This study was conducted to evaluate the effect of feeding ensiled alfalfa with waste date on ruminal fermentation characteristics, microbial protein synthesis, protozoa population and blood parameters in sheep. Eight rams were used in a 2 × 2 change over design. Each experimental period consisted of 21 days including 16 days for adaptation and 5 days for sampling. For ensiling, fresh alfalfa (Medicago sativa L.) with different levels of waste date (Phoenix dactylifera L.) were mixed together and ensiled in 100‐l containers for 45 days. Chemical composition of silages such as dry matter (DM), crude protein, NH3‐N, organic matter, NDF, ADF and pH were determined. Then, it was used as 30% (DM basis) in diets. The experimental diets were as follows: (i) control (diet containing alfalfa silage without waste date), (ii) diet containing alfalfa silage with 5 g waste date/95 g DM, (iii) diet containing alfalfa silage with 10 g waste date/90 g DM, and (iv) diet containing alfalfa silage with 15 g waste date/85 g DM. The results of this experiment showed that adding waste date to alfalfa during ensiling, improved silage quality, DM and energy level. Total protozoa population and all of holotrich, cellulolytic and entodinia in rumen fluid were increased linearly by increasing the level of waste date. Nitrogen (N) intake, urinary N excretion and nitrogen retention were affected by dietary treatments. Also, allantoin, uric acid, total purine derivatives, microbial protein synthesis, cholesterol level and blood urea nitrogen were significantly different. In conclusion, direct ensilage of alfalfa can be attained by mixing 15 g waste date/85 g DM and positive associative effects such as increased metabolizable energy (ME) and silage quality occurred.  相似文献

This study was conducted to evaluate the effects of increasing levels of wasted date (WD) in sheep (initial body weight 50 ± 2.3 kg) diets on apparent diet digestibility, rumen fermentation and microbial protein synthesis. The experimental diets contained 0 (0 WD), 9% (9 WD), 18% (18 WD) or 27% (27 WD) of wasted dates. These diets were evaluated through a 4 × 4 Latin square design with four 28‐day periods. Sheep were individually housed in metabolic cages and were fed diets ad libitum. To keep the diets isoenergetic and isonitrogenous, increasing partial replacement of WD for barley grain was used with increasing levels of soya bean meal. Increasing the amount of WD in diets of sheep did not influence (p > 0.05) diet intake and digestibility of nutrients. Inclusion of WD in diets of sheep did not affect (p > 0.05) ruminal pH but decreased (p < 0.05) NH3‐N concentration and blood urea nitrogen. Also there was no effect (p > 0.05) of the WD provision on N intake and faecal N loss of experimental animals, but urinary nitrogen excretion and retained nitrogen were affected (p < 0.05) by dietary treatments. Purine derivatives and microbial protein synthesis increased (p < 0.05) linearly with the level of WD in the diet. In conclusion, the increase in microbial protein synthesis and concomitant reduction in ruminal NH3‐N concentration and blood urea nitrogen in response to wasted date supply suggest improved efficiency of ruminal nitrogen metabolism without adverse effects on fermentation characteristics and animal performance.  相似文献

The objective of the study was to evaluate effects of Quillaja saponaria (QS) with or without Yucca schidigera extract (YS) on in vitro ruminal fermentation and methanogenesis. The culture media consisted of 400 mL of strained rumen fluid collected from two non‐lactating Holstein cows and 400 mL of artificial saliva. The culture media was anaerobically incubated with 10 g of a mixture of concentrate and oat hay (1:1, w/w) at 39°C for 24 h. The treatments were arranged as 2 × 3 factorial design experiment consisting of three levels of QS (0, 2 or 4 mL/L) and two levels of YS (0 or 2 mL/L). Treatment interactions between QS and YS were observed for pH and ammonia N concentration. CH4 production was not modified by addition of QS but decreased (P < 0.05) when added with YS. Addition of QS with YS increased (P < 0.05) propionate concentrations. Protozoa numbers were decreased (P < 0.05) by QS addition alone or with YS. Results show that QS and YS exhibited strong antiprotozoal effects and combinations of both plant extracts may have potential as safe manipulators of ruminal fermentation.  相似文献

In a replicated 3 x 3 Latin square experiment, six heifers (443 +/- 6.1 kg) fed a 61% barley grain:39% alfalfa silage diet (DM basis) were given intraruminal doses of powdered Yucca schidigera (YS). Doses of 0 (control), 20, or 60 g/d were given at 0800 daily. Ruminal content was sampled 0, 2, 4, and 6 h after dosing. Acidity, concentrations of reducing sugars, free amino acids, and peptides in the rumen were not affected (P > .05) by YS. Relative to control, ruminal ammonia concentration was reduced (P < .05) 2 h after YS dosing. Ruminal propionate concentration was increased (P < .05) by YS. Protozoal numbers in the rumen were lower (P < .05) with YS than without. Yucca did not affect (P > .05) rate or extent of in situ DM degradability. Fibrolytic, amylolytic, and proteolytic activities in ruminal contents were similar among treatments (P > .05). Dry matter intake, apparent digestibilities of DM, NDF, and CP, nitrogen balance, and microbial protein synthesis in the rumen were not affected (P > .05) by treatment. The effect of YS on ruminal ammonia concentration likely resulted from a decreased concentration of protozoa and, presumably, from ammonia binding by YS. The effect on ruminal propionate was probably a result of a selective inhibitory effect of YS on rumen microbial species.  相似文献

In this study, the effect of Fusarium toxin-contaminated triticale (FUS) at high (60%) and low (30%) concentrate proportion in ruminant rations on ruminal fermentation, microbial protein synthesis and digestibility was investigated, using in vivo and in vitro methods. Significant effects of the forage-to-concentrate ratio on ruminal degradation and digestibility of crude nutrients and detergent fibre fractions as well as on the pH value and the concentration of short chain fatty acids (SCFA) in rumen fluid were found. The production of SCFA was affected, and the degradation of crude fibre and neutral detergent fibre in the rumen was reduced by the inclusion of FUS at high concentrate proportion. The efficiency of microbial crude protein synthesis was higher in diets with 60% than in diets with 30% concentrates, but was impaired in the presence of FUS in vitro at the high concentrate level. Marginal effects of FUS on the amino acid pattern of microbial protein were detected. It was concluded that the use of FUS in high concentrate diets can influence ruminal fermentation and microbial protein synthesis at a dietary deoxynivalenol concentration below 5 mg/kg dry matter.  相似文献

Two experiments were conducted to study the effect of supplemental 100 g/day of live Bacillus cultures (2 × 1011 cell of Bacillus subtilis and Bacillus licheniformis) on rumen fermentation as well as milk yield and composition in Chinese Holstein cows. In experiment 1, investigating 3 × 10 cows, milk yield and milk protein were increased by using B. licheniformis (p < 0.05) in comparison with an unsupplemented group and the B. subtilis group. Body weight was not significantly affected by Bacillus culture supplementation (p > 0.05). Percentage of milk fat and lactose was not significantly different between treatments (p > 0.05). But milk protein increased with B. licheniformis supplementation (p < 0.05). In experiment 2, carried out with three non‐lactating ruminally and duodenally fistulated cows, results showed that B. licheniformis supplementation increased microbial crude protein flow into duodenum (p < 0.05) and decreased the ammonia nitrogen concentration in ruminal fluid at 0.5 h, 1 h, 3 h, 6 h after morning feeding (p < 0.05). Bacillus licheniformis supplementation increased total VFA and acetate concentration in ruminal fluid at 0.5 h, 1 h, 3 h, 6 h after morning feeding (p < 0.05). Bacillus subtilis had no significant effect on rumen fermentation characteristics, duodenal microbial N flow and ruminal apparent nutrient digestibility (p > 0.05). Bacillus licheniformis increased ruminal apparent nutrient digestibility of neutral detergent fibre, acid detergent fibre, and organic matter (p < 0.05).  相似文献

通过体外产气法研究了生石膏对瘤胃发酵、甲烷生成及微生态的影响。按单因子试验设计,在底物中分别按其干物质的0(对照组)、1.25%(低水平组)、2.5%(高水平组)添加生石膏,每个组设4个重复,进行24h体外发酵培养。结果显示,随生石膏添加水平升高,乙酸与总挥发性脂肪酸浓度、乙酸与丙酸的浓度比、12和24h产气量、氢的生成量及甲烷菌数量显著(P0.05)或极显著(P0.01)降低,而丙酸占总挥发性脂肪酸的比例与原虫数量极显著(P0.01)升高;生石膏添加会显著(P0.05)或显著(P0.01)降低12和24h甲烷产量,而高水平添加会显著(P0.05)降低丙酸浓度与乙酸占总挥发性脂肪酸的比例。结果表明,生石膏可通过减少氢的供应量和抑制甲烷菌生长而降低体外甲烷产量,同时会促进原虫增殖,致使2种添加水平的相对甲烷抑制潜势没有显著(P0.05)差异。  相似文献

以4只安装永久性瘤胃瘘管的白山羊作为采集瘤胃液的供体,用体外法研究了4种不同蛋白质来源(玉米-豆粕型、玉米-豆粕-棉粕型、玉米-豆粕-菜粕型、玉米-棉粕-菜粕型)的日粮对不同培养时间的发酵底物的干物质和蛋白质消化率、产气量、瘤胃pH值和NH3-N质量浓度的影响。结果表明:经24 h人工瘤胃培养后,干物质和蛋白质的消化率由高到低顺序依次为:玉米-豆粕型>玉米-豆粕-棉粕型>玉米-豆粕-菜粕型>玉米-棉粕-菜粕型。发酵底物产气量由高到低依次为:玉米-豆粕-菜粕型>玉米-豆粕-棉粕型>玉米-豆粕型>玉米-棉粕-菜粕型。瘤胃NH3-N质量浓度由高到低依次为:玉米-豆粕-棉粕型>玉米-豆粕-菜粕型>玉米-豆粕型>玉米-棉粕-菜粕型。 各组的瘤胃pH值基本一致,处于正常瘤胃生理范围。  相似文献

本试验研究日粮硝酸盐的添加量对湖羊瘤胃还原硝态氮程度和对发酵参数及血液高铁血红蛋白的影响。试验选取6头湖羊,采用随机区组试验设计,每期分别饲喂含0%,1%,2%,3%,4%,5%硝酸钾的日粮。测定喂后0,0.5,1.0,1.5,2.0,4.0,6.0,8.0 h瘤胃液硝酸盐和亚硝酸盐含量及瘤胃发酵参数。晨饲后2.0 h通过颈静脉采血,测定高铁血红蛋白含量。结果表明,湖羊在采食1.0 h后硝酸盐含量达到最高(0.952.14 g/L),之后快速消失,亚硝酸盐含量在喂后1.5 h达到最高(0.936.22 μmol/L),之后降低。高铁血红蛋白的含量随硝酸盐添加量的增加而显著升高(P<0.01),最高达0.32%。添加硝酸盐显著提高瘤胃液pH值(P<0.01)和2.0 h的氨态氮含量(P<0.01)以及乙酸/丙酸,添加量高于3%会降低微生物蛋白和总挥发性脂肪酸的浓度,2%的硝酸盐添加量微生物蛋白和总挥发性脂肪酸的合成量最高,最有利于微生物发酵。  相似文献

本试验的目的是确定饲粮中青贮银合欢添加水平对奶牛瘤胃微生物种群、氮平衡和微生物蛋白合成的影响。试验采用4×4拉丁方设计,选择12头初始体重为163±16 kg、带有瘤胃瘘管的奶牛,随机分为4个组,每组3个重复,每个重复1头牛。基础日粮以100%的水稻秸秆为原料,处理组分别用30%、60%和100%的青贮银合欢替代水稻秸秆,奶牛自由采食水稻秸秆和青贮银合欢,每天按照体重的0.2%补充浓缩料。结果表明,用60%青贮银合欢替代水稻秸秆组奶牛瘤胃微生物群,尤其是纤维素分解菌、蛋白分解菌和厌氧真菌的数量显著增加(P<0.05),各组对淀粉分解菌群无显著影响(P>0.05)。原生动物种群数量随日粮青贮银合欢添加水平的增加呈线性下降(P<0.05)(P<0.05)。此外,氮平衡和微生物蛋白合成均随着日粮青贮银合欢添加水平的升高而升高(P<0.05),其中以60%青贮银合欢组最高。结论:在本试验基础上,用60%青贮银合欢替代水稻秸秆可以显著提高奶牛体内的微生物数量和微生物蛋白的合成。  相似文献

试验旨在研究不同水平的粗蛋白(CP)及瘤胃非降解蛋白(RUP)的日粮对奶牛瘤胃代谢的影响。试验选择18头健康、体重相近的荷斯坦奶牛作为试验动物,随机分为6组,每组3头。采用2×3因子完全随机试验设计,其中CP设7.5%(低)、9%(中)1、0.5%(高)三个水平,RUP设45%CP(低)、55%CP(高)两个水平,共6个处理组,饲喂相同基础日粮,精粗比为3:7,预饲期10 d,饲喂后2、4、8、12、24 h从食道采集瘤胃液2 d,测定瘤胃液pH值,NH3-N、微生物蛋白质(BCP)、挥发性脂肪酸(VFA)含量。结果表明:日粮CP水平对瘤胃液氨氮浓度的影响极显著(P<0.01),对BCP及VFA的含量影响较小,总体而言,9%CP水平组对奶牛的瘤胃发酵略好于其他两组;日粮中高RUP(55%CP)可以显著降低瘤胃pH值且有降低NH3-N浓度的趋势,同时,对瘤胃BCP及VFA的合成有一定的促进作用。本试验条件下,日粮中CP(9%)和RUP(55%CP)可在一定程度上改善瘤胃代谢。  相似文献

对肥育期的去势中国荷斯坦奶牛饲喂TMR和精、粗分离饲料后,试验组瘤胃内的乳酸浓度显著低于对照组(P<0.05)。试验组采食后6小时时的pH值及采食后3小时时的NH3N浓度显著高于对照组(P<0.05)。乙酸和丙酸在两处理组间无显著差异,而在0小时和6小时时试验组丁酸极显著高于对照组(P<0.01),同时采食后9 h内试验组平均丁酸浓度极显著高于对照组(P<0.01)。另外,从采食后9 h内试验组平均乙酸/丙酸极显著高于对照组(P<0.01)。CMCase的活力在两处理组间无显著差异,而采食后9 h内试验组平均xylanase活力显著高于对照组(P<0.05)。总细菌数和总真菌数在试验组中表现出比对照组增加的倾向,特别是总细菌数在6小时时,总真菌数在6小时和9小时时,试验组显著高于对照组(P<0.05)。试验组粗蛋白质的消化率极显著高于对照组(P<0.01)。  相似文献

Methane emissions from ruminants enhance global warming and lead to a loss of feed energy. The emissions are low when fed brassica crops, but the factors contributing to low emissions are unknown. A meta-analysis was conducted with individual animal data collected from seven experiments. In these experiments, methane emissions were measured using respiration chambers. Animal characteristics, feed chemical composition and rumen fermentation parameters were included for the analysis using multiple regression models. Feed intake level, animal live weight and age were animal factors that were weakly and negatively related to methane yield (g/dry matter intake). The duration in which sheep were fed brassica crops was a significant contributor in the model, suggesting that the effect on emissions diminishes with time. Among a range of feed chemical composition characters, acid detergent fibre and hot-water-soluble carbohydrate contributed significantly to the model, suggesting that both structural and soluble carbohydrates affect methane formation in the rumen. There was no significant correlation between the concentration of sulphate in brassicas and emissions, but nitrate was moderately and negatively correlated with methane yield (r = −.53). Short-chain fatty acid profiles in the rumen of animals fed brassicas were different from those fed pasture, but these parameters only moderately correlated to methane emissions (r = .42). Feeding forage rape resulted in low rumen pH. The pH before morning feeding was strongly correlated to methane yield (r = .90). Rumen pH, together with microbial communities mediated by pH, might lead to low emissions. Bacteria known to produce hydrogen were relatively less abundant in the rumen contents of brassica-fed animals than pasture-fed animals. In conclusion, animal and feed factors, rumen fermentation and microbial communities all affect methane emissions to some extent. The interactions of these factors with each other thus contribute to methane emissions from brassica-fed sheep.  相似文献

Two diets differing in their degrees of ruminal degradation synchronicity between nitrogen (N) and organic matter (OM) were fed to six adult rams at a maintenance level for three months in a two‐way layout design combining a randomized block design. Adaptive changes in microbial N (MBN) supply and N balance were investigated for both diets. The in situ rumen degradation parameters of OM and N were determined for four feeds and subsequently were used to formulate the two diets to have different ruminal degradation synchronicity and to be identical with metabolizable energy (ME), metabolizable protein (MP) and ruminally degradable N concentrations. The animals were divided into two groups and fed asynchronous (ASYNC) or synchronous (SYNC) diet, respectively. On days 30, 60 and 90 of each feeding period, a five‐day metabolism trial was carried out. Animals consumed their diets thoroughly and did not show marked body weight change during the observation. The apparent digestibility of OM and N did not differ significantly between the diets throughout the periods. The MBN supply was significantly greater for ASYNC than that for SYNC (P < 0.01) and was greater on days 60 and 90 than that on day 30 (P < 0.05). Neither diets nor periods had a significant effect on the efficiency of MBN synthesis. There was a significant period effect on N retention (P < 0.05) together with tendency of diet × period interactions (P = 0.1). There was no significant dietary effect. An improvement in N retention with advancing feeding period was observed for both diets. The plasma urea concentration decreased with feeding period (P < 0.01) and there was a concomitant decrease in urinary N excretion. Plasma glucose concentration tended to increase (P = 0.07). The results suggest that lengthening feeding duration from 30 days might have caused ameliorative changes in the utilization of ME and MP, and may have improved microbe synthesis irrespective of the degradation synchronicity.  相似文献

This study aimed to investigate the effects of chitosan and whole raw soybean on nutrient intake, apparent digestibility, nitrogen utilization, microbial protein synthesis, blood metabolites, feeding behavior, ruminal fermentation, digesta kinetics, and reticular flow of nutrients of buffaloes. Four ruminally‐cannulated Murrah buffaloes (351 ± 15 kg of initial BW) were randomly assigned according to a 4 × 4 Latin square design. Treatments were arranged as 2 × 2 factorial arrangement: the first factor was whole raw soybean (WRS), and the second factor was chitosan (CHI) with or without their inclusion in diets. Intake and apparent digestibility of ether extract (p < .01; p = .04, respectively), non‐fiber carbohydrates intake (p = .03) and apparent ruminal digestibility of dry matter (p = .01) were affected by diets. An interaction effect or tendency was observed for microbial nitrogen (p = .09), concentrations, ruminal ammonia nitrogen (p = .05), total volatile fatty acid (p = .03). Association of chitosan with whole raw soybean has potential effects as a modulator of rumen fermentation; therefore, chitosan can be applied as an alternative non‐ionophore for Murrah buffaloes.  相似文献

Three yeast strains, Kluyveromyces marximanus NRRL-3234 (KM), Saccharomyces cerevisiae NCDC-42 (SC) and Saccharomyces uvarum ATCC-9080 (SU), and a mixed culture (1:1:1 ratio) were evaluated for their value as probiotics in lamb feeding in two experiment. In experiment I and II, 20 and 30 pre-weaner lambs were fed for 63 and 60 days in two and three equal groups respectively. All lambs were offered ad libitum a creep mixture and Zizyphus nummularia leaves, and yeasts were dosed orally. In experiment I, one group received no yeast, the other of the mixed culture (1.5–2 × 1010 live cells/ml). In experiment II, yeast cultivation was modified yielding 1.5–2 × 1013 live cells/ml. Lambs of the three experimental groups received 1 ml/kg live weight of one of the individual yeasts. Feed intake did not differ among groups of both experiments with the exception of SC-supplemented lambs in experiment II which showed a trend to higher intakes per kg metabolic body weight and in percentage of body weight when compared with KM- and SU-supplemented lambs. Supplementation of the mixed yeast culture had no effect on intakes of digestible crude protein and metabolisable energy, nutrient digestibility, nitrogen balance and rumen fermentation characteristics (pH, ammonia, volatile fatty acid concentration, protozoa count) and urinary allantoin as an indicator of microbial protein synthesis. The same was true for comparisons in experiment II except ciliate protozoa counts, which showed a trend to be the highest with SU and the lowest with SC. The results of present study show that the response of lambs to supplemented live yeast cultures is inconsistent, as it lacked to have an effect in the present study, and that differences among strains were small, even when supplemented at a much higher live cell count.  相似文献

对5种粗饲料,8个时间点黏附于饲料颗粒上的瘤胃微生物的羧甲基纤维素酶活的动态变化进行研究。用4只羊,分7批进行尼龙袋试验,与早晨饲喂后2h一次性投入瘤胃腹囊处,时间点取0,6,12,24,36,48,60,72h8个点。结果:对于不同饲料的瘤胃微生物(PAM)羧甲基纤维素酶活来讲,随着时间的推移,羧甲基纤维素酶活均有一个峰值出现,只是出现时间的先后顺序不一,峰值大小不一。试验中峰值出现的先后顺序为:苜蓿、披碱草、玉米秸、麦秸和谷秸(最后两者相等),时间分别为24,36,48和60h。峰值大小顺序为麦秸、玉米秸、谷秸、苜蓿、披碱草。不同饲料的平均羧甲基纤维素酶活差异显著(P<0.05),由高到低的顺序为麦秸、玉米秸、披碱草、谷秸、苜蓿。试验表明不同粗饲料对PAM羧甲基纤维素酶活有显著影响。  相似文献
