot cultures was examined. Depending on the kind of auxin and 2iP concentration in vitro cultures consisted of various number of axillary (AX) and adventitious (AD) shoots. Three different categories of AD shoots were found: leaf shoots (AD-L), node-adjoin shoots (AD-P), and base-adjoin shoots (AD-M, madshoots). The AX shoots were the least habituated (towards auxins, cytokinins and vitamins) whereas the AD-M shoots (madshoots) the most. In comparison to IAA, IBA caused dying or callusing higher number of initial explants. However, IBA generally suppressed development of AD shoots, especially madshoots whereas slightly weakened multiplication of AX shoots. IBA significantly enhanced elongation of AX shoots also. Axillary shoots obtained on IBA-media had relative long internodes and rigid, well-developed leaves. The adventitious shoots, especially base-adjoin (AD-M) ones, were easily distinguishable as were more thin and fragile, more or less vitrified, and had short internodes and smaller, sometimes unfolded leaves. Development of blueberry in vitro cultures on auxin-free and IAA-supplemented media was similar. AX shoots grown on such media resembled AD shoots. 2iP applied in higher doses along with IAA promoted much proliferation of AD than AX shoots. In contrast, 2iP applied in higher doses together with IBA stimulated significantly only growth of AX shoots whereas in general, development of adventitious shoots was not affected. Micropropagation carried out through routine method based on subculturing of shoot explants or shoot clumps on the medium supplemented with IAA (4 mg dm−3) and 2iP (10–15 mg dm−3) as well as stimulation of shoot elongation on the blank medium causes in fact the propagation of highbush blueberry through highly habituated adventitious madshoots. Replacement of IAA by IBA facilitates micropropagation of highbush blueberry cv. Herbert through axillary shoots.  相似文献

Nutrient elements and lipoxygenase (LOX) activity were determined in strawberry fruit to establish a relationship, if it exists, between nutrient ratios (N/Ca and K/Ca), and lipoxygenase activity with albinism disorder. About 33% strawberry fruit were affected by albinism. Etna had highest incidence of albinism (48.6%) and Sweet Charlie the lowest (16.2%). Dry matter content (%) was lower in albino fruit (5.23%) than normal fruit (7.36%). The concentration of N, P, and Mg did not differ significantly, but that of K (1.87 mg g−1 fresh weight) was notably higher and of Ca (0.105 mg g−1 fresh weight) was lower in albino fruit than normal fruit. Consequently, the nutrient ratios, N/Ca (9.78) and K/Ca (16.96) were higher in albino fruit than normal fruit. Cultivars differed widely in respect to dry matter (%), mineral content and nutrient ratios. LOX activity determined on dry weight or fresh weight basis was significantly higher in albino fruit than normal fruit, with significant differences among cultivars. Positive correlations existed between nutrient ratios and albinism incidence (r = +0.338), LOX activity and albinism incidence (r = +0.412), and LOX and nutrient ratios (r = +0.448). Thus, it appears from the study that calcium and LOX activity may not the basic cause of albinism in strawberry, but these may be involved in senescence or fruit ripening process, as LOX activity was lower in albino than in normal fruit.  相似文献
