A group of non-azotaemic bull terriers from families with hereditary nephritis had significant subclinical renal disease. Of the renal function tests carried out, proteinuria, almost exclusively albumin, was a reliable and early indicator of glomerular abnormality. While 24-hour urinary protein excretion (24 UPE) in the bull terriers was significantly higher than in a group of normal dogs of other breeds, urinary protein excretion (UPE) was also significantly higher when measured by simple single sample tests such as the urinary protein to urinary creatinine ratio (UP/C). UPE in bull terriers was correlated with increasing age, in contrast to lack of correlation in the normal dogs. The degree of proteinuria in affected bull terriers from which renal biopsies were taken correlated with the severity of histopathological changes which mainly affected glomeruli. Light microscopic examination revealed segmentally thickened glomerular and tubular basement membranes, thickened Bowman's capsules and adhesions between glomerular capillaries and Bowman's capsules. Other renal function tests were performed but failed to detect subclinical disease. It is suggested that hereditary nephritis (HN) in bull terriers is similar to that seen in samoyeds, dober-mann pinschers and humans.  相似文献

Background:Genetic selection in cattle has been directed to increase milk production.This,coupled to the fact that the vast majority of bovine artificial inseminations(AI)are performed using cryopreserved sperm,have led to a reduction of fertility rates over the years.Thus,seeking sensitive and specific sperm biomarkers able to predict fertility rates is of vital importance to improve cattle reproductive efficiency.In humans,sperm chromatin condensation evaluated through chromomycin A3(CMA3)has recently been purported to be a powerful biomarker for sperm functional status and male infertility.The objectives of the present study were:a)to set up a flow cytometry method for simultaneously evaluating chromatin condensation and sperm viability,and b)to test whether this parameter could be used as a predictor of in vivo fertility in bulls.The study included pools of three independent cryopreserved ejaculates per bull from 25 Holstein males.Reproductive outcomes of each sire were determined by non-return rates,which were used to classify bulls into two groups(highly fertile and subfertile).Results:Chromatin condensation status of bovine sperm was evaluated through the combination of CMA3 and Yo-Pro-1 staining and flow cytometry.Sperm quality parameters(morphology,viability,total and progressive motility)were also assessed.Pearson correlation coefficients and ROC curves were calculated to assess their capacity to predict in vivo fertility.Sperm morphology,viability and total motility presented an area under the ROC curve(AUC)of 0.54,0.64 and 0.68,respectively(P>0.05),and thus were not able to discriminate between fertile and subfertile individuals.Alternatively,while the percentage of progressively motile sperm showed a significant predictive value,with an AUC of 0.73(P=0.05),CMA3/Yo-Pro-1 staining even depicted superior results for the prediction of in vivo fertility in bulls.Specifically,the percentage of viable sperm with poor chromatin condensation showed better accuracy and precision to predict in vivo fertility,with an AUC of 0.78(P=0.02).Conclusions:Chromatin condensation evaluated through CMA3/Yo-Pro-1 and flow cytometry is defined here as a more powerful tool than conventional sperm parameters to predict bull in vivo fertility,with a potential ability to maximising the efficiency of dairy breeding industry.  相似文献

Uric acid was used as a test for liver disease before the advent of enzymology. Three old studies criticised uric acid as a test of liver function. Uric acid, as an end-product of purine metabolism in the liver, deserved re-evaluation as a liver function test. Serum totalbile acids are widely accepted as the most reliable liver function test. This study compared the ability of serum uric acid concentration to assess liver function with that of serum pre-prandial bile acids in dogs. In addition, due to the renal excretion of uric acid the 2 assays were also compared in a renal disease group. Using a control group of healthy dogs, a group of dogs with congenital vascular liver disease, a group of dogs with non-vascular parenchymal liver diseases and a renal disease group, the ability of uric acid and pre-prandial bile acids was compared to detect reduced functional hepatic mass overall and in the vascular or parenchymal liver disease groups separately. Sensitivities, specificities and predictive value parameters were calculated for each test. The medians of uric acid concentration did not differ significantly between any of the groups, whereas pre-prandial bile acids medians were significantly higher in the liver disease groups compared with the normal and renal disease group of dogs. The sensitivity of uric acid in detecting liver disease overall was 65% while the specificity of uric acid in detecting liver disease overall was 59%. The sensitivity and specificity of uric acid in detecting congenital vascular liver disease was 68% and 59%, respectively. The sensitivity and specificity of uric acid in detecting parenchymal liver disease was 63% and 60%, respectively. The overall positive and negative predictive values for uric acid in detecting liver disease were poor and the data in this study indicated uric acid to be an unreliable test of liver function. In dogs suffering from renal compromise serum uric acid concentrations may increase into the abnormal range due to its renal route of excretion.  相似文献

Because the priority of AI industry is to identify subfertile bulls, a predictive model that allowed for the prediction of 91% bulls of low fertility was implemented based on seminological (motility) parameters and DNA status assessed both as DNA fragmentation index (DFI) and by TUNEL assay using sperm of 105 Holstein–Friesian bulls (four batches per bull) selected based on in vivo estimated relative conception rates (ERCR). Thereafter, sperm quality and male fertility traits of bulls were explored by GWAS using a high‐density (777K) Illumina chip. After data editing, 85 bulls and 591,988 SNPs were retained for GWAS. Of 12 SNPs with false discovery rate <0.2, four SNPs located on BTA28 and BTA18 were significantly associated (LD‐adjusted Bonferroni <0.05) with the non‐compensatory sperm parameters DFI and TUNEL. Other SNPs of interest for potential association with TUNEL were found on BTA3, in the same chromosome where associations with non‐compensatory in vivo bull fertility were already reported. Further suggestive SNPs for sperm membrane integrity were located on BTA28, the chromosome where QTL studies previously reported associations with sperm quality traits. Suggestive SNPs for ERCR were found on BTA18 in the vicinity of a site already associated with in vivo bull fertility. Additional SNPs associated with ERCR and sperm kinetic parameters were also identified. In contrast to other, but very few GWAS on fertility traits in bovine spermatozoa, which reported significant SNPs located on BTX, we have not identified SNPs of interest in this sexual chromosome.  相似文献

Single-sire natural mating data from a beef cattle herd in tropical Australia were used to estimate heritabilities of cow fertility (hc2), heritabilities of bull fertility (hb2) and genetic correlations between cow and bull fertility (rg) within each of six genotypes. Estimates of hc2 and hb2 were low, averaging .11 and .08, respectively. The pooled estimate of rg was 0.16, indicating that cow and bull fertility are favorably genetically correlated and therefore that cow fertility could be genetically improved by indirect selection on bull fertility, or some more heritable component of bull fertility.  相似文献

The biological nature of age‐related declines in fertility in males of any species, including stallions, has been elusive. In horses, the economic costs to the breeding industry are frequently extensive. Mitochondrial function in ejaculated sperm, which is essential for sperm motility, is reflected by adenosine triphosphate production, mitochondrial oxidative efficiency and production of reactive oxygen species, and that this balance may become compromised in ageing stallions and during the process of cryopreservation. This presentation will focus on mitochondrial integrity and function as an avenue for understanding the pathophysiology of sperm when undergoing cryopreservation and male ageing. We discuss the importance of understanding the differences and similarities of sperm mitochondria to that of somatic cells regarding structure and mitochondrial biochemistry relating to sperm function. The roles of oxidative phosphorylation and glycolysis in sperm mitochondria are outlined as is the method of determining oxygen consumption and calcium homoeostasis in sperm mitochondria. The effect of acrosomal aberrations of the spermatozoa of Finnish Ayrshire bulls on the corrected non-return rate within 60 days of the first 500 inseminations was studied. The material consisted of sperm samples examined by the artificial insemination societies. All samples had been accepted for use in artificial insemination. One Giemsa-stained slide was studied for each of the 95 bulls concerned. Samples showing distinct acrosomal defects were studied by electron microscopy. Three different types of acrosomal aberration were found. One was obviously associated with subfertility in all 6 bulls in which it was detected.  相似文献

Despite several inherent characteristics that would appear to make hair a useful biopsy tissue, many problems make interpretation of data derived from hair analysis difficult. Endogenous minerals are incorporated into hair by several routes. Most attention on hair mineral incorporation has focused on element uptake within the hair follicle. Minerals incorporated within the follicle are presumably chemically or physically associated with cortical cells of the hair shaft and reflect mineral status at the time that the hair filament was synthesized. Because hair follicles in most species regularly go through cycles of intense metabolic activity and quiescence, mineral incorporation through the follicle is not a continual process. Mineral deposition onto hair does not cease when the follicle is not producing a hair fiber. The hair shaft is continuously exposed to several elements through contact with secretions from sebaceous, apocrine and eccrine glands. Significant quantities of major and trace elements of endogenous origin are adsorbed onto the hair surface via these secretions, especially when hair growth is not occurring or is very slow. For several elements significant correlations exist between mineral concentrations in hair and mineral intake. These correlations, however, are usually quite low. Non-dietary factors such as sex, age, hair color, sire, body location and contamination affect mineral levels in hair. Dietary intake of calcium, phosphorus and iron are also known to affect uptake of other elements in hair. Because many factors cause variation in mineral content in hair, hair analyses are not precise indicators of mineral status. Hair mineral analyses may be useful, however, when combined with other indicators of mineral status to provide a more precise assessment of mineral status in livestock.  相似文献
