The morphology of the retinal photoreceptors of the domestic cat (Felis catus) has been investigated by light and electron microscopy. In this species the photoreceptors can be readily differentiated and adequately described by the classical terminology of rods and cones, with rods being the more numerous. Rods are long slender cells with inner and outer segments of much the same diameter. Cones are shorter cells with a conical outer segment that tapers distally and an inner segment which is wider than the outer segment. Both rods and cones are highly polarized and compartmentalized cells and consist of an outer segment, a connecting cilium, an inner segment, a nuclear region and a synaptic process leading to an expanded synaptic ending. Morphological similarities and differences are noted between the rods and cones for the various regions of these cells.  相似文献

The morphology of the light-adapted retinal photoreceptors of the American crow (Corvus brachyrhynchos) has been investigated by light and electron microscopy. They consist of rods, single cones and double (unequal) cones present in a ratio of about 4:3:3 respectively. The rods are stout cells with a long inner segment and an outer segment that reaches to the RPE cell body. In the light-adapted state, the pigment-laden apical processes of the RPE cells surround cell photoreceptor types for most of their length. The rod inner segment displays an ellipsoid of mitochondria, a large hyperboloid of glycogen, much RER, numerous polysomes, Golgi zones and autophagic vacuoles. Single cones show a slightly tapered outer segment, a large and usually heterogeneous oil droplet and an ellipsoid of mitochondria at the apex of the inner segment. Double cones consist of a longer, stouter chief member which displays a more homogeneous oil droplet and a prominent paraboloid of glycogen and a slightly shorter and thinner accessory member with no oil droplet or paraboloid. Both members of the double cone as well as the single cones show a prominent ellipsoid and plentiful polysomes, RER and Golgi zones in the inner segment. Along the length of the contiguous membranes of the two members of the double cone are presumed interreceptor junctions. All cone photoreceptors are relatively small in diameter and hence are tightly packed. Judging by their morphology in the light-adapted state neither rods nor cones are felt to undergo photomechanical movements in this species. Although the megamitochondria (MM) were localized in various pathological conditions, normal retina of some mammalian species was reported to include MM for various physiological roles. However, it was not clearly confirmed whether the MM is present in the retina of lower vertebrate as well. In this study, we tried to show the presence of the MM in the zebrafish retina using electron microscopic technique. In all the photoreceptors including rods, cones and double cones of the zebrafish retina, MM were observed in the ellipsoid of inner segment. In the photoreceptor epllipsoid of the zebrafish retina, the mitochondria located in the central portion of the ellipsoid had a highly electron-dense matrix, which were accompanied by the mitochondria with electron-lucent matrix in the apical portion of the ellipsoid. The presence of MM was more clearly discernable in the rods, which were localized under the double cones. This finding is somewhat different from those observed in the previous studies because MM were localized in the inner segment of cones, but were not in those of rods in the case of mammalian retina. Although the exact physiological meaning for the presence of MM in some vertebrate species should be further studied, the present study could show that the MM in the ellipsoid of the retinal photoreceptors was not only restricted in some mammalian species.  相似文献

The rod inner segment displays an ellipsoid of mitochondria, an hyperboloid of glycogen, much rough ER, numerous polysomes, Golgi zones and autophagic vacuoles. The rod nucleus is located deep in the outer nuclear layer and the rod synaptic pedicle displays both invaginated and superficial synaptic sites. Single cones display a slightly tapered outer segment, a large electron lucent oil droplet and an ellipsoid of mitochondria in the apex of the inner segment. Double cones consist of a long thin chief member which shows an electron dense oil droplet and a shorter, stouter accessory cone with no oil droplet but a paraboloid of glycogen below the ellipsoid. As in the single cone, polysomes, RER and Golgi zones are present in the myoid region of both members of the double cone. All photoreceptor types have a connecting cilium joining inner and outer segments. Near the external limiting membrane, the chief and accessory cones show intercellular junctions. All cone photoreceptors are relatively small in diameter and hence tightly packed. While rods are felt to undergo retinomotor movements, cones are felt to move minimally or not at all. OBJECTIVE: To determine whether apoptosis contributes to hepatocyte loss in tunicamycin poisoning. DESIGN: Groups of four guinea pigs were given 400 microg/kg of tunicamycin subcutaneously and killed at 24 h intervals up to 72 h post-injection. Livers were examined by routine histological methods and ancillary techniques (TUNEL staining, endonuclease activation, caspase activity, and electron microscopy) to ascertain whether any hepatocyte injury was apoptotic. RESULTS: Many hepatocytes exposed to tunicamycin showed evidence of apoptosis in the form of nuclear karyorrhexis with chromatin margination and crescent formation, TUNEL-positivity, DNA laddering, elevated caspase activity and apoptotic body formation. CONCLUSION: Tunicamycin caused the death of many hepatocytes in the livers of guinea pigs by apoptosis.  相似文献

Double cones consist of a larger chief member which displays a thin tapering outer segment and an electron dense oil droplet as well as a smaller accessory cone which shows no oil droplet, an ellipsoid and a paraboloid of glycogen. As in the single cone, polysomes, RER and Golgi zones are also noted in the inner segments of both members of the double cone. Near the external limiting membrane the chief and accessory cones show membrane specializations indicative of junctions on their contiguous surfaces. All cone photoreceptors are of a smaller diameter than is normally reported for avian species. Morphogenesis of the porcine retina was studied using light microscopy from 4 weeks of gestation until birth (18 to 310 mm crown-rump length), and compared with the adult stage (6 months). Tissue samples were examined from the posterior and peripheral parts of the retina. At 18 mm the retina consists of an inner marginal layer and an outer layer of neuroblastic cells. At 18-40 mm the latter layer is divided into an inner and an outer neuroblastic layer by the transient layer of Chievitz. Subsequently, the development of the different retinal layers begins at the inner retinal border and moves progressively outwards; it also spreads from the posterior to the peripheral part of the neural retina. Many cells of the inner neuroblastic layer are prospective ganglionic cells which migrate inwards, thus forming the ganglion cell layer and the inner plexiform layer at 90 mm. At 120 mm, primitive horizontal cells appear within the outer neuroblastic layer. Separation of this layer into the inner nuclear, outer plexiform and outer nuclear layers is first evident at 180 mm. At this stage all retinal layers are present, except the layer of the photoreceptor cells which is not widespread until at 220 mm. The inner and outer segments of the photoreceptor cells lengthen considerably during the last month of gestation. During the late fetal stage the nerve fiber layer, the inner and outer plexiform layers and the layer of rods and cones all continue to increase in thickness. Concurrently, the ganglion cell layer and the inner and outer nuclear layers have reached their maximal thickness and become thinner. After the total thickness of the neural retina amounts to approximately 180 microns at two to three weeks before birth, it then thins to approximately 160 microns in the adult stage.  相似文献

Amyloid precursor protein (APP) immunocytochemistry was used to study axonal and neuronal changes in guinea pig brains exposed to tunicamycin. Substantial axonal injury was found in ischemic-hypoxic foci and more generally, but this injury was not readily appreciable in conventionally stained sections. Neuronal perikaryal APP expression was also widely distributed, possibly as an acute phase response to this neurotoxin.  相似文献

To evaluate the potential role of genetic background in the susceptibility to retinal degeneration induced by N-methyl-N-nitrosourea (MNU), female rats of the Sprague-Dawley (SD), Long-Evans (LE) and Copenhagen (CH) strains were administered 50 mg/kg MNU or saline at 7 weeks of age. Retina morphology and morphometric analysis of all rats was performed 7 days after MNU administration. Atrophy of both the peripheral and central outer retina occurred in all rat strains exposed to MNU. Decreased photoreceptor cell ratio and increased retinal damage ratio were observed. The severities of the retinal atrophy were similar among all three rat strains. In conclusion, MNU-induced photoreceptor degeneration developed consistently in all three strains regardless of the absence (SD rats) or presence (LE and CH rats) of melanin in the retina, suggesting that genetic and melanin factors did not affect photoreceptor cell death after MNU.  相似文献

In experimental infections of guinea pigs with a virulent strain of Leptospira icterohaemorrhagiae widespread hemorrhages were observed. Thrombocytopenia, prolongation of prothrombin, thrombin, partial thromboplastin and coagulation times, decrease of plasma fibrinogen, factor V, factor VIII and the presence of fibrinogen degradation products were demonstrated. Treatment of infected guinea pigs with heparin prolonged life for two to three days. The histological observations revealed that the main lesion is a severe injury of the vasculature, mainly arteries, arterioles and capillaries. Most of the endothelial cells are affected or destroyed and the muscular fibers of arteries and arterioles are injured. With Martius-Scarlet-Blue, Weigert or Picro-Mallory stains it was demonstrated that the organization seen in the vessels is not all made of fibrin. The conclusion reached was that the hemorrhages observed in experimental leptospirosis in guines pigs are due to disseminated intravascular coagulation.  相似文献

Objective To demonstrate a cost‐effective, portable, and simple‐to‐use fundus imaging system for laboratory animals. Animals Studied Albino rats, pigmented mice, albino guinea pigs, and New Zealand white rabbits. Procedure A contact fundus imaging system was designed and constructed using standard optical and mechanical components: a digital camera, an otoscope, a fiber optic light source, and standard optical lenses and mounts. Digital fundus video and photography of two albino rats, two pigmented mice, two New Zealand white rabbits, and two albino guinea pigs were obtained. For all animals examined, pupils were dilated and local anesthetic was administered. Results Digital images of the fundus were obtained in all animals. Contrast of retinal vasculature and overall image quality varied from one species to another, as the axial length, ocular optics, and retinal reflectance varied significantly across species. Light intensity and focus were optimized via the light source and lens focusing mount to produce high‐quality images for each animal. Conclusions The portable, cost‐effective contact fundus imaging system was easy to use for fundus examination of laboratory animals.  相似文献

Seven-week-old male Lewis rats received a single intraperitoneal injection of N-ethyl-N-nitrosourea (ENU) (100, 200, 400 or 600 mg/kg), and retinal damage was evaluated 7 days after the treatment. Sequential morphological features of the retina and retinal DNA damage, as determined by a TUNEL assay and phospho-histone H2A.X (γ-H2AX), were analyzed 3, 6, 12, 24 and 72 hr, 7 days, and/or 30 days after 400 mg/kg ENU treatment. Activation of the nuclear enzyme poly (ADP-ribose) polymerase (PARP) was analyzed immunohistochemically by poly (ADP-ribose) (PAR) expression in response to DNA damage of the retina. All rats that received ≥ 400 mg/kg of ENU developed retinal degeneration characterized by the loss of photoreceptor cells in both the central and peripheral retina within 7 days. In the 400 mg/kg ENU-treated rats, TUNEL-positive signals were only located in the photoreceptor cells and peaked 24 hr after ENU treatment. The γ-H2AX signals in inner retinal cells appeared at 24 hr and peaked at 72 hr after ENU treatment, and the PAR signals selectively located in the photoreceptor cell nuclei appeared at 12 hr and peaked at 24 hr after ENU treatment. However, degeneration was restricted to photoreceptor cells, and no degenerative changes in inner retinal cells were seen at any time points. Retinal thickness and the photoreceptor cell ratio in the central and peripheral retina were significantly decreased, and the retinal damage ratio was significantly increased 7 days after ENU treatment. In conclusion, ENU induced retinal degeneration in adult rats that was characterized by photoreceptor cell apoptosis through PARP activity.  相似文献

Retinal degenerations in the dog and cat are an important cause of blindness in these species. Particularly in the dog, many retinal degenerations, collectively called progressive retinal atrophy, seen in clinical practice are inherited. The clinical signs, electrophysiological findings, pathology, and underlying biochemical defects in the retina vary from breed to breed. Specific categories of inherited retinal degeneration are now recognized, and classified into early onset photoreceptor dysplasias, late-onset retinal degenerations, or retinal degenerations secondary to primary RPE dystrophy. As new inherited retinal degenerations are reported in different breeds they can generally be assigned to one these categories. Other causes of retinal degeneration include nutritional deficiencies, glaucoma, inflammation, ischemia, and toxins. Idiopathic retinal degeneration occurs in the dog with some frequency.  相似文献

Ultrasonographic examination of 40 pet guinea pigs was performed in the framework of this study. Preparation of the patient for ultrasonography and the examination procedure are described. Normal ultrasonographic findings of the abdominal organs including the liver, spleen, kidney and urinary bladder are presented and illustrated for the first time in this species. Ultrasonography was found to be a useful investigative tool in the diagnosis of abdominal diseases in guinea pigs. In comparison to other diagnostic methods like physical examination including palpation, haematological examination, radiography and diagnostic laparotomy, ultrasonography offers a rapid, non-invasive and reliable means of diagnosing abdominal diseases in this species.  相似文献

Background

There is increasing interest in recording tail damage in pigs at slaughter to identify problem farms for advisory purposes, but also for benchmarking within and between countries as part of systematic monitoring of animal welfare. However, it is difficult to draw conclusions when comparing prevalence’s between studies and countries partly due to differences in management (e.g. differences in tail docking and enrichment routines) and partly due to differences in the definition of tail damage.

Methods

Tail damage and tail length was recorded for 15,068 pigs slaughtered during three and four consecutive days at two slaughterhouses in Sweden. Tail damage was visually scored according to a 6-point scale and tail length was both visually scored according to a 5-point scale and recorded as tail length in centimetres for pigs with injured or shortened tails.

Results

The total prevalence of injury or shortening of the tail was 7.0% and 7.2% in slaughterhouse A and B, respectively. When only considering pigs with half or less of the tail left, these percentages were 1.5% and 1.9%, which is in line with the prevalence estimated from the routine recordings at slaughter in Sweden. A higher percentage of males had injured and/or shortened tails, and males had more severely bitten tails than females.

Conclusions

While the current method to record tail damage in Sweden was found to be reliable as a method to identify problem farms, it clearly underestimates the actual prevalence of tail damage. For monitoring and benchmarking purposes, both in Sweden and internationally, we propose that a three graded scale including both old and new tail damage would be more appropriate. The scale consists of one class for no tail damage, one for mild tail damage (injured or shortened tail with more than half of the tail remaining) and one for severe tail damage (half or less of the tail remaining).  相似文献

The chronic ocular lesions suffered by 50 tawny owls (Strix aluco) injured by road traffic were investigated. They included non-healing corneal erosions, cataracts and retinal scarring, and periretinal membrane formation. The intraocular pressure was significantly lower in the eyes with intraocular inflammatory pathology and higher in the eyes with irido- or cyclodialysis than in ophthalmically normal eyes. Cicatritial retinal lesions were not associated with high titres of antibodies to Toxoplasma species. The findings correlate with those observed in human beings with eye injuries due to high-speed blunt trauma.  相似文献

Blockade of potassium channels with 4-aminopryidine (4-AP) restores conduction to demyelinated axons and improves function. Unfortunately, 4-AP causes adverse effects and its clinical effects are unpredictable and limited. Derivatives of 4-AP have been tested in models of spinal cord injury in guinea pigs; three derivatives (methyl-, ethyl- and t -butyl carbamate derivatives) showed promise. This study investigates the safety and pharmacokinetics of these derivatives in dogs. Each derivative was administered orally to dogs starting at doses below effective doses in guinea pigs, and increasing the dose on sequential days. Routine blood work was performed prior to and 24 h after drug administration, blood samples were collected at intervals over 24 h after drug administration, and dogs were monitored for side effects. Derivative plasma levels were determined using high-pressure liquid chromatography. Cerebrospinal fluid (CSF) samples were taken to determine CSF levels. No adverse effects were seen even when using doses higher than those that improved conduction in spinal cord injured guinea pigs. Peak plasma levels occurred at 36.6 (ethyl), 87 ( t -butyl) and 175 (methyl) min and plasma level was related to drug dose. Penetration of the central nervous system (CNS) was good, with CSF levels higher than plasma levels for the t -butyl derivative.  相似文献

【目的】 研究H9N2亚型禽流感病毒(Avian influenza virus,AIV)对哺乳动物的传染性和致病性。【方法】 采集样品进行病毒的分离鉴定,将分离到的病毒以300 μL/只(107 EID50)的剂量通过滴鼻和肺递送方式感染豚鼠,每组各15只,对2组感染效果进行比较,然后通过分离株在豚鼠中的传播能力试验验证其气溶胶传播能力,通过间接免疫荧光试验检测分离株在人支气管上皮细胞上的复制能力。【结果】 试验分离到1株H9N2亚型AIV,命名为SD18,病毒通过肺递送和滴鼻2种攻毒方式感染豚鼠后,肺递送组的排毒量显著高于滴鼻组(P<0.05)。通过对豚鼠肺脏相关模式识别受体和抗病毒蛋白的表达检测发现,2种攻毒方式都能诱导相关的免疫因子Toll样受体3(TLR3)、TLR7、抗黏病毒蛋白(MX)和2',5'-腺苷酸激酶(OAS)表达量极显著上调(P<0.01);2组在攻毒后第6天产生抗体,并呈上升趋势。对SD18 株在豚鼠中的传播能力验证发现,SD18株具有通过直接接触和飞沫传播感染豚鼠的能力,但并未证实气溶胶的产生和传播。对SD18株感染人支气管上皮细胞BEAS-2B后发现,在感染后12 h可检测到病毒,24 h时病毒在细胞内复制能力最强,TLR3、TLR7、MX、OAS、白介素6(IL-6)、IL-8、干扰素-β(IFN-β)相关的免疫因子表达量极显著上调(P<0.01)。【结论】 H9N2亚型AIV SD18株的跨种传播能力变强,具有不经提前适应便可直接感染豚鼠的能力;SD18株具有通过飞沫传播感染豚鼠并使其产生抗体的能力且SD18株可在人支气管上皮细胞BEAS-2B上进行复制。  相似文献

The hematologic response in various animal species fed cholesterol and fat differed with the species and sex. Male rabbits, chickens and guinea pigs could not adequately comensate for the demand for increased red cell production and developed anemia. Female guinea pigs given added cholesterol and fat maintained adequate blood cell production but may have become anemic if the test period had been extended. Bone marrow production was increased in male rats and was sufficient to compensate for red cell loss. Blood and bone marrow values were not altered in female chickens by the dietary treatment. Fatty changes in the liver were common to all species fed added cholesterol and fat but gastric lesions were seen only in the rabbit.  相似文献

Hematology and serum chemistry measurements were performed on blood specimens from 12 male Dunkin-Hartley hairless guinea pigs Crl:IAF(HA)BR and 10 haired Dunkin-Hartley male guinea pigs Crl:(HA)BR. Significantly higher activities of alanine aminotransferase, aspartate aminotransferase, amylase, and creatine kinase were observed in the hairless guinea pigs as compared to the haired strain. Alkaline phosphatase activity was found to be lower in the hairless guinea pig. The hairless guinea pigs were found to have serum urea concentrations approximately 46% higher than the normal guinea pig strain. The erythrocytic mean cell volume of the hairless strain was found to be smaller, with a greater hemoglobin content. Hairless guinea pigs were found to have approximately 40% fewer leukocytes with a reversed lymphocyte:neutrophil ratio compared to the haired guinea pigs which had much higher lymphocyte counts.  相似文献
