Resistance to V. inaequalis, derived from the small-fruited species Malus floribunda 821, is determined by a major dominant gene, assigned as Vf. The material used in this paper is based on the introgression of the Vf gene from M. floribunda into commercial apple varieties. Comparing RAPD patterns of a genomic DNA sample of M. floribunda with a pooled DNA sample of resistant individuals and that of 10 susceptible commercial apple varieties, fragments were identified which are derived from M. floribunda. One of them, the fragment OPD20/600, proved to be linked to the Vf gene with a recombination value of about 0.20—0.25. It is the first DNA marker so far for scab resistance.  相似文献

Summary In studies of the inheritance of resistance, pea seedlings of seven lines in which stems and leaves were both resistant to Mycosphaerella pinodes were crossed with a line in which they were both susceptible. With seven of the crosses resistance was dominant to susceptibility. When F2 progenies of five crosses were inoculated on either stems or leaves independently, phenotypes segregated in a ratio of 3 resistant: 1 susceptible indicating that a single dominant gene controlled resistance. F2 progenies of one other cross gave ratios with a better fit to 9 resistant: 7 susceptible indicating that two co-dominant genes controlled resistance. The F2 progeny of another cross segregated in complex ratios indicating multigene resistance.When resistant lines JI 97 and JI 1089 were crossed with a susceptible line and leaves and stems of each F2 plant were inoculated, resistance phenotypes segregated independently demonstrating that leaf and stem resistance were controlled by different genes. In two experiments where the F2 progeny of the cross JI 97×JI 1089 were tested for stem and leaf resistance separately, both characters segregated in a ratio of 15 resistant:1 susceptible indicating that these two resistant lines contain two non-allelic genes for stem resistance (designated Rmp1 and Rmp2) and two for leaf resistance (designated Rmp3 and Rmp4). Evidence that the gene for leaf resistance in JI 1089 is located in linkage group 4 of Pisum sativum is presented.  相似文献

Genetic markers are a much faster and more practical alternative to classical methods for the identification of genes for scab resistance present in different apple cultivars. In our study, 28 scab-resistant cultivars, four wild sources of the resistance genes and 10 susceptible cultivars were screened for the presence of the RAPD fragments OPM18/900, OPD20/600 and OPA15/900, which are reported to be linked to the Vf gene. All three marker fragments were successfully amplified with different protocols in Vf-resistant cultivars including ‘M. floribunda 821’. No marker fragments were amplified in susceptible cultivars, three out of four Va-resistant cultivars, three out of four Vm-resistant cultivars, two Vr-resistant cultivars, ‘Antonovka PI 172612’ and ‘M. pumila R 12740-7A’. All three markers were found in the cv. ‘Nova Easygro’, reported to possess the Vr gene, and the cv. ‘Reglindis’, reported to be Va-resistant. M. atrosanguinea of unknown origin showed the presence of OPD20/600 and OPA 15/900 marker bands. The cvs. ‘Nova Easygro’, ‘Reglindis’ and M. atrosanguinea are probably carriers of the VF gene.  相似文献

Russian wheat aphid (RWA), Diuraphis noxia (Kurdjumov), is a serious pest of small grains in many countries. A previous study screened 70 genotypes, collected from different parts of Iran, for RWA resistance. Four crosses were made between two resistant lines (Shz.W-102 and Shz.W-104) and two susceptible lines (Shz.W-101 and Shz.W-103). Parents, F1, F2, and BCF1 seedlings were screened for RWA resistance in the greenhouse by artificial infection. To determine allelism, the two resistant lines were intercrossed and F1, and F2 seedlings were evaluated. Resistance in Shz.W-102 and Shz.W-104, when crossed with Shz.W-101, was controlled by one dominant gene. However, resistance in Shz.W-102 and Shz.W-104, when crossed with Shz.W-103, was controlled by two dominant genes. Genes in two resistant lines segregated independently of each other. A three-gene system was proposed to govern resistance in the lines under study . This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

Cucurbita pepo (pumpkin, squash,gourd) is an economically important species that is susceptible to the cucurbit powdery mildew fungus, Podosphaera xanthii(syn. Sphaerotheca fuliginea). ‘True French’, an open-pollinated cultivar of the Zucchini Group of C. pepo, was crossed with an unnamed powdery-mildew resistant straight neck-type accession, the resistance of which was apparently derived from an interspecific cross with a resistant wild species of Cucurbita,and resistant plants were selected in the F2 generation. This was followed by six cycles of backcross-pedigree selection for resistance, and resulted in the development of an accession true-breeding for resistance to powdery mildew and nearly isogenic to ‘True French’. The resistant and susceptible near-isogenics were crossed and seeds of the filial and backcross generations were produced. Plants of the parental accessions and their progenies were grown together in a controlled-environment chamber, exposed to the pathogenic fungus, and scored as resistant, partially resistant, or susceptible 27–33 days after sowing. The results indicated that resistance is conferred by a single incompletely dominant gene, designated Pm-0. Summary Seven wild diploid potato species, Series Tuberosa, representing 1023 clones were screened for resistance to the potato cyst nematode, Globodera pallida. Over 25% of the clones were resistant to pathotype P4A and almost 30% were resistant to pathotype P5A. The resistance in hybrid progenies of these and other resistant species with cultivated potatoes was evaluated, and over 2200 seedlings were screened. High frequencies of resistance (>50%) to P4A were found in progenies with Solanum leptophyes, S. vernei, S. gourlayi and S. capsicibaccatum, whereas resistance to P5A was found in these species as well as S. sparsipilum. The importance of nematode resistant wild species for potato breeding is discussed.  相似文献

Summary Dihaploids were produced from tetraploids resistant to potato cyst nematode (Globodera pallida (Stone)). High levels of resistance were found in the dihaploids and three were used to produce tetraploid progenies by crossing them with susceptible tetraploid cultivars. One dihaploid, PDH505, produced more highly resistant offspring than the other two, PDHs 417 and 418. The latter gave progenies whose levels of resistance were similar to those obtained from susceptible dihaploids crossed with resistant tetraploids.The differences between the progenies of the resistant dihaploids were probably due to different modes of unreduced gamete formation (PDH505 producing gametes by first division restitution (FDR) and PDHs 417 and 418 by second division restitution (SDR)) although cytological studies would be necessary to confirm this. The methods by which dihaploids could be utilised in a tetraploid potato breeding programme are discussed in relation to the mode of unreduced gamete formation.  相似文献

Summary Evaluation of Phaseolus vulgaris germplasm bank materials and progenies from a large number of crosses using red- or yellow-colored, BCMV-susceptible bean lines, crossed to purple- or grey/brown-colored, hypersensitive-resistant lines, suggested strong trait association between seed color and BCMV resistance. The cross of red-mottled I+I+ (susceptible) BAT 1255R to isogenic purple-mottled II (resistant) BAT 1255M was made to study the segregation of the two characters and to recover red-mottled resistant progenies. Summary The use of soil. naturally infested with Heterodera avenae, to select resistant heterozygotes in backcross progenies of wheat, was tested for reliability. Selfed progenies from plants selected as resistant were cultured monoxenically in test tubes with nematodes hatched from single cysts, while backcross progenies from the same parent plants were grown in pots of naturally infested soil. Cyst counts were made after two months' growth. The results showed that over 50% of the backcross lines, screened in previous generations with naturally infested soil, had been erroneously selected as resistant. The test tube cultures clearly differentiated lines carrying resistance from those which were susceptible and corroborated results from pot tests.  相似文献

A test to select Fusarium resistant seedlings of the Asiatic hybrid lily is described. Young seedlings of 28 populations, obtained from an incomplete diallel between eight parents with different levels of Fusarium resistance, were tested for resistance. Significant differences in Fusarium resistance between and within populations were detected. The average percentage of selected seedlings ranged from 34% in resistant × resistant crosses to 2% in susceptible × susceptible crosses. Although resistant descendants were obtained in susceptible × susceptible crosses, using at least one resistant parent produced higher percentages of resistant seedlings. The resistance level of the parents correlated highly with the general combining ability for Fusarium resistance based on the seedling test. For eight populations, seedlings selected for Fusarium resistance and non-tested (control) seedlings of the same cross were compared, after propagation, in a clonal test. Variation between and within populations, found at seedling level, was confirmed at clonal level. A positive selection response was found for all eight populations. In the seedling test, approximately 18% of the seedlings were selected as resistant of which 15% (2.7% of seedlings tested) appeared to be susceptible escapes. Comparison between selection at seedling level and at clonal level indicated that approximately 25% of the seedlings tested were missed (rejected resistant plants) in the seedling test. The practical use of a seedling test for Fusarium resistance in lily breeding programs is discussed.  相似文献

Summary Diploid potato clones selected for their reaction to common scab and their ability to produce 2n male gametes were used in a series of crosses to a susceptible tetraploid female parent (cv. Shepody). In addition, two tetraploid clones were also selected for their reaction to common scab and crossed with Shepody as a female parent. Results indicated that resistance to common scab can be effectively transmitted from the diploid to the tetraploid level via 4x-2x crosses. Diploid parents producing 2n pollen via either first division or second division restitution can be used to transmit scab resistance. A relatively small proportion of resistant individuals could be recovered from susceptible x susceptible crosses in both 4x-2x and 4x-4x combinations. The data support a previously developed hypothesis that scab resistance is relatively simply inherited.  相似文献

Summary Three triticale lines, Siskiyou, M2A-Beagle, and OK 77842 have been reported to possess resistance to bacterial leaf streak caused by Xanthomonas campestris, pv. translucens (Xct.). The three resistant lines were crossed to susceptible lines and crossed with each other. F2, BC1-F1, BC2-F1 plants were inoculated with a mixture of two Xct strains. The segregation data indicate the presence of a single dominant gene in each of the three resistant lines to bacterial leaf streak. Rice blast (Magnaporthe oryzae) is the most destructive and epidemic disease of rice. Use of host resistance is the best alternative for disease management. The leaf and neck blast resistance of 182 rice breeding lines were assessed for leaf and neck blast and classified relative to a susceptible check-Masuli and resistant check-Laxmi, from greenhouse experiment in 2005 and 2006. The test plants were inoculated with 105 conidial suspension/ml of M. oryzae at 21 days old seedlings for the leaf blast, and at neck base for the neck blast. Among them, for leaf blast, 77 rice lines were resistant, 43 were moderately resistant, 39 were moderately susceptible and 23 were susceptible. While among the selected 31 rice lines evaluated for neck blast, 4 lines were resistant, 4 were moderately resistant, 16 were moderately susceptible and 7 were susceptible. Leaf and neck infection was significant and positively correlated (r = 0.30, P = 0.05). The rice lines, Barkhe 1032, Barkhe 1034, Barkhe 1035, Barkhe 1036, Barkhe 2014, Barkhe 2024, Barkhe 3019, Super 3004 were resistant to leaf blast and Barkhe 1006, Barkhe 1032, Barkhe 1035, Barkhe 3004 were resistant to neck blast. The rice lines with identification # 11, 69, 137, 168, 182 from Masuli × MT4 parentage, and Barkhe 3017 were susceptible to both leaf and neck blast. Progenies of Irradiated Pusa Basmati (IPB), Kalinga III/IR64 (KIII/IR64), and Masuli/IR64 were resistant to both leaf and neck blast. However, most progenies from Masuli/MT4 showed susceptible reaction to both leaf and neck blast. Thus, rice lines form the IPB, KIII/IR64 and Masuli/IR64 will be promising resistant sources for rice blast in breeding programme.  相似文献

Summary A genomic remnant of Malus floribunda clone 821 introgressed into the cultivated apple M. x domestica Borkh. was identified using randomly amplified polymorphic DNA (RAPD) markers obtained by the polymerase chain reaction (PCR). Using a set of 59 oligonucleotide decamer primers, polymorphic DNA markers were identified among three pooled DNA samples. Based on the presence or absence of bands among bulked apple scab-resistant selections and cultivars, bulked scab-susceptible cultivars, and a M. floribunda clone 821 sample, one primer, A 15, identified amplified fragments in the scab-resistant bulked sample that was also unique to the M. floribunda clone 821. The unique band from M. floribunda clone 821 was amplified in four out of 17 scab-resistant selections/cultivars. This RAPD, designated OA15900, identifies an introgressed fragment that has as yet no known function.  相似文献

Summary Lycopersicon pimpenellifolium L3707, resistant to the late blight oomycete Phytophthora infestans was crossed with the susceptible Lycopersicon pimpenellifolium 14377 or the susceptible Lycopersicon esculentum ZH. Progeny F1 and F2 generations were scored at the 5-leaf stage for resistance against 175 field and recombinant isolates of the pathogen. F1 plants exhibited various levels of moderate resistance and F2 plants segregated 3:6:7 resistant/moderately resistant/susceptible. The data support the hypothesis that race-non-specific resistance in L3707 is controlled by two independent genes: a partially-dominant gene and a dominant epistatic gene.  相似文献

Summary Seedlings of date palm (Phoenix dactylifera L.), obtained from seven cultivars crossed with two males, were analyzed by polyacrylamide gel electrophoresis for esterase (EST), glutamate oxaloacetate transaminase (GOT), endopeptidase (ENP) and alcohol dehydrogenase (ADH) polymorphisms. Eleven, eight, five and two phenotypes were revealed for the enzymes tested, respectively. Seedlings of F1 populations derived from Bayoud (Fusarium)-resistant and low fruit quality cultivars were characterized by a high electrophoretic polymorphism, when compared with progenies of Bayoud-susceptible and high fruit quality cultivars. In almost all cases, the most frequent electrophoretic phenotypes scored for each enzyme in different F1 populations, were similar to those of the corresponding parent cultivars. Heterozygous phenotypes have been found for, at least, 3 loci; Got-2, Est-1 and Enp, indicating that such loci could be used to screen for hybrid seedlings. The expected Mendelian segregation of allozymes has been observed for these 3 loci, in many F1 populations. It seems that progenies of Bayoud-resistant cultivars are characterized by a high level of electrophoretic polymorphism. The estimation of this index and the search for genetic linkage with segregating allozymes, may be biochemical criteria useful as an aid in distinguishing date palm seedling populations derived from Bayoud-resistant cultivars and suitable for breeding programs.  相似文献

Summary The isoenzymic geneGOT-1 was used as an indicator of ploidy levels and hybridity in the progeny of a 4x × 2x (A878/5,GOT-1 abdn × ‘Gloster 69’,GOT-lcc) cross, derived from the apomictic species (Malus hupehensis), which segregated for mildew resistance. The distinction between resistant and susceptible plants was sharp. After 3 months in a greenhouse, resistant plants showed no signs of sporulation. Of the hybrid seedlings (diploid and triploid) 78% were mildew resistant. This analysis was facilitated through the use of theGOT-lc allele, as a marker of hybridity. It appears that the prospects of transferring high mildew resistance fromM. hupehensis to the cultivated apple are promising; of the 65 hybrid resistant seedlings 18 were diploid. A similar approach in earlier generations might have facilitated the earlier transfer of this resistance to the cultivated apple.  相似文献

Summary A total of 280 globe artichoke (Cynara scolymus L.) clones were assessed for reaction to naturally occurring artichoke latent virus (ALV) in field trials. Also, the reaction of 17 seed progenies of crosses which produced ALV-resistant clones in the field, 20 self progenies of parental genotypes common to these crosses, self progenies of 17 artichoke cultivars, and 7 accessions ofC. cardunculus andC. syriaca were evaluated in the glasshouse following mechanical inoculation with ALV. The local and systemic presence of ALV, and the relative virus content in infected plants were determined by ELISA.All cultivars and most of the otherC. scolymus material tested (clones and seed progenies) proved to be highly susceptible to ALV. Ten progenies were less susceptible to mechanical inoculation as shown by the moment the virus could be detected, and a slow rate of accumulation of viral antigen. Some plants in two seed progenies possessed apparently a high resistance as could be concluded from the failure of ALV to spread or to multiply in plant.Accessions ofC. cardunculus andC. syriaca were also highly susceptible to ALV. However, an interspecific hybridC. cardunculus x C. syriaca presented the highest level of resistance, the plants remained free of local and systemic infection. Ratios of resistant to susceptible plants in self progenies of resistant plants suggested that this resistance was dominant.  相似文献

An interspecific cross was made to transfer leaf rust and stripe rust resistance from an accession of Aegilops ovata (UUMM) to susceptible Triticum aestivum (AABBDD) cv. WL711. The F1was backcrossed to the recurrent wheat parent, and after two to three backcrosses and selfing, rust resistant progenies were selected. The C-banding study in a uniformly leaf rust and stripe rust resistant derivative showed a substitution of the 5M chromosome of Ae. ovata for 5D of wheat. Analysis of rust resistant derivatives with mapped wheat microsatellite makers confirmed the substitution of 5M for 5D. Some of these derivatives also possessed one or more of the three alien translocations involving 1BL, 2AL and 5BS wheat chromosomes which could not be detected through C-banding. A translocation involving 5DSof wheat and the substituted chromosome 5M of Ae. ovata was also observed in one of the derivatives. Susceptibility of this derivative to leaf rust showed that the leaf rust resistance gene(s) is/are located on short arm of 5M chromosome of Ae. ovata. Though the Ae. ovatasegment translocated to 1BL and 2AL did not seem to possess any rust resistance gene, the alien segment translocated to 5BS may also possess gene(s) for rust resistance. The study demonstrated the usefulness of microsatellite markers in characterisation of interspecific derivatives. Summary Common and durum wheat populations obtained from Sweden and originally collected in Ethiopia were screened for resistance to steum rust and leaf rust. Resistant selections of common wheat were crossed and backcrossed with either stem rust susceptible RL6071, or leaf rust susceptible Thatcher. Genetic studies, based largely on tests of backcross F2 families, showed that four of the selections had in common a recessive gene SrA. Plants with this gene were resistant (1+ infection type) to all stem rust races tested. This gene was neither Sr26 nor Sr29. The resistance of other selections, based on tests with an array of rust isolates, was due to various combinations of Sr6, 8a, 9a, 9d, 9c, 11, 13, 30, and 36. One of the selections had linked genes, Lr19/Sr25. Another selection had a dominant gene for resistance (;1 infection type) to all the races of leaf rust. With the possible exception of this gene for leaf rust resistance and SrA, no obviously new resistance was found.  相似文献
