Isoflavones in soybean were extracted in the crude form using 80% food-grade ethanol at 80 degrees C for 6 h and followed by concentration and dehydration. The soy extract contained isoflavones primarily in the forms of glucosides. In vitro antioxidant activities of the soy extract containing 20-500 ppm isoflavones were conducted using a Rancimat method. The results showed that soy isoflavone extract had strong in vitro antioxidant activity. There was a dose-dependent response for the in vitro antioxidant activity at the lower concentrations but not at the higher concentrations. In vivo antioxidant property was determined by measuring the antioxidant enzymes, superoxide dismutase, and catalase in various organs of rats that were fed with diets containing partially oxidized oil and various levels of isoflavones for up to 24 weeks. Neither short-term (8 weeks) feeding nor low isoflavone content (50 ppm) induced changes in superoxide dismutase or catalase activities in rats. Only diets containing high isoflavone contents (150 and 250 ppm) showed obvious elevated enzymatic levels in various organs. In addition, a laboratory-prepared tofu containing approximately 50 ppm isoflavones had better effects than the soy extract with the 250 ppm isoflavone group, which indicated that molecules other than isoflavones may have a synergistic effect on in vivo antioxidant enzyme inductions of tofu.  相似文献

Free radicals are considered to be important causative factors in the development of cancer and cardiovascular diseases. This relationship has led to interest in evaluating the antioxidant capacities of many dietary supplements. Fermented soybean extract is produced by symbiotic fermentation of organic soybean with 20 types of Lactobacillus and yeast. In vitro and in vivo models are used in this study to evaluate the antioxidant effect of fermented soybean extract. Several in vitro models are used to detect the antioxidant capacity of the fermented soybean extract, which is compared to vitamin C and Trolox. The results demonstrate that the fermented soybean extract has strong antioxidant activity against unsaturated fatty acid peroxidation compared to vitamin C and Trolox. By the means of the test system developed by Y. Toshiki et al., it is shown that the fermented soybean extract can function both as an antioxidant and as a free radical acceptor that can convert free radicals into harmless substances through an energy-decreasing procedure. An in vivo study examines the effects of fermented soybean extract on the activity of antioxidant enzymes. The activities of the antioxidant enzymes (AOE) including total superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPX) are measured in liver, kidney, and brain from male Sprague-Dawley rats. The activities of CAT, SOD, and GPX are increased in the liver. However, the SOD activity is decreased in the kidney. SOD and GPX activities are decreased in the brain. These results lead to the conclusion that fermented soybean extract not only has antioxidant activity but also has an effect on the activity of antioxidant enzymes in liver.  相似文献

研究大豆油体乳液的基本物理化学性质,将为其工业应用提供参考。以水为介质提取大豆油体,方法无毒,利于食用。对其在不同pH值(pH值2~8)、NaCl浓度(0~250?mmol/L)和加热处理(30~90℃,30?min)条件下的Zeta电位、平均粒径和乳析稳定性进行测定,并对其流变性进行考察。大豆油体乳液的Zeta电位为+20?mV~-40?mV(pH值2~8),等电点约为4.5。在pH值≤3和pH值≥6条件下,平均粒径均为0.4?μm左右;而在3<pH<6时,产生了乳析现象。在较高NaCl浓度下(>25?mmol/L),粒径较大和发生了乳析现象。大豆油体乳液在30~90℃加热处理时较稳定。大豆油体乳液呈现出弱凝胶的性质,其黏度随着油质量分数的降低而降低。研究表明,大豆油体乳液在一定的环境条件下是稳定的。  相似文献

Wines contain phenolic compounds that may be useful for preventing lipid oxidation as dietary antioxidants. This study was aimed at evaluating the antioxidant activity in corn oil emulsions of seventeen selected Spanish wines and two California wines. The inhibition of hydroperoxide formation at 10 microM gallic acid equivalents (GAE) varied from 8.4% to 40.2% with the red wines, from 20.9% to 45.8% with the rosé wines, and from 6.5% to 47.0% with the white wines. The inhibition of hydroperoxide formation at 20 microM GAE varied from 11.9% to 34.1% with the red wines, from 0.1% to 34. 5% with the rosé wines, and from 3.3% to 37.2% with the white wines. The inhibition of hexanal formation at 10 microM GAE varied from 23. 6% to 64.4% with the red wines, from 42.7% to 68.5% with the rosé wines, and from 28.4% to 68.8% with the white wines. The inhibition of hexanal formation at 20 microM GAE varied from 33.0% to 46.3% with the red wines, from 11.3% to 66.5% with the rosé wines, and from -16.7% to +21.0% with the white wines. The antioxidant effect decreased with increasing concentration. This antioxidant activity was related to the five main groups of phenolic compounds identified in wines by HPLC. The relative antioxidant activity correlated positively with the total phenol content of wines (by the Folin-Ciocalteu method and by HPLC), benzoic acids, anthocyanins, flavan-3-ols, and flavonols, for the inhibition of hydroperoxides and hexanal at 10 and 20 microM GAE.  相似文献

Brassica vegetables and glucosinolates contained therein are supposed to reduce the risk of cancer and to possess health-promoting properties. The benefits of a Brassica-based diet may be particularly expressed by eating sprouts, in which the glucosinolate content is higher than in mature vegetables. With this in mind, a first objective of this study was to evaluate the antioxidant properties of radish (Raphanus sativus L.) sprouts (Kaiware Daikon) extract (KDE), in which the glucosinolate glucoraphasatin (GRH), showing some antioxidant activity, is present at 10.5% w/w. The contribution of GRH to KDE's antioxidant activity was considered in two chemical assays (Trolox equivalent antioxidant capacity and Briggs-Rauscher methods). The total phenol assay by Folin-Ciocalteu reagent was performed to quantify the reducing capacity of KDE. Finally, on the basis of the putative choleretic properties of antioxidant plant extracts, the effect on the bile flow of KDE administration was investigated in an animal experimental model. The findings showed that KDE has antioxidant properties and significantly induced bile flow in rats administered 1.5 g/kg of body weight for 4 consecutive days.  相似文献

The antioxidant activity of 3-dehydroshikimic acid (DHS), an intermediate in the biosynthesis of aromatic amino acids, was evaluated in three assay systems: bulk oil (lard), liposomes, and a 10% corn oil-in-water emulsion. Upon initiation of peroxidation in the liposome or emulsion systems, DHS exhibited weak antioxidant activity. In contrast, DHS displayed strong antioxidant activity in lard, suppressing peroxidation with activity comparable to that of tert-butylhydroquinone, propyl gallate, and gallic acid and superior to that of alpha-tocopherol. Two major DHS oxidation products, gallic acid and protocatechuic acid, were identified by gas chromatography/mass spectral analysis of lard extracts; both compounds are effective antioxidants in the bulk oil system. In the liposome system, DHS remained intact throughout the assay period. A small amount of gallic acid was observed in extracts of the emulsion; however, protocatechuic acid was not detected. A mechanism to explain the different activities of DHS in the three lipid systems is proposed.  相似文献

Algae oil-in-water emulsions stabilized with 0.2% whey protein isolate (WPI) at pH 3.0 and 7.0 were chosen to evaluate antioxidant activity of a proanthocyanidin-rich extract from grape seed. In this emulsion system, (+)-catechin and ascorbic acid (620 microM) were found to be prooxidative at pH 3.0 and ineffective at pH 7.0. Grape seed extract was not able to effectively inhibit both lipid hydroperoxides and propanal formation when added to the emulsion at 124 microM. However, increasing the concentration of the grape seed extract to 620 microM resulted in inhibition of both lipid hydroperoxide and propanal formation at pH 3.0 and 7.0. None of the antioxidants tested had any effect on the physical stability of the WPI-stabilized emulsion. The superior antioxidant activity of the grape seed extract is likely due to the presence of oligomeric procyanidins which are better antioxidants compared to their monomeric counterparts.  相似文献

The genus Hibiscus thrives in a variety of climates and produces a diversity of natural compounds with bioactive properties. We have studied the chemical composition and the in vivo antioxidant properties of Hibiscus tiliaceus L. methanolic flower extract, as well as its mutagenic/antimutagenic effects. Vitamin E and some stigmasterol derivatives that might confer an antioxidant effect to the extract were present. Treatment with this extract protected several Saccharomyces cerevisiae strains defective in antioxidant defenses against H2O2 and t-BOOH cytotoxicities, showing a clear antioxidant activity. The effect is the same for all strains used, independent of the antioxidant defense disrupted, suggesting that protection may be due to molecules that act as versatile and wide spectrum nonenzymatic antioxidants, such as vitamins or phytosterols. The extract was not mutagenic in either Salmonella typhimurium or S. cerevisiae and showed a significant antimutagenic action against oxidative mutagens in S. cerevisiae.  相似文献

Fruits of the date palm (Phoenix dactylifera L. Arecaceae) are very commonly consumed in many parts of the world and are a vital component of the diet in most of the Arabian countries. This preliminary study documents for the first time its antioxidant and antimutagenic properties in vitro. There was a dose-dependent inhibition of superoxide and hydroxyl radicals by an aqueous extract of date fruit. The amount of fresh extract required to scavenge 50% of superoxide radicals was equivalent to 0.8 mg/mL of date fruit in the riboflavin photoreduction method. An extract of 2.2 mg/mL of date fruit was needed for 50% hydroxyl-radical-scavenging activity in the deoxyribose degradation method. Concentrations of 1.5 and 4.0 mg/mL completely inhibited superoxide and hydroxyl radicals, respectively. Aqueous date extract was also found to inhibit significantly the lipid peroxidation and protein oxidation in a dose-dependent manner. In an Fe(2+)/ascorbate system, an extract of 1.9 mg/mL of date fruit was needed for 50% inhibition of lipid peroxides. In a time course inhibition study of lipid peroxide, at a 2.0 mg/mL concentration of date extract, there was a complete inhibition of TBARS formation in the early stages of the incubation period that increased during later stages of the incubation. Similarly, in the high Fe(2+)/ascorbate induction system a concentration of 2.3 mg/mL inhibited carbonyl formation measured by DNPH reaction by 50%. Moreover, a concentration of 4.0 mg/mL completely inhibited lipid peroxide and protein carbonyl formation. Date fruit extract also produced a dose-dependent inhibition of benzo(a)pyrene-induced mutagenecity on Salmonella tester strains TA-98 and TA-100 with metabolic activation. Extract from 3.6 mg/plate and 4.3 mg/plate was found required for 50% inhibition of His+ revertant formation in TA-98 and TA-100, respectively. These results indicate that antioxidant and antimutagenic activity in date fruit is quite potent and implicates the presence of compounds with potent free-radical-scavenging activity.  相似文献

The antioxidant activity of oregano, parsley, olive mill wastewaters (OMWW), Trolox, and ethylenediaminetetraacetic acid (EDTA) was evaluated in bulk oils and oil-in-water (o/w) emulsions enriched with 5% tuna oil by monitoring the formation of hydroperoxides, hexanal, and t-t-2,4-heptadienal in samples stored at 37 degrees C for 14 days. In bulk oil, the order of antioxidant activity was, in decreasing order (p < 0.05), OMWW > oregano > parsley > EDTA > Trolox. The antioxidant activity in o/w emulsion followed the same order except that EDTA was as efficient an antioxidant as OMWW. In addition, the total phenolic content, the radical scavenging properties, the reducing capacity, and the iron chelating activity of OMWW, parsley, and oregano extracts were determined by the Folin-Ciocalteau, oxygen radical absorbance capacity, ferric reducing antioxidant power, and iron(II) chelating activity assays, respectively. The antioxidant activity of OMWW, parsley, and oregano in food systems was related to their total phenolic content and radical scavenging capacity but not to their ability to chelate iron in vitro. OMWW was identified as a promising source of antioxidants to retard lipid oxidation in fish oil-enriched food products.  相似文献

Antioxidant properties of prepared blueberry (Vaccinium myrtillus) extracts   总被引:3,自引:0,他引:3  
A blueberry extract (A) and two anthocyanin-derived extracts (B and C) were prepared. The contents of polyphenols, flavonoids, anthocyanins, and anthocyanin-derived pigments of the extracts were determined for the first time. The pigment profile of blueberry extract A corresponded to 15 anthocyanins, whereas extract B was mainly composed of anthocyanin-pyruvic acid adducts of the blueberry original anthocyanins and extract C was mainly composed of the respective vinylpyranoanthocyanin-catechins (portisins). The extracts' abilities to inhibit lipid peroxidation, induced by 2,2'-azobis(2-methyl-propanimidamide) dihydrochloride in a liposomal membrane system were examined. The antioxidant capacities of the extracts were evaluated through monitoring oxygen consumption and by measuring the formation of conjugated dienes. All of the extracts provided protection of membranes against peroxyl radicals by increasing the induction time of oxidation. This effect increased with the polyphenol content and with the structural complexity of the anthocyanin-derived pigments of the extracts. The pigments present in extract C seemed to induce a higher protection of the liposome membranes toward oxidation. In addition, the antiradical properties and the reducing power of the extracts were determined by using DPPH and FRAP methods, respectively. The results from these assays were in agreement with those obtained with the liposome membranes.  相似文献

The leaves and fine stems, bark, and trunk wood oils of Aniba canelilla showed yields ranging from 0.2 to 1.3%. The main volatile constituent identified in the oils was 1-nitro -2-phenylethane (70.2-92.1%), as expected. The mean of DPPH radical scavenging activity (EC 50) of the oils (198.17 +/- 1.95 microg mL(-1)) was low in comparison with that of wood methanol extracts (4.41 +/- 0.12 microg mL(-1)), the value of which was equivalent to that of Trolox (4.67 +/- 0.35 microg mL(-1)), used as antioxidant standard. The mean amount of total phenolics (TP) (710.53 +/- 23.16 mg of GAE/g) and this value calculated as Trolox equivalent antioxidant capacity (TEAC) (899.50 +/- 6.50 mg of TE/g) of the wood methanol extracts confirmed the high antioxidant activity of the species. On the other hand, in the brine shrimp bioassay the values of lethal concentration (LC50) for the oils (21.61 +/- 1.21 microg mL(-1)) and 1-nitro-2-phenylethane (20.37 +/- 0.99 microg mL(-1)) were lower than that of the wood methanol extracts (91.38 +/- 7.20 microg mL(-1)), showing significant biological activities.  相似文献

Phenolic compounds were extracted from Morton lentils using acidified aqueous acetone. The crude Morton extract (CME) was applied onto a macroresin column and desorbed by aqueous methanol to obtain a semipurified Morton extract (SPME). The SPME was further fractionated over a Sephadex LH-20 column into five main fractions (I-V). The phytochemical contents such as total phenolic content (TPC), total flavonoid content (TFC), and condensed tannin content (CTC) of the CME, SPME, and its fractions were examined by colorimetric methods. Antioxidant activity of extracts and fractions were screened by DPPH scavenging activity, Trolox equivalent antioxidant capacity (TEAC), ferric reduced antioxidant power (FRAP), and oxygen radical absorbing capacity (ORAC) methods. In addition, the compositions of active fractions were determined by HPLC-DAD and HPLC-MS methods. Results showed that the fraction enriched in condensed tannins (fraction V) exhibited significantly higher values of TPC, CTC, and antioxidant activity as compared to the crude extract, SPME, and low molecular weight fractions (I-IV). Eighteen compounds existed in those fractions, and 17 were tentatively identified by UV and MS spectra. HPLC-MS analysis revealed fraction II contained mainly kaempferol glycoside, fractions III and IV mainly contained flavonoid glycosides, and fraction V was composed of condensed tannins. The results suggested that the extract of Morton lentils is a promising source of antioxidant phenolics and may be used as a dietary supplement for health promotion.  相似文献

Interest in replacing synthetic antioxidants, namely, butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA), with natural antioxidants is increasing. The present study examined the antioxidant activity of capsaicinoid from chili pepper in heated canola oil. The oxidation was conducted at 60, 90, 120, and 180 °C by monitoring oxygen consumption and the decrease in linoleic acid and α-linolenic acid in canola oil. At 60 °C, capsaicinoid was more effective against oxidation of canola oil compared with BHT. At higher temperatures of 90, 120, and 180 °C, capsaicinoid possessed an antioxidant activity similar to or slightly weaker that that of BHT. It was found that capsaicinoid prevented canola oil from oxidation in a dose-dependent manner. To study the structure-antioxidant relationship, it was found that the trimethylsiloxy (TMS) derivatives of capsaicinoid did not exhibit any antioxidant activity, suggesting the hydroxyl moiety was the functional group responsible for the antioxidant activity of capsaicinoid. It was concluded that capsaicinoid had the potential to be further explored as a natural antioxidant in foods, particularly spicy foods.  相似文献

Electron paramagnetic resonance (EPR) and densitometry were used to measure the temperature- and rate-dependent formation of fat crystals in emulsion droplets in hardened palm kernel oil in water emulsions. The solid fat content in emulsions can be critical for the functionality of the emulsions in a wide variety of applications. Therefore, new and accessible methods are needed to monitor solid fat content in order to control the functional properties of these emulsions. EPR measurements showed that the microviscosity within the oil droplets could be measured as a function of temperature and that the storage temperature below the main fat melting point is crucial for an increase in the microviscosity, due to fat solidification. The microviscosity of the oil droplets could be an important parameter for defining functional performance (e.g., rheology and partial coalescence). Density measurements provided a simple and accurate method for monitoring changes in phase state of the oil. The phase behavior agreed well with the EPR results, demonstrating that accurate density measurements could prove to be a valuable tool for monitoring fat crystallization processes.  相似文献

该研究应用生命周期评价方法,以大豆油和地沟油分别制备1 t生物柴油为研究对象,计算生物柴油全生命周期过程中的能源消耗和周期排放,结果表明:以大豆油为原料制备生物柴油全生命周期总能耗约为地沟油的2.65倍,且以地沟油为原料制备生物柴油过程中CO2、SO2、NOx、CO和粉尘各项排放与大豆油为原料时相比分别降低了82.92%、45.68%、94.91%、53.40%和90.61%。通过对制备生物柴油生命周期排放的废气和废物对环境造成的影响进行量化分析,结果表明以大豆油为原料时生命周期环境影响潜值约为地沟油的11.70倍,其数值分别为8.42和0.72,大豆油制备生物柴油过程中对环境的影响主要是全球性的变暖,地沟油制备生物柴油过程中对环境的影响主要是地区性的酸化。  相似文献

The influence of guar gum and xanthan gum on the creaming of 10% oil in water emulsions has been investigated. The presence of very low concentrations of the polysaccharides (typically < approximately 0.075%) was found to induce depletion flocculation of the emulsion droplets and increased the rate of creaming. However, at higher polysaccharide concentrations (typically > approximately 0.1%) the creaming rate was reduced due to the increased viscosity of the aqueous phase. For most systems a delay period was noted before creaming started, which was found to be dependent on the zero shear viscosity of the continuous phase and independent of polysaccharide type. The delay period increased significantly at zero shear viscosities approaching 1 Pa s. A mathematical model has been used to fit the creaming rate profiles and a simple exponential relationship obtained between delay time and polysaccharide concentration.  相似文献

Chain-breaking antioxidants differ in their effectiveness at inhibiting lipid oxidation because of their chemical properties and physical location within a food. Our objective was how the physicochemical properties of four structurally related lipid-soluble antioxidants were related to their antioxidant activity. Antioxidants differed in the number of methyl (alpha-tocopherol and delta-tocopherol) or hydroxyl (butylated hydroxytoluene (BHT) and 4-hydroxymethyl-2,6-ditertiarybutylphenol) groups. Surface activity of the antioxidants was in the order of delta-tocopherol > alpha-tocopherol approximately 4-hydroxymethyl-2,6-ditertiarybutylphenol > BHT. Free-radical scavenging activity was similar between alpha-tocopherol and delta-tocopherol as well as BHT and 4-hydroxymethyl-2,6-ditertiarybutylphenol. In bulk menhaden oil, BHT was a more effective antioxidant than 4-hydroxymethyl-2,6-ditertiarybutylphenol while alpha-tocopherol was more effective than delta-tocopherol. In menhaden oil-in-water emulsions, BHT was a more effective antioxidant than 4-hydroxymethyl-2,6-ditertiarybutylphenol while delta-tocopherol was more effective than alpha-tocopherol. These results indicate that the surface activity and polarity of lipid-soluble antioxidants were not the only determinants of their antioxidant effectiveness in food lipids.  相似文献

The influence of EDTA on lipid oxidation in sugar beet pectin-stabilized oil-in-water emulsions (pH 6, 15% oil, wet basis), prepared from fish oil (FO) and fish oil-extra virgin olive oil (FO-EVOO) (1:1 w/w), as well as the spray-dried microcapsules (50% oil, dry basis) prepared from these emulsions, was investigated. Under accelerated conditions (80 °C, 5 bar oxygen pressure) the oxidative stability was significantly (P < 0.05) higher for FO and FO-EVOO formulated with EDTA, in comparison to corresponding emulsions and spray-dried microcapsules formulated without EDTA. The EDTA effect was greater in emulsions than in spray-dried microcapsules, with the greatest protective effect obtained in FO-EVOO emulsions. EDTA enhanced the oxidative stability of the spray-dried microcapsules during ambient storage (~25 °C, a(w) = 0.5), as demonstrated by their lower concentration of headspace volatile oxidation products, propanal and hexanal. These results show that the addition of EDTA is an effective strategy to maximize the oxidative stability of both FO emulsions and spray-dried microcapsules in which sugar beet pectin is used as the encapsulant material.  相似文献

An environmentally friendly water-based pathway to form the azide derivatives of soybean oil and fatty esters is reported. This entails first the formation of epoxides and then the azidization of the epoxides. The azidization reaction is carried out at high yields in water with only a small amount of an ionic liquid as a catalyst. The distribution of azide and alcohol functionalities on the fatty acid moiety is approximately random. This reaction has been applied to methyl oleate, methyl linoleate, soybean oil, and methyl soyate. The resulting structures have been studied by NMR.  相似文献
