建立一种新型藏羊血红素的提取方法。以藏羊血为原料,丙酮和酸性丙酮为提取溶剂,所得粗提物进行纯化后,采用红外光谱法对血红素进行定性分析,并用紫外分光光度法进行定量测定。通过进行单因素试验,可确定丙酮添加量为血液体积1倍、离心时间为15min是制备藏羊血红素的最优条件,该法制备血红素成本低,操作过程简便,生产周期短,纯度可达98.60%,且收率达到(87.91±9.04)%。 相似文献

试验以新鲜牦牛血液为原料,采用血粉法、冰醋酸法及醋酸钠法,探索制备氯化血红素的最优工艺参数,并对不同方法制备的氯化血红素纯度及得量进行对比。结果显示,血粉法的最优工艺是超声处理时间40 min,酸性丙酮添加量为120 mL,抽提时间30 min,乙酸钠添加量为9 mL,氯化血红素纯度达到96%,得量为7.38 g/L。采用冰醋酸法制备氯化血红素,当冰醋酸添加量为红细胞量的3倍时,氯化血红素纯度达到80%,得量为6.10 g/L。采用醋酸钠法制备氯化血红素,当氯仿添加量为红细胞量的2倍时,氯化血红素纯度达到90%,得量为3.44 g/L。通过对比3种制备方法,氯化血红素纯度依次为血粉法 > 醋酸钠法 > 冰醋酸法;氯化血红素得量依次为血粉法 > 冰醋酸法 > 醋酸钠法,由此可知,3种方法中,选用血粉法制备氯化血红素的量更高且更加有利于工业化生产。 相似文献

向猪血中加入抗凝剂,将血浆和血细胞分离,从血浆中提取生物活性物质.首先用硫酸铵以"三阶段盐析法"分步从血浆中提取粗纤维连接蛋白(FN)、粗免疫球蛋白和血浆杂蛋白;然后用丙酮和醋酸钠等从血细胞中提取血红素,并用氯仿、吡啶和冰醋酸等进行有机萃取纯化;最后再从溶血过滤所得的初级残渣中提取超氧化物歧化酶(SOD),并用丙酮等进行有机萃取纯化.结果表明,从1 000 mL猪血中可提取粗FN、免疫球蛋白、血浆杂蛋白、血红素和SOD分别为100 mg、5.0 g,33.0 g,10.2 g和1 945.3 U,并证明免疫球蛋白具有与抗生素同样的抑菌作用. 相似文献

以白三叶(Trifolium repens)为材料探讨了叶蛋白的提取工艺和纯化方法。对白三叶叶蛋白提取中加热时间、温度、pH值、料液比、酸的种类等单因素以及纯化试剂甲醇、乙醇、丙酮、四氯化碳、水等进行了研究。结果表明,各单因素最佳参数为加热时间9 min,温度90 ℃,pH值 4.0,料液比1∶2,沉淀的酸为硝酸。对加热时间、加热温度、pH值和料液比进行4因素3水平正交试验,发现影响叶蛋白提取因素为pH值>温度>时间>料液比,最佳提取工艺为加热时间9 min,温度80 ℃,pH值4.0,料液比1∶2。对叶蛋白纯化结果表明,纯化剂对叶蛋白纯度的影响依次为甲醇>乙醇>丙酮>四氯化碳>水,但各种纯化剂之间差异不显著(P>0.05)。 相似文献

为改进和优化金花菜(Medicago polymorpha)叶蛋白提取的工艺环节,并提高叶蛋白提取率及提取物纯度等参数指标,本研究选择长江下游农区广泛种植的金花菜为原料,通过设置较为合理科学的料液比、提取温度、加热时间、pH以及提取剂(酸)种类进行叶蛋白提取单因素试验。结果表明:各单因素试验最佳参数分别为料液比1∶3、提取温度70℃、加热时间15 min、pH 4、提取剂(酸)为盐酸。基于清洁安全生产角度,选择了单因素试验中提取指标同样较高的柠檬酸作为提取剂,设置了料液比、pH及提取温度的3因素3水平响应面试验,发现影响叶蛋白提取率的因素依次为pH、提取温度、料液比。得到最佳提取工艺:料液比1∶3、pH 4.1、提取温度72℃,相应的叶蛋白提取率为49.63%,提取物纯度为56.87%。本研究优化的技术工艺及相关参数指标可为农区豆科草类植物多元化利用及叶蛋白工业化生产提供一定的技术支撑和相关借鉴。 相似文献

On a farm where the ration of cows contained 88.0 g of butyric acid, an experimental group of cows (n = 8), producing milk containing 7.9 mg or more acetone per litre, was studied for 14 days for the effect of a 250 g supplement of sodium acetate to the ration (combined with single i.m. administration of vitamins A, D2 and E) on selected metabolism parameters and on milk production. As distinct from the control group of cows (n = 8) from the same farm which produced milk containing 3.9 mg or less acetone per litre and which were fed without sodium acetone supplements, a tendency of increased alkaemia of the organism was suggested in the experimental cows. This tendency manifested itself during the trial in increased pH values, increased base excess (BE) and standard bicarbonate (SB) in the blood, and in an increase in the pH value and net acido basic secretion in urine. A decrease was recorded in the concentration of the acetone + acetacetic acid sum, the same as beta-hydroxybutyric acid in blood and the sum of acetone and acetacetic acid in milk (P less than 0.01). An insignificant increase of the activity of gammaglutamyl transpeptidase (GMT) was recorded in the blood serum of the experimental cows and a significant increase occurred in the content of potassium (up to P less than 0.01) and urea (up to P less than 0.01) in urine. The supplement of sodium acetate to the feed ration did not influence the degree of ketonuria and the finding of urobilinogen in urine.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献
